Abstract

Parkinson's disease (PD) is characterised by motor symptoms caused by degeneration of dopaminergic neurons in the substantia nigra. The cause remains elusive and current treatments cannot halt or reverse progression. This thesis explores the effects of nicotinamide on cells associated with PD; exploring its roles in the development and protection of dopaminergic neurons, and its influence on microglia. Nicotinamide was tested on 46C mouse embryonic stem cells. Neural differentiation, indicated by Sox1-GFP expression, was identified by flow cytometry and immunocytochemistry. Neuroprotective effects of nicotinamide were assessed using a 6- hydroxydopamine primary ventral mesencephalon (VM) cell model of PD, and tyrosine hydroxylase (TH) immunocytochemistry to identify dopaminergic neurons. Effects of nicotinamide on Iba1+ microglia were assessed in VM cultures and in isolated rat cortex microglial cultures. Nicotinamide limited proliferation of 46C cells over 7 days of differentiation. Sox1-GFP expression was lower in nicotinamide-treated cultures compared to control, but peak expression occurred earlier. β-III-tubulin+ neuron numbers remained unchanged at day 7. In VM cultures, TH+ neuron numbers were higher in nicotinamide-treated cultures at days 1, 4, and 7. However, nicotinamide did not protect TH+ neurons against 6-hydroxydopamine. Nicotinamide treatment showed reversible impairment of Iba1+ microglial proliferation in VM cultures. Lower numbers of Iba1+ microglia were observed in isolated microglial cultures with 10 mM nicotinamide; however, nicotinamide did not prevent granulocyte-macrophage colony-stimulating factor-induced proliferation. Thus, nicotinamide could be utilised for novel PD treatments including promoting neuronal differentiation and restricting unwanted progenitor cell proliferation in stem cell replacement therapy. Nicotinamide may also enhance dopaminergic differentiation or survival in primary VM cultures. Finally, nicotinamide's ability to reduce microglial proliferation could be utilised as a novel therapy to slow progression at PD onset or may offer protection from immune rejection in cell replacement therapies.

6. Making sense of rewards-based crowdfunding : understanding the lived experience of nascent entrepreneurs

Abstract

Rewards-Based Crowdfunding has recently received increasing academic attention. It has been conceptualised as a source of finance that has reshaped the funding cycle. However, little is known about the lived experience of entrepreneurs using it to start a venture. In comparison to other, more traditional sources of finance, the monetary values raised are relatively small, and successful campaigns rely on a wide network of contacts on social media, who act co-creatively. Moreover, empirical research has largely used data collected by online platforms, resulting in a narrow focus on campaign success, rather than insights into enactment of the phenomenon. Therefore, research on the lived experience of entrepreneurs engaged in this phenomenon can contribute strongly to the advancement of the field. Drawing on six cases in the UK, this thesis employs an interpretivist, hermeneutic approach, adopting a sensemaking lens. Utilising in-depth interviews, online archival research and combined with the hermeneutic preunderstandings of the researcher, the study sought to gain rich insights into the phenomenon. The findings identify insights in three specific themes; (1) multi-dimensional aspirations, (2) complementary resourcing and (3) the practice of ‘entrepreneuring’. Further, from the hermeneutic consideration of the whole phenomenon, three approaches of seeking; (1) alignment, (2) sufficiency, and (3) flexibility, comprise key influencers in enacting their ventures using the phenomenon. The entrepreneurs have leveraged Rewards-Based Crowdfunding to acquire patient cashflow, market-traction and “marketing potential of persuasion” capital resources, hybrid activities which complement financial resources. Further, the findings show that the entrepreneurs leveraged alignment to increase the speed of tie formation and their ties are complex and dynamic. The identification of such approaches are significant theoretical contributions the thesis makes to resource theory, network tie theory and the broader Rewards-Based Crowdfunding literature. Furthermore, the study makes a general contribution to the study of the lived experience of entrepreneurial venture enactment. Future research should build on the insights, taking a contextual approach.

