2. Temporal and spatial patterns of Trichuris trichiura eggs: a potential threat to human health in Pakistan

Abstract

This study investigates the presence of Trichuris trichiura eggs in soil samples collected from urban areas in Lahore, Pakistan. A total of 3600 soil samples were collected over two years from Lahore’s urban regions. The detection of helminth eggs in these samples was performed using sodium hypochlorite (NaOCl) as a diagnostic technique. The study reveals an overall prevalence rate of T. trichiura at 0.97 % (35 out of 3600) in the contaminated soil samples from Lahore’s slum areas. When analyzing the data by geographical areas, the study found the highest prevalence of T. trichiura in Allama Iqbal Town (1.83 %, 11 out of 600), followed by Samanabad (1.16 %, 7 out of 600), Wapda Town (1.00 %, 6 out of 600), Gulberg (1.00 %, 6 out of 600), and Cantt (0.50 %, 3 out of 600). Conversely, Valencia Town had the lowest prevalence rate at 0.33 % (2 out of 600). However, these variations in prevalence rates were not statistically significant (p = 0.117). Prevalence rates of T. trichiura’s eggs varied significantly across different sampling seasons (p>0.001). In autumn, a total of 900 soil samples were collected, with 19 samples (2.11 %) testing positive for T. trichiura. This rate was notably higher compared to the prevalence rates observed in winter, spring, and summer, which were 0.66 %, 0.22 %, and 0.88 %, respectively. Regarding the sampling months, the study observed a significantly higher prevalence during September (3.33 %, 10 out of 300), followed by October (2.33 %, 7 out of 300), and August (1.33 %, 4 out of 300). Prevalence rates gradually decreased in other months, ranging from 1 % to 0.33 % (3 to 1 out of 300), with no parasite detection in March (0 %, 0 out of 300) (p < 0.001). This research underscores soil contamination due to fecal waste and highlights public unawareness of parasite biology, driven by open defecation practices.

Abstract

Context. Teegarden's Star is the brightest and one of the nearest ultra-cool dwarfs in the solar neighbourhood. For its late spectral type (M7.0V), the star shows relatively little activity and is a prime target for near-infrared radial velocity surveys such as CARMENES. Aims. As part of the CARMENES search for exoplanets around M dwarfs, we obtained more than 200 radial-velocity measurements of Teegarden's Star and analysed them for planetary signals. Methods. We find periodic variability in the radial velocities of Teegarden's Star. We also studied photometric measurements to rule out stellar brightness variations mimicking planetary signals. Results. We find evidence for two planet candidates, each with $1.1M_\oplus$ minimum mass, orbiting at periods of 4.91 and 11.4 d, respectively. No evidence for planetary transits could be found in archival and follow-up photometry. Small photometric variability is suggestive of slow rotation and old age. Conclusions. The two planets are among the lowest-mass planets discovered so far, and they are the first Earth-mass planets around an ultra-cool dwarf for which the masses have been determined using radial velocities., Comment: A&A 627, A49. 26 pages, 17 figures, 6 tables. Abstract

La creciente preocupación por el bienestar de los animales en sistemas de producción intensivos ha llevado al desarrollo de diferentes programas de enriquecimiento ambiental, para mejorar el grado de diversidad comportamental de los animales de granja y mejorar su bienestar. El objetivo de este trabajo fue determinar los efectos de un enriquecimiento ambiental alimenticio sobre la respuesta comportamental de ovejas gestantes, lactantes con corderos y secas. A partir del rebaño de la Facultad de Veterinaria de la Universidad Complutense de Madrid, 36 ovejas de la raza Castellana se distribuyeron aleatoriamente en 9 lotes de 4 ovejas, 3 grupos por estado fisiológico. El enriquecimiento nutricional consistió en proporcionar el 25 % de la ración de volumen, heno de alfalfa, en una malla suspendida del techo y una plataforma para facilitar el acceso, administrando también heno en el comedero habitual. Se grabaron las frecuencias y el tiempo que las ovejas dedicaron a la expresión de los distintos comportamientos. Las ovejas lactantes usaron de inmediato el enriquecimiento, mientras que las gestantes aprendieron, porque no fueron a comer a la malla el día 1, pero sí los días sucesivos. Al cuarto día la frecuencia y el tiempo que el conjunto de ovejas dedicó a comer en los diferentes sitios era mayor, así como las interacciones sociales en gestantes y secas, siendo menor el comportamiento de pie observando. Abstract

