Amalgam has been widely used as a restorative dental material for over 150 years. Most standard dental amalgam formulations contain approximately 50% elemental mercury in a mixture of copper, tin, silver, and zinc. Mercury is a highly volatile metal, which can easily vaporize to a colorless and odorless gas. It has been demonstrated that mercury is released from dental amalgam, which is increased by chewing, eating, brushing, and drinking hot liquids. Besides this, amalgam is the main occupational exposure source of mercury for dental workers. It is known that mercury exposure causes immune modulation in humans. In this study, it was aimed to evaluate the changes in neopterin levels and tryptophan (Trp) degradation in dental technicians. It was observed that low levels of occupational mercury exposure resulted in decreased neopterin, kynurenine (Kyn), and Kyn/Trp levels. Moreover, mercury and neopterin levels had a significant positive correlation in workers. The lower neopterin levels and Kyn/Trp in dental technicians compared to an unexposed group indicates a possible immune suppression with low level of occupational mercury exposure during amalgam preparation. The relationship between urinary mercury levels as an indicator of occupational mercury exposure and neopterin reminded an effect on T-cell-mediated immune response.

Psoriasis is a chronic and non-transient disease with increased epidermal proliferation in the skin. Dysregulation of the immune system is an important factor in this pathology. Inflammation markers, pro-inflammatory cytokines, and immune cells are reported to be changed in psoriasis.

Background. There is a need to identify the complex interplay between various physiological mechanisms in primary ciliary dyskinesia (PCD) and cystic fibrosis (CF). The study investigated the interaction between respiratory function, exercise capacity, muscle strength, and inflammatory and oxidant/antioxidant responses in patients with PCD and CF. Methods. The study included 30 PCD patients, 30 CF patients, and 29 age and sex-matched healthy subjects. Exercise capacity was assessed using the modified shuttle walk test (MSWT). Handgrip strength (HGS) was used to evaluate general muscle strength. Oxidative stress-inflammatory parameters were also assessed. Pulmonary function test was performed by spirometry. Regarding the forced expiratory volume in 1 second (FEV1) z-score, patients with PCD and CF were subdivided into normal, mild, and severe/moderate groups. Results. Forced vital capacity (FVC) z-scores were lower in PCD and CF patients than controls. FEV1, FEV1/FVC, peak expiratory flow (PEF), and forced mid expiratory flow (FEF25-75%) z-scores were lower in PCD than in the other groups. HGS was lower in both mild PCD and normal CF patients relative to the controls. MSWT distance was lower in severe/moderate PCD patients than controls. Catalase (CAT), glutathione S-transferase (GST), glutathione peroxidase (GPx), and malondialdehyde (MDA) levels did not differ significantly among the study groups, but superoxide dismutase (SOD) level in severe/moderate PCD, and glutathione (GSH) level in normal CF were higher than in controls. Interleukin-6 (IL-6) level was higher in patients with normal PCD and CF compared to the controls. IL-1ß level was higher in PCD compared to controls. Additionally, correlations among these parameters were also determined in some patient groups. Conclusion. Homeostasis related to respiratory function, aerobic performance, muscle strength, inflammatory response, and oxidant/antioxidant balance were affected in PCD and CF. The main goal of the present study was to detect neopterin concentrations in human tear samples and to evaluate its potential correlation with serum neopterin levels. For this purpose, 20 systemically healthy volunteers were recruited, and both tear and serum samples were synchronically collected from each individual. Enzyme-linked immunoassay (ELISA) was carried out to detect the quantity of neopterin in the samples. Mean human tear neopterin levels were observed as 3±0.56 nM while mean serum neopterin was 9±1.25 nM. Additionally, a significant positive correlation between tear neopterin and serum neopterin concentrations was observed. This is the first report to show neopterin concentration in human tears as a biological sample. Collecting tears from the individuals is a non-invasive sampling method, and as an analytical aspect detection of neopterin by ELISA in tear samples construct a valuable, practical and cheap procedure for the diagnosis and monitoring of intraocular inflammation and systemic immune-mediated diseases.

Physiologically, zinc is an essential element that has many important functions such as maintenance of immunity, cell integrity, signalling, cellular respiration, and neuromodulation. Besides zinc, folate also plays a crucial role in basic cellular reactions. Exposure to some xenobiotics may lead to folate deficiency. The aim of this study was to evaluate the effect of zinc fume on erythrocyte folate levels. For this purpose, erythrocyte folate levels and serum zinc concentrations were evaluated in 63 galvanization workers, who were exposed to zinc fume during the galvanization procedure, and 23 office personnel as a control group. End-stage renal disease (ESRD) is the inability of the kidneys to remove waste products from the blood. The most important factors causing ESRD that require hemodialysis are diabetes and hypertension. There are limited numbers of studies to evaluate tetrahydrobiopterin pathway in these patients. The aim of the study was to evaluate tetrahydrobiopterin pathway by measuring its important components, biopterin to creatinine concentrations and dihydropteridine reductase activities in diabetes and hypertension patients treated with/without hemodialysis. The patients undergoing hemodialysis were classified as diabetic nephropathy (n=21), hypertensive nephropathy (n=20) and others (n=30), while the controls consisted of healthy subjects (n=21), diabetic subjects (n=23) and hypertensive subjects (n=22) without any renal disorder. In transfusions, safety measures are used for the health of both receivers and donors. In this regard, early and sensitive markers are strongly needed. The aim of this study was to evaluate neopterin profile and tryptophan degradation in a group of Turkish blood donors. The effect of age, gender, blood type and Rh factor were also assessed. Neopterin was measured by ELISA kits. Serum tryptophan and kynurenine were determined by high performance liquid chromatography. Kynurenine/tryptophan (kyn/trp) was calculated to estimate tryptophan degradation. Tryptophan, kynurenine and kyn/trp levels were found to be higher in males compared with females (all p0.05). In routine analysis, early and reliable biomarkers are needed. In parallel with current markers, neopterin can be used to select safe donors and to maintain the safety of transfusion. In the face of our results, we recommend that donations with neopterin levels above 19 nmol/L should be excluded from the program because increased neopterin levels indicate activation of the cellular immune system.

