Abstract

We present the first catalog of very-high-energy and ultra-high-energy gamma-ray sources detected by the Large High Altitude Air Shower Observatory. The catalog was compiled using 508 days of data collected by the Water Cherenkov Detector Array from 2021 March to 2022 September and 933 days of data recorded by the Kilometer Squared Array from 2020 January to 2022 September. This catalog represents the main result from the most sensitive large coverage gamma-ray survey of the sky above 1 TeV, covering decl. from −20° to 80°. In total, the catalog contains 90 sources with an extended size smaller than 2° and a significance of detection at >5 σ . Based on our source association criteria, 32 new TeV sources are proposed in this study. Among the 90 sources, 43 sources are detected with ultra-high energy ( E > 100 TeV) emission at >4 σ significance level. We provide the position, extension, and spectral characteristics of all the sources in this catalog.

Abstract

Abstract The focal plane camera is the core component of the Wide Field-of-view Cherenkov/fluorescence Telescope Array (WFCTA) of the Large High-Altitude Air Shower Observatory (LHAASO). Because of the capability of working under moonlight without aging, silicon photomultipliers (SiPM) have been proven to be not only an alternative but also an improvement to conventional photomultiplier tubes (PMT) in this application. Eighteen SiPM-based cameras with square light funnels have been built for WFCTA. The telescopes have collected more than 100 million cosmic ray events and preliminary results indicate that these cameras are capable of working under moonlight. The characteristics of the light funnels and SiPMs pose challenges (e.g. dynamic range, dark count rate, assembly techniques). In this paper, we present the design features, manufacturing techniques and performances of these cameras. Finally, the test facilities, the test methods and results of SiPMs in the cameras are reported here.

Abstract

Observation of high energy and very high emission from Gamma Ray Bursts (GRBs) is crucial to study the gigantic explosion and the underline processes. With a large field-of-view and almost full duty cycle, the Water Cherenkov Detector Array (WCDA), a sub-array of the Large High Altitude Air Shower Observatory (LHAASO), is appropriate to monitor the very high energy emission from unpredictable transients such as GRBs. Nevertheless, the main issue for an extensive air shower array is the high energy threshold which limits the horizon of the detector. To address this issue a new trigger method is developed in this article to lower the energy threshold of WCDA for GRB observation. The proposed method significantly improves the detection efficiency of WCDA for gamma-rays around the GRB direction at 10-300 GeV. The sensitivity of the WCDA for GRB detection with the new trigger method is estimated. The achieved sensitivity of the quarter WCDA array above 10 GeV is comparable with that of Fermi-LAT. The data analysis process and corresponding fluence upper limit for GRB 190719C is presented as an example.

Abstract

The kilometer square array (KM2A) of the large high altitude air shower observatory (LHAASO) aims at surveying the northern γ-ray sky at energies above 10 TeV with unprecedented sensitivity. γ-ray observations have long been one of the most powerful tools for dark matter searches, as, e.g., high-energy γ rays could be produced by the decays of heavy dark matter particles. In this Letter, we present the first dark matter analysis with LHAASO-KM2A, using the first 340 days of data from 1/2-KM2A and 230 days of data from 3/4-KM2A. Several regions of interest are used to search for a signal and account for the residual cosmic-ray background after γ/hadron separation. We find no excess of dark matter signals, and thus place some of the strongest γ-ray constraints on the lifetime of heavy dark matter particles with mass between 105 and 109 GeV. Our results with LHAASO are robust, and have important implications for dark matter interpretations of the diffuse astrophysical high-energy neutrino emission.

Abstract

The main scientific goal of LHAASO-WCDA is to survey gamma-ray sources with energy from 100 GeV to 30 TeV. To observe high-energy shower events, especially to measure the energy spectrum of cosmic rays from 100 TeV to 10 PeV, a dynamic range extension system (WCDA++) is designed to use a 1.5-inch PMT with a dynamic range of four orders of magnitude for each cell in WCDA-1. The dynamic range is extended by using these PMTs to measure the effective charge density in the core region of air shower events, which is an important parameter for identifying the composition of primary particles. The system has been running for more than one year. In this paper, the details of the design and performance of WCDA++ are presented.

