A vidékfejlesztési szakpolitika egyre inkább felértékelődik. Úgy gondoljuk, a vidékfejlesztést nem csak agrár-megközelítésből kell szemlélni. Az EU komolyan veszi a vidéken élők problémáit, és az életszínvonal növelésére törekszik. A KAP reformok segítenek a vidéki fejlesztéspolitika térnyerésében és a vidék fejlődésének gyors és pozitív változásában. Napjainkban globális versenyről beszélhetünk. Ennek jellemzői a következők: - a kereskedelem nemzetközivé válása, - a termelés és a tőkeáramlás világméretű növekedése, - az innovációs folyamatok globalizációja. Lengyel (2003) szerint ezek a fő jellemzők, mely ismeretekkel helytállhatunk a globális versenyben. Úgy gondoljuk, csak a tudásalapú társadalom fejlődési pályáján lehetünk sikeresek. Magyarország fejlődése a természeti erőforrások használatára épül. A falusi termelés mellett az energetikai célú termelés, valamint az integrált és az ökológiai gazdálkodás is elképzelhető. Indokolt a zöldség-gyümölcs termelés és az állattenyésztés fejlesztése. Nagyon fontos a foglalkoztatás növelése és a népességmegtartás. A vidékfejlesztés szerepe térben és időben változó. A változások áttekintése fontos, hogy az ismeretek birtokában a vidéki területek fejlődése nagyobb lendülettel folytatódjon. A versenyképességre ható tényezőket sok szerző elemezte. Említhető Csath (2019), Szűcs – Marselek (2019), Chikán A. (2017), Holló – Marselek (2016) munkássága. A falvak fejlesztése során fontos szempontokat kell figyelembe venni. Rövid áttekintésünkben csak néhány, a kutatással összefüggő területre utalunk. Ilyenek pl.: a földhasználat kérdése, a mező- és vidékgazdaság helyzete, az agrár-versenyképesség, a környezetvédelem, az öntözés és vízvédelem, a zöld gazdaság és a körforgásos gazdaság.

A mezőgazdaság forradalmi vátozáson megy át. Az ipar robbanásszerű technológiai fejlődése oly mértékű, hogy az a 4. ipari forradalomnak tekinthető. Ez a folyamat begyűrűzött az agrárium területére és így már mezőgazdaság 4.0-ról is beszélünk, amelynek az agrárium teljes területén az információs technológiára alapuló innováció jelent a motorját. A mezőgazdaság esetében számos ok miatt elkerülhetetlen, a termelési hatékonyság növelése, amely a K+F+I azaz a kutatás, fejlesztés és innováció eszközeivel érhető el. Az exponenciálisan fejlődő technológia számos pozitív eredménnyel jár, hisz nő a termelés hatékonysága és volumene, emelkedik a minőség, magasabb szintű automatizációt és valós idejű optimalizációt tesz lehetővé. Mindezek ellenére veszélyeket is hordoz magában. A veszélyek közül jelen írásban a hazai gazdálkodókat markánsan terhelő problémák és az abból való kiút elvi alapjai kerülnek megvilágításra. A kevésbé fejlett országok –ideértve a Magyarországot is – agrárgazdálkodói a mezőgazdaság 4.0 által produkált technológiai innovációkat nem képesek megfelelő tempóban a saját gazdaságukba integrálni és hatékonyan felhasználni, mivel még a mezőgazdaság 3.0 vívmányait sem tudta magáévá tenni a gazdálkodók többsége, így a modern technológiához való tényleges hozzáférésük is egyre kétségesebb. Hazánkban az agrár szektor kis és középvállalkozásai különösen kitettek annak, hogy szubjektív és objektív okokból egyaránt megfosztottá válnak a modern technológiáktól. Ez a helyzet nem, vagy rendkívül nehezen oldható fel. Fogalomként az agrár depriváció szóösszetétellel fogalmazható meg legpontosabban a helyzet, mivel a megfosztottság tartós, a kitörés e helyzetből igen nehéz, a szakadék pedig a fejlett gazdaságokhoz képest akkor is nő, ha a deprivált vállalkozás technológiai színvonala stagnál, de sajnos jellemzőbb, hogy az eszközök avulásával inkább romlik, így az megfosztottság – azaz a technológiai fejlődési lemaradás és a gazdaság eszközeinek avulása – is erősödik.

Cities play a key role in EU regional policy, and to this end the Leipzig Charter called for the development of Integrated Urban Development Strategies (IVS) emphasizing an integrated approach in the 2007–2013 budget period, which was replaced in the period 2014–2020 by Integrated Settlement Development Strategies (ITS). We can find many experiments in the methodological elaboration of the measurement of regional and municipality development in the Hungarian and international literature. However, due to the complexity of the topic, no more widely accepted procedure has emerged, however, there is a common consensus that development is treated as a complex phenomenon. In the present study, we attempted to examine the development of the city of Gyöngyös through a complex system of indicators for the period 2010–2018. In addition, an important part of our research is to supplement the results obtained with statistical data and, if possible, to compare them to the subjective judgment of the residents. Our main goal was to answer how the changes of the indicator groups in the indicator system and their totality are perceived by the residents.

