The paper considers the creation of a diagnostic system based on morphological and biophysical indicators that allows us to understand the influence of the climatic component, as well as stress factors on the general physiological state of young people for further correction of preventive measures and preservation of the health of the peoples of the Far North. The aim of the study is a comprehensive assessment of the health of young people living in the Far North based on the results of bioluminescent saliva activity, stress-emotional state obtained before and after intense mental stress, as well as anthropometric and bioimpedance indicators. 117 students of the Medical Institute of Surgut State University took part in the survey at a young age (18-23 years) during the educational process (rest) and during the examination session (mental load) in the first half of the year of study (autumn-winter 2021). The stress-emotional state was assessed using the Spielberger-Khanin test, the functional state of students They were studied using bioluminescent salivary fluid testing, anthropometric and bioimpedance methods. Samples were obtained by spitting unstimulated saliva into a test tube. As a result of the study, regional features of the physique of young people were revealed, which indicates the formation of a phenotype adapted to living conditions in the northern region. The relationship between the stress-emotional status of adolescents and the level of inhibition of the bioluminescent enzymatic system by saliva samples taken before and after mental stress was shown. The value of the residual glow had an increased indicator for the state of rest and significantly reduced the intensity of the bioluminescent glow under mental stress. The methodological development obtained in the future will make it possible to effectively manage the methods and methods of diagnosing young people, taking into account the influence of external factors, climatic zones and conditions.

The issue of factors that modify the tumor process stays relevant. The effect of unilateral sciatic nerve ligation on the growth of Guerin's transplantable carcinoma and the lifespan of white outbred rats of the same age, which differed in adaptation status and aging rates, was studied.Material and Methods. The motor activity (open field test), the character and tension of the general nonspecific adaptional reactions of the body (AR) according to Garkavi–Kvakina–Ukolova, the dynamics of tumor sizes and the lifespan of rats after Guerin’s carcinoma transplantation were evaluated.Results. The effect of unilateral sciatic nerve ligation differed from the unidirectional negative effects known in tumor-bearing animals after bilateral ligation of the sciatic nerve. In groups with unilateral ligation of the sciatic nerve and a false operation, more than 40 % of animals showed an increase in lifespan compared with the maximum lifespan in the control group. At the same time, in the most cases, the tumor growth rate was similar to the control indicators or exceeded them (more 25 % of cases). A temporary inhibition of tumor growth was observed only in individual animals. There was no direct relationship between tumor growth or lifespan and the degree of decrease in the motor activity of animals 4 weeks after nerve ligation. A correlation between the changes in the ARs and lifespans of animals and, to a lesser extent, the dynamics of tumor growth was observed. The distinct negative effect of increased aging rate, measured by animal weight, on tumor development and lifespan in studied rats was shown, but not in the cases of sciatic nerve ligation. Unilateral sciatic nerve ligation had a multidirectional effect on tumor growth and lifespan in rats with different rates of aging, depending, probably, on the individual pain sensitivity and the individual features of systemic regulation of tumor-bearing animals.Conclusion. The results reflect the complex relationship between processes associated with chronic pain, oncogenesis, aging and features of neuroendocrine and immune regulation of experimental animals. The question of the reasons for the preservation of viability in animals that underwent surgery and ligation of the sciatic nerve, when the tumors reach large sizes, exceeding this indicator in the control group, needs to be clarified.

A systematic review of modern methods of experimental study of ovarian cancer using traditional (immunocompetent, genetically engineered and immunodeficient) and non-traditional (that don’t belong to the mammals) animal models,established and primary cultures of human ovarian cancer, including three-dimensional organotypic spheroids (3D- models ex vivo) is presented. The prospects of the considered models for studying of the pathogenesis of various molecular-genetic and histological variants of ovarian cancer, as well as for developing methods of personalized treatment, are discussed. The limitations of modern animal models are indicated. The greatest attention is paid to studies on immunodeficient animals using xenografts based on established cultures of human ovarian cancer cells and on tumor tissue obtained directly from the patients (patient derived xenografts, PDX). The questions of various variants of xenograft transplantation with an emphasis on the problems of orthotopic transplantation of human ovarian cancer into immunodeficient mice and the relevance of methods for local humanization in heterotopic transplantation are considered. The most promising, from the point of the author’s view, approaches to studying the effectiveness of drug therapy for ovarian cancer in immunodeficient animal models are outlined. To prepare a systematic review, a literature search was carried out on the Scopus, Web of Science, Med Line, PubMed, Cyber Leninka, RSCI databases. The analysis used literature sources indexed in the Scopus and Web of Science databases (97%) and the RSCI. More than 60% of the works amount has been published over the past 5 years.

e17549 Background: The question of the mechanisms of platinum resistance development in advanced ovarian cancer (OC) remains open. The aim of the study was to reveal the relationship between the proliferative activity of OC cells, the disruption of apoptosis processes and the changes in the activity of transporter proteins that provide the efflux of chemotherapy drugs. Methods: Patients with advanced OC (T3-4N0-3M0-1) of postmenopausal age, sensitive (SPtPs, n = 23) and resistant to platinum drugs (RPtPs, n = 17), operated on after 3-6 cycles of neoadjuvant chemotherapy, were retrospectively studied. The parameters of proliferative activity (ki67), apoptosis disturbances (p53), as well as the expression of BCRP and Pgp transporter proteins were evaluated in OC tissue using immunohistochemistry methods. Statistical analysis was performed using the Mann-Whitney and Pearson tests (χ2), as well as the Spearman's rank correlation coefficient (SC). Results: In total, the dominance of the RPtPs over the SPtPs was noted in terms of intensity of the studied indicators (p < 0.05-0.003), the most significant for Pgp expression. At the same time, moderate and high values of these indicators were observed in SPtPs in 42-61% of cases, which reduced the predictive value of the revealed differences. It was of interest that statistically significant correlations between the indicators in SPtPs differed from those in RPtPs. Thus, the following values of the SC were observed in SPtPs: +0.848 (ki67-p53), -0.675 (ki67-BCRP), -0.575 (p53-BCR). In RPtPs, another intensity and/or direction of these relations were noted: +0.521 (ki 67-p53), +0.500 (ki 67-BCRP), +0.705 (p53-BCR). There were no statistically significant relationships between ki67-Pgp expression in SPtPs and RPtPs. The direction and intensity of the studied relationships could indicate changes in cell regulation in primary OC in SPtPs compared to RPtPs. Thus, in SPtPs, they reflected a significant contribution of apoptotic disturbances to the enhancement of OC cell proliferation, which was limited by the negative feedback mechanism via BCRP activity suppression indicated by the presence of an inverse correlation ki67-BCRP. In RPtPs, this mechanism was apparently already lost, which resulted in a change in the direction of the statistical relationship ki67-BCRP from reverse to direct one. Under these conditions, the positive contribution of apoptosis disruption to the proliferative activity of OC cells could obviously be supplemented by a synergistic increase in the activity of the BCRP transporter protein, which removes platinum drugs from OC cells. Conclusions: Patients with advanced OC may develop platinum resistance due to discoordination of proliferation, apoptosis, and regulation of the activity of the BCRP transporter protein.

