ABSTRACT: The Weibull proportional hazards model was used to identify the nongenetic effects that affect length of productive life (LPL) of the Italian Mediterranean buffalo. Data were provided by the Italian National Association of Buffalo Breeders and included records of reproductive, productive, and linear type traits from 59,943 buffalo with first calving from 2002 to 2019. Data were divided into 4 geographical regions to determine whether the relationship between the investigated effects and LPL varied by region. Length of productive life was defined as the number of days from the first calving to culling for those buffaloes that were culled (uncensored) or to the date of the last test-day for those that are still alive (censored). The Weibull model included time-dependent effects of herd-year-season of calving, parity and stage of lactation, production as within-herd deviations, as well as time-independent effects of age at first calving, year of birth, type of reproduction (natural mating vs. AI), classifier, and ten linear type traits. The average duration of productive life was 1,604 days. Both year of birth and production level had a significant effect on culling risk. Moreover, culling risk decreased linearly across parities. The reference point for age at first calving relative to other classes was set at 35 months. A significant relationship between linear type traits and relative culling risk was also observed, being highest for buffaloes with the lowest linear scores. The results show that the Weibull model provides consistent and robust risk estimates. Therefore, this model would be recommended for future implementation of the first genetic evaluation of LPL in the Italian Mediterranean buffalo.

ABSTRACT: In the buffalo dairy sector, a huge effort is still needed to improve mastitis prevention, detection, and management. Electrical conductivity (EC) and total somatic cell count (SCC) are well-known indirect indicators of mastitis. Differential somatic cell count (DSCC), which represents the proportion of neutrophils and lymphocytes on the total SCC, is instead a novel phenotype collected in the dairy cattle sector in the last lustrum. As little is known about this novel trait in dairy buffalo, in the present study we explored the nongenetic factors affecting DSCC, as well as EC and total somatic cell score (SCS), in the Italian Mediterranean buffalo. The data set used for the analysis included 14,571 test-day (TD) records of 1,501 animals from 6 herds, and climatic information of the sampling locations. The original data were filtered to exclude animals with less than 3 TD per lactation and, for the investigated traits, outliers beyond 4 standard deviations. In the statistical model we included the fixed effects of herd (6 classes), days in milk (DIM; 10 classes of 30 d, with the last being an open class until 360 d), parity (6 classes, from 1 to 6+), year-season of calving (11 classes, from summer 2019 to winter 2021/2022), year-season of sampling (9 classes, from spring 2020 to spring 2022), production level (4 classes based on quartiles of average milk yield by herd), and temperature-humidity index (THI; 4 classes based on quartiles, calculated using the average temperature and relative humidity of the 5 d before sampling). Average EC, SCS, and DSCC vary across herds. Considering DIM, greater EC values were observed at the beginning and the end of lactation; SCS was slightly lower, but DSCC was greater around the lactation peak. Increased EC, SCS, and DSCC levels with increasing parity were reported. Year-season calving and year-season sampling only slightly affected the variation of the investigated traits. Milk of high-producing buffaloes was characterized by lower EC and SCS mean values, nevertheless it had slightly greater DSCC percentages. Buffaloes grouped in the highest THI classes (classes 3 and 4) showed, on average, greater EC, SCS, and DSCC in comparison to the lower classes, especially to class 2. Results of the present study represent a preliminary as well as necessary step for the possible future inclusion of EC, SCS, or DSCC in breeding programs aimed to improve mastitis resistance in dairy buffaloes.

ABSTRACT: Until now, the genetic evaluation of the Italian Mediterranean Buffalo has been mainly focused on production traits. However, female fertility affects the efficiency of the dairy industry as it is essential to maintain the profitability of dairy farms. Indeed, the estimation of its genetic component is crucial for its improvement. In this study, 3 measures of buffalo's fertility were analyzed: the age at first calving (AFC), the interval between first and second calving (CIV1), and the interval between second and successive calvings (CIV2_12). Milk yield at 270 d (MY270) was used as a correlated trait. First, genetic parameters were estimated using 7,915 buffalo cows with first calving from 1991 to 2018, then breeding values were calculated from 236,087 buffalo cows. Genetic parameters were estimated by Bayesian inference fitting a multiple-trait animal model using the GIBBS1F90 program, and BLUPF90 was used for estimation of breeding value. The heritability and repeatability estimates of fertility traits were low. The genetic correlations among fertility traits ranged from 0.10 (AFC-CIV1) to 0.92 (CIV1-CIV2_12). Genetic correlation between MY270 and fertility traits was unfavorable, ranging from 0.23 to 0.48. The results from this study can be used as a basis for the future genetic improvement of fertility traits in the Italian Mediterranean Buffaloes.

