Peculiar coordination compound Cu(Hbpt)(CH3CN)2PF6 (1) derived from 3,5-bis(4-pyridyl)-1H-1,2,4-triazole (Hbpt) is synthesized by the layer diffusion method and structurally characterized by single crystal and powder X-ray diffraction, elemental analysis, and infrared spectroscopy. Compound 1 exhibits a structure formed by cationic [Cu(Hbpt)(CH3CN2]nn+ chains and PF6– anions with abundant supramolecular interactions. Solid-state photoluminescence experiments show that compound 1 exhibits relatively strong red emission with a peak around 656 nm, and the origin of the emission band may be ascribed to the metal-to-ligand charge transfer according to the calculation of the density of states.

The Qinghai-Tibet Plateau ecosystem is sensitive and fragile, the grassland degradation and soil erosion in the plateau are still increasing. In this paper, we conducted a quantitative study to clarify the characteristics of land use/land cover change, the driving factors, and the environmental effects in the plateau. We focused on the northeastern Qinghai-Tibet Plateau as an example, and created land use change matrices and determined the relative change rate, vegetation cover, and environmental quality index using land use data and NDVI3g data from 1980 to 2015. The results show that there were significant differences in land use/land cover change in the study area around 2000, while the farmland did not increase significantly, and the water areas exhibited a decreasing trend initially followed by an increasing trend, the unused land first increased and then decreased, and residential areas increased significantly after 2000. The relative change rate of the grassland was the largest between 1980 and 2015, while most of the grassland was converted into farmland, residential land, and unused land. In addition, grassland degradation increased significantly. Considerable spatial heterogeneity in vegetation and land use/land cover was observed in the study area. The vegetation cover decreased in some areas due to the grassland degradation, however, the others areas showed improvements due to the influences of policies and climate change. The environmental index and ecological service value of the study area declined and the environment deteriorated during the study period.

E3 ubiquitin ligases play essential roles in determining the specificity of ubiquitination and subsequent protein degradation. The plant U-box (PUB) family of E3 ligases has been implicated in biotic and abiotic stress signaling and developmental events in various species. A comprehensive bioinformatics analysis identified 56 PUB genes in the grapevine (Vitis vinifera L.) genome. Based on conserved motifs, the VvPUB family was classified into seven subclasses. Expansion of this family was driven by chromosomal, segmental, and tandem duplications. Microarray expression profiling revealed that three PUB genes were preferentially expressed in pollen, four in leaf, and five in root. Moreover, a large number of PUB genes were differentially expressed under abiotic stresses and eight PUB genes likely participated in defense against powdery mildew. The microarray expression data were verified by RT-qPCR. Genome-wide identification of VvPUB genes and examination of their expression will give insights into the functions of U-box genes in grapevine.

In plants, many C2H2-type zinc finger transcription factors function in plant defense responses to biotic and abiotic stresses. Here, we report cloning and characterization of VvZFP11 which encoded a C2H2-type zinc finger protein (ZFP) in grapevine (Vitis vinifera). Sequence analysis shows that VvZFP11 contained one L-box, two C2H2-type zinc finger motifs and one ERF-associated amphiphilic repression (EAR) motif. The VvZFP11 localized to the nucleus and functional analysis shows that full-length VvZFP11 had no transcriptional activity, but VvZFP11 lacking the EAR motif had a strong transcriptional activity in yeast. In grapevine, expression of VvZFP11 was induced by salicylic acid and methyl jasmonate and also quickly responded to infection with Erisiphe necator. Arabidopsis thaliana plants overexpressing VvZFP11 were more resistant to Golovinomyces cichoracearum, and real time quantitative polymerase chain reaction revealed that defense-related genes AtPR1 and AtPDF1.2 were up-regulated in the overexpressing lines. These results suggest that VvZFP11 might play an important role in defense responses in grapevine.

We demonstrate herein the use of a one-dimensional metal-organic material as a new type of electrode material for lithium-ion batteries (LIBs) in place of the classic porous three-dimensional materials, which are subject to the size of the channel for lithium-ion diffusion and blocking of the windows of the framework by organic solvents during the charging and discharging processes. Introducing a one-dimensional coordination compound can keep organic active substances insoluble in the electrolyte during the charging and discharging processes, providing a facile and general new system for further studies. The results show that both the aromatic ligand and the metal center can participate in lithium storage simultaneously, illustrating a new energy storage mechanism that has been well-characterized by X-ray photoelectron spectroscopy, electron paramagnetic resonance spectroscopy, and cyclic voltammetry. In addition, the fact that the one-dimensional chains are linked by weak hydrogen bonds rather than strong π-π stacking interactions or covalent bonds is beneficial for the release of capacity entirely without the negative effect of burying the active sites.

MIL-101(Cr)/graphene oxides composites has been successfully synthesized via a facile route using a solvothermal treatment and fully characterized using powder X-ray diffraction (PXRD), scanning electron microscope (SEM), thermal gravimetric analysis (TGA) and infrared spectroscopy (IR). The specific capacity of MIL-101(Cr)/GO are almost twice larger than that of MIL-101(Cr) at the first discharge process and the multiple relationship maintained over 40 cycles, indicating that the electrochemical performance of MIL-101(Cr) as anode material has been improved through doping by GO.

