Search

Identity matters in science, technology, engineering, mathematics, and medicine (STEMM) because it can affect an individual's long-term sense of belonging, which may in turn affect their persistence in STEMM. Early K-12 science classes often teach students about the foundational discoveries of the field, which have been predominately made, or at least published, by White men. This homogeneity can leave underrepresented individuals in STEMM feeling isolated, and underrepresented K-12 students may feel as though they cannot enter STEMM fields. This study aimed to examine these feelings of inclusivity in STEMM through an interactive workshop that asked middle schoolers to identify scientists from images of individuals with various racial and gender identities. We found that a plurality of students had a positive experience discussing diversity in science and recognizing underrepresented individuals as scientists.

Abstract A search for full energy depositions from bosonic keV-scale dark matter candidates of masses between 65 and 1021 keV has been performed with data collected during Phase II of the GERmanium Detector Array (Gerda) experiment. Our analysis includes direct dark matter absorption as well as dark Compton scattering. With a total exposure of 105.5 kg years, no evidence for a signal above the background has been observed. The resulting exclusion limits deduced with either Bayesian or Frequentist statistics are the most stringent direct constraints in the major part of the 140–1021 keV mass range. As an example, at a mass of 150 keV the dimensionless coupling of dark photons and axion-like particles to electrons has been constrained to $$\alpha '/\alpha 1.5 \times 10^{24}$$ τ n > 1.5 × 10 24 years and for a proton $$\tau _\textrm{p} > 1.3 \times 10^{24}$$ τ p > 1.3 × 10 24 years at 90% CI. For the electron decay $$e^\text {-} \rightarrow \nu _\textrm{e} \gamma $$ e - → ν e γ a lower limit of $$\tau _\textrm{e} > 5.4\times 10^{25}$$ τ e > 5.4 × 10 25 years at 90% CI has been determined.

While climate models broadly agree on the changes expected to occur over the Arctic with global warming on a pan-Arctic scale (i.e. polar amplification, sea ice loss, and increased precipitation), the magnitude and patterns of these changes at regional and local scales remain uncertain. This limits the usability of climate model projections for risk assessments and their impact on human activities or ecosystems (e.g. fires and permafrost thawing). Whereas any single or ensemble mean projection may be of limited use to stakeholders, recent studies have shown the value of the storyline approach in providing a comprehensive and tractable set of climate projections that can be used to evaluate changes in environmental or societal risks associated with global warming. Here, we apply the storyline approach to a large ensemble of the Coupled Model Intercomparison Project Phase 6 (CMIP6) models with the aim of distilling the wide spread in model predictions into four physically plausible outcomes of Arctic summertime climate change. This is made possible by leveraging strong covariability in the climate system associated with well-known but poorly constrained teleconnections and local processes; specifically, we find that differences in Barents–Kara sea warming and lower-tropospheric warming over polar regions among CMIP6 models explain most of the inter-model variability in pan-Arctic surface summer climate response to global warming. Based on this novel finding, we compare regional disparities in climate change across the four storylines. Our storyline analysis highlights the fact that for a given amount of global warming, certain climate risks can be intensified, while others may be lessened, relative to a “middle-of-the-road” ensemble mean projection. We find this to be particularly relevant when comparing climate change over terrestrial and marine areas of the Arctic which can show substantial differences in their sensitivity to global warming. We conclude by discussing the potential implications of our findings for modelling climate change impacts on ecosystems and human activities.

Abstract In tactile sensing, decoding the journey from afferent tactile signals to efferent motor commands is a significant challenge primarily due to the difficulty in capturing population-level afferent nerve signals during active touch. This study integrates a finite element hand model with a neural dynamic model by using microneurography data to predict neural responses based on contact biomechanics and membrane transduction dynamics. This research focuses specifically on tactile sensation and its direct translation into motor actions. Evaluations of muscle synergy during in -vivo experiments revealed transduction functions linking tactile signals and muscle activation. These functions suggest similar sensorimotor strategies for grasping influenced by object size and weight. The decoded transduction mechanism was validated by restoring human-like sensorimotor performance on a tendon-driven biomimetic hand. This research advances our understanding of translating tactile sensation into motor actions, offering valuable insights into prosthetic design, robotics, and the development of next-generation prosthetics with neuromorphic tactile feedback.

Mitochondrial transplantation has been used to treat various diseases associated with mitochondrial dysfunction. Here, we highlight the considerations in quality control mechanisms that should be considered in the context of mitochondrial transplantation.

