Abstract Hydroxyapatite (HA) is a bioceramic material widely used as a bone biomimetic substitute and can be synthesized by biomineralization, according to which HA nanoparticles are formed on a polymer template. Nevertheless, little is known about the effect of ion doping and biomineralization on cell metabolism, oxidative stress, and DNA damage. In the present contribution, we report on synthesizing and characterizing biomineralized chitosan as a polymer template with HA nanoparticles doped with magnesium (MgHA) and iron ions (FeHA). The physical-chemical and morphological characterization confirmed the successful synthesis of low crystalline ions-doped HA nanoparticles on the chitosan template, whereas the biochemical activity of the resulting nanoparticles towards human osteoblasts-like cells (MG63 and HOBIT) was investigated considering their effect on cell metabolism, proliferation, colony formation, redox status, and DNA damage extent. Data obtained suggest that particles enhance cell metabolism but partially limit cell proliferation. The redox status of cells was measured suggesting a slight increase in Reactive Oxygen Species production with chitosan biomineralized with iron-doped HA, whereas no effect with magnesium-doped HA and no effect of all formulations on the oxidation level of Peroxiredoxin. On the other hand, DNA damage was investigated by COMET assay, and expression and foci γH2AX. These latter tests indicated that HA-based nanoparticles promote DNA damage which is enhanced by chitosan thus suggesting that chitosan favors the nanoparticles’ internalization by cells and modulates their biological activity. The potential DNA damage should be considered – and potentially exploited for instance in anticancer treatment – when HA-based particles are used to devise biomaterials.

Abstract Background Considering the challenges posed by nitrogen (N) pollution and its impact on food security and sustainability, it is crucial to develop management techniques that optimize N fertilization in croplands. Our research intended to explore the potential benefits of co-inoculation with Azospirillum brasilense and Bacillus subtilis combined with N application rates on corn plants. The study focused on evaluating corn photosynthesis-related parameters, oxidative stress assay, and physiological nutrient use parameters. Focus was placed on the eventual improved capacity of plants to recover N from applied fertilizers (AFR) and enhance N use efficiency (NUE) during photosynthesis. The two-year field trial involved four seed inoculation treatments (control, A. brasilense, B. subtilis, and A. brasilense + B. subtilis) and five N application rates (0 to 240 kg N ha−1, applied as side-dress). Results Our results suggested that the combined effects of microbial consortia and adequate N-application rates played a crucial role in N-recovery; enhanced NUE; increased N accumulation, leaf chlorophyll index (LCI), and shoot and root growth; consequently improving corn grain yield. The integration of inoculation and adequate N rates upregulated CO2 uptake and assimilation, transpiration, and water use efficiency, while downregulated oxidative stress. Conclusions The results indicated that the optimum N application rate could be reduced from 240 to 175 kg N ha−1 while increasing corn yield by 5.2%. Furthermore, our findings suggest that replacing 240 by 175 kg N ha−1 of N fertilizer (-65 kg N ha−1) with microbial consortia would reduce CO2 emission by 682.5 kg CO2 −e ha−1. Excessive N application, mainly with the presence of beneficial bacteria, can disrupt N-balance in the plant, alter soil and bacteria levels, and ultimately affect plant growth and yield. Hence, highlighting the importance of adequate N management to maximize the benefits of inoculation in agriculture and to counteract N loss from agricultural systems intensification.

Background: In recent years, the use of conometric systems to connect dental implant abutments and prosthetic caps has been advocated because they seem to eliminate the side effects reported when using screw- and cement-connected prosthetic restorations. Objectives: The present case study is focused on conometric connection characterization and its performance in terms of the microarchitecture of peri-implant soft tissues by using a cross-linked approach based on optical microscopy and three-dimensional imaging. Methods: Two dental implants were characterized using micro-CT and another identical one was implanted into a patient; the latter was retrieved 45 days later due to changes in prosthetic needs. Afterward, the peri-implant soft tissues were investigated using synchrotron-based phase contrast imaging, histology, and polarized light microscopy. Results: Micro-CT analysis showed perfect adhesion between the abutment and prosthetic cap; histology and polarized light microscopy showed that connective tissue was richly present around the abutment retrieved from the patient. Moreover, the quantitative evaluation of connective tissues using synchrotron imaging, supported by artificial intelligence, revealed that this tissue was rich in mature collagen, with longitudinal and transverse collagen bundles intertwined. The number and connectivity of transverse bundles were consistently greater than those of the longitudinal bundles. Conclusion: It was found that the peri-implant soft tissue was already mature and well organized after only 45 days of implantation, supporting the hypothesis that conometric connections contribute to the significant stabilization of peri-implant soft tissues.