Nanostructured NiP2@C was synthesized by a facile approach via calcination of a Ni-based metal–organic-framework (Ni-MOF-74) with adsorptive red phosphorus, and was tested as an anode material for lithium ion batteries. NiP2 nanoparticles are successfully embedded in situ in porous carbon matrix, constructing crosslinked channels for lithium ion diffusion. The in situ introduction of porous carbon round NiP2 nanoparticles greatly enhances the electronic conductivity. Benefiting from the advantages of porous carbon, the nanostructured NiP2@C electrode material showed excellent electrochemical performance with high reversibility, high rate capability, and long-term cycling stability.

MicroRNA-1294 (miR-1294) was reported to act as a tumor suppressor in several cancers. However, the biological function of miR-1294 in osteosarcoma (OS) has not been investigated. We, therefore, investigated the clinical significance and underlying mechanisms of miR-1294 in OS.Quantitative Real-Time-Polymerase Chain Reaction (qRT-PCR) was conducted to detect the levels of miR-1294. Targets of miR-1294 were validated by luciferase reporter assay and Western blot. In vitro functional assays were performed to investigate the effects of miR-217 on cell proliferation and invasion.We found miR-1294 was downregulated in OS tissues and cell lines. Downregulation of miR-1294 has a significant negative impact on the overall survival of OS patients. Overexpression of miR-1294 suppresses OS cell proliferation and invasion in vitro. Then, luciferase reporter assay validated Homeobox A9 (HOXA9) was a downstream target of miR-1294. Expression patterns of miR-1294 were inversely correlated with HOXA9 in OS tissues, strengthening the findings from the luciferase reporter assay. Further functional assays revealed that overexpression of HOXA9 could reverse the inhibition effects of miR-1294 on cell proliferation and invasion.These results suggested miR-1294 functions as a tumor suppressor in OS progression by targeting HOXA9.

The flicker noise of source follower transistors is the dominant noise source in image sensors. This paper reports a systematic study of the shallow trench isolation effect in transistors with different sizes under high temperature conditions that correspond to the quantity of empty defect sites. The effects of shallow trench isolation sidewall defects on flicker noise characteristics are investigated. In addition, the low-frequency noise and subthreshold swing degrade simultaneously in accordance to the device gate width scaling. Both serious subthreshold leakage and considerable noise can be attributed to the high trap density near the STI edge. Consequently, we propose a coincidental relationship between the noise level and the subthreshold characteristic; its trend is identical to the experiments and simulation results.

Previous case-control studies on the relationship between the CYP4F2 gene rs2108622 polymorphism and hypertension have produced contrasting results. In this study, we aimed to further evaluate the relationship between the CYP4F2 gene rs2108622 polymorphism and hypertension. We selected four case-control studies related to the CYP4F2 gene rs2108622 polymorphism and hypertension by searching PubMed, EMBase, the Chinese Biomedical Literature Database, and the Wanfang database. We utilized the Cochran Q-test and the I2 index to measure the heterogeneity across studies. To merge the odds ratio (OR) and the 95% confidence interval (95%CI), we utilized the fixed and random-effect models during the analyses. The present study included 1878 patients with hypertension and 1512 healthy control subjects. By meta-analysis, we did not find any association of the CYP4F2 gene rs2108622 polymorphism with hypertension in either genotype or allele distribution [AA+AG vs GG: OR = 1.18, 95%CI (0.91-1.54), P = 0.21; GG+AG vs AA: OR = 0.91, 95%CI (0.80-1.05), P = 0.20; A allele vs G allele: OR = 1.04, 95%CI (0.93-1.16), P = 0.53]. We concluded that the CYP4F2 gene rs2108622 polymorphism was not associated with hypertension.

This paper presents the study of massively parallel simulation of multi-physics effects in SiP using a high-performance computing scheme. An in-house time-domain finite element parallel program is developed and validated by comparing the simulated electro-thermo-mechanical responses of bonding wire arrays with commercial software. After that, thermo-mechanical responses of SiP are simulated and the parallel efficiency of our parallel program is assessed by the experiment of its strong parallel scalability. Our parallel program can reach a Speedup of 6.095 and strong scalability efficiency of 38.1% on 1024 CPU cores of 128 compute nodes.

