Abstract

Introduction: Neurogenesis is tightly regulated in space and time, ensuring the correct development and organization of the central nervous system. Critical regulators of brain development and morphogenesis in mice include two members of the p53 family: p53 and p73. However, dissecting the in vivo functions of these factors and their various isoforms in brain development is challenging due to their pleiotropic effects. Understanding their role, particularly in neurogenesis and brain morphogenesis, requires innovative experimental approaches. Methods: To address these challenges, we developed an efficient and highly reproducible protocol to generate mouse brain organoids from pluripotent stem cells. These organoids contain neural progenitors and neurons that self-organize into rosette-like structures resembling the ventricular zone of the embryonic forebrain. Using this model, we generated organoids from p73-deficient mouse cells to investigate the roles of p73 and its isoforms (TA and DNp73) during brain development. Results and Discussion: Organoids derived from p73-deficient cells exhibited increased neuronal apoptosis and reduced neural progenitor proliferation, linked to compensatory activation of p53. This closely mirrors previous in vivo observations, confirming that p73 plays a pivotal role in brain development. Further dissection of p73 isoforms function revealed a dual role of p73 in regulating brain morphogenesis, whereby TAp73 controls transcriptional programs essential for the establishment of the neurogenic niche structure, while DNp73 is responsible for the precise and timely regulation of neural cell fate. These findings highlight the distinct roles of p73 isoforms in maintaining the balance of neural progenitor cell biology, providing a new understanding of how p73 regulates brain morphogenesis. Abstract

Peptides are bioactive molecules whose functional versatility in living organisms has led to successful applications in diverse fields. In recent years, the amount of data describing peptide sequences and function collected in open repositories has substantially increased, allowing the application of more complex computational models to study the relations between the peptide composition and function. This work introduces AMP-Detector, a sequence-based classification model for the detection of peptides' functional biological activity, focusing on accelerating the discovery and de novo design of potential antimicrobial peptides (AMPs). AMP-Detector introduces a novel sequence-based pipeline to train binary classification models, integrating protein language models and machine learning algorithms. This pipeline produced 21 models targeting antimicrobial, antiviral, and antibacterial activity, achieving average precision exceeding 83%. Benchmark analyses revealed that our models outperformed existing methods for AMPs and delivered comparable results for other biological activity types. Utilizing the Peptide Atlas, we applied AMP-Detector to discover over 190,000 potential AMPs and demonstrated that it is an integrative approach with generative learning to aid in de novo design, resulting in over 500 novel AMPs. The combination of our methodology, robust models, and a generative design strategy offers a significant advancement in peptide-based drug discovery and represents a pivotal tool for therapeutic applications. Abstract

Glioblastoma (GB) is the most common and fatal type of primary malignant brain tumor for which effective therapeutics are still lacking. GB stem cells, with tumor‐initiating and self‐renewal capacity, are mostly responsible for GB malignancy, representing a crucial target for therapies. The TP73 gene, which is highly expressed in GB, gives rise to the TAp73 isoform, a pleiotropic protein that regulates neural stem cell biology; however, its role in cancer has been highly controversial. We inactivated TP73 in human GB stem cells and revealed that TAp73 is required for their stemness potential, acting as a regulator of the transcriptional stemness signatures, highlighting TAp73 as a possible therapeutic target. As proof of concept, we identified a novel natural compound with TAp73‐inhibitory capacity, which was highly effective against GB stem cells. The treatment reduced GB stem cell‐invasion capacity and stem features, at least in part by TAp73 repression. Our data are consistent with a novel paradigm in which hijacking of p73‐regulated neurodevelopmental programs, including neural stemness, might sustain tumor progression, pointing out TAp73 as a therapeutic strategy for GB. 15. The Role of Relational Reciprocity: How Students' Families Support and Influence Them during the Transition to Higher Education.

Abstract

This work is embedded in a larger qualitative study and uses grounded theory techniques to explore the reciprocal nature of familial relationships for racially and ethnically minoritized students and, in turn, how these relationships impact students' higher education experience. After analyzing semi-structured interview data using grounded theory approaches like open coding and constant comparison, themes of genuine and resistant reciprocity emerged. The findings showed that genuine familial reciprocity led to positive influences, while resistant reciprocity detracted students from their higher education journeys. Genuine reciprocity covered mutual exchanges that included emotional, temporal, and financial support. Resistant reciprocity centered on familial exchanges that left students feeling obligated to support their family financially and/or temporally. Many students used resistant reciprocity as motivation to persist despite their personal or familial circumstances. These findings contribute to a framework of relational reciprocity for college students and their families. Relational reciprocity illuminates the nature of and types of exchanges that contribute to students' persistence during higher education, whether propelled by or in spite of them. Abstract

Marine diazotrophs convert dinitrogen (N2) gas into bioavailable nitrogen (N), supporting life in the global ocean. In 2012, the first version of the global oceanic diazotroph database (version 1) was published. Here, we present an updated version of the database (version 2), significantly increasing the number of in situ diazotrophic measurements from 13 565 to 55 286. Data points for N2 fixation rates, diazotrophic cell abundance, and nifH gene copy abundance have increased by 184 %, 86 %, and 809 %, respectively. Version 2 includes two new data sheets for the nifH gene copy abundance of non-cyanobacterial diazotrophs and cell-specific N2 fixation rates. The measurements of N2 fixation rates approximately follow a log-normal distribution in both version 1 and version 2. However, version 2 considerably extends both the left and right tails of the distribution. Consequently, when estimating global oceanic N2 fixation rates using the geometric means of different ocean basins, version 1 and version 2 yield similar rates (43–57 versus 45–63 Tg N yr−1 ; ranges based on one geometric standard error). In contrast, when using arithmetic means, version 2 suggests a significantly higher rate of 223±30 Tg N yr−1(mean ± standard error; same hereafter) compared to version 1 (74 ± 7 Tg N yr−1). Specifically, substantial rate increases are estimated for the South Pacific Ocean (88±23 versus 20±2 Tg N yr−1), primarily driven by measurements in the southwestern subtropics, and for the North Atlantic Ocean (40 ± 9 versus 10 ±2 Tg N yr−1). Moreover, version 2 estimates the N2 fixation rate in the Indian Ocean to be 35 ± 14 Tg N yr−1,which could not be estimated using version 1 due to limited data availability. Abstract

