Plant root-nodule symbiosis (RNS) with mutualistic nitrogen-fixing bacteria is restricted to a single clade of angiosperms, the Nitrogen-Fixing Nodulation Clade (NFNC), and is best understood in the legume family. Nodulating species share many commonalities, explained either by divergence from a common ancestor over 100 million years ago or by convergence following independent origins over that same time period. Regardless, comparative analyses of diverse nodulation syndromes can provide insights into constraints on nodulation-what must be acquired or cannot be lost for a functional symbiosis-and the latitude for variation in the symbiosis. However, much remains to be learned about nodulation, especially outside of legumes. Here, we employed a large-scale phylogenomic analysis across 88 species, complemented by 151 RNA-seq libraries, to elucidate the evolution of RNS. Our phylogenomic analyses further emphasize the uniqueness of the transcription factor NIN as a master regulator of nodulation and identify key mutations that affect its function across the NFNC. Comparative transcriptomic assessment revealed nodule-specific upregulated genes across diverse nodulating plants, while also identifying nodule-specific and nitrogen-response genes. Approximately 70% of symbiosis-related genes are highly conserved in the four representative species, whereas defense-related and host-range restriction genes tend to be lineage specific. Our study also identified over 900 000 conserved non-coding elements (CNEs), over 300 000 of which are unique to sampled NFNC species. NFNC-specific CNEs are enriched with the active H3K9ac mark and are correlated with accessible chromatin regions, thus representing a pool of candidate regulatory elements for genes involved in RNS. Collectively, our results provide novel insights into the evolution of nodulation and lay a foundation for engineering of RNS traits in agriculturally important crops.

Abstract The redox state of arc mantle has been considered to be more oxidized and diverse than that of the mid-ocean ridge, but the cause of the variation is debated. We examine the redox state of the Cenozoic global arc mantle by compiling measured/calculated fO2 of olivine-hosted melt inclusions from arc magma and modeled fO2 based on V/Sc and Cu/Zr ratios of arc basaltic rocks. The results indicate that the redox state of Cenozoic arc mantle is latitude dependent, with less oxidized arc mantle in the low latitudes, contrasting with a near constant across-latitude trend in the mid-ocean ridges. We propose that such a latitude-dependent pattern in the arc mantle may be controlled by the variation in the redox state of subducted sediment, possibly related to a latitudinal variation in the primary production of phytoplankton, which results in more organic carbon and sulfide deposited on the low-latitude ocean floor. Our findings provide evidence for the impact of the surface environment on Earth’s upper mantle.

Abstract Background Salt sensitivity of blood pressure (SSBP) is an intermediate phenotype of hypertension and is a predictor of long-term cardiovascular events and death. However, the genetic structures of SSBP are uncertain, and it is difficult to precisely diagnose SSBP in population. So, we aimed to identify genes related to susceptibility to the SSBP, construct a risk evaluation model, and explore the potential functions of these genes. Methods and results A genome-wide association study of the systemic epidemiology of salt sensitivity (EpiSS) cohort was performed to obtain summary statistics for SSBP. Then, we conducted a transcriptome-wide association study (TWAS) of 12 tissues using FUSION software to predict the genes associated with SSBP and verified the genes with an mRNA microarray. The potential roles of the genes were explored. Risk evaluation models of SSBP were constructed based on the serial P value thresholds of polygenetic risk scores (PRSs), polygenic transcriptome risk scores (PTRSs) and their combinations of the identified genes and genetic variants from the TWAS. The TWAS revealed that 2605 genes were significantly associated with SSBP. Among these genes, 69 were differentially expressed according to the microarray analysis. The functional analysis showed that the genes identified in the TWAS were enriched in metabolic process pathways. The PRSs were correlated with PTRSs in the heart atrial appendage, adrenal gland, EBV-transformed lymphocytes, pituitary, artery coronary, artery tibial and whole blood. Abstract The electron/ion density/temperature and ion velocities observed by the ROCSAT-1 and DEMETER satellites are used to examine the daytime wavenumber-4 (WN4) feature in the equatorial/low latitude ionosphere during various months and solar activity levels of 1999–2010. A moving median process has been employed to isolate WN4 features and calculate their amplitudes, while the upward ion drift is used to estimate electric fields. The ROCSAT-1 and DEMETER ion density, ion temperature, and ion velocity generally yield prominent WN4 features over the center of Pacific Ocean, the west side of South America, the center of the Atlantic Ocean, and Southern India. The correlation coefficient between the deviation of ion density and upward ion drift is significant during high solar activity of 1999–2004, while it approaches to zero during low solar activity of 2004–2010. This confirms that the longitudinal variation of the upward ion drift is essential during high solar activity, and the associated amplitude of dynamo eastward electric field is in the range of 0.10–0.14 mV/m, which is 15–19% of daily dynamo electric field. By contrast, the deviation of the ion density and the northward field-aligned ion flow show a clear anti-correlation which yields a maximum coefficient in August during low solar activity but no correlation during high solar activity. These indicate that the longitudinal variation of the meridional field-aligned ion flow could play an important role during low solar activity, and its amplitude is in the range of 10.44–13.91 m/s, which is 10–13% of the ambient ion flows.