e15055 Background: Enhanced generation of reactive oxygen species (ROS) is an important factor in cell damage when exposed to metal nanoparticles (NPs), which can be used to develop antitumor strategies based on ROS level modulation. However, an increase in ROS can cause oxidative stress in the body. The aim of this study was to evaluate the parameters of the redox-regulating system in the blood erythrocytes of tumor-bearing rats at different efficiencies of exposure to copper NPs (CuNPs) and iron NPs (FeNPs). Methods: Outbred male rats with transplantable fusiform cell fibrosarcoma (S45) were treated with copper (group S45+CuNPs, n = 37) or iron NPs (group S45+FeNPs, n = 33). During 15 days, 35 rats received 8 intratumoral injections and 35 rats received 8 intraperitoneal injections of NPs in total doses of 10 mg/kg bw, the control S45 rats (n = 20) received saline i.p. Levels of malondialdehyde (MDA) and glutathione (GSH), the activities of catalase (CA), superoxide dismutase (SOD), and glutathione peroxidase (GPO) in the red blood cells were determined one week after the end of the treatment. Results: Complete or near complete tumor regression was recorded in 13 (35 %) rats with CuNPs treatment and 6 (27%) rats with FeNPs treatment. Decrease in tumor size by 30 to 80% in 14 (38%) rats in CuNPs group and in 14 (43%) rats in FeNPs group was observed. In other cases, tumor growth in rats with S45 treated with metal nanoparticles was similar to animals in the control group. The values of the studied parameters in rats with complete tumor regression after exposure to CuNPs did not differ from those in the group of healthy rats. Unidirectional changes in the studied parameters were observed in rats with partial S45 regression or with no effect. GPO activity increased by 43-57% (p = 0.02-0.002) and GSH level by 20-27% (p = 0.034-0.005), but MDA level was 62-65% (p = 0.027-0.007) higher than normal values. Animals with complete tumor regression after the administration of FeNPs showed changes in the activity of peroxide-inactivating enzymes: an increase in GPO by 84% (p = 0.009) and a decrease in catalase by 31% (p = 0.016) relative to normal values. In animals with partial response or without response, GPO activity increased by 75% and 57%, respectively (p = 0.031-0.042), while CA activity decreased by an average of 37% (p = 0.002-0.0002). No increase in MDA was observed in the S45+FeNPs group. The levels of MDA in rats of the control group were increased by 46% (p = 0.0032), which was combined with an increase in GSH by 19%, a decrease in catalase activity by 16%, and an increase in GPO activity by 41% (p˂0.05 in all cases), compared to the values in intact animals. Conclusions: Regardless of the route of administration, copper and iron NPs did not cause an increase in oxidative stress in rats with fibrosarcoma. The maximum antitumor effect of metal NPs was accompanied by normalization or positive reorganization of the redox-regulating system of erythrocytes.

The severity of pain and changes in the adaptational status were studied in patients with brain metastases or cervical cancer receiving xenon therapy after whole brain radiotherapy or after radical hysterectomy. Hematological indicators of the nature and tensiton of general nonspecific adaptional reactions of the body (ARs) by Garkavi-Kvakina-Ukolova, the QLQ-C15 questionnaire and a 10-point graphic visual analogue scale for the assessment of the intensity of pain were used. Xenon caused concurrent reduce in the intensity of pain and improvement of characteristics of ARs in all studied patients. The results suggested an association between the analgesic effect of xenon and the normalization of neuroimmune processes and reduced damaging effects of special antitumor treatment on the body under the influence of xenon.

Upcoming professional sports authorities seek rapid noninvasive biosensing tools for regular monitoring of athletes' physiological states. The analysis of saliva through luminescence-based biosensors has been perceived as a suitable candidate for such purposes. The present study reports a qualitative bioluminescence assay based on a coupled enzyme system that consists of bacterial luciferase (BLuc) and nicotinamide adenine dinucleotide (NADH):flavin mononucleotide (FMN) oxidoreductase (Red), BLuc-Red, for the express diagnostics of athletes' stress levels before and after physical exertion. The volunteers who participated in the study were grouped as freestyle wrestlers and students who adapted to different levels of physical activities. Under physical exertion modelling conditions, the influence of participant saliva on BLuc-Red catalyzed light emission was investigated. Results showed a significant increase in residual luminescence (Iexp , mean maximum bioluminescence intensity of the experimental measurement (Iexp ); Ic , luminescence intensity in control; Iexp /Ic , %) values for participants in the wrestler group while a decrease in the student group (P < 0.05). Such contrasting residual luminescence values in both groups were found to be dependent on the catalase activity of saliva. The proposed bioluminescence assay can be utilized as a potential nonspecific biosensing tool for determining the physical state of athletes under high loads.