We investigate herein the existence of spectral singularities (SSs) in composite systems that consist of two separate scattering centers A and B embedded in one-dimensional free space, with at least one scattering center being non-Hermitian. We show that such composite systems have an SS at $k_{c}$ if the reflection amplitudes $r^{A}\left( k_{c}\right) $ and $r^{B}\left( k_{c}\right) $ of the two scattering centers satisfy the condition $r_{\mathrm{R}% }^{A}\left( k_{c}\right) r_{\mathrm{L}}^{B}\left( k_{c}\right) e^{i2k_{c}\left( x_{B}-x_{A}\right) }=1$. Objective To observe the effectiveness of chlortetracycline (aureomycin) treatment on vulval white lesions and to explore its possible pathogenesis. Materials and methods From January 2001 to April 2011, 194 patients with vulvar non-neoplastic epithelial disorders were divided into three groups according to therapy regimens received, ie, chlortetracycline treatment group (72 cases), chlortetracycline + beclomethasone treatment group (66 cases), and beclomethasone treatment group (56 cases); their local changes of vulvar lesions were observed and efficacy of these treatment profiles was evaluated after one year. Results Effective rates of chlortetracycline group, chlortetracycline + clobetasol group and clobetasol groups were 86.1% (62/72), 87.9% (58/66), and 62.5% (35/56), respectively. There was a significant difference among these three groups (Hc = 10.7766,p = 0.0046), the curative rate of clobetasol group was markedly lower than that of the former two groups (p = 0.0072 and p = 0.0019), but was not statistical significant (p = 0.6077) when compared between the former groups. Conclusion The occurrence of vulvar non-neoplastic epithelial disorders may be associated with chlamydia and mycoplasma infection, the chlortetracycline is an effective drug for this illness, the mechanism of which might be related to killing pathogens directly and inhibiting inflammatory mediators.

The commercial conductive carbon black (CCB) with large surface area and good electrical conductivity after activation treatment is employed as conductive matrix to support sulfur for the cathode of lithium/sulfur battery. The S/activated CCB composites with 64 and 73 wt% sulfur are prepared by a simple heat treatment method. The microstructure and morphology of the as-prepared S/activated CCB composites are characterized by thermogravimetry (TG), X-ray diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). It is demonstrated that sulfur can be loaded into the pores of the host activated CCB matrix in highly dispersed state, when sulfur loading is limited to 64 wt%. While the loaded sulfur increases to 73 wt%, some crystalline sulfur nanoparticles of less than 2 nm can be observed near the pores of the host activated CCB matrix. As expected, the S/activated CCB composite with 64 wt % sulfur presents the optimized electrochemical performance, including the utilization of active sulfur, discharge capacity and cycle stability. Apparently, in order to improve the electrochemical performance of the S/activated CCB composite, the activation treatment of the commercial CCB is essential for increasing the sulfur loading.

The physiological photosynthetic characteristics and antioxidant enzyme system of the high-chlorophyll rice (Oryza sativa L.) mutant (Gc) and its wild type (Zhenshan 97B) were compared and analyzed. Resulting data showed that the total chlorophyll (Chl) and Chl b contents in the Gc mutant were significantly increased by 19.0 and 81.7%, respectively, while the increase in Chl a and thylakoid membrane protein contents was insignificant. The net photosynthetic rate (P N) was significantly higher in the mutant; stomatal conductance, intercellular CO2 concentration, and transpiration rate decreased significantly, and water-use efficiency increased significantly, indicating the higher photochemical efficiency of the mutant. The chlorophyll fluorescence parameters: electron transport rate and effective quantum yield of PSII photochemistry of the mutant were significantly higher than those of Zhenshan 97B. The nonphotochemical quenching of the mutant under light adaptation increased by 52.3%. The enzymatic activity of superoxide dismutase, peroxidas, and catalase in the mutant roots and leaves were all higher than those for the wild-type plants. It is believed that the higher activity of antioxidant enzymes in the mutant may be an important factor making difficult the photo-inactivation of Chl, and thus, increasing the content of Chl, especially Chl b.

Evaluation of CAS-OB refining slags showed that the melting temperature and viscosity were very high and could further increase during the CAS-OB refining process, causing excessive slag to stick to the snorkel with resulting operational problems. To avoid this, B2O3–CaO (mass ratio 1 : 1) and CaF2–CaO (mass ratio 1 : 1) were employed as modifiers added to the slag. The fusibility (melting temperature and viscosity) and desulphurising capacity of modified slag were investigated. Both B2O3–CaO and CaF2–CaO can effectively lower the melting temperature and viscosity of slag. The results of experiments on sulphur partition equilibrium between metal and slag indicate that the sulphur content of metal can be further decreased by the modified slag.

High density sodium lithium niobate lead free ceramics near the morphtropic phase boundary [NaxLi1−xNbO3, (LNN), x = 0.12] were prepared by the solid state reaction method. XRD patterns showed that the lattice structures were changed after polarization. The temperature dependence of the dielectric constant and dielectric loss, pyroelectric coefficient and DSC curves of LNN ceramics showed that there exist three phase transitions from room temperature up to the Curie temperature. The hysteresis loop and piezoelectric properties were measured and discussed.

On 8 November 2013, super typhoon Haiyan made landfall in Philippines. On 24 November, the Chinese hospital ship arrived in Philippines to help with disaster relief efforts. Drinking water was collected at a variety of locations, and the concentration levels of lead were determined with field test kit. The results showed that the levels of lead in 67% of total collected water samples exceeded WHO's standard. Afterwards, the local government had taken many measures to ensure a safe water supply in next few months. This is the first report about water quality in Philippines after the disaster.

