Holstein-Friesian steer beef production is renowned globally as a secondary product of the milk industry. Grass feeding is a common practice in raising Holstein steers because of its low cost. Furthermore, grass feeding is an alternative way to produce beef with a balanced n-6 to n-3 fatty acids (FAs) ratio. However, the performance and meat quality of Holstein-Friesian cattle is more likely to depend on a high-quality diet. The aim of this study was to observe whether feeding two mixed diets; a corn-based total mixed ration (TMR) with winter ryegrass (Lolium perenne) or flaxseed oil-supplemented pellets with reed canary grass haylage (n-3 mix) provided benefits on carcass weight, meat quality and FA composition compared with cattle fed with reed canary grass (Phalaris arundinacea) haylage alone. In all, 15 21-month-old Holstein-Friesian steers were randomly assigned to three group pens, were allowed free access to water and were fed different experimental diets for 150 days. Blood samples were taken a week before slaughter. Carcass weight and meat quality were evaluated after slaughter. Plasma lipid levels and aspartate aminotransferase (AST), γ-glutamyl transpeptidase (GGT), creatine kinase (CK) and alkaline phosphatase (ALP) activities were determined. Diet did not affect plasma triglyceride levels and GGT activity. Plasma cholesterol levels, including low-density and high-density lipoproteins, were higher in both mixed-diet groups than in the haylae group. The highest activities of plasma AST, CK and ALP were observed in the haylage group, followed by n-3 mix and TMR groups, respectively. Carcass weight was lower in the haylage group than in the other groups and no differences were found between the TMR and n-3 mix groups. Although the n-3 mix-fed and haylage-fed beef provided lower n-6 to n-3 FAs ratio than TMR-fed beef, the roasted beef obtained from the TMR group was more acceptable with better overall meat physicochemical properties and sensory scores. According to daily cost, carcass weight and n-6 to n-3 FAs ratio, the finishing diet containing flaxseed oil-supplemented pellets and reed canary grass haylage at the as-fed ratio of 40 : 60 could be beneficial for the production of n-3-enriched beef.

The research presents universal conditions which allow the complete splitting of a dangerous pulse (DP) in a meander microstrip line (MMSL) of any number of cascades. The verification of the conditions was performed on the example of an MMSL of two, three, four and five cascades. Attenuation (times) of a DP in the MMSL of three turns was 8.1, of four turns – 19.9 and of five turns – 33.16.

International Ocean Discovery Program (IODP) Expedition 372 combined two research topics, slow slip events (SSEs) on subduction faults (IODP Proposal 781A-Full) and actively deforming gas hydrate-bearing landslides (IODP Proposal 841-APL). Our study area on the Hikurangi margin, east of the coast of New Zealand, provided unique locations for addressing both research topics.SSEs at subduction zones are an enigmatic form of creeping fault behavior. They typically occur on subduction zones at depths beyond the capabilities of ocean floor drilling. However, at the northern Hikurangi subduction margin they are among the best-documented and shallowest on Earth. Here, SSEs may extend close to the trench, where clastic and pelagic sediments about 1.0-1.5 km thick overlie the subducting, seamount-studded Hikurangi Plateau. Geodetic data show that these SSEs recur about every 2 years and are associated with measurable seafloor displacement. The northern Hikurangi subduction margin thus provides an excellent setting to use IODP capabilities to discern the mechanisms behind slow slip fault behaviour.

Radioactive cesium (137Cs) has seriously become a human concern owing to exposure from a nuclear accident release at a nuclear plant. Many efforts have focused at the removal of radioactive cesium and remediation of a contaminated environment. To meet these demands, an effective sorbent based on magnetic composites functionalized with synthetic clay minerals was demonstrated. This sorbent shows a high removal efficiency of contaminated water containing suspended sorbents at a level of 0.5 mg ml−1 [137Cs of 84.68 Becquerel (Bq) gram (g)−1], decontaminated to 0.47 Bq g−1 (99.44 % removal efficiency) with just one treatment. The radioactive cesium is dramatically adsorbed into synthetic clay minerals. Subsequently, a rapid and easy sorbent separation from the radioactive cesium solution occurs after treatment using a magnetic field. Thus, a magnetic sodium-phlogopite sorbent can offer high potential for in situ remediation.

