Climate change poses significant challenges to the grapevine cultivation for wine production. In the last years, the occurrence of extreme weather events such as intense heat waves and prolonged period of drought increased sunburn damage with negative consequences to yield and berry composition. Short-term adaptation strategies are urgently needed to mitigate these effects. In this light, our study aimed to evaluate the efficacy of zeolite application on the canopy and irrigation during ripening on sunburn damage, yield, and berry composition in cv. Sangiovese (Vitis vinifera L.). Over 3 years (2021–2023), canopy management and irrigation treatments were arranged in a strip-plot design. Canopy management treatments included leaf removal on the cluster zone, with and without zeolite application, and no defoliated control; irrigation treatments comprised irrigation from berry softening and no irrigation. Both zeolite application and irrigation reduced sunburn damage, thus mitigating yield loss. Regarding berry composition, zeolite application did not alter the main parameters whereas irrigation led to reductions in sugar and anthocyanin concentrations. These findings suggest that zeolite application and irrigation during ripening represent promising and cost-effective solutions for grape growers facing climate change-induced challenges. However, further studies are necessary to optimize timing of irrigation to avoid detrimental effects on anthocyanin accumulation.

Foliar treatments with mineral-based compounds have aroused interest as a sustainable approach to cope with high-temperature stress in vineyards, improving the colour of grapes and wines without altering the sugar concentration. Since berry-associated microorganisms are crucial for wine production, composition and stability, the present study aims to unravel the effects of kaolin and zeolite treatments on berry quality, flavonoid gene expression and epiphytic microorganisms (total fungi, Hanseniospora uvarum, Metschnikowia pulcherrima, plant-associated bacteria and lactic acid bacteria). Experiments were performed on 'Sangiovese' during ripening in two growing seasons (2019 and 2020). Both mineral treatments were able to cause the elicitation of several flavonoid-related genes and to increase the anthocyanin concentration in berries without an alteration of the final technological maturity (TSS, TA, pH) of the grapes. Interestingly, neither kaolin nor zeolites negatively affected the analysed microbial population at harvest despite transient alterations observed in the more stressful year (2019). These results support the use of foliar mineral treatments to increase anthocyanin concentration in grape berries and protect them from excessive heat without altering their native microorganisms.

Grapevines are frequently subjected to heatwaves and limited water availability during ripening. These conditions can have consequences for the physiological health of the vines. Moreover, the situation is often exacerbated by intense solar radiation, resulting in reduced yield due to sunburn and a decline in quality. In light of these challenges, our study aimed to develop a fruit-zone cooling system designed to mitigate grape sunburn damage and improve the microclimate conditions within the vineyard. The system comprises a network of proximal sensors that collect microclimate data from the vineyard and an actuator that activates nebulizers when the temperature exceeds the threshold of 35°C. The research was conducted over two years (2022 and 2023) in Bologna (Italy) using potted Sangiovese and Montepulciano vines. These two vintages were characterized by high temperatures, with varying amounts of rainfall during the test period, significantly impacting the evaporative demand, which was notably higher in 2023. Starting from the veraison stage we compared three treatments: Irrigated control vines (WW); Control vines subjected to 50% water restriction during the month of August (WS); WS vines treated with nebulized water in the bunch area during the stress period (WS+FOG). The application of nebulized water effectively reduced the temperature of both the air around the clusters and the clusters themselves. As we expected, Montepulciano showed better single leaf assimilation rate and stomatal conductance under non-limiting water conditions than Sangiovese while their behavior was unaffected under water-scarce conditions. Importantly, for the first time, we demonstrated that nebulized water positively affected gas exchange in both grape varieties. In addition to this, the vines treated with the misting system exhibited higher productivity compared to WS vines without affecting technological maturity. In the 2023 vintage, the activation of the system prevented the ripening blockage that occurred in Montepulciano under water stress. Regarding the concentration of total anthocyanins, a significant increase in color was observed in WS+FOG treatment, suggesting a predominant role of microclimate on anthocyanin biosynthesis and reduction of oxidative phenomena. In conclusion, the fruit-zone cooling system proved to be an invaluable tool for mitigating the adverse effects of multiple summer stresses.