Monor a magyar bortermelés nagy hagyományokkal bíró területe, amely kis ültetvény méretével, de európai léptékben is jelentős, több mint 900 pincés pincefalujával különleges adottságokkal bír. A kutatás rávilágít, hogy a Monori járáson kívül nem ismert a monori pincefalu, méretéről, létezéséről sem tud a válaszadók többsége. A megkérdezettek, 46,7%-a tudott helyes választ megjelölni arra, hogy mi az OEM megjelölés, és a minta mindössze 36,4% tudta, hogy van Monori OEM. Összesen 280 fő válaszolt arra a kérdésre, hogy ha részt vett monori borrendezvényen, akkor mennyire volt elégedett. A válaszok súlyozott átlaga 7,51 volt 10-es Likert-skálán, tehát az elégedettség magas szintű. A monori bornak rendszeres fogyasztója kizárólag a Monori járásban van. Ez kiemelten jelzi, hogy a rövid ellátási láncú értékesítés nélkül a helyi termelés nem létezhetne. Emellett Budapest és környéke lehet expanziós célpont. A válaszadók 85%-a a monori borral szemben komolyabb fenntartással nincs, azt többnyire jónak értékeli. A megkérdezettek 33,1%-a 500 és 1000 Ft közötti árat fizetne egy palack borért, közel 40%-a 1000-1500 Ft közötti összeget sem sajnálna erre a célra. Ezalapján a megkérdezettek többsége, a monori palackozott borért többet fizetne, mint a korrigált országos átlagár. Egy liternyi palackozott bor és a folyóbor iránti fizetésihajlandóságot összevetve 2-2,6-szeres árat lehet elérni, amely belátható, hogy extraprofitot eredményez a palackozott termék értékesítése esetén. A válaszadók több mint 85%-a venne Monori bort, a palackozott és a folyó borra leadott preferencia szavazatok számosságát vizsgálva, a palackozott bort 1,5-szer többen vennék meg, mint a folyóbort. Szín alapján a fehér borok támogatottsága minden korosztályban a legmagasabb. A Z generációnál idősebb korosztályokban, ezt követi a vörös, majd a rozé.

Macrophages and dendritic cells (DCs) are important contributors to anti-tumor immune responses. However, these highly plastic cells are also the primary targets of tumor manipulation, which may result in the development of tumor-promoting subtypes. The effect of chemotherapeutic agents on tumor cells is an area of intense study, but little is known about their effects on innate immune cells.We investigated the effects of four chemotherapeutic drugs (two platinum-based agents; oxaliplatin and cisplatin, and two anthracyclines; doxorubicin and epirubicin) on the differentiation, function, and viability of macrophages and DCs. Macrophages and DCs were differentiated from monocytes in the presence of these chemotherapeutic drugs and we compared their cell surface receptor expression, cytokine production, and chemotactic- and T-cell-polarizing ability.We have shown that differentiation in the presence of anthracyclines dose-dependently increases CTLA-4 expression in DCs. Antineoplastic agent-driven differentiation strongly modified the CCL2- or CCL5-induced chemotactic activity of both macrophages and DCs. DCs differentiated in the presence of high-dose cisplatin and a low dose of epirubicin promoted regulatory T-cell development, whereas oxaliplatin at specific doses induced both DCs and macrophages to enhance cytotoxic T-cell responses. Furthermore, we found that inflammatory macrophages are more sensitive to doxorubicin-induced cell death than their counterparts.In summary, our results confirm that chemotherapeutic agents acting on a similar basis may have different effects on the anti-tumor immune response. Treatment with optimal dose, combinations, and timing of chemotherapy may determine tumor immunity and the metastatic potential of tumors.

A megújuló energiaforrások hasznosításának elterjesztését elsősorban energiapolitikai és klímavédelmi célkitűzések motiválják, az utóbbi években azonban egyre nagyobb hangsúlyt kapott az új beruházások vidékfejlesztési, közösségfejlesztési jelentősége is. A különböző megújuló energia ágazatok a regionálistámogatáspolitika egyik fontos célterületévé váltak, amivel párhuzamosan a potenciális befektetők érdeklődését is felkeltették. A vidéki térségek alacsony népsűrűségükkel és a természeti erőforrásokban való viszonylagos gazdagságukkal sok esetben igen kedvező feltételeket nyújtanak ahhoz, hogy ezeknek a beruházásoknak a helyszínéül szolgáljanak. A helyi gazdaságfejlesztés szempontjai alapján pozitívnak nevezhetjük, hogy ez által a térséghez kötött, belső erőforrások hasznosítására kerül sor. A gyakorlatban azonban fontos kérdéseket vet fel a beruházásokkal kapcsolatban, hogy azok milyen mértékben támaszkodhatnak helyben elérhető pénzügyi forrásokra, szakértelemre, megfelelő számú fogyasztóra. Igazán eredményes vidékfejlesztési projekteket a helyi közösség érdekeit figyelembe véve és a helyi közösség részvételével tudunk elképzelni. A kutatás arra fókuszál magyarországi és európai példák alapján, hogy a megújuló energetikai fejlesztések megvalósítása során felmerülő problémák hogyan állathatók a közösségfejlesztés szolgálatába, milyen lépések szükségesek egy hosszú távon fenntartható projekt előkészítéséhez.

