Reentry in nervous systems is the ongoing bidirectional exchange of signals along reciprocal axonal fibers linking two or more brain areas. The hypothesis that reentrant signaling serves as a general mechanism to couple the functioning of multiple areas of the cerebral cortex and thalamus was first proposed in 1977 and 1978 (Edelman, 1978). A review of the amount and diversity of supporting experimental evidence accumulated since then suggests that reentry is among the most important integrative mechanisms in vertebrate brains (Edelman, 1993). Moreover, these data prompt testable hypotheses regarding mechanisms that favor the development and evolution of reentrant neural architectures.

The Dynamic Core and Global Workspace hypotheses were independently put forward to provide mechanistic and biologically plausible accounts of how brains generate conscious mental content. The Dynamic Core proposes that reentrant neural activity in the thalamocortical system gives rise to conscious experience. Global Workspace reconciles the limited capacity of momentary conscious content with the vast repertoire of long-term memory. In this paper we show the close relationship between the two hypotheses. This relationship allows for a strictly biological account of phenomenal experience and subjectivity that is consistent with mounting experimental evidence. We examine the constraints on causal analyses of consciousness and suggest that there is now sufficient evidence to consider the design and construction of a conscious artifact.

Recent recordings of place field activity in rodent hippocampus have revealed correlates of current, recent past, and imminent future events in spatial memory tasks. To analyze these properties, we used a brain-based device, Darwin XI, that incorporated a detailed model of medial temporal structures shaped by experience-dependent synaptic activity. Darwin XI was tested on a plus maze in which it approached a goal arm from different start arms. In the task, a journey corresponded to the route from a particular starting point to a particular goal. During maze navigation, the device developed place-dependent responses in its simulated hippocampus. Journey-dependent place fields, whose activity differed in different journeys through the same maze arm, were found in the recordings of simulated CA1 neuronal units. We also found an approximately equal number of journey-independent place fields. The journey-dependent responses were either retrospective, where activity was present in the goal arm, or prospective, where activity was present in the start arm. Detailed analysis of network dynamics of the neural simulation during behavior revealed that many different neural pathways could stimulate any single CA1 unit. That analysis also revealed that place activity was driven more by hippocampal and entorhinal cortical influences than by sensory cortical input. Moreover, journey-dependent activity was driven more strongly by hippocampal influence than journey-independent activity.

Analyzing neural dynamics underlying complex behavior is a major challenge in systems neurobiology. To meet this challenge through computational neuroscience, we have constructed a brain-based device (Darwin X) that interacts with a real environment, and whose behavior is guided by a simulated nervous system incorporating detailed aspects of the anatomy and physiology of the hippocampus and its surrounding regions. Darwin X integrates cues from its environment to solve a spatial memory task. Place-specific units, similar to place cells in the rodent, emerged by integrating visual and self-movement cues during exploration without prior assumptions in the model about environmental inputs. Because synthetic neural modeling using brain-based devices allows recording from all elements of the simulated nervous system during behavior, we were able to identify different functional hippocampal pathways. We did this by tracing back from reference neuronal units in the CA1 region of the simulated hippocampus to all of the synaptically connected units that were coactive during a particular exploratory behavior. Our analysis identified a number of different functional pathways within the simulated hippocampus that incorporate either the perforant path or the trisynaptic loop. Place fields, which were activated by the trisynaptic circuit, tended to be more selective and informative. However, place units that were activated by the perforant path were prevalent in the model and were crucial for generating appropriate exploratory behavior. Thus, in the model, different functional pathways influence place field activity and, hence, behavior during navigation.

A neuronal network inspired by the anatomy of the cerebral cortex was simulated to study the self-organization of spiking neurons into neuronal groups. The network consisted of 100 000 reentrantly interconnected neurons exhibiting known types of cortical firing patterns, receptor kinetics, short-term plasticity and long-term spike-timing-dependent plasticity (STDP), as well as a distribution of axonal conduction delays. The dynamics of the network allowed us to study the fine temporal structure of emerging firing patterns with millisecond resolution. We found that the interplay between STDP and conduction delays gave rise to the spontaneous formation of neuronal groups--sets of strongly connected neurons capable of firing time-locked, although not necessarily synchronous, spikes. Despite the noise present in the model, such groups repeatedly generated patterns of activity with millisecond spike-timing precision. Exploration of the model allowed us to characterize various group properties, including spatial distribution, size, growth, rate of birth, lifespan, and persistence in the presence of synaptic turnover. Localized coherent input resulted in shifts of receptive and projective fields in the model similar to those observed in vivo.