Interspecific and intraspecific differences in the efficiency of utilization of soil phosphorus (P) are explained, in part, by plant morphological and physiological variations, which characterize the plant for nutrient acquisition. As part of a study on maize genotypes characterization for P-uptake and use efficiency, kinetic parameters of P uptake and root morphological characters were evaluated for six local and improved maize varieties, grown in nutrient solution. The kinetic parameters related to P influx were: maximal transport rate (Vmax), the Michaelis-Menten constant (Km), and the external concentration when net uptake is zero (Cmin). Measured root morphological characters were: root length, radius, and surface area. Shoot and root dry matter yield and the P content in these parts were also determined. Diferences among varieties were observed in the kinetic parameters Vmax and Km, root length and in root/shoot dry matter ratio. Lower Km values were better indicatives of P uptake ability of the varietes and were significantly correlated to higher dry matter production and P-efficiency index.

The activity of the enzyme phosphatase (P-ase) is a physiological characteristic related to plant efficiency in relation to P acquisition and utilization, and is genetically variable. As part of a study on maize genotype characterization in relation to phosphorus (P) uptake and utilization efficiency, two experiments were set up to measure phosphatase (P-ase) activity in intact roots of six local and improved maize varieties and two sub-populations. Plants were grown at one P level in nutrient solution (4 mg L-1) and the P-ase activity assay was run using 17-day-old plants for varieties and 24-day-old plants for subpopulations. Shoot and root dry matter yields and P concentrations and contents in plant parts were determined, as well as P-efficiency indexes. Root P-ase activity differed among varieties, and highest enzimatic activities were observed in two local varieties -'Catetão' and 'Caiano' -and three improved varieties -'Sol da Manhã', 'Nitrodente' and 'BR 106'. 'Carioca', a local variety, had the lowest activity. Between subpopulations, 'ND2', with low yielding and poorly P-efficient plants, presented higher root P-ase activity as compared to 'ND10', high yielding and highly P-efficient plants. In general, subpopulations presented lower P-ase activities as compared to varieties. Positive and/or negative correlations were obtained between P-ase activity and P-efficiency characteristics, specific for the genotypes, not allowing inference on a general and clear association between root-secreted phosphatase and dry matter production or P acquisition. Genotypic variability must be known and considered before using P-ase activity as an indicator of P nutritional status, or P tolerance, adaptation and efficiency under low P conditions.

AbstractThe inventory and categorization of an extensive coastal boulder assemblage originating from storm wave transport on the coastline of Fenoliga Island (Northern Adriatic Sea, southern Istria, Croatia) are presented and discussed herein. The study adopted the use of a commercial Uncrewed Aerial Vehicle (UAV) and Structure from Motion-MultiView Stereo (SfM-MVS) photogrammetry for the construction of a 3D model of the island. A Digital Elevation Model (DEM) and an orthomosaic were produced and employed for the mapping of the boulder assemblage in a GIS. In total, 592 boulders were identified and mapped. Using SfM-MVS-derived products allowed for the identified boulders to be categorized based on size classification. Amassed data relating to the boulder characteristics was inserted and stored in a GIS, including the results of a comparative assessment with historical Google Earth imagery which enabled the ‘quantification of boulder transport over a 9-year timeframe’. Field evidence indicates that boulders were created in-situ via the quarrying of bedrock strata by breaking waves causing increased water pressure within preexisting surfaces of weakness such as bedding planes and sub-vertical fractures. Once detached, the boulders were transported and deposited during storm wave events. Repeated storm events can further displace previously detached clasts.

Field symptoms of open spikelets in wheat were observed in specific cultivars and supposedly related to low B soils and differential B requirement among cultivars. This study aimed to evaluate the response of four wheat (Triticum aestivum L.) cultivars, IAC 24, IAC 60, IAC 287 and IAC 289, to increasing B concentrations in nutrient solution. The experiment was set up in a randomized complete block design, with four replicates and five B concentrations (0.0, 0.05, 0.2, 0.8 and 2.0 mg L-1), during 1997/1998, in a greenhouse. Plants were grown to maturity and evaluated for plant height, spike number and length, open spikelet number, grains per spike, plant parts dry matter, B, P, K, Ca and Mg leaf concentrations and total nutrient contents. The visual symptoms of B deficiency consisted of open spikelets, distorted spikes without grains. 'IAC 60' and 'IAC 287' had higher B efficiency, with the highest grain yields in lower B concentrations. The 'IAC 287' and 'IAC 24' were more tolerant to the highest B concentrations. 'IAC 24' required more B for grain production as compared to the other cultivars. The critical leaf B concentration for deficiency was 25 mg kg-1 of dry matter tissue for all cultivars, and for toxicity were: 44 to 45 mg kg-1 for 'IAC 60' and 'IAC 289'; 228 and 318 mg kg-1 for 'IAC 24' and 'IAC 287', respectively. Except for the highest B level in the nutrient solution, the leaf P, K, Ca and Mg concentrations and whole plant contents were in an adequate range in the plants and did not vary among cultivars.