A tanulmány a felnőttoktatás hatását vizsgálja a Miskolci kistérségben élő nők munkaerő-piaci helyzetére vonatkozóan. Kutatásunk elején több hipotézist is felállítottunk, aminek bizonyítására irodalomelemzést, statisztikai adatok analizálását és kérdőíves vizsgálatot használtunk. A vizsgálatot a Miskolci kistérségben élő 18 éves kor feletti gazdaságilag aktív nők körében végeztük, összesen 200 fő került megkérdezésre. A rendelkezésre álló adatok feldolgozásához az IBM SPSS Statistics 20 és Microsoft Excel 2007 programot használtuk fel. Kutatásunk célja az volt, hogy igazoljuk, a Miskolci kistérségben élők nők munkaerő-piaci helyzetére nincs kellő ráhatással a felnőttképzés. A kérdőíves vizsgálat eredményei azt igazolták, hogy bár érdeklődés van afelnőttképzések iránt, a kereslet és kínálat nincs összhangban. Az állam vagy nem támogatja a képzéseket vagy nincs olyan képzés, amivel szakmájában tovább tudna fejlődni és ennek hatására több forráshoz tudna jutni életminőségének javítására az abban résztvevő. A tanulmány ezért ebben az összetett témában javaslatokat kíván tenni a térség felzárkóztatására, a munkanélküliség csökkentésére, a nők hátrányosságának enyhítésére és a felnőttoktatás hatásfokának javítására vonatkozóan.

Az emberi erőforrások fokozódó jelentősége miatt az egész életen át tartó tanuláskoncepciójának megvalósítása nélkül nem beszélhetünk versenyképes tudásalapú társadalomról. A felnőttképzésnek a időről-időre megújuló, versenyképes munkaerő biztosításán túl fontos szerepe van a munkaerőpiacról kiszorult potenciális munkavállalók integrálásában. A felnőttképzésbe beiratkozottak száma alapján Borsod-Abaúj-Zemplén megye Magyarországon a második helyet foglalja el Budapestet követően. A megye hátrányos helyzetű járásaiban, településein átlag feletti azoknak a felzárkóztató képzéseknek a részaránya, amelyek az alacsonyan képzett, munkaerőpiacról kiszorult társadalmi csoportokat szólítanak meg. Kérdőíves vizsgálatunkat az ő körükben végeztük el, feltárva általános szociális helyzetüket, valamint az oktatással és munkavállalással kapcsolatos attitűdjüket.

Developing dendritic cells (DCs) from monocytes is a sensitively regulated process. One possible way for cancers to avoid immune recognition and antitumor response is the modulation of DC differentiation. Although several studies are available on the examination of tumor-associated macrophages, a comprehensive analysis focusing on the effects of tumor-formed DCs is not known to date. We provide a comparative analysis of the tumor-edited-monocyte derived DCs differentiated in the presence of adenocarcinomas (MDA, HT29, HeLa)- and primary (WM278, WM983A) or metastatic (WM1617, WM983B) melanomas. The immunomodulatory effect of tumors is mediated at least partly by secreted mediators. We investigated the impact of tumor cell-derived conditioned media on the differentiation of DCs from CD14+ monocytes, sequentially determining the phenotype, cytokine production, phagocytic, and the T cell polarizing capacity of moDCs. We completed our observations by analyzing our data with bioinformatic tools to provide objective correlations between phenotypical and functional properties of different tumor-educated moDCs. The correlation analysis revealed significant differences in the characteristics of adenocarcinomas- or melanomas-edited moDCs. We highlight the functional differences in the properties of moDCs differentiated in the presence of various cancer cell lines. We offer new information and options for the in vitro differentiation protocols of various tumor-conditioned moDCs. Our results confirm that various immunomodulatory properties of different tumor cell lines result in multiple manipulations of DC differentiation.

During tissue damage caused by infection or sterile inflammation, not only damage-associated molecular patterns (DAMPs), but also resolution-associated molecular patterns (RAMPs) can be activated. These dying cell-associated factors stimulate immune cells localized in the tissue environment and induce the production of inflammatory mediators or specialized proresolving mediators (SPMs). Within the current prospect of science, apoptotic cell death is considered the main initiator of resolution. However, more RAMPs are likely to be released during necrotic cell death than during apoptosis, similar to what has been observed for DAMPs. The inflammatory potential of many regulated forms of necrotic cell death modalities, such as pyroptosis, necroptosis, ferroptosis, netosis, and parthanatos, have been widely studied in necroinflammation, but their possible role in resolution is less considered. In this review, we aim to summarize the relationship between necrotic cell death and resolution, as well as present the current available data regarding the involvement of certain forms of regulated necrotic cell death in necroresolution.

