Ge‐rich Ge–Sb–Te alloy is a good candidate for future automotive applications due to its high crystallization temperature, which allows good data retention at elevated temperatures. Crystallization in this material is governed by elemental segregation which is key to thermal stability and device performance. In this work, elemental (Ge, Sb, Te) segregation is studied in situ during thermal annealing of Ge‐rich Ge–Sb–Te thin films using X‐ray fluorescence microscopy at ID16B beamline of ESRF with a beam size of 50 nm. Spatially resolved maps of Ge, Te, and Sb fluorescence yield are monitored and statistically analyzed as a function of temperature/time. In all investigated samples Sb appears to segregate much less than Te and Ge, indicating a lower mobility of this element. In situ, fluorescence mapping of samples doped with different amounts of carbon by ion implantation shows that carbon delays Ge and Te segregation to higher temperatures. Comparison with crystallization kinetics monitored by X‐ray diffraction shows a good correlation between the occurrence of spatially resolved chemical inhomogeneities and the appearance of crystallized phases. SUMMARY: Maintaining an optimal redox status is essential for plant growth and development, particularly when the plants are under stress. AT‐hook motif nuclear localized (AHL) proteins are evolutionarily conserved transcription factors in plants. Much of our understanding about this gene family has been derived from studies on clade A members. To elucidate the functions of clade B genes, we first analyzed their spatial expression patterns using transgenic plants expressing a nuclear localized GFP under the control of their promoter sequences. AHL1, 2, 6, 7, and 10 were further functionally characterized owing to their high expression in the root apical meristem. Through mutant analyses and transgenic studies, we showed that these genes have the ability to promote root growth. Using yeast one‐hybrid and dual luciferase assays, we demonstrated that AHL1, 2, 6, 7, and 10 are transcription regulators and this activity is required for their roles in root growth. Although mutants for these genes did not showed obvious defects in root growth, transgenic plants expressing their fusion proteins with the SRDX repressor motif exhibited a short‐root phenotype. Through transcriptome analysis, histochemical staining and molecular genetics experiments, we found that AHL10 maintains redox homeostasis via direct regulation of glutathione transferase (GST) genes. When the transcript level of GSTF2, a top‐ranked target of AHL10, was reduced by RNAi, the short‐root phenotype in the AHL10‐SRDX expressing plant was largely rescued. Anthocyanins are important secondary metabolites contributing to the red coloration of fruits, the biosynthesis of which is significantly affected by light. Glutathione S-transferases (GSTs) play critical roles in the transport of anthocyanins from the cytosol to the vacuole. Despite their importance, GST genes in mango have not been extensively characterized. In this study, 62 mango GST genes were identified and further divided into six subfamilies. MiGSTs displayed high similarity in their exon/intron structure and motif and domain composition within the same subfamilies. The mango genome harbored eleven pairs of segmental gene duplications and ten sets of tandemly duplicated genes. Orthologous analysis identified twenty-nine, seven, thirty-four, and nineteen pairs of orthologous genes among mango MiGST genes and their counterparts in Arabidopsis, rice, citrus, and bayberry, respectively. Tissue-specific expression profiling highlighted tissue-specific expression patterns for MiGST genes. RNA-seq and qPCR analyses revealed elevated expression levels of seven MiGSTs including MiDHAR1, MiGSTU7, MiGSTU13, MiGSTU21, MiGSTF3, MiGSTF8, and MiGSTF9 during light-induced anthocyanin accumulation in mango. This study establishes a comprehensive genetic framework of MiGSTs in mango fruit and their potential roles in regulating anthocyanin accumulation, which is helpful in developing GST-derived molecular markers and speeding up the process of breeding new red-colored mango cultivars. Goods and Services Tax (GST) 2017 implement in India with the vision of simple indirect tax structure and enhance economic growth. However, its impact on micro, small and medium enterprises (MSMEs) has been a topic of concern. This critical assessment study focuses on the impact of GST on MSMEs in Rajasthan State (Pareek & Sharma, 2019). The study adopts a mixed-method approach, including a survey of 200 MSMEs and in-depth interviews with 20 industry experts and government officials. The study found that the implementation of GST has resulted in several benefits for MSMEs, such as simplification of tax procedures, increased efficiency in logistics and reduced corruption. However, MSMEs have also faced several challenges such as increased compliance costs, a lack of awareness about GST and issues related to the availability of input tax credit. Indian government must take proactive measures to educate MSMEs about the GST and simplify compliance procedures (Antonyraj & Kumar, 2023). It also recommends that policymakers should revisit the GST structure to make it more MSME-friendly. Overall, this study provides insights into the impact of GST on MSMEs and highlights the need for ongoing assessment and policy reform to ensure the long-term sustainability of these enterprises (Choudhary, 2019). Ferroptosis is a form of iron-dependent cell death that has attracted significant attention for its potential role in numerous diseases. Targeted inhibition of ferroptosis could be of potential use in treating diseases: such as drug induced liver injury (DILI). Ferroptosis can be antagonized by the xCT/GSH/GPX4, FSP1/CoQ 10 , DHODH/CoQ 10 , GCH1/BH 4 , and NRF2 pathways. Identifying novel anti-ferroptosis pathways will further promote our understanding of the biological nature of ferroptosis and help discover new drugs targeting ferroptosis related human diseases. In this study, we identified the clinically used drug mifepristone (RU486) as a novel ferroptosis inhibitor. Mechanistically, RU486 inhibits ferroptosis by inducing GSH synthesis pathway, which supplies GSH for glutathione-S-transferase (GST) mediated 