The Dragon lake diversion channel (DLDC) is the only river that recharges Dragon Lake, an artificial lake in China. This paper examines the main factors influencing water quality by investigating point source and non-point source pollutants along the main route. Based on the complicated system of rivers and desilting basins, a three-dimensional water quality model using environmental fluid dynamics code (EFDC) was developed. The model of DLDC was calibrated and verified using observed data. The error ranges of river water level, total phosphorus, total nitrogen and chemical oxygen demand were within 5%, 10%, 16% and 20%, respectively, all of which meet the precision requirement. The model was employed to predict the concentrations of pollutants in the main stream under current pollution loads within a year and a flood lasting for 24 hours. The results revealed that the main pollution sources that influence the water quality of waterways were the point sources followed by the non-point pollution sources. Water quality improved when large water quantities were delivered and this trend can be described as dilution. The water quality of the Dongfeng main channel meets the requirement; however, the water quality of the Dongfeng River is somewhat poor, and the water quality of the Wei River is seriously contaminated. To address these problems, we suggest that the Dongfeng River and Wei River adopt a culvert under its riverbeds.

Polyamines are necessary for normal integrity and the restitution after injury of the gastrointestinal epithelium. The objective of this study was to investigate the effects of oral administration of putrescine and proline during the suckling period on epithelial restitution after early weaning in piglets. Eighteen neonatal piglets (Duroc × Landrace × Large Yorkshire) from 3 litters (6 piglets per litter) were assigned to 3 groups, representing oral administration with an equal volume of saline (control), putrescine (5 mg/kg BW), and proline (25 mg/kg BW) twice daily from d 1 to weaning at 14 d of age. Plasma and intestinal samples were obtained 3 d after weaning. The results showed that oral administration of putrescine or proline increased the final BW and ADG of piglets compared with the control (P0.05). Proline treatment decreased plasma D-lactate concentration but increased the villus height in the jejunum and ileum, as well as the percentage of proliferating cell nuclear antigen (PCNA) positive cells and alkaline phosphatase (AKP) activity in the jejunal mucosa (P0.05). The protein expressions for zonula occludens (ZO-1), occludin, and claudin-3 (P0.05) but not mRNA were increased in the jejunum of putrescine- and proline-treated piglets compared with those of control piglets. The voltage-gated K+ channel (Kv) 1.1 protein expression in the jejunum of piglets administrated with putrescine and the Kv1.5 mRNA and Kv1.1 protein levels in the ileum of piglets administrated with proline were greater than those in control piglets (P0.05). These findings indicate that polyamine or its precursor could improve mucosal proliferation, intestinal morphology, as well as tight junction and potassium channel protein expressions in early-weaned piglets, with implications for epithelial restitution and barrier function after stress injury.

Motivated by the physical relevance of a spectral singularity of interacting many-particle system, we explore the dynamics of two bosons as well as fermions in one-dimensional system with imaginary delta interaction strength. Based on the exact solution, it shows that the two-particle collision leads to amplitude-reduction of the wave function. For fermion pair, the amplitude-reduction depends on the spin configuration of two particles. In both cases, the residual amplitude can vanish when the relative group velocity of two single-particle Gaussian wave packets with equal width reaches the magnitude of the interaction strength, exhibiting complete particle-pair annihilation at the spectral singularity., Comment: 8 pages, 2 figures

The ferroelectric domain wall thickness of a fluoride BaMgF4 single crystal was investigated by piezoresponse force microscopy. It was found that the domain wall thickness shows a strong spatial variation in the as-grown crystal and the polarization reversal process. The original wall thickness is greater (about two to seven times) than that switched by the tip fields of the atomic force microscope. A significantly narrower domain wall was obtained in the higher tip-field. The trapped defects at the domain wall play an important role in the spatial variation of the polarization width of 180° domain wall in the BaMgF4 single crystal. (© 2008 WILEY-VCH Verlag GmbH & Co. Selectively photo-excited C-V spectroscopy has been measured in an In0.5Ga0.5As quantum dots (QDs)-embedded, three barrier-two well heterostructure. By comparing with a theoretical capacitance model, the pure capacitive contribution from In0.5Ga0.5As QDs, due to tunnelling coupling between In0.5Ga0.5As QDs and In0.18Ga0.82As quantum well, has been used to obtain the density of charges from photo-excited In0.5Ga0.5As QDs in a very straightforward manner.

An improved filter paper disk bioassay for determining activities of plant growth regulators was developed and evaluated. Indole-3-acetic acid (IAA), (±)-abscisic acid (ABA), and 6-furfurylamino-purine (kinetin) were dissolved in 95% ethanol at fixed dilutions. Specific concentrations of each growth regulator were then evenly dropped onto individual 6-cm paper disks and the solvent evaporated. The activities of the above three water-insoluble plant growth regulators in both solution and the disk assay were compared using the excised cucumber (Cucumis sativus L.) cotyledon root formation bioassay (IAA), wheat (Triticum aestivum L.) coleoptile straight growth bioassay (ABA), and cucumber cotyledon expansion bioassay (kinetin), and similar results were obtained at each concentration. The possible principle of this method has been studied using the cucumber cotyledon expansion bioassay. The results suggested that the disks, as carriers, had highly dispersed kinetin molecules on them and greatly accelerated the dissolution and diffusion of kinetin from disks to water.