Background The human elbow maintains its stability mainly through its bony structure. Stability is enhanced by ligamentous structures. To allow the ligamento-muscular reflex, which protects against strain and stress, mechanoreceptors are embedded in the ligament. This report describes the existence and the distribution of the elbow medial collateral ligaments (MCLs) mechanoreceptors. Hypothesis The bony attachment site has the highest density of mechanoreceptors, and the anterior part has the highest density of mechanoreceptors. Materials and methods Eight MCLs of elbow from fresh frozen cadavers were used. The MCLs were harvested deep to the periosteum from the medial epicondyle to the ulna. The fan-shaped ligaments were divided into six regions of interest (ROI) and stained with modified gold chloride stain. Specimens were evaluated under a light microscope. Golgi, Ruffini, and Pacinian corpuscles were found in every specimen. The number and the distribution of each mechanoreceptor in each ROI were recorded. The density of each mechanoreceptor was calculated in regards to its volume. Results Golgi, Ruffini, and Pacinian corpuscles were seen in the ligament with small nerve fibers. Ruffini corpuscles had the highest median density of all three corpuscles. The median corpuscle density was higher in the anterior than in the posterior part and higher in the bony attachment than in the mid-substance site except for Golgi corpuscle. Conclusion The three typical types of mechanoreceptors were identified in human MCL with the anterior part and bony attachment as the dominant distribution site. The metal-insulator transition in correlated materials is usually coupled to a symmetry-lowering structural phase transition. This coupling not only complicates the understanding of the basic mechanism of this phenomenon but also limits the speed and endurance of prospective electronic devices. We demonstrate an isostructural, purely electronically driven metal-insulator transition in epitaxial heterostructures of an archetypal correlated material, vanadium dioxide. A combination of thin-film synthesis, structural and electrical characterizations, and theoretical modeling reveals that an interface interaction suppresses the electronic correlations without changing the crystal structure in this otherwise correlated insulator. This interaction stabilizes a nonequilibrium metallic phase and leads to an isostructural metal-insulator transition. This discovery will provide insights into phase transitions of correlated materials and may aid the design of device functionalities.

Physical properties, including basic index properties, undrained shear strength, and thermal conductivities were measured on conventional cores retrieved from the Ulleung Basin, during the second Ulleung Basin Gas Hydrate expedition (UBGH2). The UBGH2 logged 13 sites and cored 10 sites to locate potential sites for an offshore test production. A total of 211 conventional cores were recovered. Undrained shear strength, thermal conductivity, and index properties, including porosity and grain density, were measured onboard. Mineral composition analyses and grain size analyses were completed onshore after the expedition. The averaged porosity at each site ranges from 65% to 71%, the averaged grain density at each site ranges from 2.57 to 2.66 g/cm3, and the mean grain size mostly lies between 4 μm and 25 μm, with sparsely scattered coarse grained intervals. The relatively high porosity and low grain density are due to the large portion of diatomaceous sediments. The thermal conductivities average around 0.8 W/mK, and the low porosity and the abundance of clay mineral and OAPL-A may have caused the relatively low thermal conductivity in the Ulleung Basin. The geomechanical analyses revealed a few relevant findings important for sediment physical behaviors during gas hydrate production. The particle migration in coarse grained layers during production necessitates proper measure for sand productions while fine grained layers are mostly self-filtering. The vertical deformation estimated from the compression index suggested that the subsidence induced by the pore pressure change from the depressurization is much higher than those induced by the lost of gas hydrate particles from dissociation. Massive vertical deformation in fine-grained layers has been predicted to occur due to the pore pressure change from depressurization induced gas hydrate production. The drilling mud weight induced pressure window is mostly determined by the water depth in deep sea drilling, but caution should be taken in determination of fracture pressures in hydrate-bearing sediments, because gas hydrate saturations alter Poisson's ratio.

A series of electrochemical experiments were conducted to investigate the effect of dissolved oxygen, pH and Cl− on the corrosion rate of reinforcing steel of geological disposal facility saturated with groundwater. It was found that the corrosion rate was proportional to the concentration of Cl− and dissolved oxygen which are known as a corrosive agent and an electron acceptor, respectively. The pH level also strongly influenced the corrosion rate of the reinforcing steel. Under the pore water conditions of concrete structure of geological disposal facility, i.e. pH of 10–12 and dissolved oxygen of 1 mg L−1, the corrosion rate of reinforcing steel was determined to be in the range of ∼10−8 to ∼10−9 m/year. The corrosion rates were higher than those estimated from an empirical model based on the diffusion of dissolved oxygen.

