Camellia reticulata , an ancient plant species endemic to Yunnan Province, China, remains underexplored in terms of its endophytic bacterial communities. The plant tissue pedicel serves as the connection between the flower and the stem, not only delivers nutrients but also transmits metabolic substances from endophytic bacteria to the nectar during long-term microbial colonization and probably improves the antagonistic activity of nectar against yeast. Hence, 138 isolates of endophytic bacteria have been isolated in this study from the pedicels of 12- and 60-year-old C. reticulata. Comparative analysis revealed significantly higher density of endophytic bacteria in older trees. Among these isolates, 29 exhibited inhibitory effects against nectar yeasts. Most of the isolates displayed positive results for Gram staining, catalase reaction, gelatin liquefaction, and motility. Additionally, the isolates demonstrated the ability to utilize diverse substrates, such as glucose, nitrate, and starch. Based on 16S rRNA molecular biology analysis, these isolates were identified to be 11 different species of 6 genera, with the majority belonging to Bacillus genus. Notably, C1 isolate, identified as Bacillus spizizenii , exhibited strongest antagonistic effect against three yeasts, i.e., Metschnikowia reukaufii, Cryptococcus laurentii , and Rhodotorula glutinis , with minimum inhibitory concentration values below 250 μg/mL. Major metabolites of B. spizizenii were aminoglycosides, beta-lactams, and quinolones, which possess antimicrobial activities. Furthermore, KEGG enrichment pathways primarily included the synthesis of plant secondary metabolites, phenylpropanoids, amino acids, alkaloids, flavonoids, neomycin, kanamycin, and gentamicin. Therefore, antagonistic activity of B. spizizenii against yeasts could be attributed to these antibiotics. The findings highlight the diverse endophytic bacteria associated with C. reticulata , indicating their potential as a valuable resource of bioactive metabolites. Objective: To isolate Pseudomonas aeruginosa from different clinical samples and determine the antimicrobial activity of Ceftazidime/avibactam against these isolates. Study Design: Prospective Cross-sectional study. Setting: Pathology Laboratory of Lahore General Hospital, Lahore. Period: July 2023 to June 2024. Methods: One hundred thirteen Pseudomonas aeruginosa were identified from different samples Bacterial identification was done by gram staining, bench tests, and API20NE. The antimicrobial sensitivity testing of the causative bacteria was conducted, using commercially available discs, by Kirby Bauer disc diffusion assay and reported in accordance with Clinical & Laboratory Standards Institute (CLSI) 2022. Results: Out of 113 resistant strains of Pseudomonas aeruginosa obtained from different clinical samples Ceftazidime/avibactam was only sensitive to 43.4% of strains and resistant to 56.6% of them. Conclusion: According to the findings of this study, Pseudomonas aeruginosa, a nosocomial organism is isolated from many different clinical samples. The findings of this study also indicate that even a new combination antibiotic fails to show sensitivity in more than half of the isolates. Neurolisteriosis, a common disease of small ruminants, is most often caused by Listeria monocytogenes. Here we describe 25 cases of caprine neurolisteriosis diagnosed in our laboratory over a 5-y period and compare our fluorescent antibody test (FAT) results with immunohistochemistry (IHC) and polymerase chain reaction (PCR) testing for diagnostic confirmation. Neurohistologic changes consistent with neurolisteriosis affected the pons in all cases, extending rostrally to the mesencephalon in 6 cases, caudally to the medulla oblongata in 6 cases, and/or dorsally to the cerebellum in 4 cases. Acute inflammatory changes were observed in 17 cases, and included neuroparenchymal microabscesses, neuronal necrosis and neuronophagia, axonal swelling, microgliosis and astrogliosis, and perivascular neutrophils with macrophages, lymphocytes, and plasma cells that occasionally extended to the leptomeninges. Subacute-to-chronic changes (8 cases) consisted of neuroparenchymal and perivascular clusters of macrophages with rare neutrophils, lymphocytes, and plasma cells admixed with glial nodules. Bacterial bacilli were observed within neutrophils or macrophages in H&E-stained tissue sections in 4 cases. Gram stain highlighted gram-positive bacilli in 13 cases. Native valve endocarditis owing to Mycobacterium abscessus (M. abscessus) is extremely rare and only a few cases have been reported in the literature. We present a case of a 44-year old male patient, a coronary artery disease (CAD) case with a stent in situ with pyrexia of unknown origin (PUO). He started becoming febrile following placement of stent and continued to remain febrile for duration of five months before coming to our centre. Echocardiogram showed vegetations on the aortic valve and paired blood cultures grew M. abscessus. Antibiotics including Clarithromycin were added to his treatment and a valve replacement was planned. The patient underwent valve replacement, however succumbed to the illness despite best efforts. It is difficult to diagnose M. abscessus in Gram stain as they appear as diphtheroids and may be interpreted as contaminants. Even an accurate diagnosis makes this organism difficult to treat. Objective: The primary goal of this study is to discern the optimal adjuvant treatment for patients diagnosed with bacterial vaginosis, focusing on reducing recurrence rates. Methods: This study is a double-blind clinical trial with no previous similar trials conducted to date. The study population consisted of non-pregnant, married women visiting teaching hospitals' clinics in Mashhad, complaining of vaginal discharge. After informed consent and questionnaire completion, samples were obtained from vaginal discharge surrounding the cervix of clinically diagnosed bacterial vaginosis patients. Using Gram staining, a gold standard method for bacterial vaginosis diagnosis, samples were examined under a microscope according to the Nugent score. After initial treatment with metronidazole, patients were divided into two groups receiving either vaginal or oral probiotics. Results: Of the 55 participating women, 20 were in the vaginal probiotic group and 35 were in the oral probiotic group. No significant demographic or clinical differences existed between groups at baseline. The Nugent score decreased from 8.5 to 3 in the vaginal group and from 9 to 3 in the oral group, suggesting the effectiveness of both treatments. While the difference between groups was not statistically significant, each group showed significant improvements from their initial states (p-value < 0.001). Conclusion: No significant difference was observed in the effectiveness of oral versus vaginal probiotics in reducing the recurrence of bacterial vaginosis after routine treatment. The article presents a case study of a 71-year-old woman with confusion, aphasia, and a brain mass, diagnosed with a large enhancing lesion in the left frontal lobe, suspected to be metastatic cancer. Topics include the patient's imaging findings, the evaluation and surgical intervention for brain mass diagnosis, and the consideration of metastatic cancer as a potential cause based on the presence of a pulmonary nodule and a breast nodule.

Background: The burden of sepsis worsens due to the continuation of emerging pathogens such as multidrug-resistant Pantoea species. Methods: A multicenter study was conducted between October 2019 and September 2020 at four hospitals located in central, southern, and northern parts of Ethiopia. A total of 1416 sepsis patients were recruited and blood cultures were performed. At each study site, positive cultures were characterized by their colony characteristics, gram stain, and conventional biochemical tests. All Pantoea species were identified using Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry (MALDI TOF) and subjected to whole genome sequencing (WGS) using Illumina HiSeq 2500. The phylogeny structure of Pantoea isolates was calculated using IQ-TREE v1.6.12 from single-nucleotide polymorphisms detected by Snippy v.4.6.0 and filtered by Gubbins v.2.3.4. Average nucleotide identity was estimated by using OrthoANI v.0.93.1 on Shovill v.1.1.0 assemblies. Antimicrobial resistance genes and plasmid replicons were detected using ARIBA v.2.14.6. Phylogenetic trees were visualized using iTOLv.6.5.2. Results: Multiple Pantoea species include: P. dispersa (n = 19), P. septica (n = 1), and a novel Pantoea spp. (n = 1), were identified among sepsis patients. All P. dispersa isolates and the novel Pantoea species were isolated at Dessie Referral Hospital and displayed phylogenetic clonality, including the ubiquity of an IncM1 plasmid and identical antimicrobial resistance (AMR) gene profiles, encoding blaCTX−M−15, blaTEM−1D, blaSCO−1, and aac(3)-lla. The novel Pantoea spp. isolate harboured blaCTX−M−9 and blaTEM−1D and carried an IncN3 plasmid replicon. The P. septica was isolated at Tikur Anbessa Specialized Hospital in Addis Ababa and carried no detectable acquired AMR genes. Conclusion: The emerging Pantoea spp. carrying multiple AMR genes were identified from sepsis patients. Implementation of