Abstract

La Leucosis Enzoótica Bovina (LEB) es una enfermedad infecciosa que afecta principalmente a los rodeos lecheros. Su agente causal es el virus de la leucosis bovina (BLV), perteneciente a la familia Retroviridae. Aunque la mayoría de los animales infectados son asintomáticos, algunos desarrollan linfosarcoma. La transmisión de BLV ocurre principalmente por vía horizontal, a través de prácticas iatrogénicas como aplicaciones inyectables y palpaciones rectales. De manera vertical por vía transplacentaria, calostro y leche. Se ha sugerido la posibilidad de transmisión por insectos hematófagos. En Argentina, la enfermedad ha avanzado de manera exponencial desde la década de 1970 cuando se diagnosticó por primera vez. La prevalencia de LEB varía en diferentes regiones, y se observa una mayor tasa de infección en tambos en la región central del país. El presente estudio llevado a cabo en la provincia de Misiones durante el año 2019, tuvo como objetivo actualizar información sobre la situación de BLV en 135 tambos del sector de la agricultura Familiar en la provincia de Misiones, y su distribución espacial. Se encontró una tasa de infección del 13% en rodeos y se identificó un conglomerado espacial de alta tasa en la zona donde se concentran la mayor cantidad de tambos de la provincia. La infección fue más común en la categoría vacas y en aquellos tambos de mayor tamaño, aunque estas diferencias no fueron estadísticamente significativas. Estos resultados proporcionan información actualizada sobre la presencia de BLV en tambos de Agricultura Familiar en Misiones, sugiriendo una menor prevalencia en comparación con otras regiones lecheras de Argentina. Se destaca la importancia de abordar prácticas de manejo de riesgo para prevenir la transmisión de la enfermedad.

Abstract

Ante-mortem diagnosis of bovine tuberculosis (bTB) is based mainly on the tuberculin skin test (TST) and the ɣ-IFN release assay (IGRA). Some infected animals escape screening tests, thus, limit herd sanitation. Previous reports have suggested a predominant pattern of multi-organ lesions attributable to Mycobacterium bovis (the causative agent of bTB) bacteraemia. A case–control study was conducted to investigate blood PCR as an alternative tool for improving ante-mortem detection of TST false-negative bovines. Cases comprised 70 TST false-negative bovines (cases), which were serology positive, and controls included 81 TST positive bovines; all of them confirmed as infected with M. bovis. Detection of the IS6110 target through touchdown blood-PCR (IS6110 TD-PCR) was performed. The positivity of the blood-PCR was 27.2% in the control group. This performance was similar to the 15% obtained among cases (p = 0.134). Most cases identified by the IS6110 TD-PCR exhibited focalized lesions (p = 0.002). Results demonstrated that blood-PCR could detect TST false-negative cattle, even if they are negative for IGRA. Considering that cases exhibited humoral response to M. bovis, further studies conducted in a pre-serological stage could provide evidence about the real contribution of the technique in herds.

Abstract

La Estomatitis Papular Bovina (BPSV) es una enfermedad viral caracterizada por la formación de pápulas o nódulos en morro, labios, lengua y mucosa oral. En Argentina, existen escasos registros de su presencia. Se comunica un caso de BPSV observado en terneros de un establecimiento lechero. El comportamiento clínico-epidemiológico, los hallazgos patológicos y la identificación del agente mediante biología molecular son concluyentes para la confirmación del mismo. Abstract

El síndrome conocido como “oreja caída” es un hallazgo clínico observado en los sistemas productivos bovinos. Varias son las etiologías descriptas como causa de esa patología. El objetivo del presente trabajo es describir tres casos de oreja caída en bovinos. Se estudiaron dos casos en engorde a corral y uno en guachera. Se realizaron exámenes clínicos, necropsias, histopatologías y diferentes técnicas entre ellas cultivo de hongos, de bacterias, de virus, etc. En todos los animales necropsiados se observó aumento de tamaño del ganglio de Gasser. En los estudios histopatológicos se encontró reacción granulomatosa y presencia de estructuras compatibles con criptococos que fueron positivas a la tinción de PAS. El aislamiento y tipificación de Cryptococcus neoformans de 1 animal, los resultados de los exámenes anatomo-histopatológicos, y las características estructurales y tintoriales observadas en todos los casos indican un diagnóstico compatible con criptococosis nerviosa.

