1. Transport of l-arginine and the nitric oxide synthase inhibitors NG-monomethyl-l-arginine and NG-nitro-l-arginine was investigated in human erythrocytes from healthy donors and uraemic patients on haemodialysis. 2. Although Km values for total l-arginine influx were not significantly different in erythrocytes freshly isolated from controls or uraemic patients, uraemia was associated with an increase in the Vmax for transport (826 compared with 1176 μmol h−1 l−1 of cells) which was reduced to control values after dialysis. 3. Saturable influx of l-arginine was mediated by the classical cationic amino acid transport system y+ and system y+L, known to transport cationic and neutral amino acids with higher affinity. 4. Under zero-trans conditions, the Vmax for l-arginine transport via system y+ increased from 271 to 700 μmol h−1 l−1 of cells in uraemia, while Km values increased from 44 to 94 μmol/l. Dialysis had no significant effect on the kinetic parameters altered by uraemia. 5. Under zero-trans conditions, and with system y+ inhibited by N-ethylmaleimide (0.2 mmol/l), transport of l-arginine via system y+L was unaffected by uraemia. 6. Saturable influx of NG-monomethyl-l-arginine was also mediated by systems y+ (Km = 56 μmol/l, Vmax = 353 μmol h−1 l−1 of cells) and y+L (Km = 17 μmol/l, Vmax = 51.3 μmol h−1 l−1 of cells) and, as with l-arginine, uraemia increased the transport capacity for NG-monomethyl-l-arginine. 7. Influx of the neutral nitric oxide synthase inhibitor NG-nitro-l-arginine was not readily saturable. 8. Intracellular concentrations of l-arginine and NG-monomethyl-l-arginine were significantly increased in erythrocytes from uraemic patients when compared with controls, consistent with an increased transport capacity for l-arginine and NG-monomethyl-l-arginine. 9. The present study provides evidence that system y+ mediates the increased transport of l-arginine and NG-monomethyl-l-arginine in human erythrocytes from patients with chronic renal failure. Our findings may have implications for the activity of the l-arginine—nitric oxide signalling pathway in vascular endothelial and smooth-muscle cells in uraemia.

The overall effects of oestrogen on the vasculature are beneficial as a result of its antioxidant properties and ability to enhance relaxation by modulating synthesis of endothelium-derived vasodilators and smooth-muscle cell Ca2+ signalling. Although the protective effects of oestrogen are well accepted, only limited and conflicting data are available on the cellular mechanisms mediating steroid action in the vasculature. Discrepancies in cell culture experiments may reflect differences in culture conditions, the origin of cells and/or hormonal status. For example, umbilical vein endothelial cells isolated from gestational diabetic pregnancies exhibit phenotypic changes that are maintained during prolonged cell culture [42,75]. As gender differences in vascular reactivity have long been reported [76-78], gender needs to be taken into account in interpreting vascular responses to oestrogen in animal models. Further research is required to characterize the non-genomic actions of oestrogen in the vasculature. Whether activation of non-genomic receptors by physiologically relevant plasma concentrations of oestrogen modulates the activity of endothelial cell NOS, cyclo-oxygenase and superoxide dismutase and smooth-muscle cell Ca2+ ion channel activity remains to be investigated. Prolonged exposure of the vasculature to circulating steroid hormones may more closely reflect in vivo conditions, since oestrogen is known to enhance endothelium-dependent relaxation in response to some agonists [79].

During the last 10 years, the role of oxidative stress in pancreatitis and the benefits or otherwise of antioxidants has been the subject of numerous research papers. There is general agreement that glutathione and other sulphydryl compounds are depleted while lipid peroxidation is increased in pancreatic tissue during the development of acute pancreatitis. Treatment with antioxidants has been shown to reduce acinar cell injury and oedema in various animal models of pancreatitis, suggesting that the sustained generation of reactive oxygen species depletes cellular antioxidant defences. Evidence for a role for bradykinin and nitric oxide in pancreatitis has been conflicting with some studies suggesting these agents might ameliorate pancreatic dysfunction by enhancing pancreatic blood flow and secretion in response to bradykinin-stimulated generation of nitric oxide from endothelium, while other studies suggest that nitric oxide potentiates pancreatic oxidative stress. Thus, there is clearly a need for well-designed clinical trials to evaluate the protective role of antioxidant therapy in acute pancreatitis.

The effects of exogenous insulin, glucagon and streptozotocin-diabetes on influx (15 s) of L-lysine via a cationic amino acid transporter resembling system y+ were investigated in the isolated perfused rat pancreas. In non-diabetic pancreata, transport of L-lysine was saturable with an apparent Km of 2.11 +/- 0.29 mM and Vmax of 2.21 +/- 0.20 mumol min-1 g-1 (n = 6). Bovine insulin (100 mu u ml-1) increased the maximal transport rate (Vmax = 3.49 +/- 0.30 mumol min-1 g-1, n = 4, P < 0.05) for L-lysine 1.6-fold without altering the Km. L-Lysine transport was not elevated significantly in diabetic pancreata, although insulin (100 mu u ml-1) enhanced transport to values measured in non-diabetic preparations. Human glucagon (1.5 x 10(-9) M) had no stimulatory effect on L-lysine transport. These findings provide the first evidence that exogenous insulin stimulates cationic amino acid transport activity in the exocrine pancreatic epithelium. Activation of the cationic pancreatic amino acid transporter may provide a mechanism to enhance the supply of L-arginine and thus sustain nitric oxide-mediated pancreatic secretion in response to islet hormones and secretagogues.