Cellular alignment plays a pivotal role in several human tissues, including skeletal muscle, spinal cord and tendon. Various techniques have been developed to control cellular alignment using 3D biomaterials. However, the majority of 3D-aligned scaffolds require invasive surgery for implantation. In contrast, injectable hydrogels provide a non-invasive delivery method, gaining considerable attention for the treatment of diverse conditions, including osteochondral lesions, volumetric muscle loss, and traumatic brain injury.We engineered a biomimetic hydrogel with magnetic responsiveness by combining gellan gum, hyaluronic acid, collagen, and magnetic nanoparticles (MNPs). Collagen type I was paired with MNPs to form magnetic collagen bundles (MCollB), allowing the orientation control of these bundles within the hydrogel matrix through the application of a remote low-intensity magnetic field. This resulted in the creation of an anisotropic architecture. The hydrogel mechanical properties were comparable to those of human soft tissues, such as skeletal muscle, and proof of the aligned hydrogel concept was demonstrated.In vitro findings confirmed the absence of toxicity and pro-inflammatory effects. Notably, an increased fibroblast cell proliferation and pro-regenerative activation of macrophages were observed. The in-vivo study further validated the hydrogel biocompatibility and demonstrated the feasibility of injection with rapid in situ gelation. Consequently, this magnetically controlled injectable hydrogel exhibits significant promise as a minimally invasive, rapid gelling and effective treatment for regenerating various aligned human tissues.

Since December 2019, a new form of Severe Acute Respiratory Syndrome (SARS) has emerged worldwide, caused by SARS coronavirus 2 (SARS-CoV-2). This disease was called COVID-19 and was declared a pandemic by the World Health Organization in March 2020. Symptoms can vary from a common cold to severe pneumonia, hypoxemia, respiratory distress, and death. During this period of world stress, the medical and scientific community were able to acquire information and generate scientific data at unprecedented speed, to better understand the disease and facilitate vaccines and therapeutics development. Notably, bioinformatics tools were instrumental in decoding the viral genome and identifying critical targets for COVID-19 diagnosis and therapeutics. Through the integration of omics data, bioinformatics has also improved our understanding of disease pathogenesis and virus-host interactions, facilitating the development of targeted treatments and vaccines. Furthermore, molecular biology techniques have accelerated the design of sensitive diagnostic tests and the characterization of immune responses, paving the way for precision medicine approaches in treating COVID-19. Our analysis highlights the indispensable contributions of bioinformatics and molecular biology to the global effort against COVID-19. In this review, we aim to revise the COVID-19 features, diagnostic, prevention, treatment options, and how molecular biology, modern bioinformatic tools, and collaborations have helped combat this pandemic. An integrative literature review was performed, searching articles on several sites, including PUBMED and Google Scholar indexed in referenced databases, prioritizing articles from the last 3 years. The lessons learned from this COVID-19 pandemic will place the world in a much better position to respond to future pandemics.

The adoption of artificial intelligence tools can improve production efficiency in the agroindustry. Our objective was to perform the predictive modeling of carrot yield and quality. The crop was grown in two commercial areas during the summer season in Brazil. The root samples were taken at 200 points with a 30 × 30 m sampling grid at 82 and 116 days after sowing in both areas. The total fresh biomass, aerial part, and root biometry were quantified for previous crop harvesting to measure yield. The quality of the roots was assessed by sub-sampling three carrots by the concentration of total soluble solids (°Brix) and firmness in the laboratory. Vegetation indices were extracted from satellite imagery. The most important variables for the predictive models were selected by principal component analysis and submitted to the Artificial Neural Network (ANN), Random Forest (RF), and Multiple Linear Regression (MLR) algorithms. SAVI and NDVI indices stood out as predictors of crop yield, and the results from the ANN (R2 = 0.68) were superior to the RF (R2 = 0.67) and MLR (R2 = 0.61) models. Carrot quality cannot be modeled by the predictive models in this study; however, it should be explored in future research, including other crop variables.

