Abstract: International audience; The APETALA2 (AP2)/ETHYLENE RESPONSE FACTOR (ERF) superfamily of transcriptional regulators is involved in several growth, development and stress responses processes in higher plants. Currently, the available information on the biological roles of AP2/ERF genes is derived from Arabidopsis thaliana. In the present work, we have investigated genomic and transcriptional aspects of AP2/ERF genes in the economically important perennial species, Malus ×domestica. We have identified 259 sequences containing at least one ERF domain in apple genome. The vast majority of the putative proteins display predicted nuclear localization, compatible with a biological role in transcription regulation. The AP2 and ERF families are greatly expanded in apple. Whole-genome analyses in other plant species have identified a single genomic sequence with divergent ERF, whereas in apple seven soloists are present. In the apple genome, the most noteworthy expansion occurred in sub-groups V, VIII and IX of the ERF family. Expression profiling analyses have revealed the association of ripening-involved ERF genes to scab (Venturia inequalis) pathogenesis in the susceptible Gala cultivar, indicating that gene expansion processes were accompanied by functional divergence. The presented analyses of AP2/ERF genes in apple provide evidences of shared ethylene-mediated signaling pathways in ripening and disease responses.

Abstract: International audience; Cultivated for centuries, the varieties of rose have been selected based on a number of flower traits. Understanding the genetic and molecular basis that contributes to these traits will impact on future improvements for this economically important ornamental plant. In this study, we used scanning electron microscopy and sections of meristems and flowers to establish a precise morphological calendar from early rose flower development stages to senescing flowers. Global gene expression was investigated from floral meristem initiation up to flower senescence in three rose genotypes exhibiting contrasted floral traits including continuous versus once flowering and simple versus double flower architecture, using a newly developed Affymetrix microarray (Rosa1_Affyarray) tool containing sequences representing 4765 unigenes expressed during flower development. Data analyses permitted the identification of genes associated with floral transition, floral organs initiation up to flower senescence. Quantitative real time PCR analyses validated the mRNA accumulation changes observed in microarray hybridizations for a selection of 24 genes expressed at either high or low levels. Our data describe the early flower development stages in Rosa sp, the production of a rose microarray and demonstrate its usefulness and reliability to study gene expression during extensive development phases, from the vegetative meristem to the senescent flower.

Abstract: Hydrangea macrophylla is a horticultural plant of considerable commercial interest that has been widely studied with the aim to more effectively control the different stages of its development during production. However, although floral transformation is a key factor underlying the commercial quality of the product, it remains difficult to control despite these efforts. The floral transformation sequence consists of three successive phases: floral induction (B1), floral evocation (B2), and floral organogenesis (B3). The first is a phase of vegetative organogenesis without elongation leading to the formation of a bud composed of eight phytomer primordia under inductive climatic conditions. This work shows that climatic conditions favorable to floral transformation must be continuously applied without interruption throughout phase B1 to ensure the formation of the floral bud in Hydrangea macrophylla ‘Leuchtfeuer’. In the opposite case, floral transformation is stopped and vegetative growth begins once again.

Abstract: International audience; The possibility of eradicating the pelargonium flower break virus (PFBV) and pelargonium line pattern virus (PLPV) by cryotherapy of axillary shoot apices was investigated using five Pelargonium cultivars. Viruses were detected by DAS-ELISA and their location was determined by immunolocalization. Apex culture did not permit elimination of PFBV and only 15% regenerated plants of 'Stellar Artie' cultivar were ELISA PLPV-negative. Plants regenerated from cryotherapy-treated apices were tested by DAS-ELISA after a 3-month in vitro culture period. Viruses were not detected in 25% and 50% of the plants tested for PFBV and PLPV, respectively. However, immunolocalization carried out on apices originating from cryopreserved shoot tips sampled from DAS-ELISA negative plants showed that they were still virus-infected. Using immunolocalization, PFBV and PLPV could be detected in Pelargoniuni apices, even in the meristematic dome. However, viral particles were more numerous in basal zone cells than in meristematic cells. Our results demonstrate that PFBV and PLPV are present within meristematic cells and that cryopreservation can partly reduce the quantity of these viruses in Pelargonium plants but not eliminate them totally. Additional knowledge on localization and behaviour of viruses during cryopreservation is essential to optimize cryotherapy and plant genetic resource management.