Electrophysiological effects of allopurinol on arrhythmias were studied in isolated segments of guinea pig right ventricular free walls paced from endocardium. A high-gain electrocardiogram as well as transmembrane electrical activity from endo- and epicardium were recorded. Tissues were exposed to simulated ischemia for 15 min and then were reperfused with normal Tyrode solution. Sustained or nonsustained ventricular tachycardia, bigeminy, and trigeminy with characteristics of transmural reentry occurred in early reperfusion in 75% of 20 control preparations. Arrhythmias were associated with prolongation of transmural conduction time and abbreviation of endocardial effective refractory period (ERP). Allopurinol strongly reduced the incidence of reperfusion arrhythmias (20-33%) between 10 and 100 microM, whereas either lower or higher concentrations (5 or 500 microM) were less effective (43 and 50%). Antiarrhythmic efficacy correlated with significant attenuation of reperfusion-induced transmural conduction delay (P less than 0.05 or 0.01). Allopurinol did not affect endocardial conduction times nor did it significantly alter endocardial action potential duration or ERP. Our results indicate that allopurinol exerts antiarrhythmic efficacy during reperfusion by selectively attenuating defects related to anisotropic tissue properties.

KCl at 0.134–40.2 mM significantly promoted rooting in excised cucumber ( Cucumis sativus L.) cotyledons. The effect of KCl on indole-3-acetic acid (IAA)-induced rooting was approximately additive. Several salts of potassium at 1.34 mM also significantly promoted rooting in excised cucumber cotyledons. KCl at 0.134–13.4 mM clearly stimulated rooting in mung bean ( Phaseolus radiatus L.) hypocotyls. KCl at 1.34–40.2 mM also clearly enhanced rooting in kidney bean ( Phaseolus vulgaris L.) hypocotyls.

Purpose Various ventricular assist devices (VADs) have been developed for clinical use in recent years. The aim of a multidisciplinary research team at the Fuwai Hospital of the Peking Union Medical College is to design and develop an axial flow left ventricular assist device (LVAD) for adults. Methods Using Computational Fluid Dynamics (CFD), the inflow characteristics of the axial flow pump were analyzed. After CFD analysis, the axial pump was fabricated using a 5-axis, computer numerical control (CNC) milling machine. Performances of the pump both in vitro and in vivo were tested. Results This VAD, which was developed after numerous CFD analyses for the flow characteristics of the pump, is 58.5 mm long, 30 mm wide and weighs 120 g. The pump can deliver 5 lpm for pressures of 100 mmHg over 8500 rpm. The NIH value was 0.01 g/100 L. The hemolysis, which was evaluated in an in vivo test, was a bit higher than the normal value, but remained within an acceptable range. Conclusions Performance of the pump in vitro and in vivo was considered sufficient for an LVAD. Further design improvements are being undertaken in terms of hemolysis and thrombosis to improve the biocompatibility of the pump.

Ketanserin, a selective 5-HT receptor antagonist, prolongs the QT interval of ECG in patients. The purpose of the present study was to determine whether ketanserin would block human cardiac ether-à-go-go-related gene (hERG) potassium channels.Whole-cell patch voltage-clamp technique was used to record membrane currents in HEK 293 cells expressing wild type or mutant hERG channel genes.Ketanserin blocked hERG current (I(hERG)) in a concentration-dependent manner (IC50=0.11 microM). The drug showed an open channel blocking property, the block increasing significantly at depolarizing voltages between +10 to +60 mV. Voltage-dependence for inactivation of hERG channels was negatively shifted by 0.3 microM ketanserin. A 2.8 fold attenuation of inhibition by elevation of external K+ concentration (from 5.0 to 20 mM) was observed, whereas the inactivation-deficient mutants S620T and S631A had the IC50s of 0.84 +/- 0.2 and 1.7 +/-0.4 microM (7.6 and 15.4 fold attenuation of block). In addition, the hERG mutants in pore helix and S6 also significantly reduced the channel block (2-59 fold) by ketanserin.These results suggest that ketanserin binds to and blocks the open hERG channels in the pore helix and the S6 domain; channel inactivation is also involved in the blockade of hERG channels. Blockade of hERG channels most likely contributes to the prolongation of QT intervals in ECG observed clinically at therapeutic concentrations of ketanserin.

