Nitrite accumulation in hydrogen-based denitrification (HD) has been reported as a difficulty for achieving complete denitrification. Thauera sp. has been found as the dominant bacterial species in HD previously when using a plentiful amount of HCO3−. This present study was successful in isolating Pseudomonas sp., Dietzia sp., Pannonibacter sp., Halomonas sp., Bacillus sp., and Thauera sp. These isolated strains were selected for investigating the nitrogen removal performance under the plentiful HCO3− condition. Only Pseudomonas sp. and Thauera sp. were capable of removing NO2− where the specific NO2− removal rate of Thauera sp. (36.02 ± 5.66 mgN gVSS−1 day−1) was 9 times quicker than that of Pseudomonas sp. (3.94 ± 0.80 mgN gVSS−1 day−1). The Thauera sp. strain was then tested at different HCO3− amounts. As a result, Thauera sp. had no ability to function both NO3− and NO2− removals under HCO3− deficit condition. This study provided evidence on the role of Thauera sp. and the necessity of bicarbonate in the hydrogen-based denitrification process to enhance its efficiency and to simultaneously reduce the operational cost especially for hydrogen.

The physicochemical and biological characteristics of milli-microbubbles were compared to evaluate their performance on hydrogenotrophic denitrification (HD) for groundwater treatment in remote areas. The hydrogen supply was controlled at 1.14 L/d with 40 mgN/L of NO3-N. The microbial community structure in two bubble reactors was investigated by high throughput sequencing. Microbubbles enhanced biodegradation in the HD system, providing a maximum nitrogen removal efficiency of 99%. Approximately 50% of total hydrogen was utilized for biological nitrate removal with the highest hydrogen effectiveness achieved at 1.21 g N/g H2. In contrast, millibubbles achieved less than 10% efficiency and 9.9% of total hydrogen was consumed for biological nitrogen removal. Thaueraspp., Hydrogenophagaspp. and Rhodocyclaceaeof Proteobacteriaphylum were the dominant bacteria in the microbubble reactor, whereas Methyloversatilis spp.was dominant in the millibubble reactor, in which a relatively low amount of hydrogen (0.6 mg/L) was dissolved. The differences can be attributed to the higher hydrogen transfer efficiency (45×10-3s-1) and lower rising velocity (0.31 mm/s) of the microbubbles system than the millibubbles system (2×10-3s-1and 480 mm/s). The micro-hydrogen bubble technology affords increased hydrogen effectiveness, reduced energy consumption, and modified system design. Therefore, it is more appropriate for enhancing HD.

Due to the high water demand and unsustainable water resource, wastewater reclamation and wastewater treatment prior to discharge have become current important issues. Various treatment technologies, such as biological processes, have been improved as alternatives. In this study, the biological nitrogen removal system using pure-culture Bacillus licheniformis was developed and used as an internal treatment unit in an aquarium to improve the effluent quality for water reuse. The efficiencies for NH4-N and total nitrogen (TN) removal and the quality of treated water verified the occurrence of heterotrophic nitrification and aerobic denitrification; the nitrification rate was 0.84 mg/L-h and the denitrification rate was 0.62 mg/L-h. The maximal NH4-N and TN removal efficiencies were approximately 73% at the influent NH4-N of 30 mg/L. However, the other competitive heterotroph of Pseudomonas sp. was observed, which resulted in dramatically decreasing efficiencies and an enlarged ratio of carbon consumption and nitrogen removal. Although the overall performance of the B. licheniformis system was lower than the system using mixed-culture nitrifying and heterotrophic denitrifying microorganisms, the advantages of the B. licheniformis system were ease of operation and the fact that it is a land-limited treatment system. The research is ongoing to enhance performance and maintain excellent efficiency in a long-term operation.

