Abstract

The positive transcription elongation factor b (P-TEFb) is a critical co-activator for transcription of most cellular and viral genes, including those of HIV. While P-TEFb is regulated by 7SK snRNA in proliferating cells, it is absent in quiescent and terminally differentiated cells, which has remained unexplored. In these cells, we found that CycT1 not bound to CDK9 is rapidly degraded. Moreover, productive CycT1:CDK9 interactions require phosphorylation of two threonine residues (Thr143 and Thr149) in CycT1 by PKC. Conversely, PP1 dephosphorylates these sites. Thus, PKC inhibitors or removal of PKC by chronic activation results in P-TEFb disassembly and CycT1 degradation. This finding not only recapitulates P-TEFb depletion in resting CD4+ T cells but also in anergic T cells. Importantly, our studies reveal mechanisms of P-TEFb inactivation underlying T cell quiescence, anergy, and exhaustion as well as proviral latency and terminal differentiation of cells.

6. Pb isotopic constraints on the genesis of the Toyoha indium-bearing polymetallic deposit, Hokkaido, Japan.

Abstract

The Toyoha epithermal polymetallic deposit has produced 4 700 t indium and used to be the largest indium producer in the world in the 1990s. The Pb isotopic composition of sulphides (sphalerite, galena, pyrite and wurtzite) was investigated from the indium-poor, earlier veins and the indium-rich later veins, together with the volcanic and sedimentary rocks in the Toyoha area. Data show that the Pb isotopic composition of the earlier veins fall within the range of volcanic rocks of the Toyoha area. On the other hand, the later veins yield isotopic compositions between the Toyoha volcanic rocks and the quaternary basalt of SW Hokkaido. This suggests that the sulphide and igneous rocks share a common source; e.g., Pb in both the sulphide and the magmas was derived from a deep geofluid from the subducted Pacific plate or, a fluid expelled at relatively shallow depths from the magmas significantly contributed to the ore forming fluid and metals, including the indium-rich later veins of the Toyoha deposit., The Toyoha epithermal polymetallic deposit has produced 4 700 t indium and used to be the largest indium producer in the world in the 1990s. 7. Adsorption Behavior of Divalent Metal Ions onto Surface-functionalized Mesoporous Silicate MCM-41 Having Schiff Base Structure

Abstract

Surface functionalized mesoporous silicates, MCM-41s, having 3-(2-pyridylmethylideneimino)propyl group (PI-MCM-41) or 3-(2-quinolylmethylideneimino)propyl group (QI-MCM-41) were prepared via Schiff base reaction, and the adsorption behavior of metal ions onto the modified MCM-41s was investigated. The function groups on the modified MCM-41 surface were confirmed by Fourier transform infrared spectroscopy (FT-IR), X-ray photoelectron spectroscopy (XPS), and elemental analysis. The metal ions examined, Co2+, Ni2+, Cu2+, Zn2+, Cd2+, and Pb2+, were quantitatively adsorbed on the PI-MCM-41 and QI-MCM-41, except for Mn2+. In the complexation with these metal ions, it was suggested that imine-N and heterocyclic-N atoms act as donor atoms. In addition, it was considered that the hydrophobicity derived from the organo-functional groups modified on MCM-41 contributed to improving the adsorption ability.

Abstract

This study was undertaken to investigate the extraction of In3+ and Ga3+ by 7-(4-ethyl-1-methyloctyl)-8-hydroxy-quinoline (Kelex100), which has been found to be a slow extraction process. Homogeneous liquid-liquid extraction utilizing upper critical solution temperature of 4-methyl-1,3-dioxolan-2-one (propylene carbonate, abbreviated as PC) was used to eliminate the interface and speed up the extraction, since distribution between two phases was thought to be the rate-determining step. The effect of contact time on the percentage of metal extraction was examined at 80ºC using Kelex100 dissolved in toluene and PC solutions and the difference between two solvents was evaluated. The results clearly show that the use of homogeneous liquid-liquid extraction using PC with Kelex100 is faster than traditional two phase extraction.

Abstract

Genital warts, also known as condyloma acuminata, are caused by the human papillomavirus (HPV). Rare cases have been reported in which genital warts have developed into cancerous tumors. Research has revealed more than 60 types of HPV. While HPV 6 is most often associated with genital warts, it is not found in cervical cancer. Types 16 and 18 are commonly detected in cervical cancer; other types such as 31, 33, 35, and 52b have been detected less frequently. Since HPV types 16 and 18 have been shown to be involved in causing the continuous growth of cells in tissue culture, it is believed that these viruses may play a causative role in the development of cancer of the cervix. It has been assumed that the viral DNA becomes integrated into the host genome as a part of the disease process of both warts and cancer, but little evidence to support this theory has been obtained directly from specimens of human cervical cancer. For this reason, cervical biopsy specimens were examined for the presence of human papillomavirus DNA, and to determine if the DNA is integrated into the host genome or is present in the episomal state, that is, present as unintegrated viral genome. HPV DNA was found in 62 percent of the cervical cancers, in keeping with previous reports. Many researchers suspect that the incidence would approach 100 percent if detection methods were more sensitive. Also in keeping with previous reports, HPV 16 was the most commonly identified type, with HPV 18 also frequently observed. Ten cancer specimens positive for HPV 16 DNA were analyzed for the physical state of the genes. In four specimens, both integrated DNA and viral DNA were present. In one specimen, only viral DNA could be found. Although it is impossible to rule out cancer resulting from some other cause in this specimen, the findings suggest that integration of the viral DNA into the host genome may not be necessary for the cancerous transformation of cells by HPV. Abstract

We have developed a simple agglutination (LA) method for the detection of enteric adenovirus (EAd) in stool samples from infants with acute gastroenteritis. Ad type 41 (Ad41) was detected with high sensitivity and specificity by a slide agglutination test using latex particles coated with antiAd41 antibody (LA-antiAd41). The agglutination of LA-antiAd41 with Ad41 on a glass slide was evident macroscopically within 2 min. The sensitivity of the LA method was four times higher than that of the EM method.