1. The roles of the gamma-glutamyl cycle and the anionic amino acid transport system xc- in mediating L-cystine uptake were investigated in cultured human pancreatic duct PaTu 8902 cells. This cell line exhibits morphological features of normal pancreatic duct cells and expresses gamma-glutamyl transpeptidase (gamma-GT, EC 2.3.2.2), an enzyme involved in the metabolism and regulation of intracellular glutathione (GSH). 2. Uptake of L-cystine (10 microM) was linear for up to 10 min, temperature dependent, Na+ independent, saturable (Michaelis-Menten constant (Km), 86 +/- 25 microM; maximal velocity (Vmax), 109 +/- 33 nmol (mg protein)-1 h-1) and reduced by 80-90% by a 50-fold excess concentration of L-glutamate and L-homocysteic acid, but not L-aspartate. These transport properties resemble those described for system xc-, which exchanges cystine for intracellular glutamate. 3. Acivicin, a known inhibitor of gamma-GT, decreased gamma-GT activity from 2.58 +/- 0.96 to 0.97 +/- 0.11 mU (mg protein)-1 and decreased the initial rates of L-cystine and L-glutamine uptake by 41-55%. Anthglutin (1-gamma-L-glutamyl-2-(2-carboxyphenylhyl)hydrazine), a structurally different inhibitor of gamma-GT, also caused a concentration-dependent (0.01-1 mM) decrease in gamma-GT activity and L-cystine uptake. 4. Neither acivicin nor anthglutin inhibited the uptake of L-glutamate, a poor substrate for gamma-GT. 5. In the presence of a 500-fold excess concentration of glutamate, which should abolish entry of cystine via system xc-, the remaining fraction of cystine transport was inhibited by 50% by acivicin, suggesting that transport is, in part, dependent on the activity of gamma-GT. 6. Cystine transport was also 60-80% inhibited by a series of gamma-glutamyl amino acids (5 mM) including gamma-glutamyl-glutamate, gamma-glutamyl-glutamine and gamma-glutamyl-glycine. alpha-Dipeptides inhibited cystine transport by only 6-22%. 7. These findings demonstrate that in human pancreatic duct PaTu 8902 cells, cystine uptake is mediated by system xc- (50-60%) and the gamma-glutamyl cycle. Our results provide the first evidence linking gamma-GT with cystine transport in human epithelial cells and are of relevance in view of the importance of cystine as a sulphur amino acid source for GSH synthesis in cells exposed to oxidative stress.

We have investigated changes in endometrial oxytocin receptor concentrations and prostaglandin F2 alpha release in response to exogenous oxytocin treatment in ovariectomized cows treated with progesterone and oestradiol, and made comparisons with similar treatment in cyclic cows. In long-term ovariectomized cows, endometrial oxytocin receptors were present (300 fmol mg-1 protein), but no prostaglandin F2 alpha was released in response to oxytocin treatment until after the administration of progesterone. Subsequent administration of a concentration of oestradiol sufficient to induce oestrus resulted in the downregulation of these receptors and the loss of oxytocin responsiveness, which did not reappear within 20 days in the absence of further hormone treatment. When induced oestrus was followed by further treatment with luteal phase concentrations of progesterone and oestradiol, both oxytocin receptors and oxytocin-stimulated release of prostaglandin F2 alpha reappeared by day 16 after oestrus, in a pattern similar to that seen during the luteal phase of cyclic cows. These results demonstrate how progesterone and oestradiol control the development and responsiveness of endometrial oxytocin receptors in cows, and provide a valuable model in which to investigate further the precise control of the oxytocin receptor in this species.

Three cases of multiple osteochondromatosis are reported among 843 skeletons excavated from the site of the ancient city of Jericho. Radiocarbon dating and archaeological evidence both suggest that two of the skeletons date from about 1700 BC. These skeletons represent the earliest known cases of diaphyseal aclasis.

Microvascular permeability was studied in the isolated perfused rat pancreas using a rapid multiple indicator-dilution technique. Capillary extractions, permeability-surface area products (PS), and extravascular volumes of distribution (EVV) were determined for 22Na+, 51Cr-labeled EDTA, [57Co]-cyanocobalamin (B12), and 125I-labeled insulin at various perfusion flows. Permeability to albumin was negligible. PS for Na+ and EDTA increased with increasing flow, whereas PS for cyanocobalamin and insulin approached diffusion-limited exchange at flows greater than 3 ml.min-1.g-1. Permeability coefficients for Na+, EDTA, B12, and insulin were 36, 22, 11, and 3.48 x 10(-5) cm/s, respectively, and the permeability ratio for B12/insulin (3.16) indicated restricted diffusion to insulin. In the presence of unlabeled B12 and insulin EVV (0.15-0.19 ml/g) for EDTA, B12 and insulin approximated the interstitial volume. Caerulein-induced pancreatitis or treatment with the synthetic protease inhibitor camostate had no significant effects on permeability. In caerulein-treated rats, EVV for B12 was elevated (0.17 +/- 0.01 vs. 0.28 +/- 0.06; P less than 0.01), reflecting the interstitial edema associated with this model of pancreatitis. Permeability of the rat pancreatic microvasculature is similar to that of other fenestrated tissues, but it is 10- to 20-fold greater than that of continuous capillaries. Contrary to previous assumptions, permeability does not appear to be increased after induction of acute interstitial pancreatitis.

Nitric oxide (NO) inhibits platelet function and plays a key role in the regulation of cardiovascular homeostasis. Essential hypertension is characterized by an increased risk of thrombus formation, and by an inhibition of intraplatelet NO bioactivity. We have previously shown that membrane transport of L-arginine is a rate-limiting step for platelet-derived NO synthesis. This study examined the effects of exercise on the platelet L-arginine-NO pathway and aggregation and systemic inflammation markers in 13 sedentary hypertensive patients subjected to 60 min of training activity (exercise group), predominantly aerobic, three times a week for a period of 12 weeks. Six sedentary hypertensive patients participated in the control group. After 12 weeks, L-arginine transport was significantly increased and associated with increased platelet NO synthase activity and cGMP levels and reduced platelet aggregation. Moreover, exercise training reduced plasma concentrations of fibrinogen and C-reactive protein and blood pressure. The control group did not change their previous intraplatelet L-arginine-NO results and systemic inflammatory markers levels. Thus, exercise training reduces inflammatory responses, restores NO synthesis in platelets and thereby contributes to the beneficial effects of exercise in hypertension. The present study adds exercise as a new tool to reduce morbidity and mortality associated with platelet activation in hypertension.

