Abstract

Theoretical models of β-delayed neutron emission are used as crucial inputs in r-process calculations. Benchmarking the predictions of these models is a challenge due to a lack of currently available experimental data. In this work the β-delayed neutron emission probabilities of 33 nuclides in the important mass regions south and south-west of 132Sn are presented, 16 for the first time. The measurements were performed at RIKEN using the Advanced Implantation Detector Array (AIDA) and the BRIKEN neutron detector array. The P1n values presented constrain the predictions of theoretical models in the region, affecting the final abundance distribution of the second r-process peak at A≈130.

Abstract

We introduce the first catchment dataset for large sample studies in Chile. This dataset includes 516 catchments; it covers particularly wide latitude (17.8 to 55.0° S) and elevation (0 to 6993 m a.s.l.) ranges, and it relies on multiple data sources (including ground data, remote-sensed products and reanalyses) to characterise the hydroclimatic conditions and landscape of a region where in situ measurements are scarce. For each catchment, the dataset provides boundaries, daily streamflow records and basin-averaged daily time series of precipitation (from one national and three global datasets), maximum, minimum and mean temperatures, potential evapotranspiration (PET; from two datasets), and snow water equivalent. We calculated hydro-climatological indices using these time series, and leveraged diverse data sources to extract topographic, geological and land cover features. Relying on publicly available reservoirs and water rights data for the country, we estimated the degree of anthropic intervention within the catchments. To facilitate the use of this dataset and promote common standards in large sample studies, we computed most catchment attributes introduced by Addor et al. (2017) in their Catchment Attributes and MEteorology for Large-sample Studies (CAMELS) dataset, and added several others.We used the dataset presented here (named CAMELS-CL) to characterise regional variations in hydroclimatic conditions over Chile and to explore how basin behaviour is influenced by catchment attributes and water extractions. Further, CAMELS-CL enabled us to analyse biases and uncertainties in basin-wide precipitation and PET. The characterisation of catchment water balances revealed large discrepancies between precipitation products in arid regions and a systematic precipitation underestimation in headwater mountain catchments (high elevations and steep slopes) over humid regions. We evaluated PET products based on ground data and found a fairly good performance of both products in humid regions (r > 0.91) and lower correlation (r CAMELS-CL provides unprecedented information on catchments in a region largely underrepresented in large sample studies. This effort is part of an international initiative to create multi-national large sample datasets freely available for the community. 8. Innate immune cell and severe acute respiratory syndrome coronavirus 2 interaction

Abstract

Coronavirus disease caused by the recently emerged severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) represents a major public health that has submerged the world into a crisis unprecedented in the modern era. A better understanding of the innate immune response could help to fight this pandemic and be better prepared for potential future outbreaks. Interestingly, innate immune cells can develop a non-specific memory termed trained immunity. This review details recent evidence concerning the interaction of SARS-CoV-2 with innate immune cells, in particular those in which the trained immunity activity has been demonstrated.

Abstract

This study analyzed the chemical composition of Cymbopogon citratus essential oil from Puebla, México, assessed its antioxidant activity, and evaluated in silico protein–compound interactions related to central nervous system (CNS) physiology. GC–MS analysis identified myrcene (8.76%), Z-geranial (27.58%), and E-geranial (38.62%) as the main components, with 45 other compounds present, which depends on the region and growing conditions. DPPH and Folin–Ciocalteu assays using the leaves extract show a promising antioxidant effect (EC50 = 48.5 µL EO/mL), reducing reactive oxygen species. The bioinformatic tool SwissTargetPrediction (STP) shows 10 proteins as potential targets associated with CNS physiology. Moreover, protein–protein interaction diagrams suggest that muscarinic and dopamine receptors are related to each other through a third party. Molecular docking reveals that Z-geranial has higher binding energy than M1 commercial blocker and blocks M2, but not M4 muscarinic acetylcholine receptors, whereas β-pinene and myrcene block M1, M2, and M4 receptors. These actions may positively affect cardiovascular activity, memory, Alzheimer’s disease, and schizophrenia. This study highlights the significance of understanding natural product interactions with physiological systems to uncover potential therapeutic agents and advanced knowledge on their benefits for human health.

Abstract

Purpose: To develop a knowledge-based voxel-wise dose prediction system using a convolution neural network for high-dose-rate brachytherapy cervical cancer treatments with a tandem-and-ovoid (T&O) applicator. Methods: A 3D U-NET was utilized to output dose predictions using organ-at-risk (OAR), high-risk clinical target volume (HRCTV), and possible source locations. A sample of previous T&O treatments comprising 397 cases (273 training:62 validation:62 test), HRCTV and OARs (bladder/rectum/sigmoid) was used. Structures and dose were interpolated to 1x1x2.5mm3 dose planes with two input channels (source positions, voxel identification) and one output channel for dose. We evaluated dose difference (\Delta D)(xyz)=D_(actual)(x,y,z)-D_(predicted)(x,y,z) and dice similarity coefficients in all cohorts across the clinically-relevant dose range (20-130% of prescription), mean and standard deviation. We also examined discrete DVH metrics used for T&O plan quality assessment: HRCTV D_90%(dose to hottest 90% volume) and OAR D_2cc, with \Delta D_x=D_(x,actual)-D_(x,predicted). Pearson correlation coefficient, standard deviation, and mean quantified model performance on the clinical metrics. Results: Voxel-wise dose difference accuracy for 20-130% dose range for training(test) ranges for mean (\Delta D) and standard deviation for all voxels was [-0.3%+/-2.0% to +1.0%+/-12.0%] ([-0.1%+/-4% to +4.0%+/-26%]). Voxel-wise dice similarity coefficients for 20-130% dose ranged from [0.96, 0.91]([0.94, 0.87]). DVH metric prediction in the training (test) set were HRCTV(\Delta D_90)=-0.19+/-0.55 Gy (-0.09+/-0.67 Gy), bladder(\Delta D_2cc)=-0.06+/-0.54 Gy (-0.17+/-0.67 Gy), rectum(\Delta D)_2cc=-0.03+/-0.36 Gy (-0.04+/-0.46 Gy), and sigmoid(\Delta D_2cc)=-0.01+/-0.34 Gy (0.00+/-0.44 Gy). Conclusion: 3D knowledge-based dose predictions for T&O brachytherapy provide accurate voxel-level and DVH metric estimates.