The survival of special Hungarian settlement forms (homesteads), due to the increasing urbanization phenomena, raises more and more questions. Are these forms of settlements of special, natural, landscape and cultural value? Or are the empty spaces that have lost their original function, which are relocating to the growing urban agglomerations? In our research, we sought to find out what kind of vision the Hungarian ‘farm’ could have on the 21st century Hungarian Great Plain. On the one hand, our empirical research focused on a homestead survey in the Bács-Kiskun County, where we surveyed the general condition of the farms, and on the other hand we carried out an opinion survey of the population. We have tried to compare the opinion of the urban population and the vision of the people living on the farms, to identify new functions that will fit in with the European rural development policy, financial and professional support in the future. Our results show that diversified developments can help the survival and development of homesteads.

The purpose of our study is to survey the marketing issues of small-scale food production and short supply chains (SSCs) from the consumers’ aspects, and to give insight into the effectiveness and potential of SSC marketing. The paper presents the result of a wide online consumer survey with the participation of more than 1000 consumers, mostly from the Northern Hungary region. According to our experiences, the most effective „marketing instrument” was the personal contact with producers, communication with relatives, and acquaintances. One-third part of our sample could be motivated by (this kind of) marketing and had more willingness to pay for small-producers’ goods.

Necroptosis is a regulated necrotic-like cell death modality which has come into the focus of attention since it is known to contribute to the pathogenesis of many inflammatory and degenerative diseases as well as to tumor regulation. Based on current data, necroptosis serves as a backup mechanism when death receptor-induced apoptosis is inhibited or absent. However, the necroptotic role of the proteins involved in mitochondrial apoptosis has not been investigated. Here, we demonstrated that the stimulation of several death and pattern recognition receptors induced necroptosis under caspase-compromised conditions in wild-type, but not in caspase-9-negative human Jurkat and murine MEF cells. Cerulein-induced pancreatitis was significantly reduced in mice with acinar cell-restricted caspase-9 gene knockout. The absence of caspase-9 led to impaired association of receptor-interacting serine/threonine-protein kinase 1 (RIPK1) and RIPK3 and resulted in decreased phosphorylation of RIP kinases, but the overexpression of RIPK1 or RIPK3 rescued the effect of caspase-9 deficiency. Inhibition of either Aurora kinase A (AURKA) or its known substrate, glycogen synthase kinase 3β (GSK3ß) restored necroptosis sensitivity of caspase-9-deficient cells, indicating an interplay between caspase-9 and AURKA-mediated pathways to regulate necroptosis. Our findings suggest that caspase-9 acts as a newly identified regulator of necroptosis, and thus, caspase-9 provides a promising therapeutic target to manipulate the immunological outcome of cell death.

The scientific examination of short food supply chains is justified by consumer interest and their role in the European Unions&rsquo, subsidy policy and rural development. In our current article, we present the results of a consumer survey of more than a thousand people from Hungary. It was conducted in the North Hungarian (rural) region. Our research goals were: (1) To access consumer demand for short food supply chains and to determine the characteristics of consumers who prefer local foods, and (2) to examine the role of short food supply chains in rural development by a territorial comparison. The results showed that consumers&rsquo, willingness to pay a theoretical premium for local products was high (averagely +20.7%). Compared to that, we considered their monthly purchased quantity low. We found a statistically valid, yet weak, correlation that on a monthly basis, the purchase of local products was more significant for consumers from smaller settlements and villages than those from (greater) cities. Furthermore, there were factors in some consumers&rsquo, preferences that indicated a greater interest for small producers&rsquo, goods. Although it was not possible to assess the absolute demand of the studied rural area, these results (at a certain level) confirm the potential of short food chains in rural development.

Purpose. This study is aimed at investigating the phenotype, differentiation potential, immunomodulatory properties, and responsiveness of saphenous vein vessel wall-derived mesenchymal stromal cells (SV-MSCs) to various TLR ligands and proinflammatory cytokines, as well as comparing their features to those of their bone marrow-derived counterparts (BM-MSCs). Methods. SV-MSCs were isolated by enzymatic digestion of the saphenous vein vessel wall. Phenotype analysis was carried out by flow cytometry and microscopy, whereas adipogenic, chondrogenic, and osteogenic differentiation potentials were tested in in vitro assays. For comparative analysis, the expression of different stemness, proliferation, and differentiation-related genes was determined by Affymetrix gene array. To compare the immunomodulatory properties of SV-MSCs and BM-MSCs, mixed lymphocyte reaction was applied. To investigate their responses to various activating stimuli, MSCs were treated with TLR ligands (LPS, PolyI:C) or proinflammatory cytokines (TNFα, IL-1β, IFNγ), and the expression of various early innate immune response-related genes was assessed by qPCR, while secretion of selected cytokines and chemokines was measured by ELISA. Results. The isolated SV-MSCs were able to differentiate into bone, fat, and cartilage cells/direction in vitro. SV-MSCs expressed the most important MSC markers (CD29, CD44, CD73, CD90, and CD105) and shared almost identical phenotypic characteristics with BM-MSCs. Their gene expression pattern and activation pathways were close to those of BM-MSCs. SV-MSCs showed better immunosuppressive activity inhibiting phytohemagglutinin-induced T lymphocyte proliferation in vitro than BM-MSCs. Cellular responses to treatments mimicking inflammatory conditions were comparable in the bone marrow- and saphenous vein-derived MSCs. Namely, similar to BM-MSCs, SV-MSCs secreted increased amount of IL-6 and IL-8 after 12- or 24-hour treatment with LPS, PolyI:C, TNFα, or IL-1β, compared to untreated controls. Interestingly, a different CXCL-10/IP-10 secretion pattern could be observed under inflammatory conditions in the two types of MSCs. Conclusion. Based on our results, cells isolated from saphenous vein vessel wall fulfilled the ISCT’s (International Society for Cellular Therapy) criteria for multipotent mesenchymal stromal cells, and no significant differences in the phenotype, gene expression pattern, and responsiveness to inflammatory stimuli could be observed between BM-MSCs and SV-MSCs, while the latter cells have more potent immunosuppressive activity in vitro. Further functional assays have to be performed to reveal whether SV-MSCs could be useful for certain regenerative therapeutic applications or tissue engineering purposes.

