Abstract

A study was conducted to develop a dog genome map to be utilized for linkage studies and wide genome search and comparison. Dog fibroblasts were fused after gamma irradiation with thymidine kinase-deficient hamster cells. The dog fibroblasts were irradiated with 5,000 rad. Cell lines were then determined and prepared in roller bottles. In addition, individual clones were picked and prepared at 37 degrees C in microtiter plates.

Abstract

Human T-cell leukemia virus type I (HTLV I) does not cause leukemia in all persons who are infected. Indeed, the majority of infected individuals do not have any symptoms. However, a few infected persons, although asymptomatic, may be found upon examination to have an atypical T-lymphocytosis which may either spontaneously remit or may progress to an aggressive adult T-cell leukemia or lymphoma. The recent case of a 38-year-old black woman illustrates some of the features which may complicate the diagnosis of HTLV-related T-cell leukemia. The patient lived in French Guyana, an endemic region for HTLV I infection. She was initially found to have abnormalities of the lymph nodes of the neck and back, and biopsy revealed cells which on first examination appeared to be Hodgkin's lymphoma. The patient did not have antibodies to HTLV I. She was placed on chemotherapy, but the disease progressed and the patient died three months after diagnosis. During the chemotherapy for Hodgkin's disease, the patient developed antibodies against HTLV I, which prompted a reexamination of the biopsy material. With the help of hindsight, some convoluted cells were observed which were determined to be pleiomorphic T-cell lymphoma cells. Material extracted from the biopsy samples was shown to contain genomic DNA of the HTLV I, which made it clear that the virus was present from the outset and that the patient was already infected prior to the chemotherapy. The exact relationship between the appearance of HTLV I antibodies and the chemotherapeutic treatment is not clear, but two possibilities stand out. The immunosuppressive features of chemotherapy may have permitted the unopposed replication of some infected subsets of cells. Also possible is that the chemotherapy may have activated an infection with some cells which had been only latent up to that point. Other retroviruses, including the human immunodeficiency virus (HIV), may exist only as latent genes which are not expressed until activated by some chemical or biological event. Abstract

Background: Advantages offered by canine population substructure, combined with clinical presentations similar to human disorders, makes the dog an attractive system for studies of cancer genetics. Cancers that have been difficult to study in human families or populations are of particular interest. Histiocytic sarcoma is a rare and poorly understood neoplasm in humans that occurs in 15% to 25% of Bernese Mountain Dogs (BMD). Methods: Genomic DNA was collected from affected and unaffected BMD in North America and Europe. Both independent and combined genome-wide association studies (GWAS) were used to identify cancer-associated loci. Fine mapping and sequencing narrowed the primary locus to a single gene region. Results: Both populations shared the same primary locus, which features a single haplotype spanning MTAP and part of CDKN2A and is present in 96% of affected BMD. The haplotype is within the region homologous to human chromosome 9p21, which has been implicated in several types of cancer. Conclusions: We present the first GWAS for histiocytic sarcoma in any species. The data identify an associated haplotype in the highly cited tumor suppressor locus near CDKN2A. These data show the power of studying distinctive malignancies in highly predisposed dog breeds. Impact: Here, we establish a naturally occurring model of cancer susceptibility due to CDKN2 dysregulation, thus providing insight about this cancer-associated, complex, and poorly understood genomic region. Abstract

The living water jewels have an attractive colored for example a body color, morphology, mode of taking food and have a peculiar characteristic. A number of aquarium fishes the Pterois sp is known to aquarists, while it is easy to breed and very important and interesting tasks of an aquarium keeper is to feed management. Food is playing an important role in maintaining health and preventing diseases. Providing exact diet is essential for fish growth and other activities. Overfeeding or uneaten food contributes to a deteriorating water quality by polluting the water. Some fishes in wild or culturing system are not taken regularly same food and it will be varying due to the inhabitants. The red firefish P. volitans is one of the remarkable fish in marine waters. In the present study, the food and feeding habits of lionfish P.volitans, male and female fishes were observed and due to fishes gut contents were analysis. The different food items were recorded from the guts of P. volitans during the study period. Generally, the food items found in the examined gut contents were grouped into eleven different categories. The male fish of P. volitans stomach contents as identified feed compositions in the following order: viz., Crustaceans >Fish >Zooplankton >Phytoplankton >Miscellaneous >Bivalves >Polychaetes >Gastropods >sand >Nematodes >Digested matter and the female fish of P. volitans stomach contents as identified feed compositions in the following order viz., Crustaceans >Fish >Zooplankton >Phytoplankton >Miscellaneous >Bivalves >Polychaetes >Gastropods >Sand >Nematodes >Digested matter, respectively. The present investigation very much useful for this species to be taken up for aquariums in home or office, hotel, hospital, intensive, semi intensive and mass scale culture practices in anywhere in the world.