Sleep is a ubiquitous component of animal life, and prolonged sleep deprivation is fatal in both vertebrates and invertebrates. The physiologic function of sleep, however, is not known. We propose here that sleep provides a period of time necessary to reapportion resources within neurons and neural systems that become sub-optimally distributed during active waking. Three specific examples of such reapportionment during sleep are suggested: (1) the return of the neurotransmitter, glutamate, to synaptic vesicles at presynaptic sites most active during waking, (2) the intracellular movement of mitochondria from neuronal processes to the cells soma where mitochondrial replication can occur, and (3) the readjustment of the level and distribution of neurotransmitters within the brainstem modulatory systems and elsewhere that must function in an integrated fashion during waking. Experimental approaches that might be utilized to test these hypotheses are suggested.

This paper presents evidence indicating that the signals generated by neural responses to visual input can be either enhanced by increasing or suppressed by decreasing the area of the stimuli to which attention is directed. We used magnetoencephalography (MEG) to measure the frequency-tagged steady-state visual evoked responses of 11 subjects presented with two superimposed images flickering at different frequencies. Each image consisted of seven parallel bars of equal length; in any image, all bars were either red or green and either horizontal or vertical. At randomly chosen times during the experiments, any one of the three middle bars in either image transiently increased or decreased in width. Subjects were asked to attend to one image and ignore the other and to respond to changes in bar width in the attended image with a key press. In one condition, subject responses were required for changes in any of the three central bars of the attended image. We found that visual steady-state evoked responses to the attended image were enhanced relative to those evoked by the unattended image in this condition. In a second condition, subject responses were required for changes only in the middle bar. In this condition, the responses to the attended image were suppressed relative to those of the unattended image. These results may reflect relative differences in the synchronization and desynchronization of responding neuronal populations.

Degeneracy, the ability of elements that are structurally different to perform the same function or yield the same output, is a well known characteristic of the genetic code and immune systems. Here, we point out that degeneracy is a ubiquitous biological property and argue that it is a feature of complexity at genetic, cellular, system, and population levels. Furthermore, it is both necessary for, and an inevitable outcome of, natural selection.

Excitatory synaptic activity can evoke transient and substantial elevations of postsynaptic calcium. Downstream effects of elevated calcium include the activation of the calcium-dependent protease calpain. We have developed a reagent that identifies dendritic spines in which calpain has been activated. A fusion protein was expressed that contained enhanced yellow and enhanced cyan fluorescent protein (EYFP and ECFP, respectively) linked by a peptide that included the micro-calpain cleavage site from alpha-spectrin. A PDZ-binding site fused to ECFP anchored this protein to postsynaptic densities. The fusion protein exhibited fluorescence resonance energy transfer (FRET), and diminution of FRET by proteolysis was used to localize calpain activity in situ by fluorescence microscopy. Incubation of the fusion protein with calpain in the presence of calcium resulted in the separation of EYFP and ECFP into monomeric fluorophores. In transiently transfected cell lines and dissociated hippocampal neurons, FRET was diminished by raising intracellular calcium levels with an ionophore or with glutamatergic agonists. Calpain inhibitors blocked these changes. Under control conditions, FRET levels in different dendritic spines of cultured neurons and in hippocampal slices were heterogeneous but showed robust decreases upon treatment with glutamatergic agonists. Immunostaining of cultured neurons with antibodies to a spectrin epitope produced by calpain-mediated digestion revealed an inverse correlation between the amount of FRET present at postsynaptic elements and the concentration of spectrin breakdown products. These results suggest that the FRET methodology identifies sites of synaptically induced calpain activity and that it may be useful in analyzing synapses undergoing changes in efficacy.

