Abstract The hypothalamus senses the appetite-regulating hormones and also coordinates the metabolic function in alignment with the circadian rhythm. This alignment is essential to maintain the physiological conditions that prevent clinically important comorbidities, such as obesity or type-2 diabetes. However, a complete model of the hypothalamus that relates food intake with circadian rhythms and appetite hormones has not yet been developed. In this work, we present a computational model that accurately allows interpreting neural activity in terms of hormone regulation and sleep-wake cycles. We used a conductance-based model, which consists of a system of four differential equations that considers the ionotropic and metabotropic receptors, and the input currents from homeostatic hormones. We proposed a logistic function that fits available experimental data of insulin hormone concentration and added it into a short-term ghrelin model that served as an input to our dynamical system. Our results show a double oscillatory system, one synchronized by light-regulated sleep-wake cycles and the other by food-regulated feeding-fasting cycles. We have also found that meal timing frequency is highly relevant for the regulation of the hypothalamus neurons. We therefore present a mathematical model to explore the plausible link between the circadian rhythm and the endogenous food clock.

Abstract From Darwin’s visit to nowadays, we analyzed the changes in species- and genus-level richness and turnover, equity, diversity, composition, as well as functional group and functional strategy dynamics over a 213-year historical series of the seaweed assemblage from Rio de Janeiro Bay. We developed new analysis methods to serve as a protocol for worldwide functional and bioindicator assessment of seaweed assemblages. Over the two centuries of changes tracked, the integration of taxonomic, functional group, and functional strategy analyses unraveled a trajectory of seaweed assemblage restructuring under human disturbance. Our taxonomic analyses showed a high species and genus turnover and a decline in richness, with the local extinction of the Sargassum genus, responsible for warm temperate kelp forests in the Western Atlantic. Seaweed functional group data presented a trend of morphological simplification and miniaturization, with the decline of marine forest canopy and subcanopy-forming groups favoring filamentous and crustose algae dominance. Functional strategy data showed the historical decline of competitive structuring as an assemblage driver in favor of disturbance and stress-structuring, as highlighted by the competitive strategy decrease, coupled with stress-tolerant and ruderal strategies rise across metrics. These changes were successfully portrayed by the new ecological index we proposed, the C/SR index. It is the first quantitative index for seaweed assemblages, designed to be a trackable metric for conservation, monitoring, and restoration efforts.

Abstract Seaweeds are the most phylogenetically diverse group of multicellular organisms and rank foremost among marine keystone species. Due to their taxonomic diversity and functional importance, previous studies have classified seaweeds into functional groups based on qualitative or semi-quantitative traits, such as seaweed form, anatomy, and thickness. Despite the widespread use of seaweed functional groups from basic marine ecology to coastal monitoring, it is not known how accurate such morphology-based proposals are in grouping seaweeds by their form. To address this uncertainty at the foundations of seaweed biology, we surveyed and gathered all available data on seaweed forms using PRISMA protocols. We used the surface area to volume ratio (SA:V), a quantitative and universal measure of seaweed form, to assess the distribution and diversity of seaweed morphology across 99 species from three phyla. We show that seaweed surface area to volume ratio values span 3.64 orders of magnitude and follow a continuous and exponential distribution, without any significant gaps or clusters. We also tested current functional group schemes based on morphology and anatomy and showed that only 30% to 38% of their groups showed any significant pairwise differences in morphology. Our results challenge the basis of the current functional group approach in seaweed biology and suggest that a trait-based framework based on quantitative and continuous measures of seaweed form could provide a simpler and more accurate alternative to functionally assess seaweed ecology and physiology, as well as its implications for coastal ecosystem management.

A 2018 report from the Water Quality for Human Consumption Vigilance Information System (SISÁGUA, Brazil) showed the presence of 27 pesticides in Brazilian drinking water, of which 11 have carcinogenic potential. We assessed the data for 27 municipalities in Paraná state southwest, a rural landscape with high cancer rates. We selected data from the carcinogenic potential of 11 pesticides provided by international agencies (alachlor, aldrin-diheldrin, atrazine, chlordane, DDT, diuron, glyphosate, lindane, mancozeb, molinate, and trifluralin) and estimated the number of cancer cases attributable to drinking water contamination by pesticides. Also, we correlated such findings with incidence and mortality cancer rates for ten topographies obtained from the Brazilian National Cancer Institute (INCA) database. A total of 9 cities were selected, corresponding to about 81,000 people. All towns had all pesticides quantified in the drinking water. About ten cancer cases were attributed to drinking water contamination by pesticides in 2014–2017, mainly linked to diuron and mancozeb. Concerning the consolidated incidence of cancer cases reported by the INCA, significant correlations were found regarding aldrin-diheldrin, alachlor, and atrazine for breast cancer, atrazine for prostate cancer, and mancozeb and diuron for colon cancer, among others. Regarding the consolidated mortality rates, some correlations were found between DDT and trifluralin for the breast, DDT and lindane for the prostate, and glyphosate for lung cancer. Moderate correlations were found between the estimated and consolidated cancer cases for several topographies. Our findings highlight the correlation between drinking water contamination in Paraná state southwest and its increased incidence of cancers with poor prognosis.

