1. Optimal control problem of evolution equation governed by hypergraph Laplacian

Abstract: In this paper, we consider an optimal control problem of an ordinary differential inclusion governed by the hypergraph Laplacian, which is defined as a subdifferential of a convex function and then is a set-valued operator. We can assure the existence of optimal control for a suitable cost function by using methods of a priori estimates established in the previous studies. However, due to the multivaluedness of the hypergraph Laplacian, it seems to be difficult to derive the necessary optimality condition for this problem. To cope with this difficulty, we introduce an approximation operator based on the approximation method of the hypergraph, so-called ``clique expansion.'' We first consider the optimality condition of the approximation problem with the clique expansion of the hypergraph Laplacian and next discuss the convergence to the original problem. Abstract: The PHENIX experiment measured the centrality dependence of two-pion Bose-Einstein correlation functions in $\sqrt{s_{_{NN}}}=200$~GeV Au$+$Au collisions at the Relativistic Heavy Ion Collider at Brookhaven National Laboratory. The data are well represented by L\'evy-stable source distributions. The extracted source parameters are the correlation-strength parameter $\lambda$, the L\'evy index of stability $\alpha$, and the L\'evy-scale parameter $R$ as a function of transverse mass $m_T$ and centrality. The $\lambda(m_T)$ parameter is constant at larger values of $m_T$, but decreases as $m_T$ decreases. The L\'evy scale parameter $R(m_T)$ decreases with $m_T$ and exhibits proportionality to the length scale of the nuclear overlap region. The L\'evy exponent $\alpha(m_T)$ is independent of $m_T$ within uncertainties in each investigated centrality bin, but shows a clear centrality dependence. At all centralities, the L\'evy exponent $\alpha$ is significantly different from that of Gaussian ($\alpha=2$) or Cauchy ($\alpha=1$) source distributions. Comparisons to the predictions of Monte-Carlo simulations of resonance-decay chains show that in all but the most peripheral centrality class (50%-60%), the obtained results are inconsistent with the measurements, unless a significant reduction of the in-medium mass of the $\eta'$ meson is included. In each centrality class, the best value of the in-medium $\eta'$ mass is compared to the mass of the $\eta$ meson, as well as to several theoretical predictions that consider restoration of $U_A(1)$ symmetry in hot hadronic matter., Comment: 401 authors from 75 institutions, 20 pages, 15 figures, 2 tables. v1 is version submitted to Physical Review C. Abstract: High-momentum two-particle correlations are a useful tool for studying jet-quenching effects in the quark-gluon plasma. Angular correlations between neutral-pion triggers and charged hadrons with transverse momenta in the range 4--12~GeV/$c$ and 0.5--7~GeV/$c$, respectively, have been measured by the PHENIX experiment in 2014 for Au$+$Au collisions at $\sqrt{s_{_{NN}}}=200$~GeV. Suppression is observed in the yield of high-momentum jet fragments opposite the trigger particle, which indicates jet suppression stemming from in-medium partonic energy loss, while enhancement is observed for low-momentum particles. The ratio and differences between the yield in Au$+$Au collisions and $p$$+$$p$ collisions, $I_{AA}$ and $\Delta_{AA}$, as a function of the trigger-hadron azimuthal separation, $\Delta\phi$, are measured for the first time at the Relativistic Heavy Ion Collider. Abstract: $H^2$-spatial regularity of stationary and non-stationary problems for Bingham fluids formulated with the pseudo-stress tensor is discussed. The problem is mathematically described by an elliptic or parabolic variational inequality of the second kind, to which weak solvability in the Sobolev space $H^1$ is well known. However, higher regularity up to the boundary in a bounded smooth domain seems to remain open. This paper indeed shows such $H^2$-regularity if the problems are supplemented with the so-called perfect slip boundary condition. For the stationary Bingham-Stokes problem, the key of the proof lies in a priori estimates for a regularized problem avoiding investigation of higher pressure regularity, which seems difficult to get in the presence of a singular diffusion term. The $H^2$-regularity for the stationary case is then directly applied to establish strong solvability of the non-stationary Bingham-Navier-Stokes problem, based on discretization in time and on the truncation of the nonlinear convection term., Comment: 21 pages