4-HNE detoxification. Furthermore, RU486 induced RLIP76 and MRP1 export 4-HNE conjugate contributes to its anti-ferroptosis activity. Interestingly, RU486 induced GSH/GSTs/RLIP76&MRP1 anti-ferroptosis pathway acts independent of classic anti-ferroptosis systems: including xCT/GSH/GPX4, FSP1, DHODH, GCH1, SCD1 and FTH1. Moreover, NRF2 was identified to be important for RU486's anti-ferroptosis activity by inducing downstream gene expression. Importantly, in mouse model, RU486 showed strong protection effect on acetaminophen (APAP)-induced acute liver injury, evidenced by decreased ALT, AST level and histological recovery after APAP treatment. Interestingly, RU486 also decreased oxidative markers, including 4-HNE and MDA, and induced NRF2 activation as well as GSTs, MRP1 expression. The etiology of schizophrenia (SCZ), an incredibly complex disorder, remains multifaceted. Literature suggests the involvement of oxidative stress (OS) in the pathophysiology of SCZ. Determination of selected OS markers and brain-derived neurotrophic factor (BDNF) in patients with chronic SCZ and those in states predisposing to SCZ-first episode psychosis (FP) and ultra-high risk (UHR). Determination of OS markers and BDNF levels by spectrophotometric methods and ELISA in 150 individuals (116 patients diagnosed with SCZ or in a predisposed state, divided into four subgroups according to the type of disorder: deficit schizophrenia, non-deficit schizophrenia, FP, UHR). The control group included 34 healthy volunteers. Lower activities of analyzed antioxidant enzymes and GSH and TAC concentrations were found in all individuals in the study group compared to controls (p < 0.001). BDNF concentration was also lower in all groups compared to controls except in the UHR subgroup (p = 0.01). Correlations were observed between BDNF, R-GSSG, GST, GPx activity, and disease duration (p < 0.02). A small effect of smoking on selected OS markers was also noted (rho<0.06, p < 0.03). OS may play an important role in the pathophysiology of SCZ before developing the complete clinical pattern of the disorder. The redox imbalance manifests itself with such severity in individuals with SCZ and in a state predisposing to the development of this psychiatric disease that natural antioxidant systems become insufficient to compensate against it completely. High-order harmonic generation (HHG) in solids opens new frontiers in ultrafast spectroscopy of carrier and field dynamics in condensed matter, picometer resolution structural lattice characterization and designing compact platforms for attosecond pulse sources. Nanoscale structuring of solid surfaces provides a powerful tool for controlling the spatial characteristics and efficiency of the harmonic emission. Here we study HHG in a prototypical phase-change material Ge2Sb2Te5 (GST). In this material the crystal phase can be reversibly changed between a crystalline and amorphous phase by light or electric current mediated methods. We show that optical phase-switching is fully reversible and allows for dynamic control of harmonic emission. This introduces GST as new addition to materials that enable flexible metasurfaces and photonic structures that can be integrated in devices and allow for ultrafast optical control. In the complex and harsh environment of agriculture, rolling bearings, as the key transmission components in agricultural machinery, are very prone to failure, so research on the intelligent fault diagnosis of agricultural machinery components is critical. Therefore, this paper proposes a new method based on SVD-EDS-GST and ResNet-Vision Transformer (ResViT) for the fault diagnosis of rolling bearings in agricultural machines. Firstly, an experimental platform for rolling bearing failure in agricultural machinery is built, and one-dimensional vibration signals are obtained using acceleration sensors. Next, the signal is preprocessed for noise reduction using singular value decomposition (SVD) combined with the energy difference spectrum (EDS) to solve for the interference of complex noise and redundant components in the vibration signal. Secondly, generalized S-transform (GST) is used to process vibration signals into images. Then, the ResViT model is proposed, where the ResNet34 network is used to replace the image chunking mechanism in the original Vision Transformer model for feature extraction. Finally, an improved Vision Transformer (ViT) is utilized to synthesize global and local information for fault classification. The experimental results show that the proposed method's average accuracy in rolling bearing fault classification for agricultural machinery reaches 99.08%. In addition, compared with SVD-EDS-GST-CNN, SVD-EDS-GST-LSTM, STFT-ViT, GST-ViT, and SVD-EDS-GST-ViT, the accuracy rate was improved by 3.5%, 3.84%, 4.8%, 8.02%, and 0.56%, and the standard deviation was also minimized. Background and Design: In this study, we investigated whether Glutathione S-transferase Mu 1 (GSTM1) and Glutathione S-transferase Pi 1 (GSTP1) gene polymorphisms are risk factors for psoriasis development and characteristics of psoriasis. Materials and Methods: Venous blood samples were collected from both the patients and control subjects into 2 mL EDTA tubes. DNA was isolated from 260 psoriasis patients and 200 healthy control subjects, and GSTM1 and GSTP1 gene polymorphisms were genotyped by realtime polymerase chain reaction method. Results: According to the analysis results, although the frequency of the GSTM1 null genotype was 1.33 times higher in patients compared to controls, this difference was statistically insignificant [95% confidence interval (CI): 0.91-1.93, p=0.13]. GSTP1 AG heterozygous genotype and GG homozygous polymorphic genotype frequencies also did not differ between patients and controls [odds ratio (OR): 0.80, 95% CI: 0.55-1.18, p=0.27 for AG genotype; OR: 0.74, 95% CI: 0.35-1.56, p=0.42 for GG genotype]. When both polymorphisms were evaluated based on the onset age and severity of the disease, no significant difference was found between the early onset age group and the late onset age group, nor between the mild group and the severe group. Conclusion: The results show that GSTM1 and GSTP1 polymorphisms do not have a major effect on the etiopathogenesis and clinical characteristics of psoriasis.