Abstract

Dentists could fail to notice periapical lesions (PLs) while examining panoramic radiographs. Accordingly, this study aimed to develop an artificial intelligence (AI) designed to address this problem. Materials and methods: a total of 18618 periapical root areas (PRA) on 713 panoramic radiographs were annotated and classified as having or not having PLs. An AI model consisting of two convolutional neural networks (CNNs), a detector and a classifier, was trained on the images. The detector localized PRAs using a bounding-box-based object detection model, while the classifier classified the extracted PRAs as PL or not-PL using a fine-tuned CNN. The classifier was trained and validated on a balanced subset of the original dataset that included 3249 PRAs, and tested on 707 PRAs. Results: the detector achieved an average precision of 74.95%, while the classifier accuracy, sensitivity and specificity were 84%, 81% and 86%, respectively. When integrating both detection and classification models, the proposed method accuracy, sensitivity, and specificity were 84.6%, 72.2%, and 85.6%, respectively. Conclusion: a two-stage CNN model consisting of a detector and a classifier can successfully detect periapical lesions on panoramic radiographs.

Abstract

Knowledge graphs have gained increasing popularity in the last decade in science and technology. However, knowledge graphs are currently relatively simple to moderate semantic structures that are mainly a collection of factual statements. Question answering (QA) benchmarks and systems were so far mainly geared towards encyclopedic knowledge graphs such as DBpedia and Wikidata. We present SciQA a scientific QA benchmark for scholarly knowledge. The benchmark leverages the Open Research Knowledge Graph (ORKG) which includes almost 170,000 resources describing research contributions of almost 15,000 scholarly articles from 709 research fields. Following a bottom-up methodology, we first manually developed a set of 100 complex questions that can be answered using this knowledge graph. Furthermore, we devised eight question templates with which we automatically generated further 2465 questions, that can also be answered with the ORKG. The questions cover a range of research fields and question types and are translated into corresponding SPARQL queries over the ORKG. Based on two preliminary evaluations, we show that the resulting SciQA benchmark represents a challenging task for next-generation QA systems. This task is part of the open competitions at the 22nd International Semantic Web Conference 2023 as the Scholarly Question Answering over Linked Data (QALD) Challenge.

Abstract

Question Answering (QA) is a field of study addressed to develop automatic methods for answering questions expressed in natural language. Recently, the emergence of the new generation of intelligent assistants, such as Siri, Alexa, and Google Assistant, has intensified the importance of an effective and efficient QA system able to handle questions with different complexities. Regarding the type of question to be answered, QA systems have been divided into two sub-areas: (i) factoid questions that require a single fact – e.g., a name of a person or a date, and (ii) non-factoid questions that need a more complex answer – e.g., descriptions, opinions, or explanations. While factoid QA systems have overcome human performance on some benchmarks, automatic systems for answering non-factoid questions remain a challenge and an open research problem. This work provides an overview of recent research addressing non-factoid questions. It focuses on which methods have been applied in each task, the data sets available, challenges and limitations, and possible research directions. From a total of 455 recent studies, we selected 75 papers based on our quality control system and exclusion criteria for an in-depth analysis. This systematic review helped to answer what are the tasks and methods involved in non-factoid, what are the data sets available, what the limitations are, and what is the recommendations for future research.

Abstract

There is now strong evidence to support the interest in using lactic acid bacteria (LAB)in particular, strains of lactococci and lactobacilli, as well as bifidobacteria, for the development of new live vectors for human and animal health purposes. LAB are Gram-positive bacteria that have been used for millennia in the production of fermented foods. In addition, numerous studies have shown that genetically modified LAB and bifodobacteria can induce a systemic and mucosal immune response against certain antigens when administered mucosally. They are therefore good candidates for the development of new mucosal delivery strategies and are attractive alternatives to vaccines based on attenuated pathogenic bacteria whose use presents health risks. This article reviews the most recent research and advances in the use of LAB and bifidobacteria as live delivery vectors for human and animal health.

Abstract

The β-delayed one- and two-neutron emission probabilities (P_{1n} and P_{2n}) of 20 neutron-rich nuclei with N≥82 have been measured at the RIBF facility of the RIKEN Nishina Center. P_{1n} of ^{130,131}Ag, ^{133,134}Cd, ^{135,136}In, and ^{138,139}Sn were determined for the first time, and stringent upper limits were placed on P_{2n} for nearly all cases. β-delayed two-neutron emission (β2n) was unambiguously identified in ^{133}Cd and ^{135,136}In, and their P_{2n} were measured. Weak β2n was also detected from ^{137,138}Sn. Our results highlight the effect of the N=82 and Z=50 shell closures on β-delayed neutron emission probability and provide stringent benchmarks for newly developed macroscopic-microscopic and self-consistent global models with the inclusion of a statistical treatment of neutron and γ emission. The impact of our measurements on r-process nucleosynthesis was studied in a neutron star merger scenario. Our P_{1n} and P_{2n} have a direct impact on the odd-even staggering of the final abundance, improving the agreement between calculated and observed Solar System abundances. The odd isotope fraction of Ba in r-process-enhanced (r-II) stars is also better reproduced using our new data.