Summary: Matrix stiffening by lysyl oxidase-like 2 (LOXL2)-mediated collagen cross-linking is proposed as a core feedforward mechanism that promotes fibrogenesis. Failure in clinical trials of simtuzumab (the humanized version of AB0023, a monoclonal antibody against human LOXL2) suggested that targeting LOXL2 may not have disease relevance; however, target engagement was not directly evaluated. We compare the spatial transcriptome of active human lung fibrogenesis sites with different human cell culture models to identify a disease-relevant model. Within the selected model, we then evaluate AB0023, identifying that it does not inhibit collagen cross-linking or reduce tissue stiffness, nor does it inhibit LOXL2 catalytic activity. In contrast, it does potently inhibit angiogenesis consistent with an alternative, non-enzymatic mechanism of action. Thus, AB0023 is anti-angiogenic but does not inhibit LOXL2 catalytic activity, collagen cross-linking, or tissue stiffening. These findings have implications for the interpretation of the lack of efficacy of simtuzumab in clinical trials of fibrotic diseases.

Summary: Abdominal and thoracic aortic aneurysms (AAAs, TAAs) remain a major cause of deaths worldwide, in part due to the lack of reliable prognostic markers or early warning signs. Sox6 has been found to regulate renin controlling blood pressure. We hypothesized that Sox6 may serve as an important regulator of the mechanisms contributing to hypertension-induced aortic aneurysms. Phenotype and laboratory-wide association scans in a clinical cohort found that SOX6 gene expression is associated with aortic aneurysm in subjects of European ancestry. Sox6 and tumor necrosis factor alpha (TNF-α) expression were upregulated in aortic tissues from patients affected by either AAA or TAA. In Sox6 knockout mice with angiotensin-II-induced AAA, we found that Sox6 plays critical role in the development and progression of AAA. Our data support a regulatory role of SOX6 in the development of hypertension-induced AAA, suggesting that Sox6 may be a therapeutic target for the treatment of aortic aneurysms.

Thermodynamics has predicted many different kinds of microbial metabolism by determining which pairs of electron acceptors and donors will react to produce an exergonic reaction (a negative net change in Gibbs free energy). In energy-limited environments, such as the deep subsurface, such an approach can reveal the potential for unexpected or counter-intuitive energy sources for microbial metabolism. Up until recently, these thermodynamic calculations have been carried out with the assumption that chemical species appearing on the reactant and product side of a reaction formula have a constant concentration, and thus do not count towards net concentration changes and the overall direction of the reaction. This assumption is reasonable considering microorganisms are too small (~1 μm) for any significant differences in concentration to overcome diffusion. However, recent discoveries have demonstrated that the reductive and oxidative halves of reactions can be separated by much larger distances, from millimetres to centimetres via conductive filamentous bacteria, mineral conductivity, and biofilm conductivity. This means that the concentrations of reactants and products can indeed be different, and that concentration differences can contribute to the net negative change in Gibbs free energy. It even means that the same redox reaction, simultaneously running in forward and reverse, can drive energy conservation, in an ElectroMicrobiological Concentration Cell (EMCC). This paper presents a model to investigate this phenomenon and predict under which circumstances such concentration-driven metabolism might take place. The specific cases of oxygen concentration cells, sulfide concentration cells, and hydrogen concentration cells are examined in more detail.

ABSTRACT The microbial communities of marine seep sediments contain unexplored physiological and phylogenetic diversity. Here, we examined 30 bacterial metagenome-assembled genomes (MAGs) from cold seeps in the South China Sea, the Indian Ocean, the Scotian Basin, and the Gulf of Mexico, as well as from deep-sea hydrothermal sediments in the Guaymas Basin, Gulf of California. Phylogenetic analyses of these MAGs indicate that they form a distinct phylum-level bacterial lineage, which we propose as a new phylum, Candidatus Effluviviacota, in reference to its preferential occurrence at diverse seep areas. Based on tightly clustered high-quality MAGs, we propose two new genus-level candidatus taxa, Candidatus Effluvivivax and Candidatus Effluvibates. Genomic content analyses indicate that Candidatus Effluviviacota are chemoheterotrophs that harbor the Embden–Meyerhof–Parnas glycolysis pathway. They gain energy by fermenting organic substrates. Additionally, they display potential capabilities for the degradation of cellulose, hemicellulose, starch, xylan, and various peptides. Extracellular anaerobic respiration appears to rely on metals as electron acceptors, with electron transfer primarily mediated by multiheme cytochromes and by a flavin-based extracellular electron transfer (EET) mechanism that involves NADH-quinone oxidoreductase-demethylmenaquinone-synthesizing enzymes, uncharacterized membrane proteins, and flavin-binding proteins, also known as the NUO-DMK-EET-FMN complex. The heterogeneity within the Ca. Effluviviacota phylum suggests varying roles in energy metabolism among different genera. While NUO-DMK-EET-FMN electron transfer has been reported predominantly in Gram-positive bacteria, it is now identified in Ca. Effluviviacota as well. We detected the presence of genes associated with bacterial microcompartments in Ca. Effluviviacota, which can promote specific metabolic processes and protect the cytosol from toxic intermediates.IMPORTANCEThe newly discovered bacterial phylum Candidatus Effluviviacota is widespread across diverse seepage ecosystems, marine environments, and freshwater environments, with a notable preference for cold seeps. While maintaining an average abundance of approximately 1% in the global gene catalog of cold seep habitats, it has not hitherto been characterized. The metabolic versatility of Ca. Effluviviacota in anaerobic carbon, hydrogen, and metal cycling aligns with its prevalence in anoxic niches, with a preference for cold seep environments. Variations in metabolic potential between Ca. Effluvivivax and Ca. Effluvibates may contribute to shaping their respective habitat distributions.

