To study the effect of height–diameter ratio on the uniaxial compression performance of gangue-cemented backfill columns, five groups of cylindrical specimens with height–diameter ratios of 0.5, 1.0, 2.0, 3.0, and 4.0 were prepared for uniaxial compression tests. At the same time, digital image correlation technology and acoustic emission were used for monitoring, and the stress curve, strain curve, apparent strain, acoustic emission ringing, energy, and impact number of the backfill column were recorded. The obtained data were processed and corresponded with each other with time to explore the failure characteristics of different height–diameter ratio backfill bodies. The results show that the peak stress and peak strain of gangue-cemented backfill decrease exponentially with increasing height–diameter ratio. The failure form of the backfill column changes from global failure to local failure when the height–diameter ratio is greater than 2.0, and a local strain of more than 16% can lead to the overall instability of the backfill column. When the height-to-diameter ratio of the backfill column increased from 1.0 to 4.0, the damage degree for the backfill column decreased to 11%. The proportion of damage of backfill bodies in the cumulative ringing count rapid rise region increased from 22.6% to 72.3% with increasing height–diameter ratio and reached 40.9% when the height–diameter ratio was greater than 2.0. The rising speed accelerated, and the concentration of damage was higher. The final cumulative ringing count of acoustic emission reached a maximum at a height–diameter ratio of 2.0, and the position of ringing concentration was greatly influenced by this ratio. The ratio of acoustic emission energy-to-ringing count (E/C) reacts to the intensity of energy release. The average E/C increases from 1.26 to 2.76 with increasing height–diameter ratio. Before the peak stress, the rising speed of E/C decreases above a height–diameter ratio of 2.0, and after the peak stress, the E/C rises faster above a height–diameter ratio of 2.0. Before the peak stress, the intensity of the failure of the backfill column reaches a maximum when the height–diameter ratio is 3.0, and after the peak stress, the failure process is more violent for the backfill column with a height–diameter ratio greater than 2.0. The test results provide a reference for selecting the height–diameter ratio of the backfill column in structural backfill mining.

Breast cancer (BC) is a complex disease, which causes of high mortality rate in women. Early diagnosis and therapeutic improvements may reduce the mortality rate. There were more than 74 individual studies that have suggested BC-causing hub-genes (HubGs) in the literature. However, we observed that their HubG sets are not so consistent with each other. It may be happened due to the regional and environmental variations with the sample units. Therefore, it was required to explore hub of the HubG (hHubG) sets that might be more representative for early diagnosis and therapies of BC in different country regions and their environments. In this study, we selected top-ranked 10 HubGs ( CCNB1 , CDK1 , TOP2A , CCNA2 , ESR1 , EGFR , JUN , ACTB , TP53 , and CCND1 ) as the hHubG set by the protein-protein interaction network analysis based on all of 74 individual HubG sets. The hHubG set enrichment analysis detected some crucial biological processes, molecular functions, and pathways that are significantly associated with BC progressions. The expression analysis of hHubGs by box plots in different stages of BC progression and BC prediction models indicated that the proposed hHubGs can be considered as the early diagnostic and prognostic biomarkers. Finally, we suggested hHubGs-guided top-ranked 10 candidate drug molecules (SORAFENIB, AMG-900, CHEMBL1765740, ENTRECTINIB, MK-6592, YM201636, masitinib, GSK2126458, TG-02, and PAZOPANIB) by molecular docking analysis for the treatment against BC. We investigated the stability of top-ranked 3 drug-target complexes (SORAFENIB vs ESR1 , AMG-900 vs TOP2A , and CHEMBL1765740 vs EGFR ) by computing their binding free energies based on 100-ns molecular dynamic (MD) simulation based Molecular Mechanics Poisson-Boltzmann Surface Area (MM-PBSA) approach and found their stable performance. The literature review also supported our findings much more for BC compared with the results of individual studies. Therefore, the findings of this study may be useful resources for early diagnosis, prognosis, and therapies of BC.

Abstract PTENα/β, two variants of PTEN, play a key role in promoting tumor growth by interacting with WDR5 through their N-terminal extensions (NTEs). This interaction facilitates the recruitment of the SET1/MLL methyltransferase complex, resulting in histone H3K4 trimethylation and upregulation of oncogenes such as NOTCH3, which in turn promotes tumor growth. However, the molecular mechanism underlying this interaction has remained elusive. In this study, we determined the first crystal structure of PTENα-NTE in complex with WDR5, which reveals that PTENα utilizes a unique binding motif of a sequence SSSRRSS found in the NTE domain of PTENα/β to specifically bind to the WIN site of WDR5. Disruption of this interaction significantly impedes cell proliferation and tumor growth, highlighting the potential of the WIN site inhibitors of WDR5 as a way of therapeutic intervention of the PTENα/β associated cancers. These findings not only shed light on the important role of the PTENα/β-WDR5 interaction in carcinogenesis, but also present a promising avenue for developing cancer treatments that target this pathway.

Abstract Background Postoperative central diabetes insipidus (CDI) is commonly observed in craniopharyngioma (CP) patients, and the inflammatory response plays an important role in CPs. We aimed to evaluate the predictive value of preoperative peripheral inflammatory markers and their combinations regarding CDI occurrence in CPs. Methods The clinical data including preoperative peripheral inflammatory markers of 208 CP patients who underwent surgical treatment were retrospectively collected and analyzed. The preoperative peripheral white blood cells (WBC), neutrophils, lymphocytes, monocytes, platelet (PLT), neutrophil-to-lymphocyte ratio (NLR), derived-NLR (dNLR), monocyte-to-lymphocyte ratio (MLR) and PLT-to-lymphocyte ratio (PLR) were assessed in total 208 CP patients and different age and surgical approach CP patient subgroups. Their predictive values were evaluated by the receiver operator characteristic curve analysis. Results Preoperative peripheral WBC, neutrophils, NLR, dNLR, MLR, and PLR were positively correlated and lymphocyte was negatively associated with postoperative CDI occurrence in CP patients, especially when WBC ≥ 6.66 × 109/L or lymphocyte ≤ 1.86 × 109/L. Meanwhile, multiple logistic regression analysis showed that WBC > 6.39 × 109/L in the > 18 yrs age patients, WBC > 6.88 × 109/L or lymphocytes ≤ 1.85 × 109/L in the transcranial approach patients were closely associated with the elevated incidence of postoperative CDI. Furthermore, the area under the curve obtained from the receiver operator characteristic curve analysis showed that the best predictors of inflammatory markers were the NLR in total CP patients, the MLR in the ≤ 18 yrs age group and the transsphenoidal group, the NLR in the > 18 yrs age group and the dNLR in the transcranial group. Notably, the combination index NLR + dNLR demonstrated the most valuable predictor in all groups. Conclusions Preoperative peripheral inflammatory markers, especially WBC, lymphocytes and NLR + dNLR, are promising predictors of postoperative CDI in CPs.

Accurately predicting freeway accident severity is crucial for accident prevention, road safety, and emergency rescue services in intelligent freeway systems. However, current research lacks the required precision, hindering the effective implementation of freeway rescue. In this paper, we efficiently address this challenge by categorizing influencing factors into two levels: human and non-human, further subdivided into 6 and 36 categories, respectively. Furthermore, based on the above factors, an efficient and accurate Freeway Accident Severity Prediction (FASP) method is developed by using the two-level fuzzy comprehensive evaluation. The factor and evaluation sets are determined by calculating the fuzzy evaluation matrix of a single factor. The weight matrix is calculated through the entropy method to compute the final evaluation matrix. Based on the maximum membership principle, the severity of the freeway accident is predicted. Finally, based on the experiments conducted with the traffic accident datasets in China and the US, it is shown that FASP is able to accurately predict the severity of freeway traffic accidents with thorough considerations and low computational cost. It is noted that FASP is the first attempt to achieve freeway accident severity prediction using the two-level fuzzy comprehensive evaluation method to the best of our knowledge.

