Luliconazole (LCZ) is a topical imidazole antifungal agent with broad-spectrum activity. However, LCZ encounters challenges such as low aqueous solubility, skin retention, and penetration, which reduce its dermal bioavailability and hinder its efficacy in drug delivery. The aim of the present study was to formulate, characterize, and evaluate the in vitro antifungal efficacy of luliconazole-loaded nanostructured lipid carriers (LCZ-NLCs) against a panel of resistant fungal strains. The LCZ-NLCs were synthesized using a modified emulsification-solvent evaporation technique. Characterization involved assessing parameters such as poly-dispersity index (PDI), zeta potential, encapsulation efficiency (EE %), Field Emission Scanning Electron Microscopy (FESEM), Differential Scanning Calorimetry (DSC) analysis, and Attenuated Total Reflectance Fourier Transform Infrared (ATR-FTIR). Furthermore, in vitro drug release experiments, analysis of release kinetics, cytotoxicity assessments, and in vitro antifungal susceptibility tests were performed as part of the study. The findings indicated that LCZ-NLCs displayed nanoscale dimensions, uniform dispersion, and a favorable zeta potential. The encapsulation efficiency of LCZ in NLCs was approximately 90%. FESEM analysis revealed spherical nanoparticles with consistent shape. ATR-FTIR analysis indicated no chemical interaction between LCZ and excipients. In vitro drug release experiments demonstrated that LCZ-NLCs notably improved the drug's dissolution rate. The stability testing confirmed consistent colloidal nanometer ranges in the LCZ-NLCs samples. Additionally, cytotoxicity tests revealed no toxicity within the tested concentration. Moreover, in vitro antifungal susceptibility tests demonstrated potent antifungal activity of LCZ-NLCs against the tested resistant fungal isolates. The study findings suggest that the LCZ-NLCs formulation developed in this research could be a promising topical treatment for superficial fungal infections, especially in cases of resistant infections. Varronia curassavica Jacq. is an aromatic species appertaining to the Boraginaceae family and has been mentioned for its numerous traditional uses and pharmacological properties, especially its antimicrobial and anti-inflammatory effects. The aim of the present study was to investigate the phytochemical profile and antifungal activities of the essential oils of V. curassavica, in addition to analyzing the ADMET properties of the majority components. The GC-MS analysis of V. curassavica essential oil (EOVC) comprised 97.36% of total composition, with α-pinene, β-caryophyllene, and bicyclogermacrene (44.46%, 22.87%, and 13.05%, respectively) as the main constituents among other minor/trace constituents. The antifungal activity of EOVC was evaluated against three Candida species and was observed with IC50 > 200 μg/mL. Remarkably, the combination of EOVC with fluconazole significantly reduced the IC50 required for the drug to inhibit C. tropicalis (0.003 μg/mL), C. albicans (0.7996 μg/mL), and C. krusei (17.73 μg/mL). In addition, ADME/Tox studies using α-pinene revealed that the compound poses no toxicity threats but requires caution due to its high permeability to the blood–brain barrier (BBB). Overall, the obtained results suggest that Varronia curassavica essential oil is a potentially good antifungal agent for combating fungal resistance.

The hop variety Saaz is well known over the world and is usually used for brewing of lager beers. Recently, the new related varieties Saaz Late, Brilliant, Comfort, and Shine were registered. Information about these varieties is splintered and often available only in the Czech language in regional journals. This review (i) summarizes previously published data (breeding history, genetic data, basic parameters such as yield, sensory profile, concentrations of key technologically important hop compounds), (ii) presents long-term data (2004–2021), and (iii) shows similarities/differences among these varieties. All Saaz varieties are typically fine aroma hops with a relatively low content of alpha bitter acids ranging from 5 to 7 wt%, cohumulone amounts lower than 30% rel., and hop oil content of about 1.0 wt%. Even though the new varieties have no identical chemical parameters to the original Saaz, they can substitute this established standard as well. Furthermore, the varieties Saaz Comfort and Saaz Shine show high resistance to Pseudoperonospora humuli as well as very good tolerance to drought. [ABSTRACT FROM AUTHOR]

Fungal resistance is a public health concern due to the limited availability of antifungal resources and the complexities associated with treating persistent fungal infections. Azoles are thus far the primary line of defense against fungi. Specifically, azoles inhibit the conversion of lanosterol to ergosterol, producing defective sterols and impairing fluidity in fungal plasmatic membranes. Studies on azole resistance have emphasized specific point mutations in CYP51/ERG11 proteins linked to resistance. Although very insightful, the traditional approach to studying azole resistance is time-consuming and prone to errors during meticulous alignment evaluation. It relies on a reference-based method using a specific protein sequence obtained from a wild-type (WT) phenotype. Therefore, this study introduces a machine learning (ML)-based approach utilizing molecular descriptors representing the physiochemical attributes of CYP51/ERG11 protein isoforms. This approach aims to unravel hidden patterns associated with azole resistance. The results highlight that descriptors related to amino acid composition and their combination of hydrophobicity and hydrophilicity effectively explain the slight differences between the resistant non-wild-type (NWT) and WT (nonresistant) protein sequences. This study underscores the