Evidence displays that circular RNAs (circRNAs) are considerable mediators of numerous processes in cancer development. Given that many circRNAs are not functionally characterized, our aim was to explore the function and mechanisms of circ_0051428 in thyroid cancer (TC). The analysis of circ_0051428, miR-1248 and FN1 mRNA expression was conducted using real-time quantitative polymerase chain reaction. Cell growth was observed using CCK-8 and colony formation assays. Cell migration was investigated using wound healing assay. Cell apoptosis was identified by the expression of apoptosis-related proteins (Bax and Bcl-2) using Western blotting. Animal models were established to testify the role of circ_0051428 in vivo. The assumed binding between miR-1248 and circ_0051428 or FN1 was identified using dual-luciferase reporter or RIP assay. circ_0051428 exhibits an abnormally elevated expression in TC. circ_0051428 deficiency caused inhibition of TC cell proliferation, migration, clonogenic capacity, and inhibition of tumor growth in vivo. circ_0051428 directly targeted miR-1248, and FN1 was a target downstream of circ_0051428/miR-1248 axis. circ_0051428 could sponge miR-1248 to upregulate FN1. Furthermore, miR-1248 downregulation recovered circ_0051428 deficiency-suppressed cancer cell proliferation, survival and migration. Besides, the repressive effects of FN1 knockdown on cancer cell growth, survival and migration were also partly abolished by miR-1248 downregulation. circ_0051428 targeted miR-1248 to modulate FN1 expression, thereby facilitating the malignant progression of TC, which contributed to the understanding of the molecular mechanism of TC development.

Mesocotyl elongation is a key trait influencing seedling emergence and establishment in direct-seeding rice cultivation. The phytohormone gibberellin (GA) has positive effects on mesocotyl elongation in rice. However, the physiological and molecular basis underlying the regulation of mesocotyl elongation mediated by GA priming under deep-sowing conditions remains largely unclear. In the present study, we performed a physiological and comprehensive transcriptomic analysis of the function of GA priming in mesocotyl elongation and seedling emergence using a direct-seeding japonica rice cultivar ZH10 at a 5-cm sowing depth. To look for the problems faced in the construction of the tele-critical care system, explore the framework of construction of the tele-critical care system, and verify the application effects of the established tele-critical care system.Through literature review and on-site investigation and demonstration, the causes affecting the construction of the tele-critical care system were explored. Through on-site investigation of the actual situation of the critical care department in relevant hospitals, arguing and choosing intended intensive care unit (ICU) and cooperative third-party communication and equipment companies, and through the Internet of Things and 5G communication technology, a tele-critical care system with the core hospital of the group as the center and the member institutes within the group as the nodes was built. Via the established tele-critical care system, activities such as tele-monitoring, visual remote ward rounds, remote consultation, remote teaching were carried out to verify the functions of the system.The insufficient cognition of relevant personnel, tele-medicine practice certification requirements, information security issues and the barriers of equipment information integration were the main causes affecting the construction of tele-critical care system. There were five parts in the tele-critical care system architecture foundations, including bed unit equipment and audio and video information collection system, lossless and secure transmission of collected information, real-time display of information in the remote center, real-time staff interaction between the centre and the nodal hospitals, and information cloud storage. It has been verified that patients' diagnostic and treatment information can be transmitted safely, losslessly and in real-time by a special line through private 5G network. Through this system, real-time and stable upload of audio and video information of patients and application information of monitors, ventilators and infusion work stations can be achieved; combined with tele-conference connections to conduct two-way communication with local medical staff, real-time tele-monitoring, visual remote ward rounds, remote consultation, remote teaching and other functions can be achieved.The tele-critical care system we established is feasible to construct within the medical group and can safely and effectively realize the functions of real-time tele-monitoring, visual remote ward rounds, remote consultation, and remote teaching.

