1. Quasi-interpolation for high-dimensional function approximation

Abstract

The paper proposes a general quasi-interpolation scheme for high-dimensional function approximation. To facilitate error analysis, we view our quasi-interpolation as a two-step procedure. In the first step, we approximate a target function by a purpose-built convolution operator (with an error term referred to as convolution error). In the second step, we discretize the underlying convolution operator using certain quadrature rules at the given sampling data sites (with an error term called discretization error). The final approximation error is obtained as an optimally balanced sum of these two errors, which in turn views our quasi-interpolation as a regularization technique that balances convolution error and discretization error. As a concrete example, we construct a sparse grid quasi-interpolation scheme for high-dimensional function approximation. Both theoretical analysis and numerical implementations provide evidence that our quasi-interpolation scheme is robust and capable of mitigating the curse of dimensionality for approximating high-dimensional functions.

2. Spherical quasi-interpolation using scaled zonal kernels

Abstract

We propose and study a new quasi-interpolation method on spheres featuring the following two-phase construction and analysis. In Phase I, we analyze and characterize a large family of zonal kernels (e.g., the spherical version of Poisson kernel, Gaussian, compactly-supported radial kernels), so that the underlying spherical convolution operators (upon the introduction of a scaling parameter) attains a high-order of approximation to target functions. In Phase II, we discretize the spherical integrals utilizing quadrature rules of optimal order to produce the final quasi-interpolants. Numerical experiments demonstrate that the new quasi-interpolation algorithm is robust and amenable to integrated as well as distributed ways of implementation. Moreover, the underlying error-analysis shows that by fine-tuning the scaling parameter in the radial kernels employed, the resulting quasi-interpolants achieve a well-balanced trade-off between approximation and sampling errors.

3. High-order quasi-interpolation with generalized Gaussian kernels restricted over tori

Abstract

The paper proposes a novel and efficient quasi-interpolation scheme with high approximation order for periodic function approximation over tori. The resulting quasi-interpolation takes the form of Schoenberg's tensor-product generalized Gaussian kernels restricted over circles. Notably, theoretical analysis shows that it achieves the highest approximation order equal to the order of the generalized Strang-Fix condition satisfied by the generalized Gaussian kernels. This is in sharp contrast to classical quasi-interpolation counterparts, which often provide much lower approximation orders than those dictated by the generalized Strang-Fix conditions satisfied by the kernels. Furthermore, we construct a sparse grid counterpart for high-dimensional periodic function approximation to alleviate the curse of dimensionality. Numerical simulations provided at the end of the paper demonstrate that our quasi-interpolation scheme is simple and computationally efficient.

Abstract

In vitro and in vivo comparative studies of the editing and synthetic functions of three forms of the carboxy-terminal sequence of methionyl-tRNA synthetase in Escherichia coli reveal that the C-terminal region promotes interactions between the methionine biosynthetic pathway and synthetase to transfer the excessive endogenous homocysteine to MetRS for editing. The MRS581 and wild-type MRS676 forms of C-terminal have higher editing capacity than the truncated MRS547 form. Synthetic function of in vitro MRS676 form is 25 times greater than shorter synthetase forms in each subunit.

Abstract

Soil zinc (Zn) plays a crucial role in plant growth, but excessive accumulation in the environment may lead to air, water and soil pollution. It is affected by various chemical, environmental and spatial factors. Therefore, it is important to identify the factors influencing Zn content in the landscape. The main motivation for this study is to determine the suitability of a generalized additive model (GAM) to describe change in soil Zn content due to influencing factors. A total of 1497 soil nutrient samples were collected in Fangshan District, Beijing, China. Organic matter (OM), available phosphorus (AP), available potassium (AK), alkali-hydrolyzed nitrogen (AHN) and slowly available potassium (SAK) are considered. The relationship between Zn, nutrients and geographic location (latitude & longitude) is investigated using the GAM. More precisely, the Akaike information criterion (AIC) is used to select influencing factors on Zn content and cross-validated to avoid overfitting of the multivariate model. The results show that Zn content reaches its maximum at latitude 39.8°N and longitude 115.9°E. Zinc content increases as AP increases to 150 mg/kg. When OM content is greater than 90 g/kg, Zinc content decreases with an increase in OM content. Factors that affected Zn content, in descending order of significance derived from deviance explained and adjustment coefficient of determination (Adj.R2) were AP, latitude, AHN, AK and OM. Moreover, the interactions between latitude and longitude, AHN and AP, OM and AK have significant impact on Zn.