Abstract: International audience; We present here a revised classification of Santalales, an angiosperm order that contains 18 families, 160 genera, and over 2200 species. Both nonparasitic and parasitic flowering plants occur in the traditionally circumscribed family Olacaceae whereas all other families are composed entirely of parasites. The five evolutionary radiations of aerial parasitism produced mistletoes that constitute most of the generic and specific diversity seen in the order. This classification, although based primarily upon results from molecular phylogenetic investigations, brings together all currently available information that contributes to our understanding of relationships among these plants. Monophyletic groups (clades) obtained from molecular analyses were named using a Linnaean ranked system. Four new families are named that formerly resided in Santalaceae s.l.: Amphorogynaceae, Cervantesiaceae, Comandraceae, and Nanodeaceae. A new tribal and subtribal classification for Loranthaceae is presented where nine new subtribe names are proposed.

Abstract: International audience; Internal Transcribed Spacer (ITS) of ribosomal DNA is a classical sequence used for phylogenetic analyses. Usually, concerted evolution homogenizes the numerous ITS of an organism in such a way that a single sequence is amplified. However, for vegetatively propagated plants of hybrid origin, several ITS sequences can be obtained from a single individual. By cloning ITS sequences of various taxa assigned to tribe Genisteae within Fabaceae, we have been able to show such ITS polymorphism in Cytisus x kewensis, C. x dallimorei, C. x praecox, Genista x spachiana and G. 'Porlock' among others. For these taxa, the distinct ITS sequences can be compared with sequences of putative parents found in literature. We have then confirmed that Cytisus x kewensis is a C. ardoinoi x C. multiflorus cross. We have also confirmed that Genista 'Porlock' is a cross between G.monspessulana and G. x spachiana. Genista x spachiana, commonly presented as G. canariensis x G. stenopetala cross may in fact be a more complex hybrid involving also G. monspessulana.

Abstract: Mandevilla sanderi is a plant of tropical origin of great horticultural interest because of its abundant flowering and its persistent foliage. Vegetative propagation requires the removal of leafy branches on the mother plant to produce cuttings. This loss of biomass must be compensated for by the growth of new branches thanks to the mobilization of reserves within the plant. Lack of knowledge about the physiology of this species therefore makes it necessary to characterize its different organs both at the level of their anatomic organization as well as at the level of their ability to store starch. After histological characterization of the different organs (leaves, stems, and roots), starch reserves were localized by histochemical analysis and quantified by biochemical analysis. Starch grains are mainly found in the parenchymatous cortex, the parenchymatous pith and xylem parenchyma cells, in tuberous roots and stems, and in the palisade and spongy mesophyll of leaves. In 22-week-old plants, the greatest quantity of starch is found in the leaves, whereas the tuberous roots have the highest concentration. The histological description of the different organs of Mandevilla sanderi and the localization of starch reserves allow us to assess the potential role of the different organs in plant growth and development. In the particular case of mother plant management, it is hoped that this knowledge will make it possible to optimize conditions for removing leafy branches.

This system aims to standardize area names used in biogeography and constitutes a key step towards maturity of this discipline. We point out a number of issues in this first version of ICAN. These include the lack of distinction between classification and nomenclature, the use of ranks, the criteria of area definition, the conflation of diagnosis and description, the ambiguous use of valid/available, and the system of amendments. We suggest postponing the initial date of application of this code until consistent rules are provided.