Nitrate removal during anaerobic ammonium oxidation (anammox) treatment is a concern for optimization of the anammox process. This study demonstrated the applicability and long-term stability of the coupled anammox and hydrogenotrophic denitrification (CAHD) process as an alternative method for nitrate removal. Laboratory-scale fixed bed anammox reactors (FBR) supplied with H2 to support denitrification were operated under two types of synthetic water. The FBRs showed simultaneous NH4-N and NO3-N removal, indicating that the CAHD process can support NO3-N removal during the anammox process. Intermittent H2 supply (e.g. 5 mL/min for a 1-L reactor, 14/6-min on/off cycle) helped maintain the CAHD process without deteriorating its performance under long-term operation and resulted in a nitrogen removal rate of 0.21 kg-N/m3/d and ammonium, nitrate, and dissolved inorganic nitrogen removal efficiencies of 73.4%, 80.4%, and 77%, respectively. The microbial community structure related to the CAHD process was not influenced by changes in influent water quality, and included the anammox bacteria ‘Candidatus Jettenia’ and a Sulfuritalea hydrogenivorans-like species as the dominant bacteria even after long-term reactor operation, suggesting that these bacteria are key to the CAHD process. These results indicate that the CAHD process is a promising method for enhancing the efficiency of anammox process.

High nitrate-nitrogen (NO3−–N) content is a typical feature of groundwater, which is the primary water source in the Kathmandu Valley, Nepal. Considering the Kathmandu Valley’s current problem of water scarcity, a user-friendly system for removing NO3−–N from groundwater is promptly desired. In this study, a simplified hydrogenotrophic denitrification (HD) reactor was developed for the Kathmandu Valley, and its effectiveness was evaluated by its ability to treat raw groundwater. The reactor operated for 157 days and showed stability and robustness. It had an average nitrogen removal efficiency of 80.9 ± 16.1%, and its nitrogen loading rate and nitrogen removal rate varied from 23.8 to 92.3 g–N/(m3∙d) and from 18.3 to 73.7 g–N/(m3∙d), respectively. Compared to previous HD reactors, this simplified HD reactor is a more user-friendly option for the Kathmandu Valley, as most of the materials used for the reactor were locally available and require less maintenance. The reactor is recommended for groundwater treatment at the household level. It has a current treatment capacity of 40 L/d, which can fulfill the daily requirements for drinking and cooking water in a household with 4–5 people.

The abundance and diversity of anaerobic ammonium oxidation (anammox) bacteria were assessed in 152 groundwater samples in the Kathmandu Valley, Nepal. Anammox bacterial 16S rRNA genes were detected in 54% (37/68) of samples collected in the dry season at 1.6×105–8.8×106 copies L–1, and in 60% (50/84) of samples collected in the wet season at 4.3×104–1.2×107 copies L–1. The 16S rRNA genes of “Candidatus Brocadia”, “Candidatus Anammoxoglobus”, and five new deduced anammox bacterial phylotypes were detected in the shallow groundwater samples. Diverse anammox bacteria were broadly distributed in the shallow groundwater aquifer of the Kathmandu Valley.

Shiga toxin-producing Escherichia coli (STEC) is one of the most commonly heard sources of foodborne outbreaks but presently lacks studies of in the Kathmandu Valley. This study explored the presence of STEC in river water (n = 17), groundwater (n = 83), feces (n = 50), and manure (n = 20) in the Kathmandu Valley, Nepal. Samples that were confirmed to be E. coli–positive using the Colilert assay in previous studies were selected to examine the presence of the sfmD, stx1, and stx2 genes using quantitative polymerase chain reaction. Based on Colilert/sfmD gene ratios, the percentage of viable cells of E. coli in the fecal (2.0% ± 2.5%), manure (0.1% ± 0.1%), river water (3.4% ± 3.1%), and groundwater samples (3.9% ± 5.8%) were determined. The presence of the stx1 gene was observed in the fecal (6%), river water (53%), and groundwater (2%) samples, whereas the stx2 gene was detected in the fecal (8%), manure (5%), and river water (71%) samples. Interestingly, the stx/sfmD gene ratios in the groundwater samples were greater than 100%, suggesting the presence of other stx1/stx2-harboring microorganisms. These findings indicated the importance of continuing investigation into waterborne sources of STEC contamination within the Kathmandu Valley.