Abstract

The extension of the cosmic-ray spectrum beyond 1 petaelectronvolt (PeV; 1015 electronvolts) indicates the existence of the so-called PeVatrons—cosmic-ray factories that accelerate particles to PeV energies. We need to locate and identify such objects to find the origin of Galactic cosmic rays1. The principal signature of both electron and proton PeVatrons is ultrahigh-energy (exceeding 100 TeV) γ radiation. Evidence of the presence of a proton PeVatron has been found in the Galactic Centre, according to the detection of a hard-spectrum radiation extending to 0.04 PeV (ref. 2). Although γ-rays with energies slightly higher than 0.1 PeV have been reported from a few objects in the Galactic plane3–6, unbiased identification and in-depth exploration of PeVatrons requires detection of γ-rays with energies well above 0.1 PeV. Here we report the detection of more than 530 photons at energies above 100 teraelectronvolts and up to 1.4 PeV from 12 ultrahigh-energy γ-ray sources with a statistical significance greater than seven standard deviations. Despite having several potential counterparts in their proximity, including pulsar wind nebulae, supernova remnants and star-forming regions, the PeVatrons responsible for the ultrahigh-energy γ-rays have not yet been firmly localized and identified (except for the Crab Nebula), leaving open the origin of these extreme accelerators. Observations of γ-rays with energies up to 1.4 PeV find that 12 sources in the Galaxy are PeVatrons, one of which is the Crab Nebula.

Abstract

We report the discovery of a new unidentified extended $\gamma$-ray source in the Galactic plane named LHAASO J0341+5258 with a pre-trial significance of 8.2 standard deviations above 25 TeV. The best fit position is R.A.$=55.34^{\circ}\pm0.11^{\circ}$ and Dec$=52.97^{\circ}\pm0.07^{\circ}$. The angular size of LHAASO J0341+5258 is $0.29^\circ \pm 0.06^\circ_{stat} \pm0.02^\circ_{sys}$. The flux above 25 TeV is about $20\%$ of the flux of Crab Nebula. Although a power-law fit of the spectrum from 10 TeV to 200 TeV with the photon index $\alpha=2.98 \pm 0.19_{stat} \pm 0.02_{sys}$ is not excluded, the LHAASO data together with the flux upper limit at 10 GeV set by the Fermi LAT observation, indicate a noticeable steepening of an initially hard power-law spectrum %($\alpha \leq 1.75$) spectrum with a cutoff at $\approx 50$ TeV. We briefly discuss the origin of UHE gamma-rays. The lack of an energetic pulsar and a young SNR inside or in the vicinity of LHAASO J0341+5258 challenge, but do not exclude both the leptonic and hadronic scenarios of gamma-ray production., Comment: Accepted by APJL

Abstract

Recently, the LHAASO Collaboration published the detection of 12 ultrahigh-energy γ-ray sources above 100 TeV, with the highest energy photon reaching 1.4 PeV. The first detection of PeV γ rays from astrophysical sources may provide a very sensitive probe of the effect of the Lorentz invariance violation (LIV), which results in decay of high-energy γ rays in the superluminal scenario and hence a sharp cutoff of the energy spectrum. Two highest energy sources are studied in this work. No signature of the existence of the LIV is found in their energy spectra, and the lower limits on the LIV energy scale are derived. Our results show that the first-order LIV energy scale should be higher than about 10^{5} times the Planck scale M_{Pl} and that the second-order LIV scale is10^{-3}M_{Pl}. Both limits improve by at least one order of magnitude the previous results.

Abstract

A wide field-of-view (FoV) is one of the most important goals for future ground-based γ -ray astronomy, and it is especially crucial for detailed studies of temporal and spectral characteristics of solitary events, such as γ -ray bursts (GRBs), highly variable sources like blazars, or potential very high energy (VHE) electromagnetic counterparts of gravitational waves (GWs). Atmospheric Cherenkov Telescope (ACT) based on refractive optics and operated at a very high altitude observatory may be an appropriate solution. In the present study, we report the characteristics and performance of a large FoV ACT prototype based on a refractive convex water lens. A reconstruction method for estimating the arrival direction of the air shower was developed by combining the water-lens telescope and the Extensive Air Shower (EAS) array. The Cherenkov images of the air showers were obtained by a water-lens prototype with 48 pixels, an FoV of 15 ° × 13 ° , and a median angular resolution of ∼ 0 . 9 ° .

Abstract

We report the discovery of an extended very-high-energy (VHE) gamma-ray source around the location of the middle-aged (207.8 kyr) pulsar PSR J0622+3749 with the Large High-Altitude Air Shower Observatory (LHAASO). The source is detected with a significance of 8.2σ for E25 TeV assuming a Gaussian template. The best-fit location is (right ascension, declination) =(95.47°±0.11°,37.92°±0.09°), and the extension is 0.40°±0.07°. The energy spectrum can be described by a power-law spectrum with an index of -2.92±0.17_{stat}±0.02_{sys}. No clear extended multiwavelength counterpart of the LHAASO source has been found from the radio to sub-TeV bands. The LHAASO observations are consistent with the scenario that VHE electrons escaped from the pulsar, diffused in the interstellar medium, and scattered the interstellar radiation field. If interpreted as the pulsar halo scenario, the diffusion coefficient, inferred for electrons with median energies of ∼160 TeV, is consistent with those obtained from the extended halos around Geminga and Monogem and much smaller than that derived from cosmic ray secondaries. The LHAASO discovery of this source thus likely enriches the class of so-called pulsar halos and confirms that high-energy particles generally diffuse very slowly in the disturbed medium around pulsars.