Two metal-organic lanthanum precursors, dissolved in organic solvents, were evaluated for ALD applications on 300mm diameter Si substrates using O3 as an oxidant - one with La-C bonds (Precursor I) and the other with La-N bonds (Precursor II). Precursor I has CVD-like behavior of growth rate increase with temperature in the susceptor temperature range between 240°C and 350°C. On the other hand, Precursor II exhibits ALD behavior with a saturated film growth rate in the susceptor temperature range between 240°C and 340°C. The immobilization of ABEDTA was achieved by amide formation. The presence of the metal chelating functional group in the fully deprotonated form was confirmed by FT-IR. The chelation efficiency of beads was tested by determining metal ions in supernatant using GFAAS when pH gradients from 3 to 7. Sorption experiments for radionuclides such as 14C, 99Tc, 137Cs, 90Sr, 63Ni, and 241Am were conducted using two different groundwaters (GM-1 and SS-5) and solid materials (granodiorite and fracture-filling material) sampled from the Wolsong low- and intermediate-level radioactive waste (LILW) repository, Gyeongju, Korea. The distribution coefficients of the radionuclides, Kd’s, were obtained and their sorption properties were discussed for each radionuclide. For all sorbing radionuclides, the Kd values for the fracture-filling material were observed to be higher than those for granodiorite regardless of the groundwater. The Kd values were increased in the sequence 99Tc < 14C < 90Sr < 137Cs < 63Ni < 241Am regardless of sorbent types implying that the sorption of radionuclides onto geological media is affected by their chemical behavior in accordance with geochemical environments. Anionic radionuclides such as 14C and 99Tc showed very low Kd values both for the granodiorite and fracture-filling material. The mineralogical composition of the geological media and groundwater conditions was also observed to be important in the sorption of sorbing radionuclides, especially in the case of strongly sorbing radionuclides.

Two experiments were conducted to evaluate potato protein (PP, experiment 1) and refined PP (RPP, experiment 2) obtained from Gogu valley tubers as an antimicrobial agent in broiler diets. In both the experiments, 1-d-old male Ross 308 chicks were allotted to 5 treatments and performance, nutrient retention, and microbial populations in excreta and cecum were studied. Dietary treatments were as follows: basal diet (negative control, NC), basal diet with antibiotic (positive control, PC, 10 mg/kg of avilamycin), and low, medium, or high levels of PP (0.25, 0.50, and 0.75%, respectively, in experiment 1) or RPP (200, 400, and 600 mg/kg, respectively, in experiment 2). The overall gain and retention of DM (d 20 to 21) and CP (d 20 to 21 and d 41 to 42) were greater in birds fed PC and high PP diets than birds fed the NC diet. Population of total aerobic bacteria and coliforms was lowest in the cecum and excreta of birds fed the PC diet and highest in birds fed the NC diet. An increase in dietary PP linearly improved BW gain, feed intake, and feed conversion ratio during starter phase and overall BW gain. Also, there was linear improvement in retention of DM (d 20 to 21) and CP (d 20 to 21 and d 41 to 42) and reduced populations of total aerobic bacteria and coliforms in the cecum (d 42) and excreta (d 28 and 42) due to an increase in dietary PP. In the second experiment, the PC diet and diets with increasing levels of RPP had no effect on performance and nutrient retention. Birds fed the PC diet had the lowest microbial population in excreta and cecum, whereas the population of total aerobic bacteria and coliforms in excreta and cecum decreased (linear, P0.05) as the level of RPP was increased in the diet. These results suggest that both PP and RPP obtained from Gogu valley potato tubers have in vivo antimicrobial activity.

During Ocean Drilling Program Leg 138, eleven sites were drilled. The sediments recovered are mainly composed of carbonate-rich pelagic ooze including minor diatom and radiolarian ooze. Average carbonate contents of Sites 846 and 850 are about 59% and 75%, respectively. The porosity and velocity measured in laboratory were corrected to in situ condition using empirical equation (Hamilton, 1976) and delta velocity (log value-laboratory value). The corrected laboratory porosity matches well with logging data, but it is deviating in the intervals of low carbonate content. The correlation between log porosity and the corrected porosity shows a similar trend. Depth profiles between the corrected velocity and logging data do not agree in the intervals of low carbonate content, but the intervals characterized by high carbonate content are relatively matched. Purpose: To investigate the mechanism by which pinitol inhibits tumor necrosis factor-α (TNF-α)- induced expression of matrix metalloproteinase-9 (MMP-9) and invasion of prostate cancer LNCaP cells. Methods: Reverse transcription-polymerase chain reaction (RT-PCR) together with Western blot analysis was used to analyze the expression of MMP-9 and nuclear factor-κB (NF-κB) subunits, p65 and p50, in TNF-α- treated LNCaP cells, while 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, flow cytometry, and DNA fragmentation were used to evaluate cell viability and apoptosis. MMP-9 activity and invasion were measured by gelatin zymography and matrigel invasion assay, respectively. DNA-binding activity of NF-κB and AP-1 was determined by electrophoretic mobility shift assay and luciferase activity. Results: MMP-9 activity significantly increased in response to TNF-α; however, pinitol reduced TNF-α- induced MMP-9 activity without cytotoxicity. Matrigel invasion assay showed that pinitol reduced TNF-α-induced invasion of prostate cancer LNCaP cells. Further, it downregulated the expression of MMP-9 gene induced by TNF-α-treatment. Pinitol suppressed TNF-α-induced NF-κB activity by suppressing nuclear translocation of the NF-κB subunits, p65 and p50. Conclusion: The results indicate that pinitol is a potential anti-invasive agent and acts by suppressing TNF-α-induced cancer cell invasion and specifically inhibiting NF-κB as well as downstream target genes such as MMP-9.

