400 p., En la presente Tesis Doctoral se han desarrollado nuevos agentes terapéuticos y/o de diagnóstico eficaces y selectivos contra el cáncer de mama ductal invasivo triple negativo y/o las infecciones bacterianas en heridas y quemaduras infectadas. Los agentes desarrollados, se enmarcan dentro de la subclase de los nanomateriales, y se han diseñado desde una perspectiva de la Química Inorgánica.La estrategia seguida en el presente trabajo, se ha basado en el desarrollo de nanopartículas de sílice como sistemas de liberación de fármacos, que han permitido la incorporación de distintos fragmentos elegidos de manera racional para cumplir alguna de las siguientes funciones: metales como agentes citotóxicos, fragmentos orgánicos capaces de coordinar dichos metales, fragmentos luminiscentes para proveer al material de propiedades diagnósticas, fragmentos de focalización para diseñar materiales selectivos y fragmentos con función proliferativa para mejorar la prognosis de heridas y quemaduras infectadas. Así, se han sintetizado 30 nuevos nanomateriales no clásicos de liberación de fármacos, basados en nanopartículas de sílice funcionalizadas, de los cuales 25 son agentes de targeting pasivo, mientras que 5 lo son de targeting activo, además de ser teragnósticos.

Leishmaniasis and Chagas disease are still considered neglected illnesses due to the lack of investment in research, despite the fact that almost one million new cases are reported every year. Four 7-oxo-5-phenyl-1,2,4-triazolo[1,5-a]pyrimidine (HftpO) first-row transition complexes (Cu, Co, Ni, Zn) have been studied for the first time in vitro against five different species of Leishmania spp. (L. infantum, L. braziliensis, L. donovani, L. peruviana and L. mexicana) as well as Trypanosoma cruzi, showing higher efficacy than the reference commercial drugs. UV and luminescence properties were also evaluated. As a proof of concept, anchoring of a model high-effective-metal complex as an antiparasitic agent on silica nanoparticles was carried out for the first time, and drug-release behaviour was evaluated, assessing this new approach for drug vehiculation.

Calcium carbonate, one of the most commonly found biominerals produced by organisms, has shown great potential for the development of systems with biological applications due to its excellent biocompatibility, biodegradability, and simple chemical composition. Here, we focus on the synthesis of various carbonate-based materials with vaterite phase control and their subsequent functionalization for applications in treating glioblastoma, one of the most limiting tumors currently without effective treatments. The incorporation of l-cysteine into the systems increased cell selectivity while the incorporation of manganese supplied the materials with cytotoxic capacity. Extensive characterization of the systems by infrared spectroscopy, ultraviolet-visible spectroscopy, X-ray diffraction, X-ray fluorescence, and transmission electron microscopy confirmed the incorporation of the different fragments causing selectivity and cytotoxicity to the systems. To verify their therapeutic activity, the vaterite-based materials were tested in the CT2A cell line (murine glioma) and compared to SKBR3 (breast cancer) and HEK-293T (human kidney) cell lines. These studies on the cytotoxicity of the materials have shown promising results that can encourage future in vivo studies in glioblastoma models.

[ Abstract] The pharmacological activity of organotin(IV) complexes in cancer therapy is well recognized but their large applicability is hampered by their poor water solubility. Hence, carbon dots, in particular nitrogen-doped graphene quantum dots (NGQDs), may be a promising alternative for the efficient delivery of organotin(IV) compounds as they have a substantial aqueous solubility, a good chemical stability, and non-toxicity as well as a bright photoluminescence that make them ideal for theranostic applications against cancer. Two different multifunctional nanosystems have been synthesized and fully characterized based on two fragments of organotin-based cytotoxic compounds and 4-formylbenzoic acid (FBA), covalently grafted onto the NGQDs surface. Subsequently, an in vitro determination of the therapeutic and theranostic potential of the achieved multifunctional systems was carried out. The results showed a high cytotoxic potential of the NGQDs-FBA-Sn materials against breast cancer cell line (MDA-MB-231) and a lower effect on a non-cancer cell line (kidney cells, HEK293T). Besides, thanks to their optical properties, the dots enabled their fluorescence molecular imaging in the cytoplasmatic region of the cells pointing towards a successful cellular uptake and a release of the metallodrug inside cancer cells (NGQDs-FBA-Sn).

La sociedad actual se presenta cada día más tecnificada y los estudiantes están creciendo en ella desarrollando distintas capacidades cognitivas que resultan casi imprescindible comprender y atender para poder aumentar su interés en el aprendizaje de diferentes temas. Esta realidad está llevando los profesores a reconfigurar sus estrategias pedagógicas, actualizando los métodos educativos con el fin de mejorar la calidad de la educación adaptando los contenidos que se imparten con las necesidades de la sociedad. Por ello, cada vez es más frecuente recurrir a nuevas herramientas como las TIC (Tecnologías de la Información y las Comunicaciones) y a aplicaciones lúdicas que apoyen y fomenten el proceso de aprendizaje. La aplicación de las TICs en la enseñanza, ha supuesto un avance en la transmisión y recepción del conocimiento para incentivar a los alumnos al aprendizaje de aquellas asignaturas que suponen una mayor dificultad durante su proceso formativo. Asimismo, la gamificación, que consiste en incorporar elementos de juegos como las recompensas y la competición para animar a las estudiantes a realizar tareas que normalmente se consideran aburridas, está ganando cada día más atención sobre todo gracias a su innegable carácter motivacional. De hecho, el juego es un activador en la atención y surge como alternativa para complementar los esquemas de la enseñanza tradicional. Debido a su naturaleza intrínseca, el aprendizaje de la física y de la fisicoquímica (tanto en campo farmacéutico como en general) suele resultar un proceso complicado y muchas veces difícil de entender. Los métodos tradicionales de enseñanza de la ciencia en general, y de la física y fisicoquímica en particular, hacen que estas disciplinas fundamentales sean vistas por los alumnos como algo abstracto y poco estimulante. Antibiotic resistance is a global problem and bacterial biofilms contribute to its development. In this context, this study aimed to perform the synthesis and characterization of seven materials based on silica mesoporous nanoparticles functionalized with three types of fluoroquinolones, along with Cu2+ or Ag+ species to evaluate the antibacterial properties against Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, and Pseudomonas aeruginosa, including clinical and multi-drug-resistant strains of S. aureus and P. aeruginosa. In addition, in order to obtain an effective material to promote wound healing, a well-known proliferative agent, phenytoin sodium, was adsorbed onto one of the silver-functionalized materials. Furthermore, biofilm studies and the generation of reactive oxygen species (ROS) were also carried out to determine the antibacterial potential of the synthesized materials. In this sense, the Cu2+ materials showed antibacterial activity against S. aureus and E. coli, potentially due to increased ROS generation (up to 3 times), whereas the Ag+ materials exhibited a broader spectrum of activity, even inhibiting clinical strains of MRSA and P. aeruginosa. In particular, the Ag+ material with phenytoin sodium showed the ability to reduce biofilm development by up to 55% and inhibit bacterial growth in a “wound-like medium” by up to 89.33%.
