This study hypothesized that a screening tool, SMARTIE, would improve detection of these risks pre-TAVI, and thus be predictive of postoperative delirium (POD) and 30-day mortality post-TAVI.Design: Prospective observational cohort study, using a historical cohort for comparison.Participants: A total of 234 patients (age: 82.2±6.7 years, 59.4% male) were included. Half were screened using SMARTIE.Methods: The SMARTIE cohort was assessed for cognitive deficits and depressive symptoms using the Mini-Cog test and PHQ-2, respectively. Measures of frailty included activities of daily living inventory, the Timed Up and Go test and grip strength. For the pre-SMARTIE cohort, we extracted cognitive deficits, depression and frailty symptoms from clinic charts. The incidence of POD and 30-day mortality were recorded. Bivariate chi-square analysis or t-tests were used to report associations between SMARTIE and pre-SMARTIE groups. Multivariable logistic regression models were employed to identify independent predictors of POD and 30-day mortality.Results: More patients were identified with cognitive deficits (χ2=11.73, p=0.001), depressive symptoms (χ2=8.15, p=0.004), and physical frailty (χ2=5.73, p=0.017) using SMARTIE. Abstract

The past two Decadal Surveys in Astronomy and Astrophysics recommended the completion of a space-based interferometry mission, known today as SIM PlanetQuest, for its unique ability to detect and characterize nearby rocky planets (Bahcall 1991, McKee & Taylor 2001), as well as contributions to a broad range of problems in astrophysics. Numerous committees of the National Research Council as well as NASA Roadmaps have similarly highlighted SIM as the one technology that offers detection and characterization of rocky planets around nearby stars and which is technically ready. To date, SIM remains the only program with the capability of detecting and confirming rocky planets in the habitable zones of nearby solar-type stars. Moreover, SIM measures masses and three-dimensional orbits of habitable planets around nearby stars (within 25 pc); these are the only stars for which follow-up by other techniques is feasible, such as space-based spectroscopy, ground-based interferometry, and of course TPF., Comment: ExoPTF white paper 8 pages, 2 figures

13. Non-motor and neuropsychiatric features of Parkinson's disease

Abstract

This thesis focuses on non-motor symptoms (NMS) of Parkinson’s disease (PD), particularly the neuropsychiatric symptoms of visual hallucinations, apathy and impulse control disorders. A clinical cohort study and pathology study were performed. PD patients (N=94) were recruited and each underwent neurological examination for motor aspects of the disease, detailed multi-domain cognitive assessment including executive and visuoperceptive function, and a battery of validated non-motor assessments for symptoms including mood, apathy, sleep, fatigue, psychosis, autonomic function, disability and health-related quality of life (QOL). A subgroup (N=50) also had detailed ophthalmological examination. In the autopsy study, the presence of ante-mortem visual hallucinations (VH) in PD donors was correlated with Lewy body (LB) density and distribution at different cortical locations. The clinical study examined the impact of NMS on QOL and found that NMS, particularly depression, had stronger association with QOL than motor scores, but despite this NMS were often under-reported by their treating neurologist. A significant proportion of pathologically confirmed PD patients presented exclusively with NMS and this led to misdiagnosis and, in some cases, inappropriate investigations and treatment. An aetiological model of VH was proposed and examined; factors investigated included dopaminergic medication, higher cortical function, sleep and ophthalmic pathology. Clinical associations of VH were rapid eye movement sleep behaviour disorder, autonomic function, and executive and visuoperceptive cortical function. There was however no clear association with co-existing ophthalmic pathology. In the post-mortem analysis, LB involvement at corresponding cortical areas was confirmed, including frontal (executive) and temporal and trans-entorhinal (visuoperception) cortices. A meta-analysis of studies reporting pathological gambling was performed and an association with dopaminergic medication, particularly dopamine agonist therapy, and co-existing psychopathology was confirmed.