Research experiments conducted and preliminary findings The Expedition ARA04C is a multidisciplinary research program in the Beaufort Sea, carried out in collaboration between the Korea Polar Research Institute (KOPRI), Geological Survey of Canada (GSC), Department of Fisheries and Ocean (DFO), Monterey Bay Aquarium Research Institute (MBARI), and the Alfred Wegener Institute (AWI). The Expedition ARA04C on the IBRV Araon took place from September 6 to September 24, 2013 (Figure 0.1). Multiple research experiments were undertaken to study geological processes related to degrading permafrost, fluid flow and degassing, and associated geohazards, paleo-oceanography of the Beaufort shelf and slope region, as well as physical and chemical oceanography measurement of the Arctic Ocean linked with continuous atmospheric studies. The expedition focused on two main research areas: offshore Barrow, Alaska, from September 7 to September 9, 2013, and the Canadian Beaufort Sea from September 10 to September 24, 2013. Multichannel seismic data, in conjunction with an ocean-bottom-seismometer (OBS) study were collected to support drilling proposals especially IODP pre-proposal #806 (Dallimore et al., 2012), and to verify distribution and internal structures of the offshore permafrost occurrences (Figure 0.2). The multi-channel seismic data were acquired on the outer continental shelf of the Canadian Beaufort Sea, totaling 14 lines with ~435 line-kilometers and ~4,500 shot gathers (Chapter 3). The combined multichannel seismic and OBS data will be processed post-expedition at KOPRI and the GSC, and will allow detailed velocity analyses to investigate the permafrost signature and help mapping zones of high-velocity sediments indicative of the presence of ice (Chapter 4). Individual shot gathers collected during the multichannel seismic program show clear refraction arrivals with velocities around 2000m/s in areas of expected permafrost occurrence, and shot gathers lacked such arrivals in zones where the permafrost was predicted to be absent. It is therefore expected that the OBS data, once processed, will also show clear refracted arrivals for velocity analyses. Continuous sub-bottom profiler (SBP) and multibeam data were collected along all ship tracks for detailed subsurface imaging of sediment structures and permafrost, as well as for core-site location verification (Chapter 5 and 6). During Expedition ARA04C, more than 3000 line-kilometers of SBP data were collected, co-located with multibeam and backscatter data. These data are an essential part of the study of sub-seafloor permafrost distribution and provide insights into sediment dynamics at critical boundaries, such as the shelf edge. Along the shelf edge, the occurrence of pingo-like features (PLFs) result in a rugged landscape with thousands of PLFs piercing through the otherwise laminated sediments. More than 30 crossings of this critical shelf-edge boundary were made during this expedition, which complement data acquired in 2012 with the Huntec system and 3.5 kHz data provided by ArcticNet as part of the regional multibeam map of the study area. High resolution data provided critical new insights in deep-water fluid expulsion zones. Key new data were acquired over the area of the "Gary Knolls", where PLF structures occur at the shelf edge in water depth of only 50 to 60 m. All SBP data from this expedition will be post-processed and analyzed for the presence of sub-seafloor permafrost, occurrence of the PLF structures and indications for fluid and gas migration. Multibeam and backscatter data were collected along all ship tracks, adding to the database of existing information gathered through previous expeditions to the study region. Heat flow measurements were undertaken at eight stations (Figure 0.3) to study the thermal structure of fluid expulsion features, as well as degrading permafrost along a slope-shelf transect in the eastern Mackenzie Trough (Chapter 7). The data provide critical constraints on the distribution of sub-seafloor permafrost as well as the gas hydrate stability zone around fluid expulsion features. A very important finding is the observation made at the mud volcano in 420 m water depth, where seafloor temperatures are the highest in all observed stations, indicating active mud volcanism. Geological sampling using gravity coring and multi-coring tools was performed at strategic sites to support two research objectives. The first objective was to provide key data towards ongoing international research linked to IODP pre-proposals #753 (O'Regan et al., 2010) and #806 (Dallimore et al., 2012). The second objective was to collect core to define key seismo-stratigraphic horizons critical to the understanding of geohazards in the region (Chapter 8). In total, 21 gravity cores and 12 multi-cores were taken (Figure 0.4, Table 8.3). All cores were scanned with a multi-sensor core-logger to measure physical properties (Chapter 9). Most sediment analyses on the cores will be performed post-expedition at KOPRI, GSC, and laboratories of other University-based collaborators in Canada and Germany. Onboard, sub-samples were taken from all shallow multi-cores and selected gravity cores. On selected cores from the Canadian Beaufort study region pore-waters were extracted using rhizones. These samples will be analyzed postexpedition at MBARI. Water sampling and Conductivity-Temperature-Depth (CTD) profiling was undertaken at most core sites to study physical and chemical properties of the seawater (Figure 0.5). These station-measurements were complemented by continuous waterproperty and atmospheric measurements when the Araon was underway. Most samples taken will be analyzed post-expedition at KOPRI for DIC/TA, nutrients, DOC, and POC. The pH of seawater, underway data of pCO2, CH4, and N2O, as well as a variety of subsequent calculations is required for accurate estimates in the above listed parameters. Methane was also measured with a methane sensor attached to the CTD tool and at the mud volcano in 420 m water depth, methane concentrations of more than 100-times ocean background were seen. The methane plume was also acoustically imaged with the echo sounder systems on board the IBRV Araon. Further details on the water sampling and atmospheric measurements are given in Chapter 10 and 11.