High-voltage power line insulators are crucial for safe and efficient electricity transmission. However, real-world image limitations, particularly regarding dirty insulator strings, delay the development of robust algorithms for insulator inspection. This dataset addresses this challenge by creating a novel synthetic high-voltage power line insulator image database.The database was created using computer-aided design softwares and a game development engine. Publicly available CAD models of high-voltage towers with the most common insulator types (polymer, glass, and porcelain) were imported into the game engine. This virtual environment allowed for the generation of a diverse dataset by manipulating virtual cameras, simulating various lighting conditions, and incorporating different backgrounds such as mountains, forests, plantation, rivers, city and deserts.The database comprises two main sets: The Image Segmentation Set, which includes 47,286 images categorized by insulator material (ceramic, polymeric, and glass) and landscape type (mountains, forests, plantation, rivers, city and deserts). Moreover, the Image Classification Set that contains 14,424 images simulating common insulator string contaminants: salt, soot, bird excrement, and clean insulators. Each contaminant category has 3,606 images divided into 1,202 images per insulator type.This synthetic database offers a valuable resource for training and evaluating machine learning algorithms for high-voltage power line insulator inspection, ultimately contributing to enhanced power grid maintenance and reliability.

A low-fermentable oligo-, di-, monosaccharide and polyol (low-FODMAP) diet for patients with irritable bowel syndrome (IBS) should include an adequate fiber source. Our aim was to formulate orange cookies using maize flour and type-4 resistant starch (RS4) from maize and to evaluate their properties and sensorial attributes by IBS patients. We prepared two formulations: 37.7% RS4 and 14.7% maize flour and a control with normal maize starch (MS) instead of RS4. We added orange juice and zest instead of water and evaluated their properties. The viscosity, water absorption capacity, and solubility were lower for RS4 than for MS. The width, thickness, L* and a* values of both cookies were comparable (p > 0.05), although RS4-C had a decreased b* value and higher hardness (90.6 vs. 80.1 N). The nutrient content was similar between RS4-C and MS-C, but the glycemic index of RS4-C was 63 compared to 95 of MS-C. According to IBS patients, the appearance, taste, hardness, overall quality, and perception of healthiness and nutritional value of both types of cookies were similarly high (p > 0.05). Panelists recommend the cookies. Therefore, RS4 cookies could be further investigated for their ability to improve bowel habits and re-equilibrate the microbiota of IBS patients.

Functional neuroimaging methods like fMRI and PET are vital in neuroscience research, but require that subjects remain still throughout the scan. In animal research, anesthetic agents are typically applied to facilitate the acquisition of high-quality data with minimal motion artifact. However, anesthesia can have profound effects on brain metabolism, selectively altering dynamic neural networks and confounding the acquired data. To overcome the challenge, we have developed a novel head fixation device designed to support awake rat brain imaging. A validation experiment demonstrated that the device effectively minimizes animal motion throughout the scan, with mean absolute displacement and mean relative displacement of 0.0256 (SD: 0.001) and 0.009 (SD: 0.002), across eight evaluated subjects throughout fMRI image acquisition (total scanning time per subject: 31 min, 12 s). Furthermore, the awake scans did not induce discernable stress to the animals, with stable physiological parameters throughout the scan (Mean HR: 344, Mean RR: 56, Mean SpO2: 94 %) and unaltered serum corticosterone levels (p = 0.159). In conclusion, the device presented in this paper offers an effective and safe method of acquiring functional brain images in rats, allowing researchers to minimize the confounding effects of anesthetic use.

Abstract The SARS-CoV-2 Omicron subvariant BA.5 rapidly spread worldwide and replaced BA.1/BA.2 in many countries, becoming globally dominant. BA.5 has unique amino acid substitutions in the spike protein that both mediate immune escape from neutralizing antibodies produced by immunizations and increase ACE2 receptor binding affinity. In a comprehensive, long-term (up to 9 months post primary vaccination), experimental vaccination study using male Syrian hamsters, we evaluate neutralizing antibody responses and efficacy against BA.5 challenge after primary vaccination with Ad26.COV2.S (Janssen) or BNT162b2 (Pfizer/BioNTech) followed by a homologous or heterologous booster with mRNA-1273 (Moderna) or NVX-CoV2373 (Novavax). Notably, one high or low dose of Ad26.COV2.S provides more durable immunity than two primary doses of BNT162b2, and the NVX-CoV2373 booster provides the strongest augmentation of immunity, reduction in BA.5 viral replication, and disease. Our data demonstrate the immunogenicity and efficacy of different prime/boost vaccine regimens against BA.5 infection in an immune-competent model and provide new insights regarding COVID-19 vaccine strategies.

Intestinal permeability and bacterial translocation are increased in obesity and metabolic syndrome (MS). ILC3 cells contribute to the integrity of intestinal epithelium by producing IL-22 via IL-1β and IL-23. This study investigates the role of IL-1R1 in inducing ILC3 cells and conferring protection during obesity and MS. For this purpose, C57BL/6 wild-type (WT) and IL-1R1-deficient mice were fed a standard diet (SD) or high-fat diet (HFD) for 16 weeks. Weight and blood glucose levels were monitored, and adipose tissue and blood samples were collected to evaluate obesity and metabolic parameters. The small intestine was collected to assess immunological and junction protein parameters through flow cytometry and RT-PCR, respectively. The intestinal permeability was analyzed using the FITC-dextran assay. The composition of the gut microbiota was also analyzed by qPCR. We found that IL-1R1 deficiency exacerbates MS in HFD-fed mice, increasing body fat and promoting glucose intolerance. A worsening of MS in IL-1R1-deficient mice was associated with a reduction in the ILC3 population in the small intestine. In addition, we found decreased IL-22 expression, increased intestinal permeability and bacterial translocation to the visceral adipose tissue of these mice compared to WT mice. Thus, the IL-1R1 receptor plays a critical role in controlling intestinal homeostasis and obesity-induced MS, possibly through the differentiation or activation of IL-22-secreting ILC3s.