ABSTRACT: Mastitis has detrimental effects on the world's dairy industry, reducing animal health, milk production and quality, as well as income for farmers. In addition, consumers' growing interest in food safety and rational usage of antibiotics highlights the need to develop novel strategies to improve mastitis detection, prevention, and management. In the present study we applied machine learning (ML) analyses to predict presence or absence of subclinical mastitis in Italian Mediterranean buffaloes, exploiting information collected the previous month during routine milk recording procedures, as well as climatic data. The data set included 3,891 records of 1,038 buffaloes from 6 herds located in Basilicata Region (South Italy). Prediction models were developed using 4 different ML algorithms (Generalized Linear Model, Support Vector Machines, Random Forest, and Neural Network) and 2 data set splitting approaches for the creation of the training and test sets (by record or by animal ID number, always with 80% of the data used for model training and the remaining 20% for model testing). Support Vector Machine was the best method to predict high or low somatic cell count at the subsequent test-day record in the validation set, and therefore it was used to estimate the contribution of each feature to the best model. Independently from the data set splitting approach, the most important features were somatic cell score, differential somatic cell count, electrical conductivity, and milk production. Among climatic data, the most informative were temperature and relative humidity. When the data were split by animal ID, an improvement in models' predictive performance on the test set was observed, suggesting this as the most appropriate data splitting approach in data sets with repeated measures to avoid data leakage. According to different metrics, Neural Network was the best method for making predictions on the test set. Our findings confirmed the promising role of ML methods to improve prevention and surveillance of subclinical mastitis, exploiting the large amount of data currently available to identify animals that would possibly have high somatic cell count the subsequent month.

Abstract The use of green feed for livestock breeding is an important strategy to encounter both the increasing demand for animal derived products and the perceptions of the consumers regarding animal welfare and sustainability. The aim of this study was to compare different feeding strategies in lactating water buffaloes by using a metabolomic approach. The study was carried out on 32 milking buffaloes that were randomly divided into two groups for a total period of 90 days (3 sampling times). DD Group (dry diet) received a standard total mixed ratio (TMR) characterized by dry forages and concentrates; ZG Group (zero grazing) fed an isoenergetic and isoproteic diet obtained using 30% of sorghum as green forage. Samples of milk and rumen fluid were analyzed by liquid chromatography—mass spectrometry (LC–MS) techniques. Data analyses revealed the presence of several differentially accumulated metabolites and among these, ten compounds were putatively identified in milk samples (i.e. l-carnitine, acetylcarnitine, propionylcarnitine, butyrylcarnitine, 2-methylbutyroylcarnitine, 2-hexenoylcarnitine, hexanoylcarnitine, glycerophosphocholine, δ-valerobetaine and γ-butyrobetaine) and four in rumen fluid (3-(2-hydroxyphenyl) propanoate, Indole-3-acrylic acid, oleamide (cis-9,10-octadecenoamide) and 20-carboxy-leukotriene B4). The modulation of these molecules in buffalo milk is significantly related to the green/dry based feeding and some the natural compound detected could be considered as health-promoting nutrients.

