Background and AimsThe constitutive androstane receptor (CAR) is a nuclear receptor able to recognize a large panel of xenobiotics leading to the modulation of the expression of its target genes involved in xenobiotic detoxication and energy metabolism. While CAR hepatic activity is thought to be higher in women than in men, its response to an acute pharmacological activation has never been investigated in both sexes.MethodsHepatic transcriptome, plasma and hepatic metabolome, have been analyzed inCar+/+andCar-/-male and female mice treated either with the CAR-specific agonist, 1,4-bis[2-(3,5-dichloropyridyloxy)]benzene (TCPOBOP), or with vehicle.ResultsWhile 90% of TCPOBOP-sensitive genes were modulated in a sex- independent way, the remaining 10% were almost exclusively impacted in female liver specifically. These female-specific CAR-sensitive genes were mainly involved in xenobiotic metabolism, inflammation and extracellular matrix organization. CAR activation also induced higher hepatic oxidative stress and hepatocyte cytolysis in females than in males. Data mining on human data confirmed that CAR activation may be involved in sexually-dimorphic drug-induced liver injury. Hepatic expression of flavin monooxygenase 3(Fmo3)was almost abolished and associated with a decrease of hepatic trimethylamine-N-oxide (TMAO) concentration in TCPOBOP-treated females. In line with a possible role in the control of TMAO homeostasis, CAR activation decreased platelet hyperresponsiveness in female mice supplemented with dietary choline.ConclusionsOur results demonstrate that more than 10% of CAR-sensitive genes are sex-specific and influence hepatic and systemic response such as platelet aggregation. Also, CAR activation may be an important mechanism of sexually- dimorphic drug-induced liver injury.

La croissance osseuse des vertébrés ectothermes et notamment des actinoptérygiens est discontinue au cours de la vie de l’animal suivant les conditions du milieu et des contrôles physiologiques internes : ainsi, la croissance osseuse reflète une partie de son histoire de vie. Elle voit des phases plus ou moins rapides alterner avec des phases dites d’arrêt soulignées par des lignes dites d’arrêt de croissance (LAC), visibles lorsque l’os est observé en lame mince. En zone tropicale, la croissance se produit en saison des pluies chez de nombreux poissons de lac et de rivière, tandis que les LAC se mettent en place à la saison sèche, lorsque les ressources ne sont pas suffisantes pour permettre la croissance (et sont entièrement dédiées à la survie de l’individu) ou encore lorsque les ressources sont suffisantes mais allouées à d’autres fonctions, par exemple à la reproduction ou à la guérison. Le rythme de la croissance osseuse chez ces espèces reflèterait donc en partie le rythme des saisons au long de la vie de l’animal. Si c’est bien le cas, l’étude de la croissance de pièces osseuses fossiles permettrait de reconstituer le rythme des saisons qui prévalait à l’époque où vivait le spécimen fossilisé, ce qui serait une nouvelle façon de renseigner les paléoclimats. Néanmoins, ce type d’interprétation implique une validation dans l’actuel chez des taxons proches. Dans cet objectif, nous avons testé la cohérence du signal de croissance d’une part à l’échelle d’un individu de perche du Nil (Lates niloticus), au sein et entre différentes pièces squelettiques, et d’autre part avec les données climatiques connues au long de sa vie (température et précipitations). Cette espèce est choisie car elle est bien représentée dans tous les sites à ichtyofaune depuis le Miocène en Afrique et parce qu’elle vit dans les eaux ouvertes des réseaux hydrographiques de la zone nilo-soudanaise. L’individu a été pêché dans le bassin du lac Tchad dans les environs de N’Djaména (dans le Chari) et acheté au marché aux poissons de N’Djaména en juin 2014. Nous avons sélectionné les pièces osseuses les plus fréquemment collectées et permettant une identification spécifique (prémaxillaire, maxillaire, operculaire, plusieurs épines et vertèbres), ainsi que le lacrymal et des écailles. La cyclicité isotopique (mesure des valeurs de δ18O dans l’otolithe) et de la taille des pièces squelettiques permettent de situer l’âge de l’individu entre 4 et 7 ans. Nous avons comparé le rythme de croissance osseuse entre les pièces et aux fluctuations saisonnières de température et de pluies au long de la vie de l’animal. Il en résulte que la croissance se fait au même rythme entre différentes zones d’une même pièce squelettique osseuse, entre pièces d’un même type (vertèbres, épines, écailles) et entre différentes pièces dans un même organisme. Néanmoins certaines LAC mineures ne sont pas visibles sur les écailles. De plus, chez cet individu, le rythme de croissance osseuse des cinq dernières années est fortement corrélé au rythme des saisons : la croissance est plus faible pendant les années montrant des températures et des précipitations plus faibles.En conséquence, il apparait que l’on peut utiliser les pièces fossiles de L. niloticus pour reconstituer la saisonnalité qui prévalait dans les localités néogènes et quaternaires d’Afrique au moment de leur dépôt.