Summary: Tandem running in ants is a sophisticated form of communication. Precise measurement of movement coordination by the tandem pair can shed light on social interactions. Here, we describe an integrative approach to obtain long movement trajectories of a specific tandem pair within a crowd of ants. We describe a maze-like arena, video recording and editing, and movement tracking. We integrate two pieces of image-based tracking software that have distinct individual assignment strategies. This protocol aids comparative studies of recruitment communication across species.For complete details on the use and execution of this protocol, please refer to Mizumoto et al. Summary: Communication is fundamental to the organization of animal societies, often resulting in the convergent evolution of similar social behavior across lineages. However, this similarity may conceal underlying functional and mechanistic differences. Here we combined network and information-theoretic analysis to quantify how tandem recruitment is distinguishable between two ant genera, Temnothorax and Diacamma. We show that Temnothorax uses tandem running to recruit additional recruiters, while Diacamma uses it principally to move the passive majority of their colony, a task that Temnothorax accomplishes with a different behavior, social carrying. Accordingly, the network structure of Diacamma tandems was dissimilar to that of Temnothorax, instead resembling the social-carrying networks in Temnothorax. Furthermore, our information-theoretical analysis on movement trajectories revealed that Diacamma tandem runs lack bidirectional information transfer, the signature of route learning in Temnothorax. By quantifying the diversity of similar communication systems, this study increases the resolution of our understanding of animal societies.

Abstract Background Nailfold videocapillaroscopy (NVC) is a feasible method that allows the observation of the microvascular changes that mark the course of systemic sclerosis (SSc). In previous studies, we demonstrated that the NEMO score, i.e. the cumulative number of microhaemorrhages and microthromboses, is a good indicator of the steady-state level and overtime changes of disease activity (DA) in SSc. Objectives To verify whether high NEMO scores, which mirror a very active microvascular derangement in the fingers, may be associated with the subsequent development of ischaemic digital ulcers (IDUs). Methods The NEMO score was assessed at baseline (T0) in 98 patients with SSc, all classified according to the ACR-EULAR criteria. Of them, 90 were females, 48 had the limited and 50 had the diffuse cutaneous variant of SSc. Afterwards, the patients were closely followed up for 2 years, and the appearance of new IDUs recorded at any time of the follow-up. The T0-NEMO score values of patients who developed IDUs were compared to those of patients who did not. A receiver operating curve (ROC) was constructed, and the area under the curve (AUC) calculated by plotting the sensitivity and 1-specificity of the different NEMO score values in predicting the subsequent development of IDUs. Results During the follow-up, 38 out of 98 patients developed one or more IDUs. Programming robot swarms is hard because system requirements are formulated at the swarm level (i.e., globally) while control rules need to be coded at the individual robot level (i.e., locally). Connecting global to local levels or vice versa through mathematical modeling to predict the system behavior is generally assumed to be the grand challenge of swarm robotics. We propose to approach this problem by programming directly at the swarm level. Key to this solution is the use of heterogeneous swarms that combine appropriate subsets of agents whose hard-coded agent behaviors have known global effects. Our novel global-to-local design methodology allows to compose heterogeneous swarms for the example application of self-organized task allocation. We define a large but finite number of local agent controllers and focus on the global dynamics of behaviorally heterogeneous swarms. The user inputs the desired global task allocation for the swarm as a stationary probability distribution of agents allocated over tasks. We provide a generic method that implements the desired swarm behavior by mathematically deriving appropriate compositions of heterogeneous swarms that approximate these global user requirements. We investigate our methodology over several task allocation scenarios and validate our results with multiagent simulations. The proposed global-to-local design methodology is not limited to task allocation problems and can pave the way to formal approaches to design other swarm behaviors.

Group-living animals that rely on stable foraging or migratory routes can develop behavioural traditions to pass route information down to inexperienced individuals. Striking a balance between exploitation of social information and exploration for better alternatives is essential to prevent the spread of maladaptive traditions. We investigated this balance during cumulative route development in the homing pigeon Columba livia. We quantified information transfer within pairs of birds in a transmission-chain experiment and determined how birds with different levels of experience contributed to the exploration–exploitation trade-off. Newly introduced naïve individuals were initially more likely to initiate exploration than experienced birds, but the pair soon settled into a pattern of alternating leadership with both birds contributing equally. Experimental pairs showed an oscillating pattern of exploration over generations that might facilitate the discovery of more efficient routes. Our results introduce a new perspective on the roles of leadership and information pooling in the context of collective learning.