strong infection prevention strategies and building surveillance capacity with advanced bacteriology laboratories capable of identifying multidrug-resistant emerging pathogens is strongly recommended. Background: Pertussis is a highly contagious, vaccine-preventable acute respiratory infection caused by Bordetella pertussis (B. Pertussis) and affects all age groups. Since 1947, the Global Expanded Program on Immunization (EPI) was executed, and its incidence has been effectively curtailed. The reasons for the resurgence are multifactorial including the waning of vaccine acquired immunity, the difference in vaccination strategies, the adaptation of B. Pertussis strains, and an increase in disease awareness due to the strengthening of diagnostic sensitivity and surveillance sensitivity. Notably, Hewlett and Edwards proposed that the shift in pertussis transmission patterns plays an important role in pertussis resurgence. Diphtheria is an acute infectious disease of upper respiratory tract caused by toxigenic strains of Corynebacterium Diphtheriae (CD) and Corynebacterium other than diphtheriae (COD). The organism is locally invasive and causes exotoxin - mediated illness and can lead to complications like stridor, respiratory obstruction, myocarditis, nerve palsy, renal insufficiency and death in severe cases. The objective of the present study is to isolate and identify the etiological agents in clinically suspected cases of Diphtheria and Pertussis. Materials & Methods: This Prospective study has been conducted in the department of Microbiology from Feb 2022 to Aug 2024. Samples with high clinical suspicion were received and processed. Two Throat swabs were collected in patients with clinical suspicion of Diphtheria. They were inoculated on Blood agar, Potassium Tellurite agar, Tinsdale agar. Gram staining and Albert staining were done. Molecular testing also performed for C. diphtheriae. Two nasopharyngeal swabs were collected in patients with clinical suspicion of Pertussis. They were inoculated on Bordet Gengou medium, Charcoal Agar with Cephalexin and Blood Agar and Mac Conkey agar. Gram Staining was performed and Biochemical testing was carried out. Molecular testing was carried out for B. Pertussis and B. parapertussis. Results: Out of 53 samples with clinical suspicion of Diphtheriae, 6 were positive for Corynebacterium diphtheria by culture. Molecular testing was also done for testing of rpo B gene and Tox A gene. 6 samples came to be positive for C. diphtheria rpo B gene where as one was positive for tox gene production. Out of 20 cases with strong suspicion of Pertussis, culture was positive for Bordetella pertussis in 2 samples. Molecular testing came out to be positive for one sample. Testing for the antibodies by ELISA two patients were serologically positive for Pertussis. Both Culture and molecular testing was positive in one sample. Both culture and serology was positive in one sample. Only serology was positive in one sample. The research aimed to evaluate the antioxidative and antibacterial characteristics of aqueous sumac extract on methicillin-resistant Staphylococcus aureus through in-vitro and in-vivo study. Sumac extract has been obtained through the soaking method, and its antioxidant properties were gauged using the DPPH free radical scavenging method. The minimum inhibitory concentration (MIC) of sumac extract was determined on S. aureus obtained from hospitalized patients, as well as an assessment of biofilm-formation and the release of bacterial intracellular compounds. in vivo experimentation involved injecting bacteria (108 cfu/ml) into mice, which subsequently manifested indicators of symptoms of infection, and the number of bacteria within their bloodstream was quantified. The Sumac extract demonstrated strong antioxidant properties at concentrations of 1000 mg/ml. Furthermore, the agar tests for the gram staining, mannitol, coagulase, and DNase revealed that 190 cultured bacteria samples were identified as Staphylococcus aureus. These bacteria were resistant to clindamycin, ciprofloxacin, and methicillin antibiotics, but sensitive to erythromycin and penicillin antibiotics. Additionally, the bacteria displayed significant methicillin resistance and formed a strong biofilm (65.78%). The sumac extract showed a MIC range of 125–1000 µg/ml against Staphylococcus aureus. Treatment with concentrations above the MIC was found to prevent the formation of biofilm and increase the release of bacterial intracellular compounds. Sumac extract led to a decrease in bacterial count in the blood of mice and reduced signs of infection. Sumac extract demonstrated powerful antioxidant and antibacterial effects against resistant microorganisms, suggesting its potential as a promising compound for the treatment of resistant infections in future research. Cat scratch disease rarely presents as a breast or axillary mass mimicking carcinoma both clinically and radiologically. Diagnosing breast/axillary cat scratch disease is challenging due to its rarity and nonspecific findings. Here, we reported 2 patients with breast cat scratch disease and reviewed 14 patients with cat scratch disease involving breast/axilla from the past 30 years. It mainly affects women (median age: 48), consistently presenting as axillary lymphadenopathy, and demonstrates ipsilateral breast mass in half of patients (50%, 8/16). The breast mass was most commonly located in the upper outer quadrant (88%, 7/8), indicating the possibility of disease extension from axillary adenopathy. Around half of patients (56%, 9/16) reported cat exposure. Histologically, most patients (93%, 14/15) presented as necrotizing granulomas, with characteristic stellate-shaped necrosis in 5 patients. Although pathologic differential diagnoses between cat scratch disease and cancer are straightforward, distinguishing cat scratch disease from other granulomatous mastitis poses diagnostic challenges. Silver stains should be included in the diagnostic workup panel when highly suspecting cat scratch disease clinically. However, they were only able to highlight the causative microorganism in 54% (7/18) patients, and the gram stain was negative in all 12 tested patients. In contrast, polymerase chain reaction (PCR) for the causative microorganism was consistently positive in all 3 tested patients, while serologic test confirmed diagnosis in 85% (11/13) patients; 1 patient with negative serology showed a positive PCR result. 6-Hydroxynicotinic acid emerged as a pivotal intermediate for fine chemicals. The current study aims to screen novel nicotinate dehydrogenase-producing strains. Based on the high-throughput UV coloration method, a novel nicotinate dehydrogenase-producing strain named Bacillus paramycoides was first and successfully screened from soil and gram staining as 16S rRNA gene sequencing was conducted to further confirm its accuracy. The fermentation medium formula is determined as seignette salt (5 g/L), tryptone (10 g/L), K2HPO4 (2 g/L), and nicotinic acid (4 g/L) after a single factor and orthogonal test. The activity of nicotinate dehydrogenase produced by B. paramycoides after optimization reached 1.23 U/mL, which was the highest ever reported. In addition, the enzyme characterization results revealed that the optimum reaction temperature and pH were 40 °C and 7.0, respectively. Furthermore, B. paramycoides nicotinate dehydrogenase exhibited good thermostability with more than 50% of the highest enzyme activity retained at 50 °C after 8 h incubation, which was better than ever reported. The pH stability results suggested that nicotinate dehydrogenase possessed a narrow pH ranging from 6.0 to 8.0. In addition, shaker speed demonstrated a positive relation with enzyme activity since shaker speed could affect mass transfer and dissolved oxygen concentration. Finally, it could completely transform nicotinic acid to 6-HNA within 3 h when the substrate loading is 15 g/L. Background and Objectives: This study aims to identify types of bacterial species in women visiting obstetrics/gynecology centers in Riyadh City, Saudi Arabia, during different pregnancy trimesters. Materials and Methods: This cross-sectional study was conducted among pregnant and nonpregnant women seeking care at Alyamamah Hospital, obstetrics/gynecology center, Riyadh, Saudi Arabia. A total of 110 [pregnant = 90 and nonpregnant = 20] individuals were recruited based on inclusion/exclusion criteria. Personal data, plaque index (PI), and gingival index (GI) were recorded. Bacterial samples were collected using sterile absorbent paper points from the gingival sulcus of pregnant females during the first, second, and third trimesters and compared to a control group of nonpregnant females. Thioglycolate broth containing the absorbent paper points was incubated at 37 °C for 24–48 h. After growth, the microorganisms were subjected to a Gram stain. The VITEK 2 system and conventional methods were used to identify various types of bacterial species from the gingival sulcus of pregnant and nonpregnant women. Chi-square tests and nonparametric tests were applied to the data. Results: The bacterial characterization indicated that Actinomyces naeslundii (AN) was the most predominant bacteria found in the study participants, followed by Lactobacillus fermentum (LF) (23.6%), Veillonella (VL) (10%), and unidentified organisms (9.1%). When the presence of subgingival bacterial species was compared between pregnant and nonpregnant women, a statistically significant difference was observed (p < 0.001). LF was the predominant bacteria in 9 nonpregnant women (45%) and 8 pregnant women in the first pregnancy trimester (44.4%). However, during the second (17 women: 48.6%) and third pregnancy (17 women: 45.9%) trimesters, AN becomes the predominant bacteria. A statistically significant difference was observed when the prevalence of various bacterial species was compared across the three pregnancy trimesters (p = 0.010). The plaque and gingival scores of pregnant and nonpregnant women showed no significant difference. Irritable bowel syndrome (IBS) is a prevalent gut disorder linked to changes in the gut microbiota, including lactic acid bacteria (LAB). However, research on LAB biodiversity in IBS patients is limited. This study aimed to compare LAB microbiota in healthy individuals and those with IBS through biochemical and molecular techniques. Fecal samples from 15 IBS patients and 13 healthy individuals were collected, and LAB were isolated using biochemical methods. Fifty isolates were chosen based on Gram staining and catalase tests and identified through 16S rRNA gene sequencing. A phylogenetic tree was used to analyze strain diversity, and correlation diagrams and swarm plots were employed to explore variable relationships. The study revealed a significant difference in LAB numbers between IBS and healthy subjects, with average of 5.91 and 6.63, respectively. Most bacteria were Gram‐positive cocci or bacilli, with homofermentative characteristics, except for one heterofermentative sample from the healthy group. Both IBS and healthy groups exhibited strains from Lactobacillus and Enterococcus genera, with Enterococcus faecium being predominant in both. Demographic analysis showed higher IBS prevalence among individuals aged 20–40, with IBS‐C more common in women and IBS‐D in men. The study concluded that individuals with IBS had significantly lower LAB microbiota counts, potentially impacting intestinal defense function. Background: Antibiotic-resistant Salmonella is one of the main public health concerns in the world. Isolation of Salmonella in abattoirs has been considered the core source of infection in the community from meat. Still, there is limited information on the contamination rate of cattle carcasses. Objective: This study aimed to document the occurrence and antimicrobial susceptibility profile of Salmonella species recovered from cattle carcass and abattoir personnel at Dessie, municipality abattoir, Northeast Ethiopia: Methods: A total of 336 carcass swabs of abdomen, neck, and hind limb from cattle carcasses and 24 stool samples were collected from abattoir personnel using a systematic sampling method from February to April 2019. The collected samples were transported using Cary-Blair transport media and cultivated on Selenite cysteine F-broth, Brilliant green agar, and Xylose-lysine deoxycholate agar plates to isolate Salmonella species. Gram stain, colony morphology, and biochemical tests were performed to identify the isolated bacteria. An antimicrobial susceptibility test for Salmonella was performed using the Kirby-Bauer Disc Diffusion method. Descriptive statistics; both bivariable and multivariable logistic regression analysis was performed using SPSS version 25 software. P-value < 0.05 at 95% CI was considered statistically significant. Results: The prevalence of salmonella species was 8%(27/336) from all samples.'The prevalence of Salmonella isolates in cattle carcass and abattoir personnel was 8%(25/312) and 8.3%(2/24) respectively. The antimicrobial test showed that Salmonella species were 100% resistant to ampicillin, 59.3% to trimethoprim-sulfamethoxazole, 59.3% to tetracycline, and 55.6% to amoxicillin/clavulanate. From the total antimicrobial tested bacteria, 81.5%(22/27) were resistant to three and above classes of antibiotics (drug classes). Unwashed knives, carcasses, and hands of butchers during slaughtering were significantly associated (p < 0.05) with Salmonella found in carcasses. Conclusions: Salmonella isolation rates from cattle carcasses were high, with the bacteria showing notable resistance to most tested antibiotics. The interest in region-specific ethnic fermented foods and their functional microbiota is rising. The demands for functional foods are continuously rising, so research is going on to develop nutritious food with many beneficial attributes and low safety concerns. The present study was designed to isolate and characterize lactobacilli probiotic candidates from locally resourced fermented foods (dahi, lassi, and raabadi) to make ready-to-eat fermented functional products later. Cultures were isolated from 82 fermented food samples collected from different villages. The initial experiments of gram staining, catalase test, and carbohydrate fermentation were assessed for the morphology, purity, and primary characterization on the genus level, which was verified through molecular characterization using PCR. Seven lactobacilli strains (no. MS001-MS007) were then assessed for safety, probiotic candidacy, phytase degradation, and biofilm forming abilities. All seven bacterial cultures showed no hemolytic activity and antibiotic sensitivity against more than 14 antibiotics out of 20. All seven lactobacilli isolates were able to tolerate pH 3.0, 0.3% bile 0.5% pancreatin, lysozyme (100 mg/L to 300 mg/L) and also shown possessed phytase degradation ability. All the cultures showed antioxidative potential and biofilm formation ability. Culture MS007 showed considerably higher bile salt hydrolase activity among all the isolates, whereas MS005 possessed excellent phytate degradation ability among others. Bacterial strains were identified using 16S rRNA gene sequencing. Moreover, the order of preference of isolates was calculated using the multidimensional Technique for Order of Preference by Similarity to Ideal Solution (TOPSIS) based on probiotic and other functional properties. The most promising attributes showing cultures were recognised as Limosilactobacillus fermentum MS005 and Lactiplantibacillus plantarum MS007, which could be further used for functional food product development. Background: K. pneumoniae become multidrug-resistant (MDR) and commonly poses a serious health threat to patients due to limited therapeutic options. As a result, determining the prevalence and antimicrobial susceptibility patterns of K. pneumoniae isolates from clinical specimens is substantial to patient diagnosis and treatment. Methods and materials: A retrospective cross-sectional study was conducted from July 2021 to July 2022 at the University of Gondar Comprehensive Specialized Hospital, Northwest Ethiopia. Sociodemographic and laboratory data were collected from registered books using a data collection sheet. All types of samples were collected and processed using standard procedures. Identification of K. pneumoniae was done using Gram stain, colony characterization on culture media, anda series of biochemical tests. Antimicrobial susceptibility testing was done by the Kirby Bauer disc diffusion technique. The data were entered using Epi-info version 7 and exported to SPSS version 20 for analysis. Results: Among 2600 clinical specimens, 735 (28.3%) were positive for bacteria, and K. pneumoniae isolates accounted for 147 (20%). Most of them were isolated from neonates and mainly obtained from blood specimens (81.6%). These isolates were 100% resistant to Nalidixic acid, Cefotaxime, and Cefazolin. About 84% and 83.3% of the isolates were also resistant to Ceftriaxone and Tetracycline, respectively. However, they are sensitive to Nitrofurantoin (86.6%), Imipenem (85.7%), Meropenem (79%), and Amikacin (78.3%). The overall proportion of MDR K. pneumoniae isolates accounted for 57.1%. Conclusion: The magnitude of MDR K. pneumoniae was very alarming. Background: Aseptic meningitis comprises meningeal inflammation and cerebrospinal fluid (CSF) pleocytosis without positive Gram stain and culture. Regional differences exist in the prevalence of viral etiologies of aseptic meningitis. We aimed to assess the etiologies of aseptic meningitis in immunocompetent adults, focusing on herpes simplex virus type 2 (HSV-2). Methods: This study retrospectively analyzed immunocompetent adults diagnosed with meningitis at a Korean tertiary care hospital from 2016 to 2018. Aseptic meningitis was defined through clinical and CSF analysis. We compared clinical and laboratory characteristics across viral etiologies and investigated predictors of HSV-2 meningitis. Results: A total of 98 patients (46.9% female) with aseptic meningitis were finally enrolled. The etiologies of aseptic meningitis were identified in 62 patients (63.3%), including enterovirus (28.5%), HSV-2 (16.3%), and varicella zoster virus (VZV, 15.3%). HSV-2 showed female predominance, with shorter admission times with longer hospital stays and a recurrent meningitis history. Compared to other viral etiologies, HSV-2 showed higher CSF white blood cell (WBC) counts and protein levels but lower C-reactive protein (CRP) levels. A random forest model identified previous meningitis history and serum CRP level as key