Committee Mandate Concern for structural challenges of non-conventional, special surface vessels, including uncertainties in established design methods and modelling techniques. Particular attention shall be given to mega yachts, naval craft, offshore service vessels and work boats, which can be characterized by particular structural configurations and materials (wide openings, large unsupported structures, unconventionally shaped superstructures, etc.) and/or are to sustain specific loading conditions (harsh environment, severe cyclic loads or extreme operational ones). Introduction This Specialist Committee report for the ISSC 2022 proceedings covers a range of marine vessels particularly “Special” by nature of their structural configurations. In the ISSC 2018 V.5 Special Craft report (Truelock et al., 2018) the Committee’s focus was broad and largely on highlighting “Special” craft through market segments. This report is a continuation of this Special Vessel Committee, the intent is to further discuss those markets and drill down into specific and unique structural configurations. The ISSC 2022 Special Vessel Committee has included the additional year (i.e., originally ISSC 2021) delay due to the COVID-19 global pandemic to gather further research and investigate additional topics for the report. A benchmark has been undertaken by the Committee investigating various calculation and analysis methods on window glazing as a structural material and a discussion on this study is in the Appendix. Additionally, during the Committee’s research, a few of the Committee members were able to present a paper at the 2020 High Speed Marine Vessel (HSMV) Conference, Comparative Assessment of Rule-Based Design on the Pressures and Resulting Scantlings of High Speed Powercraft (Souppez et al., 2020), which was well received by the high-speed vessel community. Finally, new markets are emerging in both commercial and government sectors, so the Committee has also included a Chapter on emerging trends to cast a light on structural uncertainties in new technologies and categories of Special Vessels. 1.1 ISSC 2018 special craft recommendations and official discusser feedback A discussion in Volume III of the ISSC 2018 V.5 Special Craft report outlines a few areas and recommendations that the 2018 Official Discusser touched on for the follow-on Committee and audience of the Committees presentation at the Congress provided, those we have addressed in this report are as follows: - The Committee name change to Special Vessels in lieu of Special Craft due to certain expectations that the word “Craft” carries. - A discussion on cyclic loading on composite structures in Chapter 2, which is a special topic relating to higher speed vessels. - The Committee focus on green, emission reducing vessels, Chapter 4, as a new emerging trend in the category of Special structural configurations. - A discussion and analysis on glazing as a structural material in Appendix A. These topics, along with a few others can be found within the report contents to address and further investigate some of these exciting topics. 1.2 Special vessels addressed The Committee Report is a consolidation of references that reflects recent research and development and topics of discussion in the areas of structural arrangements, loading, analysis technique and uncertainties in Special Vessels. Our Chapters have been categorised into types or markets of Special Vessels, Table 1.

Adiponectin (AdipoQ), a hormone abundantly secreted by adipose tissues, has multiple beneficial functions, including insulin sensitization as well as lipid and glucose metabolism. It has been reported that bone controls energy metabolism through an endocrine-based mechanism. In this study, we observed that bone also acts as an important endocrine source for AdipoQ, and its capacity in osteoblasts is controlled by the forkhead box P1 (FOXP1) transcriptional factor. Deletion of the Foxp1 gene in osteoblasts led to augmentation of AdipoQ levels accompanied by fueled energy expenditure in adipose tissues. In contrast, overexpression of Foxp1 in bones impaired AdipoQ secretion and restrained energy consumption. Chromatin immunoprecipitation sequencing (ChIP-seq) analysis revealed that AdipoQ expression, which increases as a function of bone age, is directly controlled by FOXP1. Our results indicate that bones, especially aged bones, provide an important source of a set of endocrine factors, including AdipoQ, that control body metabolism. © 2021 American Society for Bone and Mineral Research (ASBMR).

Background and Objectives. Gastric cancer (GC) is one of the common gastrointestinal tumors and the third in the mortality rate among tumors. Studies have shown that the human antigen receptor (HuR) is associated with the malignant degree of GC. Methods. We performed qRT-PCR, cell cycle assay, cell migration, and mouse transplantation model analysis in our experiments. It has been clarified that HuR and microRNAs (miRNAs) have important interplays in the regulation of tumor progression. Results. This study found microRNA-133b (miR-133b), as a HuR-sponged miRNA in GC cells. Downregulation of HuR can promote the expression of miR-133b and further affect the downstream cyclin CDC5L. The expressions of miR-133b were slightly lower in GC tissues than adjacent normal tissues. Conclusion. Our studies suggest that HuR and miR-133b are involved in the development and pathological process of GC cells.

Collaborative innovation in interdisciplinary teams is a collaborative approach to accomplish cooperative tasks and achieve innovative goals through a multidisciplinary intersection in the form of member combinations. The purpose of this study was to reveal the neural mechanisms underlying the impact of interdisciplinary collaboration on groups and individuals. Each group of subjects consisted of two people with and without a design background, who were asked to solve two realistic presented problems (RPP), and two design innovation tasks based on RPP adaptations. The study used hyperscanning of functional near-infrared spectroscopic (fNIRS) brain functional imaging techniques to simultaneously record the neural responses of interacting participants in each group. The study compared the innovation performance of interdisciplinary teams with design backgrounds in domain-specific versus domain-general innovation tasks, and the results showed differences in individual and group performance in different types of innovation tasks, with individual fluency being the most significant. The study may provide empirical evidence for the performance benefits of interdisciplinary teams in real-world collaborative co-innovation.

