There has been continuous progress in diabetes management over the last few decades, not least due to the widespread dissemination of continuous glucose monitoring (CGM) and automated insulin delivery systems. These technological advances have radically changed the daily lives of people living with diabetes, improving the quality of life of both children and their families. Despite this, hypoglycemia remains the primary side-effect of insulin therapy. Based on a systematic review of the available scientific evidence, this paper aims to provide evidence-based recommendations for recognizing, risk stratifying, treating, and managing patients with hypoglycemia. The objective of these recommendations is to unify the behavior of pediatric diabetologists with respect to the timely recognition and prevention of hypoglycemic episodes and the correct treatment of hypoglycemia, especially in patients using CGM or advanced hybrid closed-loop systems. All authors have long experience in the specialty and are members of the Italian Society of Pediatric Endocrinology and Diabetology. The goal of treating hypoglycemia is to raise blood glucose above 70 mg/dL (3.9 mmol/L) and to prevent further decreases. Abstract Introducing new grass species into cultivation has long been proposed as beneficial to increase the sustainability and diversity of productive systems. However, wild species with potential tend to show high seed dormancy, causing slow, poor, and unsynchronized seedling emergence. Meanwhile, domesticated species, such as cereals, show lower seed dormancy, facilitating their successful establishment. In this work, we conduct a review of phenotypic variation on seed dormancy and its genetic and molecular basis. This quantitative and highly heritable trait shows phenotype plasticity which is modulated by environmental factors. The level of dormancy depends on the expression of genes associated with the metabolism and sensitivity to the hormones abscisic acid (ABA) and gibberellins (GA), along with other dormancy-specific genes. The genetic regulation of these traits is highly conserved across species. The low seed dormancy observed in cereals and some temperate forages was mostly unconsciously selected during various domestication processes. Emphasis is placed on selecting materials with low seed dormancy for warm-season forage grasses to improve their establishment and adoption. Finally, we review advances in the domestication of dallisgrass, where seed dormancy was considered a focus trait throughout the process.

Se presenta un caso clínico poco frecuente de un quiste óseo aneurismático localizado en el cuboides de un niño de 13 años. Se llegó al diagnóstico mediante la tríada de síntomas, estudios por imágenes (radiografía, resonancia magnética) y anatomía patológica. El tratamiento consistió en el abordaje y curetaje minuciosos dentro de la lesión, el fresado de alta velocidad, la electrocauterización y el relleno con aloinjerto óseo liofilizado con chips cortico-esponjosos. Pese al diagnóstico tardío, la evolución clínica fue favorable con remisión completa de los síntomas y retorno a las actividades deportivas, sin recidiva local.

Chondroblastoma is a rare, benign neoplasm usually located in the epiphyses and apophyses of the long bones in the immature skeleton. Radiologically, these tumors have a classic appearance of a lytic lesion with chondroid matrix surrounded by a thin sclerotic rim. Here, we describe the case of a 5-year-old male who presented with a chondroblastoma unusually located exclusively in the metaphyseal region, which led to an elusive diagnosis. The presence of tumors outlying the traditional location or epidemiological spectrum, along with the potential for histopathological misdiagnosis, can pose a diagnostic and therapeutic challenge for the treating team.

Myositis ossificans (MO) is a benign disorder where bone forms within muscles or other soft tissues. This condition usually follows trauma and is rare in pediatric patients. Here we present the case of a 2-year-old male who developed MO of his right elbow without obvious trauma to the area. Imaging of MO in the initial phase is highly unspecific and obtaining tissue samples through a biopsy can render misleading reports. In most cases MO is a self-limited process with complete resolution, however, some cases may present a diagnostic and therapeutic challenge.

