Abstract

Apoptosis is a form of regulated cell death (RCD) that involves proteases of the caspase family. Pharmacological and genetic strategies that experimentally inhibit or delay apoptosis in mammalian systems have elucidated the key contribution of this process not only to (post-)embryonic development and adult tissue homeostasis, but also to the etiology of multiple human disorders. Consistent with this notion, while defects in the molecular machinery for apoptotic cell death impair organismal development and promote oncogenesis, the unwarranted activation of apoptosis promotes cell loss and tissue damage in the context of various neurological, cardiovascular, renal, hepatic, infectious, neoplastic and inflammatory conditions. Here, the Nomenclature Committee on Cell Death (NCCD) gathered to critically summarize an abundant pre-clinical literature mechanistically linking the core apoptotic apparatus to organismal homeostasis in the context of disease.

Abstract

Problem: Preterm birth (PTB) remains the leading cause of neonatal morbidity and mortality. Identifying women at high risk of spontaneous preterm labor (PTL) is challenging due to limited efficient diagnostic markers. Since human parturition involves inflammatory immune processes, we hypothesized that phenotyping of maternal peripheral lymphocytes might predict PTL. Therefore, we aimed to explore the relationship between maternal lymphocyte subpopulations and labor onset characterized by delivery within 7 days of admission in women hospitalized for PTL between 24 and 34 weeks of gestation. Methods of Study: Lymphocyte subpopulations were obtained from peripheral blood samples and characterized by flow cytometry: activated and regulatory T cells, natural killer and B cells, and TH1/TH2/TH17 lymphocytes. Data analysis was conducted retrospectively based on the delivery within 7 days of admission. Results: Among 167 women admitted for PTL, less than 10% delivered within 7 days post‐admission. HLA‐DR expression was significantly increased on CD4+CD8−, CD4−CD8+, and CD4+CD8+ lymphocytes in women who delivered within 7 days. Subset levels below 5% of CD4+CD8−HLA‐DR+ lymphocytes and 20% of CD4+CD8+HLA‐DR+ lymphocytes were associated with no probability of delivering within 7 days. Conclusion: Our study suggests that combining these two consecutive markers allowed us to identify 57% of women hospitalized for PTL with no probability of delivering within 7 days while retaining patients who delivered within 7 days. If prospectively validated, these markers may be able to identify patients at high risk of PTB and avoid a significant number of unnecessary admissions and healthcare costs. Abstract

Over the past decade, the Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for the definition and interpretation of cell death from morphological, biochemical, and functional perspectives. Since the field continues to expand and novel mechanisms that orchestrate multiple cell death pathways are unveiled, we propose an updated classification of cell death subroutines focusing on mechanistic and essential (as opposed to correlative and dispensable) aspects of the process. As we provide molecularly oriented definitions of terms including intrinsic apoptosis, extrinsic apoptosis, mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic cell death, NETotic cell death, lysosome-dependent cell death, autophagy-dependent cell death, immunogenic cell death, cellular senescence, and mitotic catastrophe, we discuss the utility of neologisms that refer to highly specialized instances of these processes. The mission of the NCCD is to provide a widely accepted nomenclature on cell death in support of the continued development of the field.

Abstract

La Administración Pública, por su propia naturaleza, requiere de medios personales para el cumplimiento de sus fines. La selección e incorporación de ese personal contribuye a la creación de una Administración eficiente, profesional e independiente. Sin embargo, mientras que la demanda de personal ha sido constante, los poderes públicos no han convocado los procesos selectivos necesarios para la cobertura de las plazas de naturaleza estructural. Esta situación se ha mantenido a lo largo del tiempo y ha obligado a recurrir a la contratación de personal temporal para poder garantizar la prestación de servicios. Esta anomalía ha contribuido a generar un problema endémico en la función pública española con unos niveles de temporalidad excesivos. La denuncia de esta situación ante los tribunales, tanto comunitarios como nacionales, ha obligado al legislador ha arbitrar mecanismos para la reducción de la temporalidad en el empleo público, dando lugar a la Ley 20/2021, de 28 de diciembre, de medidas urgentes para la reducción de la temporalidad en el empleo público. El presente trabajo pretende analizar la jurisprudencia en materia de abuso de la temporalidad y las medidas correctoras previstas en la Ley 20/2021, de 28 de diciembre para paliar esta situación., The Public Administration, by its nature, requires personal resources to achieve its objectives. The selection and incorporation of this personnel contribute to the creation of an efficient, professional, and independent Administration. However, while the demand for personnel has been constant, public authorities have not convened the necessary selection processes to fill structural positions. This situation has persisted over time and has forced recourse to the hiring of temporary staff to ensure the provision of services. This anomaly has contributed to generating an endemic problem in the Spanish public sector with excessive levels of temporality. The denunciation of this situation before both European Union and national courts has forced the legislator to devise mechanisms for reducing temporality in public employment, resulting in Law 20/2021, of December 28, on urgent measures for reducing temporality in public employment. Abstract

Apoptosis is a form of regulated cell death (RCD) that involves proteases of the caspase family. Pharmacological and genetic strategies that experimentally inhibit or delay apoptosis in mammalian systems have elucidated the key contribution of this process not only to (post-)embryonic development and adult tissue homeostasis, but also to the etiology of multiple human disorders. Consistent with this notion, while defects in the molecular machinery for apoptotic cell death impair organismal development and promote oncogenesis, the unwarranted activation of apoptosis promotes cell loss and tissue damage in the context of various neurological, cardiovascular, renal, hepatic, infectious, neoplastic and inflammatory conditions. Here, the Nomenclature Committee on Cell Death (NCCD) gathered to critically summarize an abundant pre-clinical literature mechanistically linking the core apoptotic apparatus to organismal homeostasis in the context of disease.

42. LAG‐3 : recent developments in combinational therapies in cancer.

Abstract

The study of anticancer immune responses and in particular the action of immune checkpoint inhibitors that overcome T cell inhibition has revolutionized metastatic patients' care. Unfortunately, many patients are resistant to these innovative immunotherapies. Over the last decade, several immune checkpoint inhibitors, currently available in the clinic, have been developed, such as anti‐PD‐1/PD‐L1 or anti‐CTLA‐4. More recently, other immune checkpoints have been characterized, among them lymphocyte activation gene 3 (LAG‐3). LAG‐3 has been the subject of numerous therapeutic studies and may be involved in cancer‐associated immune resistance phenomena. This review summarizes the latest knowledge on LAG‐3 as an immunotherapeutic target, particularly in combination with standard or innovative therapies. Indeed, many studies are looking at combining LAG‐3 inhibitors with chemotherapeutic, immunotherapeutic, radiotherapeutic treatments, or adoptive cell therapies to potentiate their antitumor effects and/or to overcome patients' resistance. We will particularly focus on the association therapies that are currently in phase III clinical trials and innovative combinations in preclinical phase. These new discoveries highlight the possibility of developing other types of therapeutic combinations currently unavailable in the clinic, which could broaden the therapeutic spectrum of personalized medicine.