A laboratory-scale chemical vapor deposition (CVD) reactor was used to perform “continuous” Hf doping experiments while the surface of a single-crystal Ni alloy was being aluminized to form an aluminide (β-NiAl) coating matrix for 45 minutes at 1150 °C. The continuous doping procedure, in which HfCl4 and AlCl3 were simultaneously introduced with H2, required a high HfCl4/AlCl3 ratio (>∼0.6) to cause the precipitation of Hf-rich particles (∼0.1 µm) at grain boundaries of the coating layer, with the overall Hf concentration of ∼0.05 to 0.25 wt pct measured in the coating layer by glow-discharge mass spectroscopy (GDMS). Below this ratio, Hf did not incorporate as a dopant into the growing coating layer from the gas phase, as the coating matrix appeared to be “saturated” with other refractory elements partitioned from the alloy substrate. In comparison, the Hf concentration in the aluminide coating layer formed on pure Ni was in the range of ∼0.1 wt pct, which was close to the solubility of Hf estimated for bulk NiAl. Interestingly, the segregation of Hf and the formation of a thin γ′-Ni3Al layer (∼0.5 µm) at the coating surface were consistently observed for both the alloy and pure-Ni substrates. The formation of the thin γ′-Ni3Al layer was attributed to an increase in the elastic strain of the β-NiAl phase, associated with the segregation of Hf as well as other refractory alloying elements at the coating surface. This phenomenon also implied that the coating layer was actually growing at the interface between the γ′-Ni3Al layer and the β-NiAl coating matrix, not at the gas/coating interface, during the early stage of the coating growth.

Highly c-axis oriented Ga-doped ZnO films (GZO) have been grown on sapphire (0001) substrates by pulsed laser deposition (PLD) method. Photoluminescence (PL) spectra indicate that Ga atoms have a large effect on the luminescent properties of ZnO films. PL spectra of GZO films show near band edge (NBE) emissions and broad orange deep-level emissions. The NBE emission shifts to higher energy region and the intensity decreases with the increase of Ga concentration. The blue shift of NBE emission results from Burstein-Moss effect. The quenching of NBE emission is ascribed to the noradiative recombination. The orange emission is related to the oxygen vacancies.

The (100) surface of a single-crystal Ni alloy was aluminized as a function of time to study the development of the resulting coating microstructure. A chemical vapor deposition (CVD) reactor, which was specially configured for short-term aluminizing experiments, was used to prepare coating specimens at 1150 °C. After 5 minutes, γ′-Ni3Al particles ∼100 nm in size randomly nucleated on the alloy surface. Within 20 minutes, a coating layer consisting of preferentially oriented, columnar β-NiAl grains was formed with the segregation of refractory elements (i.e., Ta and W) from the alloy to the coating grain boundaries. The lateral growth of the columnar grains was observed to be relatively rapid for up to 45 minutes, but slowed considerably between 45 and 180 minutes. While the columnar nature of the coating did not change significantly after 20 minutes, the surface features continually evolved, with the appearance of a small amount of the γ′ phase, which coincided with the segregation of the refractory elements to the coating surface.

We present the discharge characteristics of a rare gas halide mixture in AC- Plasma Display Panels. Studies for excimer gas of plasma display panels are performed using XeF. This excimer gas is investigated as function of its pressure and different discharge condition. In this paper, the real panel using XeF excimer gas is researched for good discharge characteristics and a full-color panel is demonstrated.

Metastasis to the appendix is extremely rare. Moreover, metastasis from a hepatocellular carcinoma (HCC) has not been reported in the English language literature. We report a case of metastasis to the appendix from a HCC in a 50-year-old man who presented with pain in the right lower quadrant of the abdomen, representing acute appendicitis. On CT, a hypervascular mass occupying the lumen of the appendix with distension of the appendiceal tip and surrounding peritoneal seeding of the HCC were observed. This is the first report of CT findings of metastasis to the appendix from a HCC.