Abstract

Strong-field ionization of polar molecules contains rich dynamical processes such as tunneling, excitation, and Stark shift. These processes occur on a sub-cycle time scale and are difficult to distinguish in ultrafast measurements. Here, with a developed strong-field model considering effects of both Coulomb and permanent dipole, we show that photoelectron momentum distributions (PMDs) in orthogonal two-color laser fields can be utilized to resolve these processes with attosecond-scale resolution. A feature quantity related to the asymmetry in PMDs is obtained, with which the complex electron dynamics of polar molecules in each half laser cycle is characterized and the subtle time difference when electrons escaping from different sides of the polar molecule is identified., Comment: 6 pages, 4 figures

Abstract

We report the discovery of a UHE gamma-ray source, LHAASO J2108+5157, by analyzing the LHAASO-KM2A data of 308.33 live days. Significant excess of gamma-ray induced showers is observed in both energy bands of 25-100 TeV and $\gt$100 TeV with 9.5 sigma and 8.5 sigma, respectively. This source is not significantly favored as an extensive source with the angular extension smaller than the point-spread function of KM2A. The measured energy spectrum from 20 to 200 TeV can be approximately described by a power-law function with an index of -2.83$\pm$ 0.18stat. A harder spectrum is demanded at lower energies considering the flux upper limit set by Fermi-LAT observations. The position of the gamma-ray emission is correlated with a giant molecular cloud, which favors a hadronic origin. No obvious counterparts have been found, deeper multiwavelength observations will help to shed new light on this intriguing UHE source., Comment: 12 pages, 7 figures, accepted by ApJL

Abstract

The main scientific goal of the LHAASO WFCTA experiment is to measure the cosmic ray energy spectra and composition from 10 TeV to 1 EeV. Cherenkov photons in the extensive air shower measured by the SiPM camera of Cherenkov telescopes can be used to reconstruct the cosmic ray energy. The absolute calibration of the camera is a crucial step to achieve the accurate measurement of the cosmic ray energy spectrum. A multi-wavelength cylindrical illuminator based on LEDs is developed and mounted inside the telescope to calibrate and monitor the camera, and the illuminator’s stability is better than 0.5% under the temperature variation from -26 to 26 ° C. A portable probe with a single photoelectron resolution of 21.6% is developed. After calibration by National Institute of Metrology, China (NIM), the probe is taken to the LHAASO site to measure the absolute photon density of the cylindrical illuminator inside the telescope. Based on the illuminator with known photon density, the photon conversion factor of the camera can be calibrated, and the overall calibration uncertainty is less than 2.6%.

Abstract

The Water Cherenkov Detector Array (WCDA) is a major component of the Large High Altitude Air Shower Array Observatory (LHAASO), a new generation cosmic-ray experiment with unprecedented sensitivity, currently under construction. The WCDA is aimed at the study of TeV $\gamma$-rays. In order to evaluate the prospects of searching for TeV $\gamma$-ray sources with the WCDA, we present in this paper a projection for the one-year sensitivity of the WCDA to TeV $\gamma$-ray sources from TeVCat (footnote: http://tevcat.uchicago.edu) using an all-sky approach. Out of 128 TeVCat sources observable to the WCDA up to a zenith angle of $45^\circ$, we estimate that 42 would be detectable for one year of observations at a median energy of 1 TeV. Abstract

The LHAASO-WFCTA experiment, which aims to observe cosmic rays in the sub-EeV range using the fluorescence technique, uses a new generation of high-performance telescopes. To ensure that the experiment has excellent detection capability associated with the measurement of the energy spectrum, the primary composition of cosmic rays, and so on, an accurate geometrical reconstruction of air-shower events is fundamental. This paper describes the development and testing of geometrical reconstruction for stereo viewed events using the WFCTA (Wide Field of view Cherenkov/Fluorescence Telescope Array) detectors. Two approaches, which take full advantage of the WFCTA detectors, are investigated. One is the stereo-angular method, which uses the pointing of triggered SiPMs in the shower trajectory, and the other is the stereo-timing method, which uses the triggering time of the fired SiPMs. The results show that both methods have good geometrical resolution; the resolution of the stereo-timing method is slightly better than the stereo-angular method because the resolution of the latter is slightly limited by the shower track length.

Abstract

The first stage of LHAASO-KM2A has been completed in Sichuan, China. KM2A is composed of electromagnetic particle detectors (ED) and muon detectors (MD), focusing on gamma ray astronomy above 30 TeV and cosmic ray physics in the “knee” region. The ED array in the first stage is installed into two clusters and the distance between the two clusters is around 230 meters. The MDs are distributed in one of the clusters. Each MD is covered with soil of 2.5 meters thick. We analyzed the lateral distribution of muons which are 140-300 meters away from Shower axis, and the primary cosmic ray energy is about 100TeV.