Plant glutathione transferases (GSTs) constitute a large and diverse family of enzymes that are involved in plant stress response, metabolism and defence, yet their physiological functions remain largely elusive. Consistent with the traditional view on GSTs across organisms as detoxification enzymes, in vitro most plant GSTs catalyse glutathionylation, conjugation of the tripeptide glutathione (GSH; γ-Glu-Cys-Gly) onto reactive molecules. However, when it comes to elucidating GST functions, it remains a key challenge that the endogenous plant glutathione conjugates (GS-conjugates) that would result from such glutathionylation reactions are rarely reported. Furthermore, GSTs often display high substrate promiscuity, and their proposed substrates are prone to spontaneous chemical reactions with GSH; hence, single-gene knockouts rarely provide clear chemotypes or phenotypes. In a few cases, GS-conjugates are demonstrated to be biosynthetic intermediates that are rapidly further metabolized towards a pathway end product, explaining their low abundance and rare detection. In this review, we summarize the current knowledge of plant GST functions and how and possibly why evolution has resulted in a broad and extensive expansion of the plant GST family. Finally, we demonstrate that endogenous GS-conjugates are more prevalent in plants than assumed and suggest they are overlooked as clues towards the identification of plant GST functions. Abstract Matching of various chalcogenide films shows the advantage of delivering multilayer heterostructures whose physical properties can be tuned with respect to the ones of the constituent single films. In this work, (Ge–Sb–Te)-based heterostructures were deposited by radio frequency sputtering on Si(100) substrates and annealed up to 400 °C. The as-deposited and annealed samples were studied by means of X-ray fluorescence, X-ray diffraction, scanning transmission electron microscopy, electron energy loss spectroscopy and Raman spectroscopy. The heterostructures, combining thermally stable thin layers (i. e. Ge-rich Ge5.5Sb2Te5, Ge) and films exhibiting fast switching dynamics (i. e. Sb2Te3), show, on the one side, higher crystallization-onset temperatures than the standard Ge2Sb2Te5 alloy and, on the other side, none to minimal Ge-segregation.

Imidacloprid is a systematic neonicotinoid widely used to combat piercing-sucking insects; however, neonicotinoids, despite having low effects on vertebrates, showed high adverse effects on honeybees (Apis mellifera L.). Glutathione-S-transferases (GST) are an important constituent of the defense system in detoxifying invading chemicals. This work examines the effects of imidacloprid on GST activity in vivo and in vitro conditions. Results revealed that there are two responses of GST activity toward IMI treatment; first, stimulation of enzyme activity to combat and detoxify the insecticide. Second, direct inhibition of GST, which is confirmed by the in vitro inhibition with IC50 887.42 ppm. At a short exposure time to IMI (2 hours) at an IMI concentration of 0.35 ppm, the enzyme was stimulated up to 113% while increasing exposure time or IMI concentration, the inhibition effect dominates.