Background: This phase 3 study evaluated the efficacy of new adjuvant chemotherapy (MFP), which intensified the mitomycin-C (MMC) plus short-term doxifluridine (Mf) for gastric cancer. Patients and methods: A total of 855 patients (424 in Mf, 431 in MFP) with pathological stage II–IV (M0) gastric cancer after D2 gastrectomy were randomly assigned to receive either Mf (MMC 20 mg m−2, followed by oral doxifluridine 460–600 mg m−2 per day for 3 months) or MFP (MMC 20 mg m−2, followed by oral doxifluridine 460–600 mg m−2 per day for 12 months with 6 monthly infusions of 60 mg m−2 of cisplatin) chemotherapy. Results: With a median follow-up of 6.6 years, there was no difference between the two groups in recurrence-free survival (RFS) (5-year RFS 61.1% in Mf and 57.9% in MFP; hazard ratio 1.10 (95% CI 0.89–1.35); P=0.39) and overall survival (OS) (5-year OS 66.5% in Mf and 65.0% in MFP; hazard ratio 1.11 (95% CI 0.89–1.39); P=0.33). Conclusion: Intensification of Mf adjuvant chemotherapy by prolonging the duration of oral fluoropyrimidine and adding cisplatin was safe but not effective to improve the survivals in curatively resected gastric cancer patients.

The pig conceptus produces a non-invasive placenta which is probably dependent on uterine secretory products during both the pre- and post-attachment periods. Since some of these factors are also produced during the oestrous cycle, uterine luminal flushings (ULF) from adult non-pregnant pigs were analysed to identify potentially important maternally-secreted growth factors or cytokines. ULF contained several proteins that were detected on Western blots by an antibody raised against the N-terminal 14 amino acids of rat pleiotrophin (PTN). One of the proteins co-migrated on SDS-PAGE gels and co-eluted from heparin-affinity columns with 18 kDa recombinant human PTN, suggesting its identity as the intact native porcine PTN (pPTN) molecule. Additional immunoreactive forms of pPTN were identified that were of lower molecular mass (14–16 kDa), had lower heparin-affinities, were more hydrophobic, and were apparently C-terminally truncated. Native pPTN was isolated from ULF using cation-exchange chromatography, heparin-affinity fast protein liquid chromatography and C4 reverse-phase high performance liquid chromatography. Structural analysis of the purified protein resulted in definitive identification of 15 out of 17 N-terminal amino acids; these were 100% conserved with the corresponding residues of human, bovine and rat PTN. These results demonstrate various biochemical properties of pPTN and suggest that, in addition to the apparent involvement of PTN in differentiation during early neonatal life, it may be delivered in uterine secretory fluids to, as yet, undefined target cells in the reproductive tract of the adult female. Purpose: Hizikia fusiforme is renowned for the possession of anti-inflammatory and anti-oxidant properties. In this study, the role of the ethyl alcohol extract of H. fusiforme (EAHF) in the induction of apoptosis in human leukemia U937 cells was investigated. Methods: Protein expression was investigated by Western blot analysis. Cell viability and apoptosis were analyzed by an MTT assay and flow cytometric analysis. Caspase activity was analyzed using a caspase-specific kit. Results: EAHF suppressed the proliferation of U937 cells in a dose-dependent manner. This effect was closely related to the induction of apoptosis via the downregulation of IAP family members such as IAP-1, IAP-2 and XIAP, as well as Bcl-2 proteins. The results also showed that caspases play an essential role in EAHF-induced apoptosis by generating of reactive oxygen species (ROS). In addition, ROS scavenging by N-acetyl-L-cysteine (NAC) and glutathione (GSH) decreased EAHF-induced apoptosis via the suppression of caspase activity. Although EAHF induced the phosphorylation of mitogenactivated protein kinases (MAPKs), treatment with MAPK inhibitors did not affect EAHF-induced apoptosis. Purpose: Hydroclathrus clathratus is a brown marine seaweed known to possess anti-cancer, anti-herpetic, and anti-coagulant activities. The present study is aimed at investigating some anti-inflammatory effects of H. clathratus . Methods: We investigated the anti-inflammatory effects of the methanol extract of H. clathratus (MEHC) by expression of mRNA and protein using RT-PCR and Western blot analysis in lipopolysaccharide (LPS)-stimulated BV2 microglial cells. The level of nitric oxide (NO) production was analyzed using Griess reaction. The release of prostaglandin E 2 (PGE 2 ) and tumor necrosis factor-α (TNF-α) were determined using sandwich ELISA. NF-κB activation was detected using EMSA methods. Results: The results obtained indicate that the extract (MEHC) inhibited LPS-induced NO, PGE2, and TNF-α production without any significant cytotoxicity (p < 0.05). MEHC also inhibited production of inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2) and TNF-α mRNA in LPS-stimulated BV2 microglial cells. In addition, MEHC significantly reduced (p < 0.05) nuclear translocation of the nuclear factor-κB (NF-κB) subunits, p50 and p65, and its DNA-binding activity in LPS-stimulated BV2 microglial cells. Conclusion: These results suggest that MEHC suppresses the induction of TNF-α, as well as iNOS and COX-2 expression, by blocking LPS-induced NF-κB activation. Summary Although winter dysentery (WD), which is caused by the bovine coronavirus (BCoV) is characterized by the sudden onset of diarrhea in many adult cattle in a herd, the pathogenesis of the WD-BCoV is not completely understood. In this study, colostrum-deprived calves were experimentally infected with a Korean WD-BCoV strain and examined for viremia, enteric and nasal virus shedding as well as for viral antigen expression and virus-associated lesions in the small and large intestines and the upper and lower respiratory tract from 1 to 8 days after an oral infection. The WD-BCoV-inoculated calves showed gradual villous atrophy in the small intestine and a gradual increase in the crypt depth of the large intestine. The WD-BCoV-infected animals showed epithelial damage in nasal turbinates, trachea and lungs, and interstitial pneumonia. The WD-BCoV antigen was detected in the epithelium of the small and large intestines, nasal turbinates, trachea and lungs. WD-BCoV RNA was detected in the serum from post-inoculation day 3. These results show that the WD-BCoV has dual tropism and induces pathological changes in both the digestive and respiratory tracts of calves. To our knowledge, this is the first detailed report of dual enteric and respiratory tropisms of WD-BCoV in calves. Comprehensive studies of the dual tissue pathogenesis of the BCoV might contribute to an increased understanding of similar pneumoenteric CoV infections in humans.