Objective To examine adrenal invasion by renal cell carcinoma (RCC), particularly by adrenal micrometastasis, to determine whether adrenalectomy should be performed during radical nephrectomy. Patients and methods From 1987 to 1994, 129 patients with RCC (90 men and 39 women, mean age 61.4 years, range 22–81) underwent radical nephrectomy with associated adrenalectomy because they had risk factors for adrenal invasion (tumour size >5 cm, or tumour of the superior pole). Pathological examinations were carried out systematically and records of these examinations reviewed. The tumour size was recorded and the frequency of invasion calculated. Results There were 10 cases where the gland was invaded; one was a synchronous contralateral metastasis and nine (7%) were ipsilateral invasions of which two were tumours in the superior pole that invaded the gland by direct extension and the other seven invaded the gland by distant metastasis, six being micrometastatic (4.7%). A single micrometastasis was found in two cases (1.5%). There was no adrenal invasion by tumours of 5 cm in diameter were considered, the ipsilateral invasion rate was 11% (9/80) and the micrometastatic rate was 7.5% (6/80). Conclusion Adrenalectomy need not be performed routinely in small tumours which are detected early, but the possibility of adrenal micrometastasis from larger tumours (>5 cm) should be considered.

Piezoelectric and ferroelectric properties of high-pressure-crystallized PVDF (polyvinylidene fluoride) film were evaluated, and ultrasonic transducers usable at high temperatures up to 200 degrees C were developed. The film crystallized in the hexagonal phase showed distinctive ferroelectric properties. The poled film exhibited persistent piezoelectric activity in a temperature range extending just below the melting point, and an electromechanical coupling factor k/sub t/ of 0.1-0.2. Ultrasonic transducers were fabricated using this film. To determine effects of RP58866 on inward rectifier K+ current (IKl), transient outward K+ current (Ito) and delayed outward rectifier K+ current (IK) in isolated cardiac myocytes.In isolated ventricular myocytes of guinea pig and dog, the effect of RP58866 on IKl, Ito, and IK were observed by the whole cell voltage-clamp technique.RP58866 decreased IKl in a concentration-dependent manner, with an IC50 of (3.4 +/- 0.8) micromol.L-1 (n = 6) at -100 mV in guinea pig ventricular cells. In dog ventricular myocytes, RP58866 inhibited Ito with IC50 of (2.3 +/- 0.5) micromol.L-1 at +40 mV. In guinea pig ventricular cells, RP58866 at 100 micromol.L-1 decreased IK: IKstep by (58 +/- 13)% at +40 mV, and IKtail by (86 +/- 17)%, respectively. RP58866 inhibited IKstep with an IC50 of (7.5 +/- 0.8) micromol.L-1, and IKtail with an IC50 of (3.5 +/- 0.9) micromol.L-1. The envelope of tail analysis suggested that both IKr and IKs were inhibited.RP58866 inhibits IKl, Ito, and IK in cardiac myocytes with a similar potency, and is not a specific IKl inhibitor.

The purpose of this study was to measure the effects of mutations on the retinol binding capability of human Repeat 1 of interphotoreceptor retinoid-binding protein (IRBP). First, we predicted important functional amino acids by several computer programs. We also noted the lack of shared functions between Tail-specific protease (Tsp) and IRBP, which bear sequence similarity, and this aided in predicting functional residues. We analyzed the effects of point substitutions on the retinol and fatty acid binding properties of Repeat 1 of human IRBP at 25 and 50 degrees C.To find residues critical to retinol binding that might affect function, a series of thirteen mutations were created by site-specific mutagenesis between positions 140 and 280 in Repeat 1 of human IRBP. These mutants were expressed, purified, and tested for binding properties. The conformations of the proteins were examined by circular dichroism (CD) scans.Seven of the mutations exhibited reduced binding capacity, and five were not expressed at high enough levels to assess binding activity. Four of the mutants were purified, and their CD scans were very similar to those of Repeat 1. Only one of the mutations did not affect binding, folding, or expression when compare to wild type Repeat 1.Several IRBP mutants containing point mutations retained native structure but lost retinol binding function. The data suggest that retinol binding is affected by many different amino acid substitutions in or near a binding pocket. That even a single point substitution can profoundly affect binding without affecting overall conformation suggests that much of Domain B (from amino acid positions 80 to 300) is involved with ligand binding. This excludes three previously proposed IRBP-retinol binding mechanisms: (1) retinol binds to a small portion of the protein repeat, (2) retinol can bind to any hydrophobic patch in the protein, and (3) native conformation is not required for retinol binding to the repeat.

We compared the structure and function of interphotoreceptor retinoid-binding protein (IRBP) related proteins and predicted domain and secondary structure within each repeat of IRBP and its relatives. We tested whether tail specific protease (Tsp), which bears sequence similarity to IRBP Domain B, binds fatty acids or retinoids, and whether IRBP possessed protease activity resembling Tsp's catalytic function. These tests helped us to learn whether the primary sequence similarities of family members extended to higher order structural and functional levels.Predictions derived from multiple sequence alignments among IRBP and Tsp family members and secondary structure computer programs were carried out. The first repeat of human IRBP (EcR1) and Tsp were expressed, purified, and tested for binding properties. Tsp was examined for fluorescence enhancement of retinol or 16-anthroyloxy-palmitic acid (16-AP) to test for ligand binding. IRBP was tested for protease activity.Tsp did not exhibit fluorescence enhancement with retinol or 16-AP. IRBP did not exhibit protease activity. The positions of critical residues needed for the ligand binding properties of retinol were predicted. Primary sequence and three-dimensional similarity was found between Domain A of IRBP Repeat 3 and eglin c.The sequence similarity of Tsp and IRBP raised the possibility that each might share the function of the other protein: IRBP might possess protease activity or Tsp might possess retinoid or fatty acid binding activity. Our studies do not support such a shared function hypothesis, and suggest that the sequence similarity is the result of maintenance of structure. The finding of similarity to eglin c in Domain A suggests the possibility of a tight interaction between Domain A and Domain B, possibly implying the need for Domain A in retinoid-binding, and suggesting that both Domains should be present in testing mutations. The positions of predicted critical amino acids suggest models in which a large binding pocket holds the retinoid or fatty acid ligand. These predictions are tested in a companion paper.