Abstract

A sub-array of the Large High Altitude Air Shower Observatory (LHAASO), KM2A is mainly designed to observe a large fraction of the northern sky to hunt for γ-ray sources at energies above 10 TeV. Even though the detector construction is still underway, half of the KM2A array has been operating stably since the end of 2019. In this paper, we present the KM2A data analysis pipeline and the first observation of the Crab Nebula, a standard candle in very high energy γ-ray astronomy. We detect γ-ray signals from the Crab Nebula in both energy ranges of 10 100 TeV and 100 TeV with high significance, by analyzing the KM2A data of 136 live days between December 2019 and May 2020. With the observations, we test the detector performance, including angular resolution, pointing accuracy and cosmic-ray background rejection power. The energy spectrum of the Crab Nebula in the energy range 10-250 TeV fits well with a single power-law function dN/dE = (1.13 0.05 0.08 ) 10 (E/20 TeV) cm s TeV . It is consistent with previous measurements by other experiments. This opens a new window of γ-ray astronomy above 0.1 PeV through which new ultrahigh-energy γ-ray phenomena, such as cosmic PeVatrons, might be discovered.

Abstract

The muonic component of the extensive air showers (EAS) is of great importance for the astroparticle physics. It carries the information about the properties of primary cosmic ray (CR) particles, such as their mass, and electromagnetic and hadronic nature. It provides a sensitive test for the hadronic interaction models, which are inevitable for describing the cascade shower development of cosmic rays in EAS experiments. The YangBaJing Hybrid Array (YBJ-HA) experiment has been in operation since the end of 2016. Surface detectors are used for the measurements of primary energy, angular direction and core position of a shower event, while underground muon detectors are used for measuring the density of muons at various locations. Using the data obtained by the YBJ-HA experiment, this work reports the first measurement of the lateral muon distribution for the primary cosmic ray energy in the 100 TeV region. The punch-through effect is evaluated via MC simulation.

Abstract

In the present paper, we investigate the correlated electron emission of atoms irradiated by a few-cycle laser pulse, with emphasis on the correlated longitudinal momentum spectra. We find that the spectra show clear v-shaped structures, in analogy to what was observed recently in long-pulse experiments. Moreover, the patterns of the spectra depend sensitively on the carrier-envelope phase as well as the laser intensity. The v-shaped structure is more pronounced at lower and higher intensities and becomes obscure at medium intensity. At a lower intensity, upon change of the phase from 0 to {pi}/2, the v-shaped structure shifts from the first quadrant to the third quadrant and the ratios between the double ionization yields in the first and third quadrants are found to increase by a few orders of magnitude. The semiclassical rescattering model is exploited in the preceding calculations and the underlying mechanisms are uncovered by analyzing the subcycle dynamics of classical trajectories.

Abstract

Objective: To examine the impact of berberine on polycystic ovary syndrome (PCOS) in mice, and to investigate the effects of berberine on the intestinal flora and the intestinal flora on PCOS. Methods: A mouse model of PCOS was established by administering dehydroepiandrosterone in combination with high fat diet, and the mouse model was given a berberine treatment. The study consisted of a blank control group (C group), a PCOS model group (M group) and a berberine treatment group (T group). During the experiment, the mice were closely monitored through timed body weight measurements and estrous cycle monitoring; intraperitoneal glucose tolerance test and insulin tolerance test were done. Upon completion of the pharmacological intervention, the wet weights of liver, ovary and fat deposits of mice were assessed and subjected to HE staining to confirm the success of PCOS modeling and the efficacy of berberine. Additionally, fecal samples were analyzed for intestinal flora through 16S rRNA analysis. Results: The PCOS model was established successfully, berberine alleviated the disturbance of estrous cycle in mice, and significantly alleviated fat accumulation and metabolic abnormalities of glucose in mice. The cross-sectional area of fat pad cells in T group was (2 858±146) μm², which was significantly lower than that in M group [(9 518±347) μm²], and the difference was statistically significant ( P <0.001). The blood glucose levels in T group were significantly lower than those in M group ( P <0.05). The composition and structure of intestinal flora in mice of M group with PCOS (compared with C group) and in mice of T group after berberine intervention (compared with M group) were significantly altered. However, alpha diversity did not change significantly among three groups ( P >0.05). Conclusion: Berberine could alleviate PCOS by intervening in the alterations of gut microbiota.