Evidence that excitatory amino acids act via N-methyl-D-aspartate (NMDA) receptors to evoke the release of catecholamines from axonal terminals and synaptosomes has been used to argue for the presence of pre-synaptic NMDA receptors. NMDA receptor agonists also generate nitric oxide (NO) which rapidly diffuses through neural tissue. We find that exogenously applied NO evokes [3H]-dopamine release from cultured neurons. This release is not blocked by the NMDA antagonist MK-801 nor by tetrodotoxin. Both NG-nitroarginine which inhibits NO synthesis, and hemoglobin which binds extracellular NO, block NMDA-evoked [3H]-dopamine release from striatal slices. A major role of endogenously-synthesized NO may be to evoke neurotransmitter release in local volumes of neural tissue.

The genomic sequences encoding a chicken Ca(2+)-dependent cell adhesion molecule (K-CAM) were recently found to be located approximately 600 base pairs upstream from the translation initiation site of a homologous protein, liver CAM. The sizes of 11 exons of the K-CAM gene are almost identical to those in the L-CAM (liver cell-adhesion molecule) gene with exon-intron junctions occurring at exactly equivalent positions. The sizes and sequences of most introns were, however, much more dissimilar. More detailed comparisons of these two genes reveal that the sequences of the last two exons (exons 15 and 16) are far more alike than are any of the other exons; they are, in fact, almost identical. The introns that separate these exons in the two genes likewise resemble one another far more than do any other corresponding introns. This degree of similarity appears to be far greater than can be accounted for by selection for protein structure, and it strongly suggests that a gene conversion event occurred between these two genes subsequent to their duplication and divergence to perform different biological functions.

In the vertebrate central nervous system, afferent axons find their appropriate target structures under the influence of local environmental cues. In many target regions, appropriate patterns of activity in the afferents are also required to establish normal mappings between the source cells and the target region. Specific mappings arise in these targets because temporal contiguity in firing is somehow transformed into spatial contiguity of synaptic contacts. In this article, we propose a theory that utilizes the covariance of a transient diffusive signal produced at active synapses with the firing of presynaptic axon terminals to account for many of these activity-dependent features of vertebrate neuroanatomy. Computer simulations of the growth of axons within a three-dimensional volume of neural tissue reveal the generality of the proposed mechanism in accounting for the self-organization of a broad range of diverse neuroanatomical structures, including those in the cerebral cortex. The proposed mechanism is consistent with detailed anatomical and physiological data, and direct experimental tests of predictions of the theory are suggested.

Several observations suggest that the Ca2(+)-dependent postsynaptic release of nitric oxide (NO) may be important in the formation and function of the vertebrate nervous system. We explore here the hypothesis that the release of NO and its subsequent diffusion may be critically related to three aspects of nervous system function: (i) synaptic plasticity and long-term potentiation in certain regions of the adult nervous system, (ii) the control of cerebral blood flow in such regions, and (iii) the establishment and activity-dependent refinement of axonal projections during the later stages of development. In this paper, we detail and analyze the basic assumptions underlying this NO hypothesis and describe a computer simulation of a minimal version of the hypothesis. In the simulation, a 3-dimensional volume of neuropil is presented with patterned afferent input; NO is produced, diffuses, and is destroyed; and synaptic strengths are determined by a set of synaptic rules based on the correlation of synaptic depolarization and NO levels. According to the hypothesis, voltage-dependent postsynaptic release of this rapidly diffusing substance links the activities of neurons in a local volume of tissue, regardless of whether the neurons are directly connected by synapses. This property is demonstrated in the simulation, and it is this property that is exploited in the hypothesis to account for certain aspects of long-term potentiation and activity-dependent sharpening of axonal arbors.

Recent experiments have revealed tightly synchronized oscillatory discharges in local assemblies of cortical neurons as well as phase coherency of oscillations at distant cortical sites. These findings are consistent with the theory of neuronal group selection, a population theory of brain function that is based on the properties of local groups of neurons. A set of computer simulations shows that cooperative interactions within and among neuronal groups can generate the observed phenomena. In the simulations, oscillations within neuronal groups are generated through local excitatory and inhibitory interactions. Different groups in general oscillate in an uncorrelated fashion. Coherency of the oscillatory activity of different neuronal groups depends crucially on reciprocal reentrant signaling and can reflect the spatial continuity of a stimulus. Separated or discontinuous features of a given stimulus can be transiently associated in a temporally coherent pattern through reentrant signaling between groups in networks responding to different aspects of that stimulus. A simulation of reentrant activity between arrays of neuronal groups selective for oriented lines and pattern motion displays cross-correlations between groups that are responsive to different parts of a stimulus contour if these parts move together. Such coherency among neuronal groups might be used in the discrimination of a stimulus from other stationary or differentially moving elements in a visual scene.