Objectives To evaluate changes in health outcomes between years 2 and 3 after discharge following COVID-19 and to identify risk factors for poor health 3-year post-discharge.Design This is a multicentre observational cohort study.Setting This study was conducted in two centres from Wuhan, China.Participants Eligibility screening has been performed in 3988 discharged laboratory-confirmed adult COVID-19 patients. Exclusion criteria were refusal to participate, inability to contact and death before follow-up. The WHO COVID-19 guidelines on defining disease severity were adopted.Results 1594 patients participated in the 1-year, 2-year and 3-year follow-ups, including 796 (49.9%) male patients, and 422 (26.5%) patients were classified in the severe disease group. 3 years after discharge, 182 (11.4%) patients still complained of at least one symptom. The most common symptoms were fatigue, myalgia, chest tightness, cough, anxiety, shortness of breath and expectoration. Fatigue or myalgia, the most common symptom cluster, frequently coexisted with chest symptoms and anxiety. Symptom persistence between years 2 and 3 was reported in 70 patients (4.4%) for which intensive care unit (ICU) admission was a risk factor (p=0.038). Of the 1586 patients who completed the chronic obstructive pulmonary disease assessment test (CAT), 97 (6.1%) scored ≥10, with older age being associated with CAT ≥10 (p=0.007).Conclusions Between years 2 and 3 after SARS-CoV-2 infection, most patients returned to an asymptomatic state, and only a few were still symptomatic. ICU admission was a risk factor for symptom persistence.

Jasmonic acid is a crucial phytohormone that plays a pivotal role, serving as a regulator to balancing plant development and resistance. However, there are analogous and distinctive characteristics exhibited in JA biosynthesis, perception, and signal transduction pathways in both herbaceous and woody plants. Moreover, the majority of research subjects have predominantly focused on the function of JA in model or herbaceous plants. Consequently, there is a significant paucity of studies investigating JA regulation networks in woody plants, particularly concerning post-transcriptional regulatory events such as alternative splicing (AS). This review article aims to conduct a comprehensive summary of advancements that JA signals regulate plant development across various woody species, comparing the analogous features and regulatory differences to herbaceous counterparts. In addition, we summarized the involvement of AS events including splicing factor (SF) and transcripts in the JA regulatory network, highlighting the effectiveness of high-throughput proteogenomic methods. A better understanding of the JA signaling pathway in woody plants has pivotal implications for forestry production, including optimizing plant management and enhancing secondary metabolite production.

