Cell death drives the magnitude and community composition of phytoplankton and can result in the conversion of particulate organic carbon to dissolved organic carbon (DOC), thereby affecting carbon cycling in the aquatic food web. We used a membrane integrity probe (Sytox Green) to study the seasonal variation in the percentage of viable cells in the phytoplankton population in an estuary in the northern Baltic Sea for 21 months. Phytoplankton cell death is an important process in marine food webs, but the viability of natural phytoplankton communities remains unexplored in many ecosystems. In this study, we measured the viability of natural pico- and nanophytoplankton communities in the central and southern parts of the Baltic Sea (55°21′ N, 17°06′ E–60°18′ N, 19°14′ E) during spring (4th–15th April 2016) to assess differences among phytoplankton groups and the potential relationship between cell death and temperature, and inorganic nutrient availability. Cell viability was determined by SYTOX Green cell staining and flow cytometry at a total of 27 stations representing differing hydrographic regimes. Three general groups of phytoplankton (picocyanobacteria, picoeukaryotes, and nanophytoplankton) were identified by cytometry using pigment fluorescence and light scatter characteristics. The picocyanobacteria and picoeukaryotes had significantly higher cell viability than the nanophytoplankton population at all depths throughout the study area. Viability correlated positively with the photosynthetic efficiency (Fv/Fm, maximum quantum yield of photosystem II) as measured on the total phytoplankton community. However, an anticipated correlation with dissolved organic carbon was not observed. We found that the abiotic factors suggested to affect phytoplankton viability in other marine ecosystems were not as important in the Baltic Sea, and other biotic processes, e.g. processes related to species succession could have a more pronounced role.

With the increasing demand for antimicrobial agents and the spread of antibiotic resistance in pathogens, the exploitation of plant oils to partly replace antibiotic emerges as an important source of fine chemicals, functional food utility and pharmaceutical industries. This work introduces a novel catalytic method of plant oils hydroxylation by Fe(III) citrate monohydrate (Fe3+-cit.)/Na2S2O8 catalyst. Methyl (9Z,12Z)-octadecadienoate (ML) was selected as an example of vegetable oils hydroxylation to its hydroxy-conjugated derivatives (CHML) in the presence of a new complex of Fe(II)-species. Methyl 9,12-di-hydroxyoctadecanoate 1, methyl-9-hydroxyoctadecanoate 2 and methyl (10E,12E)-octadecanoate 3 mixtures is produced under optimized condition with oxygen balloon. The specific hydroxylation activity was lower in the case of using Na2S2O8 alone as a catalyst. A chemical reaction has shown the main process converted of plantoils hydroxylation and (+ 16 Da) of OH- attached at the methyl linoleate (ML-OH). HPLC and MALDI-ToF-mass spectrometry were employed for determining the obtained products. It was found that adding oxidizing agents (Na2S2O8) to Fe3+ in the MeCN mixture with H2O would generate the new complex of Fe(II)-species, which improves the C-H activation. Hence, the present study demonstrated a new functional method for better usage of vegetable oils.Producing conjugated hydroxy-fatty acids/esters with better antipathogenic properties. CHML used in food industry, It has a potential pathway to food safety and packaging process with good advantages, fundamental to microbial resistance. Lastly, our findings showed that biological monitoring of CHML-minimum inhibitory concentration (MIC) inhibited growth of various gram-positive and gram-negative bacteria in vitro study. The produced CHML profiles were comparable to the corresponding to previousstudies and showed improved the inhibition efficiency over the respective kanamycin derivatives.