Abstract: International audience; The droplet-vitrification method was adapted to Pelargonium apices by optimizing the duration of the loading solution (LS) as well as the plant vitrification solution 2 (PVS2). The excised apices were dehydrated in two steps (20 min in LS and 15 min in PVS2) and then immersed directly in liquid nitrogen (LN). After thawing and unloading in the recovery solution at room temperature for 15 min, apices were plated onto semi-solid Murashige and Skoog medium. This simple protocol without any pretreatment was successfully applied to eight cultivars with a survival level ranging between 55.6 - 96.2 % and a regrowth level between 9.1% - 70.6%. These results prove the feasibility of the long-term storage of Pelargonium germplasm through cryopreservation

Abstract: In 2005, the French Systematics Society did a series of investigations into the teaching systematics in France. From these investigations it rises a decomposition of systematic into five elements, namely identification, classification, phylogeny, evolution, and the taxonomy – nomenclature unit. According to the education levels (primary education, secondary education, university), these various elements are, or not, studied. All in all, whatever is the education level, phylogeny is always approached, generally together with identification and classification. In addition, an analysis of the publication and PhD thesis indicate that two areas are dominant for plant systematics, the Paris region and Languedoc-Roussillon, and the themes approached are mainly centered on the crop plants from temperate countries and the tropical wild plants., En 2005, la Société Française de Systématique a réalisé des enquêtes sur l’enseignement de la systématique en France. De ces enquêtes il découle une décomposition de la systématique en cinq éléments de formation : l’identification, la classification, la phylogénie, l’évolution, et l’ensemble taxinomie – nomenclature. Selon les niveaux de formation (primaire, secondaire, université), ces divers éléments sont, ou non, étudiés. Globalement, quel que soit le niveau de formation, l’aspect phylogénie est toujours abordé, généralement avec ceux de l’identification et de la classification. Abstract: Dès ses premiers travaux (par exemple le Fundamenta Botanica de 1736), Linné a exposé les " règles arbitraires " de nomenclature qu'il suivait, en particulier de règles d'orthographe et de synonymie / homonymie. À la fin du xviiie et au début du xixe siècle, d'autres botanistes critiquent les règles de Linné (par exemple Heister) ou exposent les leurs (par exemple Augustin de Candolle et Lindley), mais globalement durant cette période les règles ont été implicites et relativement peu appliquées. Dans les années 1840- 1870, les premières règles de nomenclature se mettent en place tant en zoologie qu'en botanique, tel que le Code de Strickland en 1843 ou celui de de Candolle de 1867. Dans les années 1880-1890, les règles de nomenclature reviennent sur le devant de la scène avec un nombre élevé de nouveaux Codes tant en botanique qu'en zoologie, et l'expression par certains d'idées assez iconoclastes. Entre 1905 et 1907, les premiers Codes internationaux de nomenclature seront publiés tant pour la botanique que pour la zoologie. En botanique, deux courants (et donc deux Codes) vont exister à partir de 1907 (publication du " Brittonian Code "). Ce n'est qu'en 1930, que le schisme Code Américain / Code International cessera, pour fournir un Code proche de sa version actuelle, alors que pendant plus de 40 ans, aucun nouveau Code de nomenclature zoologique ne verra le jour. Au cours du xxe siècle, d'autres phénomènes vont apparaître dans le débat avec, par exemple, la scission des règles applicables aux plantes cultivées dans les années 1950 (mais dont les prémisses remontaient à 1847), puis celle des bactériologistes et des virologistes dans les années 1970-80.Enfin, au cours des années 1990-2000, plusieurs initiatives vont voir le jour : Reformed Code - 1990 ; New Biological Nomenclature - 1991 ; Biocode - 1997 et Phylocode -- 2000, dont certaines subsistent encore.