Abstract

Cellular decision making is the process whereby cells choose one developmental pathway from multiple possible ones, either spontaneously or due to environmental stimuli. Examples in various cell types suggest an almost inexhaustible plethora of underlying molecular mechanisms. In general, cellular decisions rely on the gene regulatory network, which integrates external signals to drive cell fate choice. The search for general principles of such a process benefits from appropriate biological model systems that reveal how and why certain gene regulatory mechanisms drive specific cellular decisions according to ecological context and evolutionary outcomes. In this article, we review the historical and ongoing development of the phage lambda lysis–lysogeny decision as a model system to investigate all aspects of cellular decision making. The unique generality, simplicity, and richness of phage lambda decision making render it a constant source ofmathematical modeling–aided inspiration across all of biology. We discuss the origins and progress of quantitative phage lambda modeling from the 1950s until today, as well as its possible future directions. We provide examples of how modeling enabled methods and theory development, leading to new biological insights by revealing gaps in the theory and pinpointing areas requiring further experimental investigation. Overall, we highlight the utility of theoretical approaches both as predictive tools, to forecast the outcome of novel experiments, and as explanatory tools, to elucidate the natural processes underlying experimental data.

19. Automated treatment planning framework for brachytherapy of cervical cancer using 3D dose predictions

Abstract

Objective. To lay the foundation for automated knowledge-based brachytherapy treatment planning using 3D dose estimations, we describe an optimization framework to convert brachytherapy dose distributions directly into dwell times (DTs). Approach. A dose rate kernel d ̇ ( r , θ , φ ) was produced by exporting 3D dose for one dwell position from the treatment planning system and normalizing by DT. By translating and rotating this kernel to each dwell position, scaling by DT and summing over all dwell positions, dose was computed (D calc). We used a Python-coded COBYLA optimizer to iteratively determine the DTs that minimize the mean squared error between D calc and reference dose D ref, computed using voxels with D ref 80%–120% of prescription. As validation of the optimization, we showed that the optimizer replicates clinical plans when D ref = clinical dose in 40 patients treated with tandem-and-ovoid (T&O) or tandem-and-ring (T&R) and 0–3 needles. Then we demonstrated automated planning in 10 T&O using D ref = dose predicted from a convolutional neural network developed in past work. Validation and automated plans were compared to clinical plans using mean absolute differences ( MAD = 1 N ∑ n = 1 N abs x n − x n ′ ) over all voxels (xn = Dose, N = #voxels) and DTs (xn = DT, N = #dwell positions), mean differences (MD) in organ D 2cc and high-risk CTV D90 over all patients (where positive indicates higher clinical dose), and mean Dice similarity coefficients (DSC) for 100% isodose contours. Main results. Validation plans agreed well with clinical plans (MADdose = 1.1%, MADDT = 4 s or 0.8% of total plan time, D 2cc MD = −0.2% to 0.2% and D90 MD = −0.6%, DSC = 0.99). For automated plans, MADdose = 6.5% and MADDT = 10.3 s (2.1%). The slightly higher clinical metrics in automated plans (D 2cc MD = −3.8% to 1.3% and D90 MD = −5.1%) were due to higher neural network dose predictions. The overall shape of the automated dose distributions were similar to clinical doses (DSC = 0.91). Significance. Automated planning with 3D dose predictions could provide significant time savings and standardize treatment planning across practitioners, regardless of experience.

The objective of this study was to describe the runs of homozygosity (ROH) detected in the Mexican Holstein population and to associate them with milk, fat and protein yields, and conformation final score. After imputation and genomic quality control, 4,227 genotyped animals with 100,806 SNPs markers each were used. ROH with a minimum length of 1 Mb and a minimum of 10 SNPs were included in the analysis. One heterozygous SNP marker and five missing genotypes per ROH were allowed. A total of 425,098 ROH were found in the studied population (71.83 ± 10.73 ROH per animal), with an average length and coverage of 4.80 ± 0.77 Mb, and 276.89 Mb, respectively. The average chromosome length covered by ROH was 10.40 ± 3.70 Mb. ROH between 1 and 2 Mb were the most frequent in the population (51.33%) while those between 14 and 16 Mb were the least frequent (1.20%). Long chromosomes showed a larger number of ROH. Chromosomes 10 and 20, had a greater percentage of their length covered by ROH because they presented a largest number of long ROH (>8 Mb). From the total ROH, 17 were detected in 1,847 animals and distributed among different chromosomes, and were associated with milk, fat and protein yield and percentage, and conformation final score. Of the ROH with effects on production traits, the majority were found with a length between 1 and 4 Mb. These results show evidence of genomic regions preserved by genetic selection and associated with the improvement of the productivity and functionality of dairy cattle.