Vineyard mechanical winter pruning has been spreading worldwide, and the physiological basis ascribable to it has been consolidated throughout the years. Despite labor savings and reduction of costs having been proven, the demonstration of its economic viability might be challenging. In this context, this work aims to evaluate the vine performances and the costs of different degrees of the mechanization of winter pruning over a five-year trial (2011–2015). In a vineyard of cv. Trebbiano Romagnolo (Vitis vinifera L.) located in northern Italy, three pruning treatments were laid out as follows: (a) manual pruning (MAN); (b) mechanical pre-pruning and simultaneous manual follow-up (MP + F); (c) mechanical pruning without a manual follow-up (MP). The results showed a strong increase in the node number of MP. Nevertheless, the yield compensation factors (i.e., the shoot fruitfulness and cluster weight) limited the increase in productivity. Soluble solids did not differ between the pruning treatments, while titratable acidity resulted slightly higher only on the MP berries. The MP treatment was the most economically convenient, with a vineyard surface of 1.5 hectares, while mechanical pruning with manual finishing resulted more advantageous, compared to manual pruning when the vineyard surface was greater than 2.9 hectares. The agronomic and economic results obtained in this five-year trial suggest that mechanical pruning may be profitably applied also on grapevine varieties characterized by low basal bud fruitfulness, such as Trebbiano Romagnolo.

Abstract Background In previous studies, we demonstrated that the NEMO score, i.e. the cumulative number of microhaemorrhages (MHEs) and microthromboses (MTs), observed in nailfold videocapillaroscopy was a good indicator of the steady state level of disease activity (DA) in patients with systemic sclerosis (SSc) when the European Scleroderma Study Group (EScSG) index was considered the gold standard. Aim of the study To verify whether the NEMO score could be (i) a valid tool to assess DA, even when the modified European Scleroderma Trials and Research (EUSTAR) index was considered to be the comparator, and (ii) a sensitive method to capture the DA overtime changes. Patients and methods The NEMO score and the EScSG and EUSTAR indices were contemporarily assessed at baseline (T0) and after a follow-up of 4–56 months (T1) in 98 patients with SSc. The differences (Δ) between the T1 and T0 values of the NEMO score and the EScSG and EUSTAR indices were calculated and compared to each other. Vitis vinifera L. adapts well to a scarce availability of water in the soil. However, in recent decades, the combination of thermal stress with prolonged water scarcity could have dramatic consequences on the vine’s physiological status. In this paper, we evaluated the effects of two canopy shapes and two irrigation regimes at veraison on vine physiology, yield parameters and grape composition through biochemical and molecular approaches. The water shortage strongly influenced the physiology of Sangiovese only when the stress was moderate to severe. Neither the water stress limited to veraison nor the canopy shape were able to influence the yield parameters and sugar content, and a strong induction of the expression of the genes involved in the biosynthesis of anthocyanins was recorded only in conditions of moderate-to-severe stress. This phenomenon led to an increase in the anthocyanin content in berry skins until the end of veraison. Conversely, no significant effects occurred in terms of biochemical and molecular performance after re-watering and at harvest. Though the shape of the canopy could play a role only under elevated temperature and prolonged drought, severe water stress can affect the vine physiology and berry ripening during the veraison stage.

Behavioral correlations stretching over time are an essential but often neglected aspect of interactions among animals. These correlations pose a challenge to current behavioral-analysis methods that lack effective means to analyze complex series of interactions. Here we show that non-invasive information-theoretic tools can be used to reveal communication protocols that guide complex social interactions by measuring simultaneous flows of different types of information between subjects. We demonstrate this approach by showing that the tandem-running behavior of the ant Temnothorax rugatulus and that of the termites Coptotermes formosanus and Reticulitermes speratus are governed by different communication protocols. Our discovery reconciles the diverse ultimate causes of tandem running across these two taxa with their apparently similar signaling mechanisms. We show that bidirectional flow of information is present only in ants and is consistent with the use of acknowledgement signals to regulate the flow of directional information.

