11. Antimicrobial Susceptibility Patterns of Staphylococcus aureus in Decubitus Ulcer Infections.

Abstract

Objective: Decubitus ulcers, also called bedsores or pressure ulcers, are skin and soft tissue injuries caused by sustained or prolonged pressure on the skin. Staphylococcus aureus (S. aureus) is one of the most frequently isolated bacteria in patients with decubitus ulcer infection. In this study, we aimed to retrospectively determine the antimicrobial susceptibility profile of S. aureus strains isolated from wound swab cultures of patients hospitalized in various wards. Methods: Wound swab cultures were examined from patients with clinical signs of decubitus ulcer infection during inpatient treatment in different wards with various diagnoses between January 2009 and October 2019. In these samples, the antimicrobial susceptibility profile data of 132 S. aureus strains belonging to 132 different patients who were considered clinically significant were included in our study. Results: Among all specimens, 132 (13.83%) S. aureus positivity cases were included in our study. The methicillin-resistant (MRSA) rates were 43.85% (24/56) in male patients and 57.14% (32/56) in female patients with decubitus ulcer infection. Of the S. aureus strains, 42.42% (56/132) were MRSA and 57.58% (76/132) were methicillin-susceptible (MSSA). Linezolid was found to be the most effective antibiotic among MRSA strains, whereas all MSSA strains were susceptible to amoxicillin + clavulanic acid, rifampicin, and cefoxitin. Conclusion: Antimicrobial resistant strains such as MRSA may be encountered in half of S. aureus infections and may complicate treatment options. 29. Development and Validation of a Simple and Selective Chromatographic Method for Quantification of Ceramide-NP in Skin-Simulating Liposome Formulations.

Abstract

Numerous topical products containing ceramide to restore the skin barrier function have been available on the market in recent years. A simple, rapid, and selective analytical method could contribute to the development of innovative formulations such as skin-simulating liposomes, which contain ceramides as stratum corneum components. The similarity of the physicochemical structure of ceramide and lipid excipients in liposome formulation complicates the separative and quantitative analysis. Therefore, this study aims to develop and validate a selective high-pressure liquid chromatography (HPLC) method for the simultaneous quantification of lipids in ceramide-containing skin simulating liposome formulations. The separation was performed with a reversed-phase C18 column (150 mm x 4.6 mm, 5 µm), the mobile phase system consisted of methanol: acetonitrile (60:40, v/v), the flow rate of 0.5 ml/min, the oven temperature at 45°C, injection volume of 10 µL, and UV detection at 210 nm. The method was linear (r° > 0.99) in the range of concentration from 80 µg/mL to 480 µg/mL with acceptable precision, accuracy, and selectivity for the liposome formulations. The limit of detection (LOD) values of ceramide NP (Cer-NP) was 7.90 μg/mL. The limit of quantification (LOQ) values of Cer-NP was 24.06 µg/mL. The study indicates that the method is efficient and has accuracy and selectivity for the simultaneous quantification of Cer-NP in skin-simulating liposome formulations. [ABSTRACT FROM AUTHOR]

Abstract

The Preformed Particle gels (PPGs) have widely been used and injected in low permeability rich oil zones as diverting agents to solve the conformance issues, distract displacing fluid into out of sorts swept zones. Besides, they also reduce the permeability of thief zones and high permeability fractured zones. However, the PPG propagation and plugging mechanism are still unpredictable and sporadic in manifold void space passages. PPGs have two main abilities, first, it increases the sweep efficiency, and second, it decreases the water production in mature oilfields. However, the success or failure of PPG treatment largely depends on whether it efficiently decreases the permeability of the fluid paths to an expected target or not. In this study, the different factors were studied that affect the performance of PPG in such reservoirs. PPGs were treated in different ways; treated with brine, low salinity, and high salinity brine. Also, their impacts were investigated in low/high permeability, fractured reservoirs, and void space conduit models as well. From the literature, it was revealed that the sweep efficiency can be improved through PPG, but not displacement efficiency and little impact of PPG were found on displacement efficiency. On the other hand, Low salinity water flooding (LSWF) can increase the displacement efficiency but not sweep efficiency. Hence, based on the above issues, few new techniques and directions were introduced in this study for better treatment of PPG to decrease water cut and increase oil recovery.

