Abstract Endophytes have been shown to promote plant growth and health. In the present study, a Bacillus velezensis CH1 (CH1) strain was isolated and identified from high-quality oats, which was capable of producing indole-3-acetic acid (IAA) and strong biofilms, and capabilities in the nitrogen-fixing and iron carriers. CH1 has a 3920 kb chromosome with 47.3% GC content and 3776 code genes. Compared genome analysis showed that the largest proportion of the COG database was metabolism-related (44.79%), and 1135 out of 1508 genes were associated with the function “biosynthesis, transport, and catabolism of secondary metabolites.” Furthermore, thirteen gene clusters had been identified in CH1, which were responsible for the synthesis of fifteen secondary metabolites that exhibit antifungal and antibacterial properties. Additionally, the strain harbors genes involved in plant growth promotion, such as seven putative genes for IAA production, spermidine and polyamine synthase genes, along with multiple membrane-associated genes. The enrichment of these functions was strong evidence of the antimicrobial properties of strain CH1, which has the potential to be a biofertilizer for promoting oat growth and disease resistance.

ABSTRACT SAMHD1 is an intrinsic limiting factor that effectively prevents HIV-1 infection in macrophages, dendritic cells, and resting CD4+ T cells. Extensive studies have underscored the indispensable role of the dNTPase activity of SAMHD1 in its antiviral function by primarily depleting dNTPs in quiescent cells, thereby impeding HIV-1 cDNA synthesis. However, recent advancements in understanding posttranslational modifications of SAMHD1 have revealed specific modification site mutants that maintain their ability to reduce dNTP levels while impairing the inhibition of HIV-1 replication. Thus, the precise anti-HIV-1 mechanism of SAMHD1 remains enigmatic, necessitating a comprehensive understanding of the underlying mechanisms to develop novel therapeutic strategies targeting its antiviral activity. Recent findings by Guo et al. shed light on the role of SAMHD1 as an HIV-1 core sensor in suppressing HIV-1 infection after viral cDNA synthesis through its interaction with MX2 (H. Guo, W. Yang, H. Li, J. Yang, et al., mBio 15:e01363-24, 2024, https://doi.org/10.1128/mbio.01363-24).

ABSTRACT Numerous coreceptors have been shown to facilitate hACE2-dependent or hACE2-independent infection by SARS-CoV-2. A recent study published in mBio by Yu et al. showed that the histamine receptor H1 (HRH1) functions as an alternative receptor for SARS-CoV-2 via direct binding to viral spike proteins (F. Yu, X. Liu, H. Ou, X. Li, et al., mBio e01088-24, 2024, https://doi.org/10.1128/mbio.01088-24). Furthermore, they present compelling evidence that antihistamine drugs targeting HRH1 potently inhibit SARS-CoV-2 entry. This study highlights the therapeutic potential of repurposable antihistamines against COVID-19.

ABSTRACT A recent study published in mBio by Cao et al. demonstrated that the helminth Trichinella sprialis (Ts) alleviates COVID-19-related cytokine storms in an IL-9-dependent way (Z. Cao, J. Wang, X. Liu, Y. Liu, et al., mBio 15:e00905-24, 2024, https://doi.org/10.1128/mbio.00905-24). A cytokine storm is a severe immune response characterized by the overproduction of proinflammatory cytokines, such as TNF-α and IFN-γ, leading to tissue damage and mortality in COVID-19 patients. This study indicated that IL-9 is crucial in protecting against cytokine storm syndromes associated with SARS-CoV-2 infection and proposed that anti-inflammatory molecules from Ts excretory/secretory (TsES) products could be a novel source for treating such illnesses.

Investigating oat tissue microflora during its different developmental stages is necessary for understanding its growth and anti-disease mechanism. In this study, 16S rDNA and ITS (Internally Transcribed Spacer) high-throughput sequencing technology were used to explore the microflora diversity of oat tissue. Twenty-seven samples of leaves, stems, and roots from three developmental stages, namely the seedling stage (SS), jointing stage (JS), and maturity stage (MS), underwent sequencing analysis. The analysis showed that 6480 operational taxonomic units (OTUs) were identified in the examined samples, of which 1698 were fungal and 4782 were bacterial. Furthermore, 126 OTUs were shared by fungi, mainly Ascomycota, Basidiomycota, and Mucoromycota at the phylum level, and 39 OTUs were shared by bacteria, mainly Actinobacteriota and Proteobacteria at the phylum level. The microbial diversity of oat tissue in the three developmental stages showed differences, and the α-diversity of the bacteria and β-diversity of the bacteria and fungi in the roots were higher than those of the stems and leaves. Among the bacteria species, Thiiopseudomonas, Rikenellaceae RC9 gut group, and Brevibacterium were predominant in the leaves, MND1 was predominant in the roots, and Lactobacillus was predominant in the stems. Moreover, Brevibacterium maintained a stable state at all growth stages. In the fungal species, Phomatospora was dominant in the leaves, Kondoa was dominant in the roots, and Pyrenophora was dominant in the stems. All species with a high abundance were related to the growth process of oats and antagonistic bacteria. Furthermore, connection modules were denser in bacterial than in fungal populations. The samples were treated with superoxide dismutase and peroxidase. There were 42 strains associated with SOD (Superoxide dismutase), 60 strains associated with POD (Peroxidase), and 38 strains in total, which much higher than fungi. The network analysis showed that bacteria might have more dense connection modules than fungi, The number of bacterial connections to enzymes were much higher than that of fungi. Furthermore, these results provide a basis for further mechanistic research.