This study presents a comprehensive analysis of the dimerization interfaces of fly GSTs through sequence alignment. Our investigation revealed GSTE1 as a particularly intriguing target, providing valuable insights into the variations within Delta and Epsilon GST interfaces. The X-ray structure of GSTE1 was determined, unveiling remarkable thermal stability and a distinctive dimerization interface. Utilizing circular dichroism, we assessed the thermal stability of GSTE1 and other Drosophila GSTs with resolved X-ray structures. The subsequent examination of GST dimer stability correlated with the dimerization interface supported by findings from X-ray structural analysis and thermal stability measurements. Our discussion extends to the broader context of GST dimer interfaces, offering a generalized perspective on their stability. Simple Summary: The black cutworm, Agrotis ipsilon (Hufnagel), poses a significant threat to various crops. Marjoram (Origanum majorana) and rosemary (Rosmarinus officinalis) essential oils (EOs) were investigated for their toxicity using in vitro and in silico methods. GC-MS analysis identified the main constituents, with O. majorana being more toxic than R. officinalis to A. ipsilon larvae. R. officinalis EO inhibited Na+/K+ pump activity consistently, while O. majorana showed varied effects. Both EOs influenced detoxification enzymes differently over time. Molecular docking indicated a strong binding affinity of the main EO constituents to target enzymes. These findings suggest the potential of EOs as insecticides in integrated pest management programs, particularly in organic agriculture. In this study, in vitro and in silico approaches were employed to assess the toxicity of marjoram (Origanum majorana) and rosemary (Rosmarinus officinalis) essential oils (EOs) to A. ipsilon larvae. The study determined the activities of ATPases in the larvae after treatment with the LC20 and LC70 of each EO. α-esterase and glutathione-S-transferase (GST) activities were also determined after treatment with LC10 and LC30 of each EO. Furthermore, molecular docking was employed to determine the binding affinity of terpinene-4-ol and α-pinene, the major constituents of O. majorana, and R. officinalis EOs, respectively, compared to the co-crystallized ligand of α-esterase, diethyl hydrogen phosphate (DPF). Toxicity assays revealed that O. majorana EO was more toxic than R. officinalis EO to the A. ipsilon larvae at 96 h post-treatment. However, the LC20 and LC70 of the latter significantly inhibited the activity of the Na+-K+ pump at almost all intervals. The same concentrations significantly inhibited the Mg2+/Ca2+-ATPase and Ca2+ pump at 96 h post-treatment. In contrast, O. majorana EO showed a variable effect on the Na+-K+ pump across different time intervals. On the other hand, LC10 and LC30 of both EOs showed varied effects on α-esterase and GST over time. Molecular docking revealed energy scores of −4.51 and −4.29 kcal/mol for terpinene-4-ol and α-pinene, respectively, compared to a score of −4.67 for PDF. In this study, we used Raman spectroscopy to compare the local structure of Ge2Sb2Te5 (GST) thin films with thicknesses of 90 nm and 271 nm that were crystallized through thermal annealing and laser radiation (laser annealing) during the recording of Raman spectra in situ. We found that for all crystallized films, the position of the main peaks in the Raman spectra was almost the same, and their structure corresponded to a hexagonal close packed state. The main objective is to find out the impact of the Tax Credit (ITC) in the GST structure on the utilitarian capacity and liquidity of micro, small and medium-sized enterprises (MSMEs) in India. Using a coordinated survey to gather information on working capital stocks and GST consistency, 200 MSMEs in Assam, India, were selected to conduct a quantitative assessment. The results show that there are significant areas for the relationship between better working capital conditions and the use of ITS. This suggests that MSMEs can have greater financial flexibility and fewer pay restrictions if they do register ITC claims. According to the practical implications of the assessment, policymakers should deal with the ITC system and offer additional assistance to ensure that MSMEs can fully benefit from the GST. This study is novel because it is uniquely relevant to the state of Assam, offering alternative, progressive views on the specific impact of the goods and services tax on small businesses in that state. We proposed the numerical studies and machine learning prediction for the multilayered Borophene-GST-silica-Ag-based refractive index sensor for the 1.3–1.5 µm wavelength range. The top layers of the proposed sensor incorporate Ag gratings. The proposed structure utilizes a mode based on surface plasmonic resonance, which has been created using a finite element model through computational analysis. Resonance is observed across a range of refractive index values from 1 to 2.5, which primarily aligns with the refractive indices of key biomolecules such as haemoglobin, saliva, urine, and cancerous cells. The proposed structure has also been investigated for the different physical parameters such as material height, grating space, the incident wave's wide incident angle, and the GST material's phase. We demonstrate the modulation of reflectance values in response to varying phases of the GST material (amorphous and crystalline), which can be controlled through external temperature changes. The distribution of the electric and magnetic fields of the structure is also provided in order to examine the field distribution across the grating and other layers of material. This sensor offers a wide angle of stability (0 to 80°) for the respective resonating points. An artificial neural network is employed to analyze the simulated data and make predictions regarding the behaviour of the structure. We have found the R2 = 0.97 for the proposed artificial neural network (ANN) model. The ANN