Interphotoreceptor retinoid-binding protein(IRBP) is a four-repeat protein found in the interphotoreceptor space. Each repeat can bind retinoids and fatty acids. The purpose of this study was to examine the effects of the single amino acid substitution, G239T, versus the wild type sequence of human IRBP Repeat 1, on ligand binding at equilibrium, ligand off rates, and protection of retinol from degradation.G239T was created by site-specific mutagenesis, expressed in E. coli, and purified. E. coli expressed wild type Repeat 1 (EcR1) and G239T were subjected to thermal denaturation and analyzed by circular dichroism spectroscopy. We compared the ligand binding properties by fluorescence enhancement of retinol and 16-anthroyloxy-palmitate, tryptophan quenching of the proteins by different ligands, binding competition assays, protection of retinol from degradation, and stopped-flow kinetics to measure transfer of ligands to and from model membranes.Circular dichroism, fluorescence, and absorbance spectroscopy of G239T and EcR1 showed similar wavelength scans. G239T exhibited about three-fold less fluorescence of bound all-trans-retinol or 13-cis-retinol versus EcR1. Retinol quenching of intrinsic protein fluorescence was reduced by 37% in G239T versus EcR1. Other retinoids used as quenchers produced no difference between intrinsic protein fluorescence of either G239T or EcR1; all exhibited saturable high affinity binding to each protein. Docosahexaenoic acid (DHA) served as a competitive inhibitor of retinol fluorescence enhancement with EcR1. However, DHA did not alter retinol fluorescence with G239T. 16-anthroyloxy-palmitate (16-AP) exhibited about 30% higher levels of fluorescence enhancement when bound to G239T versus EcR1. EcR1 prevented oxidative damage of all-trans-retinol, whereas G239T provided much less protection. Each protein could accept 9-cis-retinal from small unilamellar vesicles (SUVs) as measured by stopped flow kinetics. Off rates were the same in comparing G239T and EcR1 as acceptors.Despite the general similarity in shape between G239T and EcR1 and the nearly identical binding behavior with some ligands, distinct differences exist in the ligand binding properties of G239T and EcR1. Fluorescence enhancement/quenching and retinol protection experiments suggest that retinol binding is reduced by about 50% in G239T versus EcR1. The data suggest that either: (1) EcR1 contains two binding sites for retinol and G239T has lost one site or (2) EcR1 has a single binding site that is altered in G239T to reduce retinol binding. Results of all the experiments were consistent with the first model while some of the data were not consistent with the second model. Thus, it is possible that position 239, found in Domain B2 of IRBP Repeat 1, is located in or near one of two retinol binding sites.

Adventitious root formation in excised cucumber (Cucumis sativus L.) cotyledons was significantly promoted by (±)-cis-chrysanthemic acid at 0.006–1.8 mM. The effect of (±)-cis-chrysanthemic acid on indole-3-acetic acid (IAA)-induced rooting was additive. Rooting in excised cucumber cotyledons was significantly promoted by several isomers of chrysanthemic acid and sodium (±)-cis-chrysanthemate at 0.18 mM. Rooting in mung bean (Phaseolus radiatus L.) hypocotyls was also stimulated by the sodium salt at 0.06–0.6 mM. Rooting of kidney bean (Phaseolus vulgaris L.) hypocotyls was also clearly enhanced by sodium (±)-cis-chrysanthemate at 0.18–6 mM.