Studies on goat milk have mainly focused on cosmopolitan breeds and very limited information is available on local breeds, which is important for biodiversity preservation and local cheese production. The aim of this study was to evaluate the breed effect on milk yield, composition and somatic cell score (SCS) of five local Italian goat breeds (Garganica, Girgentana, Jonica, Maltese and Mediterranean Red) compared with a cosmopolitan specialized dairy breed (Saanen). A total of 60 goats (10 per breed) from an experimental farm were enrolled in the study. Milk yield, composition and SCS were recorded and analyzed every 2 weeks during the entire lactation. Data were analyzed using a mixed model with repeated measures. Saanen yielded between 0.27 and 0.62 kg day−1 more milk than the local breeds. Among local breeds, Maltese and Jonica were the most productive, with an average of 1.28 and 1.25 kg day−1, respectively, while Mediterranean Red, Garganica and Girgentana produced ≤1 kg day−1. Saanen had the highest SCS (6.81) and the lowest fat content (3.26 %). In relation to protein, Garganica showed the greatest content (3.71 %), and Saanen had a similar content to other local breeds (3.42 %) except for Maltese, which was lower (3.11 %). Saanen and Garganica had the lowest lactose percentage (4.28 % and 4.26 %, respectively). All breeds followed a similar pattern across lactation: SCS and fat and protein content peaked at the end of the lactation, whereas lactose percentage was highest at the beginning of the lactation. Differences between Saanen and the local breeds for milk yield, composition and SCS were consistent across lactation. In conclusion, local breeds produced less milk but with lower SCS and greater fat and lactose content than the Saanen cosmopolitan breed, suggesting a better milk quality.

The aim of this trial was to evaluate ghrelin response to milk administration in 20 days old buffalo calves. The trial was carried out on 5 female buffalo calves with a mean age of 21.2±2.8 days. Five blood samples were collected from each animal into EDTA tubes, starting at 07.00 until 15.00, at 2-h intervals. At 09.00, after the second blood sample, replaced milk was administered to the calves. Blood samples were immediately placed at 4°C until processing, which was performed on the same day. We used real-time reverse transcription PCR system to detect the expression of ghrelin mRNA levels in blood of buffalo calves. The aim of this study was to verify the ratio between real cheese yield at 28 hours and proteins, after values correction according to the regression analysis. Milk characteristics of subjects characterized by a 28CY/protein ratio lower (Group A; n=212) or higher (Group B; n=108) than 56 were analysed by ANOVA in order to evaluate differences between the following milk characteristics: the lactodinamographic parameters (Formagraph, Foss, DN); the complete physic-chemical composition; fat (F), protein (P), casein, lactose, ash, urea, pH and SH; the somatic cells content (SCC) using (Milkoskan); the total aerobic mesophilic flora (TAMF) by the dilution method. Chemical composition of the curd was also determined (protein, fat and ash; ASPA, 1995). The milk protein content was adjusted for the non proteic N content determined in milk as urea (corrected P); the theoretic cheese yield (ThCY) was obtained by the following formula: cheese yield =milk x[-0.88 + 3.50 x P(%)+1.23 x F(%)] x 100 -1 (Altiero et al., 1989) and the ratio between ThCY/FCY and ThCY/28CY were calculated. Other calculated variables were: FCY/Corrected P, 28CY/Corrected P, CDM/Corrected P. Regression analysis was carried out between real cheese yield at 28 hours/proteins ratio and 28CY vs. all the parameters resulted different between the two groups of buffaloes. Real cheese yield at 28 hours/proteins ratio and 28CY were corrected and ANOVA was repeated on corrected data, in order to verify the modifications of the values. These correction reduced but did not eliminate the differences.

The percentages of dry matter (DM), protein, casein, fat, lactose, solidsnot- fat and ash of milk and the characteristics of the curd (31 components and/or characteristics) were evaluated on 326 milk samples collected from 60 buffaloes every 50 days. The animals were half sib (same father or same mother ascertained by DNA test) and were bred in two farms. Relationships between the values of components and/or characteristics were also calculated. Significant relationships were found among DM percentage of milk and its components, cheese yield, curd characteristics, ratio between the studied cheese yields and protein percentage. Protein, casein and fat showed positive relationships, whereas lactose presented negative relationships. The association among proteins and real cheese yield or the dry matter of the curd that derived from 1 litre of milk (CDM), was lower than that with calculated cheese yield. This difference strongly decreased if proteins percentage, corrected for urea value, was utilized in cheese yield calculation. No relationship was found between proteins and urea in milk, but a direct relation was present between proteins and proteins adjusted for urea. This relation affects a correct estimation of cheese yield, because the percentage of true proteins in milk, corrected per urea was 4.52 vs. 4.64.