Control charts are tools of paramount importance in statistical process control. They are broadly applied in monitoring processes and improving quality, as they allow the detection of special causes of variation with a significant level of accuracy. Furthermore, there are several strategies able to be employed in different contexts, all of which offer their own advantages. Therefore, this study focuses on monitoring the variability in univariate processes through variance using the Binomial version of the ATTRIVAR Same Sample S2 (B-ATTRIVAR SS S2) control chart, given that it allows coupling attribute and variable inspections (ATTRIVAR means attribute + variable), i.e., taking advantage of the cost-effectiveness of the former and the wealth of information and greater performance of the latter. Its Binomial version was used for such a purpose, since inspections are made using two attributes, and the Same Sample was used due to being submitted to both the attribute and variable stages of inspection. A computational application was developed in the R language using the Shiny package so as to create an interface to facilitate its application and use in the quality control of the production processes. Its application enables users to input process parameters and generate the B-ATTRIVAR SS control chart for monitoring the process variability with variance. By comparing the data obtained from its application with a simpler code, its performance was validated, given that its results exhibited striking similarity.

Barred galaxies often develop a box/peanut pseudobulge, but they can also host a nearly spherical classical bulge, which is known to gain rotation due to the bar. We aim to explore how the presence of gas impacts the rotation of classical bulges. We carried out a comprehensive set of hydrodynamical N-body simulations with different combinations of bulge masses and gas fractions. In these models, both massive bulges and high gas content tend to inhibit the formation of strong bars. For low-mass bulges, the resulting bar is stronger in cases of low gas content. In the stronger bar models, bulges acquire more angular momentum and thus display considerable rotational velocity. Such bulges also develop anisotropic velocity dispersions and become triaxial in shape. We found that the rotation of the bulge becomes less pronounced as the gas fraction is increased from 0 to 30%. These results indicate that the gas content has a significant effect on the dynamics of the classical bulge, because it influences bar strength. Particularly in the case of the low-mass bulges (10% bulge mass fraction), all of the measured rotational and structural properties of the classical bulge depend strongly and systematically on the gas content of the galaxy.

Barred galaxies constitute about two-thirds of observed disc galaxies. Bars affect not only the mass distribution of gas and stars but also that of the dark matter. An elongation of the inner dark matter halo is known as the halo bar. We aim to characterize the structure of the halo bars, with the goal of correlating them with the properties of the stellar bars. We use a suite of simulated galaxies with various bar strengths, including gas and star formation. We quantify the strengths, shapes, and densities of these simulated stellar bars. We carry out numerical experiments with frozen and analytic potentials in order to understand the role played by a live responsive stellar bar. We find that the halo bar generally follows the trends of the disc bar. The strengths of the halo and stellar bars are tightly correlated. Stronger bars induce a slight increase in dark matter density within the inner halo. Numerical experiments show that a non-responsive frozen stellar bar would be capable of inducing a dark matter bar, but it would be weaker than the live case by a factor of roughly two.

IntroductionThe therapeutic efficacy of deep brain stimulation (DBS) of the subthalamic nucleus (STN) for Parkinson’s disease (PD) may be limited for some patients by the presence of stimulation-related side effects. Such effects are most often attributed to electrical current spread beyond the target region. Prior computational modeling studies have suggested that changing the degree of asymmetry of the individual phases of the biphasic, stimulus pulse may allow for more selective activation of neural elements in the target region. To the extent that different neural elements contribute to the therapeutic vs. side-effect inducing effects of DBS, such improved selectivity may provide a new parameter for optimizing DBS to increase the therapeutic window.MethodsWe investigated the effect of six different pulse geometries on cortical and myogenic evoked potentials in eight patients with PD whose leads were temporarily externalized following STN DBS implant surgery. DBS-cortical evoked potentials were quantified using peak to peak measurements and wavelets and myogenic potentials were quantified using RMS.ResultsWe found that the slope of the recruitment curves differed significantly as a function of pulse geometry for both the cortical- and myogenic responses. Notably, this effect was observed most frequently when stimulation was delivered using a monopolar, as opposed to a bipolar, configuration.DiscussionManipulating pulse geometry results in differential physiological effects at both the cortical and neuromuscular level. Exploiting these differences may help to expand DBS’ therapeutic window and support the potential for incorporating pulse geometry as an additional parameter for optimizing therapeutic benefit.

Introduction: One of the most critical complications in myelodysplastic syndromes (MDS) is the progression to acute myeloid leukemia (AML). The dynamics of clonal evolution in MDS and how acquired mutations can be used as biomarkers to track disease progression remains under investigation. Objective and method: Herein, we investigated the frequency of common myeloid clonal mutations (FLT3, NPM1, JAK2, IDH1 and IDH2) in 88 patients with MDS and 35 AML patients with myelodysplasia-related changes, followed at a single reference center in northeastern Brazil. Results: Overall, 9/88 (10%) of the MDS patients and 9/35 (26%) of the secondary AML patients had at least one mutation. While the JAK2 V617F mutation was the most frequent in the MDS patients, the FLT3, NPM1, IDH1 and IDH2 mutations were more frequently found in the secondary AML group. Furthermore, there was a higher frequency of FLT3, NPM1, IDH1 and IDH2 mutations in MDS patients classified as high-risk subtypes than in those of lower risk. Conclusion: Despite the limited sample size, our data suggest that mutations in FLT3, NPM1, IDH1 and IDH2 genes could be potential biomarkers to detect early disease progression in MDS.

Abell 1758 ( z ∼ 0.278) is a galaxy cluster composed of two structures, A1758N and A1758S, separated by ∼2.2 Mpc. The northern cluster is itself a dissociative merging cluster that has already been modeled by dedicated simulations. Recent radio observations revealed the existence of a previously undetected bridge connecting A1758N and A1758S. New simulations are now needed to take into account the presence of A1758S. We wish to evaluate which orbital configuration would be compatible with a bridge between the clusters. Using N -body hydrodynamical simulations that build upon the previous model, we explore different scenarios that could have led to the current observed configuration. Five types of orbital approaches were tested: radial, tangential, vertical, postapocentric, and outgoing. We found that the incoming simulated scenarios are generally consistent with mild enhancements of gas density between the approaching clusters. The mock X-ray images exhibit a detectable bridge in all cases. Compared to measurements of Chandra data, the amplitude of the X-ray excess is overestimated by a factor of ∼2–3 in the best simulations. The scenario of tangential approach proved to be the one that best matches the properties of the profiles of X-ray surface brightness. The scenarios of radial approach of vertical approach are also marginally compatible.