SARS-CoV-2, a positive-strand RNA virus has caused devastating effects. The standard method for COVID diagnosis is based on polymerase chain reaction (PCR). The method needs expensive reagents and equipment and well-trained personnel and takes a few hours to be completed. The search for faster solutions has led to the development of immunological assays based on antibodies that recognize the viral proteins that are faster and do not require any special equipment. Here we explore an innovative analytical approach based on the sandwich oligonucleotide hybridization which can be adapted to several biosensing devices. PolyPurine Reverse Hoogsteen hairpins (PPRHs) oligonucleotides that form high affinity triplexes with the polypyrimidine target sequences are used for the efficient capture of the viral genome. Then, a second labelled oligonucleotide is used to detect the formation of a trimolecular complex in a similar way than antigen tests do. The limit of detection is around 0.01 nM (a few femtomoles) without the use of any amplification steps. The Triplex Enhanced Nucleic Acid Detection Assay (TENADA) can be readily adapted for the detection of any pathogen requiring only the knowledge of the pathogen genome sequence.

Abstract

How important is choice of conditioning regimen in allogeneic haematopoietic stem cell transplantation (HSCT) for sickle cell disease (SCD)? We compared HSCT outcomes by conditioning regimen in paediatric patients with SCD from the EBMT registry. In 2010-2020, 251 patients aged <18 years underwent a first matched sibling donor (MSD) HSCT with conditioning based on busulfan-fludarabine (bu-flu; n = 89) or treosulfan-fludarabine (treo-flu; n = 162). In the bu-flu and treo-flu groups, 51.7% and 99.4% of patients, respectively, received thiotepa. Median follow-up was 2.7 years. Two-year overall survival (OS) was 98.7% (95% confidence interval [CI]: 90.9-99.8) with bu-flu and 99.3% (95% CI: 95.2-99.9) with treo-flu (p = 0.63). Grade III-IV acute graft-versus-host disease (GVHD) at 100 days was 2.4% (95% CI: 0.4-7.5) and 0.6% (0.1%-3.2%) for bu-flu and treo-flu respectively (p = 0.25). The 2-year incidence of extensive chronic GVHD was 1.5% (95% CI: 0.1-7.3) with bu-flu and 8.0% (95% CI: 4.1-13.3) with treo-flu (p = 0.057). These multinational data confirm the excellent curative capacity of MSD HSCT with myeloablative conditioning. Both conditioning regimens yielded excellent OS, low rates of acute and chronic GVHD, and low rates of graft failure.

Abstract

Anaplasma phagocytophilum are obligatory intracellular bacteria that preferentially replicate inside leukocytes by utilizing biological compounds and processes of these primary host defensive cells. In this study, bioinformatics analysis was conducted to further characterize A. phagocytophilum– host interactions using the neutrophil-like model of human Caucasian promyelocytic leukemia HL60 cells. We detected a hierarchy of molecules involved in A. phagocytophilum-HL60 interactions with overrepresentation in infected human cells of proteins involved in the reactive oxygen species (ROS) pathway and cell surface monocyte markers. As A. phagocytophilum phagocytosis by neutrophils is inhibited, the results suggested a possible explanation for our bioinformatics data: radical oxygen compounds could induce the killing of bacteria activating NETosis, a unique form of defense mechanism resulting in cell death that is characterized by the release of decondensed chromatin and granular contents to the extracellular space, forming neutrophil extracellular traps (NETs) to eliminate invading microorganisms. Thus, we confirmed the existence of a low NETosis induced in A. phagocytophilum- infected cells by immunofluorescence (IF) experiments. These results provide new insights into the complex mechanisms that govern immune response during A. phagocytophilum host interactions.

