Eighty consecutive cases of inpatients were enrolled. Patient’s disability was evaluated using both of the aforementioned tools, before and after the rehabilitation program.Results: The WHODAS 2.0 score was, on average, 12.21% higher than the Modified Barthel Index, before the surgical intervention. Modified Barthel Index measures could be considered as a determinant and a predictor of length of stay.Conclusion: The Modified Barthel Index is limited, since it does not consider a patient’s perspective. The WHODAS 2.0 scale fully considers a patient’s perception of disability. Therefore, both assessment scales should be administered in clinical practice, in order to provide integration of clinical information with a patient’s reported outcome measures. 4. Genetic structure at CSN1S1, CSN2 and CSN1S2 loci of four authoctonous goat breed reared in Lazio and Campania regions

Abstract

One main aspect of safeguard plans for livestock Autochthonous Genetic Types (AGTs) is to ensure economic sustainability of their farming. This encourages farmers to contribute to the pres- ervation of AGTs. The aim of this study was to characterize the genetic structure of four AGTs of endangered goats at the αS1, β and αS2 calcium sensitive caseins encoding genes CSN1S1, CSN2 and CSN1S2, respectively. The peculiar polymorphisms detected at these loci are associated with milk quali-quantitative charac- teristics and have been largely investigated in widespread dairy goat breeds. On the contrary, there are very few studies carried out in AGTs goats. This study was performed on 50 Neapolitan goats, typical of the Campania region, and on 61 goats which included the three breeds Monticellana, Capestrina and Grigia Ciociara, reared in Lazio region (Lazio AGTs goat). Genotyping of quantitative alleles at CSN1S1, CSN2 and CSN1S2 loci was performed according to previous studies. At the CSN1S1 locus a high variability was observed in all four AGTs. In particular, a higher frequency of the null allele N was observed in Neapolitan goats than Lazio AGTs goats (0.420 vs. 0.008). On the contrary a higher frequency of F allele, associated with a low αS1 casein content in milk, was observed in Lazio AGTs goat (0.442) than Neapolitan goat (0.120). All four AGTs do not carry the null alleles CSN1S1 01, CSN2 01 and CSN1S2 0 and the alleles associated with an intermediate content of αS1 (CSN1S1 E) and αS2(CSN1S2 D) caseins in milk. The genotyping data obtained pro- vide useful information regarding the productive aptitudes of the four AGTs studied. In fact, the high frequency of CSN1S1 N allele in Neapolitan goat, predisposes this breed to produce low-protein milk for fresh consumption while the high frequency of the allele CSN1S1 F in the Lazio AGTs goat addresses their milk to process- ing for the production of typical cheese with unique organoleptic and flavor characteristics. Developing safeguard plans that take these results into account will have a higher chance of success, with a consequent increase in the number of animals raised.

Abstract

Il presente documento restituisce una sintesi delle attività del progetto IMPACT ed è diviso in due parti. La prima parte evidenzia gli strumenti completati durante la durata del progetto, in particolare la rete di radar costieri per il monitoraggio delle correnti marine e la piattaforma webGIS per la consultazione dei dati raccolti ed elaborati dai partner di progetto. La seconda parte capitalizza sui suddetti strumenti per fornire sia indicazioni metodologiche che di governance. Nello specifico, la rete di radar costieri viene utilizzata per creare mappe di potenziale contaminazione e indicare in che condizioni le attività portuali possano essere più sostenibili. Le misure demografiche e i calcoli di ritenzione indicano il livello d'efficacia delle dimensioni attuali delle AMP mentre le misure di contaminazione suggeriscono di considerare un numero maggiore di stazioni nell'ottica di migliorare i piani di monitoraggio esistenti. In quest'ottica, le indicazioni rappresentano il piano d'azione e le linee guida del progetto. L'ulteriore espansione della rete di radar costieri prevista nei progetti SICOMAR plus e SINAPSI è la prova che la conclusione delle attività del progetto IMPACT sia in realtà solo un importante punto di partenza.