In this study, a novel sulfur/zinc co-doped biochar (SZ-BC) stabilizer was successfully developed for the remediation of mercury-contaminated soil. Results from SEM, TEM, FTIR and XRD revealed that biochar (BC) was successfully modified by sulfur and zinc. In the batch adsorption experiments, the sulfur-zinc co-pyrolysis biochar displayed excellent Hg(II) adsorption performance, with the maximum adsorption capacity of SZ-BC (261.074 mg/g) being approximately 16.5 times that of BC (15.855 mg/g). Laboratory-scale static incubation, column leaching, and plant pot experiments were conducted using biochar-based materials. At an additional dosage of 5 % mass ratio, the SZ-BC exhibits the most effective stabilization of mercury in soil, leading to a significant reduction in leaching loss compared to the control group (CK) by 51.30 %. Following 4 weeks of incubation and 2 weeks of leaching with SZ-BC, the residual mercury in the soil increased by 27.84 %. As a result, potential ecological risk index of mercury decreased by 92 % compared to the CK group. In the pot experiment, SZ-BC significantly enhanced the growth of Chinese cabbage, with biomass and root dry weight reaching 3.20 and 2.80 times that of the CK group, respectively. Additionally, the Translocation Factor (TF) and Bioconcentration Factor (BF) were reduced by 44.86 % and 74.43 %, respectively, in the SZ-BC group compared to the CK group. Moreover, SZ-BC can effectively improve enzyme activities and increase microbial communities in mercury-contaminated soils. The mechanisms of adsorption and stabilization were elucidated through electrostatic adsorption, ion exchange, surface complexation, and precipitation. These findings provide a potentially effective material for stabilizing soils contaminated with mercury.

Abstract In online public opinion events, key figures are crucial to the formation and diffusion of public opinion, to the evolution and dissemination of topics, and to the guidance and transformation of the direction of public opinion. Based on the four-dimensional public opinion communication supernetwork (social-psychology-opinion-convergent), this study proposes a classification and recognition algorithm of key figures in online public opinion that integrates multidimensional similarity and K-shell to identify the key figures with differentiation in online public opinion events. The research finds that the evolutionary process of public opinion events is the joint action of key figures with different roles. The opinion leader is the key figure in the global communication of public opinion. The focus figure is the core figure that promotes the dissemination of public opinion on local subnetworks. The communication figure is the “bridge” node in the cross-regional communication of public opinion. Through the algorithm verification of the case “China Eastern Airlines Passenger Plane Crash Event”, we find that the algorithm proposed in this paper has advantages in feasibility, sensitivity, and effectiveness, compared with traditional algorithms such as CI, forwarding volume, degree centrality, K-shell, and multidimensional similarity. The classification and recognition algorithm proposed in this study can not only identify multirole key figures simultaneously but also improve the recognition granularity and eliminate the interference of core-like nodes.

The natural product Desmosdumotin D (hereafter referred to as Des-D), isolated from the plant Desmos dumosus, showed potent anti-HIV activity. However, the subsequent pharmacological activity and clinical studies are limited due to the low content of Des-D in the plant. Therefore, the total synthesis path of Des-D was optimized in this paper, and the total yield was increased from 4.4% to 11.9%. Additionally, twelve analogues were obtained following the synthesis route of Des-D. The anti-HIV activity evaluation results in vitro showed that Des-D had the highest activity, with an IC50 value of 13.6 μM, and compounds 17 and 11 had the lowest anti-HIV activity, with IC50 values of 101.3 μM and 161.0 μM, respectively. Through the molecular docking of compounds Des-D and 17 with HIV-IN, the results show that phenolic hydroxyl groups and two benzene rings interact with HIV-IN and are possible pharmacodynamic groups.

The ice, cloud, and land elevation satellite-2 (ICESat-2) is the only satellite that produces photon-counting light detection and ranging data, and is equipped with the advanced topographic laser altimeter system. ICESat-2 provides sea surface height product; however, its approach of the product is unsuitable for areas with sub-surface signals. Conventional denoising methods applied to sea surface photon data of variable density involve the use of different empirical parameters. Considering the distribution of sea surface signal photons, we propose a general open-source method using a dual-signal unmixing parameter (DSUMP), which incorporates the Gaussian distribution of dual-signal peaks to determine the sea surface range. This method facilitates the direct extraction of sea surface photons under various observation conditions—day or night, strong or weak beams, and including or excluding seabed photons—without requiring any variable parameters. The elevation error by DSUMP within 0.1m accounts for more than 97%. The mean absolute error is within 0.01 m compared to sea surface photons obtained via manual extraction. Different model parameters show stable denoising accuracy, only affects operating efficiency. The proposed method introduces a novel denoising technique for extracting sea surface elevation from ICESat-2 altimetry data, and its applicability can be extended to various point cloud data with similar distributions.

Background:Asarum heterotropoides and Asarum sieboldii are commonly used in traditional Chinese medicine. However, little is known about how they differ in terms of essential oil (EO) and ethanol extract (EE) content and composition. Moreover, the effect of various geographical locations on the essential oil (EO), ethanol extract (EE), and asarinin content of different Asarum samples remains unknown. We tested four root-drying methods, i.e., soil removal and shade drying (P1), water washing and shade drying (P2), and water washing and drying at 30 °C (P3) and 40 °C (P4). We used LC-MS and GC-MS to investigate these differences. We also investigated the pharmacodynamic effects of EO and EE. Results: Overall, the EO, EE and asarinin contents of the analysed samples were 19.21–51.53 μL.g−1, 20.00–45.00 μL.g−1, and 1.268–2.591 mg.g−1, respectively. P1 treatment yielded the lowest volatile oil content compared to the other three treatments. GC-MS analysis revealed 78 EO components. Among the six major EO components, eucarvone, 3,5-dimethoxytoluene, and methyl eugenol were higher in A. heterotropoides than in A. sieboldii. However, the latter had a higher myristicin content. LC-MS analysis identified 888 EE components in roots and leaves of A. heterotropoides and A. sieboldii; 317 differentially accumulated metabolites were identified. EO and EE showed a dose-dependent reduction in the degree of swelling and an increase in the inhibition rate of drug concentration on acetic acid writhing in mice. Asarum EO proved to be more effective than EE in the pharmacodynamic study. Conclusions: We conclude that Asarum species show inter- and intra-specific differences in EO and EE content and composition, which may influence the pharmacodynamics of Asarum root extracts.

Endocrine-disrupting chemicals (EDCs) are ubiquitous in ecological environments and have become a great issue of public health concern since the 1990 s. There is a deep scientific understanding of the toxicity of EDCs. However, recent studies have found that the abnormal physiological functions of the parents caused by EDCs could be transmitted to their unexposed offspring, leading to intergenerational toxicity. We questioned whether sustained epigenetic changes occur through the male germline. In this review, we (1) systematically searched the available research on the intergenerational impacts of EDCs in aquatic and mammal organisms, including 42 articles, (2) summarized the intergenerational genetic effects, such as decreased offspring survival, abnormal reproductive dysfunction, metabolic disorders, and behavioral abnormalities, (3) summarized the mechanisms of intergenerational toxicity through paternal interactions, and (4) propose suggestions on future research directions to develop a deeper understanding of the ecological risk of EDCs.

Summary: LncRNA associated with immune cell infiltration in tumor microenvironment (TME) may be a potential therapeutic target for lung adenocarcinoma. We established a machine learning (ML) model based on 3896 samples characterized by the degree of immune cell infiltration, and further screened the key lncRNA. In vitro experiments were applied to validate the prediction. Treg is the key immune cell in the TME of lung adenocarcinoma, and the degree of infiltration is negatively correlated with the prognosis. PCBP1-AS1 may affect the infiltration of Tregs by regulating the TGF-β pathway, which is a potential predictor of clinical response to immunotherapy. PCBP1-AS1 regulates cell proliferation, cell cycle, invasion, migration, and apoptosis in lung adenocarcinoma. The results of clinical sample staining and in vitro experiments showed that PCBP1-AS1 was negatively correlated with Treg infiltration and TGF-β expression. Tregs and related lncRNA PCBP1-AS1 can be used as targets for the diagnosis and treatment of lung adenocarcinoma.