Abstract Local meteorology over the Great Barrier Reef (GBR) can significantly influence ocean temperatures, which in turn impacts coral ecosystems. While El Niño–Southern Oscillation (ENSO) provides insight into the expected synoptic states, it lacks details of anticipated sub‐seasonal weather variability at local scales. This study explores the influence of the Madden‐Julian oscillation (MJO) on Australian tropical climate, both independently and in combination with ENSO, focusing on GBR impacts. We find that during El Niño periods, including the summer of 2009/10, faster propagating MJO patterns can disrupt background warm, dry conditions, and potentially provide cooling relief via increased cloud cover and stronger winds. In La Niña periods, such as the summer of 2021/22, the MJO tends to be prevented from passing the Maritime continent, forcing it to remain in a standing pattern in the Indian Ocean. This leads to decreased cloud cover and weaker winds over the GBR, generating warm ocean anomalies.

Abstract The variability and distribution of Australia’s summer rainfall are influenced by modes of climate variability on multi-week to multi-year time scales. Here, we investigate the role of the stratospheric quasi-biennial oscillation (QBO) and demonstrate that the QBO influences rainfall variations and extremes’ responses across large regions of Australia. We find the QBO modulates convective heating to the east of the Maritime Continent and over the central South Atlantic Ocean in the austral summer. The baroclinic response and barotropic structure of the extra-tropical Rossby wave train induces anomalous circulation that affects the distribution and amount of rainfall over Australia. Our analysis and findings of QBO teleconnections with the dynamics that drive Australia’s rainfall variability and extremes represents a pathway to improve our understanding of rainfall potential predictability and scope to extend Australia’s rainfall prediction lead times.

A partial mitochondrial DNA Cytochrome Oxidase subunit I (mtCOI) gene haplotype variant of the coconut rhinoceros beetle (CRB) Oryctes rhinoceros, classed as ‘CRB-G (clade I)’, has been the focus of much research since 2007, with reports of invasions into new Pacific Island locations (e.g., Guam, Hawaii, Solomons Islands). For numerous invasive species, inference of invasion biology via whole genome is superior to assessments via the partial mtCOI gene. Here, we explore CRB draft mitochondrial genomes (mitogenomes) from historical and recent collections, with assessment focused on individuals associated within the CRB-G (clade I) classification. We found that all Guam CRB individuals possessed the same mitogenome across all 13 protein-coding genes and differed from individuals collected elsewhere, including ‘non-Guam’ individuals designated as CRB-G (clade I) by partial mtCOI assessment. Two alternative ATP6 and COIII partial gene primer sets were developed to enable distinction between CRB individuals from Guam that classed within the CRB-G (clade I) haplotype grouping and CRB-G (Clade I) individuals collected elsewhere. Phylogenetic analyses based on concatenated ATP6–COIII genes showed that only Guam CRB-G (clade I) individuals clustered together, and therefore Guam was not the source of the CRB that invaded the other locations in the Pacific assessed in this study. The use of the mtCOI and/or mtCOIII genes for initial molecular diagnosis of CRB remained crucial, and assessment of more native CRB populations will further advance our ability to identify the provenance of CRB invasions being reported within the Pacific and elsewhere.

There remains a clear deficiency in recruiting middle school students in science, technology, engineering, mathematics, and medicine fields, especially for those students entering physiology from underrepresented backgrounds. A large part of this may be arising from a disconnect between how science is typically practiced at a collegiate and K-12 level. Here, we have envisioned mitochondria and their diverse subcellular structures as an involver for middle school students. We present the framework for a workshop that familiarizes students with mitochondria, employing three-dimensional visual-spatial learning and real-time critical thinking and hypothesis forming. This workshop had the goal of familiarizing middle school students with the unique challenges the field currently faces and better understanding the actuality of being a scientist through critical analysis including hypothesis forming. Findings show that middle school students responded positively to the program and felt as though they had a better understanding of mitochondria. Future implications for hands-on programs to involve underrepresented students in science are discussed, as well as potential considerations to adapt it for high school and undergraduate students.

Serological studies in developed and developing countries using enzyme-linked immunosorbent assays have validated this technique as a rapid, noninvasive method for the diagnosis of Helicobacter pylori infections. The prevalence of serum antibodies to H pylori was studied in 473 Canadian blood donors from Manitoba, 212 healthy Japanese. and 226 healthy Americans. As expected, the seroprevalence rose progressively with age in the three populations and reached its peak (greater than 55%) in subjects 60 years of age and older. The seroprevalence did not decrease in elderly persons (60 to 99 years), indicating a persistent immune response. More detailed analysis was perfom1ed on the Canadian population. Age-adjusted prevalence rates in men and women were similar. Among young adults (15 to 29 years). farmers had a significantly higher seroprevalence rate than white-collar or blue-collar workers. but in older persons occupational rates were similar. A multiple linear regression analysis of the data confirmed that age and occupation in young adults were both significantly associated with seroprevalence of H pylori infections.