e17530 Background: Despite the progress made in diagnosis and treatment, ovarian cancer (OC) is still the third most common genital cancer. Poor results of therapy for various forms of OC stimulate the search for fundamentally new approaches to the treatment. One of the main reasons for resistance to chemotherapeutic agents includes multidrug resistance (MDR) of tumor cells. The most characterized of its various mechanisms is the elevated activity of the ABC family protein - ABCB1 (Pgp or MDR1). The purpose of the study was evaluation of the expression of this ABC transporter as a predictive factor in platinum-containing chemotherapy in OC patients. Methods: 100 patients aged 29-79 years with advanced stage IIIC - IV OC with/without ascites were recruited between 2016 and 2020. Depending on the treatment results, patients were divided into 2 groups: group 1 (n = 59) - patients with platinum sensitivity; group 2 (n = 41) - patients with platinum resistance during the treatment or within 6 months after its completion. IHC analysis was performed using rabbit polyclonal anti-MDR1 antibodies (Affinity Biosciences) diluted 1:600 and the Reveal Polyvalent HRP-DAB Detection System. The percentage and the intensity of staining were assessed: 0, 1+ weak, 2+ moderate, 3+ strong. MDR1 expression was considered positive when staining was detected in more than 10% (cut-off) tumor cells with intensities of 2+ and 3+. Statistical analysis of results was performed in the Statistica 13.0 program (StatSoftInc., USA). Results: Patients with MDR1+ prevailed in group 2 (98%), and patients with MDR1- in group 1 (32%). The average expression of MDR1 in tumor cells in group 2 (64.0±7.0) was statistically significantly higher by 1.7 times (p = 0.003) than in group 1 (37.2±6.8) by the Mann-Whitney U-test. When distributed according to Pearson's χ2 criterion, positive MDR1 as a risk factor in the platinum-resistant group increased the risks of refractory to platinum therapy by 19 times (95% CI 2.4-148.8). Conclusions: The study demonstrated predictive significance of the MDR1 biomarker in platinum treatment in patients with ovarian cancer.

e14027 Background: The existing modern standards of combination treatment of HGG patients do not provide recovery and a long-term favorable prognosis, and the increasong incidence of HGG determines the need for additional effective technologies for anticancer and decongestant therapy. One of such methods involves TMS, and we have reported its preliminary assessment earlier (DOI: 10.1200/JCO.2020.38.15_suppl.2545). In this study, we continued the observation to examine the survival of patients. Methods: Patients with HGG received combination treatment: stage 1 – surgical removal of tumors within visible unaltered tissues; stage 2 – radiation therapy (the Varian Novalis linear accelerator) to the bed of the removed tumor, single boost dose 2 Gy, total boost dose 60 Gy; stage 3 – multi-course chemotherapy: temozolomide 150 mg/m² on days 1-5 with a 23-day interval. Starting from the second day after surgery, patients of group 1 (n = 25) received 10 TMS sessions, and during radiotherapy – 15 TMS sessions. Patients of group 2 (n = 25) received combination treatment without TMS. 6 and 12 months after the surgery, survival of patients was assessed with the Kaplan-Meier method and the Log-Rank test. Results: After 6 months of the follow-up, the survival of patients in group 1 remained at 100%, while in the control group it decreased to 88.8±8.7%. The difference in the 1-year overall survival was even more pronounced: in group 1, it was 68.5±10.4%, exceeding the value in group 2 (52.0±7.5%.) The differences were statistically significant (Log-Rank test p = 0.001). Conclusions: The results confirm the effectiveness of accompanying TMS in the early postoperative period, as well as at the stage of radiation therapy. The undoubted effectiveness of the considered techniques makes it expedient to include this type of treatment in the combination therapy for HGG patients. e23517 Background: The relevance of studying multiple primary malignant tumors (MPMTs) is determined by poor understanding of their pathogenesis. Our purpose was to create an experimental model of synchronous MPMTs with the stimulation of malignant growth of tumors with different histostructure. Methods: The study included 20 male BALB/c Nude mice. The main group included mice with simultaneous subcutaneous inoculation of tumors: murine B16/F10 melanoma (0.5 ml of suspension diluted 1:20 in saline solution) below the left scapula and rat sarcoma 45 (0.5 million tumor cells in 0.5 ml of saline solution) below the right scapula. Control groups included males with melanoma or sarcoma inoculated at the same dosage and volume as in the main group. Results: In the model of synchronous MPMTs, tumors appeared faster than in controls: melanoma–by 3 times, sarcoma–by 2 times; their volume was larger: melanoma–by 2.2 times, sarcoma–by 3.2 times. Melanoma metastasized, in addition to typical sites (the lungs, spleen, liver), into sarcoma 45 to the side adjacent to the chest, under the tumor node. The survival of mice with MPMTs was lower. The morphological structure of melanoma metastasis into sarcoma 45 was represented by large lamellar-rounded epithelium-like cells of melanosarcoma type with transparent cytoplasm and nuclei with a high frequency of pathological mitosis figures. Bundles of elongated spindle-shaped melanocytes with processes of the cytoplasm were determined in some melanoma areas, as well as alveolar and concentric structures. Conclusions: Synchronous subcutaneous inoculation of murine B16/F10 melanoma and rat sarcoma 45 to male BALB/c Nude mice increases their malignant potential due to an exchange of “structural information"; as a result, sarcoma acquires impulses of proliferative activity of melanoma, which, in turn, adapts and “mimics” a tumor into which it is going to metastasize.

e17527 Background: Resistance to chemotherapy is a complex problem in the treatment of patients with ovarian cancer (OC), and this phenomenon is associated, among other things, with the presence of cancer stem cells (CSCs). The purpose of the study was to determine the amount of CSCs in tumors of OC patients with different responses to chemotherapy they receive. Methods: Samples of ovarian tumors were obtained from 100 patients (aged 29-79 years) with advanced ovarian cancer stage IIIC-IV with/without ascites. All patients recruited in 2016-2020 received standard combination treatment with surgery and platinum-containing polychemotherapy (Pt CT). Based on the results of neoadjuvant Pt CT, patients were divided into groups: patients with stabilization or progression–group 1 “without effect”; patients with complete/partial regression–group 2 with positive effect. All patients gave their informed consent for the study. Cell suspension obtained from the tumor fragments was processed using monoclonal antibodies labeled with various fluorochromes: CD45-APC-Cy7, CD44-FITС, CD133–РЕ according to the manufacturer's instructions (BD, USA). The percentage of CSCs was determined using the FACS Canto II flow cytometer (BD, USA). The amount of cells with CSC markers (CD44+, CD133+, CD44+CD133+) was calculated as a percentage from the total amount of CD45--cells. Statistics: STATISTICA 13 (StatSoftInc., USA). Results were presented as Me (LQ; UQ). The significance of differences was assessed using the Mann-Whitney test (the differences were considered significant at p- _cells, as well as CD45-CD44+ cells, in OC tumors did not differ between patients of groups 1 and 2: 79.0 (72.1; 85.9) vs. 82.8 (76.7; 89.0), and 6.2 (3.3; 9.1) vs. 6.6 (3.5; 9.8), respectively. The percentage of cancer cells with the CD45-CD133+ phenotype in group 2 was significantly lower than in group 1: by 52% (p≤0.05), 3.2 (2.4; 3.9) vs. 6.7 (3.8; 9.6), respectively. The amount of CD45-CD44+CD133- CSCs in tumors in group 1 was lower than in group 2: by 82% (p≤0.05), 0.6 (0.3; 0.7) vs. 3.3 (1.8; 4.7), respectively. Conclusions: The results demonstrated certain differences in the distribution of CSCs in tumors of OC patients with different sensitivity to chemotherapy, which may be one of the factors determining the treatment outcomes.