Abstract Osimertinib is a promising approved third-generation epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI) for treating patients with lung adenocarcinoma (LUAD) harboring EGFR-activating mutations, however, almost all patients develop resistance to Osimertinib eventually limiting the long-term efficacy. Autophagy is a vital cellular recycling process promoting Osimertinib resistance. Identifying accurate and efficient autophagy-regulatory factors is of great significance in reducing Osimertinib resistance. This study identified Cezanne, a member of the ovarian tumor protease (OTU)-deubiquitinating family, as an autophagy regulator. Cezanne was highly expressed in Osimertinib-resistant cells, and Cezanne overexpression promoted Osimertinib resistance, while chloroquine (CQ), an autophagy inhibitor, reverted this process. In the Cezanne-overexpressing cells, autophagy was activated even in the absence of autophagy inducers rapamycin and Earle’s Balanced Salt Solution (EBSS). Further study showed that Cezanne stabilized PIK3C3 by deubiquitinating K48-linked ubiquitination at Lysine 322. Surprisingly, as a compensatory mechanism of PI3P generation, PIK3C2A was shown to be upregulated by Cezanne by promoting its transcription in a POLR2A-dependent way. Based on these results, Cezanne also accelerates EGFR recycling which may explain the mechanism mediating Cezanne expression and Osimertinib resistance. In conclusion, this study establishes a new model connecting Cezanne, autophagy, and Osimertinib resistance, opening new avenues to explore the effect of Cezanne and autophagy in LUAD.

Abstract Plant hormones are important for regulating growth, development, and plant-pathogen interactions. Some of them are inhibitory to growth of fungal pathogens but the underlying mechanism is not clear. In this study, we found that hyphal growth of Fusarium graminearum was significantly reduced by high concentrations of IAA and its metabolically stable analogue 2,4-dichlorophenoxyacetic acid (2,4-D). Besides inhibitory effects on growth rate, treatments with 2,4-D also caused significant reduction in conidiation, conidium germination, and germ tube growth. Treatments with 2,4-D had no obvious effect on sexual reproduction but significantly reduced TRI gene expression, toxisome formation, and DON production. More importantly, treatments with 2,4-D were inhibitory to infection structure formation and pathogenesis at concentrations higher than 100 µM. The presence of 1000 µM 2,4-D almost completely inhibited plant infection and invasive growth. In F. graminearum, 2,4-D induced ROS accumulation and FgHog1 activation but reduced the phosphorylation level of Gpmk1 MAP kinase. Metabolomics analysis showed that the accumulation of a number of metabolites such as glycerol and arabitol was increased by 2,4-D treatment in the wild type but not in the Fghog1 mutant. Transformants expressing the dominant active FgPBS2 S451D T455D allele were less sensitive to 2,4-D and had elevated levels of intracellular glycerol and arabitol induced by 2,4-D in PH-1. Taken together, our results showed that treatments with 2,4-D interfere with two important MAP kinase pathways and are inhibitory to hyphal growth, DON biosynthesis, and plant infection in F. graminearum.

Abstract Aphid infestation poses a significant threat to crop production, rural communities, and global food security. While chemical pest control is crucial for maximizing yields, applying chemicals across entire fields is both environmentally unsustainable and costly. Hence, precise localization and management of aphids are essential for targeted pesticide application. The paper primarily focuses on using deep learning models for detecting aphid clusters. We propose a novel approach for estimating infection levels by detecting aphid clusters. To facilitate this research, we have captured a large-scale dataset from sorghum fields, manually selected 5447 images containing aphids, and annotated each individual aphid cluster within these images. To facilitate the use of machine learning models, we further process the images by cropping them into patches, resulting in a labeled dataset comprising 151,380 image patches. Then, we implemented and compared the performance of four state-of-the-art object detection models (VFNet, GFLV2, PAA, and ATSS) on the aphid dataset. Extensive experimental results show that all models yield stable similar performance in terms of average precision and recall. We then propose to merge close neighboring clusters and remove tiny clusters caused by cropping, and the performance is further boosted by around 17%. The study demonstrates the feasibility of automatically detecting and managing insects using machine learning models. The labeled dataset will be made openly available to the research community.

Abstract Autophagy, as an intracellular degradation system, plays a critical role in plant immunity. However, the involvement of autophagy in the plant immune system and its function in plant nematode resistance are largely unknown. Here, we show that root-knot nematode (RKN; Meloidogyne incognita) infection induces autophagy in tomato (Solanum lycopersicum) and different atg mutants exhibit high sensitivity to RKNs. The jasmonate (JA) signaling negative regulators JASMONATE-ASSOCIATED MYC2-LIKE 1 (JAM1), JAM2 and JAM3 interact with ATG8s via an ATG8-interacting motif (AIM), and JAM1 is degraded by autophagy during RKN infection. JAM1 impairs the formation of a transcriptional activation complex between ETHYLENE RESPONSE FACTOR 1 (ERF1) and MEDIATOR 25 (MED25) and interferes with transcriptional regulation of JA-mediated defense-related genes by ERF1. Furthermore, ERF1 acts in a positive feedback loop and regulates autophagy activity by transcriptionally activating ATG expression in response to RKN infection. Therefore, autophagy promotes JA-mediated defense against RKNs via forming a positive feedback circuit in the degradation of JAMs and transcriptional activation by ERF1.

Deregulation of cell cycles can result in a variety of cancers, including breast cancer (BC). In fact, abnormal regulation of cell cycle pathways is often observed in breast cancer, leading to malignant cell proliferation. CDK4/6 inhibitors (CDK4/6i) can block the G1 cell cycle through the cyclin D-cyclin dependent kinase 4/6-inhibitor of CDK4-retinoblastoma (cyclinD-CDK4/6-INK4-RB) pathway, thus blocking the proliferation of invasive cells, showing great therapeutic potential to inhibit the spread of BC. So far, three FDA-approved drugs have been shown to be effective in the management of advanced hormone receptor positive (HR+) BC: palbociclib, abemaciclib, and ribociclib. The combination strategy of CDK4/6i and endocrine therapy (ET) has become the standard therapeutic regimen and is increasingly applied to advanced BC patients. The present study aims to clarify whether CDK4/6i can also achieve a certain therapeutic effect on Human epidermal growth factor receptor 2 positive (HER2+) BC. Studies of CDK4/6i are not limited to patients with estrogen receptor positive/human epidermal growth factor receptor 2 negative (ER+/HER2-) advanced BC, but have also expanded to other types of BC. Several pre-clinical and clinical trials have demonstrated the potential of CDK4/6i in treating HER2+ BC. Therefore, this review summarizes the current knowledge and recent findings on the use of CDK4/6i in this type of BC, and provides ideas for the discovery of new treatment modalities.

Abstract Existing work generally classifies news headlines as a matter of short text classification. However, due to the strong domain nature and limited text length of news headlines, their classification results are usually determined by several specific keywords, which makes the traditional short text classification method ineffective. In this paper, we propose a new method to identify keywords in news headlines and expand their features from sentence level and word level respectively, and finally use convolutional neural networks (CNN) to extract and classify their features. The proposed model was tested on the Sogou News Corpus dataset and achieved 93.42 $$\%$$ % accuracy.

In spent fuel reprocessing, UO2(DBP)2 (U-DBP) can be deposited in stainless steel equipment. U-DBP must be removed by dissolution and the process must not cause corrosion to stainless steel. This study was conducted to find the best scheme for dissolution. U-DBP was manufactured by the titrimetric sedimentation method. The effects of different factors on the dissolution of U-DBP were investigated. For example, solid-liquid ratio, hydrazine carbonate solutions with different mass components, mixed solutions containing different concentrations of H2O2, and different carbonates. The results indicated that U-DBP does not have a regular crystal morphology. With the increase of the solid-liquid ratio and the mass fraction of hydrazine carbonate, the concentration of U(VI) at the dissolution equilibrium increases gradually. The addition of H2O2 has a great promotion effect on the dissolution. However, when the concentration of H2O2 is greater than 0.5 M, the dissolution solution may have an erosive effect on the stainless steel. (NH4)2CO3 can increase the dissolution capacity of dissolved U-DBP, but it may also accelerate the corrosion of stainless steel.