Luteal inadequacy is a major cause of infertility in a number of species. During the early luteal phase, progesterone production requires the rapid growth of the corpus luteum (CL), which is in turn dependent on angiogenesis. In the present study, we examined the temporal changes in vascular endothelial growth factor A (VEGFA), fibroblast growth factor 2 (FGF2) and secreted protein, acidic, cysteine-rich (osteonectin) (SPARC) during the follicular-luteal transition and CL development in the cow. Luteal VEGFA concentrations increased as the CL developed but were lower in the regressing CL. Conversely, luteal FGF2 concentrations were highest immediately postovulation in the collapsed follicle and declined as the CL developed. Furthermore, three FGF2 isoforms were present in the collapsed follicle, but only one isoform was detected in older CL. Interestingly, FGF2 concentrations increased in the regressing CL. Western blot analysis for SPARC showed the presence of two isoforms, which were constitutively expressed throughout CL development. Further studies investigated the regulation of FGF2 by LH, which showed that FGF2 concentrations in preovulatory follicular fluid were higher in those animals that had experienced an LH surge. Moreover, LH stimulated FGF2 production in dispersed luteal cells. Conversely, the LH surge had no effect on follicular fluid VEGFA concentrations. In conclusion, FGF2 was more dynamic than VEGFA and SPARC during the follicular-luteal transition, which suggests that FGF2 plays a key role in the initiation of angiogenesis at this time. Furthermore, it is likely that this is stimulated by the LH surge. The results also suggest that VEGFA and SPARC have a more constitutive, but essential, role in the development of the CL vasculature.

1. Chronic renal failure (CRF) is associated with the abnormal regulation of nitric oxide (NO) synthesis at the systemic level. The transport of L-arginine, upregulated in blood cells from uraemic patients, modulates NO synthesis in this pathological condition. The model of partial nephrectomy in rats is widely accepted as a valid model of uraemia. Because there are no reports of L-arginine transport in blood cells from uraemic rats, the aim of the present study was to investigate L-arginine transport in red blood cells (RBCs) from these rats. 2. The kinetics of L-arginine transport in RBC and plasma and the amino acid profiles of RBC were investigated in control, sham-operated and subtotally nephrectomized rats. 3. L-Arginine transport was mediated via the cationic amino acid transport system y+ and a transport system with kinetics resembling the human system y+L. In control RBC, the apparent Ki for L-leucine inhibition of L-arginine transport via system y+L was 0.16 +/- 0.02 and 4.8 +/- 2 mmol/L in the presence of Li+ and Na+, respectively. 4. The Vmax values for L-arginine transport via system y+L and system y+ were similar in RBC from control sham-operated and uraemic rats. Moreover, L-arginine concentrations in plasma and RBC were not affected by uraemia. 5. The findings of the present study provide the first evidence that L-arginine transport in rat erythrocytes is mediated by two distinct cationic transport systems with characteristics of systems y+ and y+L, which accept neutral amino acids only in the presence of Li+. In contrast with previous studies in uraemic patients, plasma levels and maximal transport rates of L-arginine were not altered in this rat model of CRF.

Early mammalian embryo development in vitro can be enhanced by co-culture with oviductal cells and by the addition of insulin-like growth factors (IGFs). This study examined the expression patterns of the oviductal IGF system in cattle in relation to the number of days after oestrus and the presence or absence of embryos. Oviducts were collected from: (i) 66 nulliparous heifers on day 3, day 6 or day 16 after insemination and from (ii) ten non-pregnant, lactating cows on day 0 or day 1 of the oestrous cycle. Oviducts were coiled, frozen whole and sectioned for in situ hybridization. Expression patterns of mRNAs encoding IGF-I, IGF-II, type 1 IGF receptor (IGF-1R), and the IFG binding proteins (IGFBP)-1, -3 and -5 were determined from autoradiographs. Separate measurements were made for the mucosa and muscle layers of the infundibulum, ampulla and isthmus. None of the parameters measured differed between heifers with or without the presence of an embryo. mRNAs encoding IGF-I and IGF-1R were present in the mucosa and muscle of all three oviductal regions, and the highest value of IGF-I mRNA was measured in heifers on day 3. IGF-II mRNA was expressed predominantly in the muscle wall. IGFBP-1 mRNA was not detectable, whereas mRNAs encoding IGFBP-3 and -5 were expressed in both the muscle and mucosa. IGFBP-3 expression was higher in cows on day 0 and day 1 of the oestrous cycle than in heifers on day 3, day 6 and day 16 after insemination. A peak of IGFBP-5 expression was reached on day 6. Locally or systemically produced IGFs, regulated by IGFBPs, may act directly on the embryo or indirectly via modulation of oviductal secretions and muscular activity to influence the success of early embryo development.

Bovine viral diarrhoea virus (BVDV) is a major pathogen of cattle and is responsible for considerable reproductive loss. In this study, the in vivo responses in six multiparous cows were investigated after a non-cytopathogenic BVDV challenge (strain Pe 515; 5 x 10(6) tissue culture infective dose 50) given 9 days before a synchronized ovulation. Six similar cows challenged with non-infectious culture medium served as controls. The experimental noncytopathogenic BVDV infection was followed by a viraemia and leucopenia at days 5-9 after challenge, but no other clinical signs of infection were detected. However, the BVDV infection altered endocrine function. Mean LH pulse frequency immediately before CIDR withdrawal was lower (Por = 0.05) in the BVDV-infected (2.17 +/- 0.34 pulses per 8 h) compared with the sham-infected (4.83 +/- 1.04 pulses per 8 h) animals. At day 3 after CIDR withdrawal, plasma oestradiol concentrations remained high (P0.05) in the infected cows (2.19 +/- 0.51 pg ml(-1)) compared with the sham-infected controls (0.72 +/- 0.29 pg ml(-1)). However, there was no difference in the peak oestradiol concentration (BVDV: 2.31 +/- 0.29 versus sham: 2.34 +/- 0.41 pg ml(-1)). In addition, non-cytopathogenic BVDV significantly (P0.05) increased the duration of the interval between ovulation and onset of the postovulatory progesterone increase (values 1.0 ng ml(-1)) (BVDV: 3.0 +/- 0.26 versus sham: 4.0 +/- 0.26 days). The viral infection also significantly (P0.01) decreased mean plasma progesterone concentrations between day 3 and day 11 after ovulation (BVDV: 2.59 +/- 0.32 versus sham: 4.13 +/- 0.27 ng ml(-1)). These data show that non-cytopathogenic BVDV viraemias during the follicular phase can modulate the secretion of gonadotrophins and sex steroids, in particular progesterone, during a synchronized oestrous cycle. Therefore, viraemias during the follicular phase may reduce the fertility of cattle by disrupting the capacity of the ovulatory follicle to form a competent corpus luteum, thereby compromising early embryo development and maternal recognition of pregnancy.