Abstract

The extraordinary phenotypic diversity of dog breeds has been sculpted by a unique population history accompanied by selection for novel and desirable traits. Here we perform a comprehensive analysis using multiple test statistics to identify regions under selection in 509 dogs from 46 diverse breeds using a newly developed high-density genotyping array consisting of >170,000 evenly spaced SNPs. We first identify 44 genomic regions exhibiting extreme differentiation across multiple breeds. Genetic variation in these regions correlates with variation in several phenotypic traits that vary between breeds, and we identify novel associations with both morphological and behavioral traits. We next scan the genome for signatures of selective sweeps in single breeds, characterized by long regions of reduced heterozygosity and fixation of extended haplotypes. These scans identify hundreds of regions, including 22 blocks of homozygosity longer than one megabase in certain breeds. Candidate selection loci are strongly enriched for developmental genes. We chose one highly differentiated region, associated with body size and ear morphology, and characterized it using high-throughput sequencing to provide a list of variants that may directly affect these traits. This study provides a catalogue of genomic regions showing extreme reduction in genetic variation or population differentiation in dogs, including many linked to phenotypic variation. The many blocks of reduced haplotype diversity observed across the genome in dog breeds are the result of both selection and genetic drift, but extended blocks of homozygosity on a megabase scale appear to be best explained by selection. Further elucidation of the variants under selection will help to uncover the genetic basis of complex traits and disease.

Abstract

The selective breeding of fish for aquaculture purposes requires the understanding of the genetic basis of traits such as growth, behaviour, resistance to pathogens and sex determinism. Access to well-developed genomic resources is a prerequisite to improve the knowledge of these traits. Having this aim in mind, a radiation hybrid (RH) panel of European sea bass (Dicentrarchus labrax) was constructed from splenocytes irradiated at 3000 rad, allowing the construction of a 1581 marker RH map. A total of 1440 gene markers providing ~ 4400 anchors with the genomes of three-spined stickleback, medaka, pufferfish and zebrafish, helped establish synteny relationships with these model species. The identification of Conserved Segments Ordered (CSO) between sea bass and model species allows the anticipation of the position of any sea bass gene from its location in model genomes. Synteny relationships between sea bass and gilthead seabream were addressed by mapping 37 orthologous markers. The sea bass genetic linkage map was integrated in the RH map through the mapping of 141 microsatellites. We are thus able to present the first complete gene map of sea bass. It will facilitate linkage studies and the identification of candidate genes and Quantitative Trait Loci (QTL). The RH map further positions sea bass as a genetic and evolutionary model of Perciformes and supports their ongoing aquaculture expansion.

Abstract

The BASSMAP consortium, funded by the European Union, has been set up to improve the understanding of the genome of European sea bass (Dicentrarchus labrax). The aim is to locate genes of known function on the physical map and to compare specific regions among perciforms. We produced an F1 cross of outbred sea bass as mapping panel and a Bacterial Artificial Chromosome library (7× redundancy and 165 kb average insert size). We use a panel of 21 wild sea bass to test for marker polymorphisms. All family genotypes are submitted to a database for pedigrees, markers and phenotypes (www.resspecies.org). These data will construct a first generation map including 162 microsatellite markers and 25 linkage groups (2n=24). A new second generation linkage map including 369 polymorphic microsatellite and AFLP markers, covering 34 linkage groups, will be disseminated via the mapping database (www.thearkdb.org). A set of 14 tissue specific cDNA banks have been constructed; 28,000 Expressed Sequence Tags (ESTs) have been sequenced in a data base (CeBiTec, U. Bielefeld). The sequences are annotated, screened for variation and mapped.