Neopterin, as a non-conjugated pteridine, is synthesized from guanosine triphosphate and its production is upregulated upon the activation of cellular immune response. Alterations of pteridines in body fluids are known to correlate well with existing diseases and stages, prognosis, clinical outcomes and survival data. It is of advantage to have a pteridine database of healthy volunteers to determine normal values. Thereby, especially in children there is no detailed study on pteridine levels. The aim of this study is to initiate the establishment of pteridine database of healthy children in our country. Serum neopterin levels were analysed by enzyme-linked immunosorbent assay. Urinary neopterin and biopterin levels and serum kynurenine, tryptophan levels and kynurenine/tryptophan ratio as an estimate of tryptophan breakdown were assessed with high-pressure liquid chromatography in serum and urine samples of 55 children aged between 3 months and 10 years. The results were evaluated within the subgroups of different ages and sex. Pteridine pathway components were found to be higher in children compared to adults. Higher levels of pteridine pathway components observed within the first years of life may reflect the rapid maturation of the immune system, and environmental adaptation and/or insufficiency of defence systems. On the other hand, it may also relate to a higher frequency of infections not (yet) manifested clinically.

Biopterin as a stable metabolite is produced by oxidation of 5,6,7,8-tetrahydrobiopterin (BH4). It is known that many diseases may cause changes in BH4 concentration and/or dihydropteridine reductase (DHPR) enzyme activity. There is only a limited number of studies correlating DHPR activity in malignancies. The main goal of the present study was to evaluate alterations in the DHPR activity and biopterin levels in patients with breast and thyroid cancers. The breast cancer patients (n=24) were compared to patients with benign breast diseases (n=19) and controls (n=30). In addition; the patients with thyroid cancer (n=17) were compared to patients with benign thyroid diseases (n=42) and the control group. This survey was conducted to investigate the contamination by multiple mycotoxins, aflatoxins (AFB1, AFB2, AFG1, and AFG2), ochratoxin A (OTA), and zearalenone (ZEA) in 61 samples of maize and 17 commercial animal feed samples, and of aflatoxin M1 (AFM1) in raw dairy milk samples (n = 20) collected from Assiut City in Upper Egypt. Multi-mycotoxin immunoaffnity columns were used for samples cleanup and mycotoxin purification. An HPLC-FLD system with an on-line post-column photochemical derivatization was used for the detection of the target toxins. AFB1 was detected in both maize (n = 15) and feed (n = 8), with only one maize sample presenting a concentration above the maximum permissible level set by the Egyptian authorities. AFB2 was observed in six maize samples and in one feed sample, with a maximum value of 0.5 µg/kg. ZEA was detected only in feed samples (n = 4), with a maximum value of 3.5 µg/kg, while OTA, AFG1, and AFG2 were under the limits of detection. For milk, all the analyzed samples (100%) were contaminated with AFM1, and 14 samples (70%) presented concentrations above the maximum permissible level in the European Union (EU) (0.05 µg/kg). The concentrations ranged from 0.02 µg/kg to 0.19 µg/kg, except that of one sample, which was under the limit of quantification. The contamination rates in maize and animal feeds are not alarming. In contrast, the consumption of dairy milk samples in Assiut City may pose public health hazards, as AFM1 levels were found to exceed the international permissible limits. Further surveys are highly recommended in order to establish a database for mycotoxin occurrence in Egypt to minimize the possible health risks in animals and humans. [ABSTRACT FROM AUTHOR]

Background: The aim of this study was to evaluate the degradation of tryptophan (Trp), neopterin production and antioxidant capacity in patients with benign and malignant thyroid disease. Methods: For this reason, the levels of tryptophan, kynurenine (Kyn) and neopterin, and superoxide dismutase (SOD) and catalase (CAT) enzyme activities in 67 thyroid patients were evaluated in our study and the results were compared with 30 healthy controls. Results: Tryptophan and kynurenine levels in thyroid patients decreased compared to the control group. Patients with thyroid disease had lower CAT activity than the control group. The neopterin and tryptophan levels in malignant and benign patients were also significantly different. Conclusion: The results of the present study suggest that thyroid disorders may lead to changes in tryptophan degradation, neopterin production and CAT enzyme activities. [ABSTRACT FROM AUTHOR]
