RESUMEN Identificar la célula productora de un metabolito bioactivo permite enfocar los esfuerzos de cultivo celular para su producción in-vitro, así como elucidar la posible función fisiológica del metabolito. La esponja marina del Caribe Discodermia dissoluta produce el policétido (+)-discodermólido, un potente antitumoral que ha llegado a fase clínica de investigación en humanos. En este trabajo se avanzó en la identificación de las células productoras mediante disociación celular, separación en fracciones por centrifugación en gradientes de densidad de Percoll® y detección de la presencia de (+)-discodermólido mediante cromatografía de capa delgada. Las fracciones mostraron células de diferentes características morfológicas, entre las que se encuentran cianobacterias, bacterias filamentosas, coanocitos, arqueocitos, entre otras. Se pudo identificar un patrón irregular pero consistente de producción de (+)-discodermólido en las fracciones que contenían uno o más tipos de células granulares de la esponja, indicando que la producción del metabolito parece estar asociada con la esponja y no con los microorganismos asociados. ABSTRACT Identification of bioactive metabolite cell producers allow driving cell culture experiments for in vitro production, just as elucidation of the possible physiologic function of metabolites. The Caribbean marine sponge Discodermia dissoluta produces the polyketide (+)-discodermolide, a potent antitumoral that has reached clinic trials in humans. In this research, progress was made in identifying the producing cells by cell dissociation, separation into fractions by centrifugation on Percoll® density gradients, and detection of (+)-discodermolide presence by thin layer chromatography. The fractions recovered showed cells of different morphological characteristics, including cyanobacteria, filamentous bacteria, choanocytes, archaeocytes, among others. It was possible to identify an irregular but consistent pattern of (+)-dicordemolide production in the fractions that contained one or more kind of sponge granular cells, indicating that metabolite production seems to be related to the sponge and not with the associated microorganisms.

Caranx hippos es uno de los recursos ícticos marinos más importantes en Colombia, especialmente en la pesca artesanal. Con el propósito de profundizar en su conocimiento biológico, se estimó la fecundidad parcial y el diámetro de los ovocitos. Las muestras provenían de faenas artesanales comerciales. A cada individuo se le midió longitud total, el peso total y el peso gonadal. La fecundidad se estimó combinando el método gravimétrico con análisis de imágenes, usando el software de libre acceso Image J®. A partir de la medición de 149523 ovocitos se obtuvo un diámetro promedio de 0.32 ± 1.24E-4 mm, y valores de fecundidad variables con un ámbito entre 291439 y 4380480 ovocitos/gónada. No se evidenció una relación directa entre la cantidad y el diámetro de los huevos con la talla y el peso del pez, lo que sugiere que no siempre las hembras más grandes son las más fecundas. Se describe el desarrollo microestructural de los oocitos maduros, encontrando diámetros promedio en alvéolo cortical y vitelogénesis de 98.6 + 33.1 µm y 207.5+ 59.6 µm, respectivamente. Dada la importancia en el país de C. hippos, este trabajo constituyó un importante aporte al conocimiento de la especie, que puede ser usado en modelos pesqueros con aplicabilidad directa en medidas de manejo. Caranx hippos is one of the most important fish resources in Colombia, particularly in artisanal fisheries. In order to deepen C. hippos biological knowledge, fecundity (number of eggs in development) and oocyte diameter was estimated. Fish samples were collected from commercial artisanal fishing trips, each fish was measured (total length, Lt) and weighted (total weight, Pt); gonads were removed, weighed and analyzed for determining fecundity using a gravimetric method combined with image analysis performed using the open access software Image J®. From the measurement of 149552 oocytes, an average diameter of 0.32 ± 1.24E-4 mm, was obtained, while fecundity varied between 291439 and 4380480 oocytes per gonad. No direct relationship was evidenced between egg number and diameter, and fish size and weight. Mature oocyte microstructural development is described, average diameters in cortical alveoli and vitellogenesis were 98.6 + 33.1 µm y 207.5+ 59.6 µm, respectively. This result suggests that larger females are not always the more fertile. Given the importance of C. hippos in Colombia, this work is a valuable contribution to the knowledge of the species, which can be used in fishery models with direct applicability to management procedures.