Abstract

En tuberculosis, el conocimiento de los diferentes tipos de lesiones, permite comprender con mayor profundidad la patología y el contagio de la enfermedad. Las cabras tienen un rol importante en transmitir esta afección a bovinos y a humanos. El objetivo del presente trabajo es describir las lesiones macroscópicas y microscópicas observadas en cabras naturalmente infectadas con Mycobacterium bovis (M. bovis). Se analizaron 55 caprinos positivos a la prueba de tuberculina, detallando las diferentes presentaciones patológicas macroscópicas y microscópicas. Entre ellas se encontraron 5 animales con lesiones cavitarias pulmonares. Se concluye que, en cabras infectadas naturalmente, existe una amplia variedad de lesiones, afectando principalmente el sistema respiratorio; algunas de ellas involucradas muy directamente con la trasmisión de la enfermedad dentro de los hatos.

6. Genotyping Mycobacterium bovis from cattle in the Central Pampas of Argentina: temporal and regional trends

Abstract

Mycobacterium bovis is the causative agent of bovine tuberculosis (TB), a disease that affects approximately 5% of Argentinean cattle. Among the molecular methods for genotyping, the most convenient are spoligotyping and variable number of tandem repeats (VNTR). A total of 378 samples from bovines with visible lesions consistent with TB were collected at slaughterhouses in three provinces, yielding 265 M. bovis spoligotyped isolates, which were distributed into 35 spoligotypes. In addition, 197 isolates were also typed by the VNTR method and 54 combined VNTR types were detected. There were 24 clusters and 27 orphan types. When both typing methods were combined, 98 spoligotypes and VNTR types were observed with 27 clusters and 71 orphan types. By performing a meta-analysis with previous spoligotyping results, we identified regional and temporal trends in the population structure of M. bovis. For SB0140, the most predominant spoligotype in Argentina, the prevalence percentage remained high during different periods, varying from 25.5-57.8% (1994-2011). By contrast, the second and third most prevalent spoligotypes exhibited important fluctuations. This study shows that there has been an expansion in ancestral lineages as demonstrated by spoligotyping. However, exact tandem repeat typing suggests dynamic changes in the clonal population of this microorganism.

Abstract

Detection and identification of bovine tuberculosis and its differentiation from micobacteriosis is fundamental during diagnoses. That is why mycobacteria laboratories improvement becomes essential in public health and veterinary medicine services. The objective of the present research is to differentiate Mycobacterium bovis and nontuberculous mycobacterias in isolates cultured from domestic and wildlife species from seven Argentinean provinces during 2004-2008. Differentiation was based on biochemical tests, phenotypic characteristics and M. bovis spolygotyping. Biochemical and phenotypic identification resulted in 20 M. bovis strains, 18 of them were confirmed by spolygotyping, and 34 nontuberculous mycobacteria strains. Thirteen species were characterized and all of them were grouped considering biological risk and pathogenic potential reported in humans and/or animals. Here we have reached advances in tuberculosis and micobacteriosis diagnoses in veterinary medicine. In this area diagnoses are often based on micro and macroscopic observation of the tubercles and skin test results. These advances are not minor as zoonotic tuberculosis is still a public health problem in Latin America.

Abstract

Bovine tuberculosis (bTB) is caused by Mycobacterium bovis and disseminated worldwide. In Argentina, the highest prevalence occurs in dairy areas. BoLA DRB3.2 is related to the adaptive immunity in mycobacterial infections. Genetic polymorphisms of this marker have been associated with resistance or susceptibility to bovine diseases. We evaluated the association between BoLA DRB3.2 polymorphisms and bTB pathology scores in dairy and beef cattle breeds of Argentina. Most bovines exhibited visible lesions compatible with tuberculosis and, furthermore, 150 (85.7%) were also positive by bacteriology. A pathology index showed a variable degree of disease, from 3 to 76 (median pathology score = 9 (IQR: 7-15)). Thirty-five BoLA DRB3.2 alleles were identified with an associated frequency from 16% to 0.3%, distributed 73% (n = 128) in heterozygosis and 27% (n = 47) in homozygosis, with 12 BoLA DRB3.2 alleles (*0101, *1101, *1501, *0201, *2707 *1001, *1002, *1201, *14011, *0501 *0902 and *0701) representing the 74.7% of the population variability. A functional analysis grouped them in 4 out of 5 clusters (A-D), suggesting a functional overlapping. Among the 90 identified genotypes, *1101/*1101, *1101/*1501 and *0101/*0101 were the most frequent (10%, 8.9% and 8.9%, respectively). No association was detected between the pathology scores and a specific DRB3.2 allele (p .05). Animals infected with M. bovis spoligotype SB0153 showed a significantly higher pathology score than those affected by the spoligotype SB0145 (p = .018). Furthermore, the Aberdeen Angus breed exhibited highest pathological scores (p .0001), which were associated with disseminated lesion, thus suggesting that the host component could be important to the disease progression.