Background: Though there are more family physicians in Canada than ever before, and residency programs are expanding, gaps in access to comprehensive care remain. This study aimed to describe and understand the role residency training experiences played in shaping practice choices, including the provision of comprehensive community-based care, among early career family physicians. Methods: A secondary analysis of sixty-three (63) qualitative interviews was conducted on data from a larger mixed method study on practice patterns and choices of early career Canadian family physicians. We utilized Braun and Clarke’s six phases of reflexive thematic analysis on portions of transcripts concerning residency training experiences. Results: Participants described positive residency training experiences that shaped practice choice with respect to preceptors and mentorship, experiences of longitudinal care, breadth of exposure, and preparedness for comprehensive clinical practice. Woven through these four domains were “points of tension” and “hidden truths”. Points of tension included: i) the promotion of an idealized professional identity and practice that was difficult to uphold, ii) lack of representation among faculty/preceptors with respect to age and gender, at some sites, and iii) frustration about the lack of opportunities for interprofessional collaborative practices that reflected training experiences. Hidden truths included: i) lack of preparation to run a business, ii) high administrative workload, iii) realities of payment models, and iv) the range of roles available for family physicians beyond the provision of comprehensive care. Conclusions: Findings highlight opportunities for educational reform supporting the transition from residency to practice alongside the importance of addressing systemic factors beyond training which impact physicians’ choices regarding comprehensive care.

Summary: It is well-understood that the science, technology, engineering, and mathematics (STEM) fields have unique challenges that discourage recruiting and retaining underrepresented minorities. Research programs aimed at undergraduates have arisen as a critical mechanism for fostering innovation and addressing the challenges faced by underrepresented minorities. Here, we review various undergraduate research programs designed to provide exposure to undergraduates, with a focus on underrepresented minorities in STEM disciplines. We provide insight into selected programs' objectives, key features, potential limitations, and outcomes. We also offer recommendations for future improvements of each research program, particularly in the context of mentorship. These programs range from broad-reaching initiatives (e.g., Leadership Alliance) to more specific programs targeting underrepresented students. By offering a nuanced understanding of each program’s structure, we seek to provide a brief overview of the landscape of diversity-focused STEM initiatives and a guide on how to run a research program effectively.

The factors that determine fibrosis progression or normal tissue repair are largely unknown. We previously demonstrated that autophagy inhibition-mediated epithelial–mesenchymal transition (EMT) in human alveolar epithelial type II (ATII) cells augments local myofibroblast differentiation in pulmonary fibrosis by paracrine signaling. Here, we report that liver kinase B1 (LKB1) inactivation in ATII cells inhibits autophagy and induces EMT as a consequence. In IPF lungs, this is caused by the down-regulation of CAB39L, a key subunit within the LKB1 complex. 3D co-cultures of ATII cells and MRC5 lung fibroblasts coupled with RNA sequencing (RNA-seq) confirmed that paracrine signaling between LKB1-depleted ATII cells and fibroblasts augmented myofibroblast differentiation. Together, these data suggest that reduced autophagy caused by LKB1 inhibition can induce EMT in ATII cells and contribute to fibrosis via aberrant epithelial–fibroblast crosstalk.

BackgroundOrgan transplantation from donors with human immunodeficiency virus (HIV) to recipients with HIV (HIV D+/R+) presents risks of donor-derived infections. Understanding clinical, immunologic, and virologic characteristics of HIV-positive donors is critical for safety.MethodsWe performed a prospective study of donors with HIV-positive and HIV false-positive (FP) test results within the HIV Organ Policy Equity (HOPE) Act in Action studies of HIV D+/R+ transplantation (ClinicalTrials.gov NCT02602262, NCT03500315, and NCT03734393). We compared clinical characteristics in HIV-positive versus FP donors. We measured CD4 T cells, HIV viral load (VL), drug resistance mutations (DRMs), coreceptor tropism, and serum antiretroviral therapy (ART) detection, using mass spectrometry in HIV-positive donors.ResultsBetween March 2016 and March 2020, 92 donors (58 HIV positive, 34 FP), representing 98.9% of all US HOPE donors during this period, donated 177 organs (131 kidneys and 46 livers). Each year the number of donors increased. The prevalence of hepatitis B (16% vs 0%), syphilis (16% vs 0%), and cytomegalovirus (CMV; 91% vs 58%) was higher in HIV-positive versus FP donors; the prevalences of hepatitis C viremia were similar (2% vs 6%). Most HIV-positive donors (71%) had a known HIV diagnosis, of whom 90% were prescribed ART and 68% had a VL