potential of ML to unravel nuanced patterns in CYP51/ERG11 sequences, providing valuable molecular signatures that could inform future endeavors in drug development and computational screening of resistant and nonresistant fungal lineages. Key message: Thchit42 constitutive expression for fungal resistance showed synchronisation with leaf augmentation and transcriptome analysis revealed the Longifolia and Zinc finger RICESLEEPER gene is responsible for plant growth and development. Pelargonium graveolens essential oil possesses significant attributes, known for perfumery and aromatherapy. However, optimal yield and propagation are predominantly hindered by biotic stress. All biotechnological approaches have yet to prove effective in addressing fungal resistance. The current study developed transgenic geranium bridging molecular mechanism of fungal resistance and plant growth by introducing cassette 35S::Thchit42. Furthermore, 120 independently putative transformed explants were regenerated on kanamycin fortified medium. Primarily transgenic lines were demonstrated peak pathogenicity and antifungal activity against formidable Colletotrichum gloeosporioides and Fusarium oxysporum. Additionally, phenotypic analysis revealed ~ 2fold increase in leaf size and ~ 2.1fold enhanced oil content. To elucidate the molecular mechanisms for genotypic cause, de novo transcriptional profiles were analyzed to indicate that the auxin-regulated longifolia gene is accountable for augmentation in leaf size, and zinc finger (ZF) RICESLEEPER attributes growth upregulation. Collectively, data provides valuable insights into unravelling the mechanism of Thchit42-mediated crosstalk between morphological and chemical alteration in transgenic plants. Abstract Pecan scab is a devastating disease that causes damage to pecan (Carya illinoinensis (Wangenh.) K. Koch) fruit and leaves. The disease is caused by the fungus Venturia effusa (G. Winter) and the main management practice for controlling the disease is by application of fungicides at 2-to-3-week intervals throughout the growing season. Besides disease-related yield loss, application of fungicides can result in considerable cost and increases the likelihood of fungicide resistance developing in the pathogen. Resistant cultivars are available for pecan growers; although, in several cases resistance has been overcome as the pathogen adapts to infect resistant hosts. Despite the importance of host resistance in scab management, there is little information regarding the molecular basis of genetic resistance to pecan scab. The purpose of this study was to elucidate mechanisms of natural pecan scab resistance by analyzing transcripts that are differentially expressed in pecan leaf samples from scab resistant and susceptible trees. The leaf samples were collected from trees in a provenance collection orchard that represents the natural range of pecan in the US and Mexico. Trees in the orchard have been exposed to natural scab infections since planting in 1989, and scab ratings were collected over three seasons. Based on this data, ten susceptible trees and ten resistant trees were selected for analysis. RNA-seq data was collected and analyzed for diseased and non-diseased parts of susceptible trees as well as for resistant trees. A total of 313 genes were found to be differentially expressed when comparing resistant and susceptible trees without disease. For susceptible samples showing scab symptoms, 1,454 genes were identified as differentially expressed compared to non-diseased susceptible samples. Many genes involved in pathogen recognition, defense responses, and signal transduction were up-regulated in diseased samples of susceptible trees, whereas differentially expressed genes in pecan scab resistant samples were generally down-regulated compared to non-diseased susceptible samples. Our results provide the first account of candidate genes involved in resistance/susceptibility to pecan scab under natural conditions in a pecan orchard. This information can be used to aid pecan breeding programs and development of biotechnology-based approaches for generating pecan cultivars with more durable scab resistance.

There is increasing evidence of clinically resistant cutaneous fungal infections. The use of combination oral antifungals is described in adults but not in paediatric patients. We present seven paediatric cases of clinically resistant fungal infections treated successfully with combination oral antifungal therapy after inadequate response to a single agent. The mechanical properties and termite and decay resistance performance of wood-based panel used in wooden houses were evaluated. The physical and mechanical properties, along with the resistance levels, of commonly used wood-based panels, including oriented strand board, structural particleboard, and particleboard for use in interior, were compared. The structural particleboard complied with the physical, mechanical, and formaldehyde emission standards of the International Organization for Standardization and Japanese Industrial Standard, surpassing the requirements for oriented strand board. The structural particleboard exhibited excellent water resistance and a consistent performance. Decay tests classified the particleboard and structural particleboard as "Resistant," with mass losses of 7.82 to 12.72% (white rot) and 14.69 to 16.55% (brown rot). Pine (Pinus densiflora) and oriented strand board exhibited no decay resistance, with mass losses exceeding 45%. The particleboard and structural particleboard demonstrated superior termite resistance, resulting in 100% termite mortality in three days without chemical treatment. The structural particleboard exhibited excellent water, decay, and termite resistance, which can be an advantage in wooden-house construction in terms of maintenance.