The L polypeptide chains of certain Bence-Jones proteins of group I have been found in three forms: monomers of molecular weight of about 20,000, dimers which monomerize in dissociating solvents, and dimers which are stable in such solvents. The L polypeptide chains of some Bence-Jones proteins of group II were found to occur naturally only as stable dimers. The L chains of normal human gamma-globulin have been obtained in a reduced unalkylated form, and a fraction of these chains was found to form stable dimers under oxidizing conditions. It is suggested that a single disulfide bond is involved in stabilization of the dimer. In experiments on the reconstitution of 7S gamma-globulin, it was found that stable dimers of L polypeptide chains did not associate appreciably with H(gamma) chains to form a soluble product. L chains in the monomeric form, both of a reduced alkylated Bence-Jones protein and of reduced unalkylated gamma-globulin, combined with H(gamma) chains to form a 7S product. After hydrolysis with papain, the 7S material containing the Bence-Jones L chains yielded fragments comparable to the fragments of papain-treated myeloma proteins. As indicated by spectrofluorometric measurements, dissociable dimers and stable dimers of the L chains of a Bence-Jones protein both underwent identical thermally induced transitions in the temperature range 48-58 degrees C. When L polypeptide chains were present in reduced alkylated gamma-globulin or reduced alkylated S fragments, no transition occurred until 65 degrees C, the coagulation temperature of gamma-globulin and S fragments. Above this temperature, L chains were released into solution. These experiments suggested that free L chains and L chains bound to H(gamma) chains have different conformational stabilities.

An exonuclease which specifically degrades double-standard DNA has been isolated from rabbit tissues. The enzyme has an approximate molecular weight of 42,000, requires a divalent metal ion as cofactor, and attacks DNA at the 5'-terminal ends, thereby liberating 5'-mononucleotides. It degrades several synthetic polydeoxynucleotides of single repeating base sequences more rapidly than DNA from natural sources. The specificity of this mammalian enzyme resembles that of several microbial enzymes (phage lambda exonuclease and DNA polymerase) which appear to be required for repair and recombination of DNA.

The chemical relations among Bence-Jones proteins, myeloma proteins, and normal gamma-globulins have been investigated by a variety of means. Starch gel electrophoresis in 8 M urea of reduced alkylated Bence-Jones proteins yielded patterns of bands corresponding to those of the light (L) polypeptide chains of the dissociated myeloma protein from the same patient. One instance in which this correspondence was found was chosen for extensive study. Chromatography on carboxymethylcellulose in 6 M urea was employed to isolate the light (L) polypeptide chains and heavy (H) polypeptide chains of the completely reduced and alkylated myeloma protein. Isolation of similarly treated Bence-Jones protein from the same patient corroborated the correspondence to the L chains of the myeloma protein. Amino acid analyses indicated that the compositions of the Bence-Jones protein and the L chains of the myeloma protein were identical. Moreover, the thermosolubility properties and spectrofluorometric behavior of the isolated L chains and Bence-Jones protein were similar. Ultracentrifugal analyses of the L chains of normal human 7S gamma-globulin showed that their molecular weight in 6 M urea was 20,000. In aqueous solution their molecular weight was 41,000, suggesting that they exist as dimers under these conditions. The L chains of normal human gamma-globulin were found to have reversible thermosolubility properties similar to those of Bence-Jones proteins. The H chains of normal human gamma-globulin did not share these properties. Using spectrofluorometric methods, characteristic molecular transitions were found upon heating Bence-Jones proteins and L chains. These transitions were indicated by an increase in the intensity of fluorescence at well defined temperatures as well as by reversible shifts in the wavelength of maximal emission. The findings suggest that Bence-Jones proteins are composed of L chains of the type found in normal and pathological gamma-globu]ins.