Abstract Shewanella species are frequently selected as model strains to investigate microbially influenced steel corrosion. This selection is due to their relevance for corrosion, but also because of their easy cultivation in aerobic media. Unfortunately, these cultivation advantages do not lead to a straight-forward interpretation of their corrosion inducing or inhibiting mechanisms. The metabolic versatility of Shewanellae indeed enables a wide variety of corrosion mechanisms. This work reviews the metabolic capacities and the extracellular electron transfer mechanisms of Shewanellae and explains how these abilities lead to the various mechanisms by which Shewanellae induce and inhibit corrosion. It should be emphasized that the medium composition (presence of electron donor, acceptor, carbon source) strongly affects which mechanism is in play. Overall, this work concludes that Shewanellae model strains offer great opportunities to study corrosion, thanks in part due to genetic engineering options, but the full complexity of their corrosion processes should always be kept mind.

Abstract We search for tri-nucleon decays of $$^{76}$$ 76 Ge in the dataset from the GERmanium Detector Array (GERDA) experiment. Decays that populate excited levels of the daughter nucleus above the threshold for particle emission lead to disintegration and are not considered. The ppp-, ppn-, and pnn-decays lead to $$^{73}$$ 73 Cu, $$^{73}$$ 73 Zn, and $$^{73}$$ 73 Ga nuclei, respectively. These nuclei are unstable and eventually proceed by the beta decay of $$^{73}$$ 73 Ga to $$^{73}$$ 73 Ge (stable). We search for the $$^{73}$$ 73 Ga decay exploiting the fact that it dominantly populates the 66.7 keV $$^{73m}$$ 73 m Ga state with half-life of 0.5 s. The nnn-decays of $$^{76}$$ 76 Ge that proceed via $$^{73m}$$ 73 m Ge are also included in our analysis. We find no signal candidate and place a limit on the sum of the decay widths of the inclusive tri-nucleon decays that corresponds to a lower lifetime limit of 1.2 $$\times $$ × 10 $$^{26}$$ 26 yr (90% credible interval). This result improves previous limits for tri-nucleon decays by one to three orders of magnitude.

Abstract The ability to detect liquid argon scintillation light from within a densely packed high-purity germanium detector array allowed the Gerda experiment to reach an exceptionally low background rate in the search for neutrinoless double beta decay of $${}^{76}$$ 76 Ge. Proper modeling of the light propagation throughout the experimental setup, from any origin in the liquid argon volume to its eventual detection by the novel light read-out system, provides insight into the rejection capability and is a necessary ingredient to obtain robust background predictions. In this paper, we present a model of the Gerda liquid argon veto, as obtained by Monte Carlo simulations and constrained by calibration data, and highlight its application for background decomposition.

β-thalassemia is a quantitative hemoglobin (Hb) disorder resulting in reduced production of Hb A and increased levels of Hb A2. Diagnosis of β-thalassemia can be problematic when combined with other structural Hb variants, so that the separation approaches in routine clinical centers are not sufficiently decisive to obtain accurate results. Here, we separate the intact Hb subunits by high-performance liquid chromatography, followed by top-down tandem mass spectrometry of intact subunits to distinguish Hb variants. Proton transfer reaction-parallel ion parking (PTR-PIP), in which a radical anion removes protons from multiply charged precursor ions and produces charge-reduced ions spanning a limited m/z range, was used to increase the signal-to-noise ratio of the subunits of interest. We demonstrate that the δ/β ratio can act as a biomarker to identify β-thalassemia in normal electrospray ionization MS1 and PTR-PIP MS1. The application of PTR-PIP significantly increases the sensitivity and specificity of the HPLC-MS method to identify δ/β ratio as a thalassemia biomarker.

The increasing frequency and duration of severe fire events in Australia further necessitate accurate and timely forecasting to mitigate their consequences. This study evaluated the performance of two distinct approaches to forecasting extreme fire danger at two- to three-week lead times for the period 2003 to 2017: the official Australian climate simulation dynamical model and a statistical model based on climate drivers. We employed linear logistic regression to develop the statistical model, assessing the influence of individual climate drivers using single linear regression. The performance of both models was evaluated through case studies of three significant extreme fire events in Australia: the Canberra (2003), Black Saturday (2009), and Pinery (2015) fires. The results revealed that ACCESS-S2 generally underestimated the spatial extent of all three extreme FBI events, but with accuracy scores ranging from 0.66 to 0.86 across the case studies. Conversely, the statistical model tended to overpredict the area affected by extreme FBI, with high false alarm ratios between 0.44 and 0.66. However, the statistical model demonstrated higher probability of detection scores, ranging from 0.57 to 0.87 compared with 0.03 to 0.57 for the dynamic model. These findings highlight the complementary strengths and limitations of both forecasting approaches. Integrating dynamical and statistical models with transparent communication of their uncertainties could potentially improve accuracy and reduce false alarms. This can be achieved through hybrid forecasting, combined with visual inspection and comparison between the statistical and dynamical forecasts. Hybrid forecasting also has the potential to increase forecast lead times to up to several months, ultimately aiding in decision-making and resource allocation for fire management.