Abstract

Significant advances in supportive care for patients with transfusion-dependent thalassemia major (TDT) have improved patients' life expectancy. However, transfusion-associated iron overload remains a significant barrier to long-term survival with good quality of life. Today, allogeneic hematopoietic stem cell transplantation (HSCT) is the current curative standard of care. Alongside selection of the best available donor, an optimized conditioning regimen is crucial to maximize outcomes for patients with TDT undergoing HSCT. The aim of this retrospective analysis was to investigate the role of busulfan-fludarabine-based and treosulfan-fludarabine-based conditioning in TDT patients undergoing HSCT. We included 772 patients registered in the European Society for Blood and Marrow Transplantation (EBMT) database who underwent first HSCT between 2010 and 2018. Four hundred ten patients received busulfan-fludarabine-based conditioning (median age 8.6 years) and 362 patients received treosulfan-fludarabine-based conditioning (median age 5.7 years). Patient outcomes were retrospectively compared by conditioning regimen. Two-year overall survival was 92.7% (95% confidence interval: 89.3-95.1%) after busulfan-fludarabine-based conditioning and 94.7% (95% confidence interval: 91.7-96.6%) after treosulfan-fludarabine-based conditioning. There was a very low incidence of second HSCT overall. The main causes of death were infections, graft-versus-host disease, and rejection. In conclusion, use of busulfan or treosulfan as the backbone of myeloablative conditioning for patients with TDT undergoing HSCT resulted in comparably high cure rates. Long-term follow-up studies are warranted to address the important issues of organ toxicities and gonadal function.

Abstract

This technical report is based on the study for the evaluation of the processes of reuse and recycling of reverse osmosis components and membranes in the Canary Islands and Macaronesia, within the DESAL+ project and in the framework of the DESAL+ LIVING LAB platform, coordinated by the Canary Islands Technological Institute (ITC) and the Canary Islands Agency for Research, Innovation and Information Society (ACIISI), with the support of the Interreg-MAC Programme. Reverse osmosis membranes could be reused in the same or another desalination plant by replacing the membranes in the first, dirtier positions with those in the last, less damaged positions. Also, by changing the best first-stage membranes to the second and vice versa, the useful life of these membranes could be extended through chemical cleaning and a second life could be given in tertiary treatment plants, reuse in industrial processes where they use special reverse osmosis membranes and degrade rapidly, in processes with leachate from landfill waste and also an interesting option is the oxidation of reverse osmosis elements to obtain nanofiltration, ultrafiltration or microfiltration membranes for the removal of physical dirt. The main categories of thermal processing recycling commonly used in industry include incineration and pyrolysis to produce energy, gas and fuel. These processes can be applied to mixed plastic waste, such as the combination of materials used in the manufacture of reverse osmosis membranes. The recycling of reverse osmosis elements from desalination plants is shown as an opportunity, nowadays existing pioneering initiatives in Europe. Energy recovery, via incineration, is feasible but is not considered in accordance with the environmental, social and political problems that this may generate. However, the recycling of the reverse osmosis elements via pyrolytic industry for fuel production can be centralized in a new industry already planned in the Canary Islands and all the osmosis membranes that are obsolete can be sent there. This is a technically and economically viable business opportunity with a promising future in today's recycling market as studied in the technical report. [ABSTRACT FROM AUTHOR]

Abstract

The growing demand for clean and efficient fuels in the world reflects the rapid growth in automotive vehicles and more stringent environmental regulations. Hence, the refining industry must employ diverse strategies to obtain a gasoline pool made up of streams from different processes focused on improving the fuel quality and the octane ratings suitable for the current demanding automotive performance. The composition of gasoline varies mainly from each country's policies, climate, environmental regulations, financial capacity, and local producer's oil refining infrastructure. However, a generally accepted composition by source of the gasoline pool is: FCC naphtha and reformate, making up about 60%, light straight-run naphtha and alkylate gasoline, around 30%, isomerate, close to 5%, and variable proportions of butane, oxygenating agents, such as methyl-ter-butyl ether (MTBE), ter-amyl-methyl ether (TAME) and/or ethanol and additives for the remaining 5%. This review is focused on the recent research in the advancement and development of catalysts for the different processes used by the oil refining industry to improve the composition and properties of gasoline fuels. A detailed section on the Fischer-Tropsch process, where liquid hydrocarbons are potentially employed to further produce clean gasoline, is also discussed. A section devoted to research on biogasoline as a potential renewable fuel is also addressed. Finally, a discussion of the physicochemical properties of the gasoline blending components is also provided. [ABSTRACT FROM AUTHOR]