Abstract

This paper describes an experimental characterization scheme of the biophysical properties of reconstituted hydrogel matrices based on indentation testing, quantification of transport via microfluidics, and confocal reflectance microscopy analysis. While methods for characterizing hydrogels exist and are widely used, they often do not measure diffusive and convective transport concurrently, determine the relationship between microstructure and transport properties, and decouple matrix mechanics and transport properties. Our integrated approach enabled independent and quantitative measurements of the structural, mechanical, and transport properties of hydrogels in a single study. We used fibrillar type I collagen as the base matrix and investigated the effects of two different matrix modifications: (1) cross-linking with human recombinant tissue transglutaminase II (hrTGII) and (2) supplementation with the nonfibrillar matrix constituent hyaluronic acid (HA). hrTGII modified the matrix structure and transport but not mechanical parameters. Furthermore, changes in the matrix structure due to hrTGII were seen to be dependent on the concentration of collagen. In contrast, supplementation of HA at different collagen concentrations altered the matrix microstructure and mechanical indentation behavior but not transport parameters. These experimental observations reveal the important relationship between extracellular matrix (ECM) composition and biophysical properties. The integrated techniques are versatile, robust, and accessible; and as matrix—cell interactions are instrumental for many biological processes, the methods and findings described here should be broadly applicable for characterizing hydrogel materials used for three-dimensional (3-D) tissue-engineered culture models.

Abstract

At the RIKEN Nishina Center, exotic neutron-rich isotopes of Ba, La, Ce, Pr, and Nd were measured. This work reports their half-lives (T1/2) and β-delayed neutron-emission probabilities (Pxn). The setup consisted of the BigRIPS in-flight separator for particle identification, the Advanced Implantation Detector Array (AIDA) for ions and β detection, and the BRIKEN neutron counter for neutron detection. Using this arrangement, 4 new T1/2 and 14 new P1n were measured. Furthermore, 38 T1/2 and 2 P1n values were remeasured, decreasing the uncertainties for some of them. In addition to improving predictions of nucleosynthesis models that describe the production of the rare-earth peak at A ∼ 160 via the r-process, these additional experimental data should help to constrain theoretical models for calculations of T1/2 and Pxn in this region.

Abstract

Background With the rapid adoption of electronic medical records (EMRs), there is an ever-increasing opportunity to collect data and extract knowledge from EMRs to support patient-centered stroke management. Objective This study aims to compare the effectiveness of state-of-the-art automatic text classification methods in classifying data to support the prediction of clinical patient outcomes and the extraction of patient characteristics from EMRs. Methods Our study addressed the computational problems of information extraction and automatic text classification. We identified essential tasks to be considered in an ischemic stroke value-based program. The 30 selected tasks were classified (manually labeled by specialists) according to the following value agenda: tier 1 (achieved health care status), tier 2 (recovery process), care related (clinical management and risk scores), and baseline characteristics. The analyzed data set was retrospectively extracted from the EMRs of patients with stroke from a private Brazilian hospital between 2018 and 2019. A total of 44,206 sentences from free-text medical records in Portuguese were used to train and develop 10 supervised computational machine learning methods, including state-of-the-art neural and nonneural methods, along with ontological rules. As an experimental protocol, we used a 5-fold cross-validation procedure repeated 6 times, along with subject-wise sampling. A heatmap was used to display comparative result analyses according to the best algorithmic effectiveness (F1 score), supported by statistical significance tests. A feature importance analysis was conducted to provide insights into the results. Results The top-performing models were support vector machines trained with lexical and semantic textual features, showing the importance of dealing with noise in EMR textual representations. The support vector machine models produced statistically superior results in 71% (17/24) of tasks, with an F1 score >80% regarding care-related tasks (patient treatment location, fall risk, thrombolytic therapy, and pressure ulcer risk), the process of recovery (ability to feed orally or ambulate and communicate), health care status achieved (mortality), and baseline characteristics (diabetes, obesity, dyslipidemia, and smoking status). Neural methods were largely outperformed by more traditional nonneural methods, given the characteristics of the data set. Ontological rules were also effective in tasks such as baseline characteristics (alcoholism, atrial fibrillation, and coronary artery disease) and the Rankin scale. The complementarity in effectiveness among models suggests that a combination of models could enhance the results and cover more tasks in the future. Conclusions Advances in information technology capacity are essential for scalability and agility in measuring health status outcomes. This study allowed us to measure effectiveness and identify opportunities for automating the classification of outcomes of specific tasks related to clinical conditions of stroke victims, and thus ultimately assess the possibility of proactively using these machine learning techniques in real-world situations.

Abstract

The β-delayed neutron-emission probabilities of 28 exotic neutron-rich isotopes of Pm, Sm, Eu, and Gd were measured for the first time at RIKEN Nishina Center using the Advanced Implantation Detector Array (AIDA) and the BRIKEN neutron detector array. The existing β-decay half-life (T 1/2) database was significantly increased toward more neutron-rich isotopes, and uncertainties for previously measured values were decreased. The new data not only constrain the theoretical predictions of half-lives and β-delayed neutron-emission probabilities, but also allow for probing the mechanisms of formation of the high-mass wing of the rare-earth peak located at A ≈ 160 in the r-process abundance distribution through astrophysical reaction network calculations. An uncertainty quantification of the calculated abundance patterns with the new data shows a reduction of the uncertainty in the rare-earth peak region. The newly introduced variance-based sensitivity analysis method offers valuable insight into the influence of important nuclear physics inputs on the calculated abundance patterns. The analysis has identified the half-lives of 168Sm and of several gadolinium isotopes as some of the key variables among the current experimental data to understand the remaining abundance uncertainty at A = 167–172.