Abstract

Aims The present work assessed the ability of two selected lactic acid bacteria (LAB) strains (Schleiferilactobacillus perolens CRL1724 and Lactococcus lactis subsp. lactis CRL1655) to inhibit the adherence of bovine mastitis pathogens to mammary epithelial cells (MAC-T) and their effects (if any) on the structure of the gland after intramammary inoculation at dry-off. Methods and Results Established bovine mammary epithelial cells (MAC-T) were used to assess the LAB strains' ability to inhibit the adherence of bovine mastitis pathogens. Monolayers of MAC-T cells were co-cultured with the LABs and then individual pathogen was added. Both strains prevented the adherence of S. aureus RC108, S. chromogenes, S. uberis UT102 and E. coli ATCC 35218. Adherence of the latter two pathogens was inhibited most strongly in vitro. To evaluate the effect of the LAB on the structure of the bovine udders, quarters were intramammary inoculated with the LAB mixture at dry-off. After slaughtering, the teats were dissected and histopathologically analysed. No modifications were identified post-inoculation in the structure of the epithelial, subepithelial and connective tissues of the mammary gland. Conclusions Probiotic strains L. lactis subsp lactis CRL1655 and S. perolens CRL1724 were both able to inhibit the adherence of a number of bovine mastitis pathogens in vitro, and that the intramammary inoculation of these strains at the established dose and concentration did not cause significant alterations in the mammary epithelium nor had undesirable effects on tissues, and may therefore be considered harmless. Significance and Impact of Study The promising findings demonstrated in this work support the potential of probiotic micro-organisms as a natural and effective alternative to prevent bovine mastitis during the dry-off period.

Abstract

Aims &Objective: Mycobacterium bovis is the causative agent of bovine tuberculosis. It is also a pathogen for other animals such as goats and humans. Molecular methods are applied to detect members of the M. tuberculosis complex in different clinical samples as milk. The Loop-Mediated Isothermal Amplification (LAMP) proceeds at a constant temperature using a strand displacement reaction. LAMP does not require a thermal cycler. It is characterized by its efficiency, rapidity, high yield of final product, robustness, sensitivity and specificity. The primers are specifically designed to recognize 6 distinct regions on the target gene. The amplified product can be detected by naked eye as a white precipitate or a color change of the reaction solution.: To detect species of the Mycobacterium tuberculosis complex in goat milk samples by IS6110-LAMP Methods: Twenty goat milk samples from animals reacting to the tuberculin test were studied by IS6110-LAMP, Touch-Down (TD) IS6110 and Rv2807 PCR. These goats had been slaughtered, their organs and milk cultured. The isolates obtained were typed by spoligotyping. DNA extraction was performed using 500uL of milk sample. It was incubated with sodium dodecyl sulfate / proteinase K and treated twice with phenol:chloroform:isoamylic alcohol and once with chloroform:isoamylic alcohol. Then NaCl was added and DNA was precipitated with isopropanol and washed with ethanol. The remaining ethanol was evaporated and the pellet resuspended in water. IS6110-LAMP reaction was performed in a 25uL volume containing Bst DNA polymerase, betaine, magnesium sulfate, dNTPs, and primers (F3, B3, FIP, BIP, LF, LR). SYBR Gold solution was added inside of the cup of the tubes that were incubated at 63°C during one hour. After that the tubes were shacked and a yellowish color indicated a positive reaction. The specificity of IS6110-LAMP was evaluated using DNA from different species of Non Tuberculous Mycobacterias (NTM) (Mycobacterium smegmatis, M. phlei, M. fortuitum, M. gordonae, M. intracellulare, M. porcinum, M. avium subsp. avium, M. avium subsp. paratuberculosis) and the close related Nocardia sp. TD IS6110 and TDRv2807 PCR were also performed with an initial step of 96°C for 3 min, followed by 8 cycles of 96°C for 1 min, annealing temperatures starting at 72°C for 1 min (decreasing by 1 °C/cycle), and 72°C for 1 min for extension. This step was followed by 30 cycles of 96°C for 1 min, 65°C for 1 min, 72°C for 2 min, and a final extension at 72°C for 8 min. The amplification products were 245 and 443 bp, respectively. Results: While 17/20 goats were confirmed by bacteriology, no isolates were obtained from milk culture. The 17 isolates were further confirmed as M. bovis by spoligotyping. Neither NTM nor Nocordia sp. templates amplified by IS6110-LAMP. The 40 % (8/20) of the samples were IS6110-LAMP positive. There was only one discordant result between IS6110-LAMP, TD-Rv2807 and TD-IS6110. Conclusion: IS6110-LAMP is a useful tool to detect species of the M. tuberculosis complex in goat milk samples that could be implemented in laboratories with low technological complexity.