In recent decades, the incidence of Cryptococcus neoformans infection, which causes cryptococcosis, has consistently increased. Fluconazole (FLU) is frequently used in the treatment of this disease, mainly in the immunocompromised population, and long-term therapy usually produces drug resistance. Research on antifungal sensitizers has gained attention as a possible means of overcoming this drug resistance. Minocycline (MINO) has an inhibitory effect in vitro on FLU-resistant Candida albicans, and the combination of MINO and FLU has a synergistic effect on FLU-resistant C. albicans. A synergistic effect of MINO/FLU has been reported against C. neoformans, but this effect has not been evaluated on FLU-resistant isolates. This study aimed to investigate the interaction of MINO and FLU against FLU-resistant C. neoformans both in vitro and in vivo. We found that the combination of MINO and FLU had a synergistic effect on FLU-resistant C. neoformans in vitro. For all FLU-resistant strains, the minimum inhibitory concentration (MIC) of FLU decreased significantly when used in combination with MINO, dropping from >128 μg/ml down to 4-8 μg/ml. Additionally, MINO and FLU had a synergistic effect on both susceptible and resistant C. neoformans biofilms, in which the MIC of FLU decreased from >256 μg/ml down to 4-16 μg/ml. Compared with FLU alone, the combination of MINO with FLU prolonged the survival rate of Galleria mellonella larvae infected with FLU-resistant C. neoformans, and also significantly decreased the fungal burden of infected larvae and reduced the tissue damage and destruction caused by FLU-resistant C. neoformans. WOS: 000400179800013, The main purpose of this study was to analyze the cytotoxic activity of an extract obtained from Centaurea fenzlii Reichardt, and the fractions eluted from this extract, in breast cancer cells. After isolation and structural analysis of the fractions were conducted, a meaningful cytotoxic effect was indicated. The goal of the analysis was to reveal the mechanism by which this effect occurs through researching the apoptotic side of these fractions and determining the amount of several proteins that are the products of the genes. Test substances were applied to breast cancer cells and the inhibitory concentration value 50 (IC50) that caused a cytotoxic effect was determined using MTT and ATP assays. The Centaurea fenzlii Reichardt dichloromethane extracts-ethyl acetate fractions (CFDCM-EAF) exhibited a stronger growth-inhibitory effect on MCF-7 cells (45.771 mu g/mL). The apoptotic effect was studied using double staining and flow cytometry. The death rate in the cells treated with the CFDCM-EAF IC50 dose was approximately 90%: 9.2% living cells, 22.8% necrotic cells, 62.3% late apoptotic cells, and 5.8% early apoptotic cells. Structural analysis of the CFDCM-EAF, which indicated significant cytotoxic effects, was performed using chromatographic methods. The Popeye domain containing (POPDC) gene family consists of POPDC1 (also known as BVES), POPDC2 and POPDC3 and encodes a novel class of cyclic adenosine monophosphate (cAMP) effector proteins. Despite first reports of their isolation and initial characterization at the protein level dating back 20 years, only recently major advances in defining their biological functions and disease association have been made. Loss-of-function experiments in mice and zebrafish established an important role in skeletal muscle regeneration, heart rhythm control and stress signaling. Patients suffering from muscular dystrophy and atrioventricular block were found to carry missense and nonsense mutations in either of the three POPDC genes, which suggests an important function in the control of striated muscle homeostasis. However, POPDC genes are also expressed in a number of epithelial cells and function as tumor suppressor genes involved in the control of epithelial structure, tight junction formation and signaling. Suppression of POPDC genes enhances tumor cell proliferation, migration, invasion and metastasis in a variety of human cancers, thus promoting a malignant phenotype. Moreover, downregulation of POPDC1 and POPDC3 expression in different cancer types has been associated with poor prognosis. However, high POPDC3 expression has also been correlated to poor clinical prognosis in head and neck squamous cell carcinoma, suggesting that POPDC3 potentially plays different roles in the progression of different types of cancer. Interestingly, a gain of POPDC1 function in tumor cells inhibits cell proliferation, migration and invasion thereby reducing malignancy. Furthermore, POPDC proteins have been implicated in the control of cell cycle genes and epidermal growth factor and Wnt signaling. Work in tumor cell lines suggest that cyclic nucleotide binding may also be important in epithelial cells. Thus, POPDC proteins have a prominent role in tissue homeostasis and cellular signaling in both epithelia and striated muscle.

Short communication, As blueberries are rich in phenolic compounds, which possess a demonstrated antimicrobial activity, it may be interesting to further elucidate its real potential and properties. As such, the main goal of this work was to evaluate the potential antimicrobial activity of Vaccinium corymbosum dry fruits and leaves aqueous extracts against several contaminants/pathogenic microorganisms while at the same time assessing the effect of V. corymbosum over lactic acid bacteria.