Diversity, equity, and inclusion (DEI) initiatives are critical for fostering growth, innovation, and collaboration in science, technology, engineering, mathematics, and medicine (STEMM). This article focuses on four key topics that have impacted many Black individuals in STEMM: know-your-place aggression, environmental microaggressions, peer mediocrity, and code-switching. We provide a comprehensive background on these issues, discuss current statistics, and provide references that support their existence, as well as offer solutions to recognize and address these problems in the STEMM which can be expanded to all historically underrepresented individuals.

Underrepresented faculty have higher burnout rates and lower grant attainment rates when compared with their non-minority counterparts. Many in science, technology, engineering, mathematics, and medicine (STEMM) disciplines, including underrepresented individuals, often have difficulty dedicating time to the writing process, with trainees often being relegated to laboratory tasks in their training years, resulting in a lack of practice in academic writing. Notably, past studies have shown that grant attainment rates of underrepresented individuals are lower than their majority counterparts. Here, we sought to consider a mechanism targeted to underrepresented individuals, although applicable to everyone, to help overcome traditional barriers to writing in STEMM. The authors have hosted a writing accountability group (WAG) that uniquely provides a format focused on physical activity and different forms of writing to strengthen both career development and award/funding attainment. Our objectives were to evaluate this unique format, thus creating a resource for individuals and institutions to learn about WAGs and expand upon the framework to formulate their own WAG. To do this, we performed a small pilot study (n = 21) to investigate attitudes towards the WAG. We present the results of a survey conducted among underrepresented WAG participants, which spanned different career stages and was highly diverse demographically. Our results show that following attendance of our WAG, individuals did not note a significant change in scales pertaining to John Henryism (high-effort coping), resilience, sense of belonging, or grit. However, significant increases were noted in the self-perceived ability to handle stress, confidence in applying for awards, appreciation for mentoring, and satisfaction of WAGs. Taken together, the results of this study suggest that our unique WAG format can have some positive results as a career and writing development opportunity and may be able to support underrepresented individuals in attaining funding at higher education institutions.

The fluorescence viewing of mitochondria is commonly performed by MitoTracker, a lipophilic cationic dye that is taken up by the mitochondria. In this forum, we highlight several issues that may occur with MitoTracker, including staining of other organelles. Our aim is to offer alternative dyes and discuss their advantages and disadvantages. We also offer options for software with alternatives to MitoTracker to expedite future experimental design.

This review provides an overview of the current methods for quantifying mitochondrial ultrastructure, including cristae morphology, mitochondrial contact sites, and recycling machinery and a guide to utilizing electron microscopy to effectively measure these organelles. Quantitative analysis of mitochondrial ultrastructure is essential for understanding mitochondrial biology and developing therapeutic strategies for mitochondrial-related diseases. Techniques such as transmission electron microscopy (TEM) and serial block face-scanning electron microscopy, as well as how they can be combined with other techniques including confocal microscopy, super-resolution microscopy, and correlative light and electron microscopy are discussed. Beyond their limitations and challenges, we also offer specific magnifications that may be best suited for TEM analysis of mitochondrial, endoplasmic reticulum, and recycling machinery. Finally, perspectives on future quantification methods are offered.

ABSTRACT Bacterial cells can vary greatly in size, from a few hundred nanometers to hundreds of micrometers in diameter. Filamentous cable bacteria also display substantial size differences, with filament diameters ranging from 0.4 to 8 µm. We analyzed the genomes of cable bacterium filaments from 11 coastal environments of which the resulting 23 new genomes represent 10 novel species-level clades of Candidatus Electrothrix and two clades that putatively represent novel genus-level diversity. Fluorescence in situ hybridization with a species-level probe showed that large-sized cable bacteria belong to a novel species with the proposed name Ca. Electrothrix gigas. Comparative genome analysis suggests genes that play a role in the construction or functioning of large cable bacteria cells: the genomes of Ca. Electrothrix gigas encode a novel actin-like protein as well as a species-specific gene cluster encoding four putative pilin proteins and a putative type II secretion platform protein, which are not present in other cable bacteria. The novel actin-like protein was also found in a number of other giant bacteria, suggesting there could be a genetic basis for large cell size. This actin-like protein (denoted big bacteria protein, Bbp) may have a function analogous to other actin proteins in cell structure or intracellular transport. We contend that Bbp may help overcome the challenges of diffusion limitation and/or morphological complexity presented by the large cells of Ca. Electrothrix gigas and other giant bacteria. IMPORTANCE In this study, we substantially expand the known diversity of marine cable bacteria and describe cable bacteria with a large diameter as a novel species with the proposed name Candidatus Electrothrix gigas. In the genomes of this species, we identified a gene that encodes a novel actin-like protein [denoted big bacteria protein (Bbp)]. The bbp gene was also found in a number of other giant bacteria, predominantly affiliated to Desulfobacterota and Gammaproteobacteria, indicating that there may be a genetic basis for large cell size. Thus far, mostly structural adaptations of giant bacteria, vacuoles, and other inclusions or organelles have been observed, which are employed to overcome nutrient diffusion limitation in their environment. In analogy to other actin proteins, Bbp could fulfill a structural role in the cell or potentially facilitate intracellular transport.