This study examined the expression patterns of oxytocin and steroid receptors in the bovine endometrium during the oestrous cycle and early pregnancy to elucidate their respective roles in the regulation of luteolysis and the maternal recognition of pregnancy. In Expt 1, uterine biopsies were collected from four cows throughout three oestrous cycles each, to provide daily samples. In Expt 2, uterine tissue was collected on days 12, 14, 16 and 18 of the oestrous cycle (n = 20) or early pregnancy (n = 16). Oxytocin receptor, oestrogen receptor alpha and progesterone receptor mRNAs were localized by in situ hybridization, and localization of oestrogen receptor and progesterone receptor was confirmed by immunocytochemistry. All three receptors showed time- and cell-specific expression patterns. Oestrogen receptor alpha increased in all regions at oestrus but high concentrations were also found in the luminal epithelium during the mid-luteal phase and in the deep glands throughout the oestrous cycle. Progesterone receptor expression was higher in the stroma than it was in the types of epithelial cell, and increased expression was observed at oestrus and during the early luteal phase. The cyclical upregulation of oxytocin receptors in the luminal epithelium on about day 16 was not related to preceding changes in the endometrial expression of either oestradiol alpha or progesterone receptors. During early pregnancy, oxytocin receptor expression was suppressed. Oestrogen receptor a concentrations increased in the non-pregnant cows and decreased in the pregnant cows between days 16 and 18, but these changes followed rather than preceded the upregulation of oxytocin receptors in the non-pregnant cows. It is concluded that the initial upregulation of oxytocin receptors in the luminal epithelium, which triggers luteolysis, is not associated directly with changes in expression of oestrogen receptor alpha.

Both the production of cytokines and the distribution of immune cells within the uterus change during early pregnancy. Evidence obtained mainly from mice indicates that these changes are important for implantation and in preventing a maternal immune response to the conceptus. The ruminant embryo also produces interferon tau at this time, the signal for the maternal recognition of pregnancy. The relationship between these events in cows was studied using uteri from three groups of animals on day 16 after observed oestrus: (i) cyclic controls, (ii) pregnant and (iii) inseminated but with no embryo present. Embryo size and the antiviral activity in uterine flushings (indicative of the interferon tau concentration) were measured. Sections of intact uterus were frozen for the localization and quantitation of CD4(+) (T lymphocytes), CD14(+) (macrophages) and CD21(+) (B lymphocytes) uterine cells by immunohistochemistry. The expression of interleukin (IL)-1alpha, IL-2, IL-6 and IL-10 mRNAs in uterine extracts was measured by RT-PCR. Neither embryo size, interferon tau concentration nor pregnancy status influenced the distribution of CD4(+), CD14(+) or CD21(+) cells in the day 16 uterus. Endometrial IL-1alpha mRNA was detected in most cows across the groups, whereas IL-2 mRNA was only present in the non-pregnant uterus. IL-6 and IL-10 mRNAs were not detectable in any uteri. In conclusion, IL-2 mRNA expression is detectable in the non-pregnant but not the pregnant uterus on day 16 and interferon t is unlikely to play a role in the redistribution of immune cells in the uterus during early bovine pregnancy.

Reactive oxygen radicals, nitric oxide, and cytokines have been implicated in the initiation of pancreatic tissue damage and impairment of the pancreatic microcirculation in acute pancreatitis. Pentoxifylline is a methylxanthine derivative with rheologic and marked anti-inflammatory properties and inhibits the production of proinflammatory cytokines. We have examined whether pentoxifylline ameliorates interstitial edema, inflammatory infiltrate, and glutathione depletion associated with cerulein-induced pancreatitis. Cotreatment of animals with pentoxifylline significantly reduced cerulein-induced pancreatic inflammation and edema and attenuated the depletion of pancreatic glutathione and the increase in serum lipase activity, nitrate, and tumor necrosis factor-alpha levels. Pentoxifylline also prevented both mitochondrial swelling and damage to mitochondrial cristae caused by cerulein. Our findings provide an experimental basis for using pentoxifylline to attenuate inflammatory responses within the pancreas in acute pancreatitis and as an adjuvant in the treatment of acute pancreatitis.