Growth in ectotherm vertebrates, especially actinopterygians, is strongly rhythmed by seasonal variation in environmental conditions and by internal physiological controls. Skeletal growth refects the somatic growth rhythm fluctuation by the alternating deposition of different growth marks, observable on thin sections. In tropical regions, growth occurs during the rainy season in many lake and river shes, while it stops during the dry season when resources are not suffcient or when energy is allocated to other functions (e.g., reproduction or healing). The growth rhythm in tropical species would therefore refect the environmental seasonal fluctuations experienced by individuals in their lifetime. Therefore, the study of the growth recorded in fossil bones would allow tracking past seasonal regimes, which would constitute a new way of inferring paleoclimates. However, this approach rst requires to be validated in extant species which are closely related to the studied fossil taxa. For this purpose, we examined the skeletal growth marks of a Nile perch (Lates niloticus) caught in June 2014 in the Chadian Chari River. This species is well represented in African fossil sites since the Miocene. Growth marks were compared within bones and between different bones, and with climatic conditions experienced during life span. Growth occurred at the same rate between the different areas of the same bone and between the different bones (vertebrae, spines, scales). The individual was estimated to be 4-year-old at time of capture. The skeletal growth rate over the past three years was strongly correlated with the environmental seasonal fluctuations, i.e., a small then a large rainy season each year. Consequently, it appears that any fossil record of Nile perch can be used to reconstruct the seasonality that prevailed in the Neogene and Quaternary localities of Africa.

International audience; Abundant and diverse fossil assemblages were recently collected from Albian-Cenomanian (mid-Cretaceous) cross-stratified beds filling a Bajocian (Middle Jurassic) paleokarst exposed near the town of Persac, Vienne, western France. They include wellpreservedfossils of bivalves, gastropods, charophytes, plant leaves and wood, bony fishes, lissamphibians, turtles, lizards, crocodyliforms, dinosaurs, pterosaurs, and mammals. During the last three years 12 m3 of sediments were screen-washed. More than 200,000 micro-remains have already been sorted out, ranging from a few mm to a 10 cm in size.The fauna consists of both freshwater and terrestrial organisms. Itoffers, thus, an inestimable milestone of scientific and patrimonial value for the mid-Cretaceous European continental ecosystems.

International audience; We here report on fossil remains of the earliest known crown-Testudo, an extant clade of Mediterranean testudinid tortoises from the late Miocene (Vallesian, MN 10) from the hominoid locality Ravin de la Pluie (RPl) in Greece. The material studied is a small, nearly complete carapace with a clearly distinct hypo-xiphiplastral hinge. This supports the sensu stricto generic assignment. This new terrestrial testudinid specimen is characterized by a possible tectiform, narrow, elongated shell with a pentagonal pygal and a long, posteriorly elevated, lenticular and rounded dorsal epiplastral lip. These unique features differ from those of other known Mediterranean hinged forms and allow the erection of the new species Testudo hellenica sp. nov. This taxon is phylogenetically close to two Greek species, the extant T. marginata and the fossil T. marmorum (Turolian, around 7.3 Ma). This record provides evidence for the first appearance of the genus Testudo sensu stricto at a minimum age of 9 Ma.