Vulvovaginitis with Candida spp. is the most common infection in women and the rate is increased during pregnancy. Antifungal prescription in pregnant women continues to present challenges and the decision must balance the risk of fetal toxicity with the benefits to the fetus and mother. Starting from the idea that clotrimazole is the most recommended antifungal in candidal vaginitis in pregnancy, we tested the sensitivity of different species of Candida spp. to other azoles, polyenes, and antimetabolites. This retrospective study (January to June 2019) assessed 663 pregnant women hospitalized for various pregnancy-related symptoms in which samples of phage secretion were taken. The laboratory results confirmed 21% of cases, indicating 140 positive mycologic samples. In this study, vaginal candidiasis was mostly related to the first trimester of pregnancy (53.57%,) and less related in the last trimester (17.14%). Candida albicans was the most frequent isolated strain in this study, accounting for 118 cases, followed by 16 strains of Candida glabrata and 6 cases of Candida krusei. The highest sensitivity for C. albicans was found in azoles, mostly in miconazole (93.2%), while C. krusei was completely resistant to polyene with low sensitivity in antimetabolites and even in some azoles, such as fluconazole. In our study, higher resistance rates to flucytosine were found, with C. glabrata and C. krusei exhibiting greater resistance than C. albicans.

Adherence to antifungals is poor in high endemic regions where antifungal resistance is high. Poor readability of prescription/over-the-counter (OTC) antifungals may contribute to poor adherence, due to the patient not fully understanding the purpose, importance, and dosage of their antifungal medicine. As there are no reports on the readability of antifungals, this study examined the readability of patient-facing antifungal information. Antifungals (n  = 16; five classes [allylamines, azoles, echinocandins, polyenes, and others—flucytosine and griseofulvin]) were selected. Readability of four sources of information, (i) summary of product characteristics, (ii) patient information leaflets (PILs), (iii) OTC patient information, and (iv) patient web-based information, was calculated using Readable software, to obtain readability scores [(i) Flesch Reading Ease [FRE], (ii) Flesch–Kinkaid Grade Level [FKGL], (iii) Gunning Fog Index, and (iv) Simple Measure of Gobbledygook (SMOG) Index) and text metrics [word count, sentence count, words/sentence, and syllables/word]. PILs, web-based resources, and OTC patient information had good readability (FRE mean ± sd = 52.8 ± 6.7, 58.6 ± 6.9, and 57.3 ± 7.4, respectively), just falling short of the ≥ 60 target. For FKGL (target ≤ 8.0), PILs, web-based resources, and OTC patient information also had good readability (mean ± sd = 8.5 ± 1.0, 7.2 ± 0.86, and 7.8 ± 0.1, respectively). Improved readability scores observed correlate with reduced words, words/sentence and syllables/word. Improving readability may lead to improved patient health literacy. Healthcare professionals, academics, and publishers preparing written materials regarding antifungals for the lay/patient community are encouraged to employ readability calculators to check the readability of their work, so that the final material is within recommended readability reference parameters, to support the health literacy of their patients/readers. Aerial roots of the Banyan tree were utilized to extract completely biodegradable natural fibers with inherent antimicrobial properties which can be used as potential raw material for health and hygiene products. Fiber extraction conditions were optimized for biological, chemical and enzymatic treatments. Bleaching and softening treatments were given for improving color, further individualization and enhancing handle of extracted fibers. The change in chemical composition due to alkali treatment was determined using FTIR. SEM was used to compare the change in surface morphology of fibers with the use of different treatment. Various physical and biological properties of the fibers were tested and compared with other lignocellulosic fibers. It was observed that the physical properties of fibers extracted from aerial roots of banyan tree were comparable to other natural cellulosic fibers like sisal, coir, jute. The fibers have a combination of properties such as low strength, moderate elongation along with good moisture regain and retention. Additionally, they exhibit excellent resistance against bacterial and fungal growth, which is quite rare in case of natural cellulosic fibers, making them suitable for potential usage, especially in hygiene products. Other applications in terms of fiber composites with suitable materials can also be envisaged.