Se estudió la influencia que ejerce la dieta sobre el desarrollo larvario del pectínido Argopecten nucleus, probando seis dietas microalgales compuestas por dietas monoalgales y mezcla de las especies Isochrysis galbana, Chaetoceros calcitrans y Tetraselmis suecica en relación 1:1, manejando una concentración por mL equivalente al peso de 40000 células de I. galbana, midiendo la respuesta a través de las variables supervivencia, crecimiento, aparición de la mancha ocular y estado de condición (larvas llenas, semillenas y vacías). Las dietas que contenían I. galbana presentaron los mayores valores en cada una de las variables medidas, en comparación con aquellas que carecían de esta. Particularmente, la dieta mixta I. galbana + C. calcitrans presentó superioridad en todas las variables, aunque en la mayoría de los casos fue estadísticamente similar a I. galbana + C. calcitrans + T. suecica, I. galbana + T. suecica e I. galbana sola. Por el contrario, las dietas que arrojaron los resultados más bajos fueron C. calcitrans y C. calcitrans + T. suecica. Los resultados obtenidos se atribuyen principalmente a la composición nutricional, especialmente de ácidos grasos altamente insaturados (HUFA por sus siglas en inglés) de cada microalga y de cada mezcla suministrada, la cual se discute detalladamente. The influence of the diet on larval growth of the scallops Argopecten nucleus was studied, testing six microalgae diets composed by single-algae and mixed diets of the species Isochrysis galbana, Chaetoceros calcitrans y Tetraselmis suecica, handling a concentration per mL equivalent to 40000 cells of I. galbana and the response through the variables surviving, growth, appearance of the eye spot and state of condition (full, half-full and empty larvae) was measured. The diets containing I. galbana showed greater results over all the measured variables, with regard to those where it was not present. In particular, the mixed diet I. galbana + C. calcitrans presented superiority over all variables, although in the most cases was statistically similar to I. galbana + C. calcitrans + T. suecica, I. galbana + T. suecica and I. galbana alone. For the opposite, the diets with the lowest results were C. calcitrans and C. calcitrans + T. suecica. The results obtained are principally attributed to the nutritional composition mainly of high unsaturated fatty acids (HUFA) of each microalgae and each mixture supply, which is discussed in detail.