In this work, we present a versatile, rapid, and low-cost manufacturing technique to develop bioceramic scaffolds that could enhance bone tissue regeneration via microwave preparation using a domestic microwave oven. The scaffolds were prepared by combining hydroxyapatite and water glass (sodium trisilicate solution), foamed by using a microwave oven, and then characterized by means of Scanning Electron Microscopy (SEM) with Energy Dispersive X-ray Analysis (EDX), mechanical properties, infrared spectroscopy (ATR-FTIR), and a density and stability test in water. Furthermore, in vitro tests were performed to verify the affinity of the scaffold for osteoclast cells. The morphology of the samples showed interconnected pores suitable for promoting tissue regeneration and vascularization, while specific mechanical properties were preserved. The physicochemical characterization and the in vitro tests presented promising results for bone regenerative applications. The scaffolds we obtained exhibited comparable properties to those fabricated using a laboratory microwave oven, including the ability to induce the formation of bone-like tissue in vitro.

Abstract Background The expression of aquaporin 4 (AQP4) and intermediate filament (IF) proteins is altered in malignant glioblastoma (GBM), yet the expression of the major IF-based cytolinker, plectin (PLEC), and its contribution to GBM migration and invasiveness, are unknown. Here, we assessed the contribution of plectin in affecting the distribution of plasmalemmal AQP4 aggregates, migratory properties, and regulation of cell volume in astrocytes. Methods In human GBM, the expression of glial fibrillary acidic protein (GFAP), AQP4 and PLEC transcripts was analyzed using publicly available datasets, and the colocalization of PLEC with AQP4 and with GFAP was determined by immunohistochemistry. We performed experiments on wild-type and plectin-deficient primary and immortalized mouse astrocytes, human astrocytes and permanent cell lines (U-251 MG and T98G) derived from a human malignant GBM. The expression of plectin isoforms in mouse astrocytes was assessed by quantitative real-time PCR. Transfection, immunolabeling and confocal microscopy were used to assess plectin-induced alterations in the distribution of the cytoskeleton, the influence of plectin and its isoforms on the abundance and size of plasmalemmal AQP4 aggregates, and the presence of plectin at the plasma membrane. The release of plectin from cells was measured by ELISA. The migration and dynamics of cell volume regulation of immortalized astrocytes were assessed by the wound-healing assay and calcein labeling, respectively. Results A positive correlation was found between plectin and AQP4 at the level of gene expression and protein localization in tumorous brain samples. Deficiency of plectin led to a decrease in the abundance and size of plasmalemmal AQP4 aggregates and altered distribution and bundling of the cytoskeleton. Astrocytes predominantly expressed P1c, P1e, and P1g plectin isoforms. The predominant plectin isoform associated with plasmalemmal AQP4 aggregates was P1c, which also affected the mobility of astrocytes most prominently. In the absence of plectin, the collective migration of astrocytes was impaired and the dynamics of cytoplasmic volume changes in peripheral cell regions decreased. Plectin’s abundance on the plasma membrane surface and its release from cells were increased in the GBM cell lines. Conclusions Plectin affects cellular properties that contribute to the pathology of GBM. The observed increase in both cell surface and released plectin levels represents a potential biomarker and therapeutic target in the diagnostics and treatment of GBMs.

Phase-contrast X-ray imaging is becoming increasingly considered since its first applications, which occurred almost 30 years ago. Particular emphasis was placed on studies that use this technique to investigate soft tissues, which cannot otherwise be investigated at a high resolution and in a three-dimensional manner, using conventional absorption-based settings. Indeed, its consistency and discrimination power in low absorbing samples, unified to being a not destructive analysis, are pushing interests on its utilization from researchers of different specializations, from botany, through zoology, to human physio-pathology research. In this regard, a challenging method for 3D imaging and quantitative analysis of collagenous tissues has spread in recent years: it is based on the unique characteristics of synchrotron radiation phase-contrast microTomography (PhC-microCT). In this review, the focus has been placed on the research based on the exploitation of synchrotron PhC-microCT for the investigation of collagenous tissue physio-pathologies from solely human samples. Collagen tissues’ elasto-mechanic role bonds it to the morphology of the site it is extracted from, which could weaken the results coming from animal experimentations. Encouraging outcomes proved this technique to be suitable to access and quantify human collagenous tissues and persuaded different researchers to approach it. A brief mention was also dedicated to the results obtained on collagenous tissues using new and promising high-resolution phase-contrast tomographic laboratory-based setups, which will certainly represent the real step forward in the diffusion of this relatively young imaging technique.