8. What are we missing? : an archaeothanatological approach to late Anglo-Saxon burials

Abstract

Archaeothanatology, a holistic approach conceived in France, examines detailed observations of the spatial positioning of skeletal elements in a grave, to characterize taphonomy and reveal funerary practices that would otherwise be archaeologically invisible. However, archaeothanatology suffers from a lack of comparative case studies exploring the effects of different burial environments upon the decomposition and disarticulation of a human corpse in the grave. Furthermore, investigations into funerary practices applying a taphonomic approach are seldom contained in published reports for burials in England. This has resulted in potentially valuable evidence for funerary practices being overlooked. This study evaluates the utility of archaeothanatology as a tool for reconstructing original burial form from an archaeological grave context, with specific focus on the identification of wooden containers from the late Anglo-Saxon period (c.A.D. 650-1100). Recent data suggests a variety of different containers for the body were commonly used but identifies so-called 'plain-earth graves' as the norm. Many containers will have been constructed entirely from wood, decomposing completely, rendering them archaeologically invisible confounding attempts to explore their prevalence. Presently various inconsistent evidence is used to identify possible wooden containers. A taphonomy-based analysis of skeletons from graves where preserved wood and metalwork provided conclusive evidence for coffins was undertaken. This information, as well as adding to our overall knowledge about decomposition in a wooden container, has been used to develop a tailored method to assist in identifying coffined burials. The method was applied to three cemeteries containing burials without surviving evidence for coffins. The results indicate that the prior default determination of 'plain-earth' in a substantial number of burials was flawed. From the sample studied, 41% were identified as decomposing in a void, 28% more than originally identified through evidence from funerary architecture alone. Thus, confirming archaeothanatology can improve burial interpretations and is beneficial to funerary archaeology.

Abstract

This article reflects on the outcomes of a workshop at the Fair Trade International Symposium in June 2023 which explored some of the ways young people are engaging in Fair Trade. Campaigners recognize the importance of giving leadership responsibilities to young people with fresh ideas and social connections to help expand the Fair Trade movement. There are opportunities to engage young people through the connections between Fair Trade and the environmental and social issues that young people are passionate about today. The workshop showcased three such initiatives: a climate and food security debate; an ethical enterprise project; and Fairtrade Fortnight events at the University of Leeds. Through these initiatives young people learned about trade (in)justice and gained a deeper understanding of the root causes of sustainability issues, whilst gaining transferable skills and building competencies to work collaboratively and creatively. Engaging with Fair Trade can provide young people with experiential learning opportunities that forward the goals of the Education for Sustainable Development (ESD) agenda and promote the relevance of the Fair Trade movement to a younger generation. Abstract

To be a desirable social partner and develop healthy relationships with peers, a child must be able to engage with peers across a variety of contexts. Understanding the factors supporting high levels of social engagement with peers is thereby essential, requiring the development of nuanced and ecologically valid indices of social engagement. Building on recent adult work, the current study explores conversational response time as a novel index of children's social engagement with peers in a dyadic context. This study further explores relationship between conversational response time and children's shyness. Fifty‐six 9‐ to 11‐year‐old children interacted with an unfamiliar peer in an unstructured setting and completed a self‐report measure of shyness. Children's behaviour was coded for their conversational RTs and overall social engagement. Faster conversational RTs were significantly related to children's own social engagement and marginally related to their partners' engagement. Moreover, higher shyness in children's partners predicted faster conversational RTs in children themselves. New directions for using conversational RT as an index of children's social engagement and implications for accounts children's social development are discussed. [ABSTRACT FROM AUTHOR]

Abstract

Young informal carers (YICs) are non-professional young individuals providing care and support in various forms, usually to immediate family members, afflicted from a diverse range of both long- and short-term health conditions. Although there is significant knowledge about the information needs of adult carers in general, information needs and information seeking characteristics of the YICs' community are understudied and are different. This study aims to identify the information needs of YICs communicated over the Internet and understanding their information seeking characteristics through a three-stage qualitative content analysis of posts written by YICs on two notable Internet forums. The analysis of 323 posts dated between March 2010 and April 2019 finds YICs' needs are categorised by two types of online expression of needs, situational and information. Situational needs are illustrations of current difficult conditions and information needs are direct requests for information. Under situational and information needs, we identify four types of needs expressed: personal and professional growth, health (self and caree), finance and relationships. In addition, the findings indicate 94.36% posts in the sample as situational needs, which depict the uncertainty experienced by YICs under caring circumstances. The findings can assist government organisations and charities by improving the indexing of advice pages of their websites appropriate to the YICs' search words, better availability of information and advertising, in addition to building quality mobile applications or digital support tools. Abstract