Abstract

1. Inosine monophosphate (IMP), is an essential component for meat flavour and microRNAs (miRNAs) play a vital role in its post-transcriptional regulation. However, the mechanism of how miRNA expression affects muscle-specific IMP deposition is unclear.2. The following study performed transcriptome sequencing and bioinformatics analysis of breast and leg muscle, which have significantly different IMP content in Jingyuan chicken. The differential miRNA-mRNAs were screened out and correlation analysis with IMP content was performed.3. A total of 39 differentially expressed miRNAs (DE miRNAs) and 666 differentially expressed mRNAs (DE mRNAs) were identified between breast muscles and leg muscles. Using miRNA-mRNA integrated analysis, 29 miRNA-target gene pairs were obtained, composed of 13 DE miRNAs and 28 DE mRNAs. Next, purine metabolism, glycolysis/gluconeogenesis, pyruvate metabolism and the biosynthesis of amino acid pathways as necessary for muscle IMP-specific deposition were identified. The differentially expressed gene PKM2 , which was significantly enriched in all four pathways, is involved in IMP anabolism in the form of energy metabolism and enzyme activity regulation. The correlation analysis suggested that the gga-miR-107-3p-KLHDC2 negative interaction may be a key regulator in IMP deposition.4. This study explores the functional mechanism of IMP-specific deposition in Jingyuan chicken muscles at the miRNA and mRNA levels and highlights multiple candidate miRNAs and mRNAs for molecular-assisted breeding.

Abstract

Objective: There are many challenges related to the treatment of coronary atherosclerotic heart disease (CAD). Studies have confirmed that Epimedium extract inhibits myocardial injury induced by myocardial ischaemia, but the mechanism of action remains unclear. This study aimed at analysed the effective components and mechanisms of Epimedium in treating CAD based on network pharmacology and molecular docking studies and to verify the mechanism in vitro., Materials and Methods: The TCMSP and UniProt databases were used to filter for the active components and drug targets of Epimedium. The GeneCards database was used to screen disease targets associated with CAD. The intersection of the drug targets of Epimedium and the disease targets of coronary heart disease was studied to identify the targets of Epimedium in the treatment of CAD. Cytoscape software was used to establish and analyse an activity-target network. The STRING database was used to analyse a protein-protein interaction (PPI) network, and proteins in the PPI network were visualized in the R language. Bioconductor software was used for GO function and KEGG pathway enrichment analyses, and visualization analysis was performed in the R language. PyMOL software was used to verify the molecular docking between selected active components of Epimedium and the targets of CAD, and the potential key effective components of Epimedium in the treatment of coronary heart disease were identified. The involvement of the PI3K/Akt pathway was validated by Western blot analysis., Results: (1) Twenty-three active compounds, including Epimedium glycoside, quercetin, luteolin, and olive resin, were screened out. There were 68 common targets of Epimedium and CAD, including IL-6, ESR1, RELA, FOS, NCOA1, CCND1, EGFR, MAPK8, VEGFA, and CASP8. The potential signaling pathways involved in the treatment of CAD by Epimedium included the human cytomegalovirus infection pathway, the PI3K-Akt signaling pathway, the TNF signaling pathway, and the HIF-1 signaling pathway. (2) Luteolin, quercetin, sitosterol, and anhydroicaritin showed strong binding to targets of CAD based on molecular docking studies. (3) Epimedium extract increased the expression of PI3K, Akt and P-Akt but decreased the expression of IL-6  in vitro., Conclusions: (1) Icariin, quercetin and luteolin may act on target proteins, including IL-6, ESR1, EGFR, MAPK8, VEGFA and CASP8, to participate in the regulation of the human cytomegalovirus infection pathway, the PI3K-Akt signaling pathway, the TNF signaling pathway and other signaling pathways in order to effectively treat CAD. (2) In vitro studies confirmed that Epimedium extract can treat CAD by upregulating PI3K, Akt and P-Akt protein expression and downregulating IL-6 protein expression in SD rat cardiomyocytes.

Abstract

Background: Winter injury often threatens phenology and walnut survival in late spring and winter in northern areas of China., Objective: This study aimed at evaluating seven walnut varieties which have superior economic traits., Materials and Methods: Mature functional leaves were observed by paraffin section and the related anatomical structure indices were measured. The stem segments were placed at six temperatures [4 (control), -10, -15, -20, -25, -30 degree C] for low temperature stress treatment, and the relevant indices were measured., Results: The thickness of palisade tissue (PT), sponge tissue (ST) and the PT/ST of Wen185 was significantly higher than in Longboxiang 2, Longboxiang 3 and Suizhuang. However, the leaf thickness (LT) of Longboxiang 3 was markedly lower than that of other varieties. With the decrease in temperature, relative electrolyte leakage (REL), proline (Pro) and soluble sugar content (SS) of all varieties increased, while superoxide dismutase (SOD) and peroxidase (POD) activities showed a rise and fall trend. According to the LT50, the cold tolerance of seven walnut varieties was ranked from high to low, viz. Yuanlin > Wen185 > Xiangling > Suizhuang > Qingxiang > Longboxiang 2 > Longboxiang 3., Conclusion: The variety Yuanlin can adapt to low temperature and should be widely promoted. doi.org/10.54680/fr22210110312.