Vascular endothelial growth factor (VEGF) has a central role in breast cancer development and progression, but the mechanisms that control its expression are poorly understood. Breast cancer tissue microarrays revealed an inverse correlation between the Forkhead transcription factor Forkhead box class O (FOXO)3a and VEGF expression. Using the lapatinib-sensitive breast cancer cell lines BT474 and SKBR3 as model systems, we tested the possibility that VEGF expression is negatively regulated by FOXO3a. Lapatinib treatment of BT474 or SKBR3 cells resulted in nuclear translocation and activation of FOXO3a, followed by a reduction in VEGF expression. Transient transfection and inducible expression experiments showed that FOXO3a represses the proximal VEGF promoter, whereas another Forkhead member, FOXM1, induces VEGF expression. Chromatin immunoprecipitation and oligonucleotide pull-down assays showed that both FOXO3a and FOXM1 bind a consensus Forkhead response element (FHRE) in the VEGF promoter. Upon lapatinib stimulation, activated FOXO3a displaces FOXM1 bound to the FHRE before recruiting histone deacetylase 2 (HDAC2) to the promoter, leading to decreased histones H3 and H4 acetylation, and concomitant transcriptional inhibition of VEGF. Platinum resistance of cancer cells may evolve due to a decrease in intracellular drug accumulation, decreased cell permeability or by an increased deactivation of the drug by glutathione (GSH). The aim of this study was (1) to investigate the effect of adenosine 5'-triphosphate (ATP) on the cytotoxicity of cisplatin in a large cell lung carcinoma cell line (H460), and (2) to examine the potential involvement of increased cisplatin uptake, GSH depletion and pyrimidine starvation by ATP in this effect. H460 cells were harvested and seeded (5% CO(2); 37 degrees C). Subsequently, cells were incubated with medium or ATP followed by an incubation with cisplatin. Cytotoxicity screening was analyzed by the sulforhodamine B (SRB) colorimetric assay, lactate dehydrogenase and caspase-3/7 activity. Pre-incubation for 72h with 0.3 and 3mM ATP strongly enhanced the anti-proliferative potency of cisplatin 2.9- and 7.6-fold, respectively. Moreover, after incubation of H460 cells with 0.3mM ATP the intracellular platinum concentration increased, indicating increased cisplatin uptake by ATP. ATP, despite lowering the LD(50) of cisplatin, did not modulate GSH levels in H460 cells. ATP itself showed a biphasic effect on H460 cell growth: 0.3mM inhibited H460 cell growth via the pyrimidine starvation effect, activation of caspase-3/7 and LDH leakage, while 3mM ATP showed no effect on cell growth. In conclusion, ATP sensitizes the H460 cells to cisplatin-induced apoptosis. The effect of 0.3mM ATP is not due to GSH depletion but involves increased cisplatin uptake and pyrimidine starvation due to ATP conversion to adenosine followed by cellular uptake.

Currently there is much interest to cultivate microalgae for the production of bulk products like lipids for biodiesel or as feedstock for industrial chemical processes. To make the production economically feasible, it is essential to develop cultivation systems in which algae convert the light with a high photosynthetic efficiency, to obtain microalgae with specific production characteristics, to optimize the culture medium, to develop cheap harvesting methods and finally professionals to implement and develop the technology. For the development of better algal cultivation systems, insight is needed on the efficiency of light conversion by these algae. Two concepts of light dilution in photobioreactor design are presented to reflect how potentially higher photosynthetic efficiencies can be achieved. Also, fast screening methods for microalgae are discussed, which can be applied to select strains with better production characteristics, to optimize the medium, to compare cultivation conditions and to investigate the toxic effects of chemicals. Apart from obtaining enhanced algal productivities a low-cost harvesting process is required. For harvesting of bulk products, as preconcentration step, controlled auto-flocculation is worthwhile investigating. Beside these research lines, more and better education for biochemical engineers should be developed to push microalgal biotechnology ahead.

Natural populations of the bloom forming cyanobacterium Microcystis are typically composed of several distinct genotypes. Using Microcystis strains that differ in growth rate, microcystin production and colony formation, we conducted a laboratory experiment in the presence and absence of a grazer, the water flea Daphnia, to investigate whether interactions among strains can be predicted from functional traits, and whether the outcome of competition between strains is influenced by a grazer. Two toxic and two non-toxic Microcystis strains, isolated from a single lake, were grown during four weeks as single strains, in all possible combinations of two strains and all together, in the presence and absence of Daphnia magna. The relative abundance of strains in the populations was assessed using denaturing gradient gel electrophoresis, and the growth rate of each strain in mixed populations was compared to its growth rate in monoculture to determine interactions between strains. The observed interactions were strain-specific, and the relative abundances of strains in mixed populations could be partially explained by taking toxicity and colony formation into account. Importantly, some of the interactions were strongly altered by the presence of Daphnia. Daphnia induced colony formation in one strain, which then became a better competitor. Daphnia grazing also caused a higher evenness in the populations, both through a weakening of competitive interactions as well as by facilitation effects. Strong facilitation effects were due to non-toxic strains benefiting from the protection offered by toxic strains in the presence of predation. Overall, our results emphasize the presence of strong competitive interactions between Microcystis strains in the absence of grazing, whereas indirect positive interactions are prevalent in the presence of a generalist grazer. Our results suggest that differences in functional traits and grazer-mediated facilitation effects may enhance coexistence of Microcystis strains, including toxic and non-toxic strains.

