Abstract

Upconversion nanoparticles (UCNPs) are under consideration for their use as bioimaging probes with enhanced optical performance for real time follow-up under non-invasive conditions. Photostable and core-shell NaYF4:Yb3+, Er3+-SiO2 UCNPs obtained by a novel and simple co-precipitation method from lanthanide nitrates or oxides were herein synthesized for the first time. The sol–gel Stöber method followed by oven or supercritical gel drying was used to confer biocompatible surface properties to UCNPs by the formation of an ultrathin silica coating. Upconversion (UC) spectra were studied to evaluate the fluorescence of UCNPs upon red/near infrared (NIR) irradiation. ζ-potential measurements, TEM analyses, XRD patterns and long-term physicochemical stability were also assessed and confirmed that the UCNPs co-precipitation synthesis is a shape- and phase-controlling approach. The bio- and hemocompatibility of the UCNPs formulation with the highest fluorescence intensity was evaluated with murine fibroblasts and human blood, respectively, and provided excellent results that endorse the efficacy of the silica gel coating. The herein synthesized UCNPs can be regarded as efficient fluorescent probes for bioimaging purposes with the high luminescence, physicochemical stability and biocompatibility required for biomedical applications. Abstract

Aerogels have recently started to be considered as "advanced materials"; therefore, as a general consideration, aerogels' toxicity testing should focus on their functionality which resides in their nanoscale open internal porosity. To assess the hazards of organic aerogels, testing at three levels may characterize their biophysical, in vitro and in vivo toxicity, defining distinct categories of aerogels. At the first level of testing, their abiotic characteristics are investigated, and the best aerogel(s) is forwarded to be tested at level 2, wherein in vitro methodologies may mainly evaluate the aerogels' cellular behavior. Within level 2 of testing, the main characteristics of toxicity are investigated and the selected aerogels are introduced to in vivo animal models at level 3. In the animal model testing, target organs are investigated along with systemic parameters of toxicity. Some study cases are presented for organic or anorganic aerogels. Within this tiered workflow, aerogels-based materials can be tested in terms of human health hazard. [ABSTRACT FROM AUTHOR]

Abstract

The aim of this research was to evaluate the effect of the addition of ascorbic acid to reduce the concentration of nitrites and nitrates in mortadella. Three formulations of mortadella were tested to which three different concentrations of ascorbic acid (0.25 g / kg, F2: 0.50 g / kg and 0.75 g F3 / kg F1) were added. Through UV-visible spectrophotometry it was quantified the initial and final concentration of nitrates and nitrites and color formation, and the residual ascorbic acid was quantified by voltammetry. The analysis of variance indicated that there was significant difference (p˂0,05) among the three treatments. Nitrate reduction of 30.78% in F1, F2 50% at 76% and the values of F3 and formed nitrite 0.41 mg / kg in F1 0.24 mg / kg was achieved in F2 and 0.04 mg / kg in F3, in the formation of nitrite. F3 showed the highest percentage reduction of nitrates and the least amount of formation of nitrite. In conclusion, the addition of ascorbic acid to mortadella reduces the residual nitrate concentration after three days, where no longer significant reduction of this preservative was presented

Im Rahmen des von der Deutschen Forschungsgemeinschaft (DFG) geförderten Graduiertenkollegs GRK 2462 "Prozesse in natürlichen und technischen Partikel-Fluid-Systemen" (PintPFS) untersuchen Forscherinnen und Forscher aus den Bereichen Bauingenieurwesen, Maschinenbau, Verfahrenstechnik und Materialwissenschaften in einem interdisziplinären Ansatz natürliche und technische Partikel-Fluid-Systeme (PFS). Da PFS im Alltag allgegenwärtig und für viele Prozesse in Industrie, Pharmazie und Lebensmittelproduktion relevant sind, stellen sie ein hochinteressantes Forschungsfeld dar, das in vielerlei Hinsicht vielversprechende und nützliche Erkenntnisse verspricht., In the framework of Research Training Group GRK 2462 “Processes in Natural and Technical Particle-Fluid-Systems” (PintPFS), funded by the German Research Foundation (DFG), researchers from the fields of civil engineering, mechanical engineering, process engineering, and materials sciences investigate natural and technical Particle-Fluid-Systems (PFS) in an interdisciplinary approach. Abstract

The access of biodegradable scaffolds to the clinical arena is constrained by the absence of a suitable sterilization technique for the processing of advanced polymeric materials. Sterilization with supercritical CO2 (scCO2) may circumvent some technological limitations (e.g., low temperature, no chemical residues on the material), although scCO2 can plasticize the polymer depending on the processing conditions used. In this latter case, the integration of the manufacturing and sterilization processes is of particular interest to obtain sterile and customized scaffolds in a single step. In this work, scCO2 was exploited as a concomitantly foaming and sterilizing agent for the first time, developing a one-step process for the production of vancomycin-loaded poly(ε-caprolactone) (PCL) bone scaffolds. The effect of the CO2 contact time on the sterility levels of the procedure was investigated, and the sterilization efficiency was evaluated against dry spores (Bacillus stearothermophilus, Bacillus pumilus and Bacillus atrophaeus). Vancomycin-loaded PCL scaffolds had relevant sustained release profiles for the prophylaxis of infections at the grafted area, even those caused by methicillin-resistant Staphylococcus aureus (MRSA). The biological performance of the scaffolds was evaluated in vitro regarding human mesenchymal stem cells (hMSCs) attachment and growth.
