3. Comparison of bacterial diversity in Bactrocera cucurbitae (Coquillett) ovaries and eggs based on 16S rRNA sequencing

Abstract

Abstract Next-generation sequencing allows for fine-scale studies of microbial communities. Herein, 16S ribosomal RNA high-throughput sequencing was used to identify, classify, and predict the functions of the bacterial communities in the eggs and ovaries of Bactrocera cucurbitae (Coquillett) (Diptera: Tephritidae), which is a pest that infests a variety of cucurbit fruits at different developmental stages. Taxonomic analyses indicate that bacteria associated with B. cucurbitae represent 19 phyla, which were spread across different developmental stages. Specifically, the egg microbiota had a higher alpha diversity than those of microbiota in the primary and mature ovaries. Significant differences were not observed between the primary and mature ovaries in terms of their microbiota’s alpha diversities. Pseudomonadota, Deinococcota, Bacteroidota, Bacillota, and Actinomycetota were the dominant phyla in all three developmental stages of B. cucurbitae, and Pseudomonadaceae and Enterobacteriaceae were the most abundant families. Owing to the unique physiological environment of the ovaries, the diversity of their bacterial community was significantly lower than that in the eggs. This study provides new insights into the structure and abundance of the microbiota in B. cucurbitae at different developmental stages and contributes to forming management strategies for this pest.

Abstract

This paper presents the first report of the occurrence of the pinion-streaked snout Schrankia costaestrigalis (Stephens, 1834) in China and the first known record of this pest’s damage to potato (Solanum tuberosum L.) in the world. This discovery allowed us to alert entomologists and farmers, as well as agricultural agencies, and to provide a diagnostic tool to prevent potential establishment and spread of this newly introduced species in the potato-growing region of China and worldwide over the next few years.

Abstract

The genus Morphostenophanes is a small arboreal group of darkling beetles that are endemic to the Oriental region. The complete mitochondrial genome of the M. sinicus population from the Manwan Town was first characterized. The mitogenome consisted of a circular DNA molecule of 15,662 bp with a 66.352% AT content. It comprises 13 protein-coding genes (PCGs), 22 tRNA genes, and two rRNA genes. The PCGs have a typical ATN (Met) start codon, except for nad1 (TTG as a start codon), and are terminated by typical TAN stop codons.

Abstract

Coronavirus disease 2019 (COVID-19) has caused more than 40,000 deaths worldwide 1 . Approximately 14% of patients with COVID-19 experienced severe disease and 5% were critically ill 2 . Studies have shown that dysregulation of the COVID-19 patients’ immune system may lead to inflammatory storm and cause severe illness and even death 3,4 . Tocilizumab treatment targeting interleukin 6 receptor has shown inspiring clinical results of severe COVID-19 patients 5 . However, the immune network with Tocilizumab treatment at single cell resolution has not been uncovered. Here, we profiled the single-cell transcriptomes of 13,289 peripheral blood mononuclear cells isolated at three longitudinal stages from two severe COVID-19 patients treated with Tocilizumab. We identified a severe stage-specific monocyte subpopulation and these cells centric immune cell interaction network connected by the inflammatory cytokines and their receptors. The over-activated inflammatory immune response was attenuated after Tocilizumab treatment, yet immune cells including plasma B cells and CD8 + T cells still exhibited an intense humoral and cell-mediated anti-virus immune response in recovered COVID-19 patients. These results provided critical insights into the immunopathogenesis of severe COVID-19 and revealed fundamentals of effectiveness in Tocilizumab treatment.

Abstract

Common ragweed (Ambrosia artemisiifolia) is a notorious invasive weed that has spread across most temperate regions of the world. The beetle (Ophraella communa) is considered to be an effective control agent against A. artemisiifolia. As an oligophagous insect, its olfactory system is extremely important for host seeking in the wild. To the best of our knowledge, there is no report on the molecular mechanisms underlying olfaction recognition in this beetle. Hence, in this study, we characterized the odorant receptor co‐receptor of O. communa and named it as 'OcomORco'. Real‐time quantitative PCR (qRT‐PCR) showed that, compared to the control treatment, RNA interference (RNAi) strongly reduced the expression of OcomORco by 89% in male and 90% in female beetles. Electroantennogram assay showed that the antennal response of both male and female beetles to four volatiles of A. artemisiifolia was significantly reduced. The injected male or female beetles lost their preference for plant leaves as observed in the behavioural tests. In addition, disruption of the expression of OcomORco resulted in a reduction of oviposition, while there was no difference in larval hatching rate between control and knockdown females. We demonstrated that OcomORco plays a vital role in olfactory perception and host search in O. communa, and it is involved in oviposition in an indirect way. [ABSTRACT FROM AUTHOR]

Abstract

Increase in atmospheric CO2 directly affects the insect physiology and behavior, and indirectly affects the herbivorous insects by affecting their hosts. The increase in atmospheric CO2 is accompanied by an increase in temperature and heat waves. Ophraella communa LeSage is a natural enemy of Ambrosia artemisiifolia (common ragweed). The development and reproduction of this beetle is weakened upon eating common ragweed grown under stress conditions. As female behavior and physiology alter after mating, the reproductive tract of males is likely to modulate reproduction and development in this species. Herein, the transcriptional profiles of testes and accessory glands from male O. communa individuals feeding on common ragweed under conditions of high CO2 concentration and heat waves and that grown under ambient CO2 concentration were compared. Differentially expressed genes (DEGs) were identified between the same tissues from beetles fed on common ragweed grown under different stress conditions. There were 3, 2, 3, 1and 5 genes related to decomposition and transport of macromolecular substances, host location, stress response, reproduction, and poisonous food-utilization. No expected response was observed in the male reproductive tract, but some of the identified DEGs might control the development of the population. The results presented here should be helpful in guiding future studies on deciphering the indirect response of other organs to high CO2 concentration and heat waves, as well as the functions of seminal fluid proteins in O. communa. Abstract

In insects, the frequency distribution of the measurements of sclerotized body parts is generally used to classify larval instars and is characterized by a multimodal overlap between instar stages. Nonparametric methods with fixed bandwidths, such as histograms, have significant limitations when used to fit this type of distribution, making it difficult to identify divisions between instars. Fixed bandwidths have also been chosen somewhat subjectively in the past, which is another problem. In this study, we describe an adaptive kernel smoothing method to differentiate instars based on discontinuities in the growth rates of sclerotized insect body parts. From Brooks’ rule, we derived a new standard for assessing the quality of instar classification and a bandwidth selector that more accurately reflects the distributed character of specific variables. We used this method to classify the larvae of Austrosimulium tillyardianum (Diptera: Simuliidae) based on five different measurements. Based on head capsule width and head capsule length, the larvae were separated into nine instars. Based on head capsule postoccipital width and mandible length, the larvae were separated into 8 instars and 10 instars, respectively. No reasonable solution was found for antennal segment 3 length. Separation of the larvae into nine instars using head capsule width or head capsule length was most robust and agreed with Crosby’s growth rule. By strengthening the distributed character of the separation variable through the use of variable bandwidths, the adaptive kernel smoothing method could identify divisions between instars more effectively and accurately than previous methods.