Abstract: We consider a general optimal control problem in the setting of gradient flows. Two approximations of the problem are presented, both relying on the variational reformulation of gradient-flow dynamics via the Weighted-Energy-Dissipation variational approach. This consists in the minimization of global-in-time functionals over trajectories, combined with a limit passage. We show that the original nonpenalized problem and the two successive approximations admits solutions. Abstract: The PHENIX experiment has performed a systematic study of identified charged-hadron ($\pi^\pm$, $K^\pm$, $p$, $\bar{p}$) production at midrapidity in $p$$+$Al, $^3$He$+$Au, Cu$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV and U$+$U collisions at $\sqrt{s_{_{NN}}}=193$ GeV. Identified charged-hadron invariant transverse-momentum ($p_T$) and transverse-mass ($m_T$) spectra are presented and interpreted in terms of radially expanding thermalized systems. The particle ratios of $K/\pi$ and $p/\pi$ have been measured in different centrality ranges of large (Cu$+$Au, U$+$U) and small ($p$$+$Al, $^3$He$+$Au) collision systems. The values of $K/\pi$ ratios measured in all considered collision systems were found to be consistent with those measured in $p$$+$$p$ collisions. However the values of $p/\pi$ ratios measured in large collision systems reach the values of $\approx0.6$, which is $\approx2$ times larger than in $p$$+$$p$ collisions. These results can be qualitatively understood in terms of the baryon enhancement expected from hadronization by recombination. Identified charged-hadron nuclear-modification factors ($R_{AB}$) are also presented. Enhancement of proton $R_{AB}$ values over meson $R_{AB}$ values was observed in central $^3$He$+$Au, Cu$+$Au, and U$+$U collisions. The proton $R_{AB}$ values measured in $p$$+$Al collision system were found to be consistent with $R_{AB}$ values of $\phi$, $\pi^\pm$, $K^\pm$, and $\pi^0$ mesons, which may indicate that the size of the system produced in $p$$+$Al collisions is too small for recombination to cause a noticeable increase in proton production., Comment: 480 authors from 78 institutions, 18 pages, 6 tables, 16 figures. v2 is version accepted for publication in Physical Review C. 9. Propagation of periodic director and flow patterns in a cholesteric liquid crystal under electroconvection

Abstract: Abstract The electroconvection of liquid crystals is a typical example of a dissipative structure generated by complicated interactions between three factors: convective flow, structural deformation, and the migration of charge carriers. In this study, we found that the periodic structural deformation of a cholesteric liquid crystal propagates in space, like a wave, under an alternating-current electric field. The existence of convection and charge carriers was confirmed by flow-field measurements and dielectric relaxation spectroscopy. Given that the wave phenomenon results from electroconvection, we suggest a possible model for describing the mechanism of wave generation. The validity of the model was examined using the Onsager variational principle. Consequently, it was suggested that wave generation can be described by four effects: the electrostatic potential, mixing entropy, anisotropic friction due to charge migration, and viscous dissipation of the liquid crystal.

10. Development of a SciFi-based beam monitor for COMET

Abstract: COMET is a leading experiment to search for coherent conversion of $\mu^- \mathrm{N}\to e^- \mathrm{N}$ with a high-intensity pulsed muon beamline, produced by the innovative slow extraction techniques. Therefore, it is critical to measure the characteristics of the muon beam. We set up a Muon Beam Monitor (MBM), where scintillation fibers (SciFi) weaved in the cross shape are coupled to silicon photomultipliers (SiPM), to measure the spatial profile and timing structure of the extracted muon beam for COMET. The MBM detector has been tested successfully with a proton beamline in China Spallation Neutron Source (CSNS) and taken data with good performance in the commissioning run called COMET Phase-$\alpha$. Experience of the MBM development, such as the mechanical structure and electronics readout, and its beam measurement results will be shared.