predictors of HSV-2 meningitis. Conclusions: This study provides insights into the etiologies of aseptic meningitis in a specific Korean region, identifying HSV-2 as a notable cause. Helicobacter pylori is a human pathogen bacterium associated with gastritis, peptic ulcer, and gastric cancer. It can be identified through the 16S rRNA gene and characterized through cagA and vacA virulence genes. Clinical cultures of H. pylori were isolated and identified from human stomach biopsies. The isolates were characterized according to their colonial and microscopic morphology, and molecular genotyping was conducted to determine the bacterial virulence. A phylogenetic analysis of the 16S rRNA gene sequencing was performed. In addition, multilocus sequence typing analysis was performed to determine the phylogeographic nature of the isolated strains. Three bacterial isolates were selected from 22 gastric biopsies, identified as H. pylori through colonial morphology, Gram staining, urease, catalase, and oxidase tests and identification of the ureC gene through end-point PCR. Amplification of 16S rRNA, urea, and tonB genes was performed, as well. Differences between the cagA and vacA genotypes were determined among the isolates. The phylogenetic analysis confirmed the identity of the three isolates as the specie Helicobacter pylori. Different genotypes were obtained for each H. pylori strain, and all the clinical isolates showed the vacA s2/m2 genotype, indicating an absence of the VacA cytotoxin. Only HCGDL-MR01 is a cagA gene carrier with a greater risk to develop a serious disease, such as stomach cancer and peptic ulcer. An 18‐year‐old healthy male complained of a 7‐day history of fever, cough, and sore throat, along with a three‐day history of left facial swelling. The x‐rays revealed subcutaneous emphysema in the chest, neck, face, and mediastinum region (Pneumomediastinum). Furthermore, an area of infiltration was visible, indicating pneumonia. Therefore, we immediately started him on intravenous antibiotics. We then moved the patient to an isolation room, considering pulmonary tuberculosis as one of the differential diagnoses. However, the Acid Fast Bacilli (AFB), Mycobacterium Tuberculosis Bacteria‐Polymerase Chain Reaction (MTB PCR), and sputum for gram stain and culture were all negative. On the other hand, the test for parainfluenza virus 3 was positive. The patient was observed with a daily chest x‐ray to monitor the progress of pneumonia and subcutaneous emphysema. Objective: To determine the antibiotic spectrum of deep skin and soft tissue infections in Pakistan. Study Design: Cross-sectional study. Setting: Department of Dermatology at Madinah Teaching Hospital, Faisalabad. Period: June, 2023 to December 2023. Methods: Inclusion criteria covered consenting patients aged 15-60, excluding those with specific conditions. The study aimed to ascertain antibiotic sensitivity patterns in deep skin and soft tissue infections. Ethical approval was obtained, and subjects meeting criteria were enrolled after informed consent, contributing valuable insights into healthcare-associated infections. Results: Examining 60 patients, the age distribution indicates a prevalent cohort up to 50 years (61.7%), highlighting potential health concerns in earlier years. Those above 50 constitute 38.3%, signifying distinct challenges in older age. With 61.7% males, Gram stain analysis reveals microbial diversity. Notably, Ciprofloxacin (90%) and Cefipime (98.3%) exhibit high resistance, while Cephradine (48.3%) and Cefoxitin (20%) show sensitivity. Meropenem and Vancomycin display moderate resistance (28.3% and 25%), while Tigecycline (10%) and Teicoplanin (1.7%) exhibit lower resistance, offering insights for effective treatment strategies. Conclusion: Our study reveals notable antibiotic resistance, with Ciprofloxacin (90%) and Cefipime (98.3%) exhibiting high resistance. Bacteriocins are proteins secreted by many species of bacteria to inhibit other bacteria, thus eliminating competitors to gain resources. Bacteria from the Lactobacillus group are known for their applications as probiotics and food preservatives. They have earned a reputation for producing substances that inhibit the growth of other microorganisms, which include organic acids, diacetyl, and bacteriocins. Produced by the ribosomes, bacteriocins are cationic proteins that inhibit other bacteria coexisting within a shared ecological habitat. Due to their potential uses in a variety of applications large-scale production of Bacteriocins would be necessary. The study aimed to identify and characterize Lactobacillus bacteria that produce potent bacteriocins and to analyze the antimicrobial activity and stability of the isolated bacteriocin under various physical and biochemical conditions. A total of 50 samples including buffalo dung, cheese, and rhizospheric region of plants were screened to isolate 8 Lactobacillus Li-1, Li-2, Li-3, Li-4, Li-5, Li-6, Li-7, and Li-8, confirmed by gram staining and other biochemical tests. The cell free supernatant from the Li-3 strain showed higher inhibition of Escherichia coli and Staphylococcus aureus, as compared to the other isolated strains. Li-3 strain was further identified as Levilactobacillus brevis strain ABRIINW-K by 16S rRNA gene sequencing. The bacteriocin isolated from this strain is a thermostable peptide (~6kDa), which is characteristic of class II bacteriocins, with potent antibacterial activity against Lactobacillus rhamnosus, Escherichia coli, and Salmonella enterica. Phytoestrogen-rich diet alters the composition of gut microbiota by enhancing the growth of beneficial bacteria and decreasing the microbial load of pathogenic organisms. Drosophila is an invertebrate model system used for research studies, as it shares 70% genetic homology with humans. The present study aimed to analyse microbiological profile of phytoestrogen rich supplement and its impact on gutmicrobiome composition in Drosophila melanogaster. The phytoestrogen rich supplement was mixed with formula 424 plain and flies were exposed to it. Gut of flies was dissected and cell suspension was prepared. Bacterial colonies were developed by streaking method. Gram staining was performed to differentiate the bacterial cells and further gut microbiome composition (Acetobacteraceae and Lactobacillales taxa) was analysed by 16S rRNA sequencing. The microbiological analysis was carried out to ascertain the microbial load of the developed product for consumption. The total bacterial count and coliform counts of the phytoestrogen rich supplement were <10 CFU/g. Also, the developed supplement exhibited minimal yeast and mold growth (<1 CFU/g). Gram staining showed gram positive (Bacilli and cocci). 16S rRNA sequencing showed significance with mild variation in similarity. It confirmed the presence of Bacillus paramycoids. The developed supplement has showed improved gut microbiome composition in the Drosophila. Hospitalized patients on mechanical ventilation are at high-risk of developing ventilator-associated pneumonia (VAP), making early and accurate diagnosis essential for the best possible treatment outcomes. This review examined various laboratory techniques, such as aerobic cultures, Gram's stain, and molecular techniques, to assess how well they diagnose VAP. We have done search strategies using Google Scholar, Medline Complete, and PubMed. Extensive statistical tools were utilized to examine studies and evaluate the diagnostic accuracy of laboratory results. Multiplex PCR was superior to Gram's stain and culture methods in terms of sensitivity (92%) and specificity (86%). On the other hand, Gram's stain showed the highest specificity (78.9%) and the lowest sensitivity (74.6%). The results of semiquantitative, quantitative, enrichment culture showed a lower specificity (75.97%) but a moderate sensitivity (78.5%). The most accurate diagnostic approach for VAP, according to a meta-analysis, was multiplex PCR-based testing, closely followed by culture methods. Beyond separate tests, multiplex PCR, culture, and Gram's stain combination increased sensitivity. Prompt and precise VAP diagnosis is essential for efficient treatment. With possible improvements through combination diagnostic techniques, multiplex PCR remains the most accurate diagnostic tool. Herbaspirillum huttiense is an opportunistic pathogen associated with rare cases of bacteremia. In this case report, H huttiense was isolated from blood samples collected from an intravenous catheter (incubated for 20.8 hours) and a peripheral vein (incubated for 14.16 hours) of a lung adenocarcinoma patient. Positive blood culture bottles were subjected to smear preparation, and Gram staining and microscopic examination revealed the presence of gram-negative rods in both aerobic bottles. We used the VITEK MS automatic microbial mass spectrometry system, VITEK 2 Compact automatic microbial analysis system, and high-throughput nucleic acid sequencing for accurate identification of the isolate. It is noteworthy that although the VITEK 2 Compact identified the isolate as Burkholderia cepacia , confirmation through VITEK MS mass spectrometry and 16S ribosomal DNA (rDNA) sequencing identified it as H huttiense. Subsequently, antimicrobial susceptibility testing was performed using the broth microdilution