The fungal development between 1 and 15 days post-inoculation was quite similar in the susceptible cv. Presto and the partially resistant cv. Texto: After conidial germination, leaf adhesion of the pathogen was achieved with mucilaginous filaments; hyphae penetrated the leaves directly with/without the formation of appressoria-like structures or via stomata; the fungus spread by epiphytic hyphae with hyphopodia and subcuticular mycelia. Intense necrotic plant cell reactions occurred under the fungal structures. At 21 days post-inoculation, typical features of fungal development were noted for each cultivar: growing hyphae emerged from stomata in cv. Presto, whereas conidiophores without conidia were observed in cv. Texto. Leaf tissues of both cultivars were strongly damaged and vesicle-like structures (assumed to be plant phenolics) were abundantly present between mesophyll cells. A real-time PCR method was developed for in planta quantification of A. dauci. Between 1 and 15 days post-inoculation, the fungal biomass was equivalent in the two cultivars and was about fourfold higher in cv. Presto than cv. Texto at 21 and 25 days post-inoculation. Taken together, our results indicated that A. dauci was able to colonize both cultivars in a similar manner during the first steps of the interaction, then fungal development in the partially resistant cultivar was restricted due to putative plant defence reactions. The results of this study enhance the overall understanding of infection processes in the A. dauci-carrot pathosystem

Abstract: International audience; Erwinia amylovora is the causal agent of fire blight of Maloideae. One of the main pathogenicity factors of this bacterium is the exopolysaccharide (EPS) of its capsule. In this paper, we used genetic transformation tools to constitutively express an EPS-depolymerase transgene in the pear (Pyrus communis L.) cv. Passe Crassane with the aim of decreasing its high susceptibility to fire blight. Expression of the depolymerase gene in 15 independent transgenic clones led, on average, to low depolymerase activity, although relatively high expression was observed at the transcriptional and translational levels. Only two of the transgenic clones (9X and 10M) consistently showed a decrease in fire blight susceptibility in vitro and in the greenhouse. These clones were also among the highest expressers of depolymerase at the RNA and enzyme activity levels. The correlation observed among all transgenic clones between depolymerase expression and fire blight resistance suggested the potential of this strategy.

Abstract: International audience; The siderophore produced by Erwinia amylovora, the causal agent of fire blight of Maloideae, is one of the virulence factors of this bacterium. The production of siderophores enables E. amylovora to overcome the conditions of iron limitation met in plant tissue, and may also protect the bacteria against active oxygen species produced through the Fenton reaction. In this paper, we have examined the ability of an iron chelator protein, encoded by the bovine lactoferrin gene, to reduce fire blight susceptibility in pear (Pyrus communis L.). Transgenic pear clones expressing this gene controlled by the CaMV35S promoter were produced by Agrobacterium tumefaciens mediated transformation. Transformants were analysed by RT-PCR and western blot to determine lactoferrin expression levels. Most transgenic clones demonstrated significant reduction of susceptibility to fire blight in vitro and in the greenhouse when inoculated by E. amylovora. These transgenic clones also showed a significant reduction of symptoms when inoculated with two other pear bacterial pathogens : Pseudomonas syringae pv. syringae and Agrobacterium tumefaciens. Moreover, we have shown that this increase in bacterial resistance was correlated with an increase in root ferric reductase level activity and leaf iron content. Despite negative effects on the growth of a few clones, our results indicate the potential of lactoferrin gene transformation to protect pear from fire blight through increased iron chelation.

Abstract: The large French research project GENIUS (2012–2019, https://www6.inra.genius-project_eng/ ) provides a good showcase of current genome editing techniques applied to crop plants. It addresses a large variety of agricultural species (rice, wheat, maize, tomato, potato, oilseed rape, poplar, apple and rose) together with some models (Arabidopsis, Brachypodium, Physcomitrella). Using targeted mutagenesis as its work horse, the project is limited to proof of concept under confined conditions. It mainly covers traits linked to crop culture, such as disease resistance to viruses and fungi, flowering time, plant architecture, tolerance to salinity and plant reproduction but also addresses traits improving the quality of agricultural products for industrial purposes. Examples include virus resistant tomato, early flowering apple and low-amylose starch potato. The wide range of traits illustrates the potential of genome editing towards a more sustainable agriculture through the reduction of pesticides and to the emergence of innovative bio-economy sectors based on custom tailored quality traits.