Technology is widely promoted as a solution to greater independence and better health for the rapidly growing UK older population. If this is to be realised, we need to understand barriers and facilitators to uptake and investigate who wants this technology and who does not express an interest in use. This analysis is based on data from a population-based cohort study, the Cognitive Function and Ageing Study (CFAS)-II, which focused on brain health in older people and included questions about access to- and interest in- internet technologies. The factors affecting willingness to use technologies that support memory and ADL were identified using binary logistic regression analysis. 541 people aged 75 years and older from Cambridgeshire, Nottingham and Newcastle responded. Older adults were more willing to use technologies directed towards improving memory (65%) than towards ADL supportive technologies (38%). Regression analysis showed that an older age (OR = 0.64, 95% CI = 0.34–0.98), female gender (OR = 0.64, 95% CI = 0.42–0.99), no access to technology including laptops and tablets (OR = 0.48, 95% CI = 0.32–0.72), and self-reported physically less slowing down (but no objective health indicators) (OR = 0.57, 95% CI = 0.36–0.88) were strongly associated with UK older adults' lesser willingness to use memory assistive technologies while not having access to laptops and tablets (OR = 0.57, 95% CI = 0.39–0.84) was associated with willingness to use ADL supportive technologies. Older people, females and those with less access to technologies should be considered as target groups by healthcare providers, policymakers, and technology producers to promote technology and support healthy and independent ageing. Abstract

Air travel mediates transboundary movement of SARS-CoV-2. To prepare for future pandemics, we sought to understand air passenger behaviour and perceived risk during the COVID-19 pandemic. This study of UK adults (n = 2103) quantified knowledge of COVID-19 symptoms, perceived health risk of contracting COVID-19, likelihood of returning to the UK with COVID-19 symptoms, likelihood to obey self-quarantining guidelines, how safe air travellers felt when flying during the pandemic (n = 305), and perceptions towards face covering effectiveness.Overall knowledge of COVID-19 symptoms was poor. Men and younger age groups (18–44) were less informed than women and older age groups (44 +). A significant proportion (21%) of the population would likely travel back to the UK whilst displaying COVID-19 symptoms with many expressing that they would not fully comply with self-isolation guidelines. Overall, males and younger age groups had a reduced perceived personal risk from contracting COVID-19, posing a higher risk of transporting SARS-CoV-2 back to the UK. Poor passenger knowledge and behaviour undermines government guidelines and policies aimed at preventing SARS-CoV-2 entry into the UK. This supports the need for stricter, clearer and more targeted guidelines with point-of-departure viral testing and stricter quarantining upon arrival.

Abstract

Cardiovascular ageing contributes to cognitive impairment. However, the unique and synergistic contributions of multiple cardiovascular factors to cognitive function remain unclear because they are often condensed into a single composite score or examined in isolation. We hypothesized that vascular risk factors, electrocardiographic features and blood pressure indices reveal multiple latent vascular factors, with independent contributions to cognition. In a population-based deep-phenotyping study (n = 708, age 18–88), path analysis revealed three latent vascular factors dissociating the autonomic nervous system response from two components of blood pressure. These three factors made unique and additive contributions to the variability in crystallized and fluid intelligence. The discrepancy in fluid relative to crystallized intelligence, indicative of cognitive decline, was associated with a latent vascular factor predominantly expressing pulse pressure. This suggests that higher pulse pressure is associated with cognitive decline from expected performance. The effect was stronger in older adults. Controlling pulse pressure may help to preserve cognition, particularly in older adults. Our findings highlight the need to better understand the multifactorial nature of vascular aging.

Variables associated with increased odds of prevalent loneliness included: prior loneliness, living alone, female gender, living in an area of higher deprivation, frequent pre-pandemic social contact at community groups, and separation from family during the pandemic, adjusted for age and sex. Weekly technology-mediated contact using telephone or video calls was associated with lower odds of loneliness. CONCLUSIONS: COVID-19 recovery plans should address loneliness in older people. Target groups should include those who have previously been lonely, people who live alone, those living in deprived areas, and those who had previously been socially active through community groups.

46. Financial Awareness Education with Apprentices in the Australian Construction Industry: Program Evaluation

Abstract

A financial awareness education program was implemented with construction industry apprentices in Victoria, Australia. The program included face-to-face delivery of education around a range of financial management issues that apprentices face as they begin their apprenticeship. The paper reports on an evaluation of the program, which included surveys at completion of the presentation, as well as focus group follow-up two months later. Both intended behaviour change and actual behaviour change, including knowledge retention, are noted in the results. The program evaluation highlights factors such as relevance and immediacy of need as crucial to determining appropriate content and successful implementation. The paper concludes by recommending inclusion of financial awareness in vocational education curricula on an ongoing basis, rather than as ad hoc delivered programs, to ensure sustainable knowledge gains and behaviour change outcomes.