Abstract Background Interstitial lung disease in systemic sclerosis (SSc-ILD) is a major cause of SSc-related death. Imunosuppressive treatment (IS) is used in patients with SSc for various organ manifestations mainly to ameliorate progression of SSc-ILD. Data on everyday IS prescription patterns and clinical courses of lung function during and after therapy are scarce. Methods We analysed patients fulfilling American College of Rheumatology (ACR)/European League against Rheumatism (EULAR) 2013 criteria for SSc-ILD and at least one report of IS. Types of IS, pulmonary function tests (PFT) and PFT courses during IS treatment were evaluated. Results EUSTAR contains 3778/11,496 patients with SSc-ILD (33%), with IS in 2681/3,778 (71%). Glucocorticoid (GC) monotherapy was prescribed in 30.6% patients with GC combinations plus cyclophosphamide (CYC) (11.9%), azathioprine (AZA) (9.2%), methotrexate (MTX) (8.7%), or mycophenolate mofetil (MMF) (7.3%). Intensive IS (MMF + GC, CYC or CYC + GC) was started in patients with the worst PFTs and ground glass opacifications on imaging. Patients without IS showed slightly less worsening in forced vital capacity (FVC) when starting with FVC 50–75% or >75%. GC showed negative trends when starting with FVC

Abstract Background Some abnormalities in nailfold videocapillaroscopy (NVC), such as the presence of micro-haemorrhages (MHEs), micro-thromboses (MTs), giant capillaries (GCs) and reduction in the number of capillaries (nCs), suggest a disease activity (DA) phase in systemic sclerosis (SSc). In a previous paper, we showed that the number of micro-haemorrhages and micro-thromboses (the so-called NEMO score) was the NVC feature more closely associated with DA. The present study was aimed at validating the NEMO score as a measure of DA in patients with SSc. Methods Two cohorts of 122 and 97 patients with SSc who were referred to two different rheumatology units, one in Milan and one in Naples, respectively, constituted the validation cohorts. The NEMO score, the total number of GCs and the mean nCs per digit were the parameters defined in each patient by eight-finger NVC. An expert operator analysed the NVCs in each of the participating units. The European Scleroderma Study Group (ESSG) index was used to define the DA level in each patient at the time of NVC examination. The activity of Italian chabasite-rich zeolitites for control of grey mould, sour rot and grapevine moth was compared to that from synthetic fungicides and insecticides in three vineyards in the Abruzzo region of Italy. Persistence of zeolitites on grapevine canopies has enabled applications to be carried out before rainfall events, which are likely to predispose for infections by grey mould and sour rot pathogens. Applications for control of Lobesia botrana (grapevine moth) were carried out after the peak of the male flight, and the presence of eggs on grape berries was also assessed. Disease and pest control were very satisfactory and comparable to those obtained from synthetic fungicides and insecticides. In addition, there were no differences among treatments for yield, berry composition, or wine ethyl alcohol, pH and total acidity. Phenolic compounds increased in wine from zeolitite-treated plants where the treatment was carried out within 15 days from grape harvest. These promising results have strategic value, because zeolitites provided simultaneous control of grey mould, sour rot and L. botrana. However, since these compounds have been tested for the first time for the control of diseases and L. botrana, the results obtained in this study require further validation.

Throughout evolution, living systems have developed mechanisms to make adaptive decisions in the face of complex and changing environmental conditions. Most organisms make such decisions despite lacking a neural architecture. This is the case of the acellular slime mold Physarum polycephalum that has demonstrated remarkable information processing and problem-solving abilities. Previous studies suggest that the membrane of P. polycephalum plays an important role in integrating and processing information leading to the selection of a resource to exploit. The cyclical contraction-relaxation pattern of the membrane changes with the local quality of the environment, and individual contractile regions within a P. polycephalum can entrain neighboring regions, providing a potential mechanism for information processing and propagation. In this study, we use an information-theoretic tool, transfer entropy, to study the flow of information in single tubule segments of P. polycephalum in a binary choice between two food sources. We test P. polycephalum tubules in two food choice conditions, where the two available options are either symmetric in their nutrient concentrations or with one more concentrated in nutrients than the other (i.e., asymmetric). We measure the contractile pattern of the P. polycephalum membrane and use these data to explore the direction and amount of information transfer along the tubule as a function of the cell's final decision. We find that the direction of information transfer is different in the two experimental conditions, and the amount of information transferred is inversely proportional to the distance between different contractile regions. Our results show that regions playing a leading role in information transfer changes with the decision-making challenges faced by P. polycephalum.