This study investigated the effect of replacing part of the dietary soybean meal with either polymer-coated urea or gelatinized starch urea on the production performance, blood indexes, and ruminal fermentation of Angus heifers. A total of 210 purebred Angus cattle (BW = 314.26 kg) were divided into three groups: the no urea group (CON), the polymer-coated urea group (PCU), and the gelatinized starch urea group (GSU); 20 g/kg polymer-coated urea or 25 g/kg gelatinized starch urea was used to replace part of soybean meal in the concentrate feed, according to the principle of isometabolic energy and isonitrogenous. The result showed that the PCU group had higher ADG and ADF apparent digestibility, while it had a lower feed–weight ratio. On the 86th day, the serum albumin (ALB) content in the PCU group was significantly higher than that in the CON group. In rumen, compared with the CON group, the contents of acetic acid and total volatile fatty acid were significantly higher in the PCU group, whereas butyric acid and propionic acid were significantly higher in the PCU group and GSU group. Ruminal bacterial diversity analysis found that the abundance of Firmicutes was higher in the PCU group at the phylum level, and an inverse result was observed in Bacteroidetes. The abundance of Paraprevotella was higher in the PCU group, whereas higher abundance of Prevotella was found in the GSU group at the genus level. These results indicate that slow-release urea can replace part of soybean meal in the diet, and the amount of substitution in this trial had no diverse effect on the performance of Angus heifers.

Objective Olecranon osteotomy and paratricipital approaches were widely used in the treatment of type C distal humerus fracture but some disadvantages exist, so a combined medial and lateral approach was designed. The objective of this study was to investigate and compare the clinical outcomes of combined medial and lateral approach with the paratricipital approach in open reduction and internal fixation of type C distal humerus fractures. Methods From May 2018 to April 2020, 37 patients with type C distal humerus fracture who accepted open reduction and internal fixation in our hospital were enrolled in this study. All cases were randomly divided into two groups according to the surgical approach: combined medial and lateral approach group (19 cases), paratricipital approach group (18 cases). All of the patients received open reduction and double vertical plates fixation. The operation and follow‐up indexes, including operation time, blood loss, incision length, triceps muscle strength, flexion‐extension arc of elbow and forearm rotation arc, were recorded and compared. Caja score was used to assess the quality of fractures reduction. Mayo Elbow Performance Score (MEPS) was used to evaluate the elbow function in the follow‐up. Complications such as incision infection, ulnar nerve injury, degenerative osteoarthritis, and heterotopic ossification were analyzed. Results The differences in age, gender, and AO classification of fractures between two groups were not statistically significant (p > 0.05). The sum of medial and lateral incision length of combined approach group was longer than the midline incision of paratricipital approach group (15.4 ± 0.8 vs. 14.6 ± 0.8, p 0.05), blood loss (71.3 ± 24.5 vs. 72.8 ± 24.6, p > 0.05), and Caja score (16.05 ± 5.67 vs. 15.56 ± 5.66, p > 0.05). During the follow‐up, the MEPS of combined approach group was higher than that of paratricipital approach group at 3 months postoperatively (80.5 ± 5.7 vs. 68.9 ± 8.1, p 0.05) and at the last follow‐up (86.8 ± 7.1 vs. 86.9 ± 7.7, p > 0.05) between the two groups. There was no significant difference in triceps muscle strength (p > 0.05), flexion‐extension arc (126.8 ± 5.3 vs. 128.9 ± 6.0, p > 0.05), and forearm rotation arc (163.2 ± 5.3 vs. 163.6 ± 4.8, p > 0.05) at the last follow‐up. Although the incidence of complication of combined approach group (15.8%) was lower than that of paratricipital approach group (22.2%), the difference was not statistically significant (p > 0.05). Conclusions The combined medial and lateral approach was an effective and safe way of open reduction and internal fixation for type C distal humerus fractures. Compared with the paratricipital approach, the combined medial and lateral approach could restore the elbow function more quickly postoperatively, and the long‐term results were comparable.

Safety concerns are progressively emerging regarding the adoption of Unmanned Aircraft Systems (UASs) in diverse civil applications, particularly within the booming air transportation system, such as in Advanced Air Mobility. The outcomes of risk assessment determine operation authorization and mitigation strategies. However, civil UAS operations bring novel safety issues distinct from traditional aviation, like ground impact risk, etc. Existing studies vary in their risk definitions, modelling mechanisms, and objectives. There remains an incomplete gap of challenges, opportunities, and future efforts needed to collaboratively address diverse safety risks. This paper undertakes a comprehensive review of the literature in the domain, providing a summative understanding of the risk assessment of civil UAS operations. Specifically, four basic modelling approaches utilized commonly are identified comprising the safety risk management process, causal model, collision risk model, and ground risk model. Then, this paper reviews the state of the art in each category and explores the practical applications they contribute to, the support offered to participants from multiple stakeholders, and the primary technical challenges encountered. Moreover, potential directions for future work are outlined based on the high-level common problems. We believe that this review from a holistic perspective contributes towards better implementation of risk assessment in civil UAS operations, thus facilitating safe integration into the airspace system.

A bi-level optimization model for the logistics UAV air route network capacity evaluation based on traffic flow allocation is designed in order to meet the future trend of large-scale and normalized operation of logistics UAVs. The maximum sorties of logistics UAVs that can be served by the air route network are the upper-bound model objective, namely, the maximum flow of the logistics UAV air route network. The impedance function is constructed by considering safety and efficiency factors, and the lower-bound model objective function with the minimum logistics UAV air route network impedance value. An improved particle swarm optimization(PSO) algorithm is combined with the method of the successive algorithm(MSA) for solving the bi-level optimization model. To verify the effectiveness of the proposed model and algorithm, a simplified logistics UAV air route network is built. The results show that the proposed algorithm obtains reliable results after 26 iterations, and most segments capacity utilization rate is more than 70%. Parametric analysis of safe separation and algorithm population size shows that the capacity of logistics UAV air route network decreases with the increase of safe separation and the decreasing trend is gradually slowed down, and the optimal algorithm population size corresponding to different safe separations also varies. Based on the study described above, a logistics UAV air route network based on actual geographic information data is constructed, and the experimental results demonstrate that the suggested technique could be used to a specific scale of logistics UAV route network capacity evaluation and had validity.