The forest is one of the most complex natural ecosystems that is one of the basic living conditions of the healthy human life due to its effects on the environment. According to their functions forests can fill security, economic, social, health, tourism, and education as well as research roles. After the end of World War I the level of forest cover decreased to 11.8% in Hungary. At this time Hungary was Europe’s fourth poorest country in forest and tree. The increasing of forest cover was an important objective in each areas since then. The location and the size of the afforested area ultimately will be appointed by the landowners’ intentions. The state can support the success of the afforestation program by various devices, and promote effectively the enforcement of public interest. The New Hungary Rural Development Programme (2007-2013) envisaged the deployment of 69,000 hectares of new forest, of which 70% were implemented. Under the measure support was granted for the first afforestation of areas withdrawn from agricultural cultivation. Despite the above average extent of forest covers in Heves County (24.5% in 2006), further opportunities were opened to increase the forest area. In afforestation terms the high priority regions includes the poor quality arable lands of feet of Mátra and Bükk mountains and the reclamation areas of Visonta open-cast lignite mine in Heves County. During the research, we worked on secondary databases with a view to examine how regional differences there are in within Heves County in the case of location of forest areas and implemented forest plantations. On the one hand we used the data of Forestry Directorate of the National Food Chain Safety Office, on the other hand we worked up the statistics of Agricultural and Rural Development Agency about afforestation subsidies supported in the framework of New Hungary Rural Development Programme (2007-2013). There are significant differences among the districts of Heves County regarding the extent of forest cover between upland and lowland areas. While Pétervására and Bélapátfalva districts were characterized by 61% and 59% forest cover in 2006, at the same time in Füzesabony, Heves and Hatvan districts the same value was slightly higher than 4%. In the period between 2006 and 2015 the rate of forestation increased in all districts of the County at least by 0.2 percent. The growth rate was more than one percent in Heves megye járásai között az erdősültség mértékét illetően jelentős különbségek figyelhetők meg a hegyvidéki és alföldi területek között. Miközben a Pétervásárai járást és a Bélapátfalvai járást 61, illetve 59%-os erdősültség jellemezte, addig a Füzesabonyi, a Hevesi és a Hatvani járásban ez az érték alig haladta meg a 4%-ot. A megye minden egyes járásában növekedett az erdősültség mértéke a 2006 ás 2015 közötti időszakban legalább 0,2 százalékponttal. 1 százalékpont feletti volt a növekedés a Pétervásárai és a Gyöngyösi járásban. Ezáltal jelentős kiterjedésű, mezőgazdasági művelésre kevéssé alkalmas terület kapott új funkciót.Pétervására and Gyöngyös district. Therefore, a significant expanse of land that is less suitable for agricultural cultivation has given a new function.

The Hungarian Chamber of Agriculture (HCA) was established in the spring of 2013 as a public body. The main tasks of the HCA are strengthening and advocacy of domestic agricultural and food sector, supporting the competitiveness of Hungarian food, furthermore the consultancy and delivering fast, accurate and reliable information to farmers. After the integration of village consultants’ network in 2014 the HCA established a unified support system for farmers. This system based on five pillars: information, consulting, training, project management and European Innovation Partnership. In Heves County 27 village consultants and 4 village consultant administrators perform advisory tasks coordinated by the chief village consultants. The scope of official duties performed by the network: crop estimation, data collection and reporting tasks associated with state rating, assessment of damages in agriculture etc. The advisors validate about 9500 farmers’ cards year by year. In our research we examined the main features of the village consultants and farmers receiving services (such as age, gender, level of education, professional experience, current scope of activities and contacts between the two groups). We hypothesized that the age and vocational qualifications of the farmers are determining the number of services used. To answer our research questions we were performed primary data collection in Heves County. We compiled two questionnaires, one for the farmers (N=150) and one for the village consultants (N=18). To answer the remaining outstanding issues we conducted interview-based survey involving 4 experts. In the course of the survey research for the village consultants we examined theirs most important activities and ranked by the number of mention. The five most common cases were the Unified Application Administration, the validation of farmers’ cards, information services in connection with former Agricultural and Rural Development Agency, monitoring data service and Chamber membership fee acknowledgment. The farmers we’ve asked were all familiar with the local village consultant and 88% of them known the office client’s time. The 69% of the respondents more than three times visited the advisor. Based on our research the village consultants completed more than 50% of administrative tasks of farmers in the 70% of cases. Overall, the village consultant network plays an important role in the life of the farmers regardless of age or level of education.