18. A common exocytotic mechanism mediates axonal and dendritic outgrowth

Abstract

Outgrowth of the dendrites and the axon is the basis of the establishment of the neuronal shape, and it requires addition of new membrane to both growing processes. It is not yet clear whether one or two exocytotic pathways are responsible for the respective outgrowth of axons and dendrites. We have previously shown that tetanus neurotoxin-insensitive vesicle-associated membrane protein (TI-VAMP) defines a novel network of tubulovesicular structures present both at the leading edge of elongating dendrites and axons of immature hippocampal neurons developing in primary culture and that TI-VAMP is an essential protein for neurite outgrowth in PC12 cells. Here we show that the expression of the N-terminal domain of TI-VAMP inhibits the outgrowth of both dendrites and axons in neurons in primary culture. This effect is more prominent at the earliest stages of the development of neurons in vitro. Expression of the N-terminal domain deleted form of TI-VAMP has the opposite effect. This constitutively active form of TI-VAMP localizes as the endogenous protein, particularly concentrating at the leading edge of growing axons. Our results suggest that a common exocytotic mechanism that relies on TI-VAMP mediates both axonal and dendritic outgrowth in developing neurons.

20. Subcellular localization of tetanus neurotoxin-insensitive vesicle-associated membrane protein (VAMP)/VAMP7 in neuronal cells: evidence for a novel membrane compartment

Abstract

The clostridial neurotoxin-insensitive soluble N-ethylmaleimide-sensitive factor attachment protein (SNAP) receptors, tetanus neurotoxin-insensitive (TI)-vesicle-associated membrane protein (VAMP)/VAMP7, SNAP23, and syntaxin 3 have recently been implicated in transport of exocytotic vesicles to the apical plasma membrane of epithelial cells. This pathway had been shown previously to be insensitive to tetanus neurotoxin and botulinum neurotoxin F. TI-VAMP/VAMP7 is also a good candidate to be implicated in an exocytotic pathway involved in neurite outgrowth because tetanus neurotoxin does not inhibit this process in conditions in which it abolishes neurotransmitter release. We have now found that TI-VAMP/VAMP7 has a widespread distribution in the adult rat brain in which its localization strikingly differs from that of nerve terminal markers. TI-VAMP/VAMP7 does not enrich in synaptic vesicles nor in large dense-core granules but is associated with light membranes. In hippocampal neurons developing in vitro, TI-VAMP/VAMP7 localizes to vesicles in the axonal and dendritic outgrowths and concentrates into the leading edge of the growth cone, a region devoid of synaptobrevin 2, before synaptogenesis. After the onset of synaptogenesis, TI-VAMP/VAMP7 is found predominantly in the somatodendritic domain. In PC12 cells, TI-VAMP/VAMP7 does not colocalize with synaptobrevin 2, chromogranin B, or several markers of endocytic compartments. At the electron microscopic level, TI-VAMP/VAMP7 is mainly associated with tubules and vesicles. Altogether, these results suggest that TI-VAMP/VAMP7 defines a novel membrane compartment in neurite outgrowths and in the somatodendritic domain.

23. A novel dual mechanism controlling the localization and function of exocytic v-SNAREs

Abstract

SNARE proteins are essential for membrane fusion but their regulation is not yet fully understood. We have previously shown that the amino-terminal Longin domain [Filippini, 2001 #8431] of the v-SNARE TI-VAMP/VAMP7 plays an inhibitory role in neurite outgrowth [Martinez-Arca, 2001 #8432; Martinez-Arca, 2000 #828]. The goal of this study was to investigate the regulation of TI-VAMP as a model of v-SNARE regulation. We show here that the Longin domain of TI-VAMP plays a dual role. Firstly, it negatively regulates the ability of TI-VAMP and the Longin/Synaptobrevin2 chimera to participate in SNARE complexes. Secondly, it interacts with the AP-3 adaptor complex and this interaction targets TI-VAMP to late endosomes. Accordingly, in mocha cells lacking AP-3d, TI-VAMP is retained in an early endosomal compartment. Furthermore, TI-VAMPc, a novel isoform of TI-VAMP lacking a one third of the Longin domain, does not interact with AP-3 and therefore it is not targeted to late endosomes, but this shorter Longin domain yet inhibits SNARE complex formation in vivo. These findings support a novel mechanism controlling both localization and function of exocytic v-SNAREs through the Longin domain and clathrin adaptors. They point to the amino-terminal domains of SNAREs as multifunctional modules responsible for the fine-tuning of SNARE function.