BackgroundEBV-associated smooth muscle tumours (EBV-SMTs) are uncommon neoplasms associated with immunodeficiency. The pathogenesis of EBV-SMTs is poorly understood. IL-2-inducible T-cell kinase (ITK), a member of the Tec family of tyrosine kinases, is the predominant Tec kinase in T cells. Researchers have shown that ITK is involved in the pathogenesis of autoimmune diseases and carcinogenesis, and the loss of ITK function due to mutation in patients can lead to EBV-associated lymphoproliferation. Multiple Epstein-Barr virus–associated smooth muscle tumours with ITK mutation have rarely been reported.Case presentationA 6-year-old female child was admitted to the hospital due to recurrent bilateral hip pain for more than one year. Tumours were found in the lung, near the intracranial cavernous sinus and in the lumbar spine and paraspinal soft tissues by CT and MRI. The patient underwent vertebral tumour biopsy, which suggested low-grade myogenic or inflammatory myofibroblastic tumours, so the patient was given three courses of chemotherapy without symptom relief or mass reduction. The patient underwent lumbar mass resection, the pathological results indicated EBV-SMT, and a novel germline homozygous deletion mutation in the ITK gene was detected. The deletion mutation in this gene has not yet been reported and the clinical significance of the pathogenicity of the variant is unknown. Intrabronchial mass resection was performed under fibre bronchoscopy, and the pathological results indicated EBV-SMT. No significant recurrence or progression was observed after more than 2 years of follow-up.ConclusionsWe present a rare case of multiple EBV-SMTs combined with ITK gene mutation. Some of the tumours were removed, and some were treated conservatively. There was no significant recurrence or progression after more than two years of follow-up. The optimal treatment regimen still needs to be further explored, and the relationship between ITK gene mutation at this locus and immunodeficiency and EBV-SMT warrants further investigation.

Synergetic trajectory planning of flights is one of the important goals of trajectory-based operation (TBO), and it is also a method to further improve the utilization of airspace resources with the increasing number of flights in recent years. In order to plan the four-dimensional trajectory (4DT) pre-tactically and comprehensively, match the flight traffic with airspace capacity, reduce congestion, potential conflicts, and fuel consumption thus improving the efficiency of flights, this paper conducts a method for synergetic trajectory planning in the en-route phase from the perspective of airlines. Firstly, the aircraft performance model, aircraft fuel consumption model, and atmospheric model are constructed according to the base of aircraft data (BADA3.11), and an airspace congestion prediction model is constructed based on the historical flow data of airspace. Secondly, a multi-objective synergetic trajectory planning model is established, and a solution method based on the non-dominated sorting genetic algorithm and simulated annealing algorithm (NSGA3-SA) for the problem of synergetic trajectory planning is designed. The simulation shows that the optimization model and the solution algorithm of NSGA3-SA can reduce fuel consumption by about 4.5% compared to the original flight plans and has a good effect on reducing congestion and avoiding conflicts. The running time of the NSGA3-SA can meet the operational requirements of the pre-tactical trajectory planning. The multi-objective optimization model and the solution algorithm proposed in this paper have great value for the research of flight plan optimization.

To improve the trajectory prediction accuracy of unmanned aerial vehicles (UAVs) with random behavior intentions, this paper presents a short-term four-dimensional (4D) trajectory prediction method based on spatio-temporal trajectory clustering. A spatio-temporal trajectory clustering algorithm is first designed to cluster the UAV trajectory segments divided by a fixed time window. Each trajectory segment is given a category label that represents some certain type of behavior characteristics, such as climbing, turning, descending, etc. The convolutional neural network (CNN) is used to identify the category label of a given trajectory segment by learning the behavior characteristics of different trajectory segments. Based on the long-short-term memory network (LSTM), a short-term trajectory prediction model for different categories of label trajectory segments is established. The global trajectory prediction includes several steps adopting the corresponding prediction models. Historical trajectory data of UAVs are used to validate the proposed prediction method. Experiment results indicate that the method can obtain obviously better prediction accuracy in a short prediction time range (0-3s) with acceptable efficiency compared to LSTM, GRU and velocity trend extrapolation.

To mitigate the problem of multiple unmanned aircraft systems (MUAS) conflicts at low altitude and ensure the operational safety, this paper proposes a Multivariate Combined Conflict Detection (MCCD) method for MUAS by combining the characteristics of nominal and probabilistic trajectory method. Firstly, the structural framework of the MCCD method is established based on the concept of potential conflict pool, and a detection pattern is derived for MUAS. Secondly, a three-dimensional conflict fast detection model is constructed by velocity obstacle methods, which can rapidly detect potential conflict risks. Thirdly, a trajectory prediction model is constructed by using bidirectional long-short term memory (Bi-LSTM) network, and then a probability-based conflict detection model can be obtained by the expected value and error distribution of trajectory prediction, which can accurately calculate the collision probability of UAS pair. By fully integrating the above models, the fast and accurate detection of MUAS conflicts is achieved. Finally, multiple conflicting trajectories are constructed to analyze the effectiveness of MCCD method, the tests indicate that the average detection time of the proposed method is less than 15ms, the false positive rate is less than 0.01 and the false negative rate is less than 0.0035. The results show that the MCCD has the accuracy advantage and better real-time performance for MUAS conflict detection compared to the method of velocity trend extrapolation, single probabilistic conflict detection and probabilistic neural network.

The meniscus is a kind of fibrous cartilage structure that serves as a cushion in the knee joint to alleviate the mechanical load. It is commonly injured, but it cannot heal spontaneously. Traditional meniscectomy is not currently recommended as this treatment tends to cause osteoarthritis. Due to their good biocompatibility and versatile regulation, hydrogels are emerging biomaterials in tissue engineering. Hydrogels are excellent candidates in meniscus rehabilitation and regeneration because they are fine-tunable, easily modified, and capable of delivering exogenous drugs, cells, proteins, and cytokines. Various hydrogels have been reported to work well in meniscus-damaged animals, but few hydrogels are effective in the clinic, indicating that hydrogels possess many overlooked problems. In this review, we summarize the applications and problems of hydrogels in extrinsic substance delivery, meniscus rehabilitation, and meniscus regeneration. This study will provide theoretical guidance for new therapeutic strategies for meniscus repair.

