Abstract

The concept of "endocrine disruptor" was first introduced in 1991 by Theodora Colborn at the Wingspread conference. This expression refers to molecules capable of interacting with the hormonal functioning of living organisms and thus causing harmful effects on health and the environment. They are unlike other substances as they are able to act via their own action mechanisms. Indeed, endocrine disruptors cause stronger effects at low doses than at high doses. They are also capable of interacting with each other, this is called the cocktail effect. That is to say that the substances, individually, have no effect but can induce it if they are present together in the body. The effects of endocrine disruptors are latent in the long term and can be transgenerational. Therefore, when a pregnant woman is exposed, her fetus is also exposed. This exposure has an impact on the baby but also on its descendants. Thus it appears that certain periods of life such as pregnancy, infancy and puberty are more vulnerable to these substances. Nowadays, we see an increase in a large number of chronic diseases in which endocrine disruptors are suspected to participate in their development. These include obesity, cardiovascular diseases, diabetes, hormone-dependent cancers, reproductive diseases and autistic or hyperactivity disorders. Therefore, it seems important to teach anyone to recognize these substances and to identify them in their daily lives. Indeed, they are omnipresent: in the air, water, soils, food, everyday objects, etc. It is crucial to offer simple prevention advice, especially on PEs in everyday consumer products.; Le concept de “perturbateur endocrinien” a été introduit pour la première fois en 1991. Cette expression fait référence à des molécules capables d'interagir avec le fonctionnement hormonal des organismes vivants et ainsi entraîner des effets néfastes sur la santé et sur l’environnement. Ce ne sont pas des substances comme les autres, elles sont capables d’agir via des modes d’action qui leur sont propres. En effet, les perturbateurs endocriniens (PE) provoquent des effets plus forts à faible dose qu’à forte dose. Ils sont aussi capables d'interagir entre eux, c’est l’effet cocktail. C'est-à-dire que les substances, individuellement, n'ont pas d’effet mais peuvent en induire si elles sont présentes ensemble dans l’organisme. Les effets des PE sont latents à long terme et peuvent être transgénérationnels. Dès lors lorsqu’une femme enceinte est exposée, son fœtus l’est aussi. Cette exposition a un impact sur le bébé mais également sur sa descendance. Ainsi il apparaît que certaines périodes de la vie comme une grossesse, la petite enfance et la puberté sont plus vulnérables face à ces substances. Aujourd'hui, nous constatons une augmentation d’un grand nombre de maladies chroniques et les PE sont suspectés de participer à leur développement. Il s’agit par exemple d'obésité, de maladies cardiovasculaires, de diabète, de cancers hormonodépendants, de pathologies de la reproduction et de troubles autistiques ou d'hyperactivité. Ainsi, il apparaît important d'apprendre à tout un chacun à reconnaître ces substances et de les identifier dans sa vie quotidienne. En effet, ils sont omniprésents : dans l’air, dans l’eau, dans les sols, dans l’alimentation, dans les objets du quotidien, etc. Il est crucial de proposer des conseils de prévention simples, notamment sur les PE présents dans les produits de consommation courante.

Abstract

A healthy ageing process is important when it is considered that one third of the population of Europe is already over 50 years old, although there are regional variations. This proportion is likely to increase in the future, and maintenance of vitality at older age is not only an important measure of quality of life but also key to participation and productivity. So, the binomial nutrition and ageing has different aspects and poses considerable challenges, providing a fertile ground for research and networks. The NutRedOx network will focus on the impact of redox active compounds in food on healthy ageing, chemoprevention and redox control in the context of major age-related diseases. The main aim of the NutRedOx network is to gather experts from Europe, and neighboring countries, and from different disciplines that are involved in the study of biological redox active food components and are relevant to the ageing organism, its health, function and vulnerability to disease. Together, these experts will form a major and sustainable EU-wide cluster in form of the NutRedOx Centre of Excellence able to address the topic from different perspectives, with the long-term aim to provide a scientific basis for improved nutritional and lifestyle habits, to train the next generation of multidisciplinary researchers in this field, to raise awareness of such habits among the wider population, and also to engage with industry to develop age-adequate foods and medicines.