En Colombia, la industria petrolera está interesada en realizar exploración de hidrocarburos offshore, actividad que ocasiona el vertimiento de sustancias perjudiciales en áreas marinas, por lo cual es fundamental conocer los efectos que pueden ocasionar sustancias producto de estas actividades, particularmente frente a organismos nativos de las zonas de influencia. Este trabajo propone un ensayo con un organismo autóctono del Caribe colombiano, que sirva como bioindicador de contaminación, para lo cual se evaluó la toxicidad aguda de un lodo de exploración offshore en base agua sobre la fecundación del erizo de mar Lytechinus variegatus, utilizando como tóxico de referencia sulfato de cobre pentahidratado. Se estandarizó el protocolo de obtención de gametos y de fecundación, así como los ensayos con el tóxico de referencia y con el lodo, para determinar la toxicidad aguda dada en términos de concentración efectiva (CE50). El porcentaje de fecundación para validar el ensayo debe ser mayor al 85% en el tratamiento control, el cual se obtuvo al fecundar 2000 ovocitos con 50 x 10(6) espermatozoides y se contaron 200 ovocitos (10%) para determinar el porcentaje de fecundación en cada una de las réplicas. La CE50 se determinó en 20.45 ± 1.90 mg.L-1 para el tóxico de referencia y 3649 mg.L-1 ± 400 mg.L-1 correspondientes a la fase suspendida particulada del lodo la cual fue preparada a una relación lodo-agua de 1:9 (v/v). Los resultados muestran el alto grado de sensibilidad de los gametos de L. variegatus a la acción del lodo de perforación para exploración offshore de hidrocarburos, lo cual evidencia la sensibilidad del organismo de prueba ante estas sustancias y su potencial como bioindicador de contaminación. In Colombia, oil industry is interested in developing offshore hydrocarbon exploration, activity that causes spills of harmful substances in marine areas. It is essential to understand the effects that those substances may cause in native organisms in the influence zones. This manuscript proposes an assay with an autochthonous organism from the Colombian Caribbean, that may be used as a bioindicator of contamination. The acute toxicity (CE50) of a fluid used in offshore exploration was assessed on basis on the fecundity of the sea urchin Lytechinus variegatus, using as a standard toxic pentahydrated cooper sulfate. The protocol for obtaining the gametes and fecundation was previously standardized with the reference toxic which is used to guarantee and validate its results in terms of effective concentration (CE50). The fecundation percentage should be greater than 85% in order to validate the assay in the control treatment, that was carried out by fecundating 2000 oocytes with 50 x 10(6) sperms. In each replicate 200 oocytes (10%) were counted in order to determine the percentage of fecundation. The obtained CE50 value of pentahydrate copper sulphate was 20.45 ± 1.90 mg.L-1 and 3649 mg.L-1 ± 400 mg.L-1 corresponding to the suspended particulate phase of mud, which was prepared from a mud-water relationship of 1:9 (v/v). The results show a high sensitivity degree of the L. variegatus gametes to the action of the drilling mud of offshore hydrocarbon exploration, which shows the sensitivity of the test organism to these substances and its potential use as bioindicator of pollution.

Entre abril y septiembre de 2010, fue estudiado el ciclo gametogénico del pectínido Argopecten nucleus en la bahía de Nenguange, Parque Nacional Natural Tayrona. Diez organismos fueron colectados cada 14 días y se procesaron sus gónadas para histología; paralelo a esto se realizó un seguimiento de la apariencia morfocromática de la gónada a 55 individuos mantenidos en un sistema de cultivo suspendido, estableciendo cinco estadios así: I = Reposo e indiferenciación; II = Desarrollo inicial; III = Desarrollo avanzado; IV = Maduro y V = Desove. El estadio gonadal predominante fue el III con un promedio de aparición de 60%, seguido del estadio V con 20.7%, después el IV con 13.5% y finalmente los estadios II y I con 5.1 y 0.6% respectivamente. Se encontró una relación significativa entre el índice gonadosomático con la precipitación (p = 0.0001), temperatura (p = 0.0015), salinidad (p = 0.0003) y clorofila a (p = 0.0285). El factor de condición (FC) mostró una correlación con la materia orgánica particulada (MOP; p = 0.0243); sin embargo, al realizar una prueba de regresión simple la MOP no afecta de manera sustancial al FC con un índice de correlación de 4.53. Between April and September 2010, the gametogenic cycle of the scallop Argopecten nucleus was studied in Nenguange Bay in Tayrona National Park. Ten organisms were collected every 14 days and processed for histology of their gonads. Parallel to this, the morfocromatic appearance of the gonads of 55 individuals kept in a suspended culture system was followed, setting up five stages as follows: I = Rest and undifferentiation; II = Initial development; III = Advanced development; IV = Mature; and V = Spawning. The predominant gonadal stage was III with an average of 60%, followed by stage V with 20.7%, followed by IV with 13.5%, and finally stages II and I with 5.1 and 0.6% respectively. We found a significant relationship between the gonadosomatic index and precipitation (p = 0.0001), temperature (p = 0.0015), salinity (p = 0.0003), and chlorophyll a (p = 0.0285). The condition factor (CF) showed a correlation with particulate organic matter (POM; p = 0.0243), however, when performing a simple regression, the test indicated that the MOP will not substantially affect the FC with a correlation index of 4.53.