Aphid infestations are one of the primary causes of extensive damage to wheat and sorghum fields and are one of the most common vectors for plant viruses, resulting in significant agricultural yield losses. To address this problem, farmers often employ the inefficient use of harmful chemical pesticides that have negative health and environmental impacts. As a result, a large amount of pesticide is wasted on areas without significant pest infestation. This brings to attention the urgent need for an intelligent autonomous system that can locate and spray sufficiently large infestations selectively within the complex crop canopies. We have developed a large multi-scale dataset for aphid cluster detection and segmentation, collected from actual sorghum fields and meticulously annotated to include clusters of aphids. Our dataset comprises a total of 54,742 image patches, showcasing a variety of viewpoints, diverse lighting conditions, and multiple scales, highlighting its effectiveness for real-world applications. In this study, we trained and evaluated four real-time semantic segmentation models and three object detection models specifically for aphid cluster segmentation and detection. Considering the balance between accuracy and efficiency, Fast-SCNN delivered the most effective segmentation results, achieving 80.46% mean precision, 81.21% mean recall, and 91.66 frames per second (FPS). For object detection, RT-DETR exhibited the best overall performance with a 61.63% mean average precision (mAP), 92.6% mean recall, and 72.55 on an NVIDIA V100 GPU. Our experiments further indicate that aphid cluster segmentation is more suitable for assessing aphid infestations than using detection models.

The hydrogenation of carbon dioxide (CO2) to produce light olefins is one of the most promising ways to utilize CO2 in power plant flue gas. However, the low concentration of CO2 (~10%) and the existence of water steam in the flue gas pose great challenges for the catalyst design. To address these problems, we introduced a Mg promoter and hydrophobic component into the Fe-based catalyst to improve the CO2 adsorption capacity and weaken the negative effects of water. The yield of light olefins on an optimized multifunctional Fe-based catalyst increased by 37% in low-concentration CO2 hydrogenation with water steam. A variety of characterizations proved that the Mg promoter played critical roles in regulating the adsorption capacity of CO2, increasing the surface electron density of Fe species, and promoting the formation of iron carbide active sites. The hydrophobic component mainly contributed to constraining the oxidation of iron carbides via water steam. It benefited from the rational design of the catalyst, showing how our multifunctional Fe-based catalyst has great potential for practical application in CO2 utilization.

Asarum heterotropoides Fr. var. mandshuricum (Maxim.) is a perennial herb native to China. Its essential oil components and asarinin are health-promoting compounds. However, cultivation under natural light can affect the growth and secondary metabolite yield of this sciophyte. Adjustment of light irradiance may be beneficial in this respect. Here, we applied six types of filtered solar irradiance by using filter films of red (I), yellow (II), blue (III), green (IV), purple (V) and 50% sunlight (VI). We measured net photosynthetic rate (APn), activity of phenylpropanoid biosynthetic enzymes, asarinin content, and essential oil content and composition. Light quality treatments V and VI resulted in increased APn, enzyme activities, asarinin content and essential oil content. Samples harvested in September 2020 had relatively higher contents and enzyme activities compared to those harvested in July 2020. Elemicin, 2,5-dimethoxytoluene and methyleugenol accumulated at higher levels in response to light quality V. Light qualities III and IV showed promising results for several compounds. Overall, we conclude that controlling the light conditions in growing environments promotes the amount of asarinin and essential oil by regulating photosynthesis and the activities of phenylpropanoid biosynthetic enzymes.

Summary: Small nucleolar RNA host genes (SNHGs) are a special family of long non-coding RNAs (lncRNAs), which not only function in a way similar to other lncRNAs but also influence the intracellular level of small nucleolar RNAs to modulate cancers. However, the features of SNHGs and their role in the prognosis and immunotherapeutic response of human cancer have not been explored. We found that SNHGs were commonly deregulated and correlated with patient survival in various cancers. The critical role of DNA methylation and somatic alterations on deregulation was also identified. SNHG family score was significantly associated with survival, multiple tumor characteristics, and tumor microenvironment. SNHG-related risk score could serve as a prognostic and immunotherapeutic response biomarker based on multiple databases. This study emphasizes the potential of SNHGs as biomarkers for prognosis and immunotherapeutic response, enabling further research into the immune regulatory mechanism and therapeutic potentials of SNHGs in cancer.

This article focuses on the mapping and rate of spread (ROS) measurement of grass fires using near infrared (NIR) images acquired by a small fixed-wing unmanned aircraft system (UAS) operating at low altitudes. A new method is proposed for spatiotemporal representation of grass fire evolution using time labeled UAS NIR orthomosaics stitched from aerial images collected at varying time stamps over different regions of fire. Furthermore, a novel NIR intensity variance thresholding method is proposed for accurate identification and delineation of grass fire fronts based on the obtained NIR mosaics in digital numbers. The proposed methods are demonstrated and validated using UAS NIR imagery acquired over a prescribed tallgrass fire in Kansas (around 13 ha.). Three NIR short time-series orthomosaics are generated at a time interval of about 2 min with a spatial registration accuracy of 1.45 m (RMSE). The mean ROS for head, flank, and back tallgrass fires are measured to be 0.28, 0.1, and 0.025 m/s.

Abstract The design of rigid vortex generators (RVG) influences the thermal performance of various technologies. We employed Discrete Adjoint-Based Optimization to show the optimal development of vortex generators. Under turbulent flow conditions, different bi-objective functions on the RVG design were examined. Specifically, we aimed at an optimal RVG shape that minimizes the pressure drop and maximizes the local heat transfer in a rectangular channel. We show that an optimal design of an RVG can be obtained using computational fluid dynamics in conjunction with the Pareto Front at a computational cost of the order ~ $$O(10^{-1})$$ O ( 10 - 1 ) . We obtained three essential vortex generator shapes based on the RVG morphing technique. Compared to the baseline geometry of a delta winglet pair DWP, the first morphed design reduced the pressure drop by $$39\%$$ 39 % , however, at the expense of a $$21\%$$ 21 % reduction in the Nusselt number. The second vortex generator design enhanced the heat transfer by $$18\%$$ 18 % , however, at the cost of a significant increase in pressure drop of about $$40\%$$ 40 % . The final morphed design achieved the highest thermal performance factor of 1.28, representing a heat transfer enhancement of $$6\%$$ 6 % with a moderate increase in pressure drop of about $$13\%$$ 13 % compared to DWP vortex generators. Furthermore, we investigated the effect of introducing different size holes on the mass reduction of vortex generators and their thermal performances. The mass of vortex generators can be reduced by $$9\%$$ 9 % and with an increase of $$7\%$$ 7 % in thermal performance factor concerning the DWP baseline. The findings of this study will lead to highly efficient lightweight heat exchangers.