Abstract: International audience; Automated identification of plants and animals have improved considerably in the last few years, in particular thanks to the recent advances in deep learning. The next big question is how far such automated systems are from the human expertise. Indeed, even the best experts are sometimes confused and/or disagree between each others when validating visual or audio observations of living organism. A picture or a sound actually contains only a partial information that is usually not sufficient to determine the right species with certainty. Quantifying this uncertainty and comparing it to the performance of automated systems is of high interest for both computer scientists and expert naturalists. This chapter reports an experimental study following this idea in the plant domain. In total, nine deep-learning systems implemented by three different research teams were evaluated with regard to nine expert botanists of the French flora. Therefore, we created a small set of plant observations that were identified in the field and revised by experts in order to have a near-perfect golden standard. The main outcome of this work is that the performance of state-of-the-art deep learning models is now close to the most advanced human expertise. This shows that automated plant identification systems are now mature enough for several routine tasks, and can offer very promising tools for autonomous ecological surveillance systems.

Abstract: The rapid and effective activation of disease resistance responses is essential for plant defense against pathogen attack. These responses are initiated when pathogen-derived molecules (elicitors) are recognized by the host. In order to create novel mechanisms for fire blight resistance in pear, we have generated transgenic pears expressing the elicitor harpin Nea from Erwinia amylovora under the control of the constitutive promoter CaMV35S. The transient expression of hrpN Ea in pear cells did not provoke any apparent damage. Therefore, stable constitutive expression of hrpN Ea was studied in seventeen transgenic clones of the very susceptible cultivar "Passe Crassane.” Most transgenic clones displayed significant reduction of susceptibility to fire blight in vitro when inoculated by E. amylovora, which was positively correlated to their degree of expression of the transgene hrpN Ea . These results indicate that ectopic expression of a bacterial elicitor such as harpin Nea is a promising way to improve pear resistance to fire blight

Abstract: International audience; High-altitude rockjasmines (genus Androsace) are a paramount example of evolutionary radiations in temperate mountains of the Northern Hemisphere. Yet, we show here that their taxonomy is incomplete and has been subject to many historical mistakes, probably due to the lack of exploration of mountains by the classical botanists who described these species. Here we wish to clarify the application of names with regard to four defined morphotypes, typical of high-elevation zones of the Western Alps, in order to set a definitive basis for morphological delimitation of a likely new species discovered in the Mont Blanc range (species not described in this work). To do so, we review the historical taxonomic treatments and positions, carefully reconsider types and species names for these morphotypes, and designate lectotypes and epitypes for each of them. In particular, we confirm the validity of names commonly used to refer to the taxa Androsace alpina, Androsace helvetica, and Androsace pubescens. We show that Androsace vandellii is an invalid name and that Androsace argentea should be used instead. Our work illustrates the utility of historical herbaria to clarify the taxonomy of complex groups of plants growing in inaccessible environments such as high-altitude regions.

Période 2000-2012. http://prodinra.inra.fr/record/256699; International audience; 'Antonovka' has long been recognised as a major source of scab (Venturia inaequalis) resistance useful for apple breeding worldwide. Both major gene resistances in the form of the Rvi10 and Rvi17 and quantitative resistance, collectively identified as VA, have been identified in different accessions of 'Antonovka'. Most of the 'Antonovka' scab resistance used in apple-breeding programmes around the world can be traced back to Schmidt 'Antonovka' and predominantly its B VIII progenies 33,25 (PI 172623), 34,6 (PI 172633), 33,8 (PI 172612) and 34,5 (PI 172632). Using genetic profile reconstruction, we have identified "common 'Antonovka' " as the progenitor of the B VIII family, which is consistent with it having been a commercial cultivar in Poland and the single source of scab resistance used by Dr. Martin Schmidt. The major 'Antonovka' scab resistance genes mapped to date are located either very close to Rvi6, or about 20-25 cM above it, but their identities need further elucidation. The presence of the 139 bp allele of the CH-Vf1 microsatellite marker known to be associated with Rvi17 (Va1) in most of the 'Antonovka' germplasm used in breeding suggests that it plays a central role in the resistance. The nature and the genetic relationships of the scab resistance in these accessions as well as a number of apple cultivars derived from 'Antonovka', such as, 'Freedom', 'Burgundy' and 'Angold', are discussed. The parentage of 'Reglindis' is unclear, but the cultivar commercialised as 'Reglindis' was confirmed to be an Rvi6 cultivar.