The current use of humanized monoclonal antibody trastuzumab for the treatment of patients with metastatic breast cancer has made evaluation of HER-2/neu status an important clinical issue. Chromogenic in situ hybridization (CISH), in which the DNA probe is detected with an immunohistochemistry (IHC)-like peroxidase reaction, has been recently developed for the assessment of HER-2/neu status in formalin-fixed breast cancer specimens. We have applied the technique of dual-colour CISH using HER-2/neu and chromosome 17 centromere probes in 27 cytological smears, and these cytological samples were obtained from scrapings of fresh breast tumours. We also investigated HER-2/neu amplification and protein overexpression in the corresponding surgical tissues by CISH and IHC using the monoclonal antibody CB11. Of the 27 cytological cases, HER-2/neu gene amplification was observed in nine cases that were positive cases (2+ and 3+) for IHC. Among the 13 IHC positive cases (2+ and 3+), four of them showed no gene amplification. Identical results for the CISH technique were obtained in the matched surgical samples. The scrape samples from fresh breast tumour offer a monolayer cell population that is especially suitable for CISH. This study has shown that the cytological smear might be a good alternative for the CISH test.

Recognizing the importance of comparisons of radionuclide calibrators in nuclear medicine and the need to provide access to activity standards that are traceable to the international measurement system, the Korea FoodDrug Administration (KFDA) as a national secondary standard dosimetry laboratory (SSDL), started a comparison program in 2002. The first two comparisons were conducted with (131)I (71 participants) and (99m)Tc (72 participants). The results indicated that only 61% of reported activities for (131)I and 65% for (99m)Tc were within +/-5% of the correct value. These numbers increased to 84% for (131)I and 83% for (99m)Tc when a +/-10% limit was applied. Follow-up action on those calibrators with results outside the +/-10% limits only produced a small improvement. There was also a marked difference in performance between the various calibrator types used. The results have shown that such comparisons are necessary to improve the quality of the measurements and to identify those radionuclide calibrators that are not functioning correctly.

Annexin I (also called lipocortin 1) is a 37-kDa member of the annexin family of proteins. It has been proposed to be involved in the regulation of cell growth and differentiation, apoptosis, and inflammation. Previously, we have reported that annexin I displays a chaperone-like function (Kim, G.Y., Lee, H.B., Lee, S.O., Rhee, H.J.Na, D.S. (1997) Biochem. Mol. Biol. Int. 43, 521-528). To determine the possibility that annexin I is a stress protein, we examined whether expression of annexin I and annexin I mRNA increases in response to stresses in A549 and HeLa cells. Treatments of cells with heat, hydrogen peroxide or sodium arsenite resulted in (a) an increase in annexin I and annexin I mRNA and (b) translocation of annexin I from the cytoplasm to the nucleus and perinuclear region. The annexin I gene promoter region, cloned upstream of a reporter gene, was inducible in response to heat, hydrogen peroxide, and sodium arsenite. These results indicate that annexin I serves as a stress protein and annexins may constitute a new class of stress proteins.