Disseminated fusariosis is treated with amphotericin B and voriconazole. To determine adequate therapy, the minimal inhibitory concentration (MIC) is used. However, MIC analysis is based on visual observation and requires a long period of fungal incubation. The measure of the minimal profile change concentration (MPCC) using MALDI-TOF MS is a quick spectral method that has presented good results in determining the antimicrobial resistance of yeasts. However, there is a lack of information on filamentous fungi. In the present work, 13 Fusarium spp. clinical isolates and two reference strains were used. MIC was obtained according to the M38-A2 protocol of the Clinical Laboratory Standards Institute, while MPPC was obtained following the initial steps of the M38-A2 protocol. Both Biotyper and the Rstudio environment were used to analyze mass spectra. For some fungal strains, the data obtained from the software MALDI Biotyper Compass 4.1 led to fuzzy heatmaps resulting in difficult interpretation, while heatmaps obtained using Rstudio tools generated better MPCC resolutions. Herein, 86.6% of the AMB MPCC values were highly correlated with the gold-standard AMB MIC. MALDI-TOF MS is a prominent tool used to determine MPCCs quicker, cost-effectively, and more accurately for Fusarium spp. strains. The morbidity and mortality of invasive fungal infections are rising gradually. In recent years, fungi have quietly evolved stronger defense capabilities and increased resistance to antibiotics, posing huge challenges to maintaining physical health. Therefore, developing new drugs and strategies to combat these invasive fungi is crucial. There are a large number of microorganisms in the intestinal tract of mammals, collectively referred to as intestinal microbiota. At the same time, these native microorganisms co-evolve with their hosts in symbiotic relationship. Recent researches have shown that some probiotics and intestinal symbiotic bacteria can inhibit the invasion and colonization of fungi. In this paper, we review the mechanism of some intestinal bacteria affecting the growth and invasion of fungi by targeting the virulence factors, quorum sensing system, secreting active metabolites or regulating the host anti-fungal immune response, so as to provide new strategies for resisting invasive fungal infection. Candida albicans, an opportunistic yeast, is the most common cause of fungal infection. In the past decade, there has been an increase in C. albicans resistance to existing antifungal drugs, which has necessitated the development of new antifungal agents. In the present study, screening 60 compounds from the JUNIA chemical library enabled us to explore an additional 11 hybrid compounds that contain pyrrolidinone rings and hydrazine moieties for their potential antifungal activities. This chemical series was identified with fair to excellent antifungal activities. Among this series, three molecules (Hyd.H, Hyd.OCH3, and Hyd.Cl) significantly reduced C. albicans viability, with rapid fungicidal activity. In addition, these three compounds exhibited significant antifungal activity against clinically isolated fluconazole- or caspofungin-resistant C. albicans strains. Hyd.H, Hyd.OCH3, and Hyd.Cl did not show any cytotoxicity against human cancer cell lines up to a concentration of 50 µg/mL and decreased Candida biofilm formation, with a significant reduction of 60% biofilm formation with Hyd.OCH3. In an infection model of Caenorhabditis elegans with C. albicans, hydrazine-based compounds significantly reduced nematode mortality. Overall, fungicidal activity was observed for Hyd.H, Hyd.OCH3, and Hyd.Cl against C. albicans, and these compounds protected C. elegans from C. albicans infection. [ABSTRACT FROM AUTHOR]

Among the most important transcription factors in plants, the v-myb avian myeloblastosis viral oncogene homolog (MYB) regulates the expression network of response genes under stresses such as fungal infection. In China, the canker disease Valsa mali threatens the survival of Malus sieversii, an ancestor of cultivated apples. Using the M. sieversii genome, we identified 457 MsMYB and 128 R2R3-MsMYB genes that were randomly distributed across 17 chromosomes. Based on protein sequence and structure, the R2R3-MsMYB genes were phylogenetically divided into 29 categories, and 26 conserved motifs were identified. We further predicted cis-elements in the 2000-kb promoter region of R2R3-MsMYBs based on the genome. Transcriptome analysis of M. sieversii under V. mali infection showed that 27 R2R3-MsMYBs were significantly differentially expressed, indicating their key role in the response to V. mali infection. Using transient transformation, MsMYB14, MsMYB24, MsMYB39, MsMYB78, and MsMYB108, which were strongly induced by V. mali infection, were functionally identified. Among the five MsMYBs, MsMYB14 and MsMYB78 were both important in enhancing resistance to diseases, whereas MsMYB24 inhibited resistance. Based on the results of this study, we gained a better understanding of the MsMYB transcription factor family and laid the foundation for a future research program on disease prevention strategies in M. sieversii. Many transcription factors and morphogenic genes were used to increase the embryogenic calli formation and regeneration efficiency in transformed plants. The myoblastosis transcription factor is the largest family and plays an important role in plant-specific processes like metabolism, development, resistance against biotic and abiotic stresses. The Arabidopsis MYB115 gene belongs to the R2R3 myoblastosis transcription factor and it is well conserved in plants. In order to evaluate the diverse function of R2R3 MYB transcription factor, rice transformation was performed using 35S promoter driven AtMYB115 gene. Detailed Southern analysis confirmed the transgene integration and copy numbers were determined. The calli from homozygous single copy line, MYB115-8-9 showed early regeneration of plantlets compared to the control plants. On the other, they didn't reveal any difference in the embryogenesis. As AtMYB115 is also involved in benzyloxy glucosinolate pathway, the resistance to sheath blight disease was evaluated. However, the plants did not show any