Understanding the architecture of a processor can be uninteresting and deterring for computer science students, since low-level details of computer architecture are often perceived to lack real-world impact. These courses typically have a strong practical component where students learn the fundamentals of the computer architecture and the handling of input/output operations through the development of simple programs in a low-level assembly programming language. Since these practical sessions require a strong involvement, student attendance and withdrawal rates are poor, lowering academic results and introducing a negative feedback loop that preconditions students to dislike them. This article introduces a new methodology for the practical sessions of Computer Organization and Design courses. This methodology disavows the use of simulators and focuses on actual hardware to promote a feeling of proximity to the execution and outcome of the programs. The proposed setup uses Raspberry Pi devices to encourage students to work autonomously, due to their low cost, capability of running an OS, and rich ecosystem of simple hardware devices. The setup is completed with RISC OS, which combines a simple window-based graphical interface with a low-level management of the hardware without requiring software abstraction layers. The article presents the methodology and the UCDebug tool, developed to help students debug their codes in RISC OS. After the introduction of the new setup at the University of Cantabria, academic results and student satisfaction have improved. The setup has also allowed to sustain a similar organization of the courses throughout the COVID-19 pandemic.

With ever-increasing incidence and high metastatic potential, cutaneous melanoma is the deadliest skin cancer. Risk prediction based on the Tumor-Node-Metastasis (TNM) staging system has medium accuracy with intermediate IIB-IIIB stages, as roughly 25% of patients with low-medium-grade TNM, and hence a favorable prognostic, undergo an aggressive disease with short survival and around 15% of deaths arise from metastases of thin, low-risk lesions. Therefore, reliable prognostic biomarkers are required. We used genomic and clinical information of melanoma patients from the TCGA-SKCM cohort and two GEO studies for discovery and validation of potential biomarkers, respectively. Neither mutation nor overexpression of major melanoma driver genes provided significant prognostic information. Conversely, expression of MGRN1 and the melanocyte-specific genes MLANA, PMEL, and TYRP1 provided a simple 4-gene signature identifying with high-sensitivity (>80%), low-medium TNM patients with adverse outcomes. Transcriptomic analysis of tumors with this signature, or from low-medium-grade TNM patients with poor outcomes, revealed comparable dysregulation of an inflammatory response, cell cycle progression, and DNA damage/repair programs. A functional analysis of MGRN1-knockout cells confirmed these molecular features. Therefore, the simple MGRN1-MLANA-PMEL-TYRP1 combination of biomarkers complemented TNM staging prognostic accuracy and pointed to the dysregulation of immunological responses and genomic stability as determinants of a melanoma outcome. Background/Objectives: Saliva is gaining importance as a diagnostic sample in pigs. The aim of this research was to evaluate a panel of salivary analytes in three porcine diseases and establish predictive models to detect them. Methods: Saliva samples were obtained from healthy pigs (n = 97) and pigs affected by meningitis due to Streptococcus suis (n = 118), diarrhea due to enterotoxigenic Escherichia coli (ETEC, n = 77), and porcine reproductive and respiratory syndrome (PRRS, n = 52). The following biomarkers were analyzed: adenosine deaminase (ADA), haptoglobin (Hp), calprotectin (Calp), aldolase, alpha-amylase (sAA), lactate dehydrogenase (LDH), total protein (TP), and advanced oxidation protein products (AOPPs). Predictive models based on binary logistic regression and decision trees combining those analytes for detecting specific diseases were constructed. Results: The results showed a different biomarker profile between the groups. S. suis and ETEC pigs showed higher values of ADA, Hp, Calp, aldolase, sAA, LDH, and TP than healthy pigs. Pigs with PRRS showed higher values of Hp, Calp, sAA, and LDH than healthy animals. The constructed predictive models showed overall accuracies of over 78% and 87% for differentiating ETEC and PRRS, respectively, whereas the models did not accurately predict S. suis infection. Conclusions: Salivary analytes show different changes in pigs depending on the disease, and the combination of these analytes can contribute to the prediction of