In the last decades, the area of agricultural land has been declined permanently in Hungary. Despite the diminishing production area, Hungary is among the leaders in terms of the proportion of agricultural land to the total area in the European Union. There are several reasons of using the arable land. As other economic sectors have gained a greater role in the production of GDP and employment, they have taken a larger area from agriculture. Real estate and infrastructure developments were justified by raising the standard of living of the population. The soil, nature, landscape and water protection aspects were also involved. In Hungary, the arable land is the most important natural resource, so it is very important to protect it. In our study, we examine the changes of arable land and uncultivated area in Hungary based on secondary databases of the Hungarian Central Statistical Office and departments of the Ministry of Agriculture. In the course of the research we focused on regional disparities.

Az utóbbi évtizedekben, úgy, mint korábban az európai nagyvárosi térségekben, Magyarországon is elindultak a szuburbanizációs folyamatok. Ez elsőként és legnagyobb volumenben a Budapesti agglomerációt érintette, kifejtve számos hatását. A folyamatosnak tekinthető kiköltözések következményeként az elővárosi lakosság növekedett, a központi településen lakók száma csökkent. Ez a folyamat később, a 2000-es évekre egyre inkább felgyorsult, és elterjedt az ország valamennyi nagyvárosi térségében. Tanulmányunk egy kis-középváros, Gyöngyös környezetére koncentrál. A lakóhelyi szuburbanizációhoz köthető jelenségeket a Gyöngyösi járás területén vizsgáltuk meg, feltárva azt, hogy mik voltak a vidéki településekre történő költözési folyamatok mozgatórugói 2001-től napjainkig. Ennek vizsgálatára szuburbanizációs index számítását végeztük el a járást alkotó települések mindegyikére a központi város kivételével. A szuburbanizációban résztvevő lakosság összetételét, motivációs jellemzőit lakossági kérdőívezés eredményei alapján elemeztük.

A tanulmány középpontjában a gazdaság fejlődésének térbeli dimenziói állnak, amelyek bemutatására a Nemzeti Adó- és Vámhivatal, a Nemzetgazdasági Minisztérium és a Központi Statisztikai Hivatal adatbázisainak feldolgozása alapján került sor. Az Észak-magyarországi régiógazdaságának térbeli folyamatait a személyi jövedelemadó, valamint a társasági adóbevallás kiemelt adatai mellett a gazdasági aktivitásra és a demográfiai folyamatokra (különös tekintettel a vándorlásra) vonatkozó információkkal elemeztük. Megvizsgáltuk, hogy a régió térszerkezetét hagyományosan meghatározó centrumok és tengelyek jelentősége hogyan változott az ezredfordulót követően. Kiemelt figyelmet fordítottunk az „Északkelet-magyarországi innovációs tengely” mentén,valamint a régió külső- és belső periféria területein lezajlott folyamatokra. Azt követően, hogy az elmaradott régiót 2005 és 2010 között további leszakadás jellemezte az országos átlaghoz képest, a 2010-es években már egyértelműen a felzárkózás jelei mutatkoztak. A belföldi jövedelem bővülése egyes periférikus térségekben kifejezetten látványos volt, azonban a vállalkozások bruttó hozzáadot tértéke továbbra is a városokra és néhány vidéki kulcstelepülésre koncentrálódik. Hosszabb távon a kedvezőtlen népesedési folyamatok szabhatnak gátat a további fejlődésnek.

Tanulmányunkban arra keressük a választ, hogy a rövid ellátási láncok sikerét mennyiben befolyásolhatja a jelenlegi helyi termék kínálat, valamint az értékesítést biztosító helyi „hagyományos" piacok, termelői piacok, biopiacok száma és területi lehelyezkedése. Az ellátási láncot tehát a termelők és az értékesítési csatorna oldaláról közelítettük meg, két minta területen (Komárom-Esztergom, Heves megye). Mindkét megye területén, elsődleges és másodlagos adatbázisok segítségével mértük fel az említett tényezőket, külön megvizsgálva térbeli eloszlásukat, és az ebből fakadó területi különbségeket. Vizsgálati eredményeinket összevetettük a rendelkezésre álló európai uniós források által kínált pályázati kiírás feltételrendszerével, lehatárolva a potenciális pályázók körét, a lehetséges REL-ek számát.

A Tudományos Ismeretterjesztő Társulat Stúdió Egyesület (TIT SE: www.tit.hu) 2018-ban meghirdette és megszervezte az „Egy napom a jövőben” gyermek-rajz versenyt és kiállítást, a Budapest XI. kerület alsó tagozatos iskolásai számára. 44 alkotás érkezett. A fotókkal illusztrált írás a kiállítás előzményeit, jellemzőit tekinti át, elemezve az itt (szülői egyetértéssel) közölt alkotásokat.The Society Dissemination of Scientific Knowledge Studio (www.tit.hu) in 2018 organised a competition and exhibition of drawings for elementary school pupils with the title „My one day in the future”. 44 drawings arrived. This event was one of the programs honouring the 50th anniversary of the Hungarian Futures Research. The published study, with photos, describes this pleasant exhibition.