A dropping nitrification reactor was proposed as a low-cost and energy-saving option for the removal of NH4+-N from contaminated groundwater. The objectives of this study were to investigate NH4+-N removal performance and the nitrogen removal pathway and to characterize the microbial communities in the reactor. Polyolefin sponge cubes (10 mm &times, 10 mm &times, 10 mm) were connected diagonally in a nylon thread to produce 1 m long dropping nitrification units. Synthetic groundwater containing 50 mg L&minus, 1 NH4+-N was added from the top of the hanging units at a flow rate of 4.32 L day&minus, 1 for 56 days. Nitrogen-oxidizing microorganisms in the reactor removed 50.8&ndash, 68.7% of the NH4+-N in the groundwater, which was aerated with atmospheric oxygen as it flowed downwards through the sponge units. Nitrogen transformation and the functional bacteria contributing to it were stratified in the sponge units. Nitrosomonadales-like AOB predominated and transformed NH4+-N to NO2&minus, N in the upper part of the reactor. Nitrospirales-like NOB predominated and transformed NO2&minus, N to NO3&minus, N in the lower part of the reactor. The dropping nitrification reactor could be a promising technology for oxidizing NH4+-N in groundwater and other similar contaminated wastewaters.

Building resilience into water systems is very important in urban areas, particularly in less-developed countries, as poor-quality systems have adverse effects on human development and poverty reduction. This mixed-methods study aims to assess the resilience of household water systems and their impact on the quality of life (QOL) after the 2015 earthquake in Nepal. Data were obtained from 1,500 households using a questionnaire and six focus group discussions. The resilience of each household's water system was assessed in terms of residents' pre- and post-earthquake perceptions of water security. Approximately 60% of the households considered their water systems to be resilient. Piped water and wells were associated with stronger perceptions of water system resilience. Participants who considered their household water system to be resilient had higher QOL than those who considered their water systems vulnerable after the earthquake. Qualitative data showed that both the quality and quantity of water deteriorated due to an earthquake. To mitigate the adverse effects of natural disasters on water security, we recommend strengthening the water system infrastructure and operations as well as implementing collaboration between governments and local people.

Groundwater available in the Kathmandu Valley is not suitable for drinking due to chemical and microbial contamination. We installed a treatment system, which was made with locally available materials and was low-cost, and supplied drinking water to the intervention site where groundwater contains high amounts of ammonia, iron, and turbidity. This research aims to evaluate the socioeconomic impact of treated water distribution. One hundred households were randomly selected and asked to use treated water for drinking, and another 100 households in the nearby community were taken randomly as a control. We conducted questionnaire surveys with the enrolled households before and five months after starting water distribution to assess the water use patterns and quality perceptions. The socioeconomic impact of the intervention was evaluated by a prepost comparison and by the difference-in-difference method. The intervention significantly enhanced most of the parameters of water quality perception, reduced the in-house water treatment, and improved the perceived water stress and quality of life. For the control site, these parameters generally became worse in the post-survey, which suggests that the survey might have affected people&rsquo, s mindset regarding water security. The system is an option for sustainable management of drinking water in the water-scarce, hard-hit areas in the developing countries.

Groundwater is a major alternative water source used to cover the deficit of water supplied by Kathmandu Upatyaka Khanepani Limited (KUKL), the authority responsible for water supply inside Kathmandu Valley. The groundwater price relative to that of KUKL affects priority of usage, and hence, groundwater resources sustainability. Therefore, taxation or subsidies on water sources become necessary based on their implication on environment. In this study, we evaluate volumetric water price, including initial investment, operation and maintenance (O&amp, M) cost for different water sources, and compare it with the water price of KUKL, Kathmandu. The results show that shallow groundwater is cheaper than KUKL&rsquo, s water. For groundwater sustainability, taxation on shallow groundwater seems necessary. For the recent water use of 97 LPCD (liters per capita per day) the taxation requirement is Nepalese Rupee (NRs.) 320/month (0.35% of total expenditure) if the initial investment for well construction and O&amp, M cost are considered, and NRs. 626 (0.7% of total expenditure) if only O&amp, M cost is considered. On the other hand, rainwater harvesting and recharging, the measures to cope with groundwater exploitation, might need 40% to 50% subsidy for their initial investment.