Abstract

International audience; RATIONALE:The potency of S-nitrosoglutathione (GSNO) as a nitric oxide (NO) donor to treat cardiovascular diseases (CVDs) has been highlighted in numerous studies. In order to study its bioavailability after oral administration, which represents the most convenient route for the chronic treatment of CVDs, it is essential to develop an analytical method permitting (i) the simultaneous measurement of GSNO metabolites, i.e. nitrite, S-nitrosothiols (RSNOs) and nitrate and (ii) to distinguish them from other sources (endogenous synthesis and diet).METHODS:Exogenous GSNO was labeled with 15 N, and the GS15 NO metabolites after conversion into the nitrite ion were derivatized with 2,3-diaminonaphthalene. The resulting 2,3-naphthotriazole was quantified by liquid chromatography/tandem ion trap mass spectrometry (LC/ITMS/MS) in multiple reaction monitoring mode after Higher-energy Collision-induced Dissociation (HCD). Finally, the validated method was applied to an in vitro model of the intestinal barrier (monolayer of Caco-2 cells) to study GS15 NO intestinal permeability.RESULTS:A LC/ITMS/MS method based on an original transition (m/z 171 to 156) for sodium 15 N-nitrite, GS15 NO and sodium 15 N-nitrate measurements was validated, with recoveries of 100.8 ± 3.8, 98.0 ± 2.7 and 104.1 ± 3.3%, respectively. Intra- and inter-day variabilities were below 13.4 and 12.6%, and the limit of quantification reached 5 nM (signal over blank = 4). The permeability of labeled GS15 NO (10-100 μM) was evaluated by calculating its apparent permeability coefficient (Papp ).CONCLUSIONS:A quantitative LC/ITMS/MS method using HCD was developed for the first time to selectively monitor GS15 NO metabolites. The assay allowed evaluation of GS15 NO intestinal permeability and situated this drug candidate within the middle permeability class according to FDA guidelines. In addition, the present method has opened the perspective of a more fundamental work aiming at studying the fragmentation mechanism leading to the ion at m/z 156 in HCD tandem mass spectrometry in the presence of acetonitrile.

Abstract

International audience; A practical experiment devoted to study the properties of common foods (lemon, tea and garlic) containing antioxidant molecules is presently described. The experimental part was based on four main steps: (i) realization of calibration curves with standard antioxidants reacting with the 1,1-diphenyl-2-picryl-hydrazyl radical (DPPH•), (ii) quantitation of antioxidant molecules in food extracts, and (iii) determination of food extract antioxidant capacity vs. DPPH•, which (iv) was linked to the content of antioxidant molecules in each food extract. The educational approach was based on both conceptual thermodynamic processes and on practical knowledge on food chemistry and also diet related to healthcare. As a result, in addition to gain experience in simple spectroscopic methods, students went deeper in the understanding of the redox mechanisms devoted to oxidative stress basic learning.

Abstract

International audience; Designing convenient substrates is a pertinent parameter that can guide stem cell differentiation. Current research is directed toward differentiating mesenchymal stem cells (MSCs) into endothelial cells (ECs). It is generally accepted that MSCs cannot be easily differentiated into ECs without high concentrations of proangiogenic factors. To guide either bone marrow-derived mesenchymal stem cells (BM-MSCs) and Wharton's jelly-derived mesenchymal stem cells (WJ-MSCs) into ECs-like phenotype, poly(allylamine-hydrochloride)/poly(styrene-sulfonate) multilayers film (PAH/PSS) was used as culture coating and compared to type I collagen (as control coating). After 2 weeks of culture and in absence of angiogenic growth factors, PAH/PSS upregulated KDR, PECAM-1, and CDH5 genes, whereas combining PAH/PSS with endothelial growth media (EGM-2®) led to the production of respective proteins by WJ-MSCs. In contrast, not fully EC-like phenotype is obtained from the differentiation of BM- or WJ-MSCs cultured on type I collagen. Thus, using PAH/PSS coating in synergy with EGM-2® appears as an ideal condition promoting WJ-MSCs differentiation into ECs-like.