Agricultural soil environments contain different types of nematodes in all trophic levels that aid in balancing the soil food web. Beneficial free-living nematodes (FLNs) consist of bacterivores, fungivores, predators, and omnivores that help in the mineralization of the soil and the top-down control of harmful plant-parasitic nematodes (PPNs). Annually, USD 125 billion in worldwide crop losses are caused by PPNs, making them a plant pathogen of great concern for growers. Farmers have started to implement the use of cover crops in agricultural systems for the protection and enrichment of soil but research on how different cover crops affect nematode populations is lacking and in demand. This study aims to determine the effects of legume and grass cover crops, Cowpea (Vigna unguiculata) and Pearl Millet (Pennisetum glaucum), as well as their mixture on the abundance and diversity of FLN and PPN populations. Soil samples were collected at the time of cover crop maturity and spinach harvest to analyze nematode communities using both morphological and DNA metabarcoding analysis. The results showed that the application of Cowpea and Pearl Millet as well as their mixture in a spinach agricultural system led to the control of PPNs and proliferation of FLN communities, with each cover crop treatment demonstrating different advantages for the various nematode feeding groups. Soil property analysis did not show a significant difference except for magnesium and total nitrogen levels, which were significantly correlated with nematode community composition. The overall findings of our study indicate that the choice of cover crop implementation by growers for spinach cultivation should be based on specific soil health conditions, which in turn promote soil fertility and a healthy nematode community. Crop wild relatives are gaining increasing attention. Their use in plant breeding is essential to broaden the genetic basis of crops as well as to meet industrial demands, for global food security and sustainable production. Solanum malmeanum (Solanum sect. Petota, Solanaceae) is a wild relative of potatoes (S. tuberosum) from Southern South America, occurring in Argentina, Brazil, Paraguay and Uruguay. This wild potato has been largely mistaken for or historically considered as conspecific with S. commersonii. Recently, it was reinstated at the species level. Retrieving information on its traits and applied uses is challenging, because the species name has not always been applied correctly and also because species circumscriptions and morphological criteria applied to recognize it have not been consistent. To overcome these difficulties, we performed a thorough literature reference survey, herbaria specimens’ identification revision and genebank database queries to review and update the information available on this potato wild relative, contributing to an increase in research on it to fully understand and explore its potential for potato breeding. Scarce studies have been carried out concerning its reproductive biology, resistance against pests and diseases as well as tolerance to abiotic stresses and evaluation of quality traits. The scattered information available makes it less represented in genebanks and genetic studies are missing. We compile, update and present available information for S. malmeanum on taxonomy, geographical distribution, ecology, reproductive biology, relationship with its closest relatives, biotic and abiotic stresses resistance and quality traits and discuss ways to overcome sexual barriers of hybridization and future perspectives for its use in potato breeding. As a final remark, we highlight that this species’ potential uses have been neglected and must be unlocked. Thus, further studies on morphological and genetic variability with molecular tools are fundamental for an efficient conservation and applied use of this promising genetic resource.

El condrosarcoma forma parte de los tumores primarios malignos más frecuentes. Las localizaciones pélvicas y de raíz de muslo requerirán abordajes amplios con la consecuente dificultad de cobertura de partes blandas. La hemipelvectomía externa es habitualmente el procedimiento de elección para estas localizaciones. Implica la resección de la extremidad inferior en bloque asociada a la hemipelvis ipsilateral, dando como resultado un gran defecto de cobertura. En algunas circunstancias, la extensa resección de partes blandas hace imposible la utilización de colgajos rotacionales locales. La reconstrucción con colgajo tipo “Fillet flaps” ofrece a estos defectos de partes blandas la opción de lograr una adecuada cobertura. El objetivo del trabajo es mostrar la resolución de un caso poco convencional de condrosarcoma de fémur proximal y su reconstrucción de partes blandas con un método nunca antes utilizado en nuestro medio.

Sex-related disparities have been recognized in incidence, pathological findings, pathophysiological mechanisms, and diagnostic pathways of non-rheumatic mitral regurgitation. Furthermore, access to treatments and outcomes for surgical and interventional therapies among women and men appears to be different. Despite this, current European and US guidelines have identified common diagnostic and therapeutic pathways that do not consider patient sex in decision-making. The aim of this review is to summarize the current evidence on sex-related differences in non-rheumatic mitral regurgitation, particularly regarding incidence, imaging modalities, surgical-derived evidence, and outcomes of transcatheter edge-to-edge repair, with the goal of informing clinicians about sex-specific challenges to consider when making treatment decisions for patients with mitral regurgitation.