Possible allelopathic effects of substances released from the macrophytes Chara globularis, Elodea canadensis, Myriophyllum spicatum on the common green alga Scenedesmus obliquus were tested in the laboratory with plastic plants and untreated medium as controls. A two-phase approach was used in which first the effects of physical presence of plants was studied (phase I) followed by the effects of plant culture filtrates (phase II). In the presence of plastic plants growth was reduced only marginally, but strong growth inhibition of Scenedesmus occurred in the physical presence of all macrophytes. In contrast, filtrates from Chara had no growth inhibitory effect on Scenedesmus. Myriophyllum filtrate reduced particle-based growth rate by 7% compared to filtration controls, while Elodea culture filtrate reduced volume-based growth by 12%, chlorophyll-based growth by 28% and particle-based growth by 15%. Photosystem II-efficiency of Scenedesmus was reduced in all three macrophyte treatments in phase I, but not in filtrates from macrophyte cultures (phase II). Thus, while enzyme activity or other physiological aspects may have been affected, the current study yielded no proof for allelopathically active compounds being directed at photosynthesis. Mean particle volume (MPV) of Scenedesmus was not influenced by macrophyte exudates and cultures remained dominated by unicells. The strong growth inhibitory effects found for Scenedesmus in the physical presence of macrophytes, but not in plastic controls, and no or weaker response in nutrient-enriched filtrates, suggest nutrient competition was a more powerful driving factor than allelochemicals. However, the experimental design does not exclude disappearance of allelochemicals during the filtration process. Cell viability is an essential touchstone in the study of the effect of medium components on cell physiology. We developed a flow-cytometric assay to determine sponge-cell viability, based on the combined use of fluorescein diacetate (FDA) and propidium iodide (PI). Cell fluorescence measurements based on incubation of cells with FDA or PI resulted in a useful and reproducible estimate of the viability of primary sponge-cell cultures. We studied the effects of temperature, ammonium, and the fungicide amphotericin B on the viability of a primary-cell culture from the marine sponge Suberites domuncula using the aforementioned flow-cytometric assay. S. domuncula cells die rapidly at a temperature of greater than or equal to22degreesC, but they are insensitive to ammonium concentrations of up to 25 mM. Amphotericin B, which is frequently used in sponge-cell culture media, was found to be toxic to S. domuncula cells. (C) 2004 Wiley Periodicals, Inc.

Starting with Savageau's pioneering work regarding demand rules for gene regulation from the 1970s, here, we choose the simplest transcription network and ask: how does the cell choose a particular regulatory topology from all available possibilities? According to the demand rules, a cell chooses an activator based regulation of a target if the target protein is required for most of the time. On the other hand, if the target protein is only required sporadically, its control tends to be via a repressor-based regulatory topology. We study the natural distribution of topologies at genome, systems, and micro-levels in E. coli and observe deviations from demand rules. Analyzing the regulation of amino acid biosynthesis, transport, and carbon utilization in E. coli and B. subtilis, and comparing choice of topology with demand, we observe an alternate pattern emerging. Simulations of networks are used to help explain the natural distribution of topologies in nature. Overall, our results indicate that the choice of topology is drawn randomly from a pool of all networks which satisfy the dynamic requirements of the cell, as dictated by physiology. In short, our results suggest that the cell picks "whatever works''.