Textile wastewater (TW) contains toxic pollutants that pose both environmental and human health risks. Reportedly, some of these pollutants, including NO3-, NO2- and reactive black 5 (RB-5) dye, can be removed via hydrogen-based denitrification (HD), however, it is still unclear how different factors affect their simultaneous removal. This study aimed to investigate the effect of H2 flow rate, the sparging cycle of air and H2, and initial dye concentration on the TW treatment process. Thus, two reactors, an anaerobic HD reactor and a combined aerobic/anaerobic HD reactor, were used to investigate the treatment performance. The results obtained that increasing the H2 flow rate in the anaerobic HD reactor increased nitrogen removal and decolorization removal rates. Further, increasing the time for anaerobic treatment significantly enhanced the pollutant removal rate in the combined reactor. Furthermore, an increase in initial dye concentration resulted in lower nitrogen removal rates. Additionally, some of the dye was decolorized during the HD process via bacterial degradation, and increasing the initial dye concentration resulted in a decrease in the decolorization rate. Bacterial communities, including Xanthomonadaceae, Rhodocyclaceae, and Thauera spp., are presented as the microbial species that play a key role in the mechanisms related to nitrogen removal and RB-5 decolorization under both HD conditions. However, both reactors showed similar treatment efficiencies, hence, based on these results, the use of a combined aerobic/anaerobic HD system should be used to reduce organic/inorganic pollutant contents in real textile wastewater before discharging is recommended.

This study investigates the contribution ratios of different groundwater recharge sources and influence of a dual monsoon system in Kofu basin, central Japan, through the hydrogen and oxygen isotopic analysis of precipitation, river water, and groundwater. The study is focused on the area of the Kamanashigawa and Midaigawa alluvial fans, which are formed by two main rivers. Precipitation isotopic content exhibits significant seasonal variability. Also, river water presents d-excess values higher than annual precipitation at plain areas (9 and 10‰), suggesting that two different air-masses contribute to precipitation, corresponding to the monsoon and pre-monsoon periods. The results of this study allow estimation of relative contributions of different sources to groundwater and influence of a dual monsoon system. The mass-balance analysis of the δ18O to identify the groundwater source indicates the river water contributes 38–100% of the recharge, while precipitation contributes 29–62% in Kamanashigawa alluvial fan. In the case of Midaigawa alluvial fan, river water contributes 77–99% in the northern part; in the southern side, 30–93% of contribution comes from precipitation. The mass-balance analysis of the d-excess indicates pre-monsoon precipitation contributes 46–68% and 39–65% to groundwater of the Kamanashigawa and Midaigawa alluvial fans, respectively.

Kathmandu Valley faces water scarcity from decades of the added burden of water storage and treatment, which imposed cost. We estimated the method-specific cost of in-house drinking water treatment (9 L/day used) based on equipment price and life, daily operation time, fuel used, and consumables replacement frequency, which were Nepalese Rupees (NRs) 23, 57, 392, 586 and 799 for chemicals, ceramic filter, boiling, Euro-Guard and reverse osmosis-ultraviolet (RO-UV) water purification, respectively. The monthly average water treatment cost was estimated based on these estimates and treatment methods used in households, obtained from a questionnaire survey of 1500 households, and its socioeconomic relationship in a Nepalese urban context was investigated. Of the households, 75% practiced at least one treatment method, (average, 1.4). The estimated monthly average cost per household was NRs 380. The fixed effects model showed that the cost was significantly higher in Lalitpur district, and in high school education households. Higher water insecurity perception of respondents was the main determinant of higher treatment cost, which was especially true in Lalitpur district. Water treatment added extra financial burden, especially for the poor households which should be averted or minimized by concerned authorities to provide adequate quantity, quality, and access to drinking water for all.

Sustainability of water supply services has been central to the main water supply agenda globally. In order to promote affordable, equitable, and sustainable water supply, this paper proposed a method to set the piped water price inside Kathmandu Valley, Nepal. The Melamchi Water Supply Project (MWSP) is currently under construction inside the valley to tide over the water deficit. Domestic water use and expenditure data were obtained from a household survey of 1500 households. Post-MWSP were developed based upon per capita water use and water price. Equity and affordability were evaluated by comparing the ratio of water expenditure to total expenditure of households (expenditure ratio: ER) in each service area. The expenditure for piped water service and alternate water sources was taken into account to calculate the total water expenditure. Sustainability was measured as the ratio of operation and maintenance cost to the total revenue of the utility (Working ratio: WR). The results show that an increase in water price by approximately 54% over the current price is necessary to meet operation and maintenance costs, assuming the WR to be 0.7. Approximately 30% increase in water use (a minimum of 80 LPCD) can be maintained post MWSP. However, water use of 135 LPCD increased the ER by more than 1%, which was the threshold of water affordability and may cause ground water stress.