resistance against the sheath blight disease. Interestingly, the overexpression of AtMYB115 in rice led to the dwarf phenotype in both T0 and T2 generation. Apart from this dwarf phenotype adventitious root formation was also observed in T2 homozygous plants. Fungi are becoming increasingly resistant, especially the new strains. Therefore, this work developed nanoemulsions (NE) containing micafungin (MICA), in order to improve its action against infections caused by Candida auris. The NEs were composed of the surfactants polyoxyethylene (20) cetyl ether (Brij 58®)/soy phosphatidylcholine at 10%, sunflower oil/cholesterol at 10%, and 80% PBS. The NEs were characterized by Dynamic Light Scattering (DLS). For the microbiological in vitro evaluation the determination of the minimum inhibitory concentration (MIC), ergosterol/sorbitol, time kill and biofilms tests were performed. Additionally, the antifungal activity was also evaluated in a Galleria mellonella model. The same model was used in order to evaluate acute toxicity. The NE showed a size of ∼ 42.12 nm, a polydispersion index (PDI) of 0.289, and a zeta potential (ZP) of −3.86 mV. NEM had an average size of 41.29 nm, a PDI of 0.259, and a ZP of −4.71 mV. Finally, both nanoemulsions showed good stability in a storage period of 3 months. Although NEM did not show activity in planktonic cells, it exhibited action against biofilm and in the in vivo infection model. In the alternative in vivo model assay, it was possible to observe that both, NEM and free MICA at 0.2 mg/l, was effective against the infection, being that NEM presented a better action. Finally, NEM and free MICA showed no acute toxicity up to 4 mg/l. NEM showed the best activities in in vitro in mature antibiofilm and in alternative in vivo models in G. mellonella. Although, NEs showed to be attractive for MICA transport in the treatment of infections caused by C. auris in vitro and in vivo studies with G. mellonella , further studies should be carried out, in mice, for example. This study developed eco-friendly protective materials for wood preservation, focusing on enhancing fungal decay resistance, insect damage prevention, and improving physical and mechanical properties. The research examined the penetration of cellulose nanocrystals (CNCs) and their functionalized compounds into wood tissue, evaluating their impact on poplar wood. Methodology included optimizing CNC production through acid hydrolysis, testing various temperature and time combinations. CNCs were then functionalized with Poly(dimethylsiloxane)-bis(3-aminopropyl) terminated (PDMS-NH), copper hydroxide, zinc oxide, and silver nanoparticles. Characterization techniques such as AFM, TEM, ESEM, XRD, FTIR, and μ-Raman spectroscopy analyzed CNCs and their derivatives. Wood samples were impregnated with CNCs and functionalized CNCs using pressure and vacuum treatments, then tested for weight gain, mass loss against white rot (Trametes versicolor) and brown rot (Coniophora puteana) fungi, resistance to insect attack (Trichoferus holosericeus), leaching resistance, and mechanical properties. Key findings included successful optimization of CNC production and functionalization, improved resistance against decay fungi (especially with CNC3/Cu treatment), elevating durability classification from "non-durable" to "low durability". However, treatments showed limited effectiveness against insect infestation. Leaching resistance varied among treatments, with CNC3/PDMS-NH performing best. Mechanical properties, particularly modulus of elasticity, improved significantly with CNC3 impregnation, especially in less degraded wood samples. The study contributes to eco-friendly wood protection systems development, demonstrating functionalized CNCs' potential to enhance wood mass loss and mechanical properties. Patients admitted to the intensive care unit or immunocompromised patients frequently develop fungal infections. Candida albicans (C. albicans) is the pathogenic fungus responsible for most invasive fungal infections. Fluconazole (FLC) is the most widely used antifungal agent in clinical practice due to its effectiveness and low cost. However, due to its widespread use, C. albicans is becoming increasingly resistant to FLC. This increase in resistance poses a significant challenge for antifungal treatments. Various attempts have been made to reverse the resistance of C. albicans to FLC, including combinations with natural compounds with low toxicity, low cost, and high antifungal efficacy. Furthermore, various natural compounds have in vitro and in vivo synergistic effects with FLC against C. albicans , particularly when treating resistant isolates. This review summarises natural compounds that, when combined with FLC, exhibit synergistic effects against C. albicans. These combinations were identified through a comprehensive search of PubMed, Web of Science, and Embase databases until March 2023. Forty-eight natural antifungal compounds with potential clinical applications were identified. The most common mechanisms underlying their synergistic effects include inhibition of drug efflux, induction of mitochondrial dysfunction, and accumulation of reactive oxygen species (ROS). onservation of historic timber structures is of great importance for cultural inheritance and community identity promotion. However, most of the current methods available for ancient architecture protection significantly affect their original appearance and aesthetic value and finding wood elements that are similar to the ones in existing historic timber structures is not easy. Here we report a simple and effective method to archaize wood, Castanopsis sclerophylla, by ferric chloride (FeCl3) treatment without significantly affecting its mechanical properties and durability. The lightness and the color indexes of treated wood are similar to the ancient wood sample. The mechanical properties of FeCl3 treated wood are not statistically different from the control. Our durability testing results indicated that FeCl3 treated wood has good decay resistance against Irpex lacteus and Trametes versicolor with a mass loss of less than 10 %. This study provides a convenient method for the restoration and protection of ancient buildings.