The Federal Aviation Administration introduced the concept of urban air mobility (UAM), a new three-dimensional transport system that operates with a fusion of manned/unmanned aerial vehicles on an urban or intercity scale. The rapid development of UAM has brought innovation and dynamism to many industries, especially in the field of logistics. Various types of unmanned aerial vehicles (UAVs) for use in transport logistics are being designed and produced. UAV logistics refers to the use of UAVs, usually carrying goods and parcels, to achieve route planning, identify risk perception, facilitate parcel delivery, and carry out other functions. This research provides a method for assessing the operational capacity of a UAV logistics route network. The concept of “logistics UAV route network operation capacity” is defined, and a bi-objective optimization model for assessing the route network’s operating capacity is developed. The first objective is to maximize the number of UAV logistics delivery plans that can be executed in a fixed operation time. The second objective is to minimize the total operational impedance value in a fixed operation time. To solve the bi-objective optimization model, the Non-dominated Sorting Genetic Algorithm-II (NSGA-II) is utilized. A UAV logistics route network with 62 nodes is developed to assess the rationale and validity of the proposed concept. The experiments show that with an increase in operation time, the route network’s optimal operational capacity gradually increases, the convergence speed of the algorithm slows down, and the optimization magnitude gradually reduces. Two key parameters—operational safety interval and flight speed—are further analyzed in the experiments. According to the experiments, as the safety interval increases, the route network’s average operational capacity steadily diminishes, as does its sensitivity to the safety interval. The average operational capacity steadily rose with the rise in flight speed, especially when the UAV logistics flight speed was between 10 m/s and 10.5 m/s. In that range, the operational capacity of the route network was substantially impacted by the flight speed.

Objective To investigate the factors, surgical treatment methods and clinical effect of internal fixation failure of intertrochanteric and subtrochanteric fractures. Methods From June 2015 to May 2019, arthroplasty and internal fixation revision were used to treat 18 cases of internal fixation failure of intertrochanteric and subtrochanteric fractures. There were 10 males and eight females, with an average age of 67.3 years (38–92 years). The 16 cases of initial intertrochanteric fractures were classified according to AO/OTA:13 cases of A2 and 3 cases of A3, the other 2 cases were subtrochanteric fractures (Seinsheimer type IV). The internal fixation failure was treated with total hip arthroplasty (6 cases), bipolar hemiarthroplasty (4 cases), revision with proximal femoral lockingplate (4 cases) and extend intramedullary nail (4 cases). Results All patients were followed up for an average of 24.7 months (range, 12 to 36 months). The average operative time was 111.4 min (range, 72 to 146 min) and the average intraoperative blood loss was 403.6 mL (range, 200 to 650 mL). The increasing number of unmanned aerial vehicles (UAVs) in low-altitude airspace is seriously threatening the safety of the urban environment. This paper proposes an adaptive collision avoidance method for multiple UAVs (mUAVs), aiming to provide a safe guidance for UAVs at risk of collision. The proposed method is formulated as a two−layer resolution framework with the considerations of speed adjustment and rerouting strategies. The first layer is established as a deep reinforcement learning (DRL) model with a continuous state space and action space that adaptively selects the most suitable resolution strategy for UAV pairs. The second layer is developed as a collaborative mUAV collision avoidance model, which combines a three-dimensional conflict detection and conflict resolution pool to perform resolution. To train the DRL model, in this paper, a deep deterministic policy gradient (DDPG) algorithm is introduced and improved upon. The results demonstrate that the average time required to calculate a strategy is 0.096 s, the success rate reaches 95.03%, and the extra flight distance is 26.8 m, which meets the real-time requirements and provides a reliable reference for human intervention. The proposed method can adapt to various scenarios, e.g., different numbers and positions of UAVs, with interference from random factors. The improved DDPG algorithm can also significantly improve convergence speed and save training time.

With the dramatic development of small unmanned aircraft systems (sUAS), how to ensure sUAS safety operation has been a growing concern. This article proposes a fast probabilistic collision detection method for sUAS based on probability density function approximations. Firstly, cylindrical collision zones for sUAS and obstacles are established by geometrical methods for simplifying collision modeling, and instantaneous collision probability for sUAS is expressed by a triple integral. Secondly, a rapid estimation algorithm is derived for instantaneous collision probability, and then the predicted collision probability in probabilistic collision detection can be obtained by the maximum of instantaneous collision probabilities during the encounter. Randomized tests indicate that the average computation time of the proposed algorithm is less than 0.001s, and the Mean Absolute Error (MAE) is less than 0.01 and the Root Mean Squared Error (RMSE) is less than 0.02. Finally, numerical simulations are carried out to analyze the influence of parameters, including crossing angle, predicted separation at the closest point of approach (CPA), and predicted time to CPA, on collision probabilities. The optimal detection time for collision detection is also discussed in the different types of encounters. The collision detection method proposed in this article can provide support for real-time collision avoidance and the definition of dynamic safety bounds for sUAS.

