Summary In this report, we describe the identification and sequencing of a novel HLA-DPB1 allele, found in an Italian haematological patient. This allele is identical to DPB1*17:01 except for a single nucleotide substitution (GACGAG) at position 57, which changes the encoded amino acid from Asp to Glu.

Summary We describe a novel HLA-B*51 allele detected by DNA direct sequencing. The sequence of this allele has been officially named B*51:78 as a confirmatory sequence. This new allele nucleotide sequence differs from HLA-B*51:01:01 for two point mutations in exon 2 where codons 79–80 change from CGG-ATC to CGC-ACC (p.Ile80Thr).

Summary Here, we describe the characterisation of a new allelic variant of HLA-B*57. The novel allele, HLA-B*5728N, was identified with sequence-based typing in a Caucasoid family. HLA-B*5728N, differs from HLA-B*5701 because of a nucleotide substitution at position 420 (C->G) resulting in a coding change from Tyrosine to a stop codon.

This paper reports the investigation results of back layered media in respect to SPT head application, taking into account both the thickness of each layer and the magnetic characteristics of the Co-Cr layer. The same media have been tested using ring heads looking for a medium compatible with both the heads.

The present paper reports experimental results obtained using neutral or polar terminated PERFLUORO-POLYETHER fluid lubricants (FOMBLIN(R)), applied by dip coating technique starting from dilute 1,1,2-tri-chlorotrifluoroethane (Delifrene)solutions. For this purpose Co-Cr floppy disks were RF sputtered in Olivetti laboratories on PET and polyimide substrates, and cut in 5.25" standard format. The lubricant layer was studied in terms of both the mechanical and the magnetic head/disk interactions. The lubricant class under investigation made it possible to achieve promising results with regard to wear life and friction reduction, with a sufficiently thin lubricant layer.

BRAF mutations define a sub-population characterized by a poor prognosis and dismal median survival. Following successful outcomes with BRAF inhibition in BRAF mutant metastatic melanoma, this approach was evaluated in metastatic colorectal cancer (mCRC). The development and combination of targeted therapies against multiple signaling pathways has proved particularly successful, with improved survival and response rates. Areas covered: This review addresses the development of therapeutic strategies with inhibitors targeting MAPK/ERK and EGFR signaling in BRAF V600E mutated mCRC, focusing on encorafenib, binimetinib and cetuximab. A pharmacological and clinical review of these drugs and the therapeutic approaches behind their optimization are presented. Expert opinion: Exploiting knowledge of the mechanisms of resistance to BRAF inhibitors has been crucial to developing effective therapeutic strategies in BRAF-V600E mutant mCRC. The BEACON trial is a successful example of this approach, using encorafenib and cetuximab with or without binimetinib in patients with previously treated BRAF V600E mutant mCRC, showing an impressive improvement in clinical outcomes and tolerable toxicity compared with chemotherapy, establishing a new standard of care in this setting.