The aim of this study was to screen for Brucella spp. buffalo embryos produced in- vitro, by using cumulus oocytes complexes (COCs) recovered from ovaries of slaughtered buffaloes naturally infected with Brucella spp. Ovaries were collected from 5 female pluriparous buffaloes slaughtered in a local abattoir. EDTA-blood samples and nasal swabs collected from each animal were used for Brucella spp. DNA detection by real-time PCR. Buffalo ovaries (n = 10) were transported to the laboratory and maintained strictly separated throughout laboratory processing. Recovered COCs were matured, fertilized and cultured in vitro until day 7. Some immature COCs, all uncleaved COCs, all blocked cleaved embryos (2 to 16 cells) and all transferable embryos (tight morulae and blastocysts) were separately analysed by real-time PCR assay. Brucella spp. DNA was detected in both blood and nasal mucus of all subjects, whereas no trace of DNA of Brucella spp. was found on either COCs or embryos. Currently, the infected or seropositive buffaloes have to be slaughtered for sanitary reasons. Interestingly, the results of this preliminary trial suggest a possible utilization of the COCs from the infected subjects of high genetic value to obtain safe embryos.

The aim of this study was to evaluate the length and the characteristics of the oestrous cycle in Italian Mediterranean buffalo cows, undergone synchronization of ovulation. The trial was performed on 32 buffaloes synchronized by the Ovsynch Program, which consists of an injection of GnRH on day 0, PGF2α on day 7 and GnRH on day 9. Starting on day 10 (Day 0 of the new cycle). Buffaloes undergone ultrasound examination of the ovaries on alternate days until the following heat. Follicular growth and corpus luteum formation and dimensions were recorded as well as the number of follicular waves. Statistical analysis was performed by ANOVA. Four animals (12.5%) did not show signs of oestrous and were excluded from the trial. The mean length of the oestrous cycle was 23.7±3.4 days. In particular, 1 animal (3.6%) showed an oestrous cycle characterized by 1 follicular wave with a length of 16 days, 17 subjects (60.7%) showed 2 follicular waves with a cycle length of 22.4±2.3 days and 10 buffaloes (35.7%) showed 3 follicular waves with a cycle of 26.8±2.0 days. These results confirm previous reports performed in buffalo species, although the cycle resulted longer in the 3-waves group.

In buffalo species embryonic mortality is considered one of the major causes of fertility loss, especially in the animals that are not mated during their reproductive period. Embryonic loss in animals mated by artificial insemination (AI) is 20-40% during seasons characterized by high number of light hours. Also in buffalo naturally mated the incidence of embryonic mortality is about 20% and a higher incidence is observed between 28- 60 days of gestation in buffaloes that conceive during increasing daylight length. A reduced capacity to secrete progesterone seems to explain in part this embryonic mortality but other as yet unidentified factors contribute between 40-50% to the embryonic losses. Treatments with hCG, GnRH agonist or progesterone on Days 5 after AI not always reduce embryonic mortality in buffalo species. Embryonic mortality in buffaloes appears to occur later (Day 25-40) than in cattle and P4 treatments should perhaps be applied later in buffaloes.

The growing rate of 240 male buffaloes fed ad libitum a diet characterized by 0.9 MFU/Kg dry matter (DM), 14% CP and 38:62 forage:concentrate ratio (Diet A) was evaluated starting from a mean age of 148 (Group 1), 218 (Group 2), 302 (Group 3), 320 (Group 4), 374 (Group 5) and 596 (Group 6) days. Data were compared at the weight of 400 kg and at the age of 550 days, values that were reached by all the subjects. The earlier administration of Diet A increased daily weight gain and reduced DM intake/die and feed conversion index compared to late administration. This trial further confirms the importance of satisfying nutritional requirements during the first months of life in buffalo species, due to the fact that these animals are not able to show a compensative weight gain like bovines.