Abstract

Longins are the only R-SNAREs that are common to all eukaryotes and are characterized by a conserved N-terminal domain with a profilin-like fold called a longin domain (LD). These domains seem to be essential for regulating membrane trafficking and they mediate unexpected biochemical functions via a range of protein-protein and intramolecular binding specificities. In addition to the longins, proteins involved in the regulation of intracellular trafficking, such as subunits of the adaptor and transport protein particle complexes, also have LD-like folds. The functions and cellular localization of longins are regulated at several levels and the longin prototypes TI-VAMP, Sec22 and Ykt6 show different distributions among eukaryotes, reflecting their modular and functional diversity. In mammals, TI-VAMP and Ykt6 are crucial for neuronal function, and defects in longin structure or function might underlie some human neurological pathologies.

27. SNAP-25 modulation of calcium dynamics underlies differences in GABAergic and glutamatergic responsiveness to depolarization

Abstract

SNAP-25 is a component of the SNARE complex implicated in synaptic vesicle exocytosis. In this study, we demonstrate that hippocampal GABAergic synapses, both in culture and in brain, lack SNAP-25 and are resistant to the action of botulinum toxins type A and E, which cleave this SNARE protein. Relative to glutamatergic neurons, which express SNAP-25, GABAergic cells were characterized by a higher calcium responsiveness to depolarization. Exogenous expression of SNAP-25 in GABAergic interneurons lowered calcium responsiveness, and SNAP-25 silencing in glutamatergic neurons increased calcium elevations evoked by depolarization. Expression of SNAP-25(1-197) but not of SNAP-25(1-180) inhibited calcium responsiveness, pointing to the involvement of the 180-197 residues in the observed function. These data indicate that SNAP-25 is crucial for the regulation of intracellular calcium dynamics and, possibly, of network excitability. SNAP-25 is therefore a multifunctional protein that participates in exocytotic function both at the mechanistic and at the regulatory level.

Abstract

The possibility of selecting the initial angular-momentum state from which antiproton-proton annihilation at rest can occur can be a fundamental tool in meson spectroscopy. With the OBELIX detector different target systems have been employed, ranging fromS-wave toP-wave dominance. The preliminary results both on meson spectroscopy and branching ratio measurements are discussed.

Abstract

A study of nˉp in-flight annihilations is presented. The 4π invariant mass spectrum resulting from the analysis of the five-prong exclusive events, following the annihilation of ∼ (100–297) MeV/c nˉ, reveals, besides a large structure centred at ∼ 1500 MeV/c2, some evidences for a new state around 1660 MeV/c2.

Abstract

The dipeptide N-acetyl-aspartyl-glutamic acid (NAAG) present in brain and spinal cord tissues may act as a neurotransmitter at excitatory synapses in the central nervous system. However, pharmacological and biochemical studies of NAAG are hampered by its rapid inactivation in vivo and in vitro by an enzyme that cleaves NAAG into N-acetyl-aspartate and glutamate. This enzyme has been previously named N-acetylated alpha-linked acidic dipeptidase (NAALADase). Based upon our earlier studies on the specificity of this enzyme, we have now designed new competitive inhibitors of this peptidase. N-Succinyl-glutamic acid, 4, was almost as potent as N-acetyl-beta-aspartyl-glutamic acid (beta-NAAG), 2, in inhibiting the hydrolysis of [Glu-3H]NAAG by rat brain membranes, with an IC50 value in the micromolar range. The analogous affinities of the substrate NAAG and of N-succinyl-glutamic acid suggest that the N-acetyl moiety is not an absolute requirement for entry into the active site of the enzyme. Therefore, the acronym NAALADase seems to be incorrect, and peptidase activity against NAAG will be used throughout this manuscript when referring to the enzyme that cleaves NAAG and whose activity is inhibited by quisqualate and beta-NAAG. Two N-acylated glutamic acid analogs, 5 and 6, were also found to be effective inhibitors of the in vitro degradation of NAAG, with Ki values in the micromolar range. Compounds 5 and 6 possess two free carboxylic functions on the N-acyl moiety, one of which could interact with the S1 subsite of the enzyme; the other could chelate the Zn++ cation involved in the catalytic hydrolysis of NAAG.(ABSTRACT TRUNCATED AT 250 WORDS)