Meniscal injuries have poor intrinsic healing capability and are associated with the development of osteoarthritis. Decellularized meniscus extracellular matrix (mECM) has been suggested to be efficacious for the repair of meniscus defect. However, main efforts to date have been focused on the concentration, crosslinking density and anatomical region dependence of the mECM hydrogels on regulation of proliferation and differentiation of adult mesenchymal stem cells (MSCs) in vitro 2D or 3D culture. A systematic investigation and understanding of the effect of mECM on encapsulated MSCs response and integrative meniscus repair by in vivo rat subcutaneous implantation and orthotopic meniscus injury model will be highly valuable to explore its potential for clinical translation. In this study, we investigated the in situ delivery of rat BMSCs in an injectable mECM hydrogel to a meniscal defect in a SD rat model. Decellularized mECM retained essential proteoglycans and collagens, and significantly upregulated expression of fibrochondrogenic markers by BMSCs versus collagen hydrogel alone in vitro 3D cell culture. When applied to an orthotopic model of meniscal injury in SD rat, mECM is superior than collagen I scaffold in reduction of osteophyte formation and prevention of joint space narrowing and osteoarthritis development as evidenced by histology and micro-CT analysis. Taken together, these results indicate mECM hydrogel is a highly promising carrier to deliver MSCs for long-term repair of meniscus tissue, while preventing the development of osteoarthritis.

Objective To investigate the surgical approach, fixation, and clinical effect of comminuted coronal shear fracture of distal humerus. Methods From March 2017 to February 2019, we had used open reduction and internal fixation to treat 19 cases of comminuted distal humeral coronal shear fracture. There were 8 males and 11 females, with an average age of 44.6 years (19–72 years). There were 10 cases on left side and 9 cases on right side. All cases were closed fractures. According to Dubberley's classification, there were six cases of type 1, six cases of type 2, and seven cases of type 3. The lateral Kocher approach, extended Kocher approach, extended Kocher approach combined with a medial incision and the olecranon osteotomy approach were used for exposure. Headless screw, Kirschner wire, and suture were used to fix the fractures. Two cases were fixed with hinged elbow fixators additionally. The follow‐up was evaluated by Mayo Elbow Performance Score (MEPS). Results All patients were followed up for an average of 17.1 months (range, 12 to 30 months). The average time of fracture union was 8.8 ± 1.9 weeks. There were three cases of degenerative osteoarthritis of elbow and one case of heterotopic ossification after operation. A total of 10 patients underwent removal of implants. At the last follow‐up, the elbow flexion‐extension arc was 130.5° ± 10.5°. The forearm rotation arc was 167.4° ± 6.1°. The MEPS was 85.8 ± 8.5, the results were classified as excellent in nine cases, good in eight, and fair in two. The excellent and good rate was 89.5%. The time of fracture union of type 1 was shorter than type 3 (P = 0.024), the elbow flexion‐ extension arc of type 1 fracture was better than type 2 (P = 0.043) and type 3 (P = 0.012), the forearm rotation arc of type 1 fracture was better than type 3 (P = 0.006), the MEPS of type 1 fracture was better than type 2 (P = 0.009) and type 3 (P = 0.002). Conclusion Open reduction and internal fixation with headless screw, Kirschner wire, and suture can be used for the treatment of comminuted distal humeral coronal shear fractures. The elbow joint function can be restored satisfactorily.

Aiming at the problem of the scheduling scheme of urban low-altitude logistics unmanned aerial vehicles (UAVs), this paper establishes a demand-based UAV scheduling scheme model using an improved simulated annealing algorithm, taking minimizing the cost of distribution as the objective function and considering restrictions such as UAV performance constraints, airspace constraints, and distribution constraints, among others. For verification, actual express data and airspace constraints in Shanghai are taken as examples. Two urban air traffic networks are constructed using road and building data. The analysis results show that the planning scheme of this model is superior to other forecasting models in terms of delivery cost and delivery time. In addition, this model can flexibly calculate the optimal scheduling scheme under the constraints of multiple parameters, according to the requirements of delivery volume, delivery distance, UAV performance, etc.

Cellular data provide a promising way for congestion detection with low cost and high coverage, and the simulation study is a feasible solution to verify the detection method. This paper presents a simulation approach that uses cellular data to detect traffic congestion on urban arterials based on the relationship between cellular data and traffic status. The virtual testbed, which includes three main modules, is developed to perform the cellular activities generation, collection, and aggregation process between cell phones and cell stations. Then, the correlation between cellular data and traffic status data is studied. Finally, three scenarios using the data from testbed are demonstrated to measure the performance of the proposed method under different conditions. The results indicate that the proposed approach is a feasible and efficient way to simulate cellular data generation, collection, and aggregation process. Also, it can be the base for further analysis to detect traffic congestion on arterials using cellular data.

Objectivechordomas are rare bone tumors with few therapeutic options. Skull base and sacrum are the two most common origin sites. Immunotherapies are emerging as the most promising approaches to fight various cancers. This study tends to identify new cell surface targets for immunotherapeutic options of skull base chordomas.Methodswe profiled 45 skull base chordoma clinical samples by immunohistochemistry for the expression of six CAR-Targets (PD-L1, B7-H3, B7-H4, VISTA, HER2 and HER3). In addition, we generated B7-H3 targeted CAR-T-cells and evaluated their antitumor activities in vitro.ResultsWe found that B7-H3 was positively stained in 7 out of 45 (16%) chordoma samples and established an expression hierarchy for these antigens (B7-H3 > HER3 > PD-L1 > HER2 = VISTA = B7-H4). We then generated a B7-H3 targeted CAR vector and demonstrated that B7-H3-CAR-T-cells recognized antigen positive cells and exhibited significant antitumor effects, including suppression of tumor spheroid formation, CAR-T-cell activation and cytokine secretion.ConclusionsOur results support B7-H3 might serve as a promising target for CAR-T-cell therapies against chordomas.