Buffalo cows were submitted to three superovulatory treatments. T1 (n = 7): PRID for 10 days (d0-d9) plus decreasing doses of 500 IU FSH/LH (12 h-intervals d7‑d10); T2 (n = 8): PRID for 11 d (d0-d10) plus 2000 IU PMSG at d7; T3 (n = 9): PRID for 11 d plus 2000 IU PMSG at d7 and decreasing doses of 175 IU FSH/LH (12 h-intervals d10‑ d11). Overall plasma inhibin‑A (In-A) concentrations correlated with large follicles (LF, diameter >6mm, R=0.83, P10 mm at d12- 13 (T1=5.0+/-1.4, T2=1.2+/-0.9, T3=8.3+/-2.3). In-A concentrations significantly rised at d11-13 of T1 and T3. In-A seems a good indicator of the follicular development during superovulation in buffalo cows, while E2 is not. Furthermore T3 was followed by better ovarian follicular responses.

The aim of this study was to verify if the administration of a luteolytic dose of PGF2α at the time of AI is able to anticipate ovulation and, hence, to increase fertility rate. Furthermore, the presence of a “farm effect” was also evaluated. The trial was performed in two commercial farms (A and B) located in Caserta on 584 buffaloes (Farm A: n=389; Farm B: n=195), synchronized by the Ovsynch-TAI Program. In each farm buffaloes were divided in two groups: treated group (n=181 and n=95, respectively in farms A and B) received an intramuscular injection of 0.524 mg cloprostenol (Estrumate®, Schering-Plough Animal health, S.p.A., Milan, Italy) at the time of AI while control group (n=208 and n=100) received an intramuscular injection of physiologic saline (0.9% NaCl). No differences were present with regard to the number of AI/subject, in terms of pregnancy rate and ovulation rate between treated and control groups. The aim of the present study was to examine whether treatment with a GnRH agonist, hCG or P4 on Day 25 after AI increased P4 concentrations and reduced the incidence of embryonic mortality (EM) in pregnant buffaloes mated in mid-winter in a Mediterranean environment. The trial was carried out in two farms characterized, in previous years, by low (LEM Group), 153 buffaloes (DIM=150±7 days), and high (HEM Group), 284 buffaloes (DIM=163±5 days), incidence of embryo mortality. Animals were synchronized by Ovsynch-TAI Program and artificially inseminated. On day 25, pregnant buffaloes were randomly assigned to four groups: Control (no treatment), GnRH agonist (buserelin acetate, 12.6 μg), hCG (1500 IU) and P4 (341 mg of P4 i.m. every 4 days for three times). Progesterone (pg/ml) was determined in milk whey on Days 10, 20 and 25 after AI in all buffaloes and in Days 30 and 45 only in buffaloes pregnant on day 25 and assigned to four groups of treatment. Pregnancy diagnosis was undertaken on Day 45 by ultrasound. All treatments increased P4 milk whey and reduced embryonic mortality in buffalo cows bred in the farm characterized by high EM.

Aim of this study was to estimate the average intercalving period of a buffalo herd/population, by using the percentage of subjects that are able to conceive within 90 days post-partum, and that, consequently, have an intercalving period lower than 400 days, in order to give a precocious judgment on its fertility. A total number of 8,845 intercalving periods were recorded in 4 farms and 4 Provinces of Campania and Lazio Regions. In all the farms the out of breeding season mating technique was adopted and a constant diet was administered throughout the year. Values were grouped for primiparous and pluriparous buffaloes and were divided in four calving periods: January-March; April-June; July-September; October-December. Within each group/farm ANOVA was performed and the means were analysed by t Student test. In each group the mean value and the percentage of subjects with an intercalving period lower than 400 days (% < 400 days) were calculated and the regression analysis was carried out between the values of the intercalving periods and the % < 400 days. Finally, an all-in regression analysis was performed by using the data of 55 groups, excluding those with less than 25 values. The regression analysis between % < 400 days and the mean value of the intercalving period was always significant, except in one case. The comparison between the real values and those calculated allows to affirm that, if the percentage of subjects with an intercalving period lower than 400 days is higher than 50%, an intercalving period lower than 450 days will be probably recorded.

The aim of this study was to evaluate the relationships between 44 components and/or characteristics of milk samples collected every 50 days from 60 buffaloes (326 samples and 14,344 values). The animals were half sib (same father or same mother) and were bred in two farms. Significant associations but with a low values of coefficient of correlation were present, demonstrating that lactodinamographic parameters marginally affect cheese yield.

