Development and validation of RP-HPLC method for the simultaneous determination of Glecaprevir & Pibrentasvir in pure form and their combined tablet dosage forms. The current separation was achieved on a Chromasil ODS Column (4.6 x 250mm, 5.0 μ) using the composition of the mobile phase of (0.1 M KH2PO4 buffer: methanol) in the ratio of (60:40) at pH 4.5. The pump flow rate was adjusted after several one milliliter per minute and both the samples were separated at 230nm. The retention times are 1.7 min and 2.3 min, for Glecaprevir and Pibrentasvir respectively with linear ranges 20 μg/ml-60 μg/ml and 50 μg/ml-150 μg/ml. The method was statistically validated for linearity, accuracy, precision and selectivity as per ICH guidelines. The drugs were subjected to stress conditions of hydrolysis, oxidation, photolysis of the RP-HPLC method. Both medications were tested under various stress forced conditions, including alkali, acid, oxidative, photodegradation, & hydrolysis degradation. The accuracy limit is the percentage recovery should be in the range of 98.0% - 101.0%. The total recovery was found to be 99.69% and 99.39% for Glecaprevir and Pibrentasvir. The validation of developed method shows that the accuracy is well within the limit, which shows that the method is capable of showing good accuracy and reproducibility. The robustness limit for mobile phase variation and flow rate variation are well within the limit, which shows that the method is having good system suitability and precision under given set of conditions. A simple, rapid, precise, accurate and sensitive reverse phase liquid chromatographic method has been developed for the simultaneous determination of Mosapride and Rabeprazole in bulk and pharmaceutical dosage form dosage form. The chromatographic method was standardized using Waters ODS (C18) RP Column, 250 mm x 4.6 mm. 5µm I.D column with UV detection at 255nm, Methanol: Ammonium Acetate buffer (pH=6) = 75:25and other conditions optimized were: flow rate (1.0 ml/minute), wavelength (255 nm), Run time was maintained at 8 minutes. The proposed method was successfully applied to the simultaneous determination of Mosapride and Rabeprazole in bulk and pharmaceutical dosage form. Optimized retention time of Mosapride and Rabeprazole is 2.069 and 2. 488. The method was linear over the range of 10µg/ml for Mosapride and Rabeprazole. The recovery was in the range of 98% to 102%. Linearity range was found to be 0-30 µg/ml for Mosapride and 0-60 µg/ml for Rabeprazole. The correlation coefficient was found to be 0.995. LOD was found to be 0.05µg/ml and LOQ was found to be 0.15µg/ml for Mosapride & The LOD was found to be 0.09µg/ml and LOQ was found to be 0.27µg/ml for Rabeprazole. The results of the forced degradation studies indicated the specificity of the developed method that has been developed. Different analytical performance parameters such as precision, accuracy, limit of detection, limit of quantification and robustness were determined according to International Conference on Harmonization (ICH) guidelines. Purpose: The present study was designed to study the Neomycin Sulfate (NS)-loaded hydrogels to heal wounds. Material and Method: NS-loaded hydrogels were prepared by simple stirring method. Results: The prepared formulation exhibited good consistency, homogeneity and gelling strength. The application of gel on healthy volunteers revealed gels to be safe for skin. It also exhibited pH between 7.4-8.0 and drug content between 100±4% evaluated through microbial assay. In-vitro drug release data showed a fast initial drug release during the first 1 h, followed by a slower and sustained release over 5 h. Conclusion: The finding suggests that NS based hydrogel is suitable for drug delivery through topical application to enhance the dermal contact time and improved patient compliance. Drug repurposing, also known as repositioning, is a technique in which existing drugs are repurposed to treat emerging and complex diseases like COVID-19. Because of the potential for shorter development timelines and lower overall costs, drug repurposing has emerged as a promising strategy. In the significant data era, artificial intelligence (AI) and network medicine provide cutting-edge information science applications to define disease, medicine, therapeutics, and identify targets with the smallest error. We present guidelines for using AI to accelerate drug repurposing or repositioning, for which AI approaches are formidable and required in this Review. We discuss how to use AI models in precision medicine, such as how AI models can accelerate COVID-19 drug repurposing. Rapidly developing, powerful, and innovative AI and network medicine technologies can help to accelerate therapeutic development. Bio-analysis is related to the analysis of analytes (drugs, metabolites, biomarkers) in biological matrix and it involves several steps from sample collection to analysis and data reporting. Sample collection from clinical or preclinical studies; then sending the samples to laboratory for analysis, later the sample preparation will be carried. Stable sample preparation method could be achieved. The role of sample preparation is to remove interferences from the biological matrix and to extract the analyte of interest and to improve analytical system performance. Sample preparation is often labor intensive and time consuming. Final step of the analysis is the quantification of analyte by using analytical instruments. Among them, Liquid chromatography-tandem mass spectrometry (LC-MS/MS) is method of choice in bioanalytical laboratories. This is due to high selectivity and high sensitivity of the LC-MS/MS technique. Development and validation of analytical method play an important role in the discovery, development and manufacturing of pharmaceuticals. Every year, number of drugs entered into the market; hence it is mandatory to develop newer analytical methods for such drugs. After the development, it becomes necessary to validate the new analytical method. Method development is the process which proves that the analytical method is acceptable for use. Validation of analytical method gives information about various stages and parameters like accuracy, precision, linearity, Limit of Detection, Limit of Quantification, specificity, range and robustness. A protocol is used to detect and measure biomolecules and metabolites in human and animal tissues using bimolecular methods. The biosanalinity method is effective at determining the number of drugs and metabolites in a biological system. New methods, the validation of existing procedures, and the analysis of samples are one of the prominent tasks for bioanalysis. Above all, a compound can be measured using several methods and identified by different methods of analysis. Drugs may be tested by several extraction techniques, including liquid extraction, solid-phase extraction, and protein precipitation in complex plasma and biological samples. To determine how the environment, matrix, or procedures impact the matrix estimation to the time of the analysis, all steps in the process must be investigated. The more detailed study of drug products can be performed with higher-pressure analytical techniques, such as high-extraction (HPLC), liquid chromatography coupled with double-mass spectrometry (LCMS/MS), and ultra-performance Liquid chromatography (UPLC). Both of them have flaws and strengths. At present, HPLC and GC usually perform biolysis. The parameters are linearity, repeatability, accuracy, selectivity, and continuity. The current studies describe the development and validation of an LC-MS-MS method for Cabozantinib in human plasma using naproxen as an internal standard using Quality by Design (QbD) (IS). For sample preparation, solidphase extraction was used and linearity was observed for drug concentrations ranging from 5 to 1000 ng/mL The method was systemically optimized using the Box-Behnken design, with mobile phase flow rate (X1), pH (X2), and mobile phase composition (X3) as method variables and retention time (Rt) (Y1) and peak area (Y2) as responses. Optimization studies revealed a reduction in the variability associated with method variables which improved method robustness. Validation studies of the developed method revealed that Cabozantinib in human plasma has good linearity, accuracy, precision, selectivity, and sensitivity. Stability studies in human plasma using freeze-thaw, bench-top, short-term and long-term cycles and auto sampler stability revealed no change in the drug's percent recovery.