Abstract

International audience; The intestinal epithelium serves as an effective barrier against the external environment, hampering the passage of potentially harmful substances (such as pathogenic microbes) that could trigger an exacerbated host immune response. The integrity of this barrier is thus essential for the maintenance of proper intestinal homeostasis and efficient protective reactions against chemical and microbial challenges. The principal consequence of intestinal barrier defects is an increase in intestinal permeability, which leads to an increased influx of luminal stressors, such as pathogens, toxins, and allergens, which in turn trigger inflammation and immune response. The fine and fragile balance of intestinal homeostasis can be altered by multiple factors that regulate barrier function, many of which are poorly understood. This review will address the role of gut microbiota as well as food supplements (such as probiotics, prebiotics, and synbiotics) in modulating gut health and regulating intestinal barrier function. In particular, we will focus on three human pathologies: inflammatory bowel disease, irritable bowel syndrome, and food allergy.

Abstract

The purpose of this in-vitro study was to compare trueness, 3D deviation, production time and costs of milled and 3D-printed resin single crowns. A total of 20 CAD-CAM resin single crowns were fabricated from 10 digital wax patterns designed on 10 tooth preparations available in a reference model. Standardized control linear measurements were performed with a CAD software. Each STL file was then used to fabricate two resins crowns - one milled and one 3D-printed. All crowns underwent physical linear measurements using a digital caliper. The crowns were then scanned using an intraoral scanner for assessing 3D deviation. Finally, time to produce a single crown, as well as costs and production rates of both methods were also compared. Both CAM methods did not present statistically significant differences in linear measurements, as compared to controls (P⟩.05). Furthermore, 3D-printed crowns had significantly greater deviations in cervical margins (P=.032) and occlusal surfaces (P=.041), as compared with milled crowns. Finally, 3D-printing took significantly longer to produce one single crown (P=.001), but with a cheaper and higher production rate than milling. These findings suggest that milling devices produce resin single crowns with smaller 3D deviations but more expensive costs, as compared with low-cost 3D printers.

Abstract

The spreading of disinformation throughout the web has become a critical problem for a democratic society. The dissemination of fake news has become a profitable business and a common practice among politicians and content producers. On the other hand, journalists and fact-checkers work unceasingly to debunk misinformation and prevent it from further spreading. In 2015, a new web markup called ClaimReview has been introduced to grant access to the fact-checking article’s meaning by search engines. It is an important initiative to fight fake news by promoting and highlighting fact-check articles among users. However, barely half of fact-checkers have adopted the ClaimReview markup so far, resulting in low findability of fact-check articles, especially in under-represented countries and languages. In this work, we investigate the viability of using Artificial Intelligence for generating ClaimReview automatically. Besides promoting fact-check articles, the automatic generating of ClaimReview is an important step towards the creation of updated multilingual knowledge base for fighting disinformation. Our experiments show noticeable results, which indicate a viable solution in a production environment. Furthermore, this work has created a benchmark that can be used in upcoming investigations in this domain.

Abstract

International audience; Severe acute respiratory syndrome coronavirus 2 virus (SARS-CoV-2) infection, the causative agent of COVID-19, now represents the sixth Public Health Emergency of International Concern (PHEIC)—as declared by the World Health Organization (WHO) since 2009. Considering that SARS-CoV-2 is mainly transmitted via the mucosal route, a therapy administered by this same route may represent a desirable approach to fight SARS-CoV-2 infection. It is now widely accepted that genetically modified microorganisms, including probiotics, represent attractive vehicles for oral or nasal mucosal delivery of therapeutic molecules. Previous studies have shown that the mucosal administration of therapeutic molecules is able to induce an immune response mediated by specific serum IgG and mucosal IgA antibodies along with mucosal cell-mediated immune responses, which effectively concur to neutralize and eradicate infections. Therefore, advances in the modulation of mucosal immune responses, and in particular the use of probiotics as live delivery vectors, may encourage prospective studies to assess the effectiveness of genetically modified probiotics for SARS-CoV-2 infection. Emerging trends in the ever-progressing field of vaccine development re-emphasize the contribution of adjuvants, along with optimization of codon usage (when designing a synthetic gene), expression level, and inoculation dose to elicit specific and potent protective immune responses. In this review, we will highlight the existing pre-clinical and clinical information on the use of genetically modified microorganisms in control strategies against respiratory and non-respiratory viruses. In addition, we will discuss some controversial aspects of the use of genetically modified probiotics in modulating the cross-talk between mucosal delivery of therapeutics and immune system modulation.

Abstract

The Ge72(n,γ) cross section was measured for neutron energies up to 300keV at the neutron time-of-flight facility n_TOF (CERN), Geneva, for the first time covering energies relevant to heavy-element synthesis in stars. The measurement was performed at the high-resolution beamline EAR-1, using an isotopically enriched GeO272 sample. The prompt capture γ rays were detected with four liquid scintillation detectors, optimized for low neutron sensitivity. We determined resonance capture kernels up to a neutron energy of 43keV, and averaged cross sections from 43 to 300keV. Maxwellian-averaged cross section values were calculated from kT=5 to 100keV, with uncertainties between 3.2% and 7.1%. The new results significantly reduce uncertainties of abundances produced in the slow neutron capture process in massive stars.