Abstract

ESAT-6, CFP-10 and EspC are virulence factors that have been extensively assayed for bovine and human tuberculosis diagnosis due their potent T-cell inducing activities. While polymorphisms of ESAT-6 and CFP-10 were analyzed, with the description of CFP-10 variants in M. tuberculosis, this fact has not been explored in M. bovis field isolates. The coding sequences of esxA (ESAT-6), esxB (CFP-10) and mb3645c (EspC) from 58 M. bovis strains exhibiting genomic variability (spoligotyping) were analyzed. Two genes –esxA and esxB – remained invariant while mb3645c exhibited one synonymous polymorphism (G to A mutation, position 66bp) in one isolate, compared to M. bovis AF2122/97 reference strain. All isolates exhibited a synonymous nucleotide polymorphism simultaneously (G to A mutation, position 255bp), compared to M. tuberculosis H37Rv reference strain. This study confirms the high conservation for ESAT-6, CFP-10 and EspC in local M. bovis field isolates and reinforce the use of these three antigens in the diagnosis of bovine tuberculosis. Abstract

Bovine tuberculosis (bTB) is a common zoonotic disease, caused by Mycobacterium bovis (M. bovis),responsible for significant economic losses worldwide. Its diagnosis is based on the detection of cellmediated immunity under the exposure to protein purified derivative tuberculin (PPD), a complex andpoorly characterized reagent. The cross-reactivity to non-tuberculous mycobacterium species (falsepositiveresults) has been crucial to develop a more proper antigen. In the present study, we selected sixM. bovis Open Reading Frames (Mb1992, Mb2031c, Mb2319, Mb2843c, Mb2845c and Mb3212c) by insilicoanalysis and evaluated them in experimental and natural infection; none of these antigens hadbeen previously assessed as diagnostic antigens for bTB. The reactivity performance was tested in animalswith both positive and negative Tuberculin Skin Test (TST) results as well as in cattle infected withMycobacterium avium subesp. paratuberculosis (MAP). The six recombinant antigens individually inducedan IFN-g response, with overall responder frequency ranging from 18.3 to 31%. Mb2845c was the mostvaluable antigen with the potential to discriminate TST-positive cattle from either TST-negative or MAPinfected animals. Mb2845c showed similar performance to that observed with ESAT-6 and PPD-B amongTST and MTC specific-PCR positive animals, although this result needs to be proven in further studieswith a higher sample size. Our data confirm the feacibility to implement bioinformatic screening toolsand suggest Mb2845c as a potential diagnostic antigen to be tested in protein cocktails to evaluate theircontribution to bTB diagnosis. Abstract

Mycobacterium bovis es el agente etiológico de la tuberculosis bovina. La confirmación diagnóstica en el laboratorio se realiza a través del aislamiento bacteriológico. Los ensayos interlaboratorio permiten evaluar el desempeno˜ de cada laboratorio participante comparándolo con otros de capacidades similares. El objetivo del presente estudio fue determinar la eficiencia en el aislamiento de M. bovis. Este estudio contó con la participación de 4 laboratorios, los cuales procesaron a ciego 25 muestras de tejidos con lesiones, seleccionadas previamente por aislamiento. El laboratorio A obtuvo la mayor proporción de aislamientos (68%), seguido del C (60%) y luego del B y D (ambos con el 52%). Las mayores concordancias se observaron entre los pares de laboratorios B-C y B-D (68%). Las diferencias pudieron deberse a factores propios de los procedimientos en cada laboratorio. Este tipo de análisis interlaboratorio permite evidenciar posibles errores en los procedimientos bacteriológicos e identificar puntos críticos del proceso para detectar M. bovis de manera eficiente. Abstract