Actualmente las poblaciones silvestres de caballitos de mar del género Hippocampus se están reduciendo debido a la demanda existente en el mercado por su uso en la medicina tradicional china, su consumo en países orientales y su atractivo como especies ornamentales, estando categorizadas todas las especies del género como amenazadas. El cultivo bajo condiciones controladas surge como una opción para la conservación y uso sostenible del recurso, sin embargo, se hace necesario desarrollar técnicas adecuadas, especialmente en la etapa de levante de las crías. En el presente estudio se evaluó el efecto del alimento vivo en el crecimiento y supervivencia de crías de Hippocampus reidi. Los primeros 11 días se alimentaron con nauplios de Artemia y rotíferos enriquecidos; del día 11 al 46 únicamente con metanauplios de Artemia enriquecidos con cuatro productos comerciales: Protein Selco Plus® (PSP), alimento para peces marinos Azoo 9 en 1® (AZ), Emulsión de Scott® (ES) y microalgas Isochrysis galbana (IG). El mayor crecimiento en términos de peso y talla se presentó en los caballitos alimentados con Artemia enriquecida con PSP (160.13 mg y 4.44 cm respectivamente), el cual fue significativamente mayor que el resto de los tratamientos, los cuales se mantuvieron en un intervalo de 81.86-97.80 mg en peso y 3.62-3.72 cm en talla (F3.12 = 9.63, p = 0.0049 en peso y F3.12 = 5.12, p = 0.0288 en talla). No se observaron diferencias significativas en la supervivencia, sin embargo el mayor porcentaje se presentó en la dieta PSP (74.42%) y el menor con ES (51.60%) (F3.12 = 1.08, p = 0.4095). Seahorses natural populations are declining, due to their demand in traditional Chinese medicine, its consumption in oriental countries and their attractive as ornamental species; then, all the Hippocampus species of the genus are considered threatened fishes. Culture under controlled conditions is an option for their conservation and sustainable use; however, it is necessary to find the appropriate techniques to culture them, especially to raise the fry. In this study the effect of live food on growth and survival of Hippocampus reidi fry was evaluated. During the first 11 days they were fed with Artemia nauplii and rotifers enriched, since the day 11 to 46 only with enriched Artemia metanauplii with four commercial products: Protein Selco Plus® (PSP), marine fish food Azoo 9 in 1® (AZ), Scott Emulsion® (SE) and Isochrysis galbana microalgae (IG). The largest growth in terms of weight and height was observed in seahorses fed with enriched Artemia with PSP (160.13 mg and 4.44 cm respectively), which was significantly higher than the other treatments, which were kept in a range of 81.86-97.80 mg in weight and 3.62-3.72 cm in size (F3.12 = 9.63, p = 0.0049 by weight and F3.12 = 5.12, p = 0.0288 in height). There were not significant differences in survival; however the highest percentage was in PSP diet (74.42%) and the lower in SE (51.60%) (F3.12 = 1.08, p = 0.4095).

This study was conducted to determine the optimal salinity for the culture of H. reidi fry during the first month of life, evaluating the effect of different salinities (27, 30, 33, and 36) on its growth and survival. The fry were distributed at three ind.L-1 density in 12 L incubators, animals were fed ad libitum throughout the experiment, survival was monitored daily, and height and weight were measured both at the beginning and end of the experiment. The salinity did not show significant differences between treatments, the highest survival rate was obtained at 27 with 17.59 ± 6.99%.