Aerial roots of the Banyan tree were utilized to extract completely biodegradable natural fibers with inherent antimicrobial properties which can be used as potential raw material for health and hygiene products. Fiber extraction conditions were optimized for biological, chemical and enzymatic treatments. Bleaching and softening treatments were given for improving color, further individualization and enhancing handle of extracted fibers. The change in chemical composition due to alkali treatment was determined using FTIR. SEM was used to compare the change in surface morphology of fibers with the use of different treatment. Various physical and biological properties of the fibers were tested and compared with other lignocellulosic fibers. It was observed that the physical properties of fibers extracted from aerial roots of banyan tree were comparable to other natural cellulosic fibers like sisal, coir, jute. The fibers have a combination of properties such as low strength, moderate elongation along with good moisture regain and retention. Additionally, they exhibit excellent resistance against bacterial and fungal growth, which is quite rare in case of natural cellulosic fibers, making them suitable for potential usage, especially in hygiene products. Anthracnose, caused by the fungal pathogen Colletotrichum lindemuthianum (Sacc. & Magnus) Lams.-Scrib., is one of the most devastating diseases in dry bean (Phaseolus vulgaris L.) with seed yield losses up to 100%. Most anthracnose resistance genes thus far identified behave in a dominant manner and were identified by seedling screening. The Middle American Diversity Panel (MDP; n=266) was screened with a modified greenhouse screening method to evaluate the response to anthracnose race 73. Thirty MDP genotypes exhibited resistance to the race of which 16 genotypes were not known to contain anthracnose resistance genes to race 73. GWAS with ~93,000 SNP markers identified four genomic regions, two each on Pv01 and Pv10, associated race 73 resistance. A likelihood-ratio-based R2 analysis indicated the peak four SNP markers are responsible for 26% of the observed phenotypic variation, where one SNP, S10_072250, explains 23% of the total variation. SNP S10_072250 is associated with a new region of anthracnose resistance and is in an intron of a ZPR1-like gene. Further greenhouse testing of the 16 resistant lines without previously known resistance to race 73 revealed various levels of resistance under various levels of disease pressure. Disease resistance was further characterized in the field using four representative genotypes. GTS-900 and Remington exhibited field resistance while Merlot and Maverick were susceptible. Field testing with two different fungicide regimes revealed the resistant genotypes had no significant disease differences. The results suggest resistance to anthracnose may differ at various growth stages and that breeders have been selecting for major genes at early seedling stages while ignoring the effect of alternative genes that may be active at later stages. The newly identified resistant lines may be related to Age Related Resistance (ARR) and could be exploited as parental sources of anthracnose resistance in addition to already known major genes. The physical localization of the multiple regions of resistance confirms the presence of two clusters of disease resistance genes on Pv01 and identifies two new regions of anthracnose resistance on Pv10 possibly associated with ARR. We isolated, purified, and structurally elucidated antifungal secondary metabolites from red soil microbes and encapsulated them into chitosan (CS)-based nanoemulsion (NE) gel (NEG).Methods: Three compounds were isolated and purified of which only one compound (Pure 2) showed potent antifungal activity (MFC: 8– 132 μg/mL), which was also significantly (P< 0.05) more efficient than fluconazole (MFC: 32– 132 μg/mL). Pure 2 was structurally elucidated using 1D- and 2D-NMR before its incorporation into NEG. The formulations were prepared by high-speed homogenization technique. Physicochemical and pharmacological characterizations of formulations (ie, droplet size, PDI, zeta potential, drug content, viscosity, SEM, FTIR, spreadability, in vitro drug release, ex vivo permeation, in vitro antifungal and in vivo antifungal activities) were assessed.Results: NMR analyses identified the compound as a derivative of phthalic acid ester (PAE). The optimized formulations displayed a droplet size < 100 nm, -ve zeta potential, and PDI < 0.45. The drug content was within the official limit of pharmacopeia (ie, 100± 10%). Insignificant changes (P> 0.05) in the viscosity of the formulations stored were observed. The morphology of the formulations indicated mesh-like structure. The FTIR study indicated that there were no interactions between the drug and other ingredients of the formulations. Optimum spreadability was observed in all formulations. NEG released 75.3± 1.12% of Pure 2 after 12 hrs while NE released 85.33± 1.88% of the compound. The skin permeation of F2 (71.15± 1.28%) was significantly different (P< 0.05) from F3 (81.80± 1.91%) in rabbits. Violacein is an important natural antimicrobial pigment that is mainly produced by Chromobacterium violaceum and Janthinobacterium lividum. It presents a significant range of effects against phytopathogenic and human fungi, besides being featured as having low toxicity, and by its important ecological role in protecting amphibian species and applications in dyed medical fabric. The hypothesis about violacein's action mechanisms against mucormycosis (Rhizopus arrhizus) and candidiasis (Candida auris) is herein discussed based on data available in the scientific literature. Significance and Impact of the Study: Infections caused by fungi in the antimicrobial‐resistance Era are of great concern since many of them are