Abstract Background Benign acute childhood myositis (BACM) is a self-limited syndrome associated with viral infections characterized by symmetric lower extremity pain typically affecting school-aged children. Evolution in rhabdomyolysis and kidney damage is rarely reported. Despite this, the acute presentation commonly concerns both parents and health care providers, often leading to unnecessary workup. The aim of the study was to determine the features and outcome of a large series of children with BACM identifying a management pathway for pediatricians in Emergency Department (ED). Methods We conducted a retrospective study of patients with BACM managed in 2 Italian pediatric ED during a period of 8 and a half years. Demographic data, clinical, and laboratory results were extracted from electronic medical records. Recurrence, complications, treatments, and outcomes were also recorded. Descriptive statistics were produced for first-episode patients and for those with recurrence of myositis. A comparison between groups was performed. Results One hundred and thirteen patients with BACM were identified. Ninety-two children (65 males) had a single episode, while ten (nine males) had recurrence. The mean age at presentation was 6.0 years (range 2–13,2). All patients had normal neurological examination and no one developed myoglobinuria, or renal failure. AimsDiabetic ketoacidosis is the most severe metabolic derangement due to prolonged insulin deficiency as in type 1 diabetes. Diabetic ketoacidosis, a life-threatening condition, is often diagnosed late. A timely diagnosis is mandatory to prevent its consequences, mainly neurological. The COVID-19 pandemic and lockdown have reduced the availability of medical care and access to hospitals. The aim of our retrospective study was to compare the frequency of ketoacidosis at the diagnosis of type 1 diabetes between the lockdown-post lockdown period and the previous two calendar years, in order to evaluate the impact of the COVID-19 pandemic.Patients and MethodsWe retrospectively assessed the clinical and metabolic data at the diagnosis of type 1 diabetes in children in the Liguria Region during 3 different time periods: calendar year 2018 (Period A), calendar year 2019 until February 23,2020 (Period B) and from February 24, 2020 onwards to March 31, 2021 (Period C).ResultsWe analyzed 99 patients with newly-diagnosed T1DM from 01/01/2018 to 31/03/2021. Briefly, a younger age at diagnosis of T1DM was observed in Period 2 compared to Period 1 (p = 0.03). The frequency of DKA at clinical onset of T1DM was similar in Period A (32.3%) and Period B (37.5%), while it significantly increased in Period C (61.1%) compared to Period B (37.5%) (p = 0.03). PH values were similar in Period A (7.29 ± 0.14) and Period B (7.27 ± 0.17), while they were significantly lower in Period C (7.21 ± 0.17) compared to Period B (p = 0.04).ConclusionsAn increase in the frequency of diabetic ketoacidosis has been documented in newly diagnosed pediatric patients in the Liguria Region during and after the lockdown period compared to previous calendar years. This increase could have been caused by the delay in diagnosis following the restrictions imposed by the lockdown with consequently reduced access to health care facilities. More information on the risks of ketoacidosis is desirable by means of social and medical awareness campaigns.

