Abstract In this study, 18 animals were fed two forage-based diets: red clover (RC) and grass silage (GS), in a crossover-design experiment in which methane (CH4) emissions were recorded in respiration chambers. Rumen samples obtained through naso-gastric sampling tubes were analysed by NMR. Methane yield (g/kg DM) was significantly lower from animals fed RC (17.8 ± 3.17) compared to GS (21.2 ± 4.61) p = 0.008. In total 42 metabolites were identified, 6 showing significant differences between diets (acetate, propionate, butyrate, valerate, 3-phenylopropionate, and 2-hydroxyvalerate). Partial least squares discriminant analysis (PLS-DA) was used to assess which metabolites were more important to distinguish between diets and partial least squares (PLS) regressions were used to assess which metabolites were more strongly associated with the variation in CH4 emissions. Acetate, butyrate and propionate along with dimethylamine were important for the distinction between diets according to the PLS-DA results. PLS regression revealed that diet and dry matter intake are key factors to explain CH4 variation when included in the model. Additionally, PLS was conducted within diet, revealing that the association between metabolites and CH4 emissions can be conditioned by diet. These results provide new insights into the methylotrophic methanogenic pathway, confirming that metabolite profiles change according to diet composition, with consequences for CH4 emissions.

The EMC effect in deuterium and helium-3 is studied using a convolution formalism that allows isolating the impact of high-momentum nucleons in short-ranged correlated (SRC) pairs. We assume that the modification of the structure function of bound nucleons is given by a universal (i.e., nucleus independent) function of their virtuality, and find that the effect of such modifications is dominated by nucleons in SRC pairs. This SRC-dominance of nucleon modifications is observed despite the fact that the bulk of the nuclear inelastic scattering cross-section comes from interacting with low-momentum nucleons. These conclusions are found to be robust to model details including nucleon modification function parametrization, free nucleon structure function, and treatment of nucleon motion effects. While existing data cannot discriminate between such model details, we present predictions for measured, but not yet published, tritium EMC effect and tagged nucleon structure functions in deuterium that are sensitive to the neutron structure functions and bound nucleon modification functions.

Most extant ice caps mantling low-relief Arctic Canada landscapes remained cold based throughout the late Holocene, preserving in situ bryophytes killed as ice expanded across vegetated landscapes. After reaching peak late Holocene dimensions ∼1900 CE, ice caps receded as Arctic summers warmed, exposing entombed vegetation. The calibrated radiocarbon ages of entombed moss collected near ice cap margins (kill dates) define when ice advanced across the site, killing the moss, and remained over the site until the year of their collection. In an earlier study, we reported 94 last millennium radiocarbon dates on in situ dead moss collected at ice cap margins across Baffin Island, Arctic Canada. Tight clustering of those ages indicated an abrupt onset of the Little Ice Age at ∼1240 CE and further expansion at ∼1480 CE coincident with episodes of major explosive volcanism. Here we test the confidence in kill dates as reliable predictors of expanding ice caps by resampling two previously densely sampled ice complexes ∼15 years later after ∼250 m of ice recession. The probability density functions (PDFs) of the more recent series of ages match PDFs of the earlier series but with a larger fraction of early Common Era ages. Post 2005 CE ice recession has exposed relict ice caps that grew during earlier Common Era advances and were preserved beneath later ice cap growth. We compare the 106 kill dates from the two ice complexes with 80 kill dates from 62 other ice caps within 250 km of the two densely sampled ice complexes. The PDFs of kill dates from the 62 other ice caps cluster in the same time windows as those from the two ice complexes alone, with the PDF of all 186 kill dates documenting episodes of widespread ice expansion restricted almost exclusively to 250–450 CE, 850–1000 CE, and a dense early Little Ice Age cluster with peaks at ∼1240 and ∼1480 CE. Ice continued to expand after 1480 CE, reaching maximum dimensions at ∼1880 CE that are still visible as zones of sparse vegetation cover in remotely sensed imagery. Intervals of widespread ice cap expansion coincide with persistent decreases in mean summer surface air temperature for the region in a Community Earth System Model (CESM) fully coupled Common Era simulation, suggesting the primary forcings of the observed snowline lowering were both modest declines in summer insolation and cooling resulting from explosive volcanism, most likely intensified by positive feedbacks from increased snow cover and sea ice and reduced northward heat transport by the oceans. The clusters of ice cap expansion defined by moss kill dates are mirrored in an annually resolved Common Era record of ice cap dimensions in Iceland, suggesting this is a circum-North-Atlantic–Arctic climate signal for the Common Era. During the coldest century of the Common Era, 1780–1880 CE, ice caps mantled >11 000 km2 of north-central Baffin Island, whereas km2 is glaciated at present. The peak Little Ice Age state approached conditions expected during the inception phase of an ice age and was only reversed after 1880 CE by anthropogenic alterations of the planetary energy balance.

Abstract Temperate reefs are at the forefront of warming-induced community alterations resulting from poleward range shifts. This tropicalisation is exemplified and amplified by tropical species’ invasions of temperate herbivory functions. However, whether other temperate ecosystem functions are similarly invaded by tropical species, and by what drivers, remains unclear. We examine tropicalisation footprints in nine reef fish functional groups using trait-based analyses and biomass of 550 fish species across tropical to temperate gradients in Japan and Australia. We discover that functional niches in transitional communities are asynchronously invaded by tropical species, but with congruent invasion schedules for functional groups across the two hemispheres. These differences in functional group tropicalisation point to habitat availability as a key determinant of multi-species range shifts, as in the majority of functional groups tropical and temperate species share functional niche space in suitable habitat. Competition among species from different thermal guilds played little part in limiting tropicalisation, rather available functional space occupied by temperate species indicates that tropical species can invade. Characterising these drivers of reef tropicalisation is pivotal to understanding, predicting, and managing marine community transformation.