Dendritic cells (DCs), as potent phagocytes engulf dead cells and present peptide fragments of tumor antigens or pathogens derived from infected cells to naive CD8+ T-lymphocytes. Dendritic cells can also induce apoptosis in target cells, thus getting an opportunity to sample their microenvironment. Here, we present that the supernatants of LPS- or CL075-activated DCs induced cell death in different cell lines, but during the differentiation to mature DCs, they lost their cytotoxic potential. Dexamethasone-pre-treated tolerogenic DCs induced less intensive death indicating that the tissue microenvironment can downregulate DC-mediated killing. Exploring the signaling of DC-induced cell death, we observed that the supernatant of activated DCs induced TNF-dependent cell death, since TNF antagonist blocked the cytotoxic activity of DCs, contrary to inhibitors of Fas and TRAIL receptors. We identified that the DC-induced killing is at least partially a RIPK1-dependent process, as RIPK1 positive target cells were more susceptible to DC-induced cell death than their RIPK1 deficient counterparts. Moreover, both the elevated phosphorylation of RIPK1 and the increase in RIPK1-caspase-8 interaction in target cells suggest that RIPK1-mediated signals contribute to DC supernatant-induced cell death. We also proved that the cytotoxic activity of DC-derived supernatant induced apoptosis in the target cells and not necroptosis, as it was completely abrogated with the pan caspase inhibitor (Z-VAD), while the necroptosis inhibitor (Nec-1) had no effect. Our work revealed that the supernatant of activated DCs induces the apoptosis of target cells in a RIPK1-dependent manner. This phenomenon could be relevant for the initiation of cross-presentation and may broaden the plethora of cytotoxic mechanisms acting against tumor cells.

Background BRAF-mutant melanoma is characterized by aggressive metastatic potential and therapeutic resistance. The innate immune receptor RIG-I has emerged as a potential target in melanoma therapies but the contributing pathways involved in anti-cancer activity are poorly characterized. Methods Baseline and ATRA-induced expression of RIG-I in nine (3 wild type and 6 BRAF-mutant) melanoma cell lines was measured with Q-PCR and Western blot. Ligand-specific stimulation of RIG-I was detected by Q-PCR and ELISA. Activation of the RIG-I-coupled IRF3, NF-κB and MAPK pathways was tested with protein array and Western blot. Cell proliferation and apoptosis was monitored by flow cytometry and cell counting. Down modulation of MKP-1 expression in melanoma cells was performed by specific siRNA. Results Short-term ATRA pre-treatment increases the expression of RIG-I in BRAF-mutant melanoma cells. Specific activation of RIG-I by 5ʹppp-dsRNA leads to increased activity of the IRF3-IFNβ pathway but does not influence NF-κB signaling. RIG-I mediates the targeted dephosphorylation of several MAPKs (p38, RSK1, GSK-3α/β, HSP27) via the endogenous regulator MKP-1 resulting in decreased melanoma cell proliferation. Conclusion RIG-I has the potential to exert anticancer activity in BRAF-mutant melanoma via controlling IFNβ production and MAPK signaling. This is the first study showing that RIG-I activation results in MKP-1-mediated inhibition of cell proliferation via controlling the p38-HSP27, c-Jun and rpS6 pathways thus identifying RIG-I and MKP-1 as novel and promising therapeutical targets.

Ultraviolet (UV) light is absorbed by nucleic acids, proteins or other endogenous chromophores, such as porphyrins, flavins and melanin, triggering biological processes in skin cells. Both UV-induced mutations in melanocytes and changes in the immune microenvironment are understood to play a role in the development of cutaneous melanoma. The degree of UV-induced stress and the protection against this stress are influenced by both intracellular and intercellular molecular interactions. The present review summarizes the known major molecular biological changes induced by UV light in the skin that play a role in melanoma initiation and promotion. Nevertheless, cutaneous melanoma is not a homogenous disease, and the interaction of variable environmental exposure and different genetic susceptibility and other host factors lead to the formation of melanomas with different biological behavior and clinical characteristics. This review highlights the challenges in the understanding of how UV radiation contributes to the formation of cutaneous melanoma, and reviews the new results of photobiology and their link to tumor genetics and tumor immunology with potential implications on melanoma prevention and therapeutic strategies. The information presented here is expected to add clarity to ongoing research efforts in this field to aid the development of novel strategies to prevent and treat melanoma.

Serotonin is a monoamine neurotransmitter that signals through a wide array of receptors (5-HT1–7) many of which are also involved in immune processes. Dendritic cells (DCs) are crucial players in immune defense by bridging innate and adaptive immune responses via their vast repertoire of pattern recognition receptors and antigen-presenting capability. Although serotonin is known to influence immunity at many levels, cell type-specific expression and function of its receptors remains poorly understood. Here we aimed to study 5-HT1–7 expression and function in CD1a− and CD1a+ human monocyte-derived DCs (moDCs). We found that the 5-HT2B receptor-subtype is solely expressed by the inflammatory CD1a+ moDC subset. Specific 5-HT2B activation potently inhibited TLR2, TLR3, and TLR7/8-induced proinflammatory cytokine and chemokine (TNF-α, IL-6, IL-8, IP-10, IL-12) but not type I interferon-β responses. 5-HT2B agonism also interfered with the polarization of CD1a+ moDC-primed CD4+ T cells towards inflammatory Th1 and Th17 effector lymphocytes. Here we report the subset-specific expression and immunomodulatory function of 5-HT2B in human moDCs. Our results expand the biological role of 5-HT2B which may act not only as a neurotransmitter receptor, but also as an important modulator of both innate and adaptive immune responses.