model of this structure also presented the values of mean square error (MSE), mean absolute error (MAE), and root mean square error (RMSE) for different epoch values. Results shown in this research can help to sense the wide range of biomolecule samples whose refractive index ranges from 1 to 2.5. The phrase "One Nation, One Tax" refers to the fact that all taxes must be paid in one location nationwide. The report thoroughly examines the effects of GST in India. The study offers sentiment analysis and bibliometric visualization of GST. It was discovered that the government implemented the GST in order to tax everyone in the nation and stop the flow of illicit money. Several genetic variations are associated with acute myeloid leukemia (AML) susceptibility, including the GSTP1 Ile105Val polymorphism. Even with the existing meta-analysis conducted on the topic, no consensus has been reached since none of the studies available performed in-depth data analysis. Hence, we performed an updated systematic review and meta-analysis in this paper to obtain more precise estimates. We searched various databases and calculated the odds ratio (OR) and 95% confidence interval (CI) to examine whether the GSTP1 Ile105Val polymorphism is associated with AML susceptibility. Further statistical analysis was also done to obtain more accurate and reliable findings. A total of 15 studies are included in the systematic review, but only 9 were included in the meta-analysis due to the studies deviating from the Hardy-Weinberg equilibrium. The analysis showed significantly increased susceptibility to AML in the allelic, co-dominant, and recessive models. Furthermore, subgroup analysis noted increased AML susceptibility in the non-Asian population. Comparing the proportions of the genotypes and alleles showed a significantly higher proportion of the Val/Val genotype and Val allele in the non-Asian cohort. Introduction: The biosynthesis of secondary metabolites like anthocyanins is often governed by metabolic gene clusters (MGCs) in the plant ancestral genome. However, the existence of gene clusters specifically regulating anthocyanin accumulation in certain organs is not well understood. Methods and results: In this study, we identify MGCs linked to the coloration of cotton reproductive organs, such as petals, spots, and fibers. Through genetic analysis and map-based cloning, we pinpointed key genes on chromosome A07, such as PCC/GhTT19, which is involved in anthocyanin transport, and GbBM and GhTT2-3A, which are associated with the regulation of anthocyanin and proanthocyanidin biosynthesis. Our results demonstrate the coordinated control of anthocyanin and proanthocyanidin pathways, highlighting the evolutionary significance of MGCs in plant adaptation. The conservation of these clusters in cotton chromosome A07 across species underscores their importance in reproductive development and color variation. Our study sheds light on the complex biosynthesis and transport mechanisms for plant pigments, emphasizing the role of transcription factors and transport proteins in pigment accumulation. The main objective is to find out the impact of the Tax Credit (ITC) in the GST structure on the utilitarian capacity and liquidity of micro, small and medium-sized enterprises (MSMEs) in India. Using a coordinated survey to gather information on working capital stocks and GST consistency, 200 MSMEs in Assam, India, were selected to conduct a quantitative assessment. The results show that there are significant areas for the relationship between better working capital conditions and the use of ITS. This suggests that MSMEs can have greater financial flexibility and fewer pay restrictions if they do register ITC claims. According to the practical implications of the assessment, policymakers should deal with the ITC system and offer additional assistance to ensure that MSMEs can fully benefit from the GST. This study is novel because it is uniquely relevant to the state of Assam, offering alternative, progressive views on the specific impact of the goods and services tax on small businesses in that state.

Polymorphisms of the uridine-diphospho-glucuronosyltransferase 1A1 (UGT1A1) gene, hepatic solute carrier organic anion transporter 1B1/B3 (SLCO1B1/3) gene, and glutathione S-transferase (GST) gene have been associated with significant hyperbilirubinemia in some populations. This study aims to determine whether the variation of UGT1A1, SLCO1B1/3 and GST genes play an important role in neonatal hyperbilirubinemia in Turkish newborn infants.

Environmental pollution is a global issue, and Kosovo is no exception, grappling with extensive mining activities. The research aims to assess the influence of industrial pollution in Drenas and Mitrovica using oxidative stress biomarkers (malondialdehyde MDA and the amount of glutathione transferase GST) and biochemical indicators (total proteins) in three plant species (Achillea millefolium, Hypericum perforatum and Plantago lanceolata). Plants were collected within a one-kilometer radius around industrial complexes in Drenas and Mitrovica, with Peja serving as a control point. In the homogenate of plant leaves, total proteins were determined using the Lowry method, MDA levels were determined using the Health Packer method, and GST activity was assessed using Habig method. GST activity was higher in the three plant species collected in Drenas and Mitrovica compared to those in Peje, the control point. Apart from Hypericum perforatoum in Mitrovica, the study revealed a significant increase in MDA levels in the three plant species from industrial areas compared to those in the control point. Generally, plants collected in industrial areas exhibited lower total protein levels than those in Peja. A negative correlation was observed between MDA levels and total proteins of Achillea millefolium and Plantago lanceolata, but not in Hypericum perforatum. The findings underscore the impact of pollution on the three plant species investigated in the study.