Objective. To investigate the role of acetylsalicylic acid (ASA) in reducing the incidence of cardiovascular (CV) events in an Italian multicentre rheumatoid arthritis (RA) inception cohort. Methods. The clinical charts of RA patients consecutively admitted to 4 Italian centres for their 1st visit from November 1, 2000, to December 31, 2015, and followed up till December 2016 were retrospectively investigated for the incidence of CV events. Patients were subdivided into two groups, namely, ASA- and non-ASA-treated groups. The Kaplan–Meier curve and log-rank test were used to investigate differences in event-free survival. Cox regression analysis was carried out to identify factors associated with CV event occurrence. Results. Seven hundred forty-six consecutive RA patients were enrolled and followed up for a median of 5.6 years (range 2.9–8.9 years). The incidence rate (IR) of CV events was 8/1000 person-years (p-ys) in the overall cohort. The IR of CV events was significantly lower in the ASA-treated group with respect to the non-ASA-treated group (IR 1.7 vs. 11.8/1000 p-ys; p=0.0002). The CV event-free rate was longer in ASA-treated patients than in non-ASA-treated patients (log-rank test 12.8; p=0.0003). At multivariable analysis, arterial hypertension (HR 9.3) and hypercholesterolemia (HR 2.8) resulted to be positive predictors and ASA (HR 0.09) and hydroxychloroquine (HCQ) (HR 0.22) to be negative predictors. Conclusion. The IR of CV events in our Italian multicentre cohort was lower than that reported in other European and non-European cohorts. Low-dose ASA may have a role in the primary prophylaxis of CV events in RA patients.

A fluid challenge with a rapid infusion of saline helps to discriminate between pre- and post-capillary pulmonary hypertension (PH) and allows unmasking hidden post-capillary PH. Systemic sclerosis (SSc) patients may present with biventricular systolic and diastolic dysfunction. The aim of this study was to evaluate the hemodynamic changes of the pulmonary circulation in SSc patients without PH after a fluid challenge. Twenty-five SSc patients and 25 controls underwent right heart catheterization in basal conditions and after volume loading with saline infusion of 7 mL/kg over 5–10 min. At baseline, there was no difference in hemodynamics between SSc patients and controls. Rapid volume loading resulted in a significant increase in pressures and flows in both groups. Increases in right atrial pressure (3 ± 1 vs. 2 ± 1 mmHg, P = 0.03), mean pulmonary artery pressure (5 ± 1 vs. 3 ± 1 mmHg, P 18 mmHg suggesting latent left heart failure. Even if differences are small and not diagnostic for heart failure, SSc patients without PH have a larger increase in pulmonary vascular pressures and a smaller increase in cardiac output than controls after an acute volume loading, probably due to subclinical left ventricular diastolic dysfunction.

The study of phenolic maturity in Vitis vinifera L. requires a multidisciplinary approach to understand how the evolution of berry flavonoids and cell wall material influence the colour and the textures of red wine. This is a challenging issue which involves researchers of viticulture and enology, and the results of their work are of particular interest for the producers of high-quality red wines. This review reports the current knowledge regarding phenolic maturity, describing the sensorial traits of the different compounds, the evolution of berry flavonoids and the methodologies used to evaluate their characteristics. Finally, the role of cell wall material in influencing the extractability of anthocyanins and proanthocyanidins was shown. By means of a critical review of the results, it can be hypothesised that prolonged ripening improved colour characteristics and mouthfeel properties, thanks to the higher amounts of extractable skin flavonoids associated with lower amounts of seed proanthocyanidins, and to the increased affinity of the cell wall material for the proanthocyanidins most involved in the perception of unpleasant astringency.

High temperatures and the anomalous distribution of rainfall during the growing season may have a negative impact on grapevine yield and berry composition. In recent years, many studies have focused on the application of agronomical techniques to reduce the negative impact of heat waves on secondary metabolites such as phenols. In particular, treatments with kaolin have shown positive effects on reducing canopy temperatures, enhancing the accumulation of anthocyanins. In regard to the above, three treatments were evaluated: untreated control (C), kaolin (CAO), and chabasite-rich zeolitites (ZEO) applications on cv. Sangiovese in order to verify the cooling effects on leaves and bunches, and the impact on gas exchange, yield parameters, berry composition, and on both chemical and sensory notes of wine. Minerals were sprayed twice around the veraison on the entire canopy at a 3% concentration. The results showed that the application of the minerals was able to reduce the berry temperatures in both years of the trial as compared to the untreated control (C), without affecting vine gas exchange, yield, and soluble solid accumulation. Furthermore, the cooling effect determined an increase in anthocyanin on both the grapes and the wine. At testing, CAO and ZEO wines stood out regarding greater color intensity and were preferred by the judges.

