Prion hastalıkları, insanları ve hayvanları etkileyen ölümcül nörodejeneratif hastalıklardır. İnsanlarla yakın temasta olan kedilerin prion proteinindeki varyasyonlar ve bunların prion proteini üzerindeki etkileri iyi bilinmemektedir. Bu çalışmada sokak kedilerinde prion protein genindeki varyasyonların araştırılması ve biyoinformatik araçlar kullanılarak FSE ile ilişkili varyasyonların değerlendirilmesi amaçlanmıştır. Bunun için kedi DNA örnekleri (n:258) prion protein genini hedefleyen bir PCR tarafından incelendi ve dizilendi. Varyasyonların prion proteini üzerindeki etkileri biyoinformatik araçlarla tahmin edildi. Daha sonra bu polimorfizmlerin protein üç boyutlu yapısı üzerindeki etkileri ve bu polimorfizmlerin kedi nöral hücre adezyon molekülü 1 (NCAM1) ile etkileşimleri analiz edildi. Elde edilen sonuçlara göre, analiz edilen DNA örneklerinden 25 dizi verisi elde edilmiştir. Bu dizi verileri arasında SNP'ler olmak üzere 10 farklı varyasyon ve 108 bp uzunluğunda bir insersiyon/delesyon polimorfizmi tespit edildi. Tespit edilen SNP'lerden beşi (c314A>G, c.454T>A, c.579G>C, c.642G>C ve c.672G>C) ilk kez tespit edildi. Biyoinformatik bulgular, c.579G>C (Q193H) varyasyonunun yüksek amiloid eğilimleri ile birlikte prion proteini üzerinde zararlı etkileri olduğunu göstermiştir. Ayrıca c.454T>A (Y152N) ve c.457G>A (E153K) prion proteini üzerinde zararlı etkilere sahipti. Sonuç olarak, bu çalışma sokak kedilerinin prion protein varyasyonlarını ve bunların prion proteini üzerindeki zararlı etkilerini ilk kez ortaya koymaktadır., Prion diseases are deadly neurodegenerative diseases that affect humans and animals. Variations in the prion protein of cats in close contact with humans and their effects on prion protein are not well known. In this study, it was aimed to investigate the variations in the prion protein gene in stray cats and to evaluate the variations associated with FSE using bioinformatics tools. For this, cat DNA samples (n:258) were analyzed and sequenced by a PCR targeting the prion protein gene. The effects of variations on the prion protein were estimated by bioinformatics tools. Then, the effects of these polymorphisms on the three-dimensional structure of the protein and the interaction of these polymorphisms with cat neural cell adhesion molecule 1 (NCAM1) were analyzed. According to the results obtained, 25 sequence data were obtained from the analyzed DNA samples. Among these sequence data, 10 different variations, including SNPs, and a 108 bp long insertion/deletion polymorphism were detected. Five of the detected SNPs (c314A>G, c.454T>A, c.579G>C, c.642G>C and c.672G>C) were detected for the first time. Bioinformatics findings showed that the c.579G>C (Q193H) variation has deleterious effects on prion protein with high amyloid affinities. Also, c.454T>A (Y152N) and c.457G>A (E153K) had deleterious effects on the prion protein. As a result of interaction analysis, S105R variant showed the highest affinity for NCAM1 protein, while 108 bp deletion and Q224H variant did not interact with NCAM1 protein. In conclusion, this study reveals for the first time the prion protein variations of stray cats and their deleterious effects on prion protein.