Abstract

Gammaherpesviruses establish lifelong infection and are associated with a variety of cancers, including B cell lymphomas. These viruses manipulate the B cell differentiation process to establish lifelong infection in memory B cells. Specifically, gammaherpesviruses infect naive B cells and promote entry of both infected and uninfected naive B cells into germinal centers, where the virus usurps rapid proliferation of germinal center B cells to exponentially increase its cellular latent reservoir. In addition to facilitating the establishment of latent infection, germinal center B cells are thought to be the target of viral transformation. In this study, we have uncovered a novel proviral role of host interleukin 17A (IL-17A), a well-established antibacterial and antifungal factor. Loss of IL-17A signaling attenuated the establishment of chronic gammaherpesvirus infection and gammaherpesvirus-driven germinal center response in a route of inoculation-dependent manner. Further, IL-17A treatment directly supported gammaherpesvirus reactivation and de novo lytic infection. This study is the first demonstration of a multifaceted proviral role of IL-17 signaling. IMPORTANCE Gammaherpesviruses establish lifelong infections in a majority of humans and are associated with B cell lymphomas. IL-17A is a host cytokine that plays a well-established role in the clearance of bacterial and fungal infections; however, the role of IL-17A in viral infections is poorly understood. In this study, we show that IL-17A signaling promoted the establishment of chronic gammaherpesvirus infection following the mucosal route of infection, viral lytic replication, and reactivation from latency. Abstract

Triggering receptor expressed on myeloid cells-1 (TREM-1) is expressed on neutrophils and monocyte/macrophages and amplifies Toll-like receptor-mediated inflammation during infection. TREM-1 also exists in an antagonistic soluble form (sTREM-1) that has been used as a peripheral biomarker in sepsis, though the mechanisms of its release are not entirely clear. The requirement of TREM-1 in single microbial infections is controversial, with some studies showing a protective role and others a contribution to immunopathology. Furthermore, the role of membrane-bound and sTREM-1 in polygenic infections is currently unknown. In a mouse co-infection model where preceding viral infection greatly enhances bacteria co-infection, we now determine a mechanisms for the striking increase in sTREM-1 and the loss of TREM-1 on surface of neutrophils. We identified a matrix metalloproteinase (MMP)-9 cleavage site in TREM-1 and that the increase of MMP-9 in bronchoalveolar lavage fluid mirrors sTREM-1 release. In vitro studies with neutrophils and MMP-9 and the reduction of sTREM-1 in vivo after MMP-9 inhibition verifies that this enzyme cleaves TREM-1. Intriguingly, MMP-9 inhibition significantly reduces bacterial load and ensuing immunopathology in a co-infection model. This highlights MMP-9 inhibition as a potential therapeutic via blocking cleavage of TREM-1.

Abstract

Objective: Ovarian cancer is the most lethal gynecologic cancer worldwide, since most patients are diagnosed at an advanced stage. To improve the early diagnosis and treatment of ovarian cancer, we performed a integrated analysis of transcription profile and genetic variations to study on the molecular pathogenesis in ovarian cancer., Methods: mRNA expression profiles of ovarian cancer and normal controls downloaded from ArrayExpress database were applied to identify differentially expressed genes (DEGs). The chromosomal distributions of these DEGs were established using DAVID. Then, DNASeq data from the Cancer Genome Atlas (TCGA) were extracted to analyze gene mutational information including the number of mutations (mut), the number of mutational genes (mutG) and chromosomal distributions of mutations. Statistical method was offered to carrying on correlation analysis of gene mutations and differential expression., Results: A total of 1732 DEGs were identified, and the chromosomal distributions of 97 genes were unknown. These DEGs were most significantly distributed on chromosome 4 with p value = 1.34E-7. Chromosome 1 enriched the most DEGs (11.56%). Statistical algorithm showed that DEGs presented significantly positive correlation with mut (p = 0.000009) and mutG (p = 0.00001). In 48.7% DEGs, gene mutations were found., Conclusions: We conducted scientific analysis on integration of DEGs in expression profiles and genetic mutations in ovarian cancer, displayed the correlation of differential expression and genetic variations. The result indicated that expression profiles were significantly correlated to genotype.

29. Investigation of the effect of a disposal endotracheal catheter and metal catheter combination application in inhalation injury patients.