Abstract

We propose a general formalism to characterize orientational frustration of smectic liquid crystals in confinement by interpreting the emerging networks of grain boundaries as objects with a topological charge. In a formal idealization, this charge is distributed in point-like units of quarter-integer magnitude, which we identify with tetratic disclinations located at the end points and nodes. This coexisting nematic and tetratic order is analyzed with the help of extensive Monte Carlo simulations for a broad range of two-dimensional confining geometries as well as colloidal experiments, showing how the observed defect networks can be universally reconstructed from simple building blocks. We further find that the curvature of the confining wall determines the anchoring behavior of grain boundaries, such that the number of nodes in the emerging networks and the location of their end points can be tuned by changing the number and smoothness of corners, respectively., To appear in Physical Review Letters

Abstract

Babiano-Suárez, V., et al., i-TED is an innovative detection system which exploits Compton imaging techniques to achieve a superior signal-to-background ratio in (n, γ) cross-section measurements using time-of-flight technique. This work presents the first experimental validation of the i-TED apparatus for high-resolution time-of-flight experiments and demonstrates for the first time the concept proposed for background rejection. To this aim, the Au(n, γ) and Fe(n, γ) reactions were studied at CERN n_TOF using an i-TED demonstrator based on three position-sensitive detectors. Two CD detectors were also used to benchmark the performance of i-TED. The i-TED prototype built for this study shows a factor of ∼ 3 higher detection sensitivity than state-of-the-art CD detectors in the 10 keV neutron-energy region of astrophysical interest. This paper explores also the perspectives of further enhancement in performance attainable with the final i-TED array consisting of twenty position-sensitive detectors and new analysis methodologies based on Machine-Learning techniques., This work has been carried out in the framework of a project funded by the European Research Council (ERC) under the European Union’s Horizon 2020 research and innovation programme (ERC Consolidator Grant project HYMNS, with grant agreement No. 681740). 38. Pembrolizumab plus eribulin in hormone-receptor-positive, HER2-negative, locally recurrent or metastatic breast cancer (KELLY): An open-label, multicentre, single-arm, phase ? trial

Abstract

BACKGROUND: Pembrolizumab has modest activity if used in patients with hormone-receptor-positive (HR+), HER2-negative, previously treated metastatic breast cancer (BC). Our study investigated whether there would be any clinical benefit in combining chemotherapy with pembrolizumab in a similar patient population. METHODS: This single-arm, phase ? trial enrolled women aged =18 years with HR+, HER2-negative, inoperable, locally recurrent or metastatic BC. Patients were previously treated with hormonal therapy and 1-2 chemotherapy regimens for locally recurrent and/or metastatic BC. On each 21-day cycle, patients received intravenous pembrolizumab 200mg on day 1 and eribulin 1·23mg/m2 on days 1 and 8. The primary endpoint was the clinical benefit rate. Analysis of safety and activity was carried out in all patients who met the screening criteria and received at least 1 dose of study treatment. The trial is registered at ClinicalTrials.gov, NCT03222856. RESULTS: Of the 44 patients enrolled between January 29 and October 17, 2018, clinical benefit was achieved in 25 (56·8%, 95% confidence interval [CI]: 41·0-71·7), objective response in 18 (40·9%, 95% CI: 26·3-56·8), median progression-free survival was 6·0 months (95% CI: 3·7-8·4), and 1-year overall survival was 59·1% (95% CI: 45·8-76·2). The most common treatment-emergent adverse events (AEs) of any grade were neutropenia (20 [45·5%]), anaemia (17 [38·6%]), alopecia (19 [43·2%]), asthenia (19 [43·2%]), diarrhoea (14 [31·8%]), fatigue (14 [31·8%]), and peripheral neuropathy (12 [27·3%]). Serious AEs occurred in 14 (31·8%) patients including febrile neutropenia (3 [6·8%]), neutropenia (2 [4·5%]), fever (2 [4·5%])and peripheral neuropathy (2 [4·5%]). Immune-related AEs occurred in 11 (25·0%) patients. One (2·3%) patient died of cardiac arrest unrelated to study treatment. CONCLUSION: Pembrolizumab plus eribulin demonstrates encouraging antitumour activity in patients with heavily pre-treated, HR+, HER2-negative, locally recurrent or metastatic BC. The safety and tolerability of the combination is similar to eribulin or pembrolizumab monotherapy.

Experiments were performed at the n_TOF facility at CERN via the time-of-flight technique, over a wide neutron energy range, for all stable germanium isotopes (70,72,73,74, and 76). Results for Ge-70 [Phys. Rev. C 100, 045804 (2019)] and Ge-73 [Phys. Lett. B 790, 458 (2019)] are already published. In the field of nuclear structure, such measurements allow to study excited levels close to the neutron binding energy and to obtain information on nuclear properties. Abstract

We measured the neutron capture-to-fission cross-section ratio (α ratio) and the capture cross section of $^{235}$U between 0.2 and 200 eV at the n_TOF facility at CERN. The simultaneous measurement of neutron-induced capture and fission rates was performed by means of the n_TOF BaF$_{2}$ Total Absorption Calorimeter (TAC), used for detection of γ rays, in combination with a set of micromegas detectors used as fission tagging detectors. The energy dependence of the capture cross section was obtained with help of the $^{6}$Li(n,t) standard reaction determining the n_TOF neutron fluence; the well-known integral of the $^{235}$U(n,f) cross section between 7.8 and 11 eV was then used for its absolute normalization. The α ratio, obtained with slightly higher statistical fluctuations, was determined directly, without need for any reference cross section. To perform the analysis of this measurement we developed a new methodology to correct the experimentally observed effect that the probabilities of detecting a fission reaction in the TAC and the micromegas detectors are not independent. The results of this work have been used in a new evaluation of $^{235}$U performed within the scope of the Collaborative International Evaluated Library Organisation (CIELO) Project, and are consistent with the ENDF/B-VIII.0 and JEFF-3.3 capture cross sections below 4 eV and above 100 eV. However, the measured capture cross section is on average 10% larger between 4 and 100 eV.