The hypothesis that cyanobacteria can be controlled by commercially available ultrasound transducers was tested in laboratory experiments with cultures of the cyanobacteria Anabaena sp., Cylindrospermopsis raciborskii and Microcystis aeruginosa and the green alga Scenedesmus obliquus that were grown in the absence or presence of ultrasound (mix of 20, 28 and 44 kHz). The Scenedesmus experiment also included a treatment with the zooplankton grazer Daphnia magna. Chlorophyll-a and biovolume-based growth of Anabaena was significantly lower in ultrasound exposed cultures than in controls. Particle based growth rates were higher in ultrasound treatments. Filaments were significantly shorter in ultrasound exposed cultures reflecting filament breakage. Photosystem II efficiency was not affected by ultrasound. In Cylindrospermopsis chlorophyll-a based growth rates and photosystem II efficiencies were similar in controls and ultrasound treatments, but biovolume-based growth was significantly lower in ultrasound exposed cultures compared to controls. Despite biovolume growth rates of the filamentous cyanobacteria were reduced in ultrasound treatments compared to controls, growth remained positive implying still a population increase. In Microcystis and Scenedesmus growth rates were similar in controls and ultrasound treatments. Hence, no effect of ultrasound on these phytoplankton species was found. Ultrasound should not be viewed ``environmental friendly'' as it killed all Daphnia within 15 mm, releasing Scenedesmus from grazing control in the cultures. Based on our experiments and critical literature review, we conclude that there is no music in controlling cyanobacteria in situ with the commercially available ultrasound transducers we have tested. We investigated whether in vitro L1210 growth inhibition by alpha-difluoromethylomithine (DFMO; 740 mu M) and 4-amidinoindan-l-one 2'-amidinohydrazone (CGP 48664A; 1.7 mu M) ii reversible with N-1-acetylspermine (N-1-acSp). Influences of N-1-acSp dose (1-100 mu M), time (0-12 h at 100 mu M), aminoguanidine (AG, 1 mM) and cell numbers (at 1 mu M N-1-acSp) on percentage S-phase, polyamine contents and viability were determined. DFMO/CGP 48664A decreased percentage S-phase from 58 to 26%, decreased spermidine (Sd) and sl,ermine (Sp) contents 3-fold, but did not affect viability. With increasing N-1-acSp dose, S-phase percentage and Sd contents increased concomitantly, reaching plateau values that were comparable with those of untreated controls. S-phase and Sd content increased from 4-6 h after N-1-acSp administration, reaching plateau values from 11 and 6 h, respectively. N-1-acSp content was dose dependent and increased linearly to reach plateau values from 8 h. AG did not affect any of these parameters. Addition of 1 mu M N-1-acSp to decreasing numbers of DFMO/CGP 48664A-treated cells caused increasing S-phase percentage, Sd and N-1-acSp contents. We conclude that cell cycle kinetics of cultured L1210 cells can be manipulated by the induction of growth inhibition with DFMO/CGP 48664A and its subsequent abolishment with N-1-acSp. N-1-acSp accumulation rate and its subsequent conversion to Sd is relatively slow compared with intracellular Sd needs. The data support the notion that Sd is the most important polyamine for growth. We describe a method for the profiling of polyamines, N-acetylated polyamines and the polyamine analogues N-1,N-11 bis(ethyl)norspermine (BE-3-3-3) and 1,19-bis(ethylamino)-5,10,15-triazanonadecane (BE-4-4-4-4) in L1210 murine leukaemia cells by capillary gas chromatography with nitrogen-phosphorus detection. The method makes use of four internal standards. Prepurification comprises deproteinization, isolation with Sep-Pak silica at pH 9.0, conversion to heptafluorobutyryl derivatives and postderivatization organic fluid extraction. Within-and between-series precisions (given as C.V.s) for analysis of 1-2x10(6) cells were: putrescine 5.5 and 29.4%; spermidine 1.6 and 7.1%; and spermine 3.2 and 7.6%, respectively. Recoveries relative to the respective internal standards, were in the 70.6-104.7% range. Accuracy and precision of measurements of BE-4-4-4-4 can probably be improved by the introduction of a separate pentamine internal standard. We conclude that the method can be used for studying the effect of BE-3-3-3 and BE-4-4-4-4, and possibly their metabolites, on polyamine homeostasis (biosynthesis, retroconversion, transport, terminal catabolism) and polyamine function. In this study, the inhibition effect of some organic acids as natural antimicrobial agents and also, the efficiency of potassium sorbate which is known and used as a food preservative were researched to compare with each other. Saccharomyces cerevisiae ATCC 9763 and Kluyveromyces lactis ATCC 8585 strains were activated in Malt Ekstract Broth. The inhibitory effects of some concentrations of potassium sorbate (0, 300, 500, 750, 1000 mg L-1) and organic acids which are acetic, lactic and citric acids (0, 0.1, 0.3, 0.5, 0.7, 1.0%) at pH 6.0 was researched. It was determined that the inhibitory effect of K-sorbate and organic acid concentrations are quite different. According to the results of this research, acetic acid concentrations are the most effective and citric acid is the second effective for inhibition of S. cerevisiae, on the other hand, for K. lactis as K-sorbate is the most effective one, acetic acid is the second effective inhibitor. But, lactic acid has the weakest effect for both of yeasts.