The popularity of unmanned aerial vehicle (UAV) technology has made UAV logistics transportation possible. However, based on the current development status of logistics UAVs, there are difficulties in using UAVs directly for door-to-door logistics transportation. Therefore, it is necessary to establish UAV parcel-receiving stations that can gather logistics needs in a small area. The construction of stations allows the UAVs to transport back and forth between the distribution warehouse and the established stations, enabling customers to send and receive packages at the more convenient stations. Based on the current situation, it is a more appropriate air–ground cooperative transport mode to solve the “last-mile” cargo transportation problem. In this paper, a bi-layer UAV parcel-receiving station and public air-route planning method is proposed to explore the interaction between station location and public route planning, and is solved with a genetic algorithm and max–min ant system (GA-MMAS). The model proposed in this paper can determine the location of the stations and plan the public air routes between the warehouse and stations simultaneously. Simulation results show that the planning results of the bi-layer optimization model proposed in this paper meet the requirements of station location and public air-route planning. Compared with the layered planning results, the cost of the upper-layer model is reduced by 5.12% on average, and the cost of the lower-layer model is reduced by 4.48%.

Background: To improve the grade of beef marbling has great economic value in the cattle industry since marbling has the traits of high quality and comprehensive nutrition. And because of the marbling’s importance and complexity, it is indispensable to explore marbled beef at multiple levels. This experiment studied the relationship between fecal metabolites and marbling characters, and further screened biomarkers. Results: We performed fecal metabolomics analysis on 30 individuals selected from 100 crossbreed cattle (Luxi Yellow cattle ♀ × Japanese Wagyu cattle ♂), 15 with an extremely high-grade marbling beef and 15 with an extremely low-grade marbling beef. A total of 9959 and 8389 m/z features were detected in positive ionization and negative ionization mode by liquid chromatography-mass spectrometry (LC-MS). Unfortunately, the sample separation in the PCA is not obvious, and the predictive ability of the orthogonal partial least squares discrimination analysis (OPLS-DA) model is not good. However, we got six differential metabolites filtered by VIP > 1 and p < 0.05. After that, we used weighted correlation network analysis (WGCNA) and found out a module in each positive and negative mode most related to the trait of marbling beef, and then identified three metabolites in positive mode. By further annotation of the Kyoto encyclopedia of genes and genomes (KEGG), it was found that these metabolites involved a variety of metabolic ways, including sphingomyelin metabolism, linoleic acid metabolism, glycerophospholipid metabolism, and so on. Finally, receiver operating characteristic (ROC) analysis was used to evaluate the predictability of metabolites, and the result showed that SM(d18:0/16:1(9Z)) (AUC = 0.72), PC(15:0/18:2(9Z,12Z)) (AUC = 0.72), ADP (AUC = 0.71), PC(16:0/16:0) (AUC = 0.73), and 3-O-Sulfogalactosylceramide (d18:1/18:0) (AUC = 0.69) have an accuracy diagnosis. Conclusions: In conclusion, this study supports new opinions for the successive evaluation of marbling beef through metabolites. Furthermore, six non-invasive fecal metabolites that can evaluate beef marbling grade were found, including SM(d18:0/16:1(9Z)), PC(15:0/18:2(9Z,12Z)), ADP, PC(16:0/16:0), and 3-O-Sulfogalactosylceramide.

With the development of civil aviation, the number of flights keeps increasing and the flight delay has become a serious issue and even tends to normality. This paper aims to prove that Stacking algorithm has advantages in airport flight delay prediction, especially for the algorithm selection problem of machine learning technology. In this research, the principle of the Stacking classification algorithm is introduced, the SMOTE algorithm is selected to process imbalanced datasets, and the Boruta algorithm is utilized for feature selection. There are five supervised machine learning algorithms in the first-level learner of Stacking including KNN, Random Forest, Logistic Regression, Decision Tree, and Gaussian Naive Bayes. The second-level learner is Logistic Regression. To verify the effectiveness of the proposed method, comparative experiments are carried out based on Boston Logan International Airport flight datasets from January to December 2019. Multiple indexes are used to comprehensively evaluate the prediction results, such as Accuracy, Precision, Recall, F1 Score, ROC curve, and AUC Score. The results show that the Stacking algorithm not only could improve the prediction accuracy but also maintains great stability.

Although numerous studies have indicated that chronic stress causes cognitive dysfunction with the impairment of synaptic structures and functions, the relationship between cognitive deficits induced by repeated restraint stress and the level of NMDA receptors in the subregion of the hippocampus has been relatively unknown until now. In this study, 3-week-old male Sprague-Dawley rats were exposed to repeated restraint stress for seven consecutive days, their cognitive functions were evaluated through behavioral tests, and then they were sacrificed for electrophysiological, morphological, and biochemical assays. Chronic repeated restraint stress led to cognitive and electrophysiological impairments, with a reduced density of dendritic spines. We also found that the protein level of NMDA receptors only increased in the hippocampal CA3 region. Nevertheless, repeated restraint stress-induced cognitive and synaptic dysfunction were effectively reversed by Ro25-6981, an inhibitor of the GluN2B receptor. These findings suggest that repeated restraint stress-induced synaptic and cognitive deficits are probably mediated through NMDA receptors.

Background/Aims: Current drug therapies for osteoarthritis (OA) are not practical because of the cytotoxicity and severe side-effects associated with most of them. Artemisinin (ART), an antimalarial agent, is well known for its safety and selectivity to kill injured cells. Based on its anti-inflammatory activity and role in the inhibition of OA-associated Wnt/β-catenin signaling pathway, which is crucial in the pathogenesis of OA, we hypothesized that ART might have an effect on OA. Methods: The chondro-protective and antiarthritic effects of ART on interleukin-1-beta (IL-1β)-induced and OA patient-derived chondrocytes were investigated in vitro using cell viability assay, glycosaminoglycan secretion, immunofluorescence, quantitative reverse transcription-polymerase chain reaction, and western blotting. We also used OA model rats constructed by anterior cruciate ligament transection and medial meniscus resection (ACLT+MMx) in the joints to investigate the effects of ART on OA by gross observation, morphological staining, immunohistochemistry, and enzyme-linked immunosorbent assay. Results: ART exhibited potent anti-inflammatory effects by inhibiting the expression of proinflammatory chemokines and cytokines, including interleukin (IL)-1β, IL-6, tumor necrosis factor alpha, and matrix metallopeptidase-13. It also showed favorable chondro-protective effect as evidenced by enhanced cell proliferation and viability, increased glycosaminoglycan deposition, prevention of chondrocyte apoptosis, and degeneration of cartilage. Further, ART inhibited OA progression and cartilage degradation via the Wnt/β-catenin signaling pathway, suggesting that it might serve as a Wnt/β-catenin antagonist to reduce inflammation and prevent cartilage degradation. Conclusion: In conclusion, ART alleviates IL-1β-mediated inflammatory response and OA progression by regulating the Wnt/β-catenin signaling pathway. Thereby, it might be developed as a potential therapeutic agent for OA.

