Abstract

Small molecules can display certain electronic and structural features that enable their use for metal-sensing applications in different fields. Of the reported metal sensors, there has been increasing interest in copper sensing in the past decade, given the biological importance of copper as well as its presence as a potential contaminant in water and fuels. Molecules used for copper(II) sensing generally consist of a fluorophore/chromophore and a ligand for selective metal ion recognition. This review article focuses on literature contributions since the year 2010 concerning small molecule copper(II) sensors that provide a naked-eye color response in solution. We present molecular structural features and sensing mechanisms for the colorimetric and fluorometric detection of copper(II) ions in different environmental, agricultural, and biological samples. In addition, the sensing performance of these chemosensors is compared and discussed, which could aid in the future design of chemosensors for copper(II). Finally, we outline the challenges and future prospects of fluorophore/chromophore–ligand chemistry in applications of small molecules for fluorometric and colorimetric assays of copper(II).

Abstract

The metal-binding capabilities of the spiropyran family of molecular switches have been explored for several purposes from sensing to optical circuits. Metal-selective sensing has been of great interest for applications ranging from environmental assays to industrial quality control, but sensitive metal detection for field-based assays has been elusive. In this work, we demonstrate colorimetric copper sensing at low micromolar levels. Dimethylamine-functionalized spiropyran (SP1) was synthesized and its metal-sensing properties were investigated using UV-vis spectrophotometry. The formation of a metal complex between SP1 and Cu2+ was associated with a color change that can be observed by the naked eye as low as ≈6 μM and the limit of detection was found to be 0.11 μM via UV-vis spectrometry. Colorimetric data showed linearity of response in a physiologically relevant range (0-20 μM Cu2+) with high selectivity for Cu2+ ions over biologically and environmentally relevant metals such as Na+, K+, Mn2+, Ca2+, Zn2+, Co2+, Mg2+, Ni2+, Fe3+, Cd2+, and Pb2+. Since the color change accompanying SP1-Cu2+ complex formation could be detected at low micromolar concentrations, SP1 could be viable for field testing of trace Cu2+ ions.

Abstract

Acute flaccid myelitis (AFM) has caused motor paralysis in >560 children in the United States since 2014. The temporal association of enterovirus (EV) outbreaks with increases in AFM cases and reports of fever, respiratory, or gastrointestinal illness prior to AFM in >90% of cases suggest a role for infectious agents. Cerebrospinal fluid (CSF) from 14 AFM and 5 non-AFM patients with central nervous system (CNS) diseases in 2018 were investigated by viral-capture high-throughput sequencing (VirCapSeq-VERT system). These CSF and serum samples, as well as multiple controls, were tested for antibodies to human EVs using peptide microarrays. EV RNA was confirmed in CSF from only 1 adult AFM case and 1 non-AFM case. In contrast, antibodies to EV peptides were present in CSF of 11 of 14 AFM patients (79%), significantly higher than controls, including non-AFM patients (1/5 [20%]), children with Kawasaki disease (0/10), and adults with non-AFM CNS diseases (2/11 [18%]) (P = 0.023, 0.0001, and 0.0028, respectively). Six of 14 CSF samples (43%) and 8 of 11 sera (73%) from AFM patients were immunoreactive to an EV-D68-specific peptide, whereas the three control groups were not immunoreactive in either CSF (0/5, 0/10, and 0/11; P = 0.008, 0.0003, and 0.035, respectively) or sera (0/2, 0/8, and 0/5; P = 0.139, 0.002, and 0.009, respectively).IMPORTANCE The presence in cerebrospinal fluid of antibodies to EV peptides at higher levels than non-AFM controls supports the plausibility of a link between EV infection and AFM that warrants further investigation and has the potential to lead to strategies for diagnosis and prevention of disease.