The distribution of benthic macroinvertebrates was studied in deep Lake Motosu (oligotrophic lake), Japan. Bathymetrical sampling surveys were carried out using a standard Ekman grab at each of 16 stations in the lake on 8 March 2013. The average densities of the benthic community for all the stations were 2185 ind./m2, comprised principally of Tubifex tubifex (Muller) 23.4%, Asellus hilgendorfi Bovallius 4.9% and chironomids 65.8%. T. tubifex and chironomids inhabited the entire lake bottom, with higher densities in the shallower region (25 m>) than in the deeper region (ca. 120 m). On the other hand, the density of A. hilgendorfi increased with water depth and was highest in the deeper regions (>70 m). The densities of chironomid larvae were lower in the deeper region, with fewer chironomid larvae located in the center of the lake (637 ind./m2). The most abundant species, Micropsectra yunoprima Sasa, exhibited the widest distribution from 10.0 m to 121.0 m (mean density 926 ind./m2), followed by Procladius choreusi (Meigen) from 10 m to 74.1 m (mean density 365 ind./m2). The results of this study were compared to previous data by Kitagawa (1973) on chironomid larvae. The densities of these chironomid larvae increased throughout the whole lake in 2013. The water quality has not undergone a major change since the 1970’s. In this study, the number of M. yunoprima larvae has shown a tendency to increase, especially in shallower regions; where they are widely distributed, suggesting the continuation of favorable bottom condition for larvae since the 1970’s., Article, Europian Journal of Environmental Science. 5(1):35-40 (2015)

Water security is one of the central global issues today. This study aimed to describe and test the validity and reliability of a household water insecurity scale, and to assess the impact of household water insecurity on psychological distress among 371 women living in urban Nepal. A household interview survey was conducted using a structured questionnaire. Approximately 70% of the participants experienced collecting less water than they needed. Four domains of household water insecurity emerged from the principal components factor analysis: (i) difficulties in house-work related to water, (ii) lost opportunity costs and social interactions related to water, (iii) an insufficient safe water supply, and (iv) difficulties in basic activities related to water. Multivariate regression analysis showed that the domains of ‘lost opportunity costs and social interactions’, ‘difficulties in house-work’ and ‘difficulties in basic activities’ were associated with psychological distress among women. The impact of household water insecurity on psychological distress differed by water supply service levels. The current household water insecurity scale is a simple instrument which can be used to prioritise the target population for water interventions. Further research should be conducted towards the development of a universally applicable measurement tool.

This study estimated spatial and seasonal variation of water quality to understand characteristics of Nakdong river basin, Korea. All together 11 parameters (discharge, water temperature, dissolved oxygen, 5-day biochemical oxygen demand, chemical oxygen demand, pH, suspended solids, electrical conductivity, total nitrogen, total phosphorus, and total organic carbon) at 22 different sites for the period of 2003-2011 were analyzed using multivariate statistical techniques (cluster analysis, principal component analysis and factor analysis). Hierarchical cluster analysis grouped whole river basin into three zones, i.e., relatively less polluted (LP), medium polluted (MP) and highly polluted (HP) based on similarity of water quality characteristics. The results of factor analysis/principal component analysis explained up to 83.0%, 81.7% and 82.7% of total variance in water quality data of LP, MP, and HP zones, respectively. The rotated components of PCA obtained from factor analysis indicate that the parameters responsible for water quality variations were mainly related to discharge and total pollution loads (non-point pollution source) in LP, MP and HP areas; organic and nutrient pollution in LP and HP zones; and temperature, DO and TN in LP zone. This study demonstrates the usefulness of multivariate statistical techniques for analysis and interpretation of multi-parameter, multi-location and multi-year data sets.
