5. Plant Regeneration through Leaf Culture of Yacon

application/pdf, 論文(Article), ヤーコン(Smallanthus sonchifolius)は, 南米アンデス高原原産のキク科に属する塊根作物で, フラクトオリゴ糖が多量に含まれていることから注目されている.ヤーコンは種子繁殖が困難であり, またウイルス病の感染も考えられるため, 組織培養による苗の増殖が求められている.種々の組み合わせのオーキシンとサイトカイニンを添加したMS培地でヤーコンの葉片を培養したところ, 0.1 mg・liter^ 2, 4D, 0.1 mg・liter^ BAPを添加した培地でもっとも効率的にカルスが誘導された.これらのカルスを1.0 mg・liter^ BAPと0.1 mg・liter^ GA_3を添加したHaR培地(Paterson・Everett, 1985)で培養したところ, 不定芽に由来するシュートがカルス上に形成された.さらに, これらのカルスを分割して1カ月間隔で継代培養し, 約1年間にわたってシュートの再分化を継続させることができた.得られたシュートは0.1 mg・liter^ NAAを添加した1/2MS培地で発根し, 順化・鉢出しを経て圃場で順調に生長した.親植物および再分化植物から抽出したDNAについてAFLP解析を行ったところ, 13中12の再分化植物で親植物とは異なる増幅断片が得られ, ソマクローナル変異が生じていることが示唆された., Yacon (Smallanthus sonchifolius), a member of Asteraceae, is an ethnic food of Andeans. Interest in this crop is increasing because of its high fructo-oligosaccharide eontent in the tuberous roots. Due to the low productivity of viable seeds and virus infections through vegetative propagation, effective methods of in vitro propagation of yacon is being sought. To accomplish this, leaf pieces of yacon were cultured on the gelrite-solidified MS media, supplemented with various combinations of auxins and cytokinins. The most effective means of inducing calli was achieved on a MS medium that was supplemented with 0.1 mg・liter^ 2, 4-D and 0.1 mg・liter^ BAP. Shoot regeneration occurred when the calli were subcultured on the HaR medium (Paterson and Everett, 1985), supplemented with 1.0 mg・liter^ BAP and 0.1 mg・liter^ GA_3. By sub culturing those calli at a monthly interval, the shoot regeneration continued up to the 10th subculture. The regenerated shoots rooted ; the plantlets were well-acclimatized and transplanted successfully to the field. AFLP analysis conducted on the extracted DNA from the regenerants revealed 12 out of the 13 possessed different amplified fragments from those of the mother plants, indicating that the somaclonal mutation took place during the course of callusing and plant regeneration.

8. Novel process for SiO_2/Si selective etching using a novel gas source for preventing global warming

Abstract

A novel gas source replacing fluorocarbon feed gases has been developed for preventing global warming. The novel gas source was designed to generate fluorocarbon species from polytetrafluoroethylene by CO_2 laser ablation. The species generated from the gas source were introduced into an electron cyclotron resonance (ECR) plasma employing Ar gases. To characterize the gas source, CF_x (x=1–3) radical densities with and without plasmas were measured by infrared diode laser absorption spectroscopy. In the ECR plasma employing the novel gas source, CF_x (x =1–3) radical densities were estimated to be of the order of 10^12–10^13 cm^-3. The gas source has been applied to the selective etching of SiO_2 to Si using the ECR plasma. As a result, the etching characteristics by ECR plasma employing the novel gas source were equivalent to those by a conventional ECR plasma employing C_4F_8 gas. Therefore, this novel gas source is applicable to etching processes for preventing global warming.

9. Early clinical outcomes of 3D-conformal radiotherapy using accelerated hyperfractionation without intracavitary brachytherapy for cervical cancer