Abstract

The Spitzer Space Telescope, NASA's Great Observatory for infrared astronomy, was launched 2003 August 25 and is returning excellent scientific data from its Earth-trailing solar orbit. Spitzer combines the intrinsic sensitivity achievable with a cryogenic telescope in space with the great imaging and spectroscopic power of modern detector arrays to provide the user community with huge gains in capability for exploration of the cosmos in the infrared. The observatory systems are largely performing as expected and the projected cryogenic lifetime is in excess of 5 years. This paper summarizes the on-orbit scientific, technical and operational performance of Spitzer. Subsequent papers in this special issue describe the Spitzer instruments in detail and highlight many of the exciting scientific results obtained during the first six months of the Spitzer mission., Comment: Accepted for publication in the Astrophyscial Journal Supplement Spitzer Special Issue, 22 pages, 3 figures. Abstract

The resolution of SARS-CoV-2 replication hinges on cell-mediated immunity, wherein CD8+ T cells play a vital role. Nonetheless, the characterization of the specificity and TCR composition of CD8+ T cells targeting non-spike protein of SARS-CoV-2 before and after infection remains incomplete. Here, we analyzed CD8+ T cells recognizing six epitopes from the SARS-CoV-2 nucleocapsid (N) protein and found that SARS-CoV-2 infection slightly increased the frequencies of N-recognizing CD8+ T cells but significantly enhanced activation-induced proliferation compared to that of the uninfected donors. The frequencies of N-specific CD8+ T cells and their proliferative response to stimulation did not decrease over one year. We identified the N222-230 peptide (LLLDRLNQL, referred to as LLL thereafter) as a dominant epitope that elicited the greatest proliferative response from both convalescent and uninfected donors. Single-cell sequencing of T cell receptors (TCR) from LLL-specific CD8+ T cells revealed highly restricted Vα gene usage (TRAV12-2) with limited CDR3α motifs, supported by structural characterization of the TCR–LLL–HLA-A2 complex. Lastly, transcriptome analysis of LLL-specific CD8+ T cells from donors who had expansion (expanders) or no expansion (non-expanders) after in vitro stimulation identified increased chromatin modification and innate immune functions of CD8+ T cells in non-expanders. These results suggests that SARS-CoV-2 infection induces LLL-specific CD8+ T cell responses with a restricted TCR repertoire. Although SARS-CoV2 epitope characterization has been the focus of extensive research, these efforts have largely focused on the spike protein. Abstract

The spatial and temporal variation of plasmaspheric mass throughout the inner magnetosphere as a function of changing geomagnetic and solar wind conditions is a consequence of its transport throughout the system. It is proposed that the global measurement of 30.4 -nm EUV emission from plasmaspheric He+ can be used to derive the mass transport pathways, as well as the dynamic conditions and locations of when specific pathways are active. This is to be accomplished using observations obtained by the Extreme Ultraviolet Instrument (EUV) on the Imager for Magnetopause -to-Aurora Global Exploration Mission (IMAGE). Questions to be addressed by this approach include evaluation of whether plasmaspheric transport fully accounts for the plasma lost through erosion a nd plume formation, through inward compression of the plasmapause, or by plasma redistributed back into the ionosphere? Does plasmaspheric erosion begin near 3h MLT, sweeping plasma dawnward and duskward as some report? Can the mass in a persistent plume be accounted for by continued loss from within the body of the plasmasphere or must the ionosphere be an active source? These questions are part of the broader unresolved morphology of the global circulation of plasmaspheric plasma through the magnetosphere to be lost into the solar wind, recirculated, or returned to the ionosphere. An initial demonstrations of the methodology and corresponding results will be presented.

Abstract

Establishing the abundance and physical properties of regolith and boulders on asteroids is crucial for understanding the formation and degradation mechanisms at work on their surfaces. Using images and thermal data from NASA's Origins, Spectral Interpretation, Resource Identification, and Security-Regolith Explorer (OSIRIS-REx) spacecraft, we show that asteroid (101955) Bennu's surface is globally rough, dense with boulders, and low in albedo. The number of boulders is surprising given Bennu's moderate thermal inertia, suggesting that simple models linking thermal inertia to particle size do not adequately capture the complexity relating these properties. At the same time, we find evidence for a wide range of particle sizes with distinct albedo characteristics. Our findings imply that ages of Bennu's surface particles span from the disruption of the asteroid's parent body (boulders) to recent in situ production (micrometre-scale particles).