e21583 Background: Multiple primary malignant tumors (MPMTs) are characterized by the presence of several primary neoplasms in one patient. The purpose of the study was to create an experimental model of MPMTs with one dominating tumor. Methods: The study included 21 female BALB/c Nude mice. The main group included mice with simultaneous subcutaneous inoculation of tumors: Guerin's carcinoma (0.5 million tumor cells in 0.5 ml of saline solution) under the right scapula and B16/F10 melanoma (0.5 ml of suspension diluted 1:20 in saline solution) under the left scapula. Control groups included females with melanoma or carcinoma inoculated at the same dosage and volume as in the main group. Results: In a MPMT model, tumors appeared 3 times faster than in controls and demonstrated larger volumes: melanoma – by 7.5 times, carcinoma – by 2.2 times; the survival of mice with MPMTs decreased. Carcinoma in a MPMT model metastasized to melanoma and almost completely suppressed its growth. Melanoma was represented by a small “island” of tumor tissue 3-4 mm in diameter and was located just under the skin at the site of injection of melanoma cells. The light part of the same loose pasty consistency as the dark part, with a diameter of 6-7 mm, was located around the dark “center” of melanoma. The rest part of the tumor located under the left scapula looked like an elongated grayish-pink node of a dense elastic consistency - just like the tumor located under the right scapula, which was much larger in volume. The right and left tumors did not merge with each other; there was a small distance of about 2-3 mm between them. A small lesion of caseous necrosis, 6–7 mm in diameter, was recorded in the center of the right tumor node of Guerin's carcinoma; there was no necrosis in the left tumor. Smaller size, the absence of necrosis and visually more “young” carcinoma tissue on the left indicated its later appearance than that on the right, which, in combination with the remnants of melanoma fused to the left tumor and the absence of “contact” between the left tumor and the right one, indicated the metastatic nature of Guerin’s carcinoma on the left. B16/F10 melanoma did not metastasize. Conclusions: In simultaneous subcutaneous inoculation of murine B16/F10 melanoma and rat Guerin’s carcinoma to female BALB/c Nude mice, carcinoma cells metastasized to melanoma and suppressed its growth.

e21557 Background: 10% of cancer patients have comorbidities accompanied by chronic pain. Neurotrophins and fibrinolytic system are involved in carcinogenesis and pain pathogenesis. The purpose of the study was to measure levels of neurotrophins in white matter of the brain of urokinase-deficient (uPA–) mice with B16/F10 melanoma growing in presence of chronic neuropathic pain (CNP). Methods: The study included female mice С57ВL/6 (normal genome uPA+, n = 40) and C57BL/6-Plautm1.1BugThisPlauGFDhu/GFDhu (urokinase gene-knockout uPA–, n = 28) with B16/F10 melanoma (M) implanted subcutaneously 2 weeks after bilateral sciatic nerve ligation (CNP model). Intact mice (I) were controls. Levels of brain-derived neurotrophic factor BDNF, nerve growth factor NGF-β and neurotrophins 3 and 4 (NT3, NT4) were measured by ELISA in white matter of the brain after 3 weeks of tumor growth in presence of CNP. Results: Tumor volume in (uPA–) females by week 3 of carcinogenesis was 0.04 cm3, which was 70 times smaller than in (uPA+) females. Tumor volume in (uPA–) females with CNP was 5.76 cm3, which was 144 times larger than in (uPA–) females without CNP, and in (uPA+) females – 2.5 cm3. The brain of I (uPA–) showed higher levels of NT3 (by 1.3 times, p < 0.05), NT4 (by 2.6 times) and NGF-β (by 1.9 times, p < 0.05) and lower BDNF (by 1.7 times, p < 0.05), compared to I (uPA+). Both strains of mice with M or CNP demonstrated decreased levels of NGF-β, more pronounced in animals with a combination of these factors. (uPA–) females with CNP+M showed a decrease of NT3 and BDNF by 2 times, with NGF-β 2.2 times higher than in (uPA+) mice. Conclusions: The study revealed underlying differences in levels of neurotrophins in the brain of (uPA–) females which could contribute to the creation of conditions for the inhibition of tumor growth. Changes in the levels of NGF-β in mice with melanoma or CNP were nonspecific. Changes in the BDNF, NT3 and NGF-β balance in the brain of (uPA–) mice may be part of the mechanism of greater stimulation of melanoma growth under the influence of CNP.

The activity of key ferments in the Krebs cycle (SDH) and glycolysis (α-GPDH) in peripheral blood lymphocytes in animals and humans with cancer pathology is considered herein. Identified is a close relationship between the fermentative activity and non-specific integral reactions of an organism, the type of which can be regulated by factors of electromagnetic nature. Monitoring of signal energetic and adaptive reactions permits to adequately evaluate an efficacy of treatment and predict recovery.

This review considers the current data on the use of nanoparticles of biogenic metals and their oxides in antineoplastic treatment, the role that the metals play in important regulatory and metabolic processes, their immunotropic effects, and the possible effects on the electromagnetic parameters of cell–cell interactions. Analysis of the available data and original in vivo experimental results indicates that the antitumor potential of these agents is underestimated. Avenues of further research that may contribute to the development of new effective anticancer nanotechnologies are discussed.