comorbidities related to SARS‐CoV‐2 infection. This mini‐review is a critical analysis of violacein use, which is a natural bacterial secondary metabolite product featured by a great versatility of biological activity, a fact that turns it into a likely new strategy against phytopathogenic and human fungal infections produced by different micro‐organism species. The current manuscript aims to show violacein's potential as a new strategy against mucormycosis (caused by Rhizopus arrhizus), an opportunist disease that may affect the respiratory tract and the skin, and candidiasis (caused by Candida auris), a disease that has different presentations, from local lesions in the skin and mucosae, to a severe disseminated form. These two important infections must be taken into consideration in severe COVID‐19 cases. Summary: Histone lysine methylations (HLMs) are implicated in control of gene expression in different eukaryotes. However, the role of HLMs in regulating desirable crop traits and the enzymes involved in these modifications are poorly understood.We studied the functions of tomato histone H3 lysine methyltransferases SET Domain Group 33 (SDG33) and SDG34 in biotic and abiotic stress responses. SDG33 and SDG34 gene edited mutants were altered in H3K36 and H3K4 methylations, and expression of genes involved in diverse processes and responses to biotic and abiotic stimuli.The double but not the single mutants show resistance to the fungal pathogen Botrytis cinerea. Interestingly, single mutants were tolerant to drought and the double mutant showed superior tolerance and plant growth consistent with independent and additive functions. Mutants maintained higher water status during drought and improved recovery and survival after lapse of drought.Notably, diminution of H3K4 and H3K36 trimethylation and expression of negative regulators in challenged plants contributes to stress tolerance of the mutants. Mutations in SDG33 and SDG34 are likely to remove predisposition to biotic and abiotic stress by disrupting permissive transcriptional context promoting expression of negative regulatory factors. The change in the physical and mechanical characteristics of thiokol sealants under the influence of micromycetes isolated in the tropical climate of the Republic of Cuba and micromycetes used as test cultures according to GOST (State Standard) 9.049–91 was studied. • The expression of PtoASMT is induced by different abiotic stresses and phytohormones. • PtoASMT has diurnal expression rhythm, but it is not involved in the seasonal growth of poplar. • Overexpression of PtoASMT can promote the accumulation of MT and enhance the drought tolerance of poplar. • Overexpression of PtoASMT enhances the resistance of poplars to fungal diseases by raising the expression of PR genes. In recent years, the increase in extreme climates, such as persistent high temperatures and drought, has adversely affected the growth and development of fast-growing trees. Melatonin (MT) plays an important role in plant responses to biotic and abiotic stresses, yet there is a lack of research on the specific role of limiting enzyme genes for MT biosynthesis in fast-growing woody plants. In this study, we investigated the function of PtoASMT , a key rate-limiting enzyme encoding gene for MT biosynthesis, which can be induced by drought, salt, and the phytohormones ABA, SA and JA. Our results show that: (1) PtoASMT was widely expressed in all tissues of poplar, but was highly expressed in petioles, moderately expressed in roots, stems, shoots and young leaves, exhibiting a typical diurnal expression rhythm in leaves, with the encoded protein localized on chloroplasts; (2) the content of MT was significantly promoted in overexpressing PtoASMT transgenic poplar plants, but there were no obvious differences in their growth and development; (3) overexpressing PtoASMT plants exhibited stronger drought tolerance, accumulating less reactive oxygen species (ROS) under drought stress relative to wild-type plants, whereas knockout PtoASMT plants were more sensitive and accumulated more ROS; (4) overexpressing PtoASMT plants were more resistant to fungi Dothiorella gregaria than WT plants, while knockout plants showed higher sensitivity; meanwhile, the expression of disease resistance-related genes (PRs and JAZ10) was significantly altered. We conclude that PtoASMT enhances the resistance of poplar to drought and Dothiorella gregaria by mediating MT biosynthesis in poplar. Disseminated fusariosis is treated with amphotericin B and voriconazole. To determine adequate therapy, the minimal inhibitory concentration (MIC) is used. However, MIC analysis is based on visual observation and requires a long period of fungal incubation. The measure of the minimal profile change concentration (MPCC) using MALDI-TOF MS is a quick spectral method that has presented good results in determining the antimicrobial resistance of yeasts. However, there is a lack of information on filamentous fungi. In the present work, 13 Fusarium spp. clinical isolates and two reference strains were used. MIC was obtained according to the M38-A2 protocol of the Clinical Laboratory Standards Institute, while MPPC was obtained following the initial steps of the M38-A2 protocol. Both Biotyper and the Rstudio environment were used to analyze mass spectra. For some fungal strains, the data obtained from the software MALDI Biotyper Compass 4.1 led to fuzzy heatmaps resulting in difficult interpretation, while heatmaps obtained using Rstudio tools generated better MPCC resolutions. Herein, 86.6% of the AMB MPCC values were highly correlated with the gold-standard AMB MIC. MALDI-TOF MS is a prominent tool used to determine MPCCs quicker, cost-effectively, and more accurately for Fusarium spp. strains. However, better statistical analyses could help measure the technique’s limit detection.