Abstract: This paper is concerned with the Cauchy problem of a multivalued ordinary differential equation governed by the hypergraph Laplacian, which describes the diffusion of ``heat'' or ``particles'' on the vertices of hypergraph. We consider the case where the heat on several vertices are manipulated internally by the observer, namely, are fixed by some given functions. This situation can be reduced to a nonlinear evolution equation associated with a time-dependent subdifferential operator, whose solvability has been investigated in numerous previous researches. In this paper, however, we give an alternative proof of the solvability in order to avoid some complicated calculations arising from the chain rule for the time-dependent subdifferential. 18. Fresnel Microfacet BRDF: Unification of Polari-Radiometric Surface-Body Reflection

Abstract: Computer vision applications have heavily relied on the linear combination of Lambertian diffuse and microfacet specular reflection models for representing reflected radiance, which turns out to be physically incompatible and limited in applicability. In this paper, we derive a novel analytical reflectance model, which we refer to as Fresnel Microfacet BRDF model, that is physically accurate and generalizes to various real-world surfaces. Our key idea is to model the Fresnel reflection and transmission of the surface microgeometry with a collection of oriented mirror facets, both for body and surface reflections. We carefully derive the Fresnel reflection and transmission for each microfacet as well as the light transport between them in the subsurface. This physically-grounded modeling also allows us to express the polarimetric behavior of reflected light in addition to its radiometric behavior. That is, FMBRDF unifies not only body and surface reflections but also light reflection in radiometry and polarization and represents them in a single model. Experimental results demonstrate its effectiveness in accuracy, expressive power, and image-based estimation.

Abstract: Abstract Background Inattention due to attention‐deficit/hyperactivity disorder (ADHD) can lead to forgetfulness. Transient epileptic amnesia (TEA) can cause forgetfulness, similar to ADHD. We report a patient with ADHD who developed TEA. Case Presentation The patient was a 40‐year‐old woman with ADHD. She has been prone to forgetfulness since childhood. Two years before visiting our outpatient clinic, she had begun to occasionally forget events that had occurred several days earlier. However, she was largely unaware of the emergence of new amnestic symptoms. She had also begun to experience various other amnestic symptoms 2 months before she visited our clinic, which prompted her to visit our outpatient clinic. The combination of a detailed interview, electroencephalography (EEG) examination, and consideration of TEA enabled us to diagnose her with TEA and provide treatment accordingly. In our patient, daily forgetfulness due to ADHD delayed the recognition of new additional forgetfulness attributed to TEA. Conclusion Psychiatrists need to consider TEA when patients with ADHD present with changes in or exacerbation of forgetfulness.

Abstract: Abstract NLR family proteins act as intracellular receptors. Gene duplication amplifies the number of NLR genes, and subsequent mutations occasionally provide modifications to the second gene that benefits immunity. However, evolutionary processes after gene duplication and functional relationships between duplicated NLRs remain largely unclear. Here, we report that the rice NLR protein Pit1 is associated with its paralogue Pit2. The two are required for the resistance to rice blast fungus but have different functions: Pit1 induces cell death, while Pit2 competitively suppresses Pit1-mediated cell death. During evolution, the suppression of Pit1 by Pit2 was probably generated through positive selection on two fate-determining residues in the NB-ARC domain of Pit2, which account for functional differences between Pit1 and Pit2. Consequently, Pit2 lost its plasma membrane localization but acquired a new function to interfere with Pit1 in the cytosol. These findings illuminate the evolutionary trajectory of tandemly duplicated NLR genes after gene duplication.