Summary: Maximizing Access to Research Careers (MARC) programs are aimed to increase diversity in science, technology, engineering, math, and medicine (STEMM) fields. However, limited programs and eligibility requirements limit the students who may apply to similar programs. At Winston-Salem State University, we piloted a series of workshops, collectively termed Project Strengthen, to emulate some of the key aspects of MARC programs. Following the workshop, Project Strengthen students showed a significant increase in their understanding of essential educational development skills, such as writing personal statements, applying to graduate school, studying for the GRE, and seeking summer internships. This suggests Project Strengthen may be a potential lower cost comparable option than MARC to make up for current deficiencies in preparedness for graduate school. We also provide educational materials from Project Strengthen, including a clear framework for this seminar series, six ready-made PowerPoints to share with trainees that have been demonstrated to be effective.

Cable bacteria are centimeter-long filamentous microbes that conduct electrons via internal wires, thus coupling sulfide oxidation between sediment layers. Here, Bjerg et al. show that the anoxic part of oxygen-respiring cable bacteria attracts swarms of other bacteria, which appear to transfer electrons to cable bacteria via soluble metabolites.

Abstract Background Routine follow-up of patients hospitalised with COVID-19 is recommended, however due to the ongoing high number of infections this is not without significant health resource and economic burden. In a previous study we investigated the prevalence of, and risk factors for, persistent chest radiograph (CXR) abnormalities post-hospitalisation with COVID-19 and identified a 5-point composite score that strongly predicted risk of persistent CXR abnormality at 12-weeks. Here we sought to validate and refine our findings in an independent cohort of patients. Methodology A single-centre prospective study of consecutive patients attending a virtual post-hospitalisation COVID-19 clinic and CXR as part of their standard clinical care between 2nd March – 22nd June 2021. Inpatient and follow-up CXRs were scored by the assessing clinician for extent of pulmonary infiltrates (0–4 in each lung) with complete resolution defined as a follow-up score of zero. IntroductionSarcoidosis is a highly variable disease in terms of organ involvement, type of onset and course. Associations of genetic polymorphisms with sarcoidosis phenotypes have been observed and suggest genetic signatures.MethodsAfter obtaining a positive vote of the competent ethics committee we genotyped 1909 patients of the deeply phenotyped Genetic-Phenotype Relationship in Sarcoidosis (GenPhenReSa) cohort of 31 European centers in 12 countries with 116 potentially disease-relevant single-nucleotide polymorphisms (SNPs). Using a meta-analysis, we investigated the association of relevant phenotypes (acute vs. sub-acute onset, phenotypes of organ involvement, specific organ involvements, and specific symptoms) with genetic markers. Subgroups were built on the basis of geographical, clinical and hospital provision considerations.ResultsIn the meta-analysis of the full cohort, there was no significant genetic association with any considered phenotype after correcting for multiple testing. In the largest sub-cohort (Serbia), we confirmed the known association of acute onset with TNF and reported a new association of acute onset an HLA polymorphism. Multi-locus models with sets of three SNPs in different genes showed strong associations with the acute onset phenotype in Serbia and Lublin (Poland) demonstrating potential region-specific genetic links with clinical features, including recently described phenotypes of organ involvement.DiscussionThe observed associations between genetic variants and sarcoidosis phenotypes in subgroups suggest that gene–environment-interactions may influence the clinical phenotype. In addition, we show that two different sets of genetic variants are permissive for the same phenotype of acute disease only in two geographic subcohorts pointing to interactions of genetic signatures with different local environmental factors. Our results represent an important step towards understanding the genetic architecture of sarcoidosis.

Fire danger poses a pressing threat to ecosystems and societies worldwide. Adequate preparation and forewarning can help reduce these threats, but these rely on accurate prediction of extreme fire danger. With the knowledge that climatic conditions contribute heavily to overall fire danger, this study evaluates the skill with which episodes of extreme fire danger in Australia can be predicted from the activity of large-scale climate driver patterns. An extremal dependence index for extreme events is used to depict the historical predictive skill of the Australian Bureau of Meteorology’s subseasonal climate prediction system in replicating known relationships between the probability of top-decile fire danger and climate driver states at a lead time of 2–3 weeks. Results demonstrate that the El Niño Southern Oscillation, Southern Annular Mode, persistent modes of atmospheric blocking, Indian Ocean Dipole and Madden-Julian Oscillation are all key for contributing to predictability of fire danger forecasts in different regions during critical fire danger periods. Northwest Australia is found to be particularly predictable, with the highest mean index differences (>0.50) when certain climate drivers are active, compared with the climatological index mean. This integrated approach offers a valuable resource for decision-making in fire-prone regions, providing greater confidence to users relying on fire danger outlooks for key management decisions, such as those involved in the sectors of national park and forest estate management, agriculture, emergency services, health and energy. Furthermore, the results highlight strengths and weaknesses in both the Australian Fire Danger Rating System and the operational climate model, contributing additional information for improving and refining future iterations of these systems.