Candida albicans, an opportunistic yeast, is the most common cause of fungal infection. In the past decade, there has been an increase in C. albicans resistance to existing antifungal drugs, which has necessitated the development of new antifungal agents. In the present study, screening 60 compounds from the JUNIA chemical library enabled us to explore an additional 11 hybrid compounds that contain pyrrolidinone rings and hydrazine moieties for their potential antifungal activities. This chemical series was identified with fair to excellent antifungal activities. Among this series, three molecules (Hyd.H, Hyd.OCH3, and Hyd.Cl) significantly reduced C. albicans viability, with rapid fungicidal activity. In addition, these three compounds exhibited significant antifungal activity against clinically isolated fluconazole- or caspofungin-resistant C. albicans strains. Hyd.H, Hyd.OCH3, and Hyd.Cl did not show any cytotoxicity against human cancer cell lines up to a concentration of 50 µg/mL and decreased Candida biofilm formation, with a significant reduction of 60% biofilm formation with Hyd.OCH3. In an infection model of Caenorhabditis elegans with C. albicans, hydrazine-based compounds significantly reduced nematode mortality. Overall, fungicidal activity was observed for Hyd.H, Hyd.OCH3, and Hyd.Cl against C. albicans, and these compounds protected C. elegans from C. albicans infection.

Comparative studies were carried out of the fungal resistance of thiokol sealants to micromycetes isolated in the tropical climate of the Republic of Cuba and micromycetes used as test cultures according to GOST (State Standard) 9.049–91. The growth of both groups of microorganisms was revealed on samples of sealants vulcanized with manganese dioxide, and almost no growth was detected on samples of sealants vulcanized with sodium dichromate. In this study, an efficient Agrobacterium-mediated genetic transformation method followed by Tc chitiI gene for enhanced expression in peanut (Arachis hypogaea L.) cv ICG 7827 is reported. A. tumefaciens strain LBA 4404 harboring the plant transformation plasmid pBinAR contains the Tc chitI gene and nptII gene as a selectable marker. The precultured leaflet (LL) explants were infected with A. tumefaciens containing Tc chitiI gene and cocultivated on SIM (MMS+10 mg/L BAP+1 mg/L NAA+2 mg/L AgNO3) for four days. After co-cultivation, these explants were transferred to selection medium (SIM+75 mg/L Kan+250 mg/L Cefotaxime). In T0 generation, 70 % of transformation efficiency was recorded. T1 generation derived from the primary transgenic (T0) events revealed a Mendelian inheritance pattern (3:1) for the Tc chit I transgene. T1 transgenic peanut plants were tested for resistance against C. arachidicola, C. personatum, and P. arachidis by infection with the microspores using detached leaf assay. The Tc chitI gene expressed in T1 transgenic plants have shown longer incubation, longer latent period, and lower infection frequencies than non-transformed (WT) plants. A significant negative correlation was recorded between chitinase activity and the frequency of infection to the C. arachidicola, C. personatum and P. arachidis pathogens. [ABSTRACT FROM AUTHOR]

Free and microencapsulated thyme essential oils (Zataria multiflora Boiss) were incorporated into solvent and waterborne polyurethane coatings separately to enhance decay resistance of the biocide free coatings. The essential oil was extracted by hydro-distillation in a Clevenger-type apparatus for 4 h. Poly (methyl methacrylate) (PMMA) microcapsules containing thyme oil as an active ingredient were prepared through solvent evaporation method with oil in water emulsion system. Fungal resistance of the coated hornbeam wood (Carpinus betulus) against white rot fungus Coriolus versicolor CTB 863 A and brown rot fungus Coniophora puteana BAM Ebw. 15 was tested according to European EN 113 standard before and after six-cycle accelerated aging test (ASTM D1037). The results revealed that the core-shell capsules were formed properly, and their sizes were in the range of 5–50 µ. The encapsulation efficiency determined by UV–visible spectrophotometer at λ = 275 nm was 67%. The free essential oil was not efficient enough to improve the fungal resistance, while the microencapsulated oil enhanced the resistance even after the accelerated aging via a controlled-release mechanism as well as protected the susceptible ingredients through the shielding effect of the polymeric shell. TCMTB (2-(thiocyanomethylthio)-benzothiazole), as a broad spectrum antifungal agent, has found a wide spread usage in processing of pickled, wet-blue and crust leathers and became one of the most used fungicides in leather although it suffered some drawbacks related to its toxicity and handling hazards. Monitoring TCMTB usage and the amounts in leather while keeping the antifungal effectiveness has become a necessity. In this study; 5 different TCMTB based fungicides with similar concentrations sold under different trade names were applied to wet-blue sheep skins. Then the leathers and baths were evaluated for their TCMTB content by using UV Spectrophotometer and HPLC methods to determine the concentration of the active content in fungicides and residue in leather. Meanwhile the effect of the biocides on the fungal resistance of leathers was tested by the use of two methods: tropical chamber and ASTM D4576.