Abstract

Objective. The aim of this paper was to make an objective evaluation of the scientific nature and effectiveness of a combination application of two catheters by investigating and comparing the effects and the occurrence of complications using a single metal catheter application, a single disposal endotracheal catheter application, and a combination application of these two catheters separately in tracheotomy patients. Method. A study was carried out on 85 patients with a metal catheter from 1990 to 1997, 62 patients with a disposal endotracheal catheter from 1998 to 2002, and 56 patients with an on-vent disposal endotracheal catheter and an off-vent metal catheter from 2003 to November 2008, defined respectively as groups A, B, and C, in order to observe catheter balloon rupture and catheter olisthe during intubation, as also sputum adhesion to the endotracheal catheter wall after application, and to assess the number of cases of patients with airway bleeding, the number of patients with endotracheal catheter blockage, and the time lapse between ventilator withdrawal and extubation. Results. Patients in groups B and C were observed to have balloon rupture; the number of patients with catheter olisthe and airway bleeding was in both cases less than in group A (p < 0.05); and in groups A and C the number of patients with catheter blockage was in both cases less than in group B (p < 0.05) - extubation time was also shorter than in group B, but without statistical significance (p > 0.05). Macroscopic observation found that there was a large amount of sputum adhering to the lumen of the open disposal endotracheal catheter and that the lumen became significantly narrow, while the metal catheter became non-significantly narrow. Conclusion. In inhalation injury patients we found that the effect of the combination application of the disposal endotracheal catheter and the metal catheter was better than that of the single application.

Abstract

Host genetic factors and environment factors including hepatitis B virus (HBV) genotype are widely viewed as common basis of the different outcomes of HBV infection. Human leucocyte antigen (HLA) plays an important role in immunological reaction to HBV infection. The study aimed to explore whether the HLA-DQB1 allele polymorphisms are associated with the outcomes of HBV infection in the Chinese Han population. One hundred and thirty-four asymptomatic HBV carriers and 139 chronic hepatitis B patients were recruited in this case-control study in Beijing, China. Sequence-specific primers-polymerase chain reaction was used to detect 13 alleles of HLA-DQB1 gene. The frequency distributions of alleles in two groups were analysed using sas 9.1.2 software. After adjustment of confounders the frequencies of HLA-DQB1*0503 allele and *0303 allele in chronic hepatitis B group were statistically significant lower than those in asymptomatic HBV carrier group (P = 0.04; P = 0.05), and the frequency of exposure to alcohol consumption in patients with chronic hepatitis B was clearly higher than that in asymptomatic HBV carrier group (P = 0.004). HLA-DQB1*0503 allele and *0303 allele are independently resistant genetic factors to chronic hepatitis B, and alcohol consumption is the risk factor of chronic hepatitis B.

Abstract

Objective: To explore the modulating effects of thymopentin (TP-5) on the immune system of severely burned patients., Methods: Sixty patients with severe burn were enrolled in this study and were randomly divided into treatment (T, n = 30) and control (C, n = 30) groups. The T lymphocytes and NK cells in peripheral blood in two groups were determined on the 2, 5, 8, 15 and 22 postburn days (PBDs). The patients in T group were treated with TP-5 in a dose of 1 mg/d for 20 days beginning on 2 PBD., Results: There was no difference of T lymphocytes and NK cells between the two groups on 2 PBD, and there was no difference of total number in T lymphocytes between the two groups on 5, 8, 15 and 22 PBDs. But the NK cell number and CD4/CD8 ratio in C group were much lower than those in T group., Conclusion: The function of T lymphocytes and NK cells in severely burned patients could be obviously improved by TP-5, which played immune modulating roles.

Abstract

Tandem pore domain acid-sensitive K(+) channel 3 (TASK-3) is a new member of the tandem pore domain potassium channel family. A cDNA encoding a 365- amino acid polypeptide with four putative transmembrane segments and two pore regions was isolated from guinea pig brain. An orthologous sequence was cloned from a human genomic library. Although TASK-3 is 62% identical to TASK-1, the cytosolic C-terminal sequence is only weakly conserved. Analysis of the gene structure identified an intron within the conserved GYG motif of the first pore region. Reverse transcriptase-polymerase chain reaction analysis showed strong expression in brain but very weak mRNA levels in other tissues. Cell-attached patch-clamp recordings of TASK-3 expressed in HEK293 cells showed that the single channel current-voltage relation was inwardly rectifying, and open probability increased markedly with depolarization. Removal of external divalent cations increased the mean single channel current measured at -100 mV from -2.3 to -5.8 pA. Expression of TASK-3 in Xenopus oocytes revealed an outwardly rectifying K(+) current that was strongly decreased in the presence of lower extracellular pH. Substitution of the histidine residue His-98 by asparagine or tyrosine abolished pH sensitivity. This histidine, which is located at the outer part of the pore adjacent to the selectivity filter, may be an essential component of the extracellular pH sensor.

34. [Experimental circumferential tracheal replacement with rib and costal muscle ring in dog.].