Abstract Generated by the immune system post-infection or through vaccination, the effectiveness of antibodies against emerging SARS-CoV-2 variants is crucial for protecting individuals from the COVID-19 pandemic. Herein, a platform for the multiplexed evaluation of SARS-CoV-2 neutralizing antibodies against various variants was designed on the basis of near-infrared (NIR) surface enhanced fluorescence by nano-plasmonic gold chip (pGOLD). Antibody level across variants (Wild-type, Alpha, Beta, Delta, Omicron) was confirmed by the sera from recovered-individuals who were unvaccinated and had infected with Wild-type, Delta, Omicron variants. However, the neutralizing activity against Omicron variant was markedly decreased for individuals infected by Wild-type (~ 5.6-fold) and Delta variant (~ 19.1-fold). To the opposite, neutralizing antibody from individuals recovered from Omicron variant infection showed weak binding strength against non-Omicron variants. Antibody evolution over time was studied with individuals 196–530 days post Wild-type infection. Decreasing IgG antibody titer accompanied by increasing IgG binding avidity with elongated post-infection period were observed for the sera from Wild-type recovered-individuals with different post-infection times, suggesting that after the primary infection, a great number of antibodies were generated and then gradually decreased, while the antibody matured over time. By comparing the IgG level of individuals vaccinated for 27–51 days with individual post-infection, we found that ca. 1 month after two doses of vaccination, the antibody level was comparable to that of 500 days post-infection, and vaccination could enhance IgG avidity more efficiently. This work demonstrated a platform for the multiplexed, high-throughput and rapid screening of acquired immunity against SARS-CoV-2 variants, providing a new approach for the analysis of vaccine effectiveness, immunity against emerging variants, and related serological study. Abstract Fusarium head blight (FHB), mainly caused by Fusarium graminearum, is one of the most devastating diseases in wheat and barley worldwide. In addition to causing severe yield losses, F. graminearum produces deoxynivalenol (DON), a trichothecene mycotoxin which is harmful to human health and serves as an important virulence factor. Currently, changes in global climate and tillage systems have made FHB epidemics more frequent and severe. During the past decade, considerable efforts have been deployed to reveal the pathogenic mechanisms of F. graminearum, identify resistance genes in wheat, and breed FHB-resistant varieties. In this review, we highlight recent advances in FHB pathogenesis, F. graminearum-wheat interaction, and wheat defense mechanisms. This review contains four main sections: (1) signal sensing and transduction associated with the pathogenesis of F. graminearum; (2) regulation and functions of DON during wheat infection; (3) roles of F. graminearum-secreted enzymes and effectors in facilitating pathogen infection of wheat; (4) wheat components involved in interactions with F. graminearum. A comprehensive understanding of the molecular interactions between F. graminearum and wheat will contribute to the development of novel and efficient strategies for controlling FHB.

Seed placement information for evaluating planters is obtained by tedious manual methods like using a pogostick or a ruler. To obtain more information and reduce the time required. There is a need for an automated system. The goal of this study is to design such a system with the use of GPS and seed detection.A 12-row planter was instrumented with cameras and a GPS. The planter was operated at planting speeds of 9.66 kmph and 12.87 kmph with a seed population of 74,131 and 86,4868 seeds per hectare respectively and seed spacing was measured manually using a ruler. YOLOR-P6, YOLOR-CSPX, YOLOX-S, YOLOX-M, YOLOX-L, YOLOX-TINY, and YOLOV4 were trained to detect seeds on a different dataset. An algorithm was designed to estimate the distance between two consecutive seeds by filtering out old detections from the detector with the use of the IOU metric and GPS stream.The seed spacing estimator was evaluated on Jensen–Shannon Divergence (JSD), mean, standard deviation, ΔCount, and RMS metrics. Results indicate that the developed system along with the algorithm was able to perform better at 9.66 kmph with JSD distances of 0.223 and 0.244 (74,131 and 86,4868 seeds per hectare respectively) compared to 0.298 and 0.258 (74,131 and 86,4868 seeds per hectare respectively) at 12.87 kmph. Moreover, this algorithm was able to reduce the total time taken to detect seed and estimate seed spacing information from 2hrs by manual method to 1 min 14 seconds using YOLOR-CSPX.

Summary: The lateral habenula (LHb) has been considered a moderator of social behaviors. However, it remains unknown how LHb regulates social interaction. Here, we show that the hydroxymethylase Tet2 is highly expressed in the LHb. Tet2 conditional knockout (cKO) mice exhibit impaired social preference; however, replenishing Tet2 in the LHb rescues social preference impairment in Tet2 cKO mice. Tet2 cKO alters DNA hydroxymethylation (5hmC) modifications in genes that are related to neuronal functions, as is confirmed by miniature two-photon microscopy data. Further, Tet2 knockdown in the glutamatergic neurons of LHb causes impaired social behaviors, but the inhibition of glutamatergic excitability restores social preference. Mechanistically, we identify that Tet2 deficiency reduces 5hmC modifications on the Sh3rf2 promoter and Sh3rf2 mRNA expression. Interestingly, Sh3rf2 overexpression in the LHb rescues social preference in Tet2 cKO mice. Therefore, Tet2 in the LHb may be a potential therapeutic target for social behavior deficit-related disorders such as autism.

The vegetable plug seedling plays an important role in improving vegetable production. The process of plug seedling contributes to high-quality vegetable seedlings. The substrate composition and chemical fertilizer are widely studied to promote seedling growth. However, little is known about the effect of beneficial bacteria in the rhizosphere microbial community and vegetables’ growth during plug seedling. The use of beneficial microbes to promote vegetable seedling growth is of great potential. In this study, we showed that the Serratia marcescens strain LYGN1 enhanced the growth of cucumber and pepper seedlings in plug seedling cultivation. The treatment with LYGN1 significantly increased the biomass and the growth-related index of cucumber and pepper, improving the seedling quality index. Specifically, LYGN1 also improved the cucumber and pepper root system architecture and increased the root diameter. We applied high-throughput sequencing to analyze the microbial community of the seedlings’ rhizosphere, which showed LYGN1 to significantly change the composition and structure of the cucumber and pepper rhizosphere microbial communities. The correlation analysis showed that the Abditibacteriota and Bdellovibrionota had positive effects on seedling growth. The findings of this study provide evidence for the effects of Serratia marcescens LYGN1 on the cucumber and pepper rhizosphere microbial communities, which also promoted seedling quality in plug seedling cultivation.

Tremor is an involuntary, rhythmic movement disorder. Despite its prevalence, the underlying pathophysiology remains poorly understood, and effective treatment options are limited. Animal models are essential in enhancing our understanding of the mechanisms of tremorogenesis and developing new therapeutic interventions. Although tremor is amenable to measurement by automated systems, visual observation is still the most prevalent method for recording tremor in animal studies. This review gives a brief summary of two behavioral methods that enable quantitative measurement of forelimb tremor (the press-while-licking task) and whole-body tremor (the force-plate actometer) in rodents. These methods utilize force transducer and computing technologies to generate high-resolution force-time waveforms for automated detection and characterization of tremor. The focus will be on the sensitive, precise, and quantitative measurement of tremors induced in rodents by low-dose pharmacological agents, brain lesion, physical training, and genetic mutations. The methods reviewed here provide new tools that can facilitate preclinical assessment of treatment strategies for tremor.

BackgroundSystematic reviews and meta-analyses based on observational studies have shown voluntary medical male circumcision (VMMC) may reduce HIV risk among men who have sex with men (MSM). There is a lack of randomized controlled trial (RCT) data assessing the efficacy of VMMC. ObjectiveThe primary objective of this study is to assess the efficacy of VMMC for preventing HIV acquisition among MSM who primarily engage in insertive anal sex. MethodsA multicenter RCT will be conducted among MSM in 8 cities in China. Eligible participants are men aged 18-49 years who self-report ≥2 male sex partners in the past 6 months, predominantly practice insertive anal sex, and are willing to undergo circumcision. Interested men who satisfy inclusion criteria will be tested for HIV 1 month before enrollment and at enrollment, and only those who are HIV negative will be enrolled. At baseline, all enrolled participants will be asked to report sociodemographic characteristics and sexual behaviors; provide a blood sample for HIV, syphilis, and herpes simplex virus type 2 testing; and provide a penile swab for human papillomavirus testing. Participants will be randomly assigned to the intervention or control group. Those in the intervention group will receive VMMC and undergo a web-based weekly follow-up assessment of postsurgery healing for 6 consecutive weeks. All participants will be tested for HIV at 3-, 6-, 9-, and 12-month follow-ups. All participants will also be asked to report sexual behaviors and undergo repeat herpes simplex virus type 2 and human papillomavirus testing at 6- and 12-month follow-ups. The primary end point is HIV seroconversion. Secondary end points are the safety and satisfaction with VMMC and the changes in sexual behaviors after VMMC. The grouped censored data will be analyzed by intention-to-treat approach. ResultsRecruitment for the RCT began in August 2020 and continued through July 2022. Data collection is expected to be completed by July 2023, and full data analysis is going to be completed by September 2023. ConclusionsThis study will be the first RCT to assess the efficacy of VMMC in preventing HIV infection among MSM. Results from this trial will provide preliminary evidence for the potential efficacy of VMMC to reduce incident HIV infection among MSM. Pre-driven longwall retracement roadway (PLRR) is commonly used in large mine shaft. The support crushing disasters occur frequently during the retracement, and roof management is necessary. Taking the 31107 panel as research background, the roof breaking structure of PLRR is analyzed. It is concluded that the roof cutting with vertical hydraulic fracture (HF) at a specified position, that is, fixed-length roof cutting, can reduce support load and keep immediate roof intact. The extended finite element method (XFEM) is applied to simulate hydraulic fracturing. The results show that both the axial and transverse hydraulic fracturing cannot effectively create vertical HFs. Therefore, a novel construction method of vertical HF based on the stress shadow effect (SSE) is proposed. The stress reversal region and HF orientation caused by the prefabricated hydraulic fracture (PF) are verified in simulation. The sub-vertical HFs are obtained between two PFs, the vertical extension range of which is much larger than that of directional hydraulic fracturing. The new construction method was used to determine the field plan for fixed-length roof cutting. The roof formed a stable suspended structure and deformation of the main PLRR was improved after hydraulic fracturing.