Abstract: Abstract Background Four-dimensional flow magnetic resonance imaging (MRI) enables blood flow visualization. The absence of left atrial vortex flow (LAVF) has been implicated in the development of thrombus formation and arrhythmias. However, the clinical relevance of this phenomenon in patients with congenital heart disease (CHD) remains unclear. This study aimed to unravel the relationship of LAVF with left atrial functions in patients with CHD. Results Twenty-five participants who underwent cardiac MRI examinations were included (8 postoperative patients with CHD aged 17–41 years and 17 volunteers aged 21–31 years). All participants were in sinus rhythm. Four-dimensional flow MRI (velocity encoding 100 cm/s) assessed the presence of LAVF, and its relationship with left atrial function determined by transthoracic echocardiography was explored. LAVF was detected in 16 patients. Upon classification of the participants based on the presence or absence of LAVF, 94% of participants in the LAVF group were volunteers, while 78% of those in the without LAVF group were postoperative patients. Participants without LAVF had a significantly lower left atrial ejection fraction (61% vs. 70%, p = 0.019), reservoir (32% vs. 47%, p = 0.006), and conduit (22% vs. 36%, p = 0.002) function than those with LAVF. Conclusions LAVF occurred during the late phase of ventricular systole, and left atrial reservoir function may have contributed to its occurrence. Many postoperative patients with CHD experienced a loss of LAVF. LAVF may indicate early left atrial dysfunction resulting from left atrial remodeling.

23. Morphogenesis of a chiral liquid crystalline droplet with topological reconnection and Lehmann rotation

Abstract: Abstract Morphogenesis is a hierarchical phenomenon that produces various macroscopic structures in living organisms, with high reproducibility. This study demonstrates that such structural formation can also be observed in a chiral liquid crystalline droplet under a temperature gradient. Through specific control of the temperature change process, we were able to switch the final structure obtained as a result of the formation via the appearance and reconnection of loop defects in the transient state during structure formation. Simultaneously, the existence of the gradient resulted in a characteristic rotational phenomenon called Lehmann rotation, which was prominently induced in the transient state. By demonstrating three-dimensional measurements of the flow field, we revealed the existence of Marangoni convection in the state. Consequently, it is indicated that the convection results in high-speed Lehmann rotation and large structural deformation with topological changes, thereby playing a significant role in the structure formation.

Abstract: A system with equation and dynamic boundary condition of Cahn-Hilliard type is considered. This system comes from a derivation performed in Liu-Wu (Arch. Ration. Mech. Anal. 233 (2019), 167--247) via an energetic variational approach. Actually, the related problem can be seen as a transmission problem for the phase variable in the bulk and the corresponding variable on the boundary. The asymptotic behavior as the coefficient of the surface diffusion acting on the boundary phase variable goes to 0 is investigated. By this analysis we obtain a forward-backward dynamic boundary condition at the limit. We can deal with a general class of potentials having a double-well structure, including the non-smooth double-obstacle potential. We illustrate that the limit problem is well-posed by also proving a continuous dependence estimate. Moreover, in the case when the two graphs, in the bulk and on the boundary, exhibit the same growth, we show that the solution of the limit problem is more regular and we prove an error estimate for a suitable order of the diffusion parameter., Comment: 27 pages