To explore the role of glutathione S-transferase gene (GST) in the accumulation of anthocyanin in Brassica juncea, one GST gene related to the anthocyanidin accumulation was cloned from near-isogenic lines of B. juncea with purple stalk and green stalk, and named as BjGSTF12. In this study, the bioinformatics characteristics of BjGSTF12 encoding protein and promoter were analyzed, the expression level of BjGSTF12 and the relationship with total anthocyanidin content were analyzed in B. juncea lines with purple stalk and green stalk. The results were as follow: (1) BjGSTF12 genes from B. juncea were successfully cloned, whose the full length of BjGSTF12 in genome and cDNA was 808 bp and 651 bp, encoding a protein of 216 amino acids. The BjGSTF12 contained the typical domains of GST proteins, namely GST_N and GST_C. However, their sequences of BjGSTF12 did not exhibit any differences between the two lines of B. juncea. (2) BjGSTF12 was closely related to AtGSTF12 in Arabidopsis, and both belonged to the φ subfamily of GST family. (3) The BjGSTF12 promoter sequences were cloned from two Brassica juncea strains of green stalk and purple stalk, and they exhibited four base mutations/insertions. However, the types and numbers of cis-acting elements did not show obvious differences between the two strains, including nine MYB transcription factor binding sites, one hormone response element, and three abiotic corresponding elements. (4) The total anthocyanidin content in B. juncea of purple stalk was significantly higher than that in green stalk ones, and the expression levels of BjGSTF12 in two lines were found to be similar to the total anthocyanidin content in both lines. (5) Protein interaction network analysis revealed that BjGSTF12 may interact with the key enzymes of anthocyanidin biosynthesis, glycosylation modification, and transporter proteins. In summary, BjGSTF12 is likely to play a key role in the accumulation of anthocyanidin in B. juncea by regulating its biosynthesis, modification, and transportation through interactions with other proteins. This study provides a theoretical reference for further study on the function of GST and the mechanism of anthocyanidin accumulation in B. juncea.

Abstract Background In paddy fields, the noxious weed barnyard grass secretes 2,4-dihydroxy-7-methoxy-2H-1,4-benzoxazin-3(4H)-one (DIMBOA) to interfere with rice growth. Rice is unable to synthesize DIMBOA. Rice cultivars with high or low levels of allelopathy may respond differently to DIMBOA. Results In this study, we found that low concentrations of DIMBOA (≤ 0.06 mM) promoted seedling growth in allelopathic rice PI312777, while DIMBOA (≤ 0.08 mM) had no significant influence on the nonallelopathic rice Lemont. DIMBOA treatment caused changes in the expression of a large number of glutathione S-transferase (GST) proteins, which resulting in enrichment of the glutathione metabolic pathway. This pathway facilitates plant detoxification of heterologous substances. The basal levels of GST activity in Lemont were significantly higher than those in PI312777, while GST activity in PI312777 was slightly induced by increasing DIMBOA concentrations. Overexpression of GST genes (Os09g0367700 and Os01g0949800) in these two cultivars enhanced rice resistance to DIMBOA. Conclusions Taken together, our results indicated that different rice accessions with different levels of allelopathy have variable tolerance to DIMBOA. Lemont had higher GST activity, which helped it tolerate DIMBOA, while PI312777 had lower GST activity that was more inducible. The enhancement of GST expression facilitates rice tolerance to DIMBOA toxins from barnyard grass root exudates.

Cotton is one of the most widely used industrial products in various industries and is attacked every year by the cotton bollworms, Helicoverpa armigera Hübner (Lepidoptera: Noctuidae). In this study, we investigated the effect of the spinosad insecticide on some biological and bio chemical parameters of the cotton bollworm. Bioassays were performed by using 3rd instar larvae fed with artificial diets containing lethal (LC50), and sublethal (LC15) concentrations of spinosad. Lethal and sublethal concentrations were estimated to be 0.90 and 0.35, mg a.i./kg, respectively. Compared to the control, lethal and sublethal concentrations of spinosad significantly increased larval and pupal development time, decreasing their weight. In addition, only LC50 of spinosad significantly reduced the fecundity and lifespan of adults as well as the hatching rate of the pest compared to LC15 and control. Moreover, lethal and sublethal concentrations of spinosad reduced carbohydrate and glycogen content and did not change lipid content. A significant decrease in protein content was observed only after exposure to LC50. At LC50 the activities of glutathione S-tran sferase (GSTs), a- an d ß- esterase were significantly reduced compared to LC15 and control. The results of this study showed that LC50 and LC15 of spinosad affect several biochemical and biological parameters of H. armigera. The enzymes known as glutathione S-transferases (GST), which are present in many evolved organisms, are essential for the defense against reactive oxygen species. GSTs have a role in the development of defenses against biotic and abiotic challenges, especially defending plants from various stresses such as drought, salinity, and heavy metal. This study uses a genome-wide investigation of the GST gene family in Phaseolus vulgaris to pinpoint several distinctive traits. 