Abstract This work presents a systematic theoretical analysis and experimental validation of a novel coherent absorber which is printed through conventional inkjet-printing technology. The new absorber consists of a single resistively loaded sheet printed on a conventional plastic sheet, resulting a low complexity and passive design. The low-cost and easily fabricated absorber is frequency independent, polarization insensitive, wide-angle and we demonstrate its absorbance reconfigurability using a remote illumination as a control signal. Theoretical, numerical and experimental results are in good agreement. Specifically, experimental results shown that near perfect absorption (i.e., $$100\%$$ 100 % ) can be achieved using a printed sheet of thickness $$\lambda /215$$ λ / 215 .

The SARS-CoV-2 variant of concern Omicron has quickly spread. Here, Zou et al. develop a high-throughput neutralization test for Omicron SARS-CoV-2 and show that patients with previous non-Omicron infections do not develop robust neutralization against Omicron.

Statistical process control (SPC) is one of the most powerful techniques for improving quality, as it is able to detect special causes of problems in processes, products and services with a remarkable degree of accuracy. Among SPC tools, X¯ and R control charts are widely employed in process monitoring. However, scenarios involving vague, imprecise and even subjective data require a type-2 fuzzy set approach. Thus, X¯ and R control charts should be coupled with interval type-2 triangular fuzzy numbers (IT2TFN) in order to add further information to traditional control charts. This paper proposes a performance analysis of IT2TFN and X¯ and R control charts by means of average run length (ARL), standard deviation of the run length (SDRL) and RL percentile. Computer simulations were carried out considering 10,000 runs to obtain ARL, SDRL and the 5th, 25th, 50th, 75th and 95th RL percentiles. Simulation results reveal that the proposed control charts increased fault detection capability (speed of response) and slightly reduced the number of false alarms in processes under control. Moreover, it was observed that, in addition to superior performance, IT2TFN X¯-R control charts proved to be more robust and flexible when compared to traditional control charts.

Founded on a circular economy perspective, the possible effect of targeted supplementation with leftover feed from dairy cows (i.e., intensive system) upon the productive economic performance of crossbred–rangeland goats (i.e., extensive system) in northern arid Mexico was assessed. Multiparous goats (n = 38) with similar body condition score (BCS) and body weight (BW) were randomly assigned during the deep anestrus season (i.e., March–April, 25° N) into two groups: (1) the control-non-supplemented group (CONT; n = 19; BCS: 1.76 ± 0.06; BW: 44.3 ± 2.5 kg) and (2) the supplemented group (SUPL; n = 19; BCS: 1.76 ± 0.07; BW: 43.7 ± 1.8 kg). While the SUPL group received 400 g goat d−1 of dairy cow feed leftovers prior to grazing, both groups went daily to the rangeland (i.e., ≈8 h). The study considered an experimental period of 36 d with an experimental breeding of 11 d (d0–d10). Previously, on days −20, −10, −1 preceding the male-to-female interaction, the anovulatory status of goats was confirmed through ultrasonographic scanning. Prior to mating, the males were separated from goats and treated for a period of 3 weeks (i.e., every 3rd d) with testosterone (i.e., 50 mg i.m.). The response variables evaluated considered goats induced to estrus (GIE, %), goats ovulating (GO, %), ovulation rate (OR, units), pregnancy rate-1 (PRd36, %), pregnancy rate-2 (PRd50, %), embryo mortality-d50 (EMO, %), potential kidding index-d50 (PKId50,%), kid weight at birth simples (KWBS, kg), potential litter efficiency at birth (PLEB, kg), and potential litter efficiency at weaning (i.e., d21 post kidding), either expressed as kg head−1 (PLEW1) or USD head−1 (PLEW2). Although no differences (p > 0.05) occurred for GIE and PRd50, increases in the phenotypic expression of OR (1.42 vs. 0.73), PRd36 (68.4 vs. 36.8), EMO (23.0 vs. 0), PKId50 (74.7 vs. 26.8), and KWBS (4.1 vs. 3.3) occurred (p < 0.05) in the SUPL goats. To have a more integrative evaluation regarding the out-of-season reproductive outcomes, three bio-economic efficiency indices per goat exposed to males were generated: PLEB (4.3 vs. 0.6 kg), PLEW1-kg (7.7 vs. 3.1 kg), and PLEW2-USD (63.9 vs. 26.1 USD), which also favored (p < 0.05) to the SUPL goats. The last result occurred despite no differences (p > 0.05) regarding BW, BCS, and serum glucose concentrations between experimental groups. Furthermore, applying the main research outcomes from this specific study toward the large-scale goat production system in the Comarca Lagunera—one of the largest dairy goat production hubs in The Americas—denoted promising expectations, either from an economic or productive–reproductive standpoint. Certainly, goat producers from the region would increase their potential annual income just from the sale of kids by close to 250%; that is from MUSD 1.1 to 3.9. This result should reduce food insecurity and economic stress, as well as enhance the livelihoods of the goat keepers and their families.