Abstract

Nanocrystal technology enables the production of solid drug particles with a particle size below 1000 nm without the need for any carrier system. By reducing the size of the particles of drugs with low solubility in water, increasing the surface area and decreasing the thickness of the diffusion layer leads to an increase in k the solubility rate. Accordingly, the increasing concentration gradient in the absorption zone promotes permeation and absorption by passive diffusion between the intestinal lumen and blood. Therefore, it is very important to improve and / or increase their bioavailability by using the nanocrystal technology approach for drug molecules belonging to BCS Class II and IV. Top-down and bottom-up methods are used to obtain nanometer-sized drug particles. In the pharmaceutical industry, top-down methods, which are subdivided into wet milling (BWM) and high pressure homogenization (HPH), are preferred in the ball mill because of its ease of application, repeatability and scalability. With nanocrystal technology, it is aimed to create new dosage forms by providing less side effects, lower doses and faster onset of action of drug molecules that are already approved in the pharmaceutical industry. It is also aimed that new drug molecules can be formulated with better bioavailability., Nanokristal teknolojisi partikül boyutu 1000 nanometre (nm)’nin altında, herhangi bir taşıyıcı sisteme ihtiyaç duymadan katı ilaç partikülerinin üretilmesini sağlar. Sudaki çözünürlüğü düşük ilaçların partiküllerinin boyutunun küçültülmesi ile, yüzey alanlarının artması ve difüzyon tabakasının kalınlığının azaltılması k çözünürlük hızının da artışına yol açar. Buna bağlı olarak, absorpsiyon bölgesinde artan konsantrasyon gradienti bağırsak lümeni ve kan arasındaki pasif difüzyon yoluyla permeasyonu ve emilimi teşvik etmektedir. Dolayısıyla Biyofarmasötik Sınıflandırma Sistemi (BCS) Sınıf II ve IV’e ait ilaç molekülleri için nanokristal teknolojisi yaklaşımını kullanarak biyoyararlanımlarını geliştirmek ve/veya arttırmak oldukça önemlidir. Nanometre boyutunda ilaç partikülü elde edebilmek için yukarıdan aşağıya (top-down) ve aşağıdan yukarıya (bottom-up) yöntemlerinden yararlanılmaktadır. İlaç endüstrisinde uygulama kolaylığı, tekrar edilebilirliği ve ölçeklendirilebilmesi nedeniyle bilyeli değirmende yaş öğütme (BWM) ve yüksek basınçlı homojenizasyon (HPH) olarak alt bölümlere ayrılan yukarıdan aşağıya yöntemleri tercih edilmektedir. Nanokristal teknolojisi ile ilaç endüstrisinde hâlihazırda tedavide onaylanmış olan ilaç moleküllerinin daha az yan etki, daha düşük dozlar ve daha hızlı etki başlangıcı sağlayarak yeni dozaj formlarının geliştirilmesi ve yeni ilaç moleküllerinin daha iyi bir biyoyararlanımla formüle edilebilmesi amaçlanmaktadır.

Abstract

In this study, naftifine (a topical antifungal drug) loaded poly(vinyl) alcohol (PVA)/sodium alginate (SA) nanofibrous mats were prepared using the single-needle electrospinning technique. The produced nanofibers were crosslinked with glutaraldehyde (GTA) vapor. The morphology and diameter of the electrospun nanofibers were studied by scanning electron microscopy (SEM). SEM images showed the smoothness of the nanofibers and indicated that the fiber diameter increased with crosslinking and drug loading. Atomic force microscopy (AFM) images confirmed the uniform production of the scaffolds, and elemental mapping via energy dispersive X-ray spectroscopy (EDS) showed the uniform distribution of the drug within the nanofibers. An attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy study demonstrated that naftifine has sufficient secondary interactions with the polymer blend. The crosslinking treatment decreased the burst drug release effectively and the release mechanism followed Korsmeyer-Peppas Super Case-II transport. Overall, these findings suggest the potential use of naftifine-loaded PVA/SA nanofibers as a topical antifungal drug delivery system.