Cattle are the host and main reservoir of the etiologic agent of bovine tuberculosis, Mycobacterium bovis; although other mammalian species, including humans, are susceptible. The tuberculin test and/or slaughterhouse surveillance is the diagnostic method used by control programs all around the world to control and eradicate the disease. In order to compare different tuberculosis diagnostic tests and to reach disease confirmation, a study was performed in a group of 14 steers of Friesian breed, reacting positively to tuberculin test. Three ante-mortem assays were performed according to the type of sample: the gamma interferon (IFN-gamma) test (which quantifies the release of this cytokine by sensitized lymphocytes in whole blood in response to purified protein derivative (PPD) and recombinant ESAT-6 and CFP10 proteins); PCR and bacteriologic culture from nasal swab and intradermal tuberculin test. These assays were taken at different times to assess the evolution of clinical parameters. Post-mortem examination showed macroscopic and microscopic tuberculosis lesions with acid-fast bacillus and positive cultures. By spoligotyping, we observed that all the isolates showed the same pattern. The positive results based on comparison to lesions observed ranged from 58% to 75% for the IFN-gamma assays, to 72% for cultures, and ranged from 50% to 90% for PCR in nasal swabs. In conclusion, in a herd infected by the same strain, ante-mortem direct and immune-diagnostic parameters change, suggesting that several tests are needed for a faster control of infection at herd level.

Abstract

Mycobacterium bovis is the causative agent of bovine tuberculosis (TB), a disease that affects approximately 5% of Argentinean cattle. Among the molecular methods for genotyping, the most convenient are spoligotyping and variable number of tandem repeats (VNTR). A total of 378 samples from bovines with visible lesions consistent with TB were collected at slaughterhouses in three provinces, yielding 265 M. bovisspoligotyped isolates, which were distributed into 35 spoligotypes. In addition, 197 isolates were also typed by the VNTR method and 54 combined VNTR types were detected. There were 24 clusters and 27 orphan types. When both typing methods were combined, 98 spoligotypes and VNTR types were observed with 27 clusters and 71 orphan types. By performing a meta-analysis with previous spoligotyping results, we identified regional and temporal trends in the population structure of M. bovis. For SB0140, the most predominant spoligotype in Argentina, the prevalence percentage remained high during different periods, varying from 25.5-57.8% (1994-2011). By contrast, the second and third most prevalent spoligotypes exhibited important fluctuations. This study shows that there has been an expansion in ancestral lineages as demonstrated by spoligotyping. Abstract

Mycobacterium bovis is the causative agent of bovine tuberculosis, a disease that affects approximately 5% of Argentine cattle. The aim of this research was to study if it is possible to infer the degree of virulence of different M. bovis genotypes based on scorified observations of tuberculosis lesions in cattle. In this study, we performed association analyses between several parameters with tuberculosis lesions: M. bovis genotype, degree of progression of tuberculosis, and animal age. For this purpose, the genotype was determined by spoligotyping and the degree of bovine tuberculosis gross lesion was quantified with a score based on clinical observations (number, size, and location of granulomas along with histopathologic features). This study was performed with naturally infected cattle of slaughterhouses from three provinces in Argentina. A total of 265 M. bovis isolates were obtained from 378 pathological lesion samples and 192 spoligotyping and VNTR (based on ETR sequences) typing patterns were obtained. SB0140 was the most predominant spoligotype, followed by SB0145. The spoligotype with the highest lesion score was SB0273 (median score of 27 ± 4.46), followed by SB0520 (18 ± 5.8). Furthermore, the most common spoligotype, SB0140, had a median score of 11 ± 0.74. Finally, the spoligotype with the lowest score was SB0145 (8 ± 1.0). ETR typing of SB0140, SB0145, SB0273, and SB0520 did not subdivide the lesion scores in those spoligotypes. In conclusion, SB0273 and SB0520 were the spoligotypes with the strongest association with hypervirulence and both spoligotypes were only found in Río Cuarto at the south of Córdoba province. Abstract

The effect of intramammary inoculation of Lactobacillus perolens CRL 1724 on bovine udders at drying off was evaluated through histological examination of the canal and cistern tissues. The persistence of the strain in the udder 7 d post inoculation was also determined. Lb. perolens CRL 1724 was recovered from all mammary quarters and no clinical signs or teat damage were observed after inoculation of 106 cfu/ml. The udders showed a normal structural aspect and there were no modifications of the milk appearance. Lb. perolens CRL 1724 cells were evidenced on the surface of the epithelial cells of the cistern without causing any morphological modifications or cell alterations. Lb. perolens CRL 1724 produces a mild inflammatory reaction, characterized by recruitment of neutrophils to the epithelial zone and a slight hyperaemia into blood vessels. This preliminary study provides important information for further studies directed towards the inclusion of Lb. perolens CRL 1724 in the design of probiotic products for preventing bovine mastitis in non-lactating dairy cows.