Abstract Antenna arrays and multi-antenna systems are essential in beyond 5G wireless networks for providing wireless connectivity, especially in the context of Internet-of-Everything. To facilitate this requirement, beamforming technology is emerging as a key enabling solution for adaptive on-demand wireless coverage. Despite digital beamforming being the primary choice for adaptive wireless coverage, a set of applications rely on pure analogue beamforming approaches, e.g., in point-to-multi point and physical-layer secure communication links. In this work, we present a novel scalable analogue beamforming hardware architecture that is capable of adaptive 2.5-dimensional beam steering and beam shaping to fulfil the coverage requirements. Beamformer hardware comprises of a finite size Maxwell fisheye lens used as a scalable feed network solution for a semi-circular array of monopole antennas. This unique hardware architecture enables a flexibility of using 2 to 8 antenna elements. Beamformer development stages are presented while experimental beam steering and beam shaping results show good agreement with the estimated performance.

The impact of pulse timing is an important factor in our understanding of how to effectively modulate the basal ganglia thalamocortical (BGTC) circuit. Single pulse low-frequency DBS-evoked potentials generated through electrical stimulation of the subthalamic nucleus (STN) provide insight into circuit activation, but how the long-latency components change as a function of pulse timing is not well-understood. We investigated how timing between stimulation pulses delivered in the STN region influence the neural activity in the STN and cortex. DBS leads implanted in the STN of five patients with Parkinson's disease were temporarily externalized, allowing for the delivery of paired pulses with inter-pulse intervals (IPIs) ranging from 0.2 to 10 ms. Neural activation was measured through local field potential (LFP) recordings from the DBS lead and scalp EEG. DBS-evoked potentials were computed using contacts positioned in dorsolateral STN as determined through co-registered post-operative imaging. We quantified the degree to which distinct IPIs influenced the amplitude of evoked responses across frequencies and time using the wavelet transform and power spectral density curves. The beta frequency content of the DBS evoked responses in the STN and scalp EEG increased as a function of pulse-interval timing. Pulse intervals 4 ms produced modest, but non-significant growth. Beta frequency activity in the scalp EEG and STN LFP response was maximal when IPIs were between 1.5 and 4.0 ms. These results demonstrate that long-latency components of DBS-evoked responses are pre-dominantly in the beta frequency range and that pulse interval timing impacts the level of BGTC circuit activation.

ABSTRACT Objectives: To evaluate the impact of COVID-19 on clinical and surgical practice, educational activities, health and lifestyle behavior of Brazilian urology residents. Materials and Methods: A web-based survey was sent to 468 Brazilian urology residents from postgraduate years (PGY) 3 to 5 to collect data on clinical practice and training after 4 months of COVID-19. We also assessed health-related and behavior changes, rate of infection by SARS-CoV-2, deployment to the front line of COVID-19, residents’ concerns, and access to personal protective equipment (PPE). Results: Massive reductions in elective and emergency patient consultations, diagnostic procedures and surgeries were reported across the country, affecting PGY 3 to 5 alike. Most in-person educational activities were abolished. The median damage to the urological training expected for 2020 was 6.0 [3.4 – 7.7], on a scale from 0 to 10, with senior residents estimating a greater damage (P< 0.001). Educational interventions developed included online case-based discussions, subspeciality conferences and lectures, and grand rounds. Most senior residents favored extending residency to compensate for training loss and most younger residents favored no additional training (p< 0.001). Modifications in health and lifestyle included weight gain (43.8%), reduced physical activity (68.6%), increased alcoholic intake (44.9%) and cigarette consumption (53.6%), worsening of sexual life (25.2%) and feelings of sadness or depression (48,2%). Almost half were summoned to work on the COVID-19 front-line and 24.4% had COVID-19. Most residents had inadequate training to deal with COVID-19 patients and most reported a shortage of PPE. Residents’ concerns included the risk of contaminating family members, being away from residency program, developing severe COVID-19 and overloading colleagues. Conclusions: COVID-19 had a massive impact in Brazilian urology residents´ training, health and lifestyle behavior, which may reflect what happened in other medical specialties. Studies should confirm these findings to help developing strategies to mitigate residents’ losses.

No-till (NT) farming is a major soil conservation strategy. In the Brazilian Cerrado, soybean is the main grain crop, and strategies that improve the cultivation of cover crops in the off-season in succession to soybean could facilitate NT for large cultivated areas. In the municipality of Nova Xavantina, MT, Brazil, three experiments were carried out in a randomized block design with four replications. This work aimed to evaluate the establishment of Urochloa ruziziensis (ruzigrass) in succession to soybean as a function of ruzigrass seed treatment, sowing density and phenological stage, and soybean cultivar allelopathic potential. In the first experiment, treatment of ruzigrass seeds with six doses (0, 150, 300, 450, 600 and 750 mL of commercial product 100 kg−1 of seeds) of fipronil + thiophanate-methyl + pyraclostrobin was evaluated. In the second experiment, two ruzigrass interseeding periods (soybean phenological stages R5.5 and R7) and five ruzigrass seed densities (4.3, 6.4, 8.6, 10.7 and 12.9 kg ha−1 VPS) were tested. In the third experiment, the interference of soybean cultivars in the establishment of ruzigrass and the potential allelopathic effect of the cultivars on forage germination were evaluated, six soybean cultivars RR (Roundup Ready®) were used: P98Y70; TMG 115; TMG 132; M 8527; P98Y11; TMG 123. Our results showed that seed treatment facilitated forage establishment in the interseeding system, and the use of high sowing rates at phenological stage R5.5 of soybean promoted greater ruzigrass plant development. The effects of the soybean cultivars on ruzigrass germination varied, suggesting allelopathic effects of the deposition of petioles and soybean leaves on ruzigrass seeds. Thus, an appropriate interseeding strategy can improve forage production in the off-season and favor NT through correct management of forage establishment in succession to soybean.