The study of collective behavior has traditionally relied on a variety of different methodological tools ranging from more theoretical methods such as population or game-theoretic models to empirical ones like Monte Carlo or multi-agent simulations. An approach that is increasingly being explored is the use of information theory as a methodological framework to study the flow of information and the statistical properties of collectives of interacting agents. While a few general purpose toolkits exist, most of the existing software for information theoretic analysis of collective systems is limited in scope. We introduce Inform, an open-source framework for efficient information theoretic analysis that exploits the computational power of a C library while simplifying its use through a variety of wrappers for common higher-level scripting languages. We focus on two such wrappers here: PyInform (Python) and rinform (R). Inform and its wrappers are cross-platform and general-purpose. They include classical information-theoretic measures, measures of information dynamics and information-based methods to study the statistical behavior of collective systems, and expose a lower-level API that allow users to construct measures of their own. We describe the architecture of the Inform framework, study its computational efficiency and use it to analyze three different case studies of collective behavior: biochemical information storage in regenerating planaria, nest-site selection in the ant Temnothorax rugatulus, and collective decision making in multi-agent simulations.

Carbamylation is a post-translational modification that mostly affects proteins with low turnover, such as dermal proteins. Carbamylated proteins accumulate in skin in an age-dependent manner, contributing to tissue alterations. As dermis is affected by systemic sclerosis (SSc) and anti-carbamylated protein antibodies (anti-CarP Ab) are found in SSc patients, we sought to evaluate the specificity of anti-CarP Ab and their relationship with clinical parameters reflecting skin involvement in SSc. This study investigated serum samples and clinical data from 124 patients with SSc. Anti-CarP Ab were affinity purified from pooled SSc sera, and their specificity was assessed by western blotting and ELISA with carbamylated proteins from two species (human and bovine albumin; human fibrinogen). Anti-CarP Ab were measured in SSc serum samples and in 41 healthy aged-matched individuals. Affinity-purified anti-CarP Ab recognized carbamylated epitopes irrespective of the protein type or species origin. Among environmental factors, temperature is the one that poses serious threats to viticulture in the present and future scenarios of global climate change. In this work, we evaluated the effects on berry ripening of two thermal regimes, imposed from veraison to harvest. Potted vines were grown in two air-conditioned greenhouses with High Temperature (HT) and Low Temperature (LT) regimes characterized by 26 and 21°C as average and 42 and 35°C as maximum air daily temperature, respectively. We conducted analyses of the main berry compositional parameters, berry skin flavonoids and berry skin transcriptome on HT and LT berries sampled during ripening. The two thermal conditions strongly differentiated the berries. HT regime increased sugar accumulation at the beginning of ripening, but not at harvest, when HT treatment contributed to a slight total acidity reduction and pH increase. Conversely, growing temperatures greatly impacted on anthocyanin and flavonol concentrations, which resulted as strongly reduced, while no effects were found on skin tannins accumulation. Berry transcriptome was analyzed with several approaches in order to identify genes with different expression profile in berries ripened under HT or LT conditions. The analysis of whole transcriptome showed that the main differences emerging from this approach appeared to be more due to a shift in the ripening process, rather than to a strong rearrangement at transcriptional level, revealing that the LT temperature regime could delay berry ripening, at least in the early stages. Moreover, the results of the in-depth screening of genes differentially expressed in HT and LT did not highlight differences in the expression of transcripts involved in the biosynthesis of flavonoids (with the exception of PAL and STS) despite the enzymatic activities of PALs and UFGT being significantly higher in LT than HT. This suggests only a partial correlation between molecular and biochemical data in our conditions and the putative existence of post-transcriptional and post-translational mechanisms playing significant roles in the regulation of flavonoid metabolic pathways and in particular of anthocyanins.

Aims : To test the effects of varying degrees of vigour on vine growth, cropping, grape composition and wine quality. Methods and results: The study was conducted in 2008-2009 in a cv. Sangiovese (V. vinifera L.) vineyard (Tuscany). Two uniform zones marked by low (LV) and high (HV) vigour vines were pinpointed using an NDVI (normalized difference vegetation index) map. Soil analysis showed similar texture in both zones, but total soil nutrients were lower in LV than in HV. While only LV vines showed soil water content close to wilting point in 2008, they demonstrated lower leaf area and yield and higher berry sugar and anthocyanin concentrations compared to HV vines. Chemical and wine tasting analysis of the wines made in 2009 showed that the LV wines had better sensory attributes than the HV wines, despite their excessive ethanol content. Conclusions: The differences in vigour, yield and must and wine quality of LV compared to HV vines were linked to variations in soil fertility and water retention capacity. Precision vineyard management practices like supplementary fertilization and irrigation should be used to increase vigour and yield and to decrease sugar content in LV grapes. Cover crop may be used in HV vines to decrease their vigour and yield. Significance and impact of the study: The study confirms that the evaluation of within-field variability is crucial for site-specific vineyard management.