Blaumeise by Agrobacterium tumefaciens following the development of an efficient regeneration system using leaf discs excised from 12 to 15 weeks old meristem-derived vitroplants. Explants were cultivated on solid B5 medium complemented with maltose 110 mM, BAP 10 μM and NAA 0.5 μM. A low light regime of 17 μmol m −2 s −1 improved regeneration frequency up to 86%. For transformation, leaf discs were inoculated and co-cultivated with two disarmed A. tumefaciens strains, EHA 101 and LBA 4404, both carrying the binary vector pFAJ3000 which contained the nptII selectable gene and the GUS reporter gene. A pre-culture period of 3 days and a short co-cultivation duration (1 day) improved the efficiency of transformation. Inoculation of only 10 min with agitation including (or not) vacuum infiltration was sufficient. If selection on kanamycin containing medium was applied after a 2 weeks culture period on shoot regeneration medium, the percentage of explants forming kanamycin-resistant shoots increased from 3.3 to 13.3%. Integration and expression of the introduced transgene were confirmed by histochemical GUS assay, PCR and Southern blot analysis. Flowering of transgenic plants in glasshouse occurred 10 months after acclimatization.

Abstract: According to the history of the cultivated carrot, root colour can be considered as a structural factor of carrot germplasm. Therefore, molecular variations of carotenoid biosynthesis genes, these being involved in colour traits, represent a good putative source of polymorphism related to diversity structure. Seven candidate genes involved in the carotenoid biosynthesis pathway have been analysed from a sample of 48 individual plants, each one from a different cultivar of carrot (Daucus carota L. ssp. sativus). The cultivars were chosen to represent a large diversity and a wide range of root colour. A high single nucleotide polymorphism (SNP) frequency of 1 SNP per 22 bp (mean pi (sil) = 0.020) was found on average within these genes. The analysis of genetic structure from carotenoid biosynthesis gene sequences and 17 putatively neutral microsatellites showed moderate genetic differentiation between cultivars originating from the West and the East (F (ST) = 0.072), this being consistent with breeding history, but not previously evidenced by molecular tools. Surprisingly, carotenoid biosynthesis genes did not exhibit decay of LD (mean r (2) = 0.635) within the 700-1,000 bp analysed, even though a fast decay level of LD is expected in outcrossing species. The high level of intralocus LD found for carotenoid biosynthesis genes implies that candidate-gene association mapping for carrot root colour should be useful to validate gene function, but may be unable to identify precisely the causative variations involved in trait determinism. Finally this study affords the first molecular evidence of a genetic structure in cultivated carrot germplasm related to phylogeography.

Abstract: Alternaria leaf blight (ALB), caused by Alternaria dauci, is one of the most damaging foliar diseases of carrot worldwide. The aim of this study was to compare different methods for evaluating levels of carrot resistance to ALB. Three techniques were investigated by comparison with a visual disease assessment control: in vivo conidial germination, a bioassay based on a drop-inoculation method, and in planta quantification of fungal biomass by quantitative PCR (Q-PCR). Three carrot cultivars showing different degrees of resistance to A. dauci were used, i.e. a susceptible cultivar ( Presto) and two partially resistant genotypes (Texto and Bolero), challenged with an aggressive or a very aggressive isolate of A. dauci. Both partially resistant genotypes produced a higher mean number of germ tubes per conidium (up to 3.42 +/- 0.35) than the susceptible one (1.26 +/- 0.18). The drop-inoculation results allowed one of the partially resistant genotypes (Bolero, log(10)(S+1) = 1.34 +/- 0.13) to be distinguished from the susceptible one (1.90 +/- 0.13). By contrast, fungal growth measured by Q-PCR clearly differentiated the two partially resistant genotypes with log(10)(I) values of 2.77 +/- 0.13 compared to the susceptible cultivar (3.65 +/- 0.13) at 15 days post-inoculation. This result was strongly correlated (r(2) = 0.91) with the disease severity index scored at the same date. Data obtained with the different assessment methods strongly suggest that the Texto and Bolero genotypes have different genetic resistance sources.