The methods for activating antitumor resistance mechanisms that were developed using the concept of the periodic system of general nonspecific adaptational reactions of the body are briefly reviewed. The principles that allow effective impacts with electromagnetic radiation and biologically active substances are described. The criteria and concepts of the theory of adaptational reactions are compared to some of the concepts and categories of synergetics. The specific features in the dynamics of the studied parameters that are induced by effective impacts are considered. The antistress nature of the systemic effect that is caused by ferromagnetic nanoparticles on animals with grafted tumors is shown. Possible mechanisms that underlie the regression of large tumors that is caused by two different factors, viz., modulated electromagnetic radiation and magnetite nanoparticles, are discussed. The cases where the order parameter changed with the development of antistress unresponsiveness and regression of experimental tumors caused by a combined electromagnetic exposure are analyzed.

e16698 Background: The coexistence of neuroendocrine tumors and pancreatic adenocarcinoma is rare, and treatment of such mixed tumors is challenging due to the differences in their natural course and response to systemic therapy. There is growing evidence that vitamins affect the biology of pancreatic tumors. The purpose of the study was to measure concentrations of retinol (RET), α-tocopherol (α-TCP) and diene conjugates (DC) in the blood of patients with pancreatic cancer in order to reveal its pathogenetic characteristics. Methods: Blood levels of RET and α-TCP (ELISA methods, Cloud-Clone Corp, USA), their ratio and DC concentrations (biochemical method) were measured before treatment if 42 patients with pancreatic cancer: adenocarcinoma (AC), T1-3N0-1M0, n = 9; AC with a neuroendocrine component (AC+NE) (up to 30%), n = 21; neuroendocrine tumors (NET), T1-3N0-1M0, n = 12. 22 healthy men of similar age were controls. All patients gave their voluntary informed consent for the study. Results: RET levels in all patient were statistically significantly lower than in controls: in AC by 3.8 times, in AC+NE by 1.9 times, in NET by 3.7 times (p = 0.0000). Concentrations of α-TCP in AC were 1.6 times (p = 0.0011) lower than in controls, in AC+NE were similar, and in NET α-TCP was 1.5 times higher than in controls (p = 0.0000). The ratio of α-TCP/RET in all patients significantly exceeded control values: in AC by 2.2 times, in AC+NE by 1.6 times, in NET by 5.7 times (p = 0.0000). Levels of DC in all patients were higher than in controls: in AC by 2.5, in AC+NE by 2.1, in NET by 2.7 times (p = 0.0001). Conclusions: Changes in serum levels of RET and α-TCP differ in patients with AC, NET and mixed tumors, which causes changes in the balance of vitamins and can contribute to a prooxidant effect, as evidenced by an increase in DC levels.

e15650 Background: The aim of the study was to assess the effect of metallic nanoparticles on growth of transplanted ascitic tumor sarcoma 37 in mice. Methods: Nanoparticles (NP) of transition metals were experimentally developed and kindly provided us by Prof. V.B. Borodulin. Ultradispersed metallic powders (NP Cu, NP Zn, NP Fe) 30-100 nm sized were dispersed by ultrasound in 0.85% NaCl before use and daily administrated intraperitoneally to tumor-bearing mice with ascitic sarcoma 37. Injections of NP 500 µl (10 µg/ml) were performed for 4 days, and 4 days after the completion of the course mice were sacrificed. The control group received 0.85% NaCl. Ascitic fluid volume was measured and living cells were counted. Results: In all the tumor-bearing mice the progression of ascites was noted: it was maximal in controls, less in animals having received NP Fe and NP Cu, minimal – in mice after injection of NP Zn. The volume of the latter was 0.78±0.31 ml in comparison with control data: 1.79±0.66 ml (p < 0.05); it was also less than in mice after administration of NP Cu (2.16±0.68 ml) and NP Fe (3.3±1.3 ml). Similar difference was seen in the amount of tumor cells in ascitic fluid: (1.67±0.5)х106/ml after the injection of NP Zn and (27.7±2.86)х106/мл in the control group. Effect of NP Cu and NP Fe was less pronounced than of NP Zn though obvious in spite of the noted tendency of the increase of the ascitic fluid volume. The most significant changes were observed when absolute amount of living tumor cells was counted per one mouse – it was minimal after NP Zn administration (0.636±0.2)х106/ml and maximal in the control group (23.98±10.5)х106/ml), i.e. only 2.6% of tumor cells had survived after NP Zn injection. Conclusions: Our results demonstrate that metallic NP were able to produce antitumor in vivo without any supplements like laser or hyperthermia: the effect was likely to depend upon the metal and was the highest in NP Zn compared to NP Cu and Fe.

e15616 Background: The question of the effects of comorbid diseases on oncogenesis has been little studied else. The increase in the frequency of dorsopathies and arthritis with age determines the important of the question about their influence on malignant process. The aim of the research was to study the features of the tumor growth and animal's condition under chronic neurogenic and inflammatory pain. Methods: In experiments on 54 white unbred male rats of 280-390 g, with transplantable Guerin's carcinoma (сG), two experimental models of chronic pain were used - unilateral sciatic nerve ligation (SNL) according to G.J. Bennet (neurogenic pain) and injections of formalin (Fm) into the ankle joint of the back paw (inflammatory pain). Formalin injection is known to be used to form both acute and chronic pain. The dynamics of tumors size, level of motor activity (MAL) in open field test, hematological parameters (by the blood analyzer «Exigo EOS vet», Sweden), adaptation status (AS) by Selye-Garkavi-Kvakina and also the lifespan of the animals (LS) were evaluated. Results: Unlike the unidirectional negative influence of chronic pain known in tumor-bearing animals in the cases of bilateral SNL, two opposite effects were observed with unilateral SNL and the formalin injection. Along with accelerated tumor growth in some animals (up to 62%), increase of LS of more than a third of the rats of the main groups by 3-30% in comparative with maximal LS in the control group was noted. In these cases, often the death of animals occurred with the larger tumors than in the control groups. In the cases of unilateral SNL, a relation between changes in the characteristics of the AS and the LS was noted. In the cases of Fm injections, animals with diverse LS differed in the features of the inflammatory reaction and the dynamics of the MAL. The prognostic value of shifts of the MAL in chronic inflammatory pain and tumor growth is assumed. Conclusions: A nonlinear relationship between the processes caused by chronic pain, oncogenesis, and individual characteristics of systemic regulation was shown. The question about the mechanisms of adaptation to the malignant process, promoting the body's viability in large tumors in conditions of chronic neurogenic and inflammatory pain is of interest.