Abstract: The PHENIX experiment reports systematic measurements at the Relativistic Heavy Ion Collider of $\phi$-meson production in asymmetric Cu$+$Au collisions at $\sqrt{s_{_{NN}}}$=200 GeV and in U$+$U collisions at $\sqrt{s_{_{NN}}}$=193 GeV. Measurements were performed via the $\phi\rightarrow K^{+}K^{-}$ decay channel at midrapidity $|\eta|<0.35$. Features of $\phi$-meson production measured in Cu$+$Cu, Cu$+$Au, Au$+$Au, and U$+$U collisions were found to not depend on the collision geometry, which was expected because the yields are averaged over the azimuthal angle and follow the expected scaling with nuclear-overlap size. The elliptic flow of the $\phi$ meson in Cu$+$Au, Au$+$Au, and U$+$U collisions scales with second-order-participant eccentricity and the length scale of the nuclear-overlap region (estimated with the number of participating nucleons). At moderate $p_T$, $\phi$-meson production measured in Cu$+$Au and U$+$U collisions is consistent with coalescence-model predictions, whereas at high $p_T$ the production is in agreement with expectations for in-medium energy loss of parent partons prior to their fragmentation. Abstract: Introduction: Young adults are hesitant to receive the coronavirus disease 2019 (COVID-19) vaccination owing to concerns regarding adverse events despite the effectiveness of vaccines in preventing SARS-CoV-2 infection-associated serious illness, hospitalization, and death. Methods: A retrospective cohort study was conducted in Gifu University students receiving the mRNA-1273 vaccine and boosters to elucidate the real incidence of adverse events and factors that prevent them. We examined the adverse events and identified potential risk factors through a self-administered questionnaire on the participants’ physical condition after COVID-19 vaccination. Results: Focal/systemic adverse events were highly frequent among university students after receiving the COVID-19 vaccine; however, there were no life-threatening cases or hospitalizations over two years. Abstract: Abstract Background Subacute myelo‐optico‐neuropathy (SMON) is a neurological disorder associated with the administration of clioquinol, particularly at very high doses. Although clioquinol has been used worldwide, there was an outbreak of SMON in the 1950s–1970s in which the majority of cases were in Japan, prompting speculation that the unique genetic background of the Japanese population may have contributed to the development of SMON. Recently, a possible association between loss‐of‐function polymorphisms in NQO1 and the development of SMON has been reported. In this study, we analyzed the relationship between NQO1 polymorphisms and SMON in Japan. Methods We analyzed 125 Japanese patients with SMON. NQO1 loss‐of‐function polymorphisms (rs1800566, rs10517, rs689452, and rs689456) were evaluated. The allele frequency distribution of each polymorphism was compared between the patients and the healthy Japanese individuals (Human Genomic Variation Database and Integrative Japanese Genome Variation Database), as well as our in‐house healthy controls. Results The frequencies of the loss‐of‐function NQO1 alleles in patients with SMON and the normal control group did not differ significantly. Conclusion We conclude that known NQO1 polymorphisms are not associated with the development of SMON.

29. Usefulness of health checkup‐based indices in identifying metabolic dysfunction‐associated steatotic liver disease

Abstract: Abstract Aims The application of indices in the context of metabolic dysfunction‐associated steatotic liver disease (MASLD) remains unexplored. We aimed to validate the ability of alanine aminotransferase (ALT), fatty liver index (FLI), and hepatic steatosis index (HSI) to identify MASLD during health checkups. Methods We recruited 627 participants and utilized their health checkup data and ultrasound to assess the potential of using ALT, FLI, and HSI as indices for MASLD; this was indicated by the area under the curve (AUC) and restricted cubic spline (RCS) model. The optimal, rule‐out (sensitivity ≥90%), and rule‐in (specificity ≥90%) cutoff values of each index for identifying MASLD were reported. Results Among participants with a median age of 46 years, the prevalence of MASLD was 28% in total (38% in males and 18% in females). RCS models confirmed a linear association between indices and MASLD. ROC analyses indicated that the AUC of ALT in identifying MASLD was 0.79 for the total cohort, 0.81 for males, and 0.69 for females. The optimal, rule‐out, and rule‐in cutoff values for ALT were 21, 13, and 29, respectively. Similarly, the AUC of FLI/HSI in identifying MASLD was 0.90/0.88 for the total cohort, 0.86/0.85 for males, and 0.93/0.90 for females. Considering the reference cutoff values, distinct cutoff values were observed between the sexes for FLI, while HSI had similar cutoff values. Conclusion This study demonstrated that ALT > 30 IU/L is a reasonable cutoff value to rule‐in MASLD. ALT, FLI, and HSI are reliable indices for identifying MASLD during health checkups.

Abstract: This paper proposes a stretchable-substrate-impregnated fabric for deployable high-gain antenna for small satellites. Constellations of small satellites offer worldwide coverage of wireless communication and earth observation; high-gain antennas are required to realize high-speed communication and high-resolution radar. This paper aims to develop a CubeSat onboard high-gain reflectarray antenna that can be folded compactly and deployed in space. The novelty of this paper is the stretchable-substrate-impregnated fabric, which can realize low stowage volume with high fabrication tolerance and low resistivity with single-piece reflectarray to avoid gain degradation. The origami-folding-pattern compatible reflection elements can avoid creases on the elements, avoiding gain reduction after folding. A $17\times 17$ -element reflectarray using the stretchable-substrate-impregnated fabric and the origami-folding-pattern compatible reflection elements is designed, fabricated, and characterized. The folded volume of the reflectarray is 27 mm in height and 58 mm in diameter, and the measured fabrication tolerance of the reflection element was $20 ~\mu \text{m}$ at most. Electrical antenna measurements confirmed low resistivity of 1.20 $\text{m}\Omega /\Box $ and a high realized gain of 23.6 dBi, comparable with conventional flexible printed circuit reflectarray antennas. The simulated and measured antenna efficiency is 25.3% and 25.1%, respectively. The simulated and measured 1-dB gain down bandwidth is 1.6% and 1.5%, respectively.