method, following the guidelines for nonfermenting gram-negative bacilli provided by the Clinical and Laboratory Standards Institute. This case highlights the possibility of misidentification of H huttiense as B cepacia by VITEK 2 Compact in certain situations, emphasizing the importance of considering uncommon pathogens, such as H huttiense , in the context of bacteremia in cancer patients. Background: Vancomycin-resistant enterococci (VRE) have become an increasing public health concern in the past few decades, being associated with serious multidrug-resistant (MDR) infections. This study was conducted to investigate the role of diarrheic pet animals as potential reservoirs for virulent extensively drug-resistant (XDR) VRE and their threat on human health. Materials and Methods: Rectal swabs were collected from 153 diarrheic pet animals (80 dogs and 73 cats). The collected swabs were cultured on CHROMagarTMVRE for the isolation of vancomycin-resistant Enterococcus faecalis and Enterococcus faecium, and then suspected colonies were identified as enterococci after Gram staining, conventional biochemical tests, and molecular techniques. VRE were basically identified using the disk diffusion method; however, molecular identification of vanA and vanB genes was carried out among confirmed VRE isolates. Moreover, three virulence genes (cytolysin A, cylA; enterococcal surface protein, esp; and hyaluronidase, hyl) were investigated in VRE isolates. Thereafter, VRE strains that harbored virulence genes were tested for antimicrobial susceptibility. Results: Eighteen out of 153 animals (11.8%) were positive for VRE, which were obtained from 15% and 8.2% of the examined dogs and cats, respectively. None of the obtained isolates carried the vanA gene, whereas the vanB gene was detected in E. faecalis (4/10) with a prevalence rate (40%). Of the obtained VRE isolates, five possessed esp and/or cylA, while all strains were negative for the hyl gene. Furthermore, four virulent VRE isolates exhibited an XDR pattern, and one isolate was MDR. Conclusion: Diarrheic pet animals could represent a potential zoonotic reservoir for virulent XDR vancomycin-resistant E. faecalis, which may have serious public health implications. Background: In February 2024, our hospital confirmed a case of ocular infection with Clostridium tertium caused by a salute gun explosion. The patient sought medical attention at our hospital due to a salute gun explosion injury in the right eye. Two days ago, a patient mistakenly believed that the fuse was not ignited when firing a salute gun. When observing, the salute gun exploded and injured his right eye. The patient immediately went to the local hospital for treatment. The CT scan of the local hospital showed rupture of the right eyeball. For additional diagnosis and treatment, the patient came to our hospital. The patient in this case has an acute onset, severe condition, no additional systemic diseases, and no history of drug or food allergies. Methods: Intraocular exploration, cranial CT, local and systemic anti infection treatment. Pathogen examination items: bacterial smear, bacterial culture and identification. Venous blood test items: blood routine, liver function, kidney function, and coagulation function. Results: Intraocular exploration showed conjunctival congestion and edema in the right eye, corneal haze and edema, shallow anterior chamber, anterior chamber hemorrhage, and unclear intraocular structure. Clinical treatment: debridement and suturing of right eye rupture + repair of eyeball rupture + removal of intraocular foreign body + repair of superior rectus muscle detachment + anterior chamber flushing + anterior chamber shaping + suture of eyelid laceration. Pathogen examination item: Eye secretion bacterial smear (Gram staining): A large number of gram-positive bacilli were found, and the secretion bacterial culture and identification (MALDI-TOF MS): Clostridium tertium. Auxiliary examination: Blood routine (venous blood): White blood cells 10.89 x 109/L, neutrophil count 9.65 x 109/L, whole blood hypersensitive C-reactive protein 20.28 mg/L, renal function: urea 9.15 mmol/L, uric acid 428.5 μmol/L, fasting glucose 6.48 mmol/L, no further abnormalities observed. Clinical drug treatment plan: Tetanus human immunoglobulin 250 IU im, tobramycin eye drops 0.1 g ext qd, vancomycin 0.5 g ih qd, levofloxacin 0.5g ivgtt qd, aluminum magnesium suspension 15 mL po bid, potassium chloride sustained-release tablets 0.5 g po qd. After 7 days of treatment, the patient's body temperature returned to normal, conjunctival congestion and edema decreased, anterior chamber hemorrhage decreased, corneal incision closed properly, and the patient improved and was discharged. Conclusions: This article reports a case of ocular infection caused by a salute gun explosion with Clostridium tertium. Clostridium tertium was quickly and accurately identified by a mass spectrometer, and reasonable treatment measures were adopted clinically. The patient improved and was discharged. Background: The incidence of hypermucoviscous Klebsiella pneumoniae (hmvKp), which complicates communityacquired pneumonia, has been increasing recently. This study aimed to detect hypermucoviscous K. pneumoniae and determine its antimicrobial susceptibility pattern in adult patients with community-acquired pneumonia in Northwest Ethiopia. Methods: This cross-sectional study included 39 K. pneumoniae isolates identified by using Gram stain, culture, and biochemical tests from 312 adult patients with community-acquired pneumonia at the University of Gondar Comprehensive Specialized Referral Hospital from April to June 2021. The hypermucoviscous strains were identified by using the string test. Antimicrobial susceptibility testing was performed by using the Kirby-Bauer disk diffusion method. Data were entered by using EPI data version 4.6 and were analyzed by using SPSS version 20. A pvalue ≤ 0.05 at a 95% confidence interval was considered statistically significant. Results: Overall, 35.9% (n = 14) of the 39 K. pneumoniae isolates were hypermucoviscous phenotype. The mean age of the hmvKp group was lower than of the cKp group (36.93 ± 12.573 vs. 53.52 ± 19.556 years, p = 0.007). All hmvKp isolates were resistant to amoxicillin-clavulanic acid and trimethoprim-sulfamethoxazole. Azithromycin resistance in the hmvKp strains was significantly higher than in the cKp group (p = 0.012). Conclusions: This study demonstrates that the hmvKp phenotype causes community-acquired pneumonia and a full resistance to amoxicillin-clavulanic acid and trimethoprim-sulfamethoxazole. Antimicrobial resistance was higher in the hmvKp strain than in the classic strains. Background: Actinomyces spp. are most commonly found in human commensal flora; however, they have also been shown to cause suppurative infections. We present a case of a rare Actinomyces funkei bacteraemia from an infected deep vein thrombosis in a patient who went on to develop pulmonary cavities secondary to septic emboli. Infected thrombi and septic emboli have been associated with other Actinomyces spp. in the literature, often posing a diagnostic challenge and, in some cases, causing drastic clinical deterioration in patients. The literature regarding Actinomyces funkei is scarce and to our knowledge there are no reports of a relationship between this Actinomyces subspecies and infected thrombi or septic emboli. Case presentation: The patient was a 39-year-old known intravenous drug user who presented with a groin injecting site sinus and systemic symptoms. The bacteria was first observed by gram staining of a blood culture sample after 48 h of incubation and the species was identified using matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) as Actinomyces funkei. Sputum cytology/histology with cell block revealed a branching gram-positive species suspicious of slow growing bacteria or fungus. CT imaging of his lower limb and chest revealed an extensive DVT with inflammatory changes and pulmonary cavities respectively. The patient was treated with Ceftriaxone before being discharged with a 6-month course of Linezolid. He made a good recovery with reduction in size of the cavitating lung lesions on follow-up imaging. Conclusions: This case report presents a difficult-to-diagnose bacterial infection in an intravenous drug user, complicated by bacteraemia and secondary septic emboli. Relatively little is known about Actinomyces funkei, and therefore this report aims to increase clinician awareness of diagnosis, management, and complications. [ABSTRACT FROM AUTHOR]