Summary Community-acquired bacterial meningitis (CABM) is the main cause of morbidity and mortality in children. The epidemiology of CABM is regional and highly dynamic. To clarify the diagnostic status and epidemiological characteristics of children with CABM in this region, and pay attention to the disease burden, so as to provide evidence for the prevention and treatment of CABM. By retrospective case analysis, the clinical data of 918 CABM cases in children aged 0–14 years in Zhejiang Province from January, 2019 to December, 2020 were collected. The etiological diagnosis rate of CABM in children was 23.1%, the annual incidence rate 4.42–6.15/100,000, the annual mortality rate 0.06–0.09/100,000,the cure and improvement rate 94.4%, and the case fatality rate 1.4%. The total incidence of neuroimaging abnormalities was 20.6%. The median length of stay for CABM children was 20(16) days, with an average cost of 21,531(24,835) yuan. Abstract The China Seismo-Electromagnetic Satellite (CSES), with a sun-synchronous orbit at 507 km altitude, was launched on 2 February 2018 to investigate pre-earthquake ionospheric anomalies (PEIAs) and ionospheric space weather. The CSES probes manifest longitudinal features of four-peak plasma density and three plasma depletions in the equatorial/low-latitudes as well as mid-latitude troughs. CSES plasma and the total electron content (TEC) of the global ionosphere map (GIM) are used to study PEIAs associated with a destructive M7.0 earthquake and its followed M6.5 and M6.3/M6.9 earthquakes in Lombok, Indonesia, on 5, 17, and 19 August 2018, respectively, as well as to examine ionospheric disturbances induced by an intense storm with the Dst index of − 175 nT on 26 August 2018. Anomalous increases (decreases) in the GIM TEC and CSES plasma density (temperature) frequently appear specifically over the epicenter days 1–5 before the M7.0 earthquake and followed earthquakes, when the geomagnetic conditions of these PEIA periods are relatively quiet, Dst: − 37 to 19 nT. In contrast, TEC and CSES plasma parameter anomalies occur globally in the southern hemisphere during the storm days of 26–28 August 2018. The CSES ion velocity shows that the electric fields of PEIAs associated with the M7.0 earthquake are 0.21/0.06 mV/m eastward and 0.11/0.10 mV/m downward at post-midnight/post-noon on 1–3 August 2018, while the penetration electric fields during the storm periods of 26–28 August 2018 are 0.17/0.45 mV/m westward/downward at post-midnight of 02:00 LT and 0.26/0.26 mV/m eastward/upward at post-noon of 14:00 LT. Spatial analyses on CSES plasma discriminate PEIAs from global effects and locate the epicenter of possible forthcoming large earthquakes. CSES ion velocities are useful to derive PEIA- and storm-related electric fields in the ionosphere.

AbstractThe intelligent monitoring of electric locomotive loading is crucial in unmanned underground systems. A CNN-based monitoring scheme with migration learning was proposed to address efficiency, abnormality, and data acquisition challenges. Locomotive loading datasets are transformed, augmented, and equalized. Our model improves performance and training by modifying the fully connected layer, using optimized learning rate decay and adaptive algorithms. Training in PyTorch, the optimized VGG19-EL migration network achieves 99.85% recognition for 2-classifications, while the optimized RESNET50-EL migration network achieves 97.3% for 10-classifications. Overall, this study proposes a reliable and efficient model for liberating workers and monitoring locomotive loading.

Acute kidney injury (AKI) frequently emerges as a consequential non-neurological sequel to traumatic brain injury (TBI), significantly contributing to heightened mortality risks. The intricate interplay of oxidative stress in the pathophysiology of TBI underscores the centrality of the Keap1-Nrf2/HO-1 signaling pathway as a pivotal regulator in this context. This study endeavors to elucidate the involvement of the Keap1-Nrf2/HO-1 pathway in modulating oxidative stress in AKI subsequent to TBI and concurrently explore the therapeutic efficacy of dimethyl fumarate (DMF). A rat model of TBI was established via the Feeney free-fall method, incorporating interventions with varying concentrations of DMF. Assessment of renal function ensued through measurements of serum creatinine and neutrophil gelatinase-associated lipocalin. Morphological evaluation of renal pathology was conducted employing quantitative hematoxylin and eosin staining. The inflammatory response was scrutinized by quantifying interleukin (IL)-6, IL-1β, and tumor necrosis factor-α levels. Oxidative stress levels were discerned through quantification of malondialdehyde and superoxide dismutase. The apoptotic cascade was examined via the terminal deoxynucleotidyl transferase dUTP deletion labeling assay. Western blotting provided insights into the expression dynamics of proteins affiliated with the Keap1-Nrf2/HO-1 pathway and apoptosis. The findings revealed severe kidney injury, heightened oxidative stress, inflammation, and apoptosis in the traumatic brain injury model. Treatment with DMF effectively reversed these changes, alleviating oxidative stress by activating the Keap1-Nrf2/HO-1 signaling pathway, ultimately conferring protection against AKI. Activating Keap1-Nrf2/HO-1 signaling pathway may be a potential therapeutic strategy for attenuating oxidative stress-induced AKI after TBI.