Iso-nitrogenous and iso-lipidic diets containing 0%, 3%, 6%, 9%, and 12% hydrolyzed porcine mucosa (namely, HPM0, HPM3, HPM6, HPM9, and HPM12) were prepared to evaluate their effects on the growth performance, muscle nutrition composition, texture property, and gene expression related to muscle growth of hybrid groupers (Epinephelus fuscoguttatus ♀ × Epinephelus lanceolatus ♂). Groupers were fed to apparent satiation at 08:00 and 16:00 every day for a total of 56 days. It was found that the weight gain percentage in the HPM0, HPM3, and HPM6 groups did not differ (P > 0.05). Triple-negative breast cancer (TNBC) is one of the most lethal subtypes of breast cancer (BC), and it accounts for approximately 10%–20% of all invasive BCs diagnosed worldwide. The survival rate of TNBC in stages III and IV is very low, and a large number of patients are diagnosed in these stages. Therefore, the purpose of this study was to identify TNBC-causing molecular signatures and anti-TNBC drug agents for early diagnosis and therapies. Five microarray datasets that contained 304 TNBC and 109 control samples were collected from the Gene Expression Omnibus (GEO) database, and RNA-Seq data with 116 tumor and 124 normal samples were collected from TCGA database to identify differentially expressed genes (DEGs) between TNBC and control samples. A total of 64 DEGs were identified, of which 29 were upregulated and 35 were downregulated, by using the statistical limma R-package. Among them, seven key genes (KGs) were commonly selected from microarray and RNA-Seq data based on the high degree of connectivity through PPI (protein–protein interaction) and module analysis. Out of these seven KGs, six KGs (TOP2A, BIRC5, AURKB, ACTB, ASPM, and BUB1B) were upregulated and one (EGFR) was downregulated. We also investigated their differential expression patterns with different subtypes and progression stages of BC by the independent datasets of RNA-seq profiles from UALCAN database, which indicated that they may be potential biomarkers for early diagnosis. Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses with the proposed DEGs were performed using the online Enrichr database to investigate the pathogenetic processes of TNBC highlighting KGs. Then, we performed gene regulatory network analysis and identified three transcriptional (SOX2, E2F4, and KDM5B) and three post-transcriptional (hsa-mir-1-3p, hsa-mir-124-3p, and hsa-mir-34a-5p) regulators of KGs. Finally, we proposed five KG-guided repurposable drug molecules (imatinib, regorafenib, pazopanib, teniposide, and dexrazoxane) for TNBC through network pharmacology and molecular docking analyses. These drug molecules also showed significant binding performance with some cancer-related PTM-sites (phosphorylation, succinylation, and ubiquitination) of top-ranked four key proteins (EGFR, AURKB, BIRC5, and TOP2A). Therefore, the findings of this computational study may play a vital role in early diagnosis and therapies against TNBC by wet-lab validation.

With the development of technologies, multiple primary lung cancer (MPLC) has been detected more frequently. Although large-scale genomics studies have made significant progress, the aberrant gene mutation in MPLC is largely unclear. In this study, 141 and 44 lesions from single and multiple primary lung adenocarcinoma (SP- and MP-LUAD) were analyzed. DNA and RNA were extracted from formalin-fixed, paraffin-embedded tumor tissue and sequenced by using the next-generation sequencing-based YuanSu450TM gene panel. We systematically analyzed the clinical features and gene mutations of these lesions, and found that there were six genes differently mutated in MP-LUAD and SP-LUAD lesions, including RBM10, CDK4, ATRX, NTRK1, PREX2, SS18. Data from the cBioPortal database indicated that mutation of these genes was related to some clinical characteristics, such as TMB, tumor type, et al. Besides, heterogeneity analysis suggested that different lesions could be tracked back to monophyletic relationships. We compared the mutation landscape of MP-LUAD and SP-LUAD and identified six differentially mutated genes (RBM10, CDK4, ATRX, NTRK1, PREX2, SS18), and certain SNV loci in TP53 and EGFR which might play key roles in lineage decomposition in multifocal samples. These findings may provide insight into personalized prognosis prediction and new therapies for MP-LUAD patients.

In this paper, underwater dissimilar aluminum alloy laser cladding has been investigated for the first time through a laser nozzle. The influences of water environment on laser cladding of dissimilar aluminum alloy were researched under optimized process parameters. With the transfer of cladding environment from air to water, the height of cladding zone (CZ) and the cladding angle increased, but the width of CZ and the depth of melting zone (MZ) decreased. The microstructure of MZ and CZ were columnar dendrites and equiaxial dendrites, but the underwater microstructure was much smaller than that of the in-air. Both underwater and in-air CZ were composed of Aluminum phase and Al0.5Fe3Si0.5 phase, while the formation of intermetallic compound increased in the in-air CZ. The water environment was beneficial to reduce the magnesium burning loss in the CZ. In the underwater CZ, although the decrease of hard phase compound (Al0.5Fe3Si0.5) formation made microhardness value go down, the microhardness value went up with the increase of magnesium content and the refinement of microstructure. Finally, under the combined action of these three factors, the average microhardness value of underwater cladding layer was higher than that of the in-air cladding layer.

Accurate identification of molecular targets of disease plays an important role in diagnosis, prognosis, and therapies. Breast cancer (BC) is one of the most common malignant cancers in women worldwide. Thus, the objective of this study was to accurately identify a set of molecular targets and small molecular drugs that might be effective for BC diagnosis, prognosis, and therapies, by using existing bioinformatics and network-based approaches. Nine gene expression profiles (GSE54002, GSE29431, GSE124646, GSE42568, GSE45827, GSE10810, GSE65216, GSE36295, and GSE109169) collected from the Gene Expression Omnibus (GEO) database were used for bioinformatics analysis in this study. Two packages, LIMMA and clusterProfiler, in R were used to identify overlapping differential expressed genes (oDEGs) and significant GO and KEGG enrichment terms. We constructed a PPI (protein–protein interaction) network through the STRING database and identified eight key genes (KGs) EGFR, FN1, EZH2, MET, CDK1, AURKA, TOP2A, and BIRC5 by using six topological measures, betweenness, closeness, eccentricity, degree, MCC, and MNC, in the Analyze Network tool in Cytoscape. Three online databases GSCALite, Network Analyst, and GEPIA were used to analyze drug enrichment, regulatory interaction networks, and gene expression levels of KGs. We checked the prognostic power of KGs through the prediction model using the popular machine learning algorithm support vector machine (SVM). We suggested four TFs (TP63, MYC, SOX2, and KDM5B) and four miRNAs (hsa-mir-16-5p, hsa-mir-34a-5p, hsa-mir-1-3p, and hsa-mir-23b-3p) as key transcriptional and posttranscriptional regulators of KGs. Finally, we proposed 16 candidate repurposing drugs YM201636, masitinib, SB590885, GSK1070916, GSK2126458, ZSTK474, dasatinib, fedratinib, dabrafenib, methotrexate, trametinib, tubastatin A, BIX02189, CP466722, afatinib, and belinostat for BC through molecular docking analysis. Using BC cell lines, we validated that masitinib inhibits the mTOR signaling pathway and induces apoptotic cell death. Therefore, the proposed results might play an effective role in the treatment of BC patients.