55 Pv-GST proteins have been identified in P. vulgaris. The molecular weights of these proteins range from 15.02 kDa to 47.99 kDa; the range of amino acid numbers 132 to 420 and the range of theoretical isoelectric points 5.03 to 9.61 were identified. Pv-GST genes are estimated to have at least 2 and a maximum of 10 exons, with an average of 4 exons. Phylogenetic analysis was performed with GST proteins from Arabidopsis thaliana, Glycine max, and Phaseolus vulgaris species, and subfamilies of these GSTs were identified. Using RNAseq data, the expression profiles of Pv-GST genes in leaf tissue of common bean during drought and salinity stress were identified. Using the obtained sequence data, primers for qRT-PCR were designed. Changes in the expression profiles of GST genes caused by salt and drought stress and melatonin treatments in two different common bean cultivars were determined by qRT-PCR experiments. Under drought and salt stress, the expression levels of GSTs decreased, while melatonin treatment with few exceptions increased these expression levels. As a result of this study, it was observed that Pv-GST genes may play a role in the growth and development of bean and may be involved in the response to abiotic stresses. Moreover, the results of this study will provide a basis for functional gene research and the expression profiles and qRT-PCR results under different stress conditions in common bean will provide a valuable contribution to the understanding of the functions of the GST gene family. BACKGROUND: Chlorantraniliprole (CAP) is a diamide insecticide with high efficacy against many pest insects, including the black cutworm, Agrotis ipsilon. Agrotis ipsilon is a serious pest causing significant yield losses in crops. Glutathione‐S‐transferases (GSTs) belong to a family of metabolic enzymes that can detoxify a wide range of pesticides. However, little is known about the functions of GSTs in CAP tolerance in A. ipsilon. RESULTS: A cDNA sequence (designated AiGSTu1) encoding an unclassified GST was identified from A. ipsilon. AiGSTu1 is highly expressed during the 3rd‐instar larval and the pupal stages. Most of the mRNA transcripts were found in larval Malpighian tubules. Exposure to CAP strongly enhanced AiGSTu1 expression, GST activity, hydrogen peroxide (H2O2) and malondialdehyde levels in larvae. H2O2 treatment upregulated the transcription level of AiGSTu1, suggesting that CAP‐induced oxidative stress may activate AiGSTu1 expression. The activity of recombinant AiGSTu1 was inhibited by CAP in a dose‐dependent manner. Metabolism assay results demonstrated that AiGSTu1 is capable of depleting CAP. Overexpression of AiGSTu1 enhanced the tolerance of Escherichia coli cells to H2O2 and the oxidative stress inducer, cumene hydroperoxide. Silencing of AiGSTu1 by RNA interference increased the susceptibility of A. ipsilon larvae to CAP. CONCLUSION: The findings of this study provide valuable insights into the potential role of AiGSTu1 in CAP detoxification and will improve our understanding of CAP tolerance in A. ipsilon. © 2023 Society of Chemical Industry. SUMMARY: The color of purple carrot taproots mainly depends on the anthocyanins sequestered in the vacuoles. Glutathione S‐transferases (GSTs) are key enzymes involved in anthocyanin transport. However, the precise mechanism of anthocyanin transport from the cytosolic surface of the endoplasmic reticulum (ER) to the vacuoles in carrots remains unclear. In this study, we conducted a comprehensive analysis of the carrot genome, leading to the identification of a total of 41 DcGST genes. Among these, DcGST1 emerged as a prominent candidate, displaying a strong positive correlation with anthocyanin pigmentation in carrot taproots. It was highly expressed in the purple taproot tissues of purple carrot cultivars, while it was virtually inactive in the non‐purple taproot tissues of purple and non‐purple carrot cultivars. DcGST1, a homolog of Arabidopsis thaliana TRANSPARENT TESTA 19 (TT19), belongs to the GSTF clade and plays a crucial role in anthocyanin transport. Using the CRISPR/Cas9 system, we successfully knocked out DcGST1 in the solid purple carrot cultivar 'Deep Purple' ('DPP'), resulting in carrots with orange taproots. Additionally, DcMYB7, an anthocyanin activator, binds to the DcGST1 promoter, activating its expression. Compared with the expression DcMYB7 alone, co‐expression of DcGST1 and DcMYB7 significantly increased anthocyanin accumulation in carrot calli. However, overexpression of DcGST1 in the two purple carrot cultivars did not change the anthocyanin accumulation pattern or significantly increase the anthocyanin content. These findings improve our understanding of anthocyanin transport mechanisms in plants, providing a molecular foundation for improving and enhancing carrot germplasm. Advances in online geospatial technologies (GST) have expanded access to K‐12 classrooms which has implications for the support teachers require to effectively integrate GSTs to promote learning. Previous studies have shown the impact of GST‐integrated lessons on student engagement, spatial thinking skills, and/or content knowledge; however, most of these studies have been small in scope and scale and frequently focus on the affordances of the technology, without addressing the context of the implementation and student characteristics for whom GST is most impactful. We attempt to address some of these gaps. Our program scaled an effective GST‐focused professional learning and development program to a national audience through a facilitator development model. This paper explores the student characteristics and lesson factors that resulted in student interest in science and technology and careers in those fields. After teaching a Geospatial Inquiry lesson created during a teacher workshop, teachers (n = 82) submitted the lessons and surveys on the implementation of Geospatial Inquiry lessons. The implementation surveys and lessons were scored