Efficient evaluation of the transient free-surface Green function with sufficient accuracy is the key to employ time-domain panel method in solving hydrodynamic problems. In this paper, series and asymptotic expansions for the Green function and its two spatial derivatives are deduced in detail for the evaluation. New residual functions are derived straightforwardly from the resulted asymptotic formulas and it is concluded that the present residual functions decay rapidly than existing choices. The ultimate algorithm is the combination of Chebyshev approximations and asymptotic expressions, along with a special partitioning line introduced to speed up the evaluation. After comprehensive numerical validation, it is pointed out that the present algorithm owns at least 6 digits of accuracy and sufficient efficiency for practical application. Any efficient method for evaluating the Green function will use the asymptotic expansions. Furthermore, it should be pointed out that the present residual functions can also be conveniently applied to the scheme of interpolation in a precomputed table.

In China, aflatoxin B1 (AFB1) and sterigmatocystin (STC) have been detected in rice but there is no published information on the mycobiota in toxin-contaminated rice samples. Occurrence of AFB1 and STC was evaluated in 100 rice samples. Out of 78 samples of polished rice, 12 and 23 samples had detectable levels of AFB1 and STC, respectively, at mean levels of 0.14 μg/kg and 0.39 μg/kg, respectively. The highest level of STC in polished rice was 2.76 μg/kg in a sample from Hubei province; out of 22 samples of unhusked rice, STC was detected in 12 samples at a mean level of 0.91 μg/kg; however, only one sample contained detectable amounts of AFB1 at a level of 0.2 μg/kg. Seven samples that were positive for both AFB1 and STC were selected and evaluated for the presence of mycobiota; species of Aspergillus and Penicillium predominated. Twelve species of Penicillium and fifteen species of Aspergillus were isolated, of which P. fellutanum and A. candidus were the most frequently found. Further studies are now required to determine which of these species produce AFB1 and STC.

Background Most countries set regulatory values for the total trace element (TE) concentrations in soil, although there is growing interest in using a risk-based approach to evaluate the bioavailable TE using dilute salt extractants or other soil parameters, including pH and organic carbon. The present study compares the current regulatory system (based on total TEs and pH) and a risk-based approach using 0.01 mol L-1 CaCl2 to estimate the bioavailable fraction. Results In total, 150 paired samples of Chinese flowering cabbages (Brassica parachinensis) and their growth soils were collected, and the total and extractable concentrations of chromium (Cr), cadmium (Cd), lead (Pb), nickel (Ni), copper (Cu), zinc (Zn), arsenic (As) and mercury (Hg), as well as soil pH and organic matter content, were measured. No more than 3.33% of the edible parts exceeded Chinese food safety standards, even when growing in soils exceeding the current regulatory thresholds by over 50%. The total soil Cd (1.5 mg kg-1 ), as well as the extractable concentrations of Cd (0.1 mg kg-1 ), Ni (0.03 mg kg-1 ) and Zn (0.1 mg kg-1 ), are the key factors affecting the TE concentrations in B. parachinensis. Conclusion Our findings suggest that the current soil regulatory guidelines for safe production of B. parachinensis are overly strict and conservative. A risk-based approach based on the extractable TE concentrations would provide a better indication for plant uptake of soil TEs and avoid the waste of farmlands that can still produce safe vegetables. Future research should focus on providing crop-specific available TE concentration guidelines to promote effective utilization of farmlands. © 2021 Society of Chemical Industry.

Neuregulin‐1 (NRG‐1) is reported to be cardioprotective through the extracellular‐regulated protein kinase (ERK) 1/2 pathway in myocardial ischaemia‐reperfusion injury (MIRI). NOX4‐induced ROS activated NLRP3 inflammasome and exacerbates MIRI. This study aims to investigate whether NRG‐1 can suppress NOX4 by ERK1/2 and consequently inhibit the NLRP3/caspase‐1 signal in MIRI. The myocardial infarct size (IS) was measured by TTC‐Evans blue staining. Immunohistochemical staining, real‐time quantitative PCR (RT‐qPCR) and Western blotting were used for detection of the factors, such as NOX4, ERK1/2, NLRP3, caspase‐1 and IL‐1β .The IS in the NRG‐1 (3 μg/kg, intravenous) group was lower than that in the IR group. Immunohistochemical analysis revealed NRG‐1 decreased 4HNE and NOX4. The RT‐qPCR and Western blot analyses revealed that NRG‐1 mitigated the IR‐induced up‐regulation of NOX4 and ROS production. Compared with the IR group, the NRG‐1 group exhibited a higher level of P‐ERK1/2 and a lower level of NLRP3. In the Langendorff model, PD98059 inhibited ERK1/2 and up‐regulated the expression of NOX4, NLRP3, caspase‐1 and IL‐1β, which exacerbated oxidative stress and inflammation. In conclusion, NRG‐1 can reduce ROS production by inhibiting NOX4 through ERK1/2 and inhibit the NLRP3/caspase‐1 pathway to attenuate myocardial oxidative damage and inflammation in MIRI.