Different regulator-free nutrient media were compared to determine the effects of the mineral solution, agar and sucrose concentrations on seedling development from mature embryos. HortScience 19, 507-509] mineral solutions were more efficient for the development of aerial parts than nutrient-poor KN [Knop, W., 1884. Bereitung einer concentrierten nahrstofflosung fur pflanzen. Landwersuhssat 30, 292-294] and WT [Withe, P.R., 1936. Plant tissue cultures. Bot. Rev. 2, 419-437] solutions. Reducing the agar concentration enhanced fresh matter production and balanced seedling development. When tested at different concentrations, sucrose was found to orient mature embryo development, with the best results obtained at concentrations of 2-4%, whereas high concentrations (6 and 12%) mainly inhibited elongation of the aerial parts. Plantlets obtained previously from in vitro cultured embryos were propagated by axillary budding. High bud proliferation (six shoots per explant) was achieved when using 17.8 mu M benzyladenine (BA) combined with 0.5 mu M indole-3-butyric acid (IBA). The addition of 2.9 mu M gibberellic acid (GA(3)) to the propagation medium did not improve axillary shoot yields and on average, media with GA(3) produced 2.3-2.6 elongated stems per cultured explant. Abstract: International audience; We formulate a spatially realistic population-genetic model for ascertaining the synergetic effect between genetic and spatial composition of the host population on the pathogen spread reinforced by evolutionary processes. We show that spatial arrangement of host genotypes is crucial to the efficacy of host genetic diversification. In particular, the reductive effect of multigenic resistance on the pathogen density can be produced by a random patterning of monogenic resistances. Random patterns can reduce both density and genetic diversity of the pathogen population and delay invasion promoted by sexual recombination. By contrast, patchy distributions diversify pathogen population and, hence, reduce the efficacy of resistance genes. The proposed approach provides theoretical support for studying fast emergence and spread of novel pathogen genotypes carrying multiple virulence genes. It has a practical applicability to design innovative strategies for the most appropriate deployment of plant resistance genes.

Abstract: International audience; Fire blight caused by the bacteria Erwinia amylovora is one of the most important diseases of pear. This causal agent produces a siderophore (desferrioxamine), which was identified as one of its virulence factors. This protein enables the bacteria to overcome conditions of iron limitation encountered in host tissues, and may also protect the bacteria against active oxygen species. Previous experiments indicate that the use of an iron chelator protein, encoded by the bovine lactoferrin gene, reduces fire blight symptoms in some transgenic pear clones. The aim of the present work is to test the ability of a more efficient iron chelator, plant ferritin, to reduce fire blight susceptibility in pear. In the literature, ferritin genes have been overexpressed under the control of a constitutive promoter in different transgenic plant species for various purposes. In several cases, the constitutive expression of ferritin produced negative effects such as reduced growth and chlorophyll content. Therefore, we decided to place the exogenous ferritin gene from pea under the control of a pathogen inducible promoter (sgd24) in comparison with a constitutive promoter (CaMV 35S). Two pear varieties, 'Conference' (CF) and 'Passe-Crassane' (PC) were transformed using both constructs. Transformation rates depended on variety and construct. They were respectively of 12 and 4.3% for CF and PC using the sgd24-ferritin construct. Only PC was transformed with the 35S-ferritin construct, with a transformation rate of 2%. First analyses of the transgenic clones by RT-PCR showed the expression of pea ferritin in both constructs and in all clones. The transgenic clones were acclimatized in greenhouse and exhibited normal growth. Quantification of ferritin gene expression, ferritin protein accumulation, and evaluation of fire blight resistance are underway.