35. Advantages and Challenges of Using Physics Curricula as a Model for Reforming an Undergraduate Biology Course

Abstract

We report on the development of a life sciences curriculum, targeted to undergraduate students, which was modeled after a commercially available physics curriculum and based on aspects of how people learn. Our paper describes the collaborative development process and necessary modifications required to apply a physics pedagogical model in a life sciences context. While some approaches were easily adapted, others provided significant challenges. Among these challenges were: representations of energy, introducing definitions, the placement of Scientists' Ideas, and the replicability of data. In modifying the curriculum to address these challenges, we have come to see them as speaking to deeper differences between the disciplines, namely that introductory physics--for example, Newton's laws, magnetism, light--is a science of pairwise interaction, while introductory biology--for example, photosynthesis, evolution, cycling of matter in ecosystems--is a science of linked processes, and we suggest that this is how the two disciplines are presented in introductory classes. We illustrate this tension through an analysis of our adaptations of the physics curriculum for instruction on the cycling of matter and energy; we show that modifications of the physics curriculum to address the biological framework promotes strong gains in student understanding of these topics, as evidenced by analysis of student work. Abstract

CRM197 is a genetically detoxified mutant of diphtheria toxin (DT) that is widely used as a carrier protein in conjugate vaccines. Protective immune responses to several bacterial diseases are obtained by coupling CRM197 to glycans from these pathogens. Wild‐type DT has been described in two oligomeric forms: a monomer and a domain‐swapped dimer. Their proportions depend on the chemical conditions and especially the pH, with a large kinetic barrier to interconversion. A similar situation occurs in CRM197, where the monomer is preferred for vaccine synthesis. Despite 30 years of research and the increasing application of CRM197 in conjugate vaccines, until now all of its available crystal structures have been dimeric. Here, CRM197 was expressed as a soluble, intracellular protein in an Escherichia coli strain engineered to have an oxidative cytoplasm. The purified product, called EcoCRM, remained monomeric throughout crystallization. The structure of monomeric EcoCRM is reported at 2.0 Å resolution with the domain‐swapping hinge loop (residues 379–387) in an extended, exposed conformation, similar to monomeric wild‐type DT. The structure enables comparisons across expression systems and across oligomeric states, with implications for monomer–dimer interconversion and for the optimization of conjugation. Abstract

August 21, 2017 provided a unique opportunity to investigate the effects of the total solar eclipse on high frequency (HF) radio propagation and ionospheric variability. In Marshall Space Flight Center's partnership with the US Space and Rocket Center (USSRC) and Austin Peay State University (APSU), we engaged citizen scientists and students in an investigation of the effects of an eclipse on the mid-latitude ionosphere. Activities included fieldwork and station-based data collection of HF Amateur Radio frequency bands and VLF radio waves before, during, and after the eclipse to build a continuous record of changing propagation conditions as the moon's shadow marched across the United States. Post-eclipse radio propagation analysis provided insights into ionospheric variability due to the eclipse.

Abstract

Fetal body composition is an important determinant of body composition at birth, and it is likely to be an important determinant at later stages in life. The purpose of this work is to provide a comprehensive overview by presenting data from previously published studies that report on body composition during fetal development in newborns and the infant/child through 5 years of age. Understanding the changes in body composition that occur both in utero and during infancy and childhood, and how they may be related, may help inform evidence-based practice during pregnancy and childhood. We describe body composition measurement techniques from the in utero period to 5 years of age, and identify gaps in knowledge to direct future research efforts. Available literature on chemical and cadaver analyses of fetal studies during gestation is presented to show the timing and accretion rates of adipose and lean tissues. Quantitative and qualitative aspects of fetal lean and fat mass accretion could be especially useful in the clinical setting for diagnostic purposes. The practicality of different pediatric body composition measurement methods in the clinical setting is discussed by presenting the assumptions and limitations associated with each method that may assist the clinician in characterizing the health and nutritional status of the fetus, infant and child. It is our hope that this review will help guide future research efforts directed at increasing the understanding of how body composition in early development may be associated with chronic diseases in later life.