32. Usefulness of a questionnaire for assessing the relationship between eating behavior and steatotic liver disease among Japanese male young adults

Abstract: Abstract This study aimed to reveal the relationship between eating behavior and nonalcoholic fatty liver disease (NAFLD)/metabolic dysfunction-associated steatotic liver disease (MASLD) in young adults and suggest a questionnaire for eating behavior assessment. We included 322 male graduate students at Gifu University. Diagnoses of NAFLD and MASLD were based on the presence of hepatic steatosis on ultrasonography. Eating behavior was assessed using the eating behavior questionnaire (EBQ) recommended by the Japan Society for the Study of Obesity. We assessed the eating behaviors associated with NAFLD and MASLD using logistic regression, decision tree, and random forest analyses. The median age of the participants was 22 years, and 16% and 11% had NAFLD and MASLD, respectively. The EBQ total score was significantly higher in participants with MASLD than in those without MASLD (102 vs. 90 points, P = 0.006) and in those with NAFLD than in those without NAFLD (97 vs. 90 points, P = 0.007). Among eating behavior categories, the decision tree and random forest analyses revealed that “perception of constitution and weight” was the strongest contributor for NAFLD/MASLD. Our study revealed that eating behavior assessed with the EBQ is robustly associated with NAFLD and MASLD in young male adults.

Abstract: The PHENIX collaboration presents a systematic study of $\pi^0$ production from $p$$+$$p$, $p$$+$Al, $p$$+$Au, $d$$+$Au, and $^{3}$He$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV. Measurements were performed with different centrality selections as well as the total inelastic, 0%--100%, selection for all collision systems. For 0%--100% collisions, the nuclear modification factors, $R_{xA}$, are consistent with unity for $p_T$ above 8 GeV/$c$, but exhibit an enhancement in peripheral collisions and a suppression in central collisions. The enhancement and suppression characteristics are similar for all systems for the same centrality class. It is shown that for high-$p_T$-$\pi^0$ production, the nucleons in the $d$ and $^3$He interact mostly independently with the Au nucleus and that the counter intuitive centrality dependence is likely due to a physical correlation between multiplicity and the presence of a hard scattering process. These observations disfavor models where parton energy loss has a significant contribution to nuclear modifications in small systems. Nuclear modifications at lower $p_T$ resemble the Cronin effect -- an increase followed by a peak in central or inelastic collisions and a plateau in peripheral collisions. The peak height has a characteristic ordering by system size as $p$$+$Au $>$ $d$$+$Au $>$ $^{3}$He$+$Au $>$ $p$$+$Al. For collisions with Au ions, current calculations based on initial state cold nuclear matter effects result in the opposite order, suggesting the presence of other contributions to nuclear modifications, in particular at lower $p_T$., Comment: 554 authors from 81 institutions, 21 pages, 13 figures, and 3 tables. Abstract: An asymptotic analysis for a system with equation and dynamic boundary condition of Cahn-Hilliard type is carried out as the coefficient of the surface diffusion acting on the phase variable tends to 0, thus obtaining a forward-backward dynamic boundary condition at the limit. This is done in a very general setting, with nonlinear terms admitting maximal monotone graphs both in the bulk and on the boundary. The two graphs are related by a growth condition, with the boundary graph that dominates the other one. It turns out that in the limiting procedure the solution of the problem looses some regularity and the limit equation has to be properly interpreted in the sense of a subdifferential inclusion. However, the limit problem is still well-posed since a continuous dependence estimate can be proved. Moreover, in the case when the two graphs exhibit the same growth, it is shown that the solution enjoys more regularity and the boundary condition holds almost everywhere. Abstract: Abstract Background CD38 has been established as an important therapeutic target for multiple myeloma (MM), for which two CD38 antibodies are currently approved—daratumumab and isatuximab. CD38 is an ectoenzyme that degrades NAD and its precursors and is involved in the production of adenosine and other metabolites. Aim Among the various mechanisms by which CD38 antibodies can induce MM cell death is immunomodulation, including multiple pathways for CD38‐mediated T‐cell activation. Patients who respond to anti‐CD38 targeting treatment experience more marked changes in T‐cell expansion, activity, and clonality than nonresponders. Implications Resistance mechanisms that undermine the immunomodulatory effects of CD38‐targeting therapies can be tumor intrinsic, such as the downregulation of CD38 surface expression and expression of complement inhibitor proteins, and immune microenvironment‐related, such as changes to the natural killer (NK) cell numbers and function in the bone marrow niche. There are numerous strategies to overcome this resistance, which include identifying and targeting other therapeutic targets involved in, for example, adenosine production, the activation of NK cells or monocytes through immunomodulatory drugs and their combination with elotuzumab, or with bispecific T‐cell engagers.