Both growth and branching processes were phenotyped on the consecutive annual shoots of different axes within a tree. For each studied trait, predicted values (best linear unbiased predictors, BLUPs) of the genotypic (G) effect or its interaction with tree age (G×A) and climatic year (G×Y) were extracted from mixed linear models of repeated data. These BLUPs, which are independent from autocorrelations between repeated measurements, were used for QTL mapping. QTL detection power was improved by this two-step approach. For each architectural process, numerous QTLs were detected and some particularly interesting co-localised in common genomic regions, for internode lengthening, top diameter, and number and percentage of axillary shoots. When several QTLs were detected for a given trait, global models were estimated, which explained a maximum of 40% of the total variance for both internode length and top diameter and 28% for branching. QTLs detected for BLUPs of G×Y effects were interpreted as resulting from the interaction between genetic maximal potential of growth and climatic factors, while those for G×A effects were interpreted in relation to tree ontogeny. Most of the latter ones were found to be concomitant with key development stages during which the trait average started to decrease, but with different magnitudes depending on genotype

Période 2000-2012. http://prodinra.inra.fr/record/256699; International audience; An integrated consensus genetic map for apple was constructed on the basis of segregation data from four genetically connected crosses (C1 = Discovery x TN10-8, C2 = Fiesta x Discovery, C3 = Discovery x Prima, C4 = Durello di Forli x Fiesta) with a total of 676 individuals using CarthaGene(R) software. First, integrated female-male maps were built for each population using common female-male simple sequence repeat markers (SSRs). Then, common SSRs over populations were used for the consensus map integration. The integrated consensus map consists of 1,046 markers, of which 159 are SSR markers, distributed over 17 linkage groups reflecting the basic chromosome number of apple. The total length of the integrated consensus map was 1,032 cM with a mean distance between adjacent loci of 1.1 cM. Markers were proportionally distributed over the 17 linkage groups (chi(2) = 16.53, df = 16, p = 0.41). A non-uniform marker distribution was observed within all of the linkage groups (LGs). Clustering of markers at the same position (within a 1-cM window) was observed throughout LGs and consisted predominantly of only two to three linked markers. The four integrated female-male maps showed a very good colinearity in marker order for their common markers, except for only two (CH01h01, CH05g03) and three (CH05a02z, NZ02b01, Lap-1) markers on LG17 and LG15, respectively. Abstract: International audience; Eutypine is a toxin produced by Eutypa lata, the causal agent of eutypa dieback of grapevines. An eutypine detoxifying gene (Vr-ERE) encoding an NADPH-dependent aldehyde reductase converts eutypine into the corresponding alcohol, eutypinol, a non-toxic form of the toxin. A variety of phytotoxic compounds containing an aldehyde group can act as substrates for this enzyme, therefore opening the possibility to use Vr-ERE as an alternative selection system for plant transformation. Our preliminary experiments with apple and pear have demonstrated the following: 1) among the various substrates of VR-ERE, benzaldehyde (BD), a naturally occurring compound, is able to inhibit adventitious bud regeneration from apple as well as pear in vitro leaves, at concentrations ranging from 1 to 1.5 mM; 2) constitutive expression of Vr-ERE in transgenic ‘Greensleeves’ apple does not provoke any abnormal phenotype after one year of growth in greenhouse, but aldehyde reductase activity is reduced in all transgenic clones; 3) production of transgenic pear using BD instead of kanamycin as selection pressure is feasible, but the efficiency of the selection pressure seems low. In conclusion, the efficiency of Vr-ERE as a selective gene to produce apple and pear transgenic plants is not yet proven and further research is needed