The signalling mechanisms involved in the induction of nitric oxide synthase and l-arginine transport were investigated in bacterial lipopolysaccharide (LPS)- and interferon-gamma (IFN-gamma)-stimulated rat cultured aortic smooth muscle cells (RASMCs). The expression profile of transcripts for cationic amino acid transporters (CATs) and their regulation by LPS and IFN-gamma were also examined. Control RASMCs expressed mRNA for CAT-1, CAT-2A and CAT-2B. Levels of all three transcripts were significantly elevated in activated cells. Stimulated CAT mRNA expression and l-arginine transport occurred independently of protein kinase C (PKC), protein tyrosine kinase (PTK) and p44/42 mitogen-activated kinases (MAPKs), but were inhibited by the p38 MAPK inhibitor SB203580, which at 3 microM caused maximum inhibition of both responses. Induction of NO synthesis was independent of p44/42 MAPK activation and only marginally dependent on PKC, but was attenuated markedly by the PTK inhibitors genistein and herbimycin A. SB203580 differentially regulated inducible NO synthase expression and NO production, potentiating both processes at low micromolar concentrations and inhibiting at concentrations of >/=1 microM. In conclusion, our results suggest that RASMCs constitutively express transcripts for CAT-1, CAT-2A and CAT-2B, and that expression of these transcripts is significantly enhanced by LPS and IFN-gamma. Moreover, stimulation of l-arginine transport and induction of NO synthesis by LPS and IFN-gamma appear to be under critical regulation by the p38 MAPK, since both processes were significantly modified by SB203580 at concentrations so far shown to have no effect on other signalling pathways. Thus, in RASMCs, the p38 MAPK cascade represents an important signalling mechanism, regulating both enhanced l-arginine transport and induced NO synthesis.

The effects of the oxidant tert-butylhydroperoxide (t-buOOH) on carbachol-stimulated pancreatic secretion in the vascularly perfused rat pancreas have been studied in parallel with [Ca2+]i signalling and amylase output in perifused rat pancreatic acinar cells. Perfusion of the pancreas with t-buOOH (0.1–1 mM) caused a rapid and irreversible inhibition of carbachol-stimulated (3×10−7 M) amylase and fluid secretion. Pre-perfusion of the pancreas with vitamin C and dithiothreitol or a cocktail of GSH and GSH-precursor amino acids provided only marginal protection against the deleterious effects of t-buOOH, even though GSH levels were elevated significantly. In perifused pancreatic acini, repetitive [Ca2+]i spikes evoked by carbachol (3×10−7 M) were sustained for 40 min. t-buOOH (1 mM) acutely increased the amplitude and duration of Ca2+ spikes, then attenuated Ca2+ spiking and subsequently caused a marked and sustained rise in [Ca2+]i. t-buOOH-induced alterations in carbachol-stimulated [Ca2+]i signalling and amylase release in perifused pancreatic acini were prevented by vitamin C. Although vitamin C restored impaired Ca2+ signalling and maintained amylase output in pancreatic acini, it seems likely that oxidative stress inhibits fluid secretion irreversibly in the intact pancreas, resulting in a loss of amylase output. Thus, perturbations in [Ca2+]i signalling may not fully explain the secretory block caused by oxidative stress in acute pancreatitis.

1. In this study the mechanisms of the acute vasodilator action of bacterial lipopolysaccharide (LPS) were investigated in the rat Langendorff perfused heart. 2. Infusion of LPS (5 microg ml(-1)) caused a rapid and sustained fall in coronary perfusion pressure (PP) of 59 +/- 4 mmHg (n = 12) and a biphasic increase in NO levels determined in the coronary effluent by chemiluminescent detection. Both the fall in PP and the increase in NO release were completely abolished (n = 3) by pretreatment of hearts with the NO synthase inhibitor L-NAME (50 microM). 3. LPS-induced vasodilatation was markedly attenuated to 5 +/- 4 mmHg (n 3) by pretreatment of hearts with the B2 kinin receptor antagonist Hoe-140 (100 nM). 4. Vasodilator responses to LPS were also blocked by brief pretreatment with mepacrine (0.5 microM, n = 3) or nordihydroguaiaretic acid (0.1 microM, n = 4) and markedly attenuated by WEB 2086 (3 microM, n = 4). 5. Thirty minutes pretreatment of hearts with dexamethasone (1 nM), but not progesterone (1 microM), significantly modified responses to LPS. The action of dexamethasone was time-dependent, having no effect when applied either simultaneously with or pre-perfused for 5 min before the administration of LPS but inhibiting the response to LPS by 91 +/- 1% (n = 4) when pre-perfused for 15 min. The inhibition caused by dexamethasone was blocked by 15 min pretreatment with the glucocorticoid receptor antagonist RU-486 (100 nM) or by 2 min pre-perfusion of a 1:200 dilution of LCPS1, a selective antilipocortin 1 (LC1) neutralizing antibody. 6. Treatment with the protein synthesis inhibitor, cycloheximide (10 microM, for 15 min) selectively blunted LPS-induced vasodilatation, reducing the latter to 3 +/- 5 mmHg (n = 3), while having no effect on vasodilator responses to either bradykinin or sodium nitroprusside. 7. These results indicate that LPS-induced vasodilatation in the rat heart is dependent on activation of kinin B2 receptors and synthesis of NO. In addition, phospholipase A2 (PLA2) is activated by LPS resulting in the release of platelet-activating factor (PAF) and lipoxygenase but not cyclo-oxygenase products. These effects are dependent on de novo synthesis of an intermediate protein which remains to be identified.

1. Transport of l-arginine was investigated under zero-trans conditions in human erythrocytes from healthy donors and patients with heart failure. 2. Saturable influx of l-arginine was mediated by the classical cationic amino acid transport systems y+ and y+L. 3. The Vmax for l-arginine transport via system y+ increased from 292 to 490 μmol h−-1 l−-1 of cells in heart failure. 4. With system y+ inhibited by N-ethylmaleimide (0.2 mmol/l), the Vmax for the transport of l-arginine via system y+L was unaffected in erythrocytes from patients with heart failure. 5. The inhibition of l-arginine and l-leucine influx by NG-monomethyl-l-arginine was similar in erythrocytes from control and heart failure patients. 6. Plasma l-arginine levels were reduced in patients with heart failure (59 μmol/l) compared with controls (125 μmol/l). Plasma from patients with heart failure also contained the endogenous l-arginine analogue NG-monomethyl-l-arginine, which was undetectable in plasma from controls. 7. Intracellular concentrations of l-arginine and NG-monomethyl-l-arginine were significantly elevated in erythrocytes from patients with heart failure compared with controls, consistent with an increased transport capacity for l-arginine and NG-monomethyl-l-arginine. 8. The present study provides the first evidence that system y+ mediates the increased transport of l-arginine in human erythrocytes from patients with chronic heart failure. These findings are similar to our previous results obtained in patients with chronic renal failure. Since both pathologies seem to present with an increased synthesis of nitric oxide, studies of l-arginine transport in erythrocytes may provide a valuable paradigm to study abnormalities of the l-arginine-nitric oxide signalling pathway.