Abstract

The neutron capture cross sections of several unstable nuclides acting as branching points in the s process are crucial for stellar nucleosynthesis studies. The unstable $^{171}$Tm(t$_{1/2}$=1.92 yr) is part of the branching around mass A∼170 but its neutron capture cross section as a function of the neutron energy is not known to date. In this work, following the production for the first time of more than 5 mg of $^{171}$Tm at the high-flux reactor Institut Laue-Langevin in France, a sample was produced at the Paul Scherrer Institute in Switzerland. Two complementary experiments were carried out at the neutron time-of-flight facility (n_TOF) at CERN in Switzerland and at the SARAF liquid lithium target facility at Soreq Nuclear Research Center in Israel by time of flight and activation, respectively. The result of the time-of-flight experiment consists of the first ever set of resonance parameters and the corresponding average resonance parameters, allowing us to make an estimation of the Maxwellian-averaged cross sections (MACS) by extrapolation. The activation measurement provides a direct and more precise measurement of the MACS at 30 keV: 384(40) mb, with which the estimation from the n_TOF data agree at the limit of 1 standard deviation. This value is 2.6 times lower than the JEFF-3.3 and ENDF/B-VIII evaluations, 25% lower than that of the Bao et al. compilation, and 1.6 times larger than the value recommended in the KADoNiS (v1) database, based on the only previous experiment. Our result affects the nucleosynthesis at the A∼170 branching, namely, the $^{171}$Yb abundance increases in the material lost by asymptotic giant branch stars, providing a better match to the available pre-solar SiC grain measurements compared to the calculations based on the current JEFF-3.3 model-based evaluation.

Abstract

This research is implemented through IKY scholarships programme and co-financed by the European Union (European Social Fund - ESF) and Greek national funds through the action entitled Reinforcement of Postdoctoral Researchers, in the framework of the Operational Programme Human Resources Development Program, Education and Lifelong Learning of the National Strategic Reference Framework (NSRF) 2014-2020., Neutron-induced fission cross sections of isotopes involved in the nuclear fuel cycle are vital for the design and safe operation of advanced nuclear systems. Such experimental data can also provide additional constraints for the adjustment of nuclear model parameters used in the evaluation process, resulting in the further development of fission models. In the present work, the Np-237(n,f) cross section was studied at the EAR2 vertical beam-line at CERN's n_TOF facility, over a wide range of neutron energies, from meV to MeV, using the time-of-flight technique and a set-up based on Micromegas detectors, in an attempt to provide accurate experimental data. Preliminary results in the 200 keV - 14 MeV neutron energy range as well as the experimental procedure, including a description of the facility and the data handling and analysis, will be presented., IKY scholarships programme, European Social Fund (ESF) European Commission, Greek national funds through the action entitled Reinforcement of Postdoctoral Researchers

44. Influence of Gingival Contour on Marginal Fit of CAD-CAM Zirconia Copings on Implant Stock Abutments

Abstract

Intraoral scanning subgingival finish lines has been described as challenging. The purpose of this study was to assess the impact of gingival contour around margins of implant stock abutments on marginal fit of Computer-aided Design - Computer-aided Manufacturing (CAD-CAM) zirconia copings. This in-vitro study was conducted on 40 analogues of implant stock abutments that were embedded into individual phantoms composed by a resin block. All 40 phantoms underwent two intraoral scans - one with (test group) and one without artificial gingiva (control group) - using a closed system intraoral scanner. Zirconia copings were then digitally designed and milled, followed by high-speed sintering, before being analyzed for marginal adaptation and internal surface roughness with scanning electron microscopy. Statistically significant differences between groups were assessed with the Mann-Whitney test. Median marginal gap values were 149.78 μm (95% CI: 112.39-216.66) for the test group and 94.90 μm (95% CI: 83.89-107.74) for the control group. A statistically significant difference was found between groups (p=0.0001). However, there were no subjective differences between groups for internal surface roughness. Within the limitations of this study, the present findings suggest that a gingival contour one millimeter higher than the finishing line affects marginal adaptation of CAD-CAM zirconia copings.

Abstract

Background: Nuclear waste management is considered amongst the major challenges in the field of nuclear energy. A possible means of addressing this issue is waste transmutation in advanced nuclear systems, whose operation requires a fast neutron spectrum. In this regard, the accurate knowledge of neutron-induced reaction cross sections of several (minor) actinide isotopes is essential for design optimization and improvement of safety margins of such systems. One such case is Pu240, due to its accumulation in spent nuclear fuel of thermal reactors and its usage in fast reactor fuel. The measurement of the Pu240(n,f) cross section was previously attempted at the CERN n_TOF facility EAR1 measuring station using the time-of-flight technique. Due to the low amount of available material and the given flux at EAR1, the measurement had to last several months to achieve a sufficient statistical accuracy. This long duration led to detector deterioration due to the prolonged exposure to the high α activity of the fission foils, therefore the measurement could not be successfully completed. Purpose: It is aimed to determine whether it is feasible to study neutron-induced fission at n_TOF/EAR2 and provide data on the Pu240(n,f) reaction in energy regions requested for applications. Methods: The study of the Pu240(n,f) reaction was made at a new experimental area (EAR2) with a shorter flight path which delivered on average 30 times higher flux at fast neutron energies. This enabled the measurement to be performed much faster, thus limiting the exposure of the detectors to the intrinsic activity of the fission foils. The experimental setup was based on microbulk Micromegas detectors and the time-of-flight data were analyzed with an optimized pulse-shape analysis algorithm. Special attention was dedicated to the estimation of the non-negligible counting loss corrections with the development of a new methodology, and other corrections were estimated via Monte Carlo simulations of the experimental setup. Results: This new measurement of the Pu240(n,f) cross section yielded data from 9meV up to 6MeV incident neutron energy and fission resonance kernels were extracted up to 10keV. Conclusions: Neutron-induced fission of high activity samples can be successfully studied at the n_TOF/EAR2 facility at CERN covering a wide range of neutron energies, from thermal to a few MeV.