41. How Long Would You Like to Live? A 25-year Prospective Observation of the Association Between Desired Longevity and Mortality

Abstract: Background: Desired longevity represents how strongly people esteem possible extensions of their own lifetime. The association between desired longevity and mortality risk has been reported in only one prospective study, which examined a small sample of older participants. We aimed to examine the hypothesis that desired longevity at middle-age predicted long-term survival. Methods: In the prospective cohort study, residents aged 40–64 years were asked how long they would like to live and asked to choose one from three options: longer than, as long as, or shorter than the life expectancy. We used Cox proportional hazards model to calculate the hazard ratios (HRs) and 95% confidence intervals (CIs) for all-cause and cause-specific mortality according to the three groups for desired longevity, treating the “longer than” group as the reference. We conducted mediation analysis to investigate the mechanism for the association between desired longevity and mortality. Results: We recruited 39,902 residents to the study. Risk of all-cause mortality was significantly higher in the “shorter than” group (HR 1.12; 95% CI, 1.04–1.21). The association was independent of sex, age, marital status, education, medical history, and health status. Regarding cause of death, mortality risk of cancer (HR 1.14; 95% CI, 1.00–1.29) and suicide (HR 2.15; 95% CI, 1.37–3.38) were also higher in the “shorter than” group. The unhealthy lifestyle mediated this association with all-cause mortality by 30.4%. Conclusion: Shorter desired longevity was significantly associated with an increased risk of all-cause mortality, and mortality from cancer and suicide. Lifestyle behaviors particularly mediated this association.

Abstract: Leucine is a branched-chain amino acid that is present in protein, and it is an essential factor in activating the mechanistic target of the rapamycin complex 1 signaling pathway and increasing muscle protein synthesis. However, the loss of digestive function after total gastrectomy leads to impaired protein absorption, potentially failing to stimulate muscle protein synthesis. Therefore, this study aimed to investigate whether muscle protein synthesis is enhanced by oral skim milk administration after total gastrectomy. Male Sprague Dawley rats were divided into total gastrectomy (TG) and sham surgery (S) groups. After five weeks postoperatively, we orally administered skim milk to achieve 3.1 g protein/kg body weight and collected blood and gastrocnemius muscle. The gastrocnemius muscle weight was significantly lower in the TG group than in the S group (p < 0.05). The increase in plasma leucine concentration was significantly lower in the TG group than in the S group (p < 0.05). The skeletal muscle protein synthesis and the phosphorylation of p70S6K and 4E-BP1 showed a similar increase in both groups. Even after TG, muscle protein synthesis was stimulated by consuming skim milk, accompanied by a sufficient rise in plasma leucine concentration.