e16697 Background: High mortality rates in patients with pancreatic tumors and especially with neuroendocrine tumors necessitate the study of their molecular features. Our purpose was the comparative analysis of the intensity of lipid peroxidation and functioning of the enzymatic unit of the antioxidant system in the blood of patients with pancreatic tumors depending on the presence of a neuroendocrine component in them. Methods: The study included 42 patients with pancreatic cancer before the treatment and 22 donors. Parameters of lipid peroxidation (LPO) - the content of malondialdehyde (MDA) and diene conjugates, the activity of superoxide dismutase (SOD) and catalase were measured in the plasma and red blood cells, and the content of SOD1, SOD3 and catalase were measured in the blood plasma by ELISA. The results were analyzed depending on the histotype of removed tumors: pancreatic neuroendocrine tumors (n = 12), pancreatic adenocarcinoma (n = 9), pancreatic adenocarcinoma with a neuroendocrine component from 1% to 10-30% (n = 21). Results: The most pronounced changes in the blood oxidative status relative to the group of donors were observed in neuroendocrine cancer: an increase in plasma MDA levels by 30% (p = 0.006), diene conjugates in plasma and red blood cells by 2.6 and 2.9 times (p≤0.0003), a decrease in SOD and catalase activity in red blood cells by 39.7%, and 46.5% (p = 0.000000), reduced (by 20%, p = 0.047) plasma catalase activity and the minimal ratio of activity to content of both antioxidant enzymes (37.2% and 54.8% of donor levels, p < 0.0002). Conclusions: The LPO intensification most typical of neuroendocrine cancer is promoted by more significant inhibition of enzymes of the first-line antioxidant defense in red blood cells and decreased plasma catalase activity, compared to the values in adenocarcinoma.

e13088 Background: The development of breast cancer (BC) and ovarian cancer (OC), as well as their chemoresistance may be due to the presence of mutations in many genes, but most often in BRCA1/2 and CHEK2. Early diagnosis and subsequent preventive interventions improve the survival of patients with mutations in the BRCA1/2 genes. Genotyping of the gene inactivation events predicts a high sensitivity of BC and OC to platinum-containing cytostatics. Our purpose was to study the spectrum of germline mutations in the BRCA1/2 and CHEK2 genes in BC and/or OC patients in the South of Russia. Methods: The study included 622 patients of 10 nationalities with BC and/or OC from the South of Russia. Genomic DNA was isolated from peripheral blood leukocytes; mutations were detected by Real-Time PCR in the BRCA1 (185delAG, 300T > C, 2080delA, 4154delA, 5382insC, 3819delGTAAA, 3875delGTCT), BRCA2 (6174delT), CHEK2 (1100delC, IVS2+1G > A, 470T > C) genes. Results: Mutations in the BRCA1/2 gene were found in 13.5% of cases, in CHEK2 – 6.2%. In the BRCA1/2 gene, the frequency of 5382insC mutation was 76.2%; 4153 delA – 9.5%; 300T > G – 7.1%; 2080delA – 3.6%; 185delAG, 3875delGTCT, 6174delT - 1.2% each. In the CHEK2 gene: 470Т > C – 73.1%; IVS2+1G > A – 23.1%; 1100delC – 3.8%. Two patients showed a combination of 5382insC mutation in the BRCA1 gene and 470T > C mutation in the CHEK2 gene. The observed prevalence of mutations in BRCA1 was generally consistent with that for European countries (p = 0.062). Identification of 470T > C mutation in CHEK2 in two cases confirmed the final diagnosis of Li-Fraumeni syndrome (OMIM: 609265); however, we failed to establish a family history of cancer in one patient. A high-risk cancer group was formed for prophylactic purposes based on the data of genotyping for major mutations in the BRCA1/2 and CHEK2 genes in patients with BC/OC and their healthy relatives. Conclusions: The frequency of BRCA1/2 and CHEK2 mutations in the studied multinational population of Caucasian patients with BC and/or OC was 17.2%. The 5382insC BRCA1/2 (76.2%) and 470Т > C CHEK2 (73.1%) mutations were the most frequent which corresponded to the occurrence of these SNPs in populations of European countries.

Aim The aim hereof is to study the glutathione system peculiarities in the cardiac mitochondria in experimental animals against the background of a tumor process, chronic pain and the combined effect both of chronic pain and the tumor process. Materials and methods The experiment has been carried out in female mice of the C57BL / 6 line (n = 28), aged 8 weeks, with an initial body mass of 21-22 g. The animals have been divided into the following groups: an intact group as the reference (n = 7), a test group with a reproduction of the chronic pain model (n = 7), a comparison test group (B16/F10), which covered the animals with standard subcutaneously inoculated melanoma B16/F10 (n = 7), and the main experimental group (chronic pain + B16/F10) of mice with B16/F10 melanoma inoculated 3 weeks after the chronic pain model development (n = 7). All rodents have been decapitated with a guillotine upon completion of 3 weeks of the experiment (on 21 day of the experiment). After decapitation, the animal hearts have been quickly removed with the use of coolants, and mitochondria have been isolated. In the obtained mitochondrial samples, using standard IFA test systems, the following levels have been determined: reduced glutathione (GSH) in nmol/L , oxidized glutathione (GSSG) in nmol/L, glutathione peroxidase-1 (GPx-1) in ng/mL, glutathione peroxidase-4 (GPx-4) in ng/mL, glutathione reductase (GR) in ng/ mL, glutathione -S- transferase (GST) in ng/mL and superoxide dismutase-2 (SOD-2) in pg/mL. The statistics data have been calculated using the Statistica 6.0 software. Results In case of a chronic pain and a tumor process, in the cardiac mitochondria a low concentration of GPx-1, SOD-2 and a high level of GSSG are detected. The amount of GR increases in case of chronic pain and decreases under a malignant process. The combined effect of a chronic pain and a malignant process leads to a decrease in GSH against the background of an elevated GSSG level. The enzymatic component of the glutathione system (GPx-1, GR, GST) in the cardiac mitochondria in case of the combined actions and effects is characterized by a cumulative potential. The GSH/GSSG ratio in all experimental groups is statistically significantly lower than the intact values. Conclusion According to the results from this study, we may conclude that chronic pain and a tumor process exert systemically an effect on the animal's organism, but in doing so they affect different members of the regulation in the LPO-AOP system. A disorder in the glutathione system performance triggers an activation of molecular oxygen and a change in the redox potential that in its turn affects the subcellular level of the regulation and, in general, the entire cellular metabolism. The revealed abnormalities in the energy system of the cardiac mitochondria can be considered as stressor disorders. The combined effect of a chronic pain and a malignant process is treated by the organism as a double stress with an imbalance in the antioxidant glutathione system.