Aun cuando Argopecten nucleus es un pectínido nativo del Caribe colombiano que presenta gran potencial para cultivo, su producción a gran escala se enfrenta con la escasez de semilla en el medio natural. Por tal motivo, la producción de semilla en el laboratorio es la fuente más prometedora. Dentro de los numerosos estudios que se han realizado al respecto, no se ha logrado un consenso sobre la alimentación y densidad larval durante el cultivo. El presente estudio determinó el efecto conjunto de la densidad inicial de siembra y de la ración alimenticia, utilizando tres densidades (5, 10 y 15 larvas mL-1) y tres concentraciones de microalgas Isochrysis galbana (20000, 40000 y 70000 células mL-1). Se evaluó la supervivencia larval, el crecimiento, la deformidad, el estado de condición de la glándula digestiva (llena, semillena y vacía) y la aparición de la mancha ocular. Se determinó que no hubo interacción entre los factores con respecto a la supervivencia, crecimiento y deformidad, pero sí sobre el estado de condición. No se obtuvieron diferencias significativas en la supervivencia con ninguna de las combinaciones probadas, mientras que la densidad larval ejerció gran influencia sobre el crecimiento y el estado de condición, siendo mayor en los cultivos con 10 y 15 larvas mL-1. En ninguno de los casos hubo influencia significativa por parte de la concentración de alimento. Aun así, los porcentajes obtenidos con la combinación de 10 larvas mL-1 y 20000 células mL-1 de alimento (L10-20C) en la mayoría de las variables medidas presentaron los mayores valores. En este experimento no se obtuvieron larvas con mancha ocular. Argopecten nucleus is a scallop bivalve native from the Colombian Caribbean, with great potential culture, but the critic point for a large-scale production is the scarce seed spat available in the environment. For this reason, its production in the laboratory is the most promising source. There is no consensus about feeding with respect to the larval sowing density after several studies, since generally each factor is worked separately. Therefore, in this research we studied the joint effect of the larval initial density and the feed ration, testing three different densities, 5, 10, and 15 larvae mL-1 and three feed concentrations, 20000, 40000, and 70000 cells mL-1 of Isochrysis galbana, over the surviving, growth, condition state (full, halffull, and empty larvae), deformity or malformation, and development of the eye spot. There was not interaction between factors with regard to surviving, growth, and deformity, but over condition state. There were not significant differences with regard to survival with any of combinations tested, whereas larval density make a greatly influence in the growth and condition state, being higher in the 10 and 15 larvae mL-1 culture for the first variable and in the 10 larvae mL-1 culture for the second one. There was no significant influence by feed concentration in any case. Even so, the percentages obtained with the combination of 10 larvae mL-1 and 20000 cells mL-1 of food (L10-20C) was higher in most cases. There were not eye spot larvae in this experiment.

Como un aporte para determinar la factibilidad biológica y tecnológica del cultivo de Nodipecten nodosus en el Caribe colombiano, entre abril y noviembre de 2007 se evaluó el efecto de dos densidades de cobertura (30 y 50 %) sobre su crecimiento y supervivencia. En jaulas suspendidas en el mar a 5 m de profundidad se colocaron juveniles entre 60 y 65 mm de longitud. Se realizaron mediciones quincenales de la longitud de la concha y de la supervivencia. De igual manera se registró la temperatura, salinidad, seston total y materia orgánica particulada para determinar la relación presentada con estos factores; N. nodosus alcanzó una longitud de 84.7 y 87.9 mm a 30 y 50 % de densidad, respectivamente. Los tratamientos (30 y 50 %) no difirieron en la supervivencia, pero sí en el crecimiento. Se observó una relación directa de la tasa de crecimiento con la materia orgánica y el seston total. La supervivencia mantuvo una tendencia similar hasta julio en las dos densidades, presentando a su vez una relación directa con la materia orgánica particulada. Los resultados observados demuestran que N. nodosus puede soportar altas densidades, lo que permite tener un mayor aprovechamiento del arte de cultivo, aumentando así la productividad del mismo. As a contribution to determine the biological and technological feasibility of the culture of N. nodosus in the Colombian Caribbean, the effect of two cover densities (30 and 50 %) upon growth and survival was evaluated between April and November 2007. In cages suspended in the sea at 5 m depth juveniles between 60 and 65 mm length were placed. Shell lengths and survival were measured each 15 days. In the same manner, temperature, salinity, seston and particulate organic matter were measured in order to determine relationships with these factors; N. nodosus reached a length of 84.7 and 87.9 mm at densities of 30 and 50 %, respectively. Treatments affected growth but did not affect significantly survival. A direct relation between growth and organic matter and between growth and total seston were detected. Survival maintained a similar trend until July at both densities, presenting in turn a direct relation to particulate organic matter. Results demonstrate that N. nodosus presents a good development at high densities, which allow to have a greater utilization of the culture art, hence increasing its productivity.