Uterine growth factors are potential effector molecules in embryo growth signaling pathways. Pig uterine luminal flushings contained a heparin-binding growth factor (HBGF) that required 0.8 M NaCl for elution from heparin columns and was termed HBGF-0.8. This factor, which was heat- and acid-labile and of Mr 10,000 as assessed by gel filtration, stimulated DNA synthesis in fibroblasts and smooth muscle cells but not endothelial cells. Two forms of HBGF-0.8, termed HBGF-0.8-P1 and HBGF-0.8-P2, exhibited differential heparin-binding properties. SDS-polyacrylamide gel electrophoresis showed that each form of HBGF-0.8 migrated with an apparent Mr of 10, 000 under reducing conditions. Amino acid sequencing revealed the N-terminal sequence EENIKKGKKXIRTPKI for HBGF-0.8-P1 and ENIKKGKKXIRT for HBGF-0.8-P2. These sequences corresponded, respectively, to residues 247-262 and 248-259 of the 349-residue predicted primary translation product of porcine connective tissue growth factor (pCTGF). 10-kDa CTGF-mediated fibroblast DNA synthesis was modulated by exogenous heparin, and CTGF-immunoreactive proteins of 10, 16, and 20 kDa were present in unfractionated uterine luminal flushings. These data reveal the identity of a novel growth factor in uterine fluids as a highly truncated form of CTGF and show that the N-terminal two-thirds of the CTGF primary translation product is not required for mitogenic activity or heparin binding.

Specular laser reflectance (He-Ne laser) has been used to monitor in-situ and in real time the growth rate and the index of refraction of oxide films during chemical vapor deposition. This technique has been implemented on an inverted vertical stagnation-flow reactor equipped with a solid-source delivery system. Yttria deposited on silicon has been chosen as a starting material to characterize the reactor and the precursor delivery system capabilities. The experimental reflectance curves have been fitted to a simple three-layer (gas/film/substrate) model allowing the determination of the growth rate and of the refractive index. The growth rate has been studied as a function of various key processing parameters: the source feeding rate, the powder packing density, the oxygen partial pressure and the total pressure. The change in reflectivity has also been recorded during pulsed-delivery growth. Nanometer-scale resolution is obtained which demonstrates that this method can be extended to the study of multilayer oxide structures.

Accumulating evidence suggests that uterine luminal fluids contain a variety of polypeptide growth factors and cytokines that, it is speculated, have roles in the development, growth and differentiation of the uterus and, during pregnancy, in the growth and survival of the embryo. Although epidermal growth factor (EGF) has previously been identified by radioimmunoassay and immunohistochemistry in the pig uterus, there have been no detailed studies of the secreted EGF protein. EGF was therefore purified from uterine flushings and uterine fluids of nonpregnant pigs of mixed breed using a variety of ion-exchange chromatography steps. Uterine flushings and fluids contained an anionic factor(s) that at 4 degrees C competed with 125I-labelled mouse EGF for binding to EGF receptors on an endometrial carcinoma cell line and stimulated DNA synthesis in Balb/c mouse 3T3 fibroblasts. As analysed by gel filtration, uterine fluids contained a 3-6 kDa factor that stimulated 3T3 cell DNA synthesis and was a competitor of cellular 125I-labelled EGF binding. Gel filtration further revealed that uterine flushings and fluids contained, respectively, 45 kDa and 40-70 kDa moieties that were mitogenic and that bound to the EGF receptor. SDS-PAGE and western blotting using an antiserum specific for pig EGF revealed immunoreactive forms of EGF of approximately 25 kDa in partially purified uterine flushings. It is concluded that uterine secretory EGF occurs, at least in part, as high molecular mass proteins. The ability of these high molecular mass EGFs to bind to and activate the EGF receptor suggests that they may be authentic ligands for the EGF receptor in utero.

This paper presents the results of transmission electron microscopy (TEM) studies on the distribution of polystyrene/polyisoprene/polystyrene (SIS) triblock copolymer compatibilizer in polystyrene-polyethylene homopolymer blends. The principal independent variable studied is the major-phase rheological properties (PSmajor/PEminor and PEmajor/PSmimor blends). The polyisoprene block was preferentially stained by OSO4 and could be clearly identified in cryo-ultramicrotomed TEM sections. The microstructural data show that when the Theologically strong PS phase is the major constituent, the compatibilizer follows the classical model localizing itself at the PS/PE interface. In the PEmajor/PSminor blends, however, the SIS compatibilizer is not localized at the PE/PS interface. Instead microdispersions of PS in SIS form which in turn are dispersed in the PE matrix. While the various microstructures are affected by the mixing protocol and polymer rheology, quiescent annealing after mixing indicates that the compatibilizer distribution is in part driven by thermodynamic effects.