Abstract In vitro expansion of mesenchymal stem cells (MSCs) has been implicated in loss of multipotency, leading to impaired chondrogenic potential and an eventual therapeutic effect, as reported in our previous study. However, the precise regulatory mechanism is still unclear. Here, we demonstrate that endoplasmic reticulum (ER) stress and the unfolded protein response (UPR) were involved in transformation of MSCs induced by in vitro culture based on the comparative profiling of in vitro cultured bone marrow MSCs at passage 3 (P3 BMSCs) vs. fresh P0 BMSCs by microarray analysis. Indeed, RT-PCR and Western blot analysis showed significantly lower expression levels of three key UPR-related molecules, ATF4, ATF6 and XBP1, in P3 BMSCs than P0 BMSCs. Further, we found that UPR suppression by 4-phenylbutyrate (4-PBA) reduced the chondrogenic potential of P0 BMSCs and further cartilage regeneration. Conversely, UPR induction by tunicamycin (TM) enhanced the chondrogenic differentiation of P3 BMSCs and the therapeutic effect on cartilage repair. Thus, the decline in the chondrogenic potential of stem cells after in vitro culture and expansion may be due to changes in ER stress and the UPR pathway.

Purpose: To investigate the early and mid-term results of open reduction and internal fixation (ORIF) with transarticular external fixation (TEF) but no deltoid ligament repair (DLR) in the treatment of supination-external rotation type IV equivalent (SER IV E) ankle fractures (AO/OTA classification 44-B 3.1) and provide evidence for clinical practice. Methods: This study cohort consisted of 22 patients with SER IV E ankle fractures that underwent ORIF with TEF but no DLR between December 2011 and December 2014. There were 13 males and 9 females, mean age 38.9 years (range, 17–73 years). Eight cases involved the left side and 14 the right side. The causes of fractures included road traffic accidents (11 cases), falling from height (6 cases) and sports injuries (5 cases). The mean period of hospitalization was 9.8 days (range, 6–14 days). For all the patients, MRI and three-dimensional CT were done before surgery and X-rays done preoperatively and during follow-ups. The external frame was kept for 8–10 weeks. The preoperative American Orthopedic Foot and Ankle Society (AOFAS) ankle-hindfoot score was 56.86 ± 4.400, the Medical Outcomes Short Form 36-item (SF-36) questionnaire score was 57.41 ± 4.102 and the visual analog score (VAS) was 5.50 ± 1.058. Patients' main complaints about inconvenience of daily life were also recorded. Results: All the 22 patients were followed up for 24–63 months (mean, 33.6 months). None of them developed nonunion during the follow-up; pin site infection was observed in one patient and posttraumatic osteoarthritis in another. At the final follow-up, the average AOFAS score, SF-36 score and VAS score were respectively 90.59 ± 5.096, 79.59 ± 5.394 and 1.82 ± 1.181, which were significantly improved compared with the preoperative data (t = 26.221, p

Purpose: To investigate the mid-term curative effects of the treatment of Pipkin type IV femoral head fractures using a reconstruction plate and bioabsorbable screws and provide the evidence for clinical practice. Methods: From February 2010 to September 2014, 21 patients with Pipkin type IV femoral head fractures were treated surgically. There were 13 males and 8 females with an average age of 41.1 years (range, 20–65 years). The causes of the fractures included traffic accidents (13 cases), falls from a height (four cases), heavy lifting injuries (three cases), and sport injury (one case). All patients were followed up with radiography and three-dimensional reconstruction computed tomography and other checks and any complications were actively managed. Closed reduction of fracture-dislocation of the hip was attempted under general anesthesia using the Kocher-Langenbeck approach. Femoral head fractures were treated with internal fixation or excision based on the size of the fracture fragments, whereas acetabular fractures were fixed with a reconstruction plate and screws following anatomic reduction. Results: The incisions healed by primary intention in all patients after surgery, without any infection, deep venous thrombosis, or other complications. All 21 patients were followed up for 36–76 months, with an average follow-up duration of 49 months. Postoperative imaging data showed that all dislocations and fractures were anatomically reduced, and bony union of the fractures was achieved. Heterotopic ossification was found in four patients, post-traumatic osteoarthritis in three, and avascular necrosis of the femoral head in two. At the final follow-up, the assessment of hip joint function according to the Thompson-Epstein scoring scale was excellent in 10 cases, good in six cases, fair in three cases, and poor in two cases. The rate of excellent and good functional outcomes was 76.1%. Conclusion: The mid-term curative effects of a reconstruction plate and bioabsorbable screws in the treatment of Pipkin type IV femoral head fractures is significant, and such the treatment can significantly improve the patient's joint function and quality of life. Objective: The aim of this study was to investigate the optimal timing for the resection of heterotopic ossification (HO) of the elbow. Methods: We retrospectively reviewed 42 patients who were treated operatively for heterotopic ossification of the elbow from March 2010 to December 2014 at our institution. The patients were divided into early (before 12 months) and late (after 12 months) excision groups. In the early excision group (17 patients), the average time from the initial injury to HO excision was 7.4 (3–11) months, and in the late excision group (25 patients), the average time was 33.5 (12–240) months. Every patient was evaluated by range of motion (ROM), the Mayo Elbow Performance Score (MEPS), postoperative complications and HO recurrence. Results: The preoperative mean ROM in the late excision group was greater than that of the early excision group, suggesting that the ROM is expected to increase even without surgery. Depth images play an important role in 3-D applications. However, due to the limitation of depth acquisition equipment, the acquired depth images are usually in limited resolution. In this paper, a spatially adaptive tensor total variation-Tikhonov model is proposed to solve this problem. The tensor total variation regularization is adopted to maintain sharp edges that reflect latent discontinuities in the real world, while the Tikhonov regularization ensures that depth changes smoothly inside objects. Furthermore, a fused edge map is proposed to indicate edge regions and balance both regularization terms. In edge regions, tensor total variation regularization is predominant, thus edge blurring artifacts are suppressed. In non-edge regions, Tikhonov regularization plays a more important role to suppress staircasing artifacts. Specifically, texture edges are removed in the fused edge map, and texture copying artifacts are avoided. Experimental results demonstrate the effectiveness and superiority of the proposed framework. Moreover, the proposed method yields much sharper edges and a lower percentage of bad pixels.