Objective: To compare the SDAI values to DAS28 scores in RA patients undergoing different DMARD regimens. Methods: The SDAI is an unweighted numerical sum of five outcome parameters: tender and swollen joint count (based on 28-joint assessment), patient and physician global assessment of disease activity (visual analogue scale: 0-10 cm) and level of C-reactive protein (mg/dl). 80 patients (F/M 68/12; age between 20-68 years, median 52) with active rheumatoid arthritis were prospectively enrolled in the study. The patients were randomly assigned to one of four groups according to the therapeutic regimens: group I: Methotrexate (MTX) 15 mg/weekly + salazopyrin 2 g/daily; group II: MTX 15 mg/weekly + infliximab 3 mg/Kg at time 0, 2, 4 and every 8 weeks; group III: MTX 15 mg/weekly + etanercept 25 mg/twice weekly; group IV: MTX 15 mg/weekly + adalimumab 40 mg/every other week. SDAI and DAS28 were determined at baseline and after 6 months in each patient. Mean changes in SDAI values were compared to those detected in DAS 28 at baseline and after 6 months. Results: SDAI and DAS 28 were found to be significantly correlated at baseline. Moreover, changes in SDAI over time paralleled those in DAS, and were found to be significantly correlated. Conclusions: SDAI is a valid measure of response to treatment in RA patients undergoing different therapeutic regimens.

Aims: Recent trials on Sangiovese vines have shown that hand defoliation of shoot basal leaves at pre-bloom is effective in reducing fruit set and yield, leading to better grape composition and quality. The present work was performed to assess whether similar outcomes could be obtained by a more economically viable mechanical approach, which appears to be extremely attractive in cultivars such as Sangiovese, marked by high or very high yield potential and heavy, fairly compact clusters quite sensitive to rot. Methods and results: The trial was designed to compare pre-bloom mechanical defoliation (MD), hand defoliation (HD) and no defoliation (C) on Sangiovese vertical shoot positioned and spur pruned cordon de Royat trained vines. In the HD treatment, the first six basal leaves of each shoot were removed (70 % of leaf area), whereas in the MD treatment 33 % of the leaf area was removed from the basal part of the shoots. HD and MD compared to C reduced fruit set (HD = 29.8%; MD = 24.2%; C = 35.5 %), yield per shoot (HD = 546 g; MD = 516 g; C = 764 g), cluster weight (HD = 292 g; MD = 272 g; C = 382 g) and berry weight (HD = 2.17 g; MD = 2.31 g; C = 2.45 g), but improved total soluble solids (HD = 23.0 °Brix; MD = 22.5 °Brix; C = 20.8 °Brix) and total anthocyanins (HD = 837 mg/kg of grapes; MD = 744 mg/kg of grapes; C = 647 mg/kg of grapes). Leaf photosynthesis, measured in 2007, increased only temporarily after HD and MD as compared to control. Conclusions: The pre-bloom HD of shoot basal leaves confirmed its positive effect on crop yield control and grape composition, leading to better grape quality. The pre-bloom MD of the basal part of the shoots maintained most of the advantages associated with HD, although only half of the leaf area removed by hand was removed by the machine. Significance and impact of the study: Pre-bloom mechanical defoliation can replace hand defoliation and partially replace the time-consuming and costly manual cluster thinning technique, which is often used in high yield cultivars such as Sangiovese.

The engineering of large-scale decentralised systems requires sound methodologies to guarantee the attainment of the desired macroscopic system-level behaviour given the microscopic individual-level implementation. While a general-purpose methodology is currently out of reach, specific solutions can be given to broad classes of problems by means of well-conceived design patterns. We propose a design pattern for collective decision making grounded on experimental/theoretical studies of the nest-site selection behaviour observed in honeybee swarms (Apis mellifera). The way in which honeybee swarms arrive at consensus is fairly well-understood at the macroscopic level. We provide formal guidelines for the microscopic implementation of collective decisions to quantitatively match the macroscopic predictions. We discuss implementation strategies based on both homogeneous and heterogeneous multiagent systems, and we provide means to deal with spatial and topological factors that have a bearing on the micro-macro link. Finally, we exploit the design pattern in two case studies that showcase the viability of the approach. Besides engineering, such a design pattern can prove useful for a deeper understanding of decision making in natural systems thanks to the inclusion of individual heterogeneities and spatial factors, which are often disregarded in theoretical modelling.