Tumors are composed of abnormally transformed cell types and tissues that differ from normal tissues in their genetic and epigenetic makeup, metabolism, and immunology. Molecular compounds that modulate the immune response against neoplasms offer promising new strategies to combat cancer. Inhibitors targeting the indoleamine-2,3-dioxygenase 1 enzyme (IDO1) represent one of the most potent therapeutic opportunities to inhibit tumor growth. Herein, we assess the biochemical role of IDO1 in tumor metabolism and immune surveillance, and review current diagnostic and therapeutic approaches that are intended to increase the effectiveness of immunotherapies against highly aggressive and difficult-to-treat IDO-expressing cancers.

The cytoplasmic nucleotide oligomerization domain 2 (NOD2) receptor recognizes the bacterial cell wall component muramyl dipeptide (MDP). NOD2 ligation initiates the nuclear factor kappa B and the mitogen-activated protein kinase cascades. However, administering MDP alone is insufficient to elicit strong cytokine responses in various immune cells, including dendritic cells (DCs). Because the simultaneous presence of various microbial products and cytokines in inflamed tissues modulates DC function, we initiated this study to examine how interferon gamma (IFNγ), a central modulator of inflammation, affects the NOD2-mediated signaling pathway in human conventional DCs (cDCs). Synergistic stimulation of DCs with MDP and IFNγ increased the expression of CD40, CD80, CD83, CD86, and human leukocyte antigen DQ proteins and significantly elevated the production of pro-inflammatory cytokines IL-1β, IL-6, IL-12, and tumour necrosis factor (TNF), as well as anti-inflammatory cytokine IL-10. Furthermore, the simultaneous presence of MDP and IFNγ was necessary to decrease IkBα protein levels. By investigating various mechanisms implicated in MDP- and IFNγ-mediated signaling pathways, we revealed that the increased production of pro-inflammatory cytokines is highly dependent on the X-linked inhibitor of apoptosis protein (XIAP) but not on cellular IAP1 and IAP2. We also found that the NOD2 signaling pathway is regulated by the mammalian target of rapamycin (mTOR) but is not affected by phosphatidylinositol-3 kinase or signal transducer and activator of transcription 1 inhibition. Our results demonstrate, for the first time, that IFNγ positively affects NOD2-mediated signaling in human cDCs, in a manner considerably dependent on XIAP and partially dependent on mTOR.

How results typical nationwide are also typical for this area which type of direct sales is mostly used here. In former studies market mainly traditional market is prior. The second most widespread way of sales is selling from the house or selli ng on - farm. For farms of producers selling this way is typical to produce several fruits and vegetables are small plant forms and that they do not employ outside workers .

Type I and III IFNs are crucial, soluble components of potent antiviral responses. It has been explored recently that mTOR is involved in the regulation of IFN-α/β production by pDCs, albeit its role in the induction of IFN responses in cDCs remained unrevealed. In this study, we demonstrate that the PI3K/mTOR pathway is indispensable for eliciting intact type I and III IFN responses in moDCs stimulated with polyI:C. The inhibition of mTOR functionality by rapamycin impairs the pIRF3 and also a few members of the MAPK family, suggesting that mTOR contributes to the activation of multiple signaling pathways in the presence of viral antigens. Furthermore, rapamycin-treated moDCs show decreased capacity to prime IFN-γ secretion by naive CD8+ T-lymphocytes. As in moDCs, mTOR-mediated regulation is also essential for the production of type I and III IFNs in circulating CD1c+ DCs. To our best knowledge, these results demonstrate for the first time that mTOR has an impact on the functional activities of cDCs via modulating the outcome of IFN secretion.

The glucocorticoid-induced tumor necrosis factor receptor (GITR) is a member of the tumor necrosis factor receptor superfamily. Attachment of GITR to its ligand (GITRL) regulates diverse biological functions, including cell proliferation, differentiation, and survival. In this study, the extracellular region of human GITRL (hGITRL) was cloned, expressed, and purified. The coding sequence of the extracellular region of hGITRL was isolated from human brain cDNA and inserted in pET20b vector. The hGITRL was expressed in Escherichia coli BL21 (DE3) Star at 37 and 25 °C. The majority of the protein was found in inclusion bodies. We identified three important factors for efficient refolding of hGITRL: a ratio of GSH/GSSG, pH, and addition of polyethylene glycol. The renaturated protein was purified by Ni-NTA chromatography. The overall yield of the expression and refolding was higher than 50 mg/l E. coli culture grown at 37 °C. Size exclusion chromatography showed that hGITRL exists as mixture of various multimeric forms in solution. We tested the association of recombinant hGITRL with THP-1 and U937 cell lines and its activity to promote extracellular signal-regulated protein kinase phosphorylation. The results showed that the recombinant protein was biologically active.