Abstract The field of wireless communication has witnessed tremendous advancements in the past few decades, leading to more pervasive and ubiquitous networks. Human bodies are continually exposed to electromagnetic radiation, but typically this does not impact the body as the radiation is non-ionizing and the waves carry low power. However, with progress in the sixth generation (6G) of wireless networks and the adoption of the spectrum above 100 GHz in the next few years, higher power radiation is needed to cover larger areas, exposing humans to stronger and more prolonged radiation. Also, water has a high absorption coefficient at these frequencies and could lead to thermal effects on the skin. Hence, there is a need to study the radiation effects on human tissues, specifically the photothermal effects. In this paper, we present a custom-built, multi-physics model to investigate electromagnetic wave propagation in human tissue and study its subsequent photothermal effects. The proposed finite-element model consists of two segments—the first one estimates the intensity distribution along the beam path, while the second calculates the increase in temperature due to the wave distribution inside the tissue. We determine the intensity variation in the tissue using the radiative transfer equation and compare the results with Monte Carlo analysis and existing analytical models. The intensity information is then utilized to predict the rise in temperature with a bio-heat transfer module, powered by Pennes’ bioheat equation. The model is parametric, and we perform a systematic photothermal analysis to recognize the crucial variables responsible for the temperature growth inside the tissue, particularly for terahertz and near-infrared optical frequencies. Our numerical model can serve as a benchmark for studying the high-frequency radiation effects on complex heterogeneous media such as human tissue.

The adoption of a no-till system presents difficulties in maintaining the potential crop yield under long-term conditions when compacted soil layers are observed in the field. The research objective was to assess the benefits of soil intervention in a traditional area of no-till system while considering the production parameters of corn (Zea mays L.) under tropical conditions. The area was installed considering 24 experimental units, including 6 treatments, a 3 × 2 scheme, and 4 repetitions (3 soil management systems and 2 crop hybrids). The soil scarification was carried out at two depths, 0.15 m and 0.30 m, to compare its influence on corn yield. The number of days of plants’ emergence, seed depth, longitudinal distribution of seedlings, phytotechnical parameters, and crop yield in the field were evaluated. The dataset was subjected to the F Test and Tukey test (p < 0.05) to compare the means by each attribute and treatment. The adopted hybrids, as well as soil-management systems in the experimental area, did not interfere with the quality of the sowing and quantitative parameters of the crop (CV = 15% for normal distribution on plants’ spacing within rows). So, there is no need to scarify soil with the aim of increasing corn yield under the evaluated field conditions.

Mitral regurgitation (MR) is a broadly diffuse valvular heart disease (VHD) with a significant impact on the healthcare system and patient prognosis. Transcatheter mitral valve interventions (TMVI) are now well-established techniques included in the therapeutic armamentarium for managing patients with mitral regurgitation, either primary or functional MR. Even if the guidelines give indications regarding the correct management of this VHD, the wide heterogeneity of patients’ clinical backgrounds and valvular and heart anatomies make each patient a unique case, in which the appropriate device’s selection requires a multimodal imaging evaluation and a multidisciplinary discussion. Proper pre-procedural evaluation plays a pivotal role in judging the feasibility of TMVI, while a cooperative work between imagers and interventionalist is also crucial for procedural success. This manuscript aims to provide an exhaustive overview of the main parameters that need to be evaluated for appropriate device selection, pre-procedural planning, intra-procedural guidance and post-operative assessment in the setting of TMVI. In addition, it tries to give some insights about future perspectives for structural cardiovascular imaging.

Tidal notches, long regarded as reliable indicators of mean sea level, have been extensively studied along carbonate coasts in the central Mediterranean Sea. Previous studies revealed a correlation between the genesis of tidal notches and tidal range, lithology, cliff foot depth, and wave energy. In the 2020 Geoswim campaigns at Lampedusa, the southernmost island of the Pelagie archipelago (Italy), and in Gozo Island (Malta), ‘anomalous’ tidal notches were identified. Unlike normal notches observed elsewhere, those in Lampedusa’s southern bays exhibited a particular behaviour—constantly deepening in the inner part of the bays, reaching a maximum depth of approximately 30 cm below sea level and narrowing inwards. Similar phenomena were previously observed near Marseille (France). As confirmed by the literature, all these areas are tectonically stable. Time-lapse images, alongside measurements of morphometric parameters, were collected during the survey. Our hypothesis indicates that a combination of marine factors influenced by local marine conditions driven by the local morphology of the small bays exposed to southern quadrants contribute to the formation of these unique landforms. The latter manifests higher lowering erosion rates slightly below the mean sea level in sheltered areas, challenging conventional notions about tidal notch formation.