BackgroundHigh interleukin (IL)-17A levels are characteristically found in the skin of systemic sclerosis (SSc) individuals. Our aim was to investigate whether the dermal expression of IL-17A and related IL-17 family members (i.e. IL-17C, IL-17E and IL-17F) could distinguish fibrotic from healthy skin and could show similarities in SSc and morphea, two disorders with presumed distinct pathogenesis, but characterized by skin fibrosis.MethodsBiopsies were obtained from the involved skin of 14 SSc, 5 morphea and 8 healthy donors (HD) undergoing plastic surgery. Immunohistochemistry/immunofluorescence techniques were coupled to a semi-automated imaging quantification approach to determine the presence of the IL-17 family members in the skin. The in vitro effects induced by the IL-17 family members on fibroblasts from normal and SSc individuals were assessed by ELISA and RIA.ResultsPositive cells for each of the IL-17 isoforms investigated were present in the dermis of all the individuals tested, though with variable frequencies. SSc individuals had increased frequency of IL-17A+ (p = 0.0237) and decreased frequency of IL-17F+ (p = 0.0127) and IL-17C+ cells (p = 0.0008) when compared to HD. Similarly, morphea individuals had less frequent IL-17C+ cells (p = 0.0186) in their skin but showed similar number of IL-17A+ and IL-17F+ cells when compared to HD. Finally, IL-17E+ cells were more numerous in morphea (p = 0.0109) and tended to be more frequent in SSc than in HD. Fibroblast production of IL-6, MMP-1 and MCP-1 was enhanced in a dose-dependent manner in the presence of IL-17E and IL-17F, but not in the presence of IL-17C. None of the cytokine tested had significant effect on type I collagen production. Of interest, in SSc the frequency of both IL-17A and IL-17F positive cells increased with disease duration.ConclusionsThe frequency of IL-17A and IL-17F distinguish SSc to morphea individuals while dermal expression of IL-17C (low) and IL-17E (high) identifies a fibrosis-specific motif. The specific IL-17C/IL-17E cytokine combination may thus play a role in the development of fibrosis.

Centromere-associated protein A (CENP-A), a common autoimmune target in a subset of systemic sclerosis patients, appears to have no role to explain why its corresponding auto-antibodies are more frequently found in the limited than the diffuse form of systemic sclerosis. Therefore, we investigated the fine specificity of anti-CENP-A antibodies as a first step to understanding their role in systemic sclerosis pathology. We focused on the amino-terminal portion of CENP-A spanning amino acids 1 to 17 (Ap(1-17)), which represents, along with Ap(17-30), an immunodominant epitope of the protein. Peptide Ap(1-17) was used to purify antibodies from 8 patients with systemic sclerosis. Anti-Ap(1-17) antibodies specifically reacted with human CENP-A but did not cross-react with CENP-B or Ap(17-30). Panning of a phage display peptide library with anti-Ap(1-17) antibodies from 2 patients identified two novel, partially overlapping motifs, and as the result of the alignment of specific phage clone insert sequences. Anti-Ap(1-17) IgG from the 8 patients had different reactivities to isolated phage clone insert sequences. Scanning the Swiss-Prot database revealed a large number of different types of proteins containing the two Ap(1-17) antigenic motifs. These data show that anti-CENP-A(1-17) antibodies are generated independently from anti-CENP-B antibodies and display great heterogeneity in their specificity by recognizing different motifs within that peptide sequence. This finding, along with the widespread interspecies and human tissue distribution of the two motifs, suggests that the number of motif-expressing proteins which can be the potential target of these antibodies is markedly higher than that estimated from the peptide-based epitope spreading model.

Systemic sclerosis (SSc) is an orphan, complex, inflammatory disease affecting the immune system and connective tissue. SSc stands out as a severely incapacitating and life-threatening inflammatory rheumatic disease, with a largely unknown pathogenesis. We have designed a two-stage genome-wide association study of SSc using case-control samples from France, Italy, Germany, and Northern Europe. The initial genome-wide scan was conducted in a French post quality-control sample of 564 cases and 1,776 controls, using almost 500 K SNPs.