For the first time, five lead-resistant bacteria were isolated from the marine water and sediments of Versova Creek, Mumbai, India, and analysed for their bioflocculation-assisted bioremediation potential. Based on 16 S rRNA sequencing and BLAST, the isolates matched the genera Bacillus and Enterobacter. They exhibited robust growth in media amended with 1600 mg/l of lead nitrate, with a minimum inhibitory concentration (MIC) of 2000 mg/l. The optimum growth parameters were determined to be 37ºC and pH 7. Gram staining and colony morphology identified PbR01 and PbR02 as Gram-positive, while PbR03, PbR04, and PbR05 were Gram-negative. Bacillus sp. AKVPbR02 demonstrated exceptional lead removal efficiency, eliminating 74% and 89.5% of Pb(II) at an initial concentration of 400 mg/l after 72 and 120 h of incubation, respectively. Enterobacter sp. AKVPbR04 also exhibited high removal efficiency, achieving 89.49% removal by the 5th day (120 h) of incubation. Other strains, including Enterobacter sp. AKVPbR03, Bacillus sp. AKVPbR01, and Enterobacter sp. AKVPbR05, removed 87.70%, 83.52%, and 82.76% of Pb(II), respectively, by the same incubation period. The isolates displayed diverse biochemical profiles. All demonstrated urease activity except PbR05, while only PbR02 exhibited β-glucuronidase activity. Most isolates could metabolize a range of sugars, including sucrose, D-sorbitol, and D-trehalose. Nitrogen source utilization varied, with all isolates capable of utilizing lysine and ornithine except PbR05. Antibiotic sensitivity testing revealed varied resistance and susceptibility patterns. For example, PbR01, PbR02, PbR04, and PbR05 were sensitive to azithromycin and erythromycin, while PbR03 exhibited resistance. Background. Recent studies have revealed that antibiotics are yielding unsatisfactory outcomes in certain patients with acne. Therefore, attention was drawn to antibiotic resistance as a rapidly growing problem that might lead to treatment failure. Objective. The study aims to assess the antibiotic susceptibility patterns of Cutibacterium acne isolates in a sample of Egyptian patients to the antibiotics most frequently prescribed for acne vulgaris. Methods. A total of 155 patients with moderate to severe acne were included in the study. Skin swab samples were collected and inoculated. Cutibacterium acnes was identified based on colonial morphology, Gram stain, and biochemical reactions and further confirmed by VITEK 2 automated systems. Antibiotic susceptibility tests were performed using minimum inhibitory concentrations determined by the Epsilometer test. Results. Cutibacterium acnes was detected in 50 samples (32.2%), alone in 17 (10.9%), and in combination with Staphylococci in 33 (21.3%) cases. The results of the antibiotic susceptibility testing revealed high resistance to erythromycin, followed by clindamycin, tetracycline, trimethoprim/sulfamethoxazole, and doxycycline. Conclusion. Background: Invasive Aspergillosis is a fungal infection caused by Aspergillus species, typically posing life-threatening risks to immunocompromised individuals. While occurrences in immunocompetent hosts are rare, a recent case report documented fulminant pulmonary aspergillosis in an immunocompetent patient during autopsy. Here, we present a case of invasive aspergillosis in an immunocompetent woman, manifesting with disseminated lesions. Case presentation: A 29-year-old Asian woman presented to our hospital in March 2022, reporting chest pain and shortness of breath persisting for two months. Upon examination, she appeared thin and unwell, with no notable abnormalities otherwise. Radiographic imaging revealed an ill-defined lesion in her left lung. Subsequent bronchoscopy and lavage were performed, followed by initiation of empirical antibiotic therapy. Lavage results were negative for gram staining, culture, and ZN staining for AFB, but revealed numerous septate hyphae on fungal smear. Histopathological examination indicated chronic granulomatous inflammation with septal fungal hyphae, indicative of aspergillosis. Subsequent culture confirmed Aspergillus species, prompting initiation of voriconazole therapy. Remarkably, the patient exhibited significant improvement, with weight gain and restored appetite observed within a short period. Within 2 months of treatment, her symptoms resolved, and she resumed near-normal daily activities. Conclusion: This case highlights the diagnosis of aspergillosis in an immunocompetent individual presenting with disseminated nodular lesions across the lungs, mediastinum, and abdomen. There is an enormous diversity of life forms present in the extremely intricate marine environment. The growth and development of seaweeds in this particular environment are controlled by the bacteria that settle on their surfaces and generate a diverse range of inorganic and organic chemicals. The purpose of this work was to identify epiphytic and endophytic bacterial populations associated with ten common marine macroalgae from various areas along the Mediterranean Sea coast in Alexandria. This was done to target their distribution and possible functional aspects. Examine the effects of the algal habitat on the counting and phenotypic characterization of bacteria, which involves grouping bacteria based on characteristics such as shape, colour, mucoid nature, type of Gram stain, and their ability to generate spores. Furthermore, studying the physiological traits of the isolates under exploration provides insight into the optimum environmental circumstances for bacteria associated with the formation of algae. The majority of the bacterial isolates exhibited a wide range of enzyme activities, with cellulase, alginase, and caseinase being the most prevalent, according to the data. Nevertheless, 26% of the isolates displayed amylolytic activity, while certain isolates from Miami, Eastern Harbor, and Montaza lacked catalase activity. Geographical variations with the addition of algal extract may impact on the enumeration of the bacterial population, and this might have a relationship with host phylogeny. The most significant observation was that endophytic bacteria associated with green algae increased in all sites, while those associated with red algae increased in Abu Qir and Miami sites and decreased in Eastern Harbor. At the species level, the addition of algal extract led to a ninefold increase in the estimated number of epiphytic bacteria for Cladophora pellucida in Montaza. Notably, after adding algal extract, the number of presented endophytic bacteria associated with Codium sp. increased in Abu Qir while decreasing with the same species in Montaza. Background: Ocular infections, including conjunctivitis, keratitis, and endophthalmitis, significantly impact eye health, leading to morbidity, visual impairment, and blindness if not treated. Aims and Objectives: This study focuses on reviewing and analyzing the prevalence of bacterial and fungal pathogens in ocular infections and assessing their antimicrobial susceptibility patterns at Vijayanagar Institute of Medical Sciences, Ballari. Materials and Methods: A cross-sectional study was conducted by collecting ocular infection samples from January 01, 2022, to December 31, 2022. Samples, including corneal scrapings and conjunctival swabs, underwent initial procedures such as Gram staining and were inoculated on various agars. Antimicrobial susceptibility testing was performed using the Kirby-Bauer disk diffusion method. Results: Out of 404 evaluated samples, corneal scrapings were predominant, with a significant incidence of fungal and bacterial growth. Aspergillus spp. and Pseudomonas spp. were the most common fungal and bacterial isolates, respectively. Antimicrobial susceptibility testing revealed that Gram-negative bacilli showed more susceptibility to gentamycin, ceftriaxone, and ciprofloxacin, whereas Grampositive cocci were more susceptible to tetracycline and doxycycline. Conclusion: The study highlights the cornea as the most common source of ocular infections, with Aspergillus and Pseudomonas spp. being the predominant fungal and bacterial pathogens, respectively. Identifying the prevalent species in this geographical area aids in early diagnosis and treatment, thereby preventing morbidity and improving patient outcomes. The poultry market is troubled by Salmonellosis infections which has a large negative impact due to morbidity and decreased productivity. Salmonella pullorum causes a disease named Salmonellosis which affects the poultry industries and has long been a significant obstacle to the development of nations. Due to the occurrence of resistance to most of the antibiotics in the poultry, treatment of infection now becomes difficult and challenging. So, alternative therapy is needed to reduce the burden of bacterial load and to treat the infection. The main objectives of this study were to isolate Salmonella pullorum from the layer poultry birds with to check antibiotic resistance of Salmonella pullorum and to isolate lytic phages against Salmonella pullorum. Poultry fecal samples were collected from poultry farms in different areas. For isolation and purification of Salmonella pullorum, Salmonella Shigella agar (SS-agar) was used. Black centered colonies were observed on SS agar and non-lactose fermenter on MacConkey's agar. For the confirmation of bacterial isolates, gram staining yielded pink-red rods as observed microscopically. For citrate utilization isolates also +ve test, catalase production and methyl red reaction and negative for VP and indole reaction. The disk diffusion method on Mueller Hinton was performed to check the susceptibility pattern of Salmonella pullorum. Bacterium showed resistance to amoxicillin followed by tetracycline and ceftazidime, while sensitive to chloramphenicol and kanamycin. Sewage water was collected for the isolation of phages from different sewage lines of poultry farms. Bacteriophages against Salmonella pullorum were isolated through agar overlay method. Qualitative urinalysis using the Sternheimer stain is a