Diverse characteristics of El Niño Southern Oscillation (ENSO) events challenge the traditional view of tropical coupled ocean-atmosphere systems. The probability of a transition from one type of event to another is influenced by multiple factors of which many are projected to change. Here we assess the likelihood of ENSO transitions in observations and climate models, including a distinction between events that peak in the Eastern Pacific (EP) and Central Pacific (CP). We find that the initial ENSO state influences the likelihood of certain transitions and that some transitions are not physically possible or stochastically likely. For example, transitions to CP events are more likely than EP events except from a neutral state. We also find that El Niños tend to occur as singular events compared to La Niñas. While consecutive El Niño and La Niña events of EP type are possible, opposing EP events do not occur in succession. We identify several transitions likely driven by internal dynamical processes including neutral conditions to El Niño, CP El Niño to another El Niño, EP El Niño to CP La Niña, CP La Niña to CP El Niño and La Niña, and EP La Niña to neutral and CP El Niño. Projections of future transitions show an increased probability of transitions to CP El Niño events while transitions to EP La Niña events become less frequent under a high-emissions scenario. Accordingly, transitions to these events become more and less likely, respectively. We also find changes in the likelihood of specific transitions in a warming world: consecutive CP El Niño events become more likely while EP El Niño events become less likely to transition into CP La Niña events. These changes are expected to occur as early as 2050 with some changes to be accelerated by the end of the 21st century.

Abstract Sericea lespedeza (Lespedeza cuneata) is an invasive nonnative legume that commonly occurs on sites managed for northern bobwhite (Colinus virginianus). Sericea reduces bobwhite habitat availability by outcompeting native plants that provide food and cover. Sericea can be controlled with postemergence herbicides such as glyphosate as well as a mixture of triclopyr with fluroxypyr, but these herbicides do not provide preemergence control. Imazapic has been effective at controlling a limited number of planted sericea seeds, but the efficacy of imazapic at various application rates to control sericea seedbank response following treatment with a postemergence herbicide has not been evaluated. We designed a field experiment to evaluate sericea control and changes in the plant community as related to bobwhite habitat with 2 postemergence herbicides followed by preemergence treatment of imazapic at 3 sites in Tennessee and Alabama, USA, 2018–2022. Specifically, we treated sericea with postemergence broadcast applications of glyphosate or triclopyr with fluroxypyr in 2018 and applied spot treatment with the same herbicides in 2019–2021. We applied imazapic at 4 rates following prescribed fire in 2019 and 2022. We measured coverage of sericea, native forbs, annual grasses, perennial grasses, and bobwhite food plants before treatment in 2018 and after all treatments in 2022. Nearly all postemergence treatments reduced sericea coverage, but imazapic did not increase control compared to postemergence herbicides alone. Perennial grasses used for nesting and bobwhite foraging were maintained following treatments. We recommend managers use either glyphosate or triclopyr with fluroxypyr to control sericea postemergence and consider using a low rate of imazapic to reduce annual grasses if coverage is a problem on the site.

IntroductionMany studies in mice have demonstrated that cardiac-specific innate immune signaling pathways can be reprogrammed to modulate inflammation in response to myocardial injury and improve outcomes. While the echocardiography standard parameters of left ventricular (LV) ejection fraction, fractional shortening, end-diastolic diameter, and others are used to assess cardiac function, their dependency on loading conditions somewhat limits their utility in completely reflecting the contractile function and global cardiovascular efficiency of the heart. A true measure of global cardiovascular efficiency should include the interaction between the ventricle and the aorta (ventricular-vascular coupling, VVC) as well as measures of aortic impedance and pulse wave velocity.MethodsWe measured cardiac Doppler velocities, blood pressures, along with VVC, aortic impedance, and pulse wave velocity to evaluate global cardiac function in a mouse model of cardiac-restricted low levels of TRAF2 overexpression that conferred cytoprotection in the heart.ResultsWhile previous studies reported that response to myocardial infarction and reperfusion was improved in the TRAF2 overexpressed mice, we found that TRAF2 mice had significantly lower cardiac systolic velocities and accelerations, diastolic atrial velocity, aortic pressures, rate-pressure product, LV contractility and relaxation, and stroke work when compared to littermate control mice. Also, we found significantly longer aortic ejection time, isovolumic contraction and relaxation times, and significantly higher mitral early/atrial ratio, myocardial performance index, and ventricular vascular coupling in the TRAF2 overexpression mice compared to their littermate controls. We found no significant differences in the aortic impedance and pulse wave velocity.DiscussionWhile the reported tolerance to ischemic insults in TRAF2 overexpression mice may suggest enhanced cardiac reserve, our results indicate diminished cardiac function in these mice.