1 page.-- Workshop for the analysis of the impact of perkinsosis to the European shellfish industry, September 12-14th 2007, Vigo (Spain), The prevalence variation of Perkinsus olseni in carpet-shell clams (Ruditapes decussatus) was determined over a year (July 2000-July 2001) in two important production areas of Galicia (NW Spain) (Ría of Pontevedra and Ría of Arosa). Mortality rates, temperature and salinity profiles were also measured. In the Ría of Pontevedra production area, the prevalence of Perkinsus sp., showed two annual peaks (Winter and Summer time) while in the Ría of Arosa only one prevalence peak was detected in early spring. Significant differences in the prevalence of this pathogen between areas were established. Although temperature and salinity have a marked influence in the disease dynamics of Perkinsus sp., resulting in a seasonal pattern of variation in both prevalence and intensity of infection, no significant correlation could be established among these two environmental parameters and the prevalence of Perkinsus sp., in none of the two production areas. Monthly mortality rates varied between 2 and 12 % and no statistically significant relation could be established between the presence of Perkinsus olseni, the environmental parameters and the mortality rates

In this study, we have determined the histopathology of the infection by the protozoan parasite Perkinsus atlanticus in three exploitable clam species (carpet-shell clam Ruditapes decussatus, Manila clam R. philippinarum, and the species R. pullastra) from Galicia (NW of Spain). In histological preparations from infected animals, typical mature trophozoites of Perkinsus, containing lipid droplets and vacuole but no vacuoplast, were observed. The trophozoites were usually in groups surrounded by a light halo. Pre-palintomic tomonts, as well as palintomic tomonts with a variable number of daughter cells, were also present in the host tissues. Less frequently, immature trophozoites with an undifferentiated cytoplasm were observed. The presence of P. atlanticus cells was always associated with a strong hemocytic infiltration of the surrounding tissues. Occasionally, the parasite cells were internalized by granular hemocytes. The morphology and distribution of the P. atlanticus were similar in the three clam species studied, this being the first time that R. pullastra from Galicia has been found infected by P. atlanticus. The morphology of the parasite's life cycle stages and the histopathology of the infection resemble those reported on other species of the genus Perkinsus.

Los procesos quorum sensing (QS) y quorum quenching (QQ) permiten a las comunidades bacterianas coordinar acciones para adaptarse a los cambios ambientales e incluso formar biopelículas. En esta investigación se evaluaron los mecanismos de comunicación celular usados para la cooperación o competencia y el proceso de formación de biopelículas entre bacterias cultivables aisladas de sedimentos marinos del área de Santa Marta en el Caribe colombiano. Para ello, se realizó la identificación molecular de las cepas mostrando que son especies del género Bacillus, estas fueron capaces de formar biopelículas bajo diferentes condiciones, comprobándose su viabilidad mediante la tinción LIVE/DEAD, permaneciendo vivas después de 24 horas. Los resultados obtenidos del bioensayo para QQ indicaron que dos de las once cepas lograron inhibir la producción de violaceína en Chromobacterium violaceum ATCC 31532. Mientras que en los ensayos para detectar QS no fue posible identificar mediante cromatografía de gases- espectrometría masas CG-MS el autoinductor-2 en los Cultivos de las bacterias. Se infiere, que la interrupción de la comunicación bacteriana que algunas de estas especies realizan podrían estar regulando la expresión de genes de otras bacterias en su entorno como mecanismo de competencia. También, que la capacidad de formar biopelículas de estas bacterias contribuiría a la adaptación al estrés ambiental y favorece el biofouling. Maestría