The aim of this study was to evaluate the effects of two different diets on fertility rate in 64 Mediterranean buffalo cows. The animals were divided in two groups: Group S (n=32) received a total mixed ration based on haysilage of Italian ryegrass (Lolium multiflorum), hay and concentrates, whereas group H (n=32) was fed only hay and concentrates. Both diets consisted of 50-55 % forages and 45-50 % concentrates and contained 0.91 MFU/kg dry matter (DM) and 16% crude protein/DM. From February until September the cows were located together with bulls and pregnancy diagnosis was performed monthly. Furthermore, blood and milk samples were monthly collected on 20 animals in each group. Statistical analysis was performed by χ2 and t Student tests. Thirty adult buffaloes were used for this study. The animals were synchronized by PRID+PMSG+PGF2α and artificially inseminated. The ovaries were ultrasonografically examined to study the preovulatory follicle (PF) and the successive corpus luteum (CL). At the same time, blood sample were collected for P4 determination carried out by RIA. On the basis of echogenic appearance, the corpora lutea (CLs) were classified as belonging to grade 1, 2 or 3. Significant difference was found in the diameter of PF and in that of CL in relation to the CL echogenic appearance. Well defined granular CL, distinguishable from the ovarian stroma and dark grey in colour (grade 3) resulted to have a bigger diameter compared with CLs belonging to a grade 1 or 2, and resulted to derive from a larger PF. Positive correlation was found between plasma P4 concentration and CL diameter. The PF diameter was larger in buffaloes become pregnant after AI respect to the non-pregnant. The CL diameter, also, was significantly larger in the pregnant buffalo compared to the non-pregnant since the first observation at day 7, while significant difference in P4 value was found starting from day 21 after AI.

The aim of this work was to determine the effect of intensive feeding (IF group) or pasture system (PS group) on the onset of puberty and on the ovarian size in buffaloes. The trial was carried out on 26 prepubertal 8-months old buffalo heifers weighing a mean of 134 kg (n =13 per group). The heifers were weighed monthly and, starting from the 13th month of age, blood samples were collected every 10 days to determine the concentration of plasma progesterone by RIA. Heifers were considered to have achieved puberty and cyclic ovarian activity when plasma progesterone levels exceeded 1 ng/ml for two samples with a lower intermediate value. In buffalo the overall in vitro embryo production efficiency is lower than cattle, mainly due to the lower cleavage rate (Gasparrini, 2002). In fact in our experience, comparable embryo yields are obtained in the two species only in relation to the cleaved oocytes. It is thought that a major limiting factor is the quality of the frozen semen. It has been demonstrated that freezing results in acrosomal damage, leakage of enzymes, alterations in ionic strength and pH, complete withdrawal of the hydration shell of protein in the solution and loss of motility (Meur et al., 1988). On the other hand we cannot role out that the lower cleavage rate is related to the failure in the accomplishment of oocyte competence after maturation. In vitro embryo production (IVEP) is largely applied world wide to animal breeding. One of the principal steps of the IVEP is represented by embryo culture (Khurana and Niemann., 2000). In the past, embryos were grown in co-culture systems with other cells such as oviductal epithelial cells, cumulus cells, Buffalo rat liver (BRL) and VERO cells (Duszewska et al., 2000). These cells are able to supply the nutrients for embryo development by their replication and metabolism. Nevertheless, the metabolic activity of these cells is also responsible of an early lowering of pH in the culture medium: that needs to be changed every two days. Furthermore, with this culture system it is impossible to standardize all the procedure: in fact the result is dependent from several variables, as the quality of the cells and their concentration in co-culture. The use of defined culture media is necessary to acquire a better comprehension of metabolism and biochemical requirements for IVEP........

The authors described the effects of diets characterized by different energy density and forages concentrations on reproductive activity of primiparous and pluriparous lactating buffalo cows, undergone the out of season breeding technique. Productive and reproductive data of a buffalo farm in Salerno were collected. Furthermore, the diets administered to the animals from 1998 to 2003 (6 years) were also recorded. The components of the diet were monthly analysed according to the method described by ASPA (1980). The fertility in primiparous buffaloes resulted significantly better (P< 0.01) during the last year when the diet were characterized by high energy and less quantity of forage.