Type II phosphatidylinositol (PtdIns) 4-kinases produce PtdIns 4-phosphate, an early key signaling molecule in phosphatidylinositol cycle, which is indispensable for T cell activation. Type II PtdIns 4-kinase alpha and beta have similar biochemical properties. To distinguish these isoforms Epigallocatechin gallate (EGCG) has been evaluated as a specific inhibitor. EGCG is the major active catechin in green tea having anti-inflammatory, antiatherogenic and cancer chemopreventive properties. The precise mechanism of actions and molecular targets of EGCG in early signaling cascades are not well understood. In the present study, we have shown that EGCG inhibits type II PtdIns 4-kinases (alpha and beta isoforms) and PtdIns 3-kinase activity in vitro. EGCG directly bind to both alpha and beta isoforms of type II PtdIns 4-kinases with a Kd of 2.62 mu M and 1.02 mu M, respectively. Type II PtdIns 4-kinase-EGCG complex have different binding pattern at its excited state. Both isoforms showed significant change in helicity upon binding with EGCG. EGCG modulates its effect by interacting with ATP binding pocket: the residues likely to be involved in EGCG binding were predicted by Autodock. Our findings suggest that EGCG inhibits two isoforms and could be a key to regulate T cell activation. Published by Elsevier Inc.

Risk factors for intramammary infections caused by coagulase-negative staphylococci, contagious major pathogens and environmental major pathogens in early lactating heifers were evaluated at the herd, heifer and quarter levels. In total, 764 quarters of 191 dairy heifers in 20 randomly selected farms in Flanders (Belgium) were sampled. Quarter milk samples were collected between 1 and 4 days in milk and between 5 and 8 days in milk for bacteriological culture. Data were analyzed using multivariable, multilevel logistic regression analysis. Higher average herd milk somatic cell count (>200,000 cells/mL), not having an effective fly control strategy, contact with lactating cows prior to calving and moderate to severe udder edema prior to calving increased the odds of intramammary infections caused by contagious major pathogens. Poor heifer hygiene and lack of mineral/vitamin supplementation prior to calving were risk factors for intramammary infection caused by environmental major pathogens. Teat apex colonization with coagulase-negative staphylococci prior to calving seemed to protect quarters against intramammary infections caused by major pathogens. Poor heifer hygiene before calving, a non-clipped udder and not practicing of teat dipping prior to calving increased the odds of intramammary infection with coagulase-negative staphylococci. Although management is important in the prevention and control of intramammary infections in early lactating heifers, most variation in the prevalence of intramammary infections resided at the heifer and quarter levels, indicating that the susceptibility for intramammary infections around calving is mainly determined by heifer and quarter characteristics.

Histone deacetylase (HDAC) inhibitors arrest cancer cell growth and cause apoptosis with low toxicity thereby constituting a promising treatment for cancer. In this study, we investigated the anti-tumor activity in lung cancer cells of the novel cyclic amide-bearing hydroxamic acid based HDAC inhibitors SL142 and SL325. In A549 and H441 lung cancer cells both SL142 and SL325 induced more cell growth inhibition and cell death than the hydroxamic acid-based HDAC inhibitor suberoylanilide hydroxamic acid (SAHA). Moreover, the combination treatment using retinoid drugs ATRA or 9-cis RA along with SL142 or SL325 significantly induced more apoptosis and suppressed colony formation than the single use of either. The expression of the retinoic acid receptors RARα, RARβ, RXRα and RXRβ were unchanged with the treatment. However a luciferase reporter construct (pGL4. RARE 7x) containing seven tandem repeats of the retinoic acid responsible element (RARE) generated significant transcriptional activity after the combination treatment of retinoic acids and SL142 or SL325 in H441 lung cancer cells. Moreover, apoptosis-promoting Bax expression and caspase-3 activity was increased after the combination treatment. These results suggest that the combination treatment of SL142 or SL325 with retinoic acids exerts significant anti-tumor activity and is a promising therapeutic candidate to treat human lung cancer.

Currently there is much interest to cultivate microalgae for the production of bulk products like lipids for biodiesel or as feedstock for industrial chemical processes. To make the production economically feasible, it is essential to develop cultivation systems in which algae convert the light with a high photosynthetic efficiency, to obtain microalgae with specific production characteristics, to optimize the culture medium, to develop cheap harvesting methods and finally professionals to implement and develop the technology. For the development of better algal cultivation systems, insight is needed on the efficiency of light conversion by these algae. Two concepts of light dilution in photobioreactor design are presented to reflect how potentially higher photosynthetic efficiencies can be achieved. Also, fast screening methods for microalgae are discussed, which can be applied to select strains with better production characteristics, to optimize the medium, to compare cultivation conditions and to investigate the toxic effects of chemicals. Apart from obtaining enhanced algal productivities a low-cost harvesting process is required. For harvesting of bulk products, as preconcentration step, controlled auto-flocculation is worthwhile investigating. Beside these research lines, more and better education for biochemical engineers should be developed to push microalgal biotechnology ahead.