The routine use of extracorporeal cardiopulmonary resuscitation (ECPR) is not recommended for patients with cardiac arrest. However, ECPR is considered for selected patients with cardiac arrest of reversible cause. Extracorporeal membrane oxygenation (ECMO) provides temporary cardiopulmonary support and adequate perfusion to the end organs, thereby shortening ischemic organ time and minimizing complications. One indication for ECPR therapy is prolonged ventricular fibrillation despite optimal conventional CPR.Here, we report a successful recovery case from ECPR, in which the patient suffered from refractory ventricular fibrillation and was predisposed to severe hyperkalemia. Ventricular fibrillation failed to respond despite prolonged conventional CPR and defibrillation management for 32 min. After successfully initiating ECPR 54 min after cardiac arrest, spontaneous circulation returned sooner. He demonstrated clear consciousness after treatment and was discharged without any neurological disability on day 11.

Alkanes and alkenes are high-value platform chemicals that can be synthesized by microorganisms, utilizing organic residues from agri-food industries and municipalities, thereby offering an alternative opportunity in resource recovery. Current research and technological advancements for the biosynthesis of alkanes and alkenes are mainly impeded by low product titers, obstructing the bioprocess upscaling and large-scale applications. Thus, current scientific investigations aim to improve productivity by utilizing natural and engineered metabolic pathways in various microbial chassis to suppress competing metabolic pathways, coupled with bioprocess optimization. Additionally, to reduce costs, research is being conducted on utilizing inorganic carbon sources such as CO2 to promote the green synthesis of alkanes and alkenes. Therefore, this review critically discusses the opportunities and challenges in alkane and alkene biosynthesis, aiming to examine the current technological advancements. In this review, the limitations of five major metabolic pathways for alkane and alkene biosynthesis are thoroughly discussed, highlighting their shortcomings. Additionally, various techniques, including metabolic engineering, autotrophic metabolic pathways, and new non-biosynthetic routes, are investigated as potential methods to enhance product titers. Furthermore, this review offers valuable insights into the economic and environmental aspects of alkane and alkene biosynthesis while also presenting perspectives for future research directions.

Due to the clinical similarities between pulmonary embolism (PE) and myocardial infarction (MI), physicians often encounter challenges in promptly distinguishing between them, potentially missing the critical window for the correct emergency response. This paper presents a biosensor, termed the PEMI biosensor, which is designed for the identification and quantitative detection of pulmonary embolism or myocardial infarction. The surface of the working electrode of the PEMI biosensor was modified with graphene oxide and silk fibroin to immobilize the mixture of antibodies. Linear sweep voltammetry was employed to measure the current-to-potential mapping of analytes, with the calculated curvature serving as a judgment index. Experimental results showed that the curvature exhibited a linear correlation with the concentration of antigen FVIII, and a linear inverse correlation with the concentration of antigen cTnI. Given that FVIII and cTnI coexist in humans, the upper and lower limits were determined from the curvatures of a set of normal concentrations of FVIII and cTnI. An analyte with a curvature exceeding the upper limit can be identified as pulmonary embolism, while a curvature falling below the lower limit indicates myocardial infarction. Additionally, the further the curvature deviates from the upper or lower limits, the more severe the condition. The PEMI biosensor can serve as an effective detection platform for physicians.

Complexity theory suggests that educational research should consider students' community contexts, because individual students' outcomes are interrelated with community characteristics. While some prior literature finds that higher education enrollment choices are influenced by geography, scholars have largely overlooked the importance of geography and community-level characteristics in admissions outcomes, especially in countries with distinct segments of academic and vocational universities. In this study, we adopt a geo-spatial perspective to examine the spatial distribution of student enrollments at a highly selective Taiwanese technological university, Taipei Tech. Geocoding students' home addresses, our dataset includes geodemographic characteristics and university enrollment data for villages throughout Taiwan. Our analysis shows that geodemographic segmentation of enrollment patterns is only partially driven by socioeconomic conditions of villages and primarily driven by geographic proximity. We also confirm that spatial clustering is evident and that it influences enrollment patterns. Additionally, we provide evidence of a spatial spillover effect; after controlling for village-level characteristics, we show that a village tends to have more students enrolled at Taipei Tech when nearby villages have more students enrolled at Taipei Tech. Although universities such as Taipei Tech have adopted a non-traditional admissions policy to increase diversity, we show that students from remote villages rarely used the policy to enroll at Taipei Tech. Conversely, villages with more well-educated residents were more likely to benefit from the non-traditional admission pipeline. Our findings suggest that offering alternate admissions policies may only partially address challenges related to the geography of opportunity.