Based on a circular economy approach, we evaluated the possible effect of targeted supplementation with leftover feed from dairy cows (i.e., intensive system) on the reproductive performance of crossbred/rangeland goats (i.e., extensive system) in arid Northern Mexico. During the deep-anestrous season (i.e., March–April, 25° North), multiparous goats (n = 38) with a similar body weight (BW) and body condition score (BCS) were randomly assigned to two groups: (1) supplemented group (SG; n = 19; BCS: 1.76 ± 0.07; BW: 43.7 ± 1.8 kg), receiving 400 g goat d−1 of dairy-cow-feed leftovers prior to grazing; and (2) non-supplemented group (NS; n = 19; BCS: 1.76 ± 0.06; BW: 44.3 ± 2.5 kg). Both groups were directed to the rangeland for a period of ≈8 h. While the experimental period lasted 36 d, the experimental breeding considered 11 d (d0–d10). The anovulatory status of goats was ultrasonographically confirmed on days −20, −10, and −1 prior to male–female interaction. Previously, bucks were separated for 3 weeks from the experimental females and received exogenous testosterone every third day (i.e., 50 mg i.m.) prior to mating. With respect to the response variables, namely BW, BCS, blood glucose levels (BGLs), estrus induction (GIE, %), latency to estrus (LTE, h), estrus duration (ED, h), and luteal tissue volume (LTV, mm), no differences (p > 0.05) occurred between experimental groups. However, the response variables, namely goats ovulating (GO, %; 78.9 vs. 47.3), total number of corpuses luteum (TNCL, n; 27 vs. 13), ovulation rate (OR, n; 1.42 vs. 0.73), multiple ovulation (MO, %; 73.3 vs. 55.5), and pregnancy rate on d 36 (PRd36, %, 68.4 vs. 36.8), favored (p < 0.05) the SG over the NS goats. Our results demonstrate that connecting the circularity of two divergent ruminant production systems (i.e., cow-intensive and goat-extensive) by using dairy cows’ feed leftovers as a targeted supplementation strategy in anestrous goats under a marginal-rangeland production system enhanced out-of-season reproductive outcomes (i.e., ovulation rate and pregnancy rate), thus benefiting marginal goat producers and their families.

ABSTRACT Influenza A virus (IAV) infections are a leading cause of mortality worldwide. Excess mortality during IAV epidemics and pandemics is attributable to secondary bacterial infections, particularly pneumonia caused by Streptococcus pneumoniae. Resident alveolar macrophages (rAMs) are early responders to respiratory infections that coordinate initial host defense responses. Maresin conjugates in tissue regeneration (MCTRs) are recently elucidated cysteinyl maresins that are produced by and act on macrophages. Roles for MCTRs in responses to respiratory infections remain to be determined. Here, IAV infection led to transient decreases in rAM numbers. Repopulated lung macrophages displayed transcriptional alterations 21 days post-IAV with prolonged susceptibility to secondary pneumococcal infection. Administration of a mix of MCTR1 to 3 or MCTR3 alone post-IAV decreased lung inflammation and bacterial load 48 and 72 h after secondary pneumococcal infection. MCTR-exposed rAMs had increased migration and phagocytosis of Streptococcus pneumoniae, reduced secretion of CXCL1, and a reversion toward baseline levels of several IAV-induced pneumonia susceptibility genes. Together, MCTRs counter regulated post-IAV changes in rAMs to promote a rapid return of bacteria host defense. IMPORTANCE Secondary bacterial pneumonia is a serious and common complication of IAV infection, leading to excess morbidity and mortality. New host-directed approaches are needed to complement antibiotics to better address this important global infectious disease. Here, we show that harnessing endogenous resolution mechanisms for inflammation by exogenous administration of a family of specialized proresolving mediators (i.e., cys-MCTRs) increased macrophage resilience mechanisms after IAV to protect against secondary infection from Streptococcus pneumoniae.

Deep brain stimulation (DBS) is a well-established therapy for the motor symptoms of Parkinson’s disease (PD), but there remains an opportunity to improve symptom relief. The temporal pattern of stimulation is a new parameter to consider in DBS therapy, and we compared the effectiveness of Temporally Optimized Patterned Stimulation (TOPS) to standard DBS at reducing the motor symptoms of PD. Twenty-six subjects with DBS for PD received three different patterns of stimulation (two TOPS and standard) while on medication and using stimulation parameters optimized for standard DBS. Side effects and motor symptoms were assessed after 30 min of stimulation with each pattern. Subjects experienced similar types of side effects with TOPS and standard DBS, and TOPS were well-tolerated by a majority of the subjects. On average, the most effective TOPS was as effective as standard DBS at reducing the motor symptoms of PD. In some subjects a TOPS pattern was the most effective pattern. Finally, the TOPS pattern with low average frequency was found to be as effective or more effective in about half the subjects while substantially reducing estimated stimulation energy use. TOPS DBS may provide a new programing option to improve DBS therapy for PD by improving symptom reduction and/or increasing energy efficiency. Optimizing stimulation parameters specifically for TOPS DBS may demonstrate further clinical benefit of TOPS DBS in treating the motor symptoms of Parkinson’s disease.