Abstract

Nanoparticles have emerged as versatile tools in the realm of thermal therapy, offering precise control and feedback mechanisms for targeted treatments. This review explores the intersection of nanotechnology and thermal therapy, focusing on the utilization of nanoparticles for image-guided interventions and temperature monitoring. Starting with an exploration of local temperature dynamics compared to whole-body responses, we delve into the landscape of nanomaterials and their pivotal role in nanomedicine. Various physical stimuli employed in therapy and imaging are scrutinized, laying the foundation for nanothermal therapies and the accompanying challenges. A comprehensive analysis of nanomaterial architecture ensues, delineating the functionalities of magnetic, plasmonic, and luminescent nanomaterials within the context of thermal therapies. Nano-design intricacies, including core–shell structures and monodisperse properties, are dissected for their impact on therapeutic efficacy. Furthermore, considerations in designing in vivo nanomaterials, such as hydrodynamic radii and core sizes at sub-tissue levels, are elucidated. The review then delves into specific modalities of thermally induced therapy, including magnetically induced hyperthermia and luminescent-based thermal treatments. Magnetic hyperthermia treatment is explored alongside its imaging and relaxometric properties, emphasizing the implications of imaging formulations on biotransformation and biodistribution. This review also provides an overview of the magnetic hyperthermia treatment using magnetic nanoparticles to induce localized heat in tissues. Similarly, optical and thermal imaging techniques utilizing luminescent nanomaterials are discussed, highlighting their potential for light-induced thermal therapy and cellular-level temperature monitoring. Finally, the application landscape of diagnosis and photothermal therapy (PTT) is surveyed, encompassing diverse areas such as cancer treatment, drug delivery, antibacterial therapy, and immunotherapy. The utility of nanothermometers in elucidating thermal relaxation dynamics as a diagnostic tool is underscored, alongside discussions on PTT hyperthermia protocols. Moreover, the advancements in nanoparticle magnetic imaging and implications of imaging formulations especially in creating positive MRI contrast agents are also presented. This comprehensive review offers insights into the evolving landscape of nanoparticle-based image-guided thermal therapies, promising advancements in precision medicine and targeted interventions, underscoring the importance of continued research in optimization for the full potential of magnetic hyperthermia to improve its efficacy and clinical translation. Abstract

No discussion of challenges for chemistry in molecular imaging would be complete without addressing the elephant in the room-which is that the purest of chemical compounds needs to interact with a biological system in a manner that does not perturb normal biology while still providing efficacious feedback to assist in diagnosis of disease. In the past decade, magnetic resonance imaging (MRI) agents long considered inert have produced adverse effects in certain patient populations under certain treatment regimens. More recently, inert blood pool agents have been found to deposit in the brain. Release of free metal is often suspected as the culprit but that hypothesis has yet to be validated. In addition, even innocuous agents can cause painful side effects during injection in some patients. In this brief review, we summarize known biological effects for gadolinium- and iron-based MRI contrast agents, and discuss some of the potential mechanisms for the observed biological effects, including the potential role of phosphorus imbalance, related to kidney disease or cancer, in destabilizing gadolinium-based chelates and precipitating free gadolinium.This article is part of the themed issue 'Challenges for chemistry in molecular imaging'.

Abstract

In this work we synthesize molecular switches that are responsive to cysteine, homocysteine, and glutathione; three redox systems that make up the majority of the body's antioxidant defenses. Synthesized spiropyran isomers with conjugation-ready linkages showed good selectivity of response to these major antioxidant thiols over nucleophilic amino acids; however the position of the linking group can affect selectivity and reversibility of the switching response. An isomer with selectivity for cysteine against GSH and Hcy was identified.

Abstract

It is currently unclear whether changes in viral communities will ever be predictable. Here we investigate whether viral communities in wildlife are inherently structured (inferring predictability) by looking at whether communities are assembled through deterministic (often predictable) or stochastic (not predictable) processes. We sample macaque faeces across nine sites in Bangladesh and use consensus PCR and sequencing to discover 184 viruses from 14 viral families. We then use network modelling and statistical null-hypothesis testing to show the presence of non-random deterministic patterns at different scales, between sites and within individuals. We show that the effects of determinism are not absolute however, as stochastic patterns are also observed. In showing that determinism is an important process in viral community assembly we conclude that it should be possible to forecast changes to some portion of a viral community, however there will always be some portion for which prediction will be unlikely.

Abstract

This review focuses on the green synthesis methods, challenges, and applications of manganese oxide (Mn3O4) nanoparticles investigated in the past five years. Mn3O4 nanoparticles offer some unique properties that are attributed in part to the presence of mixed oxidation states of manganese (i.e. +2 and +3) in the particle, which can be utilized in a wide range of redox-sensitive applications, such as in developing supercapacitive energy storage materials. In addition, the green synthesis of Mn3O4 nanoparticles through plant extracts has potential uses in sustainable nanotechnology. Various plant extract-mediated synthesis techniques for Mn3O4 nanoparticles have been investigated and presented. By comparing the size and structure of the synthesized Mn3O4 nanoparticles, we have observed a consistent pattern of obtaining spherical particles with a size ranging from 16 to 50 nm. The morphology of the generated Mn3O4 nanoparticles can be influenced by the annealing temperature and the composition of the plant extract used during the nanoparticle synthesis. Additionally, numerous applications for the greenly produced Mn3O4 nanoparticles have been demonstrated. Mn3O4 nanoparticles derived from plant extracts have been found to possess antimicrobial properties, supercapacitive and electrochemical capabilities, and excellent pollutant degradation efficiency. However, the magnetic properties of these nanoparticles synthesized by plant extracts are yet to be explored for potential biomedical applications. Finally, challenges to existing synthetic methods and future perspectives on the potential applications of these green synthesized Mn3O4 nanoparticles are highlighted. [ABSTRACT FROM AUTHOR]