Aim The aim hereof is a comprehensive study of the mental, adaptive and hormonal status in patients with lung cancer with varying prevalence of the malignant process. The obtained results are compared with data on the psychosomatic state and subclinical disorders in the thyroid system in patients with cardiovascular diseases. Materials and methods The characteristics of general non-specific adaptation reactions by the organism (AR), the level of personal and situational anxiety, the severity of depression, and the reactions to the disease are studied in 28 lung cancer patients with resectable and unresectable tumors. The “Antistress” expert software is used for the qualitative and quantitative evaluation of AR. The level of the adrenocortical and thyroid axis hormones in blood serum is studied in patients with resectable tumors. Results The relationship between the AR characteristics and the malignant process prevalence, a decrease in the ACTH concentration, an elevated level of cortisol and thyroxine are noted. The mental status has been characterized by non-depressiveness and dominance of the euphoric reaction to the disease. In patients with resectable tumors without detected metastases, who have shown the euphoric reaction, the maximum scores of AR are observed with a higher thyroxine concentration as compared with those in patients with metastases and low adaptation status, who have demonstrated the hypochondriacal type of the response. Conclusion The relationship between the characteristics of AR in patients with lung cancer and the cancer process prevalence is shown herein. It is assumed that thyroxine is involved in the formation of the positive psychoemotional states in the studied patients. The combination of the euphoric reaction with the maximum AR scores in patients with resectable lung cancer without detected metastases indicates that there is a psychophysiological mechanism of restraining tumor development; at the same time, it demonstrates that there is the need to refine the concept of the euphoric type of the response. The obtained results indirectly count in favor of the cardiovascular diseases therapy approaches, which involve the use of thyroxin drugs in accordance with the algorithms of activation therapy.

The mechanism of local effects of broadband stochastically organized electromagnetic radiation (BBSO EMR) on formalin-induced edema accompanied by pain was studied at the level of integral adaptational reactions (AR). Leukogram evaluation criteria by Garkavi-Kvakina-Ukolova were used to identify types of AR and their tension (reactivity levels). Significant differences were found between the adaptation body response formed in formalin-induced edema by the type of unproductive reaction of over-activation (OA) and an integral mechanism of the bioadaptive effect of BBSO EMR - the development of stable AR of the calm (CA) and elevated activation (EA) at high reactivity levels.

The paper presents a brief review on how some concepts on the organism general adaptational reactions developed, including their role in increasing the unspecific resistance in the organism. Demonstrated is periodicity of the patterns of anti-stressor type reactions (training, calm and elevated activation) and stress in a wide range of stimulation values. The concepts on the organism adaptational reactions and reactivity levels have created the theoretical basis for practical applications of the activation therapy aimed at health improvement, prevention and treatment of various diseases, including oncology. Evidence data on the applications of the activation therapy as a promising approach to the organism state control are confirmed by the originators of this scientific discovery, their followers and medical experts.

The paper reveals logical relationships between events, facts and results of research studies and exemplary materials of practical applications of the adaptational reactions. It is demonstrated that the stress reaction by H. Selye and the anti-stressor type adaptational reactions by L. Kh. Garkavi, E.B. Kvakina and M.A. Ukolova have become the cornerstone in development of scientifically grounded approaches to control of the organism states.

Knowledge of laws of the different organism reactions to external and internal influences in case both of general unspecific adaptational reactions of different types and the periodic system of the repetitive pattern reactions offers us a possibility to target and control the organism resistance level using the activation therapy methods. Given are some examples of using the available biologically active means and the programmed modes of their dosing which demonstrate simplicity and efficacy of the stable health state restoration and maintenance.

This paper presents a brief overview of the results of original research of biological responses to the effect of low doses of substances exhibiting cytostatic and regulatory effects, as well as to weak electromagnetic radiation of different frequency ranges. The possibility of obtaining pronounced antitumor, antistress, and geroprotective effects was shown. The relation between the systemic mechanisms of implementation of these effects and structural rearrangements in biological fluids, as well as areas that seem promising for optimization of the therapeutic properties of informational effect, are discussed.

e14683 Background: The rates of physical and chemical interactions are associated with water isotopic composition. The ability of deuterium-depleted water (DDW) to influence free radical, metabolic and regulatory processes determines the need to change the drinking diet at aging, tumor growth antitumor chemotherapy (CT). Our purpose was to study the influence of DDW on the state of aging female rats, tumor growth and the CT effectiveness. Methods: The study included 37 aging outbred white female rats (250-280 g) with transplanted sarcoma 45 (S-45) receiving chemotherapy with cyclophosphamide (CF). DDW intake (46±2 ppm, 25-30 mL a day) was started 5 weeks before S-45 transplantation. CF was administered intraperitoneally on day 10 at a dose of 50 mg/kg twice with a 5-day interval. We determined tumor volume by Shrek, deuterium concentrations in the blood plasma (NMR spectrometer JEOL JNM-ECA 400MHz) and inner organs (mass spectrometer DELTAplus H/Device, Finnigan, Germany), estrous cycle stages and types of the nonspecific adaptational reactions of organism by Garkavi-Kvakina-Ukolova (1975). Results: DDW administration before the tumor transplantation significantly balanced the stages of the estrous cycle and increased the incidences of the antistress adaptational reactions development by 1.6 times (p < 0.05); external signs of rejuvenation were observed (changes in hair state and motor activity). After S-45 transplantation, its growth prior to CT was suppressed (tumor volume 0.25±0.07 cm3 vs. 0.47±0.10 cm3 in controls, p < 0.1). DDW increased the effect of CF by 4.8 times by day 15 (0.88±0.67 cm3 vs. 4.22±1.43 cm3 with CF alone, p < 0.05). Concentrations of deuterium in the blood plasma decreased by 33%, in the inner organs (liver, kidney and heart) – by 6-14% (p < 0.05). Conclusions: DDW has an antistress, geroprotective , antitumor and CT-optimizing effect.