BACKGROUND: The aberrant expression of microRNAs (miRNAs) has been found in various types of cancer. miR-205 was reported to be upregulated in laryngeal squamous cell carcinoma (LSCC) tissues, however, the mechanisms by which miR-205 functions as a regulator of LSCC are largely unknown RESULTS: In this study, Real-time qPCR and Western blot assay showed that expression of miR-205 was upregulated and expression of cyclin-dependent kinase 2-associated protein 1 (CDK2AP1) was downregulated in LSCC tissues. The expression levels of miR-205 were negatively related to those of CDK2AP1 in LSCC tissues and cell lines. Moreover, we found that miR-205 was the upstream regulator of CDK2AP1 and could suppress the CDK2AP1 expression in LSCC cells. 3-(4,5-dimethylthiazal-2-yl)-2,5-diphenyl-tetrazolium bromide assays and transwell invasion assay were performed to test the proliferation and invasion of LSCC cells. Gelatin zymography was used to detect the activity of MMP2 and MMP9. CDK2AP1, c-Myc and CyclinD1 expression in cells was assessed with Western blotting. We found that miR-205 was the upstream regulator of CDK2AP1 and could suppress the expression of CDK2AP1 in LSCC cells. In addition, miR-205 significantly induced cell proliferation and invasion by suppressing CDK2AP1 expression. Consistent with miR-205 inhibitors, overexpressed CDK2AP1 suppressed the activity of MMP2 and MMP9 and c-Myc and CyclinD1 expression in LSCC cells CONCLUSION: These findings help us to better elucidate the molecular mechanisms of LSCC progression and provide a new theoretical basis to further investigate miR-205 as a potential biomarker and a promising approach for LSCC treatment

The popularity of big data from cell phone communication records makes cellular handoff probe system become an optimal choice for the collection of traffic speeds, especially for the large-scale freeway network. As a probe-based technique, the issues related to sample size are vital for the accuracy of cellular handoff probe system. Based on the current findings from simulation and field test, this study made an extensive discussion about the sample size problems using the field data. The subsequent investigation began with the correlation analysis between the sample size and the potentially influencing factors (the length of handoff link and traffic volume). Furthermore, a Weibull distribution model was set up to fit sample size for different handoff links. And then, the impact of sample size on the accuracy of cellular handoff probe system was following. Finally, based on the above explorations, several concluding remarks were presented as a reference for determining appropriate sample size to achieve high precision.

The internal structural details of unknown structures are very helpful to our personnel in carrying out activities during anti-terrorism operations and emergency disaster relief operations in urban environments. According to multipath identification and propagation mechanism separation technology, a building structure perspective imaging algorithm based on the ray-tracing model is proposed in this paper by making full use of the rich multipath delay power spectrum information and gradually filtering the multipath. Based on the imaging results taking overall consideration of all the propagation factors involved, the presented study is used to obtain the intricate geometric structure of indoor buildings. The original approach is then verified by using measured data, and the proposed algorithm is further optimized by extracting the internal wall structure of the building scene based on the Hough transform algorithm. In comparison with the real wall of the building, the average error of the inverse building wall position using simulated data is 0.071 m, and the average error of the inverse building wall position using the measured data is 0.355 m. To evaluate the effectiveness of limb-salvage treatment for osteosarcoma in adolescent patients, a comprehensive search on PubMed, Embase and Cochrane Library was conducted. Studies with a clear diagnosis of osteosarcoma were included and duplicate publications, studies without full text or incomplete information, those with an inability to extract data, divergent definitions of exposure, animal experiments, reviews, and systematic reviews were excluded. The data were analyzed using STATA 15.1. The findings of the present study revealed that overall survival (OS) and progression-free survival (PFS) of patients with osteosarcoma in the limb-salvage treatment group were significantly longer than those in the amputation treatment group [hazard ratio (HR)=0.71; 95% confidence interval (CI): 0.63-0.80; P=0.000 vs. HR=0.60; 95% CI: 0.48-0.76; P=0.000]. Additionally, the five-year OS rate for patients in the limb-salvage treatment group was higher than that in the amputation group [odds ratio (OR)=4.48; 95% CI: 2.74-7.31; P=0.000]. However, the local recurrence rate was notably higher in the limb-salvage treatment group compared with the amputation treatment group (OR=2.68; 95% CI: 1.50-4.77; P=0.001). Furthermore, the results indicated no significant difference in distant metastasis rates between the limb-salvage treatment group and the amputation treatment group (OR=0.32; 95% CI: 0.10-1.06; P=0.062). In conclusion, the present meta-analysis underscores the potential of limb-salvage therapy for adolescent patients with osteosarcoma. The OS and PFS of patients undergoing limb-salvage surgery are longer than those of amputation, with a higher five-year OS rate and a similar rate of distant metastasis.