Uveal melanoma (UM) is the most common primary malignancy of the adult eye but lacks any FDA-approved therapy for the deadly metastatic disease. Thus, there is a great need to dissect the driving mechanisms for UM and develop strategies to evaluate potential therapeutics. Using an autochthonous zebrafish model, we previously identified MITF, the master melanocyte transcription factor, as a tumor suppressor in GNAQQ209L -driven UM. Here, we show that zebrafish mitfa-deficient GNAQQ209L -driven tumors significantly up-regulate neural crest markers, and that higher expression of a melanoma-associated neural crest signature correlates with poor UM patient survival. We further determined how the mitfa-null state, as well as expression of GNAQQ209L , YAPS127A;S381A , or BRAFV600E oncogenes, impacts melanocyte lineage cells before they acquire the transformed state. Specifically, examination 5 days post-fertilization showed that mitfa-deficiency is sufficient to up-regulate pigment progenitor and neural crest markers, while GNAQQ209L expression promotes a proliferative phenotype that is further enhanced by YAPS127A;S381A co-expression. Finally, we show that this oncogene-induced proliferative phenotype can be used to screen chemical inhibitors for their efficacy against the UM pathway. Overall, this study establishes that a neural crest signature correlates with poor UM survival, and describes an in vivo assay for preclinical trials of potential UM therapeutics.

Abstract

The main objective of this study was to examine the explanatory models of secondary school students, about the structure of the animal eukaryotic cells before and after an didactic intervention, based from the cognitive model of science (Giere, 1992) and the constructivist learning cycle (Sanmartí, 2000). The research took place in two stages: a. Descriptive and interpretative stage, we categorized the explanatory models identified in 18 families of models and b. The pre-experimental stage, it identifies whether there were significant differences between the models before and after the educational intervention by Wilcoxon test and data randomization, ANOVA. The main findings showed that didactic intervention encourages the building of explanatory models, which are characterized by more specialized language, new relationships between the organelles and cellular functions and the ability to incorporate new elements to enrich the school cell model, demonstrating that these models evolve progressively (RodrÍguez and Moreira, 1999; Justi, 2006).

Abstract

El uso de árboles leguminosos como sombra para cafetales podría ser parte de futuras estrategias agrícolas en el trópico para enfrentar el cambio climático. El llamado “guamo” (Inga spp., Fabaceae) es una leguminosa beneficiosa utilizada tradicionalmente para amortiguar el microclima y mejorar las propiedades del suelo. Sin embargo, aún no se sabe si la cercanía de las plántulas de guamo sembradas trae beneficios a los cultivos de café cuando se establecen o si este beneficio llega con el tiempo. Para examinar si la proximidad del guamo afectó el crecimiento de las plántulas de café bajo diferentes condiciones de fertilización, sembramos 360 plántulas de guamo de tres meses de edad y 90 plántulas de café de la misma edad en una parcela de 1800 m2 utilizando un diseño aleatorio estratificado de 90 parcelas. y 270 subtramas. Asignamos al azar tres tratamientos (guamo, café, y testigo sin fertilizante), dos fertilizantes (gallinaza comercial y copolímero orgánico), y el testigo. Se tomaron medidas de crecimiento mensual durante un año. Las plántulas crecieron más con gallinaza (23,4% y 27,8% de diámetro mayor que la obtenida con el copolímero y el testigo, respectivamente) mientras que la altura fue 19,9% mayor que la obtenida con el copolímero y 22,6% mayor que la del testigo. La evidencia respalda que el estiércol de pollo fue un fertilizante orgánico eficaz y que la presencia de guamo no benefició (vía facilitación) ni perjudicó (vía competencia interespecífica) a los árboles jóvenes de café durante el establecimiento de la plantación. 4% y 27,8% mayor de diámetro que el obtenido con el copolímero y el control, respectivamente) mientras que la altura fue 19,9% mayor que la obtenida con el copolímero y 22,6% mayor que el control. The use of leguminous trees as shade for coffee plantations could be part of future agricultural strategies in the tropics to face climate change. The so-called “guamo” (Inga spp., Fabaceae) is a beneficial leguminous tree traditionally used for buffering microclimate and improving soil properties. However, it is not yet known whether the closeness of planted guamo seedlings brings benefits to coffee crops when they are established or whether this benefit comes with time. To examine whether the proximity of guamo affected the growth of coffee seedlings under different fertilization conditions, we planted 360 three-month-old guamo seedlings and 90 coffee seedlings of the same age in an 1800 m2 plot using a stratified randomized design consisting of 90 plots and 270 subplots. We randomly assigned three treatments (guamo, coffee, and control without fertilizer), two fertilizers (commercial chicken manure and organic copolymer), and the control. Monthly growth measures were taken for a year. Seedlings grew the most with chicken manure (23.4% and 27.8% greater diameter than that obtained with the copolymer and the control, respectively) while the height was 19.9% greater than that obtained with thecopolymer and 22.6% greater than the control. The evidence supports that chicken manure was an effective organic fertilizer and the presence of guamo neither benefitted (via facilitation) nor damaged (via interspecific competition) the coffee saplings during the establishment of the plantation.