Parkinson’s disease is a neurological disease with cardinal motor signs including bradykinesia and tremor. Although beta-band hypersynchrony in the cortico-basal ganglia network is thought to contribute to disease manifestation, the resulting effects on network connectivity are unclear. We examined local field potentials from a non-human primate across the naïve, mild, and moderate disease states (model was asymmetric, left-hemispheric dominant) and probed power spectral density as well as cortico-cortical and cortico-subthalamic connectivity using both coherence and Granger causality, which measure undirected and directed effective connectivity, respectively. Our network included the left subthalamic nucleus (L-STN), bilateral primary motor cortices (L-M1, R-M1), and bilateral premotor cortices (L-PMC, R-PMC). Results showed two distinct peaks (Peak A at 5–20 Hz, Peak B at 25–45 Hz) across all analyses. Power and coherence analyses showed widespread increases in power and connectivity in both the Peak A and Peak B bands with disease progression. For Granger causality, increases in Peak B connectivity and decreases in Peak A connectivity were associated with the disease. Induction of mild disease was associated with several changes in connectivity: (1) the cortico-subthalamic connectivity in the descending direction (L-PMC to L-STN) decreased in the Peak A range while the reciprocal, ascending connectivity (L-STN to L-PMC) increased in the Peak B range; this may play a role in generating beta-band hypersynchrony in the cortex, (2) both L-M1 to L-PMC and R-M1 to R-PMC causalities increased, which may either be compensatory or a pathologic effect of disease, and (3) a decrease in connectivity occurred from the R-PMC to R-M1. The only significant change seen between mild and moderate disease was increased right cortical connectivity, which may reflect compensation for the left-hemispheric dominant moderate disease state.

The SARS-CoV-2 pandemic have been affecting millions of people worldwide, since the beginning of 2020. COVID-19 can cause a wide range of clinical symptoms, which varies from asymptomatic presentation to severe respiratory insufficiency, exacerbation of immune response, disseminated microthrombosis and multiple organ failure, which may lead to dead. Due to the rapid spread of SARS-CoV-2, the development of vaccines to minimize COVID-19 severity in the world population is imperious. One of the employed techniques to produce vaccines against emerging viruses is the synthesis of recombinant proteins, which can be used as immunizing agents. Based on the exposed, the aim of the present study was to verify the systemic and immunological effects of IM administration of recombinant Nucleocapsid protein (NP), derived from SARS-CoV-2 and produced by this research group, in 2 different strains of rats (Rattus norvegicus); Wistar and Lewis. For this purpose, experimental animals received 4 injections of NP, once a week, and were submitted to biochemical and histological analysis. Our results showed that NP inoculations were safe for the animals, which presented no clinical symptoms of worrying side effects, nor laboratorial alterations in the main biochemical and histological parameters, suggesting the absence of toxicity induced by NP. Moreover, NP injections successfully triggered the production of specific anti-SARS-CoV-2 IgG antibodies by both Wistar and Lewis rats, showing the sensitization to have been well sufficient for the immunization of these strains of rats. Additionally, we observed the local lung activation of the Bronchus-Associated Lymphoid Tissue (BALT) of rats in the NP groups, suggesting that NP elicits specific lung immune response. Although pre-clinical and clinical studies are still required, our data support the recombinant NP produced by this research group as a potential immunizing agent for massive vaccination, and may represent advantages upon other recombinant proteins, since it seems to induce specific pulmonary protection.

Longitudinal and behavioral preclinical animal studies generate complex data, which may not be well matched to statistical approaches common in this literature. Analyses that do not adequately account for complexity may result in overly optimistic study conclusions, with consequences for reproducibility and translational decision-making. Recent work interrogating methodological shortcomings in animal research has not yet comprehensively investigated statistical shortcomings in the analysis of complex longitudinal and behavioral data. To this end, the current cross-sectional meta-research study rigorously reviewed published mouse or rat controlled experiments for motor rehabilitation in three neurologic conditions to evaluate statistical choices and reporting. Medline via PubMed was queried in February 2020 for English-language articles published January 1, 2017- December 31, 2019. Included were articles that used rat or mouse models of stroke, Parkinson's disease, or traumatic brain injury, employed a therapeutic controlled experimental design to determine efficacy, and assessed at least one functional behavioral assessment or global evaluation of function. 241 articles from 99 journals were evaluated independently by a team of nine raters. Articles were assessed for statistical handling of non-independence, animal attrition, outliers, ordinal data, and multiplicity. Exploratory analyses evaluated whether transparency or statistical choices differed as a function of journal factors. A majority of articles failed to account for sources of non-independence in the data (74-93%) and/or did not analytically account for mid-treatment animal attrition (78%). Ordinal variables were often treated as continuous (37%), outliers were predominantly not mentioned (83%), and plots often concealed the distribution of the data (51%) Statistical choices and transparency did not differ with regards to journal rank or reporting requirements. Statistical misapplication can result in invalid experimental findings and inadequate reporting obscures errors. Clinician-scientists evaluating preclinical work for translational promise should be mindful of commonplace errors. Interventions are needed to improve statistical decision-making in preclinical behavioral neurosciences research.

The Brazilian Midwest has significant spatiotemporal variability in terms of precipitation and air temperature, making it more vulnerable to the occurrence of extreme weather events. The objective of this study is to characterize the trend of extreme climatic events regarding precipitation and air temperature in the Brazilian Midwest, and to analyze their relationship with Pacific and Atlantic Sea Surface Temperature anomalies (SSTAs). We used daily precipitation and air temperature data measured at 24 conventional weather stations. Pacific and Atlantic SSTA data were obtained from the Climate Prediction Center. The frequency of hot extremes had increased, while that of cold extremes had decreased significantly, thus highlighting the consistent warming across the Brazilian Midwest. The precipitation extremes had greater variability than the temperature extremes. Precipitation intensity increased in Amazonia, with no change in annual precipitation volume. The precipitation extremes in the Brazilian Savanna, Pantanal, and the Atlantic Forest did not have a well-defined pattern but indicated a trend towards a decrease in days with intense precipitation events. In general, the Equatorial Pacific and Atlantic Ocean (TNAI and TSAI) SSTAs were negatively correlated with precipitation extreme indices and positively correlated with air temperature extreme indices in the Amazon. However, the North Atlantic SSTAs were positively correlated with precipitation and air temperature extreme indices in the Brazilian Savanna and Pantanal. In addition, the Pacific SSTAs were positively correlated with precipitation intensity in the Atlantic Forest. Thus, the variability of the trends of precipitation and air temperature extreme indices in the Brazilian Midwest was observed, and it was surmised that this measure was significantly related to Pacific and Atlantic SSTAs.