Abstract Transcranial focused ultrasound with the InSightec Exablate system uses thermal ablation for the treatment of movement and mood disorders and blood brain barrier disruption for tumor therapy. The system uses computed tomography (CT) images to calculate phase corrections that account for aberrations caused by the human skull. This work investigates whether magnetic resonance (MR) images can be used as an alternative to CT images to calculate phase corrections. Phase corrections were calculated using the gold standard hydrophone method and the standard of care InSightec ray tracing method. MR binary image mask, MR-simulated-CT (MRsimCT), and CT images of three ex vivo human skulls were supplied as inputs to the InSightec ray tracing method. The degassed ex vivo human skulls were sonicated with a 670 kHz hemispherical phased array transducer (InSightec Exablate 4000). 3D raster scans of the beam profiles were acquired using a hydrophone mounted on a 3-axis positioner system. Focal spots were evaluated using six metrics: pressure at the target, peak pressure, intensity at the target, peak intensity, positioning error, and focal spot volume. Targets at the geometric focus and 5 mm lateral to the geometric focus were investigated. There was no statistical difference between any of the metrics at either target using either MRsimCT or CT for phase aberration correction. As opposed to the MRsimCT, the use of CT images for aberration correction requires registration to the treatment day MR images; CT misregistration within a range of ± 2 degrees of rotation error along three dimensions was shown to reduce focal spot intensity by up to 9.4%. MRsimCT images used for phase aberration correction for the skull produce similar results as CT-based correction, while avoiding both CT to MR registration errors and unnecessary patient exposure to ionizing radiation.

Stroke remains a major burden on patients, families, and healthcare professionals, despite major advances in prevention, acute treatment, and rehabilitation. Preclinical basic research can help to better define mechanisms contributing to stroke pathology, and identify therapeutic interventions that can decrease ischemic injury and improve outcomes. Animal models play an essential role in this process, and mouse models are particularly well-suited due to their genetic accessibility and relatively low cost. Here, we review the focal cerebral ischemia models with an emphasis on the middle cerebral artery occlusion technique, a “gold standard” in surgical ischemic stroke models. Also, we highlight several histologic, genetic, and in vivo imaging approaches, including mouse stroke MRI techniques, that have the potential to enhance the rigor of preclinical stroke evaluation. Together, these efforts will pave the way for clinical interventions that can mitigate the negative impact of this devastating disease.

Distributions of branched glycerol dialkyl glycerol tetraethers (brGDGTs) are frequently employed for reconstructing terrestrial paleotemperatures from lake sediment archives. Although brGDGTs are globally ubiquitous, the microbial producers of these membrane lipids remain unknown, precluding a full understanding of the ways in which environmental parameters control their production and distribution. Here, we advance this understanding in three ways. First, we present 43 new high-latitude lake sites characterized by low mean annual air temperatures (MATs) and high seasonality, filling an important gap in the global dataset. Second, we introduce a new approach for analyzing brGDGT data in which compound fractional abundances (FAs) are calculated within structural groups based on methylation number, methylation position, and cyclization number. Finally, we perform linear and nonlinear regressions of the resulting FAs against a suite of environmental parameters in a compiled global lake sediment dataset (n = 182). We find that our approach deconvolves temperature, conductivity, and pH trends in brGDGTs without increasing calibration errors from the standard approach. We also find that it reveals novel patterns in brGDGT distributions and provides a methodology for investigating the biological underpinnings of their structural diversity. Warm-season temperature indices outperformed MAT in our regressions, with the mean temperature of months above freezing yielding the highest-performing model (adjusted R2 = 0.91, RMSE = 1.97 ∘C, n = 182). The natural logarithm of conductivity had the second-strongest relationship to brGDGT distributions (adjusted R2 = 0.83, RMSE = 0.66, n = 143), notably outperforming pH in our dataset (adjusted R2 = 0.73, RMSE = 0.57, n = 154) and providing a potential new proxy for paleohydrology applications. We recommend these calibrations for use in lake sediments globally, including at high latitudes, and detail the advantages and disadvantages of each.

Abstract In semiarid agricultural regions, aquifers have watered widespread economic development. Falling water tables, however, drive up energy costs and can make the water toxic for human consumption. The study area is located in central Mexico, where arsenic and fluoride are widely present at toxic concentrations in well water. We simulated the holistic outcomes from three pumping scenarios over 100 years (2020–2120); (S1) pumping rates increase at a similar rate to the past 40 years, (S2) remain constant, or (S3) decrease. Under scenario S1, by 2120, the depth to water table increased to 426 m and energy consumption for irrigation increased to 4 × 109 kWh/yr. Arsenic and fluoride concentrations increased from 14 to 46 μg/L and 1.0 to 3.6 mg/L, respectively. The combined estimated IQ point decrements from drinking untreated well water lowered expected incomes in 2120 by 27% compared to what they would be with negligible exposure levels. We calculated the 100‐year Net Present Value (NPV) of each scenario assuming the 2020 average crop value to water footprint ratio of 0.12 USD/m3. Without drinking water mitigation, S1 and S3 yielded relative NPVs of −5.96 × 109 and 1.51 × 109 USD, respectively, compared to the base case (S2). The relative NPV of providing blanket reverse osmosis treatment, while keeping pumping constant (S2), was 11.55 × 109 USD and this gain increased when combined with decreased pumping (S3). If a high value, low water footprint crop was substituted (broccoli, 1.51 USD/m3), the net gains from increasing pumping were similar in size to those of implementing blanket drinking water treatment.