Abstract

The isotope used in this research was supplied by the United States Department of Energy Office of Science by the Isotope Program in the Office of Nuclear Physics., The neutron capture cross section of 154Gd was measured from 1 eV to 300 keV in the experimental area located 185 m from the CERN n_TOF neutron spallation source, using a metallic sample of gadolinium, enriched to 67% in 154Gd. The capture measurement, performed with four C6D6 scintillation detectors, has been complemented by a transmission measurement performed at the GELINA time-of-flight facility (JRC-Geel), thus minimising the uncertainty related to sample composition. An accurate Maxwellian averaged capture cross section (MACS) was deduced over the temperature range of interest for s process nucleosynthesis modelling. We report a value of 880(50) mb for the MACS at kT = 30 keV, significantly lower compared to values available in literature. Abstract

In the measurement of neutron capture cross-sections of fissile isotopes, the fission channel is a source of background which can be removed efficiently using the so-called fission-tagging or fission-veto technique. For this purpose a new compact and fast fission chamber has been developed. The design criteria and technical description of the chamber are given within the context of a measurement of the 233U(n, ������) cross-section at the n_TOF facility at CERN, where it was coupled to the n_TOF Total Absorption Calorimeter. For this measurement the fission detector was optimized for time resolution, minimization of material in the neutron beam and for alpha-fission discrimination. The performance of the fission chamber and its application as a fission tagging detector are discussed., This work was partially supported by the French NEEDS/NACRE Project and by the European Commission within HORIZON2020 via the EURATOM Project EUFRAT.

Abstract

The idea of slow-neutron capture nucleosynthesis formulated in 1957 triggered a tremendous experimental effort in different laboratories worldwide to measure the relevant nuclear physics input quantities, namely (n, γ) cross sections over the stellar temperature range (from few eV up to several hundred keV) for most of the isotopes involved from Fe up to Bi. A brief historical review focused on total energy detectors will be presented to illustrate how advances in instrumentation have led to the assessment of new aspects of s-process nucleosynthesis and to the progressive refinement of stellar models. A summary will be presented on current efforts to develop new detection concepts, such as the Total-Energy Detector with γ-ray imaging capability (i-TED). Abstract

The design and operation of innovative nuclear systems requires a better knowledge of the capture and fission cross sections of the Pu isotopes. For the case of capture on 242Pu, a reduction of the uncertainty in the fast region down to 8-12% is required. Moreover, aiming at improving the evaluation of the fast energy range in terms of average parameters, the OECD NEA High Priority Request List (HPRL) requests high-resolution capture measurements with improved accuracy below 2 keV. The current uncertainties also affect the thermal point, where previous experiments deviate from each other by 20%. A fruitful collaboration betwen JGU Mainz and HZ Dresden-Rossendorf within the EC CHANDA project resulted in a 242Pu sample consisting of a stack of seven fission-like targets making a total of 95(4) mg of 242Pu electrodeposited on thin (11.5 μm) aluminum backings. This contribution presents the results of a set of measurements of the 242Pu(n, γ) cross section from thermal to 500 keV combining different neutron beams and techniques. The thermal point was determined at the Budapest Research Reactor by means of Neutron Activation Analysis and Prompt Gamma Analysis, and the resolved (1 eV - 4 keV) and unresolved (1 - 500 keV) resonance regions were measured using a set of four Total Energy detectors at the CERN n_TOF-EAR1. Abstract

This research is co-financed by Greece and the European Union (European Social Fund-ESF) through the Operational Programme Human Resources Development, Education and Lifelong Learning in the context of the project "Strengthening Human Resources Research Potential via Doctorate Research" (MIS-5000432), implemented by the State Scholarships Foundation (IKY). Also, the authors would like to acknowledge the support of the European Commission under the CHANDA project (7th Framework Programme)., Feasibility, design and sensitivity studies on innovative nuclear reactors that could address the issue of nuclear waste transmutation using fuels enriched in minor actinides, require high accuracy cross section data for a variety of neutron-induced reactions from thermal energies to several tens of MeV. The isotope Am-241 (T-1/2= 433 years) is present in high-level nuclear waste (HLW), representing about 1.8 % of the actinide mass in spent PWR UOx fuel. Its importance increases with cooling time due to additional production from the beta-decay of Pu-241 with a half-life of 14.3 years. The production rate of 241Am in conventional reactors, including its further accumulation through the decay of Pu-241 and its destruction through transmutation/incineration are very important parameters for the design of any recycling solution. In the present work, the Am-241(n,f) reaction cross-section was measured using Micromegas detectors at the Experimental Area 2 of the n_TOF facility at CERN. For the measurement, the U-235(n,f) and U-238(n,f) reference reactions were used for the determination of the neutron flux.