e21035 Background: Lack of prognostic markers of melanoma to assess its progression, metastasis (MTS), and treatment effectiveness, determines the feasibility of studying biophysical aspects of the transmission of events from cellular to extracellular environment. Receipt and circulation of pathological proteins of melanoma (M) in the blood carries important information, and its trap is the phase transition of biofluids into a solid state. The purpose of the study was to determine markers of melanoma at the supramolecular level in solid-state samples of the blood serum (BS). Methods: 240 samples of BS obtained prior to the surgery from 60 patients with M were studied. The preparations were prepared by the Shatokhina-Shabalin method by creating a closed "analytic cell": dehydration of a BS drop (10 mcL) under a cover glass at 20–240С, humidity 65–70%, for 72 hours. The folding of M proteins was assessed by a polarized light microscopy (Leica DM LS2 microscope, x400-640) using photo and video control and the Morphotest program. Results: The initial stages of M development were characterized by the early appearance of single combined melanoform crystals — anisotropic microspherolites (Mi) embedded in the center of a large anisomorphone (lAM). As M progressed, Mi aggregation with basic lAMs was enhanced. Anisotropy of Mi increased, revealing clear sectors filled with melanin. In the later stages of M growth, melanoform aggregations dominated with point, filamentous, and chained melanin structures. The manifestation of MTS was accompanied by the destruction of the aggregations and the filling of the analytic cell space with numerous Mi. Conclusions: Correspondence of crystallogenesis of M melanomorphic markers in BS in tumor progression allows using the studied supramolecular factors to predict the development of melanoma.

e14179 Background: Previously, regression of the developed transplanted tumors under the influence of magnetite nanoparticles (NPs) as monofactor was shown. The purpose of the study was to reveal signs of intercellular interactions in the zone of tumors regressed under the influence of magnetite NPs. Methods: Experiments were carried out on 14 white outbred male rats (180-200 g) with transplanted sarcoma 45 of an initial volume of 0.7-1.3 cm3. After 6 peritumoral injections of magnetite NPs (10 ± 2 nm) in the form of magnetic fluid AM-01 ("AM-Kub", Ekaterinburg) twice a week in a single dose of 17.7 mg/kg, 5 out of 7 rats of the main group demonstrated a regression of the tumor from 1.5-3 cm3 to 0.7 ± 0.2 cm3 (in the control group – 9.7 ± 1.9 cm3, p < 0.01). Ultrathin sections of the tumor tissue of 12 animals (including 7 rats of the main group) were examined in electron microscope JEOL JEM-1011 (Japan). Results: Under effective action of magnetite NPs in tumor tissue, in addition to necrosis, apoptosis and autophagy were marked. In the case of apoptosis, chromatin condensation in compact masses and separation of apoptotic bodies were observed. Significantly more common autophagic cell death was identified by violation of the integrity of the cell membrane and the presence of autophagosomes. Numerous signs of activation of cell-cell interactions with participation of immune cells were observed. Diverse groups of 2-4 contacting cells included macrophages, lymphocytes, plasmocytes, degranulated mast cells, neutrophils in various combinations with distinct signs of the metabolic activity. Invagination of macrophages cytoplasm into the cytoplasm of tumor cells, close adjacency of the cytoplasmic membranes of neutrophils and lymphocytes at considerable length, tight intertwining of mast cell cytoplasmic processes and engulfing of the mast cell granules by macrophages were noted. Conclusions: The results supplement the characteristics of immune processes during the self-dependent antitumor action of magnetite NPs.

e13511 Background: Development of alternative methods of non-invasive therapy in neurooncology is determined by the need to prevent the continued growth of glioblastoma and radiation-induced complications. Usage of electromagnetic influence for these reasons is determined by the oscillatory nature of intracellular processes and results of previous studies on the effect of weak electromagnetic radiation in tumor growth. The purpose of the study was to reveal the effect of a pulsed magnetic field (PMF) and ionizing radiation (IR) on human T98G glioblastoma cells. Methods: Human T98G glioblastoma cells were cultured in the RPMI-1640 medium (Biolot, Russia) supplemented with 10% fetal bovine serum (Thermo Scientific HyClone, USA). The culture was seeded in the Biofil plates (at least 8x105 cells per 3 mL of the medium), incubated (CB 150 Binder, Germany) and exposed to IR 10 Gy (TheratronEquinox-BestTheratronics) for 11.7 min. and/or PMF 15 mT or 300 mT with a sequence of frequencies 0.3-3.0-9.0 Hz for 7 min. (Neiro-MS/D by Neirosoft, Russia). The efficiency criteria were lethality and the mitotic activity of human T98G glioblastoma cells. Results: The highest cellular lethality was registered 3 hours (18.7 vs. 5.2% in controls, p≤0.05) and a day after IR exposure. PMF 15 or 300 mT alone increased lethality by 2.5-2.8 times compared to controls (p < 0.05). IR plus PMF (15 mT) did not caused increased lethality. At the same time, the most pronounced decrease in the mitotic activity a day after exposure (by 4.7 times, p < 0.05) was registered in PMF alone (15 mT). Conclusions: The inhibitory effect of PMF on the viability of human glioblastoma cells indicates the prospects for its use as an accompanying treatment in neurooncology.

e15619 Background: Associations between the psychosomatic state of cancer patients and the disease course, treatment and complications are poorly studied. Our purpose was to study adaptation and mental statuses of patients with biliopancreatoduodenal cancer (BPDC) at different disease stages with various volumes of surgical intervention. Methods: Parameters of the psychosomatic state of 32 patients with BPDC were studied before treatment, 1 day after surgery and at the dismissal from hospital. Adaptation statuses were assessed by a character and intensity of the general nonspecific adaptation reactions (AR) with scoring by the Antistress program. Mental statuses were evaluated by the levels of personal (PA) and situational anxiety (SA) by the Spielberger-Khanin method. Patients with pancreatoduodenectomy (PDE, 19 patients) and those with palliative intervention for a bypass anastomosis (BA, 13 patients) were compared. Patients with stage II disease prevailed in the PDE group and stage III-IV – in the BA group. Results: Patients with PDE had a higher adaptation status than patients with BA (p < 0.05). It resulted in lower rates of most adverse AR prior to the surgery (21% vs. 61%, respectively) and a higher AR score 1 day after the surgery (187±39 and 80±24, respectively). The AR score of 18 and lower 1 day after PDE was associated with postoperative complications. A tendency to a higher number of patients with a high PA was found in the PDE group compared to the BA group (58 and 31%, respectively) which could affect the time of pathology detection in patients of different groups. Patients of the two groups showed differences in the SA dynamics determined by the volume of surgical intervention. Conclusions: The association between the psychosomatic state of BPDC patients and the disease stage and volume of surgical intervention was demonstrated. The AR scoring 1 day after the surgery allows prognosis of postoperative complications.