34. Rare Case of Disseminated Histoplasmosis Mitral Valve Endocarditis in Florida.

Abstract

Objective: Rare disease Background: Histoplasma capsulatum is prevalent in the mid-eastern United States and is an environmental fungus that causes human infection by the inhalation of its spores. It is commonly associated with areas containing large amounts of bird excrement and can survive for years in the soil. Only 1% of infected individuals develop disseminated histoplasmosis or Histoplasma endocarditis. Case Report: A 61-year-old man with atrial fibrillation had 8 months of fatigue, low-grade fevers, night sweats, and unexplained weight loss presented to the Emergency Department. He worked and lived in Central Florida and although he raised cattle, he denied exposure to birds or bats with regularity. A transesophageal echocardiogram confirmed a sessile echo density on the atrial surface of the mitral valve. His microbial Karius cell-free DNA test from his blood sample was positive for Histoplasma capsulatum, and he was immediately given intravenous liposomal amphotericin for 2 weeks. A tissue valve was used to successfully replace his mitral valve along with a coronary artery bypass and a maze procedure for his persistent atrial fibrillation and atrial futter. The diagnosis of mitral valve endocarditis from disseminated histoplasmosis was confirmed by pathological analysis, and he was sent home on long-term itraconazole maintenance treatment. Conclusions: Surgical intervention in combination with anti-fungal medication can be a lifesaving intervention for disseminated histoplasmosis. A thorough history is particularly important when evaluating a patient with an unknown infectious source, especially assessing for risk factors, including exposure to environmental factors, workplace, and animals. 49. Risk to Resilience: Exploring Protective Factors for Students Experiencing Homelessness at a Traditional High School and a Modified Comprehensive School

Abstract

In 2008-2009, almost one million children experiencing homelessness were enrolled in school; this number has increased by 41% between 2009 and 2011. Unfortunately, this trend has continued to increase; according to the most recent data from the National Center for Homeless Education, the number of homeless students has reached a record of 1,056,794. These students are one of the most marginalized and victimized populations in schools. They experience more daily stressors and are more at risk than their housed peers. Unfortunately, there is a dearth in educational research about the conditions, within the school context, that promote resilience in this population that is highly at-risk. Using a conceptual framework that draws upon literature on educational risks, resilience, school structures, and school climate with respect to homeless students, this comparative qualitative case study investigated the protective factors offered by two school models for homeless students. The study compared the institutional structures, school climate, and experiences of students served by distinct types of programs: a traditional school and an alternative school designed specifically for homeless children and youth. By examining the impact of different school settings, the study contributes to a better understanding of the challenges, successes, and recommended research-based interventions designed to help a vulnerable student population. Findings suggest resilience-promoting protective factors recommended for a general student population apply to homeless students. Abstract

The following papers is a technical note about a new endoscopic access to lateral ventricle through occipital horn to place ventricles catheters, 20 patient was operated using Deck System of endoscopy with lens of 30 grades, 2,7 mm of diameter and 30 cm of larger, sheets and working canals including. CT scan was performing 24 hours after surgery and optimal position was obtained in 95%. Median distance from occipital bon to frontal horn of the ventricles was 10,5 cm. Any patient present dysfunction of the system and only one patient had a hematoma of the surgical area. Key words: Ventricular- peritoneal shunt, ventricular catheters, occipital horn. El siguiente trabajo constituye una nota tecnica sobre un nuevo proceder endoscopico de colocacion de cateteres ventriculares permanente a traves del cuerno occipital. Se colocaron 20 cateteres permanentes utilizando el sistema DECK con un endoscopio Hopkins II de 30[degrees], diametro 2,7 mm y longitud 30 cm, con vainas y canales de trabajos. La posicion del cateter fue evaluada por tomografia de craneo realizada a las 24 horas de la cirugia, obteniendose una posicion a (optima) en el 95% de los casos, con una distancia promedio de 10,5 cm desde la tabla externa del hueso occipital al cuerno frontal. En ninguno de los casos disfunciono el sistema derivativo y un solo paciente presento como complicacion un hematoma del lecho quirurgico.