Irrigated agriculture is indispensable to the Lower Mekong River Basin (LMB), which ensures food security and provides livelihoods for tens of millions of people. Irrigation, agricultural production, hydropower and aquatic ecosystem health are intertwined in LMB, so it is necessary to adopt a holistic approach to analyze irrigation problems. Here, we discuss the challenges and opportunities of LMB irrigation. Bibliometric analysis is carried out to determine the characteristics and patterns of watershed irrigation literature, such as the importance of authors, affiliated institutions, and their distribution in China. Based on bibliometric analysis, research topics are determined for thematic review. Firstly, we investigated the factors that directly affect the demand and supply of irrigation water and associated crop yield impacts. Secondly, we analyzed the influence of water availability, land use and climate change on agricultural irrigation. Thirdly, we analyzed the adverse effects of improper irrigation management on the environment, such as flow pattern change, ecosystem deterioration and land subsidence caused by groundwater overexploitation. Fourthly, the time–space mismatch between water supply and demand has brought serious challenges to the comprehensive water resources management in cross-border river basins. In each specific application area, we sorted out the technologies in which remote sensing technology is used. We hope that this review will contribute to in-depth research and decision analysis of remote sensing technology in agricultural irrigation.

Abstract Parkinson’s disease (PD), one of the most common neurodegenerative disorders, is characterized by progressive neurodegeneration of dopaminergic (DA) neurons in the substantia nigra pars compacta (SNpc). DJ-1 acts essential roles in neuronal protection and anti-neuroinflammatory response, and its loss of function is tightly associated with a familial recessive form of PD. However, the molecular mechanism of DJ-1 involved in neuroinflammation is largely unclear. Here, we found that wild-type DJ-1, rather than the pathogenic L166P mutant DJ-1, directly binds to the subunit p65 of nuclear factor-κB (NF-κB) in the cytoplasm, and loss of DJ-1 promotes p65 nuclear translocation by facilitating the dissociation between p65 and NF-κB inhibitor α (IκBα). DJ-1 knockout (DJ-1 −/−) mice exhibit more microglial activation compared with wild-type littermate controls, especially in response to lipopolysaccharide (LPS) treatment. In cellular models, knockdown of DJ-1 significantly upregulates the gene expression and increases the release of LPS-treated inflammatory cytokines in primary microglia and BV2 cells. Furthermore, DJ-1 deficiency in microglia significantly enhances the neuronal toxicity in response to LPS stimulus. In addition, pharmacological blockage of NF-κB nuclear translocation by SN-50 prevents microglial activation and alleviates the damage of DA neurons induced by microglial DJ-1 deficiency in vivo and in vitro. Thus, our data illustrate a novel mechanism by which DJ-1 facilitates the interaction between IκBα and p65 by binding to p65 in microglia, and thus repressing microglial activation and exhibiting the protection of DA neurons from neuroinflammation-mediated injury in PD.

Abstract Background Interferon regulatory factor-1 (IRF1) affects the proliferation of colorectal cancer (CRC). Recombinant interferon inducible protein 35 (IFI35) participates in immune regulation and cell proliferation. The aim of the study was to examine whether IRF1 affects the radiation sensitivity of CRC by regulating IFI35. Methods CCL244 and SW480 cells were divided into five groups: blank control, IFI35 upregulation, IFI35 upregulation control, IFI35 downregulation, and IFI35 downregulation control. All groups were treated with X-rays (6 Gy). IFI35 activation by IRF1 was detected by luciferase reporter assay. The GEPIA database was used to examine IRF1 and IFI35 in CRC. The cells were characterized using CCK-8, EdU, cell cycle, clone formation, flow cytometry, reactive oxygen species (ROS), and mitochondrial membrane potential. Acute ischemic stroke (AIS) is a primary cause of mortality and morbidity worldwide. Currently, no clinically approved immune intervention is available for AIS treatment, partly due to the lack of relevant patient classification based on the peripheral immunity status of patients with AIS. In this study, we adopted the consensus clustering approach to classify patients with AIS into molecular subgroups based on the transcriptomic profiles of peripheral blood, and we identified three distinct AIS molecular subgroups and 8 modules in each subgroup by the weighted gene co-expression network analysis. Remarkably, the pre-ranked gene set enrichment analysis revealed that the co-expression modules with subgroup I-specific signature genes significantly overlapped with the differentially expressed genes in AIS patients with hemorrhagic transformation (HT). With respect to subgroup II, exclusively male patients with decreased proteasome activity were identified. Intriguingly, the majority of subgroup III was composed of female patients who showed a comparatively lower level of AIS-induced immunosuppression (AIIS). In addition, we discovered a non-linear relationship between female age and subgroup-specific gene expression, suggesting a gender- and age-dependent alteration of peripheral immunity. Taken together, our novel AIS classification approach could facilitate immunomodulatory therapies, including the administration of gender-specific therapeutics, and attenuation of the risk of HT and AIIS after ischemic stroke.

Pyroptosis, defined as programmed cell death, results in the release of inflammatory mediators. Recent studies have revealed that pyroptosis plays essential roles in antitumor immunity and immunotherapy efficacy. Long noncoding RNAs (lncRNAs) are involved in a variety of biological behaviors in tumor cells, although the roles and mechanisms of lncRNAs in pyroptosis are rarely studied. Our study aimed to establish a novel pyroptosis-related lncRNA signature as a forecasting tool for predicting prognosis and ascertaining immune value. Based on lung adenocarcinoma (LUAD) patients from The Cancer Genome Atlas (TCGA), we performed Pearson’s correlation analysis to identify pyroptosis-related lncRNAs. After differentially expressed gene analysis and univariate Cox regression analysis, we selected prognosis-related and differentially expressed lncRNAs. Finally, we performed multivariate Cox regression analysis to establish the three pyroptosis-related lncRNA signature. Kaplan–Meier (KM) survival analyses and receiver operating characteristic (ROC) curves indicated the excellent performance for predicting the prognosis of LUAD patients. At the same time, we applied multidimensional approaches to further explore the functional enrichment, tumor microenvironment (TME) landscape, and immunotherapy efficacy among the different risk groups. A nomogram was constructed by integrating risk scores and clinical characteristics, which was validated using calibrations and ROC curves. Three lncRNAs, namely, AC090559.1, AC034102.8, and AC026355.2, were involved in this signature and used to classify LUAD patients into low- and high-risk groups. Overall survival time (OS) was higher in the low-risk group than in the high-risk group, which was also validated in our LUAD cohort from Shandong Provincial Hospital. TME landscape analyses revealed that a higher abundance of infiltrating immune cells and a greater prevalence of immune-related events existed in the low-risk group. Meanwhile, higher expression of immune checkpoint (ICP) genes, higher immunophenoscore (IPSs), and greater T cell dysfunction in the low-risk group demonstrated a better response to immunotherapy than the high-risk group. Combined with predictions from the Tumor Immune Dysfunction and Exclusion (TIDE) website, we found that LUAD patients in the low-risk group significantly benefited from programmed cell death 1 (PD-1) and cytotoxic T-lymphocyte–associated protein 4 (CTLA4) immune checkpoint blockade (ICB) therapy compared with those in the high-risk group. Furthermore, drug susceptibility analysis identified potential sensitive chemotherapeutic drugs for each risk group. In this study, a novel three pyroptosis-related lncRNA signature was constructed, which could accurately predict the immunotherapy efficacy and prognosis in LUAD patients.