Vascular endothelial cells synthesize nitric oxide (NO) from the terminal guanidine-nitrogen atom of the cationic amino acid L-arginine (Palmer, R et al, 1988). Although basal NO synthesis is not limited by arginine availability, under conditions of stimulated NO production or following substrate deprivation exogenous L-arginine appears to be rate-limiting (Palmer, R. et al., 1988). We have now investigated whether bradykinin, an agonist which is known to release NO release, modulates the endothelial cell L-arginine transporter.

Twenty-four Scottish Blackface ewes (mean weight 50.0 +/- 0.1 kg with ovulation rate 1.3 +/- 0.1) were randomly divided into 4 groups of 6 animals. Under general anesthesia, following the collection of a timed sample of ovarian venous blood, the ovaries of these animals were collected either on Day 10 of the luteal phase or 12, 24, and 48 h after a luteolytic dose of a prostaglandin (PG) F2 alpha analogue (cloprostenol 100 micrograms i.m.) administered on Day 10. All follicles greater than 3 mm were dissected from the ovaries and incubated in Medium 199 (M199) at 37 degrees C for 2 h, following which the granulosa cells were harvested and incubated in triplicate for 24 h in M199 with or without ovine FSH or ovine LH. Plasma and culture media samples were assayed for inhibin, estradiol (E2), androstenedione (A4), and testosterone (T) by specific RIA. After correcting for hematocrit, ovarian secretion rates were calculated from the product of the plasma concentration and flow rate. The rate of ovarian inhibin secretion during the luteal phase was similar from ovaries categorized on the basis of presence of luteal tissue (1.0 +/- 0.3 and 0.9 +/- 0.5 ng/min for CL present and absent, respectively), confirming that the ovine CL does not secrete appreciable amounts of inhibin. Inhibin secretion was higher (p less than 0.05) at 12 h after PG-induced luteolysis but not at 24 or 48 h compared to values for luteal phase control ewes. Although ovaries containing large estrogenic follicles (greater than or equal to 4 mm in diameter and classified as estrogenic from in vitro criteria) secreted the most inhibin (55%; p less than 0.05), both ovaries containing large nonestrogenic follicles (33%) and small (11%; less than 4 mm in diameter) follicles secreted appreciable amounts of inhibin. This contrasted strongly with E2 where greater than 80% of the steroid was secreted by large estrogenic follicles. The rate of ovarian inhibin secretion was positively correlated (p less than 0.05) with the rate of E2, A4, and T secretion. Overall, there was no significant effect of stage of cycle on follicular inhibin content after 2 h incubation in vitro, release of inhibin by follicles incubated in vitro, or synthesis of inhibin by granulosa cells cultured in vitro. FSH and LH had no effect on the production of either inhibin or estradiol by cultured granulosa cells. Follicular diameter was positively correlated (p less than 0.001) with follicular inhibin and steroid release. Follicular inhibin content after 2 h incubation in vitro was more highly correlated with inhibin release by incubated follicles (r = 0.7; p less than 0.001) than with inhibin synthesis by granulosa cells in vitro (0.4; p less than 0.01).(ABSTRACT TRUNCATED AT 400 WORDS)

Amino acid transport in the isolated perfused exocrine pancreas is mediated by at least four parallel transporters, exhibiting differential sensitivity to islet hormones, diabetes, fasting and dietary composition. Insulin and streptozotocin-induced diabetes stimulate amino acid influx via the Na+-independent Systems asc (L-serine) and y+ (L-lysine). Moreover, activation of transport by insulin is dependent upon extracellular Ca2+ and inhibited by somatostatin-14. Elevation of transport induced by diabetes may not be mediated by glucagon, as exogenous glucagon fails to stimulate transport. Although system L (L-phenylalanine) is unaffected by insulin and diabetes, fasting for 72 h activates transport and refeeding for 24 h restores transport to fed levels. Adaptive responses in the activities of systems L and y+ induced by fasting and changes in dietary protein may involve changes in intracellular amino acid levels which are known to influence counter-transport. Elucidating the mechanisms regulating uptake and incorporation of amino acids into pancreatic enzymes is of clinical relevance, since 11 C-labelled amino acids are currently used as PET imaging probes for regional tissue metabolism in pancreatitis.

Transport of short-chain and long-chain neutral amino acids across the basolateral membrane of the epithelium in the perfused cat salivary gland has been studied using a rapid (less than 30 s) single circulation paired-tracer dilution technique. Amino acid uptake was measured by comparing the venous dilution profiles for a tritiated amino acid and D-[14C]mannitol (extracellular reference) following a bolus intra-arterial injection of a mixture containing both molecules. Unidirectional influx (v) was estimated from the maximal tracer uptake (Umax), the perfusate flow (F) and the perfusate amino acid concentration (Ca): v = [-F . ln (1-Umax)] . Ca. L-alanine influx was saturable and apparently mediated by a single entry system (Km = 0.83 +/- 0.11 mM and Vmax = 655 +/- 32 nmol/min . g). These kinetic constants were considerably lower than our previously reported values for L-phenylalanine: Km = 6.4 mM and Vmax = 1719 nmol/min . g. In cross-inhibition experiments performed over a wide range of concentrations (0.05-24 mM), influx of L-alanine and L-phenylalanine could be further discriminated, since both L-phenylalanine (Ki = 22 mM) and L-alanine (Ki = 19 mM) behaved as poor competitors. Removal of Na+ from the perfusate resulted in a selective inhibition of L-alanine and L-serine influx, whereas influx of the long-chain neutral amino acids L-leucine, L-phenylalanine and L-tryptophan remained unaffected. Although prolonged perfusion of glands with dinitrophenol (0.8 mM for 20-30 min) caused a variable but net inhibition of unidirectional uptake, it markedly enhanced the tracer efflux of L-leucine, L-phenylalanine, L-tyrosine and the basic amino acid L-lysine. It appears that at least two separate neutral amino acid transport systems are operative at the blood-tissue interface of the salivary epithelium: (i) a Na+-dependent alanine-serine-cysteine preferring type of carrier exhibiting a high affinity for amino acids with short, polar or linear side chains and (ii) a Na+-independent leucine preferring type of carrier selective for large neutral amino acids.