common method in Japan for identifying bacteriuria, but there is a lack of studies examining its test characteristics. In this study, we aimed to investigate the sensitivity and specificity of the Sternheimer stain for urine culture results and compare it with the sensitivity and specificity of the Gram stain. Our goal was to determine the usefulness of the Sternheimer stain in identifying bacteriuria. Among 986 patients aged 16 years or older from whom samples for both urinalysis and urine culture were obtained at the emergency room of Tenri Hospital from January 2019 to December 2019, 342 patients with pyuria, defined as the presence of 10 or more white cells per cubic millimeter in a urine specimen, who had not received prior antimicrobial therapy were included. Urine cultures were used for comparison to determine the sensitivity and specificity of Sternheimer and Gram stain in this patient group. A positive Sternheimer stain result was defined as bacteriuria ≥ (1+), and that of Gram stain was defined as ≥ 1/1 field of high-power (× 1000) oil immersion. Using urine culture results for comparison, the sensitivity of Sternheimer stain was 92.2%, the specificity was 48.5%, the positive likelihood ratio was 1.79, and the negative likelihood ratio was 0.16. Objective: Unknown etiology. Background: Meningococcal meningitis is rare in Japan; however, when outbreaks do occur, they predominantly involve domestically infected cases rather than those contracted overseas. Case Report: A Japanese man with diabetes in his 50s experienced fever and loss of consciousness, with no history of international travel. In our hospital, gram-negative diplococci were detected in the cerebrospinal fluid (CSF) of the patient by Gram staining, although the rapid agglutination test and cultures of blood and CSF were negative. Multiplex polymerase-chain reaction (PCR) testing returned positive results for meningococcus and parechovirus. Brain MRI revealed a finding of meningitis, but there were no indications of encephalitis. To determine the serotype and genotype, we sent the sample to the National Institute of Infectious Diseases, which identified the serogroup and sequence type (ST) as type B and 2057, respectively. Despite the unknown antimicrobial susceptibility, the patient responded well to empirical treatment with ceftriaxone at 2 g every 12 h, and was discharged with remaining symptoms of dizziness, headache, difficulty hearing in the left ear, and tinnitus in the left ear. Conclusions: In Japan, vaccines covering serogroups A, C, and W/Y are available but not routinely administered. According to epidemiological surveillance reports, serogroup B is the second most common cause of meningococcal meningitis in Japan, yet there is no corresponding vaccine available in the country. This case has prompted a review of the epidemiology of meningococcus in Japan, encompassing strategies for vaccination and hospital infection control to prevent droplet transmission, which includes post-exposure prophylaxis when no prior measures have been implemented. Background: Ruthenibacterium lactatiformans, a Gram-stain-negative, rod-shaped, obligate anaerobic bacterium of the Oscillospiraceae family, has not been previously reported in human infections. This study reports the first case of bacteraemia and potential vertebral osteomyelitis caused by Ruthenibacterium lactatiformans. Case presentation: An 82-year-old man with a history of diabetes, chronic renal failure, and prior spinal surgery for spondylolisthesis and spinal stenosis presented with fever and lower back pain. Magnetic resonance imaging revealed multiple vertebral osteomyelitis lesions. Initial blood cultures identified methicillin-resistant Staphylococcus aureus (MRSA), which prompted vancomycin treatment. However, repeated blood cultures not only confirmed persistent MRSA, but also detected Gram-negative bacilli (GNB). Despite surgical removal of the spinal hardware and antimicrobial therapy, the patient's osteomyelitis worsened, necessitating transfer for further management. Subsequent analysis using 16S rRNA gene sequencing identified the GNB as Ruthenibacterium lactatiformans. Conclusions: This is the first documented instance of human infection with Ruthenibacterium lactatiformans, signifying its pathogenic potential in vertebral osteomyelitis. The involvement of anaerobic bacteria and the possibility of polymicrobial infections complicate the diagnosis and treatment of vertebral osteomyelitis. Community‐acquired pneumonia (CAP) is a major cause of morbidity and mortality worldwide. It also contributes significantly to hospital admissions, particularly in low‐income countries such as Ethiopia, where it accounts for major public health problems. This could be attributed to the increasing prevalence of antibiotic‐resistant pathogens in CAP patients. This study aimed to identify and assess the antibiotic resistance patterns of bacterial isolates from CAP patients at the Adama Hospital Medical College in Adama City, Ethiopia. A cross‐sectional study was conducted from November 10, 2022, to November 30, 2023. Demographic, clinical data, and sputum samples were collected from patients with CAP (n = 369). Sputum samples were subjected to standard microbiological procedures, including culture, Gram staining, and a panel of different biochemical tests for the identification of pathogenic bacterial isolates. The Kirby–Bauer disc diffusion method was used for drug susceptibility testing. Descriptive statistics were computed by using SPSS (version 26). Of the 369 patients with CAP, bacterial pathogens were identified in 31.7% (n = 117, 95% CI: 27.0%–36.7%). The most common isolates were Moraxella catarrhalis (n = 15; 12.8%), Staphylococcus aureus (n = 15; 12.8%), Klebsiella pneumoniae (n = 12; 10.3%), Escherichia coli (n = 11; 9.4%), Pseudomonas aeruginosa (n = 11; 9.4%), Enterobacter species (n = 11; 9.4%), and Citrobacter species (n = 11; 9.4%). Among the identified isolates, resistance rates were high in Enterobacteriaceae, followed by Gram‐positive bacteria, and non‐Enterobacteriaceae. Overall, 68 (58.1%) of the identified bacterial isolates were multidrug resistant (MDR), with K. pneumoniae accounting for the highest proportion of multidrug resistant isolates (91.7%), while P. aeruginosa accounted for the lowest proportion (9.1%) of MDR isolates. This study revealed a high prevalence (31.7%) of bacterial pathogens in CAP patients and higher (58.1%) MDR bacterial pathogens. 【Objective】The causative agent was isolated from the diseased Micropterus salmoides tissue. The physiological properties of the pathogenic bacteria were identified, and their virulence genes and drug resistance genes were analyzed, with an aim to provide guidance for the scientific use of drugs in clinical practice.【Method】Samples were collected from pathologic tissues of diseased M. salmoides from a fish breeding factory in Zhanjiang, Guangdong Province, China. The bacteria were isolated by dilution coating method, the 16S rRNA sequence comparison and biochemical tests were performed to identify the species, and the virulence genes, drug resistance genes and drug sensitivity characteristics were also identified. 【Result】Bacteria with round colony morphology, smooth milky white surface and opaque color were isolated from pathologic tissues of M. salmoides, and numbered as MSB-2, which was identified as Aeromonas in Gram-negative bacteria through phenotypic identification by Gram staining and physiological and biochemical reactions. By amplifying the 16S rRNA gene sequence and constructing a phylogenetic tree, the bacterium was further identified as A. veronii. The strain had virulencerelated genes such as act, aer and aexT and resistance-related genes such as tetC, tetA and sull. The results of the drug sensitivity test showed that the bacterium was sensitive to carbenicillin, doxycycline, cefalexin, neomycin, gentamicin, cefradine, minocycline, cefazolin and etc. Artificial regression of healthy M. salmoides infected with A. veronii showing clinical signs similar to those of diseased fish, which were the same as those of the naturally occurring disease, such as congestion of the ventral and pectoral fin bases, redness and swelling of the anus, and obvious enlargement of the abdomen, etc. And a large amount of fluid in the abdominal cavity was found by autopsy.【Conclusion】The above results indicate that A. veronii is the pathogenic bacterium causing this disease of M. salmoides, and the approved fishery drugs such as neomycin and doxycycline can be used to control the disease during the breeding process. 【Objective】In recent years, probiotics have been widely used in aquaculture, but the probiotics derived from Trachinotus ovatus been rarely reported. In the study, a strain was isolated from the intestine of Trachinotus ovatus and its biological characteristics were analyzed, which provided a theoretical basis its application in T. ovatus culture.【Method】 The bacteria were isolated and identified through the sterile collection of intestinal contents from the healthy T. ovatus. And the morphological observation, gram staining, biochemical identification, 16S rRNA gene sequence analysis, drug sensitivity test, salt tolerance test, acid production ability, growth curve determination, and virulence gene identification of the typical bacterial colonies were performed.