The application of information and communication technology in court enhances case management efficiency. This brief note discusses the experience with e-courts in China. The experience has been predominantly positive. The important step going forward is to integrate state-of-the-art technology with best practices so that the use of technology becomes a driving force for judicial reform in China.

Ship parametric roll is one of the main reasons for marine accidents and is introduced into the second-generation intact stability criteria by the International Maritime Organization (IMO) recently. In this paper, a 6-DOF three-dimensional time-domain model based on the IRF (Impulse Response Function) method is constructed to predict large-amplitude ship motions and investigate the phenomenon of parametric roll in head waves as well as major factors. The F-K forces and the restoring forces are calculated on the instantaneous wet surface while the radiation and diffraction forces are kept linear and transformed from frequency-domain results calculated with the three-dimensional Havelock form translating-pulsating source green function method. The proposed weakly nonlinear time-domain model is used to simulate motions of the C11 containership, which predicts the occurrence of the parametric roll successfully and shows a good agreement with the experimental data in amplitude. The inner mechanism of parametric roll is revealed by investigating the time-history and resonance frequencies of restoring forces and coefficients numerically.

Objective Apolipoprotein C3 (ApoC3) is a regulator of triglyceride metabolism and inflammation, and its plasma levels are positively correlated with the progression of diabetic nephropathy (DN) in patients. However, the role and underlying mechanism of ApoC3 in DN remain unclear. Methods Diabetes was induced in ApoC3 transgenic (Tg) and knockout (KO) mice by injection of streptozotocin. We studied the effect of ApoC3 on type 1 DN after 4 months of diabetes. Plasma glucose and lipid levels, renal function parameters and inflammation- and fibrogenesis-related gene and protein expression levels were studied. In vitro, human mesangial cells (HMCs) were incubated with high levels of glucose or/and triglyceride-rich lipoproteins (TRLs) with a high or low ApoC3 content isolated from Tg or wild-type (WT) mice, respectively, to explore the mechanisms of ApoC3 on development of DN. Results We found that compared to WT mice, Tg mice exhibited hypertriglyceridemia (HTG), aggravated early renal function injury and inflammation, enlarged glomerular and mesangial surface areas, renal lipid deposition and elevated fibrogenesis-related gene expression levels after 4 months of diabetes. ApoC3 overexpression activated the renal Toll-like receptor 2 (TLR2) and nuclear factor-κB (NF-κB) signaling pathways and increased the renal gene and protein expression levels of the downstream inflammatory factors TNF-α, VCAM-1 and MCP-1. Unfortunately, we did not find that ApoC3 deficiency had an obvious protective effect against DN. In vitro, we found that TRLs with a high ApoC3 content increased the gene and protein expression levels of inflammation- and fibrogenesis-related factors in HMCs compared to those following administration of the same concentration of TRLs with a low ApoC3 content. These effects of ApoC3 were inhibited by blockade of TLR2 or NF-κB. Conclusions These findings suggest that ApoC3 aggravates early-stage DN by activating the renal TLR2/NF-κB pathway which is partially independent of HTG.

Leaf size is an important agronomic trait directly affecting yield in rice, and thus understanding the genes determining leaf size is important in breeding. In this study, one Leaf Mutant 7 (lm7) with small leaf size was isolated using ethyl methane sulphonate (EMS) mutagenesis from the japonica Zhenggeng 1925. MutMap by whole genome resequencing of phenotypic bulks revealed that LM7 is likely located in the 133 kb region on chromosome 7 using F2 population from a cross between lm7 and wild-type (WT) Zhenggeng 1925. The candidate gene encoding heat shock protein OsHSP40 for LM7 was functionally validated. Disruption of this gene in Oshsp40 mutants significantly reduced the leaf size compared with that of WT in rice. Microscopic examination showed that OsHSP40 modulated leaf size via regulating the veins formation and cell size/cell number. Nucleotide diversity analysis indicated that a single nucleotide polymorphism (SNP) variation of C to T in the coding region of OsHSP40 may cause small leaves among rice accessions. Therefore, the natural variation of OsHSP40 contributing to leaf size might be useful for rice breeding.