The multigrain model for polymerization of olefins over solid catalysts is used to predict kinetic behavior, molecular weights, and polydispersities. The effects of intraparticle and external boundary layer transport resistance on the kinetic behavior and polymer properties are explored. Means for the experimental detection of intraparticle diffusion resistance are suggested. The importance of catalyst physical properties, such as the porosity, and the catalyst loading is illustrated through simulation. Finally, the hypotheses of diffusion resistance and site heterogeneity as explanations for the broad molecular weight distributions of olefin polymers are critically evaluated, and molecular weight distribution control in industrial catalysts is discussed.

We have applied the paired-tracer dilution method to the study of transport processes in a mixed mammalian muscle preparation, the perfused rat hindlimb. The method is suitable for the characterization of the kinetic parameters of sugar and amino acid transport and its regulation by hormones, contractile activity, hypoxia, etc. Insulin stimulates sugar transport by increasing the Vmax. of the process 2-3 fold, but its affinity is unchanged. Starvation increases the affinity of sugar transport in perfused skeletal muscle.

Capillary permeability and cellular uptake of noradrenaline by the isolated artificially perfused rabbit heart was measured using rapid (less than 30 s) single-circulation tracer-dilution techniques. In a single coronary circulation capillary extractions of L-[14C]noradrenaline and D-[3H]mannitol (extracellular reference) relative to an intravascular marker, 125I-labelled albumin, were similar and above 60%. The 'apparent' volume of distribution for tracer noradrenaline was 2.5-fold larger than that measured for D-mannitol (0.32 ml g-1) suggesting cellular uptake of the amine. Unidirectional noradrenaline uptake was estimated by directly comparing coronary sinus dilution profiles of L-[3H]noradrenaline and D-[14C]mannitol. Michaelis-Menten saturation kinetics based on a single-entry system were determined (Km = 2.8 +/- 1.5 microM, Vmax = 2.1 +/- 0.5 nmol min-1 g-1, n = 4) by perfusing hearts with varying concentrations of L-noradrenaline (1-10 microM). Various known inhibitors of noradrenaline uptake were investigated to determine whether uptake was mediated by neuronal (uptake1) and/or extraneuronal (uptake2) mechanisms. Desipramine (5 microM), imipramine (5 microM) and metaraminol (2 microM) resulted in a 66-94% inhibition of noradrenaline influx. In comparison, the steroids, 17 beta-oestradiol (1 microM) and corticosterone (10 microM), and the noradrenaline metabolite normetanephrine (5 microM) caused virtually no inhibitory effects. The beta-adrenergic antagonist propranolol (5 microM) was also relatively ineffective. These results together with the kinetic constants estimated suggest that the rapid noradrenaline uptake reflects transport into adrenergic neurones lying in the coronary interstitium. The high resolution of this paired-tracer dilution technique has permitted a 'non-invasive' study of neuronal uptake mechanisms and its application may be of clinical value.

The specificity and kinetics of L-lysine influx across the basolateral surface of the cat salivary epithelium have been investigated in the perfused cat submandibular gland using a high-resolution, paired-tracer dilution technique. L-lysine influx was measured at several different perfusate concentrations (0.05-2.5 mM) and was found to be saturable. A Michaelis-Menten analysis based on a single entry site gave a Km of 0.49 +/- 0.08 mM and a Vmax of 231 +/- 20 nmol/min X g. The uptake of L-lysine was highly stereospecific and markedly inhibited by L-arginine (0.25-2.5 mM). The inhibitor constant (Ki) was 0.23 mM, suggesting that the carrier had a greater affinity for L-arginine than L-lysine. When the inhibitory effects of L-histidine (0.5-10 mM) were examined the Ki, estimated at 10 mM, was 4.6 mM. Nine other neutral amino acids (L-alanine, L-serine, L-cysteine, glycine, L-proline, L-homoserine, L-leucine, L-phenylalanine and L-glutamine), and an acidic amino acid (L-aspartate) were also tested at 10 mM and, although several caused inhibition, the Ki was always at least 20 times higher than the measured Km for L-lysine. It is concluded the carrier is highly specific for the L-form of the basic amino acids. The sodium dependence of L-lysine influx was investigated over a range of L-lysine concentrations (0.05-1 mM), and total removal of sodium from the perfusate had no effect on L-lysine influx. In the presence of sodium, L-homoserine, an amino acid not normally present in animal tissues, inhibited L-lysine influx (Ki = 13 mM). This inhibition was not observed in the absence of sodium, and contrasts with the observation that the inhibitory action of L-histidine was sodium independent. The present data suggest that a specific cationic amino acid transport system is operative in the basolateral membrane of the cat salivary epithelium. The properties of this system appear to be similar to the system y+ which has been described in several other cell types.