【Result】Based on the morphological characteristics, gram staining results biochemical characteristics, and 16S rRNA gene sequence alignment of the isolated bacterium, the results showed that it shared 100% similarities with Enterococcus faecalis, therefore, the isolated bacterium was identified as E. faecalis. The colony of the isolated strain was light grayish white, and Gram-positive. The biochemical indicators including esculin, cellobios, maltose, mannose and salicylic acid were tested positive. The results of the drug sensitivity test showed that the isolated bacterium exhibited varying degrees of resistance to drugs such as piperacillin, polymyxin B, penicillin, trimethoprim-sulfamethoxazole and oxacillin, exhibited intermediate sensitivity to ciprofloxacin, medemycin and ceftriaxone and sensitive to 11 drugs such as vancomycin, cefoperazone, ofloxacin and etc. The results of the salt tolerance test showed that the isolated strain could grow at 0%-8% NaCl concentrations, showing salt tolerance to some extent. The results of acid production capacity and growth curve measurements indicated that the pH of the culture media rapidly decreased and the acid production capacity was enhanced as the proliferation of the strain. The identification results of virulence genes showed that the isolated strains possessed four virulence genes including efaA, agg, as and fsrA.【Conclusion】A potential probiotic strain was successfully isolated from the intestine of T. ovatus and identified as E. faecalis. The isolated strain has good salt tolerance and acid production capability, and it contains functional genes associated with intestinal adhesion, which will provide a theoretical basis for further application of the strain. Pseudomonas aeruginosa (P. aeruginosa) is a multidrug-resistant (MDR) environment associated microorganism that causes infections in animals and human. The present study aimed to determine antibiotic susceptibility patterns and molecular characteristics of P. aeruginosa. The molecular typing of P. aeruginosa isolates was also performed. Poultry meat and meat product samples (n=110) were collected. The samples were inoculated on MacConkey and cetrimide agar for cultural identification and isolation. Gram staining and biochemical tests were performed for confirmation. The confirmed isolates of P. aeruginosa were then used for antibiotic susceptibility testing against commonly used antibiotics including meropenem, amikacin, aztreonam, azithromycin, enrofloxacin, and gentamicin. The virulence genes including exoU, exoS, lasR, rhlR, algD, pslD, and blaTEM were detected in selected MDR isolates of P. aeruginosa. Among the poultry meat samples, P. aeruginosa was more prevalent (50%) in fresh meat samples than others [frozen meat (40%) and meat products (8%)]. Confirmed isolates were Gram-negative, catalase positive, oxidase positive, beta-hemolytic, and citrate-positive while colorless growth on MacConkey's agar and green color growth on cetrimide agar was observed. In antibiotic sensitivity testing, results indicated that in poultry meat and meat product isolates, all P. aeruginosa isolates were highly resistant to azithromycin (83%). High resistance rate was also observed for aztreonam (67%), gentamicin (58%) and enrofloxacin (50%) and all P. aeruginosa isolates were highly sensitive to meropenem (84%) followed amikacin (75%). In molecular detection of virulence genes, poultry meat samples, exoU, algD, and their genes showed comparable prevalence (80%). The prevalence of drug resistance genes in P. aeruginosa from poultry meat samples was as follows; blaTEM (95%), tetR (0%), exoU (25%), exoS (30%), algD (80%), pslD (85%), lasR (85%) and rhlR (80%). In summary, the presence of virulent genes in P. aeruginosa isolates enables them to harbor antibiotic resistance and acts as an MDR public health pathogen with the potential to transfer to humans via the food chain. Fungal and bacterial bioaerosols have the potential to induce biodeterioration in books and archival materials stored within libraries through their metabolic processes. The progression of these processes is dependent on microclimatic factors, including relative humidity and temperature. At the national level, research on biodeterioration in libraries and investigations into bioaerosols in indoor environments have been limited. These topics are of great significance as they have an impact on both public health and book preservation. This study aims to determine the microbial load within the indoor air of the library situated on the campus of Universidad Militar Nueva Granada in Cajicá, Colombia. Simultaneously, it seeks to investigate the microclimatic conditions of this environment. We used a MAS-100 Eco air sampler with an airflow rate of 100 L/min for a duration of 2.5 minutes. Nutrient agar and rose Bengal were employed as culture media. The samples were incubated for 8 days, and colony counting was performed on Petri dishes. For fungal identification, we used lactophenol blue staining, and Gram staining was employed for bacterial classification. Microclimatic conditions were continuously monitored using a data logger equipped with a USB connection. The results revealed microbial loads ranging from 220 to 921 CFU/m³, with temperatures fluctuating between 18.2 and 18.6°C, and relative humidity levels within the range of 60.8% to 64.7%. The identification of fungal genera, including Aspergillus, Penicillium, and Cladosporium, as well as both Gram-positive and Gram-negative bacteria, was successfully achieved. An interesting observation emerged, indicating a corresponding increase in the concentration of fungal aerosols as relative humidity levels rose. Objectives 1. To determine the etiological profile of suspected cases of bacteremia 2. To establish the antibiotic susceptibility pattern of bacterial isolates Materials and Methods: The retrospective study was carried out from January 2022 to December 2022 at Islamabad International Hospital. A total of 639 blood culture isolates were processed manually onto blood agar, chocolate agar, MacConkey’s agar, and bile-esculin agar and were identified on their morphological appearance using gram staining. Isolates were further identified at the species level using the Analytic Profile Index (API). Antibiotic susceptibility testing was performed using the Disk Diffusion method as per Clinical and Laboratory Standards Institute guidelines (M-100 Version 2021) Results: Amongst 639 blood culture isolates, only 66 (10.32%) were culture-positive. Prevalent gram-negative bacilli included Salmonella typhi (n= 19) and Escherichia coli (n= 8) whilst Enterococcus (n=4), and Staphylococcus aureus (n=4) were the predominant gram-positive cocci. Salmonella typhi and S. aureus showed 100% resistance to ciprofloxacin whilst enterococcus displayed 75% resistance. Overall, all strains (n=6) of K. pneumoniae and 63% of E. coli showed resistance to over 4 antibiotics. Conclusion: Gram-negative bacteria were the main cause of BSIs, where the predominant microorganism was S. typhi. Remarkably all S. typhi isolates were completely resistant to ciprofloxacin. Probiotics can enhance the growth performance and disease resistance of aquatic animals. Probiotics can influence digestive physiology through the secretion of exogenous digestive enzymes, facilitating the utilization of nutrients, and resulting in improvements in growth performance. The objectives of the present study were to reduce fish diseases, minimize the usage of antibiotics, and promote national policy on aquatic health management in Oman through enhancing immune stimulation mechanisms with probiotics from indigenous Omani products and finally to promote Oman's aquaculture industry. Thirty-five kinds of edible products were screened. After the primary screening process, only 3 products contained appropriate amounts of probiotics, therefore the present study focused on 3 probiotics from Omani products, i.e., dried date, date vinegar, and camel milk. Gram staining, VITIK2, and DNA tests were used for the identification and characterization of bacterial strains. DNA sequence revealed that the bacteria isolated from dried dates were identified as Bacillus licheniformis. Those from date vinegar were identified as Bacillus vallismortis. A viability test with pepsin, pH 3 showed that the bacteria isolated from camel's milk (Bacillus stratospheric) had a greater tolerance at low pH followed by bacteria isolated from dried date (B. licheniformis). Providencia genus is known to harbor certain opportunistic pathogens capable of causing human infections. Here, we report two strains of multidrug-resistant bacteria initially identified as Providencia rettgeri by mass spectrometry, but genome analysis revealed their ANI (79.84–84.20%) and dDDH (21.1–25.6%) values to fall below the accepted species threshold for known Providencia species. We therefore propose that these isolates be recognized as a novel species, Providencia xianensis sp. nov. Alarmingly, both strains, isolated from locations far apart, exhibited resistance to last-resort antibiotics, indicating their possible wide distribution, underscoring the urgency for immediate attention and enhanced surveillance for this emerging multidrug-resistant pathogen.