Este estudio comprende el análisis de los depósitos limolíticos basales de la For- mación Lumbrera superior (unidad cuspidal del Grupo Salta), de edad Eoceno medio, los cuales se encuentran ubicados en la localidad de El Simbolar y Río Juramento, provincia de Salta. Los resultados de los análisis petrográficos, macro y microfaciales, granulométricos y mineralógicos se exponen en este trabajo con la finalidad de discutir el posible origen eólico y el escenario paleogeográfico que pudo haber contribuido a su formación. La Formación Lumbrera superior está formada por espesas sucesiones de material mayormente limolítico de intenso color rojizo, de distribución mantiforme, y groseramente estratificado en la cual se reconocieron cinco litofacies sedimentarias: Limolita bioturbada, Limolita calcárea, Arenisca fina blanca, Heterolítica y Ceniza volcánica; con evidencias de desarrollo de rasgos pedogenéticos en todas las litofacies. Las características sedimentarias sugieren un paleoambiente de llanuras vegetadas con áreas topográficamente más bajas que quedaban temporalmente inundadas. Los estudios granulométricos y mineralógicos de la fracción fina muestran una participación de más del 50% de la fracción limo y un patrón de distribución de tamaño de grano predominantemente bimodal, donde la moda principal se encuentra en el rango de limo fino (7,8 a 15,6 µm) en la mayoría de las muestras analizadas y la moda secundaria es inferior a la fracción arena muy fina (63 a 125 µm). La mediana de este conjunto de muestras está representada por la fracción limo medio (15,6 a 31 µm) y limo fino (7,8 a 15,6 µm). La distribución granulométrica de estas muestras es similar a la mostrada por el loess pampeano cuaternario y el polvo atmosférico actual. Asimismo, la mineralogía de la Formación Lumbrera es homogénea y no presenta cambios sustanciales a lo largo de la columna estratigráfica analizada, reconociéndose dos grupos de minerales: componentes detríticos (cuarzo, plagioclasa, illita/moscovita) y componentes auti- génicos (calcita, analcima, hematita). Las características sedimentológicas permiten reconocer a estos depósitos como paleoloess, siendo una posible fuente del detrito el inicio del levantamiento, en forma contemporánea, de la Cordillera de los Andes. Además se atribuye como fuente secundaria del material, a la actividad del arco volcánico durante el Eoceno - Oligoceno temprano en la región oriental de Chile (20°S - 25°S).

A lo largo del presente trabajo se recupera parte de la experiencia transitada durante los años 2018 y 2019 por el Centro de Investigaciones en Seguridad, Estado y Sociedad (CISES), en el marco del Proyecto de Extensión denominado “Seguridad y Derechos Humanos”. Este último tenía como objetivos específicos: 1) analizar la información proveniente de fuentes judiciales y periodísticas referidas a diferentes casos de abusos policiales en la Provincia de Santa Fe; 2) explorar la normativa y protocolos vigentes a nivel nacional e internacional que abordan el uso racional de la fuerza policial; 3) promover instancias de encuentro e intercambio entre agentes de la Policía y representantes del Ministerio de Seguridad y de la Secretaría de Derechos Humanos de la Provincia de Santa Fe. El informe procura plasmar las experiencias y resultados obtenidos durante el año de trabajo, así como los obstáculos, debates y aprendizajes que se presentaron producto del intercambio entre el personal policial y los miembros del equipo que participaron de los talleres realizados en Recreo y en Rosario, en octubre de 2019.

Next-generation genome mapping through nanochannels (Bionano optical mapping) of plant genomes brings genome assemblies to the ‘nearly-finished’ level for reliable and detailed gene annotations and assessment of structural variations. Despite the recent progress in its development, researchers face the technical challenges of obtaining sufficient high molecular weight (HMW) nuclear DNA due to cell walls which are difficult to disrupt and to the presence of cytoplasmic polyphenols and polysaccharides that co-precipitate or are covalently bound to DNA and might cause oxidation and/or affect the access of nicking enzymes to DNA, preventing downstream applications. Here we describe important improvements for obtaining HMW DNA that we tested on Solanum crops and wild relatives. The methods that we further elaborated and refined focus on • Improving flexibility of using different tissues as source materials, like fast-growing root tips and young leaves from seedlings or in vitro plantlets. • Obtaining nuclei suspensions through either lab homogenizers or by chopping. • Increasing flow sorting efficiency using DAPI (4′,6-diamidino-2-phenylindole) and PI (propidium iodide) DNA stains, with different lasers (UV or 488 nm) and sorting platforms such as the FACSAria and FACSVantage flow sorters, thus making it appropriate for more laboratories working on plant genomics.The obtained nuclei are embedded into agarose plugs for processing and isolating uncontaminated HMW DNA, which is a prerequisite for nanochannel-based next-generation optical mapping strategies.