1. The unidirectional influx of amino acids into the guinea-pig syncytiotrophoblast was measured using a single circulation paired-tracer dilution technique which allows separate characterization of both fetal and maternal interfaces. An in situ preparation perfused through the fetal circulation was used to examine the fetal side, while an isolated preparation perfused through both the fetal and maternal circulations was used to study both interfaces simultaneously. 2. On the fetal side the maximal uptake (Umax) determined at tracer concentrations was high for the short-chain neutral amino acid alanine (76%) and the long-chain neutrals, leucine (75%), phenylalanine (90%) and tyrosine (82%) and for the basic amino acid lysine (65%). In contrast, Umax was negligible for α-methylaminoisobutyric acid and taurine, a β-amino acid. 3. The uptake of alanine and phenylalanine on the fetal side was inhibited by both short-chain (alanine, serine, cysteine) and long-chain (phenylalanine, methionine, leucine) neutral amino acids. d-alanine had no effect on l-alanine uptake whereas d-phenylalanine significantly inhibited that of l-phenylalanine. Diaminobutyric acid, lysine and arginine were effective inhibitors of alanine uptake but had no effect on phenylalanine uptake. 4. On the maternal side uptake of alanine, phenylalanine and lysine was measured. Over a wide range of concentrations self-inhibition of alanine influx was similar to the cross-inhibition observed with phenylalanine. In contrast, the influx of phenylalanine, which was strongly self-inhibited, was only partially cross-inhibited by alanine. 5. Influx of alanine and phenylalanine was measured at various perfusate concentrations and was found to be saturable on both maternal and fetal sides. The data were fitted to a single hyperbola and, on the maternal side, the Km for alanine (10.3±2.7 m m, mean± s.e., n = 3) was three-fold higher than the value measured for phenylalanine (3·1±0·8 m m). On the fetal side the Km values for alanine (8·4±1·4 m m, n = 4) and phenylalanine (11·9±1·9 m m, n = 3) were similar. 6. The uptake of alanine, phenylalanine and lysine appeared to be highly sodium-dependent accounting for 40-70% of the total influx. However, the inhibited fractions were found to be different on the two sides of the placenta. 7. The results of uptake, cross-inhibition and Na+-dependency experiments suggest the presence of an alanine-serine-cysteine (ASC) type system and a leucine (L) type system with markedly overlapping specificities at both the fetal and maternal interfaces. Separate kinetic characterization of a two carrier system was not possible under the conditions of these experiments. However, kinetic parameters for the over-all transport of alanine and phenylalanine were measured.

Organic and inorganic reagents have been tested for destruction or inactivation of the aflatoxins present in peanut and cottonseed meals. The treatments were made in a special laboratory-scale reactor, and were evaluated by determination of the aflatoxins in the products by thin layer chromatography. In some instances, a larger pilot-plant scale reactor was used. Ammonia, methylamine, sodium hydroxide and formaldehyde reduced aflatoxin levels and appear practical for large scale treatments. Effects of various reaction parameters including time, temperature and moisture content on the efficiency of these reagents are presented.

Samples of completely refined peanut oil, semirefined pecan oil, imported edible grade olive oil and crude safflower seed oil have been examined for composition, spectral transmittance and other properties. Compositions were determined by means of the modified Bertram oxidation method and application of the iodine-thiocyanogen number technique.

On the basis of spectrophotometric data on the characteristic spectral absorptions observed in oxidizing substrates with and without added γ-tocopherol, it has been shown that the red color which develops on accelerated oxidation of fat substrates containing this tocopherol results from its conversion to chroman-5,6-quinone. The chroman-5,6-quinone thus produced possesses relatively little antioxidant activity.

Eleven cottonseed meals have been prepared by batch extractions of a given lot of cottonseed with various acetone-hexane-water mixtures using several different extraction schedules. These meals, together with eight meals of commercial origin and a commercial soybean meal, have been subjected to chemical evaluation and assayed for protein quality using the growing chick as a test animal. In general the acetone-hexane-water meals were superior to the commercial cottonseed meals for promoting the growth of the chicks and, considering all the meals, a linear correlation was obtained between the logarithm of the weight gains and the available lysine contents of the meals. The free and total gossypol contents of the cottonseed meals appeared to have little or no influence on the growth rates.

A peanut meal contaminated with ca. 600 ppb aflatoxins was treated with formaldehyde alone and in combination with calcium hydroxide in a benchscale reactor, operated both sealed and at atmospheric pressure. In general, thin layer chromatographic assays revealed that addition of calcium hydroxide to formaldehyde caused greater inactivation of the toxins than did formaldehyde alone. With the reactor sealed and 25% moisture in the meal, treatments for 1 hr with 0.5% and 1.0% formaldehyde plus 2.0% calcium hydroxide yielded products having 3 and 1 ppb aflatoxins, respectively, whereas under reflus at atmospheric pressure with 20% meal moisture, 1 hr treatment with 1.0% calcium hydroxide yielded a product with 5 ppb aflatoxins.

A detailed mathematical model for polymerization of olefins over solid catalysts, the Multigrain Model, is used as the basis for this study. This model takes account of the micros trueture of the growing particle and considers diffusion of monomer in two distinct regimes. Using the model, polymerization rate behavior of the growing polymer particles in gas or slurry polymerization may be predicted. Criteria for the importance of intra particle monomer diffusion and heat transfer under conditions of industrial interest are presented graphically. In addition, the mass and heat transfer resistances in the external film are evaluated for catalysts of varying activity. The effects of mass and heat transfer and catalyst decay on reaction rate profiles, activation energies and particle overheating phenomena are illustrated.

Amino acid tmransport at the luminal side of brain capillary endothelium and the basal side of salivary epithelium were compared using a nondestructive, first-circulation, paired-tracer dilution method. In the brain, the reference molecule was an intravascular marker (Crone's method) whereas inthe salivary gland the reference was an extracellular marker of similar size to the test molecule. The unidirectional flux was related to a maximal uptake, U: U = 1 --test concentration/reference concentration Uptake and cross-inhibition experiments in brain suggest the presence of only a long-chain neutral L-amino acid transport system (leucine, phenyl-alanine, tryptophan, tyrosine, valine, methionine). In contrast, results in the salivary gland suggest 4 transport systems: 1. large neutral, 2. small neutral (alanine, serine), 3. basic (lysine) and 4. acidic (aspartic, glutamic). The same method was applied to localize binding sites. Ouabain bound very significantly to the salivary epithelium but not at all to the brain endothelium. The method described is of interest since it can be extended to any organ and possibly to man.