different diseases. (1) Background: the penalty kick is an action in football that decides many matches. The aim of this research was to establish the success rate of penalty kicks in Spain between the seasons 2012/2013 and 2019/2020 considering different key performance variables. (2) Methods: A total of 941 penalty kicks were analysed from eight seasons of the first division of the Spanish men's football league (La Liga). An observational methodology was used and, in order to obtain the results, different analytical techniques were employed: descriptive and chi square with a significance level of p < 0.05. (3) Results: The effectiveness of penalty kicks was 76.7%. Most of the penalties were taken by the home team (60.9%), with a tie on the scoreboard (44.2%) and within the last 30 min of the match (41.9%). The highest success rate was observed for balls kicked to the upper side of the goal (100%), even though the highest number of penalties were taken low, to one of the two sides of the goal (57.0%). Up to 5.6% of the penalties were not kicked inside the goal. (4) Conclusions: Effectiveness does not depend on home–visitor status, player laterality, score or remaining match time, studied independently. (1) Background: The analysis of women's tennis performance has not been extensively explored by the scientific community, necessitating further research to understand the tactical actions occurring in matches. This research aimed to examine the chance of winning a point in professional women's tennis based on the key variables that influence performance in the sport. (2) Methods: Data from 3239 points were examined across three distinct court surfaces, sourced from the final rounds (starting with the quarterfinals) of three Grand Slam tournaments in the 2021 season. An observational methodology was employed, using various analysis techniques: descriptive and chi-square analyses, with a significance level of p < 0.05. (3) Results: The probability of winning a point on the first serve was 61% on clay, 70% on grass, and 69% on hard courts. For second serves, the probability of winning the point varied between 55% and 57%, depending on the court surface. Additionally, the majority of points, ranging from 70% to 71%, concluded with short rallies, involving one to four shots. On clay courts, the server won up to 65% of points with a first serve and a short rally, while the success rate increased to 75% on both grass and hard courts. For medium-length rallies (5–8 shots), the probability of winning the point dropped to 55–57%. The point outcome (winner, forced error, and unforced error) varied according to court surface, serve type, and rally length. The first near-infrared fluorescent turn-on sensor for the detection of nitroxyl (HNO), the one-electron reduced form of nitric oxide (NO), is reported. The new copper-based probe, CuDHX1, contains a dihydroxanthene (DHX) fluorophore and a cyclam derivative as a Cu(II) binding site. Upon reaction with HNO, CuDHX1 displays a five-fold fluorescence turn-on in cuvettes and is selective for HNO over thiols and reactive nitrogen and oxygen species. CuDHX1 can detect exogenously applied HNO in live mammalian cells and in conjunction with the zinc-specific, green-fluorescent sensor ZP1 can perform multicolor/multianalyte microscopic imaging. These studies reveal that HNO treatment elicits an increase in the concentration of intracellular mobile zinc.

In this report, different handling conditions at slaughterhouse were studied to assess changes in salivary biomarkers. For this purpose, finishing pigs were divided into two groups, one in which handling was improved to minimize stress (Group A, n = 24, transported and stabled at the slaughterhouse at low density without mixing with unfamiliar animals throughout the whole process) and another one in which animals had a more stressful handling process (Group B, n = 24, transported and stabled at high density with unfamiliar animals). Saliva samples were taken the day before transport to the slaughterhouse at 8:00 a.m. (B0) and 12:00 a.m. (B4), and the day of slaughter just after unloading animals at the slaughterhouse at approximately 8:00 a.m. (S0) and after 4 h of lairage at approximately 12:00 a.m. (S4). Group B showed significantly higher cortisol, total esterase activity, oxytocin, adenosine deaminase and haptoglobin levels than the Group A at both S0 and S4 sampling times, and higher levels of calprotectin and creatine kinase at S4 sampling time. This report indicates that differences in the way in which the pigs are handled at the slaughterhouse can lead to changes in salivary biomarkers and opens the possibility of the use of biomarker at slaughter to monitor handling conditions.