Abstract

Purpose/Objective: To evaluate the outcome of cervical cancer patients unable to undergo conventional intracavitary brachytherapy (ICBT) treated with 3D-conformal radiotherapy (3DCRT) alone using accelerated hyperfractionation (AHF). Methods and Materials: We reviewed the records of 7 patients who had received definitive radiotherapy with 3DCRT alone using AHF for cervical cancer between 2002 and 2005. FIGO stage was IB (1), IIB (2), IIIA (1), IIIB (2), and IVA (1). The reason we did not perform ICBT was due to patient refusal. In 1 patient with stage IB, a total dose of 65.4 Gy was delivered by local irradiation (LI) only. In 1 patient with stage IIIA, a total dose of 60 Gy was delivered by LI only. In 5 patients with Stage IIB-IV, a median total dose of 70.8 Gy was delivered by combination of whole pelvic irradiation (median dose of 45 Gy) with LI. Median overall treatment time was 42 days. Results: Median follow-up for survival patients was 17 months. Out of 7 patients, 6 patients had CR and 1 patient had PR. The response rate was 100%.0 The 2-year local control rate was 85.7%.0 Of these patients, 5 are alive without disease and 1 is alive with lung metastasis. Conclusions: Our outcomes suggest that 3DCRT using AHF may be a promising as a definitive treatment for cervical cancer when ICBT is not able to be performed.

Abstract

A combination of external beam radiotherapy (EBRT) and intracavitary brachytherapy (ICBT) is well established as the standard radical radiotherapy (RT) for cervical cancer. However, it is sometimes necessary to perform EBRT alone for patients where ICBT is not feasible. For these patients, we initiated EBRT alone with three-dimensional conformal radiotherapy (3DCRT). The purpose of this study is to evaluate the results of EBRT alone without ICBT for patients with cervical cancer. Sixteen patients were treated with EBRT alone between 2002 and 2009. There were three stage IIB, six stage IIIB and seven patients with stage IVA disease. A total of 10 patients were treated with a median dose of 66 Gy with a median overall treatment time (OTT) of 40 days delivered by a concomitant boost (CCB), and a median dose of 60 Gy with a median OTT of 47 days was administered for six patients by conventional fractionation (CF). The 3-year overall survival (OAS) and local control (LC) rates were 43.8% and 75.0%, respectively. The 3-year LC rate was 90.0% for the CCB group, 50.0% for the CF group (P = 0.0692); 100% for OTT ≤42 days, 42.9% for OTT ≥43 days (P = 0.0095). No severe acute and late adverse effects were encountered for any of the patients. These outcomes suggest that EBRT with a CCB program may be a promising radical treatment for cervical cancer that provides better LC with minimal complications, especially in cases where ICBT cannot be performed.

Abstract

A new method has been devised for the determination of mercury in electrothermal atomic absorption spectrometry. The mercury-adsorbed ion-exchange resin was heated in a carbon furnace, and the absorption peak of mercury appeared in the “drying” period without interference from the peak of the resin. This method made it possible to determine 0.04ppm of mercury.

Abstract

We aimed to evaluate the feasibility and efficacy of hypofractionated involved-field radiation therapy (IFRT) omitting elective nodal irradiation (ENI) with concurrent chemotherapy for locally advanced non-small-cell lung cancer (NSCLC). Between July 2004 and July 2006, ten patients with locally advanced NSCLC were included in this study. One had stage IIIA and 9 had stage IIIB disease. The treatment consisted of IFRT in fractions of 2.5 Gy and weekly carboplatin (CBDCA)/paclitaxel (PTX). Hypofractionated IFRT with a median total dose of 65 Gy with median percent total lung volume exceeding 20 Gy (V20) of 20.2%, and a median of five courses of chemotherapy with weekly CBDCA (area under the curve, 1.5-2.0)/PTX (30-35 mg/m(2)) were given to all patients. The median survival time and the 1-, 2-, and 3-year overall survival rates were 29.5 months and 90.0%, 58.3%, and 43.8%, respectively. No elective nodal failure was encountered during the median follow up of 18.2 months. No acute or late toxicities of grade 3 or worse were observed. No in-field recurrence occurred in the group with a total dose of 67.5 Gy or more, but there was such recurrence in 83.3% of those in the group with less than 67.5 Gy. Hypofractionated IFRT with weekly CBDCA/PTX was a feasible treatment regimen. Hypofractionated IFRT with a total dose of 67.5 Gy or more could be a promising modality to improve the treatment results in patients with locally advanced NSCLC.

Abstract

Electronic and optical properties of screw-vacancy γ-In2Se3 were studied by using the first-principles calculations. We found that the dangling-bond bands appear around the valence-band top.