To be able to study the effect of mixing as well as any other parameter on productivity of algal cultures, we designed a lab-scale photobioreactor in which a short light path (SLP) of (12 mm) is combined with controlled mixing and aeration. Mixing is provided by rotating an inner tube in the cylindrical cultivation vessel creating Taylor vortex flow and as such mixing can be uncoupled from aeration. Gas exchange is monitored on-line to gain insight in growth and productivity. The maximal productivity, hence photosynthetic efficiency, of Chlorella sorokiniana cultures at high light intensities (1,500 micromol m(-1) s(-1)) was investigated in this Taylor vortex flow SLP photobioreactor. We performed duplicate batch experiments at three different mixing rates: 70, 110, and 140 rpm, all in the turbulent Taylor vortex flow regime. For the mixing rate of 140 rpm, we calculated a quantum requirement for oxygen evolution of 21.2 mol PAR photons per mol O(2) and a yield of biomass on light energy of 0.8 g biomass per mol PAR photons. The maximal photosynthetic efficiency was found at relatively low biomass densities (2.3 g L(-1)) at which light was just attenuated before reaching the rear of the culture. When increasing the mixing rate twofold, we only found a small increase in productivity. On the basis of these results, we conclude that the maximal productivity and photosynthetic efficiency for C. sorokiniana can be found at that biomass concentration where no significant dark zone can develop and that the influence of mixing-induced light/dark fluctuations is marginal.

Peroxisome proliferator-activated receptor gamma (PPAR gamma) gene is a nuclear receptor that is involved in thyroid tumourigenesis. Recently, our group has shown that follicular carcinomas underexpressing PPAR gamma protein are more prone to develop distant metastases, to invade locally, to present poorly differentiated areas and to persist after surgery. Aneuploidy is also observed in some thyroid tumours, particularly in the more advanced cases. The aim of the present study was to investigate the association of PPAR gamma expression with the degree of differentiation and ploidy status of benign and malignant thyroid neoplasias. DNA cytometric studies, ploidy and S-phase fraction (SPF) determination, and quantitative RT-PCR analysis of molecular markers specific for thyroid follicular cells, namely Tg (thyroglobulin), TSHR (TSH receptor) and NIS (Na+/I- symporter) were compared between thyroid lesions with positive or negative PPAR gamma protein expression. We observed that PPAR gamma-negative tissues expressed lower levels of Tg mRNA [4.66x10(6) a.u. (arbitrary units) +/- 1.49x10(6)], and were more frequently aneuploid (36%), and presented higher SPF (3.1%+/- 0.4) than PPAR gamma-positive samples (Tg mRNA = 2.54x10(7) a.u. +/- 9.72 x 10(6). P=0.0006; aneuploidy=8%, P=0.0031; SPF=2.2%+/- 0.2, P=0.0430). A similar trend was also observed for TSHR and NIS mRNA, although not reaching statistical significance. This study showed that underexpression of PPAR gamma is associated with poor tumour differentiation, aneuploidy and higher cell proliferative activity. Therapies designed to modulate expression of PPAR gamma may have an impact on the growth of thyroid neoplasias. publishersversion published

1 - Article; 1-Hydroxymethylene-1,1-bisphosphonic acids (HMBPs) are important drugs for the treatment of a variety of bone diseases. Since these compounds have no chromophore, their detection is challenging and mass spectrometry (MS) appears to be an appropriate sensitive tool. Our work deals with the analysis by electrospray ionization tandem mass spectrometry (ESI-MSn) of the well-known nitrogen-containing HMBP alendronate and of three analogues, considered as potential prodrugs. These four molecules share a common structure with different protecting groups on the phosphonic acid and on the amine functions. We describe the dissociation mechanisms of nitrogen-containing HMBPs in positive ion mode and we compare, in negative ion mode, our results with literature data. In both modes, the dissociations are essentially losses of ROH, and of phosphorus-containing species (HPO2, ROP(OH)(2) and ROPO(OH)(2)), where R = H, C6H5, or CH3OC6H5. Submerged aquatic macrophytes are important in shallow aquatic ecosystems because they stabilize the macrophyte-dominated state by increasing water transparency in various ways. One of these is the excretion of allelopathic substances inhibitory to phytoplankton, but it is still controversial whether this mechanism can be important in the field. We developed a model that describes phytoplankton growth including the effects of two different macrophytes ( Chara sp. and Stratiotes aloides ), which are both known to excrete allelopathic substances. With this model we analysed singular and combined effects of shading (1), sediment resuspension (2), competition for nutrients (3) and allelopathy (4) on the development of phytoplankton. Both macrophyte species had a considerable effect on phosphorus levels in the water. Shading of phytoplankton by Chara sp. was estimated to be negligible. This macrophyte species has a growth form close to the sediment. The allelopathic activity of Chara sp. caused a phytoplankton growth reduction of ca. 5–10%, while the most pronounced effect seems to be related to reduction of resuspension. S. aloides floats on the water surface during spring and summer. Therefore, it has a potentially high shading effect. Additionally, this species has a relatively strong allelopathic capacity, sometimes resulting in a reduction of phytoplankton between 50 and 80%. The model suggests that the relative contribution of allelopathy in situ is low or negligible for charophytes. Only for macrophyte species that have a high allelopathic potential, like S. aloides , it may be a significant effect.