Abstract

Based on multiquadric trigonometric spline quasi-interpolation, the paper proposes a scheme for numerical differentiation of noisy data, which is a well-known ill-posed problem in practical applications. In addition, in the perspective of kernel regression, the paper studies its large sample properties including optimal bandwidth selection, convergence rate, almost sure convergence, and uniformly asymptotic normality. Simulations are provided at the end of the paper to demonstrate features of the scheme. Both theoretical results and simulations show that the scheme is simple, easy to compute, and efficient for numerical differentiation of noisy data. [ABSTRACT FROM AUTHOR]

44. Tim3 Foxp3 Treg Cells Are Potent Inhibitors of Effector T Cells and Are Suppressed in Rheumatoid Arthritis.

Abstract

Rheumatoid arthritis (RA) is a chronic autoimmune disorder. Earlier studies have demonstrated that regulatory T (Treg) cells, the main cell type mediating immune tolerance, appeared to be enriched in the inflamed synovial tissues. It is still unclear why the Treg cells in RA patients are unable to limit exacerbated inflammation. Here, we found that the frequency of Tim3Foxp3 Treg cells, which were potent suppressors of proinflammatory responses, was downregulated in RA patients. Reduction in Tim3Foxp3 Treg frequency was correlated with increased RA disease activity. Furthermore, we observed that Tim3Foxp3 Tregs were expressed more interleukin (IL)-10 than Tim3Foxp3 Tregs. CD4CD25Tim3 T cells had higher capability of inhibiting interferon (IFN)-γ and tumor necrosis factor (TNF)-α secretion from T cells and peripheral blood mononuclear cells (PBMCs) than CD4CD25Tim3 T cells. Compared to that in healthy individuals, CD4CD25 T cells in RA patients were less potent in suppressing IFN-γ and TNF-α production from PBMCs. Blocking Tim3 on CD4CD25 T cells from healthy controls resulted in an elevation of IFN-γ and TNF-α production from PBMCs, suggesting that Tim3 expression on CD4CD25 T cells was required for optimal Treg function. However, this phenomenon was not observed in RA patients. In conclusion, our study suggested that the CD4CD25Foxp3 Treg cells from RA patients demonstrated a reduction of Tim3 and were less functional than Treg cells from healthy controls in a Tim3-related manner. 47. Inactivation of glycogen synthase kinase-3β up-regulates β-catenin and promotes chondrogenesis.

Abstract

This study aimed to investigate whether inhibition of glycogen synthase kinase-3β (GSK-3β) could promote chondrocytes proliferation. The expression pattern of GSK-3β was firstly determined by immunohistochemistry (IHC) in normal mouse. Tibias were then isolated and cultured for 6 days. The tibias were treated with dimethylsulfoxide (control) or GSK-3 inhibitor SB415286 (SB86). Length of tibias was measured until 6 days after treatment. These bones were either stained with alcian blue/alizarin red or analyzed by IHC. In addition, GSK-3β and β-catenin were analyzed by Western blot. Finally, cartilage-specific GSK-3β deletion mice (KO) were generated. Efficiency of GSK-3β deletion was determined through Western blot and IHC. After treated by inhibitor SB86, the overall length of growth plate was not changed. However, growth of tibia in SB86 group was increased by 31 %, the length of resting and proliferating was increased 13 % (P < 0.01), whereas the length of hypertrophic was decreased by 57 % (P < 0.01). Besides, the mineralized length was found to be significant longer than the control group (P < 0.05). In KO mice, growth plate and calvaria tissue both exhibit significant reduction of GSK-3β (P < 0.05) whereas the lengths of tibias in KO were almost same compared with control mice. Finally, an increase amount of β-catenin protein was observed in SB86 (P < 0.05). In addition, significantly increased β-catenin was also found in the growth plate of KO mice (P < 0.05). Inhibition of GSK-3 could promote longitudinal growth of bone through increasing bone formation. Besides, the inactivation of GSK-3β could lead to enhancing β-catenin, therefore promote chondrocytes proliferation. [ABSTRACT FROM AUTHOR]