Abstract

Background: To evaluate the feasibility of using vascularized rib and costal muscle ring to repair large circumferential defect of thoracic trachea in dog., Methods: Eighteen adult mongrel dogs were used in this study. The third rib with adjacent intercostal muscles was engineered to a ring with vascular pedicle , which replaced five or eight rings of trachea., Results: Eleven of eighteen dogs were alive after operation , and the longest one had survived for 14 months. These dogs were active , although a slight stricture at the newly formed trachea was found. Microscopic examination showed the trachea had good blood supply , and the substitution of pleural epithelium by ciliated epithelium was observed., Conclusions: Circumferential tracheal replacement with vascularized rib and intercostal muscle ring is easily managed and has less complications. The results suggest that this technique might be clinically used in the future.

Abstract

Objective: To explore noxious stimulation-induced morphological changes of nitric oxide synthase (NOS) positive neurons in the tongue., Methods: The histochemical method of reduced nicotinamide adenine dinucleotide phosphate(NADPH)-diaphorase activity was used., Results: NOS positive neuron bodies in the tongue were obviously enlarged, the staining intensity increased and nerve fibers became thicker after injury. At the body and tip of the tongue, near the body cells of NOS positive neurons deeply stained fine granular particles were seen, and structures of plum blossom form were seen along the route of the nerve fiber extending all the way to the terminal. NOS positive nerve fibers in the striated muscles and around the gland in the tongue root became thicker, and bead structures were seen in some of the fibers., Conclusion: NOS positive neurons of the tongue, when noxiously stimulated, are inflicted with pathological changes, which suggest that nitric oxide might play a role in the transmission and control of the message related to pathological lesions.

Abstract

Objective: To investigate the possible cause and molecular genetic mechanism of matrilineal nonsyndromic sensorineural deafness, the authors analyzed an extensive matrilineal nonsyndromic sensorineural deafness family., Methods: PCR amplification of the nt1555 and nt7445 of the mitochondrial DNA, combined with PCR-SSCP, PCR-RFLP and sequence to analyze the family., Results: The authors found a homoplasmic A to G transition at position 1555(A1555G) of the mitochondrial 12SrRNA gene from all the patients, and four matrilineal relatives of this family, but the mutation was not found in the normal spouses of the family and controls (100 normal persons)., Conclusion: The A1555G mutation may be one of the major factors that cause deafness in this family.

Abstract

Objective: To investigate the mechanism of Astragalus Angelica Mixture (AAM) effect of regulating lipid metabolism disturbance in nephrotic rats., Methods: To examine the effects of AAM on serum albumin, lipid levels, and activities of lipoprotein lipase (LPL) and lecithin-cholesterol acyltransferase (LCAT), which are key enzymes for catabolism of lipid, in immune-induced nephrotic hyperlipidemia rats and exogenous hyperlipidemia rats., Results: Serum albumin was reduced, lipid increased significantly, LPL activity decreased significantly and the LCAT activity was relatively insufficient in nephrotic rats. Activities of LPL and LCAT increased significantly in AAM treated nephrotic rats. There were no change of activities of LPL and LCAT in exogenous hyperlipidemia rats and AAM showed no effect on the activities of these two enzymes., Conclusion: The effect of AAM in regulating lipid metabolism might be due to enhancing the clearance of triglyceride and cholesterol-rich lipoprotein by improving the activities of LPL and LCAT.

Abstract

The complete nucleotide sequence of prune dwarf ilarvirus (PDV) RNA 3 has been determined from cloned viral cDNAs. The PDV RNA 3 is 2129 nucleotides and contains two large open reading frames (ORFs) separated by an intergenic region of 72 nucleotides. The 5' proximal ORF (ORF-1) is 882 nucleotides, encoding a gene product which shares homology with putative cell-to-cell movement proteins of related viruses, including tobacco streak virus (TSV) and alfalfa mosaic virus (AIMV). The downstream ORF (ORF-2) is 657 nucleotides and encodes a gene product which shares primary sequence homology and structural features with AIMV coat protein. Furthermore, when expressed in bacteria, this ORF produces a polypeptide which comigrates with authentic PDV coat protein and reacts with PDV coat protein antiserum. Hybridization data suggest that the genomic organization of PDV RNAs 3 and 4 is similar to that of TSV, the only other ilarvirus for which sequence information is published. The 3' untranslated region (UTR) of PDV RNA 3 is similar to that of TSV and AIMV, containing a potential stem-loop structure followed by the sequence AUGC, a structure which may signal binding of coat protein and activation of genome replication. However, a striking feature of the deduced PDV coat protein sequence is the absence of a "zinc-finger" motif thought to function in binding of the coat protein to the 3'-UTR in ilarviruses and AIMV. This result suggests that the zinc-finger motif is not a required aspect of coat protein activation of replication in ilarviruses.