for alignment to the principles of high‐quality Geospatial Inquiry. Students (n = 1924) completed a post‐lesson retrospective survey and indicated the extent to which their perceptions and attitudes toward science and technology changed because of the lesson. Results indicate that teacher GST performance is associated with increases in student outcomes. Students with previous exposure to science activities were more likely to have increased interest and excitement in science and careers in science but decreased interest in technology careers. Students who had previous exposure to technology activities had increased interest and excitement in technology and careers in technology but decreased interest in science careers. Geospatial Inquiry lessons also had a significant impact on students who are traditionally underrepresented in STEM fields. After participating in the lessons, students who identify as female reported higher engagement and interest in science and higher interest in science careers. Students who identified as Black or Hispanic also reported higher interest and excitement in science and technology, and students who identified as Black reported marginally higher interest in science careers. Over-expression of glutathione S-transferase (GST) can promote Cisplatin resistance in hepatocellular carcinoma (HCC) treatment. Hence, inhibiting GST is an attractive strategy to improve Cisplatin sensitivity in HCC therapy. Although several synthesized GST inhibitors have been developed, the side effects and narrow spectrum for anticancer seriously limit their clinical application. Considering the abundance of natural compounds with anticancer activity, this study developed a rapid fluorescence technique to screen “green” natural GST inhibitors with high specificity. The fluorescence assay demonstrated that schisanlactone B (hereafter abbreviated as C1) isolated from Xue tong significantly down-regulated GST levels in Cisplatin-resistant HCC cells in vitro and in vivo. Importantly, C1 can selectively kill HCC cells from normal liver cells, effectively improving the therapeutic effect of Cisplatin on HCC mice by down-regulating GST expression. Considering the high GST levels in HCC patients, this compound demonstrated the high potential for sensitizing HCC therapy in clinical practice by down-regulating GST levels.

Fusarium head blight (FHB) is a devastating fungal disease of hexaploid wheat (Triticum aestivum). Several genetic loci were previously identified that control FHB resistance in wheat, including Fhb1. Fhb7, a major QTL conferring resistance to FHB, controlling for mycotoxin deoxynivalenol (DON) production, has been introgressed into soft red winter wheat (SRWW). As an exotic QTL, Fhb7 is associated with linkage drag, affecting agronomic and end-use quality performance. This study outlines a breeding strategy for introducing and pyramiding Fhb7 into SRWW breeding populations that already possessed Fhb1 and harbored some additional disease-resistance genes. In addition to the Fhb1-Fhb7 pyramiding, we developed gene-based markers for both genes and examined them on 57 SRWW breeding lines. Our data showed that 15 out of 57 breeding lines possessed both Fhb1 and Fhb7 resistant alleles. Two years of phenotypic data from the inoculated and misted irrigation field showed that the combination of Fhb1-Fhb7 lowers mycotoxin DON accumulation in kernels, which provides protection for end-users and the milling industry. The Fhb gene-pyramided lines, with the additional regionally important disease resistance genes, produced in this breeding pipeline showed reasonable agronomic traits and can be used in crossing programs for the widespread introgression in elite wheat cultivars.

Abstract Type 2 diabetes mellitus is believed to be associated with microvascular complications which include diabetic retinopathy, nephropathy, and neuropathy. Oxidative stress plays a predominant role in the pathogenesis of DN and also influences metabolic endeavor and its hemodynamic pathways to possess various associations with renal complications, and one such is diabetic nephropathy which is the insignificant cause of end-stage renal disease. Renal injury in DN is predominantly related to the inclined oxidative stress, with influential metabolic endeavor and its hemodynamic pathways. Hyperglycemia, an hallmark feature of diabetes, promotes conditions of the diabetic patients responsible for higher reactive oxygen species production, which ultimately leads to increased oxidative stress, and this is considered to be the important event in the initiation of DN. Pertaining to oxidative stress, ROS is generated mostly by the variety of important pathways, in which this paves the way for antioxidant therapeutic approach preventing the initiation and progression/aggravation of tubular injury in DN. The most salient antioxidant enzymes including superoxide dismutase, catalase, glutathione-S-transferase, and glutathione peroxidase are considered as prime elements involved in the assembly and discharge of reactive metabolites. Therefore, this review highlights that antioxidant gene polymorphisms also postulate that this in these antioxidant genes may be a major cause for the pathogenesis of DN. Hence, it could also answer many questions put forth by researchers, and clinicians detecting the single-nucleotide polymorphism of these antioxidant genes and targeting therapeutic approach can enhance the genetic changes and help to reduce severity at the early stages of DN. Additionally, this literature review also shows the importance of regional population studies on detecting the SNPs of antioxidant gene which in turn reflects the status of oxidative stress involved in the pathogenesis of DN associated with T2D.