Cell viability is an essential touchstone in the study of the effect of medium components on cell physiology. We developed a flow-cytometric assay to determine sponge-cell viability, based on the combined use of fluorescein diacetate (FDA) and propidium iodide (PI). Cell fluorescence measurements based on incubation of cells with FDA or PI resulted in a useful and reproducible estimate of the viability of primary sponge-cell cultures. We studied the effects of temperature, ammonium, and the fungicide amphotericin B on the viability of a primary-cell culture from the marine sponge Suberites domuncula using the aforementioned flow-cytometric assay. S. domuncula cells die rapidly at a temperature of greater than or equal to22degreesC, but they are insensitive to ammonium concentrations of up to 25 mM. Amphotericin B, which is frequently used in sponge-cell culture media, was found to be toxic to S. domuncula cells. (C) 2004 Wiley Periodicals, Inc.

The retinoid 4-(N-hydroxyphenyl)retinamide (4HPR, fenretinide) has both growth inhibitory and apoptosis-inducing effects on a number of cancer cell lines in vitro and in vivo and has been entered into a number of oncological trials. However, little is known about its mechanism(s) of action or its effects on normal cells such as fibroblasts. In this study, the effects of fenretinide on both epidermoid carcinoma cells of vulva (cell line A431) and normal human dermal fibroblasts, both as monolayers and also grown in 3D cell culture systems, have been investigated. The 3D cell culture system contained normal human fibroblasts embedded in a type I collagen gel with the carcinoma cells seeded on top of the collagen gel, which mimics the epidermoid carcinoma. Fenretinide significantly inhibited the rate of DNA synthesis of carcinoma cells, while there was little effect on fibroblasts on monolayers, at 10(-6)-10(-5) M, which are clinically attainable doses. Fenretinide at 5 x 10(-6) M induced apoptosis characterised by cell shrinkage, membrane blebbing, nuclear condensation and/or fragmentation, and cell detachment in carcinoma cells, but not fibroblasts from monolayers. Fenretinide also reduced the viability of carcinoma cells in the 3D cell culture system without affecting fibroblasts. These data show that fenretinide may preferentially induce apoptosis in epidermoid carcinoma cells.

We investigated whether in vitro L1210 growth inhibition by alpha-difluoromethylomithine (DFMO; 740 mu M) and 4-amidinoindan-l-one 2'-amidinohydrazone (CGP 48664A; 1.7 mu M) ii reversible with N-1-acetylspermine (N-1-acSp). Influences of N-1-acSp dose (1-100 mu M), time (0-12 h at 100 mu M), aminoguanidine (AG, 1 mM) and cell numbers (at 1 mu M N-1-acSp) on percentage S-phase, polyamine contents and viability were determined. DFMO/CGP 48664A decreased percentage S-phase from 58 to 26%, decreased spermidine (Sd) and sl,ermine (Sp) contents 3-fold, but did not affect viability. With increasing N-1-acSp dose, S-phase percentage and Sd contents increased concomitantly, reaching plateau values that were comparable with those of untreated controls. S-phase and Sd content increased from 4-6 h after N-1-acSp administration, reaching plateau values from 11 and 6 h, respectively. N-1-acSp content was dose dependent and increased linearly to reach plateau values from 8 h. AG did not affect any of these parameters. Addition of 1 mu M N-1-acSp to decreasing numbers of DFMO/CGP 48664A-treated cells caused increasing S-phase percentage, Sd and N-1-acSp contents. We conclude that cell cycle kinetics of cultured L1210 cells can be manipulated by the induction of growth inhibition with DFMO/CGP 48664A and its subsequent abolishment with N-1-acSp. N-1-acSp accumulation rate and its subsequent conversion to Sd is relatively slow compared with intracellular Sd needs. The data support the notion that Sd is the most important polyamine for growth.