This paper reports a new method to grow vertically-aligned carbon nanotubes (VA-CNT) in a cold-wall system, using a two-phase deposition step at low pressure. The first phase consists of the flow of NH3 for a short period together with the C2H2 flow to ensure catalyst activation. In the second phase, NH3 flow is replaced by H2 flow during the remaining growth time, to obtain tall VA-CNT. Previous tests with only one of the reduction gases flowing with C2H2 during the deposition step demonstrated that the NH3 flow contributes to increasing the VA-CNT uniformity throughout the substrate, while the H2 flow contributes to obtaining taller VA-CNT. The conclusions drawn during these tests lead to the recipe with a two-phase deposition step. The VA-CNT obtained with this new recipe reached a maximum height of 160 µm and a non-uniformity of 21.9 %, which falls within the state-of-the-art values. Furthermore, this process takes place at low pressure and only the chamber stage is heated, which allows a cheaper production of this material on a large scale.

Lahars are erosive floods, mixtures of water and pyroclastic detritus, known for being the biggest environmental disaster and causing a large number of fatalities in volcanic areas. Safety measures have been recently adopted in the threatened territories by constructing retaining dams and embankments in key positions. More disastrous events could be generated by the difficulty of maintaining these works in efficiency and for the changed risk conditions originating from their presence and the effects of their functioning. LLUNPIY/3r, a version of the cellular-automaton model LLUNPIY for lahar simulations, is presented. The growing frequency of lahars in the Vascún valley of Tungurahua volcano (Ecuador), probably due to the effects of the climatic change, has recently produced smaller and less dangerous events, sometimes favored by the collapse of ponds generated by small landslides. An investigation is performed here in order to reproduce such situations in a controlled way by the use of LLUNPIY/3r simulations. Using precise field data, points are individuated where dams by backfill, which are easy to collapse, can produce the formation of ponds; LLUNPIY/3r simulations permit projecting the triggering of small lahars by minor rainfall events or projecting, in the case of more rainfall, the anticipation of lahar detachment, avoiding simultaneous and dangerous confluence with other lahars.

Abstract Background One of the key steps in the management of chronic diseases in animals including Johne’s disease (JD), caused by Mycobacterium avium subsp. paratuberculosis (MAP), is the ability to track disease incidence over space and time. JD surveillance in the U.S. dairy cattle is challenging due to lack of regulatory requirements, imperfect diagnostic tests, and associated expenses, including time and labor. An alternative approach is to use voluntary testing programs. Here, data from a voluntary JD testing program, conducted by the Minnesota Dairy Herd Improvement Association, were used to: a) explore whether such a program provides representative information on JD-prevalence in Minnesota dairy herds, b) estimate JD distribution, and, c) identify herd and environmental factors associated with finding JD-positive cows. Milk samples (n = 70,809) collected from 54,652 unique cows from 600 Minnesota dairy herds between November 2014 and April 2017 were tested using a MAP antibody ELISA. Participant representativeness was assessed by comparing the number of JD-tested herds with the number of herds required to estimate the true disease prevalence per county based on official statistics from the National Agricultural Statistical Services. Multivariable logistic regression models, with and without spatial dependence between observations, were then used to investigate the association between herd status to JD (positive/negative), as indicated by milk ELISA results, and available covariates at the herd level. Results Within the study population, at least one test-positive cow was found in 414 of 600 (69%) herds. Results indicated that large herds that test frequently and herds located in loamy or silt soils are more likely to have at least one MAP test-positive cow. After adjusting for herd size, testing frequency, and soil type, there was no spatial dependence in JD risk between neighboring dairies within 5 to 20 km. Furthermore, the importance of collecting data on herd management, feed, and biosecurity for insightful interpretations was recognized. The study suggested that, although limited, the voluntary testing database may support monitoring JD status. Conclusions Results presented here help elucidate the spatial characteristics of JD in Minnesota and the study may ultimately contribute to the design and implementation of surveillance programs for the disease.

The Delta variant of SARS-CoV-2 has caused many breakthrough infections in fully vaccinated individuals. While vaccine status did not generally impact the number of viral RNA genome copies in nasopharyngeal swabs of breakthrough patients, as measured by Ct values, it has been previously found to decrease the infectious viral load in symptomatic patients. We quantified the viral RNA, infectious virus, and anti-spike IgA in nasopharyngeal swabs collected from individuals asymptomatically infected with the Delta variant of SARS-CoV-2. Vaccination decreased the infectious viral load, but not the amount of viral RNA. Furthermore, vaccinees with asymptomatic infections had significantly higher levels of anti-spike IgA in their nasal secretions compared to unvaccinated individuals with asymptomatic infections. Thus, vaccination may decrease the transmission risk of Delta, and perhaps other variants, despite not affecting the amount of viral RNA measured in nasopharyngeal swabs.

Teenagers generally present mild to no symptoms of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. In the present report, we present the case of a 14-year-old boy with Angelman syndrome (AS) who presented with severe COVID-19 symptoms. He spent 20 days in the ICU with elevated inflammatory biomarkers (C-reactive protein and D-dimer) and increased peaks of neutrophil-to-lymphocyte ratio, which is uncommon for teenagers diagnosed with COVID-19. Although he showed physiological instability, he was able to produce neutralizing antibodies, suggesting a functional immune response. The literature concerning the immune response to infections in patients with AS is still poor, and to our knowledge, this was the first report of a patient with AS diagnosed with COVID-19. As such, the present study may alert other patients with AS or other rare diseases that they lack a competent immune response and could suffer severe consequences of SARS-CoV-2 infection.