BackgroundAt present, fish meal (FM) resources are in short supply, and competition for food between humans and animals is becoming increasingly critical. Finding non-grain protein sources that can replace FM is the key to solving the rapid development of aquaculture.MethodsSeven trial diets were prepared with 0 g/kg (EP0), 30 g/kg (EP3), 60 g/kg (EP6), 90 g/kg (EP9), 120 g/kg (EP12), 150 g/kg (EP15), and 180 g/kg (EP18) of enzyme-digested poultry by-product meal (EPBM) by replacing of FM. A total of 630 hybrid groupers (Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂) were equally portioned into 21 tanks. At 8:00 and 16:00 each day, groupers were fed until they were full for a cumulative period of 8 weeks.ResultsThe results showed that 30 g/kg of EPBM significantly increased the rates of weight gain and special growth (P < 0.05). Significantly higher activities of serum glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, catalase, and superoxide dismutase were observed in the EP3 group (P < 0.05). The categories and numbers of the top 10 dominant bacteria in the phylum and genus levels were not significantly influenced by feed (P > 0.05). In the proximal intestine and distal intestine, there were significantly higher expressions of SNAT3, LAAT1, CAT2, and CAT1 in the EP3 group compared with the EP0 group (P < 0.05). In the EP3 group, the expressions of PepT1, LAAT1, B0, +AT, and CAT2 were significantly increased in MI than those in all other groups (except the EP0 group, P < 0.05).ConclusionWhen FM was replaced by 30 g/kg of EPBM, growth performance, antioxidant capacity, and the ability to transport amino acids and peptides of hybrid grouper were significantly improved.

Cholangiocarcinoma (CHOL) is highly malignant and has a poor prognosis. This study is committed to creating a new prognostic model based on hypoxia related genes. Here, we established a novel tumor hypoxia-related prognostic model consisting of 6 hypoxia-related genes by univariate Cox regression and the least absolute shrinkage and selection operator (LASSO) algorithm to predict CHOL prognosis and then the risk score for each patient was calculated. The results showed that the patients with high-risk scores had poor prognosis compared with those with low-risk scores, which was verified as an independent predictor by multivariate analysis. The hypoxia-related prognostic model was validated in both TCGA and GEO cohorts and exhibited excellent performance in predicting overall survival in CHOL. The PPI results suggested that hypoxia-related genes involved in the model may play a central role in regulating the hypoxic state. In addition, the presence of IDH1 mutations in the high-risk group was high, and GSEA results showed that some metabolic pathways were upregulated, but immune response processes were generally downregulated. These factors may be potential reasons for the high-risk group with worse prognosis. The analysis of different immune regulation-related processes in the high- and low-risk groups revealed that the expression of genes related to immune checkpoints would show differences between these two groups. We further verified the expression of the oncogene PPFIA4 in the model, and found that compared with normal samples, CHOL patients were generally highly expressed, and the patients with high-expression of PPFIA4 had a poor prognosis. In summary, the present study may provide a valid prognostic model for bile duct cancer to inform better clinical management of patients.

Background: Lung cancer is the most common cause of cancer-related deaths worldwide. LKB1-mutant lung adenocarcinoma (LUAD) is a unique subtype of this deadly cancer. LKB1 mutations cause functional changes in a variety of cell processes, including immune functions, that affect prognosis. To date, the potential role of immunity in the prognosis of LKB1-mutant LUAD is not well understood.Methods: We systematically analyzed immune-related genes in LUAD samples from The Cancer Genome Atlas (TCGA) database. ESTIMATE and CIBERSORT algorithms were used to explore the immune microenvironment. A prognostic risk model was constructed, and prognostic, immune function, drug sensitivity, and model specificity analyses were performed to identify the effectiveness of the model.Results: Our results showed that LKB1 mutations suppressed immune function in LUAD. A three-gene signature was constructed to stratify patients into two risk groups. The risk score was an independent predictor for overall survival (OS) in multivariate Cox regression analyses [hazard ratio (HR) > 1, p = 0.002]. Receiver operating characteristic (ROC) curve analyses confirmed that the risk score has better performance than clinicopathological characteristics. Functional analysis revealed that the immune status was different between the risk groups. ZM.447439 was an appropriate treatment for the high-risk group of patients. This risk model is only suitable for LKB1-mutant tumors; it performed poorly in LUAD patients with wild-type LKB1.Conclusion: Our findings indicate the potential role of immunity in LKB1-mutant LUAD, providing novel insights into prognosis and guiding effective immunotherapy.

Malignant glioma is the most common solid tumor of the adult brain, with high lethality and poor prognosis. Hence, identifying novel and reliable biomarkers can be advantageous for diagnosing and treating glioma. Several galectins encoded by LGALS genes have recently been reported to participate in the development and progression of various tumors; however, their detailed role in glioma progression remains unclear. Herein, we analyzed the expression and survival curves of all LGALS across 2,217 patients with glioma using The Cancer Genome Atlas (TCGA), Chinese Glioma Genome Atlas (CGGA), and Rembrandt databases. By performing multivariate Cox analysis, we built a survival model containing LGALS1, LGALS3, LGALS3BP, LGALS8, and LGALS9 using TCGA database. The prognostic power of this panel was assessed using CGGA and Rembrandt datasets. ESTIMATE and CIBERSORT algorithms confirmed that patients in high-risk groups exhibited significant stromal and immune cell infiltration, immunosuppression, mesenchymal subtype, and isocitrate dehydrogenase 1 (IDH1) wild type. Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), CancerSEA, and Gene Set Enrichment Analysis (GSEA) showed that pathways related to hypoxia, epithelial-to-mesenchymal transition (EMT), stemness, and inflammation were enriched in the high-risk group. To further elucidate the function of LGALS in glioma, we performed immunohistochemical staining of tissue microarrays (TMAs), Western blotting, and cell viability, sphere formation, and limiting dilution assays following lentiviral short hairpin RNA (shRNA)-mediated LGALS knockdown. We observed that LGALS expression was upregulated in gliomas at both protein and mRNA levels. LGALS could promote the stemness maintenance of glioma stem cells (GSCs) and positively correlate with M2-tumor-associated macrophages (TAMs) infiltration. In conclusion, we established a reliable survival model for patients with glioma based on LGALS expression and revealed the essential roles of LGALS genes in tumor growth, immunosuppression, stemness maintenance, pro-neural to mesenchymal transition, and hypoxia in glioma.

ObjectivePrimary ventricular lymphoma (PVL) is an extremely rare and commonly misdiagnosed disease. Previous studies were predominantly case reports, and literature regarding the diagnosis and treatment of PVL is limited. Therefore, this study aimed to evaluate the characteristics of patients with PVL.MethodsThe data of patients with pathologically confirmed PVL were assessed. Epidemiological data, imaging findings, surgery, pathological results, and prognosis were retrospectively analyzed. A systematic review of relevant literature was also conducted.ResultsA total of eight patients with PVL were identified. The main symptom was increased intracranial pressure. Radiographically, five patients had single lesion and three had multiple lesions; typical findings on magnetic resonance imaging included hypointensity on T1- and T2-weighted imaging, adjacent brain edema, and homogeneous enhancement on contrast-enhanced T1-weighted images. Preoperatively, six cases were misdiagnosed and two cases did not get a definite diagnosis. Craniotomy was performed on all patients, and four achieved gross total resection. Hydrocephalus was relieved after surgical resection in four patients. Pathology revealed diffuse large B-cell lymphoma in all patients. Only one patient had a severe complication. A total of three patients received concomitant adjuvant treatment, whereas five patients refused any adjuvant therapy. At the time of follow-up, the median survival time of patients was 15 months.ConclusionPrimary ventricular lymphoma mainly presented with symptoms of increased intracranial pressure and had several imaging characteristics for the diagnosis, but the condition still tends to be misdiagnosed. Surgical resection is a feasible treatment for patients with isolated nodules, especially those with acute obstructive hydrocephalus.