Whether a ligand can stabilize the protein from thermal denaturation, 4. Whether the four repeats can be further classified into two groups. Our rationale was to make each repeat from the human cDNA and then examine structural and functional characteristics.Individual repeats were produced in E. coli and the whole protein was expressed in baculovirus. Binding properties with all-trans-retinol were characterized by ligand fluorescence enhancement. The quenching of protein fluorescence by retinol, 9-cis-retinal, all-trans-retinoic acid, beta-ionine, alpha-ionine, trans-parinaric acid, and DHA was also examined. Binding curves were analyzed by nonlinear regression. Prevention of retinol decomposition was measured by absorption spectroscopy. Circular dichroism was examined in the far UV range to study protein secondary structure and the near UV range to study ligand binding effects on the tryptophan environment.Temperature dependent denaturation suggests that EcR1 is the most stable of the four repeats. Each repeat possesses the capability of binding 9-cis-retinal, all-trans-retinol, all-trans retinoic acid, docosahexaenoic acid, alpha- and beta-ionine, and trans-parinaric acid. Protein fluorescence quenching by retinol and retinol fluorescence enhancement assays yielded similar binding parameters for each repeat. Each expressed repeat prevents the degradation of retinol in aqueous solutions.The data contrast with the idea that two or more repeats are needed to bind one molecule of ligand. Each repeat binds both retinoids and analogs, suggesting that each has multiple ligand binding sites or one binding site with affinity for different ligands. Together, the results suggest that each repeat retains all functions of the whole protein. However, there are distinguishing characteristics among the repeats in their ligand binding properties, though the four repeats cannot be classified into just two distinctive groups. Last, these data fit well with the current model of multiple binding sites in IRBP derived from quadruplication of an ancestral monomeric binding protein.

A variety of class III antiarrhythmic agents have been shown to block the delayed rectifier current, but their effects on other K+ currents, particularly in human tissues, are less clear. We studied the concentration-dependent actions of the class III compounds d-sotalol, E-4031 and ambasilide on the transient outward current (I(to)) and the ultra-rapid delayed rectifier current (I(Kur)) in human atrial myocytes. d-Sotalol and E-4031 failed to alter I(to) or I(Kur) at concentrations up to 500 and 50 microM, respectively. In contrast, ambasilide produced a concentration-dependent inhibition of I(to) and I(Kur), with statistically significant effects at 10 microM and maximum effects at 100 microM. The 50% inhibitory concentration of ambasilide averaged 23 +/- 2 microM and 34 +/- 3 microM for I(to) and I(Kur) respectively. Ambasilide did not alter the voltage-dependence of activation or inactivation of I(to), or the voltage-dependence of I(Kur), and it did not affect I(to) recovery from inactivation. On the other hand, ambasilide accelerated I(to) inactivation, by introducing a more rapid component that accelerated with increasing drug concentration. Furthermore, block of both I(to) and I(Kur) developed over time after the onset of depolarization, with time constants of 5.8 +/- 0.8 msec and 2.5 +/- 0.4 msec at concentrations of 10 and 50 microM for I(to) and 6.1 +/- 0.8 msec and 2.1 +/- 0.3 msec at 10 and 50 microM for I(Kur). We conclude that neither d-sotalol nor E-4031 affects I(to) or I(Kur), whereas ambasilide produces efficacious open-channel block of both currents, in human atrial myocytes.

Erectile dysfunction associated to low circulating androgens is characterized by a low testosterone due to hypogonadism of hypothalamic-pituitary or testicular origin. Long term androgen administration is unacceptable unless biology has demonstrated hypogonadism with a low testosterone level not related to hyperpolactinemia. Intramuscular testosterone or transdermal dihydrotestosterone can be used with a similar clinical effect but a different biological impact regarding aromatase activity. Whatever may be the type of androgen supplementation, one should use low dosage with frequent administration in order to obtain stable and physiologic plasmatic values. On account of androgen impact on prostate and cardiovascular system, careful pretreatment screening should eliminate an occult prostate cancer and a vascular thrombosis risk in a complaining and informed patient. Clinical and biological parameters should be periodically followed throughout androgen therapy.

lymph node involvement in renal cell carcinoma is factor of very poor prognosis. In a series of 55 node-positive patients, 33 (60%) had simultaneous renal vein or vena cava invasion and 32 (58.2%) had metastases. Gross lymph node involvement was found in 39 patients (70.9%). Patients without venous invasion or metastasis may have a prolonged survival. In this group, those with microscopic nodal involvement can be cured, as the 10 and 15-year the actuarial survival rate is 54.5% Formal lymphadenectomy might have played a role in these results. Surgery can be performed when vein invasion is present without metastasis, but the prognosis is generally poor. Survival does not seem to be influenced by surgery when metastasis is present, regardless of the vein status.

We report 13 cases of renal oncocytoma. Urinary symptoms occurred in only 3 cases. The patient's age ranged from 41 to 74 years with an average of 62.3 years. The mean tumor diameter was 5.6 cm (range: 1.5-14). Diagnostic features of ultrasonography, CT scan and, in some instances, angiography were suggestive of renal oncocytoma in 2 patients, but never affirmative, 4 patients were treated by partial nephrectomy. No local or metastatic recurrence was observed with a mean follow-up of 30.8 months, ranging from 6 to 96 months. We assume that the term renal oncocytoma should be restricted to tumors exclusively composed of regular oncocytic cells with an eosinophilic granular mitochondria-rich cytoplasm and an absence of malignant potential. Diagnostic imaging characteristics may sometimes suggest the diagnosis of renal oncocytoma, but cannot eliminate the main differential diagnosis, i.e. granular renal cell adenocarcinoma. When the tumor is small and unifocal, nephron sparing surgery may be considered. Whether or not the diagnosis has been confirmed by fine needle aspiration, conservative surgery must be controlled by intraoperative frozen sections of the tumor and surgical margins.
